BACKGROUND: Aging-associated left ventricular dysfunction promotes cardiopulmonary fibrogenic remodeling, Group 2 pulmonary hypertension (PH), and right ventricular failure. At the time of diagnosis, cardiac function has declined, and cardiopulmonary fibrosis has often developed. Here, we sought to develop a molecular positron emission tomography (PET)-magnetic resonance imaging (MRI) protocol to detect both cardiopulmonary fibrosis and fibrotic disease activity in a left ventricular dysfunction model. METHODS AND RESULTS: Left ventricular dysfunction was induced by transverse aortic constriction (TAC) in 6-month-old senescence-accelerated prone mice, a subset of mice that received sham surgery. Three weeks after surgery, mice underwent simultaneous PET-MRI at 4.7 T. Collagen-targeted PET and fibrogenesis magnetic resonance (MR) probes were intravenously administered. BackgroundData coordination across multiple health care facilities has become increasingly important for many emerging health care applications. Distrust has been recognized as a key barrier to the success of such applications. Leveraging blockchain technology could provide potential solutions tobuild trust between data providers and receivers by taking advantage of blockchain properties such as security, immutability, anonymity, decentralization, and smart contracts. Many health technologies have empirically proven that blockchain designs fit well with the needs of health care applications with certain degrees of success. However, there is a lack of robust architecture to provide a practical framework for developers to implement applications and test the performance of stability, efficiency, and scalability using standard blockchain designs. A generalized blockchain model is needed for the health care community to adopt blockchain technology and develop applications in a timely fashion. ObjectiveThis study aimed at building a generalized blockchain architecture that provides data coordination functions, including data requests, permission granting, data exchange, and usage tracking, for a wide spectrum of health care application developments. MethodsAn augmented, 3-layered blockchain architecture was built on a private blockchain network. The 3 layers, from bottom to top, are as follows: (1) incorporation of fundamental blockchain settings and smart contract design for data collection; (2) interactions between the blockchain and health care application development environment using Node.js and web3.js; and (3) a flexible development platform that supports web technologies such as HTML, https, and various programing languages. Two example applications, health information exchange (HIE) and clinical trial recruitment, were developed in our design to demonstrate the feasibility of the layered architecture. Case studies were conducted to test the performance in terms of stability, efficiency, and scalability of the blockchain system. ResultsA total of 331,142 simulated HIE requests from accounts of 40,000 patients were successfully validated through this layered blockchain architecture with an average exchange time of 11.271 (SD 2.208) seconds. We also simulated a clinical trial recruitment scenario with the same set of patients and various recruitment criteria to match potential subjects using the same architecture. Potential subjects successfully received the clinical trial recruitment information and granted permission to the trial sponsors to access their health records with an average time of 3.07 seconds. ConclusionsThis study proposes a generalized layered blockchain architecture that offers health technology community blockchain features for application development without requiring developers to have extensive experience with blockchain technology. The case studies tested the performance of our design and empirically proved the feasibility of the architecture in 2 relevant health application domains.

While all reversible receptor-targeting radioligands for positron emission tomography (PET) can be displaced by competition with an antagonist at the receptor, many radiotracers show limited occupancies using agonists even at high doses. [11C]Raclopride, a D2/D3 receptor radiotracer with rapid kinetics, can identify the direction of changes in the neurotransmitter dopamine, but quantitative interpretation of the relationship between dopamine levels and radiotracer binding has proven elusive. Agonist-induced receptor desensitization and internalization, a homeostatic mechanism to downregulate neurotransmitter-mediated function, can shift radioligand-receptor binding affinity and confound PET interpretations of receptor occupancy. In this study, we compared occupancies induced by amphetamine (AMP) in drug-naive wild-type (WT) and internalization-compromised β-arrestin-2 knockout (KO) mice using a within-scan drug infusion to modulate the kinetics of [11C]raclopride. We additionally performed studies at 3 h following AMP pretreatment, with the hypothesis that receptor internalization should markedly attenuate occupancy on the second challenge, because dopamine cannot access internalized receptors. Without prior AMP treatment, WT mice exhibited somewhat larger binding potential than KO mice but similar AMP-induced occupancy. At 3 h after AMP treatment, WT mice exhibited binding potentials that were 15 % lower than KO mice. At this time point, occupancy was preserved in KO mice but suppressed by 60 % in WT animals, consistent with a model in which most receptors contributing to binding potential in WT animals were not functional. These results demonstrate that arrestin-mediated receptor desensitization and internalization produce large effects in PET [11C]raclopride occupancy studies using agonist challenges.

The application of far-infrared blankets has shown certain benefits in health promotion and therapy, such as improving blood circulation and alleviating muscle pain. However, the effects of such blankets on increasing deep sleep, reducing blood pressure, enhancing memory, dilating microvessels for blood flow, reducing chronic inflammation, and decreasing fatigue remain to be studied. We aim to investigate the effects of the DAZZEON αSleep® far-infrared blanket on these indicators. This study adopted a double-blind design, recruiting 24 male participants aged over 45 years, divided into two groups of 12 each: (A) a placebo group and (B) a DAZZEON αSleep® group. The participants used the blanket every night for two weeks, with sleep records taken using a wearable device and blood pressure, blood oxygen levels, arterial stiffness, and surface temperature measured before and after the intervention. Blood samples were collected for an analysis of inflammation and sleep-related blood indicators (serotonin and melatonin), and exercise tests were conducted to assess fatigue improvement. Compared with before the intervention, the blanket significantly increased changes in grip strength and reaction time. Additionally, it significantly increased blood serotonin, melatonin, and nitric oxide concentrations (p < 0.05), thus significantly increasing deep sleep and REM sleep durations (p < 0.05) and improving subjective sleep quality (p < 0.05). This study confirmed that using the DAZZEON αSleep® far-infrared blanket for 14 consecutive days helps to improve blood circulation, reduce vascular age and arterial stiffness, increase serotonin and melatonin levels, and improve sleep quality, as well as enhances muscle strength and reaction time.

To explore the influence of slightly acidic electrolyzed water (SAEW) with different pH and available chlorine contents (ACC) on the germination of tartary buckwheat (Fagopyrum tataricum), the germination rate was taken as the index, and the germination process of tartary buckwheat sprouts with SAEW was optimized by single factor and response surface experiments. Moreover, to obtain good quality selenium-enriched tartary buckwheat sprouts, the effects of different Na2SeO3 concentrations on tartary buckwheat were studied on the basis of SAEW germination, with the total selenium content and germination rate as indexes. The optimum technological parameters were determined as follows: soaking time 2.3 h, pH6.5, germination temperature 21 ℃ and germination time 108 h. Under these conditions, the germination rate was 84.96%, which was close to the predicted value of the response surface group (85.36%), being 14.3% and 11.71% higher than those of distilled water and tap water groups, respectively. The optimal concentration of Na2SeO3 was 60 mg/L, under which the total selenium content of tartary buckwheat sprouts was 15.5 mg/L, and the germination rate was 85.63%, which were both higher than the predicted and actual values of response surface group. This study provided a theoretical and scientific basis for the development of SAEW and selenium-enriched treatment of tartary buckwheat sprouts.

We sought to characterize early or late onset atopic asthma with fungal or non-fungal sensitization (AAFS or AANFS) and non-atopic asthma (NAA) in children and adults in the Severe Asthma Research Program (SARP). SARP is an ongoing project involving well-phenotyped patients with mild to severe asthma. METHODS: Phenotypic comparisons were performed using Kruskal-Wallis or chi-square test. Genetic association analyses were performed using logistic or linear regression. Background As a common geriatric chronic disease, sarcopenia has a high prevalence in patients with coronary heart disease (CHD), which increases the risk of adverse events such as falls and fractures, calf circumference, upper arm circumference, and Ishii score have been shown to be valuable in screening for sarcopenia, but are less commonly used in CHD patients. Objective To evaluate the accuracy and consistency of calf circumference, upper arm circumference, and Ishii score in screening for combined sarcopenia in hospitalized elderly patients with CHD. Methods A total of 213 CHD patients treated at the Department of Cardiology, Affiliated Hospital of North Sichuan Medical College from January 1, 2023 to May 31, 2023, were selected for the study, the body composition test instrument was used to detect indicators such as muscle mass and upper arm circumference, the measurement of calf circumference was performed, and Ishii score was calculated simultaneously. Spearman's rank correlation analysis was used to explore the correlation of upper arm circumference, calf circumference and Ishii score with indicators such as muscle mass, the diagnostic effect of upper arm circumference, calf circumference and Ishii score on sarcopenia in different people was analyzed by plotting receiver operating characteristic (ROC) curves, and the optimal diagnostic cut-off values were determined, which were calculated as Kappa values to determine the consistency of the upper arm circumference, calf circumference and Ishii score with the diagnostic criteria of sarcopenia according to the Chinese Expert Consensus (2023) Criteria for the Prevention and Treatment of Sarcopenia in the elderly. Taiwan has been promoting its national health insurance (NHI), which provides people with appropriate medical resources; however, health insurance education is missing from schools, and teachers lack relevant teaching abilities. This study describes the development of the health insurance education empowerment program (HIEEP) to promote effective teaching on educators. A two-group parallel-controlled trial was conducted with health education teachers, in which participants learned to discuss its importance through demonstration and practice. The questionnaire measured their teaching effectiveness in HIEEP using a baseline measurement and a one-week follow-up measurement. Of 196 people that started the trial, 186 completed it. The intervention effects were evaluated using the Johnson-Neyman method. The results showed that in the experimental group, the program raised all teachers' variable scores, which evaluated health insurance meaning, teaching competence, self-determination, and impact. The empowerment program can provide teachers will allow for more effective health insurance education.

In practice, it is often necessary to add retarders to β-hemihydrate phosphogypsum (β-HPG) to prolong its setting time. In this paper, the effects of modified amino acid (XK) on physical and mechanical properties, hydration characteristics, and microstructure of β-HPG were investigated using various methods including fluidity, setting time, strength, hydration heat, X-ray diffraction (XRD), scanning electron microscope (SEM), mercury intrusion porosimetry (MIP), and X-ray photoelectron spectroscopy (XPS). The findings indicate that the addition of XK enhances the fluidity of β-HPG, and as the content of XK increases, there is an occurrence of prolonged setting time but a concurrent decrease in both the compressive and flexural strength. In addition, the incorporation of XK hampers the hydration process of β-HPG, resulting in a reduction in the hydration exothermic rate. Microscopic analysis reveals that while XK does not alter the final hydration products, it does affect the growth pattern of dihydrate gypsum crystals because of surface adsorption, and specifically, the crystals become short and columnar, with a looser crystal structure. Furthermore, XK leads to larger pores and deterioration of the pore structure of β-HPG.
