The poultry industry stands as a cornerstone in securing global food stability, both in the present and foreseeable future. To continue to uphold this critical role, the exploration of innovative, low-input feed sources is imperative. Hemp emerges as a promising candidate, offering locally sourced ingredients that not only reduce feed costs but also maintain high nutritional standards for livestock. This study delved into the utilisation of whole hemp seeds in the diet of 100 Naked-neck Kabir chicks that were divided randomly into two dietary groups, one fed a control diet (CTRL) and one fed a diet deprived of soybean meal with 10% inclusion of Carmagnola hemp seeds (HEMP). Birds were slaughtered at 81 days of age. Physicochemical characteristics of breast, thigh, and liver, besides the antioxidant content and the lipid oxidative status, were investigated. The HEMP birds exhibited higher daily weight gain and final live weight compared to the CTRL group. Results revealed improvements in the fatty acid profiles and antioxidant content in the HEMP group, leading to good levels of key nutrients such as α-tocopherol, retinol, and n-3 polyunsaturated fatty acids. These findings underscore the potential of hemp seeds as a feed ingredient, standing poised as a sustainable ally in fortifying the poultry productive chain.

In the European Union, energy consumption in agriculture and forestry is less intense than in other economic sectors. However, fossil and non-renewable sources, particularly oil, are predominant, with higher percentages than in different economic sectors. In this context, this study aims to compare the environmental impact generated by equine and mechanical traction for viticulture practices in an organic production context, using life cycle assessment (LCA). Three scenarios were considered: business as usual (i.e., tractor-grown grapes) and horse-grown grapes but with two feeding options (tractor-grown hay and horse-grown hay). The results show that the tractor-grown grape scenario has higher impacts in five impact categories whereas the horse-grown scenario shows higher Global Warming, and to a minor extent Land use. Specifically, the worst results of the control scenario are in the mid-point category of Inorganic Respiratory due to fuel use, which produces P M 2.5. Fuel use is also the cause of the highest environmental burdens in other categories such as terrestrial Ecotoxicity. A sensitive analysis of the results was performed considering the endpoint damage categories. The outcomes show that, in general, the impact of horses is quite stable even under potentially different conditions. Some categories (ecosystem quality, global impact) are only slightly affected by variation in growth time, while all other categories (human health, resources, climate change) experience greater variation.

This study explores the potential of rabbits as a sustainable solution for poverty alleviation and food security in Burkina Faso, a country facing socio-economic and environmental challenges. In the context of limited resources and employment opportunities, rabbits offer advantages such as small size, short generation interval and high reproductive capacity. The research focuses on characterising the morphology of Burkina Faso’s local rabbit population, comparing it with two Italian breeds: a local medium-growing breed called Leprino di Viterbo (LV) and a fast-growing hybrid (a crossbred New Zealand × California, NZC). Utilising path analysis and principal component analysis, the study identifies key morphometric traits crucial for breeding programs and examines the influence of specific traits on body weight (BW) gain and heat stress resilience through ‘Transpiration Indexes.’ Findings highlight the importance of variables like chest girth (CG), abdominal girth (AG), rump width (RW) and nose to shoulders length (NSL) in influencing body weight (BW) and reveal significant size and shape differences among the breeds. The study suggests potential climate adaptation in Burkina Faso’s rabbit population, providing practical insights for breeding programs in resource-constrained regions. This research not only advances scientific understanding but also provides practical insights for breeding programs in resource-constrained regions, emphasising the importance of certain morphological traits for heat dissipation.

Specific characteristics of the eggs from nucleus populations of Ancona (AN), Bianca di Saluzzo (BS), Bionda Piemontese (BP), Livorno, Mericanel della Brianza (MB), Mugellese (MU), Siciliana (SI) and Valdarnese (VA) chicken breeds kept in University Research Centres across Italy were evaluated. The physical characteristics considered were whole egg weight and shell, yolk and albumen proportion. The chemical characteristics considered were dry matter (DM), ash and protein content of the yolk and albumen, and the yolk fat. Among the breed groups evaluated in this study, BS and BP produce large eggs that are rich in total crude protein and lipids, with abundant egg whites. MB and MU produce small eggs with a high yolk percentage. AN and SI hens produce medium-small-sized eggs. The heaviest yolks were observed in AN eggs, and the highest shell percentage was found in SI eggs. LB and VA produce medium-large-sized eggs with low yolk lipid content.

ABSTRACT: The optimization of animal welfare, meat quality, environmental impact, and economic sustainability in alternative poultry farming can be achieved by modulating several productive factors and improving the synergy between the chicken genotype and the outdoor environment. The objective of the study was to characterize 4 slow-growing chicken genotypes reared in free range conditions. Eight hundred chickens (SGs; 25 chickens/replicates/genotype/enrichment) belonging to the following genotypes, Red JA57 (RJ), Naked Neck (NN), Lohmann Dual meat-type (LD), and an Italian crossbreed (Robusta Maculata x Sasso, CB). were utilised and slaughtered at 81 d: The grazing areas were alternatively provided with enrichment constituted by strips of sorghum plants (ENR) or only grass (NO ENR). Productive performance (daily weight gain, daily feed intake, feed conversion ratio, live weight) were recorded weekly. Behaviour observations (walking and grass pecking), carcass and meat quality of breast and drumstick were also assessed in 15 chickens/replicate/genotypes/enrichment. Results demonstrated that both LD and CB showed the highest walking activity, but the different strains were differently capable of using the foraging resources (eating grass). The better productive performance was recorded in RJ followed by NN, CB and LD. In LD and CB, the different walking activities also affected the physico-chemical profiles (lower pHu, WHC, and lipids) of the breast and drumstick. The oxidative status was worse in CB than in the other groups (lower tocols, higher carbonyls), in both meat cuts. Fatty acid profile was also related to the genetic strain: a higher amount of n-3 polyunsaturated fatty acids was recorded both in the breast and drumstick of RJ and NN. The Healthy Fatty Index resulted excellent in all the chicken genotypes. In conclusion, the environment/animal interaction resulted as an important factor affecting the adaptability of genotypes to an extensive rearing system. All four genotypes, to different extents, showed good adaptability and production performance, with the exception of LD and CB, which were too light for the commercial supply chain requirements.

ABSTRACT: The present study aimed to compare the dynamics of growth of various chicken genotypes exposed to heat stress, low-input diets, and free-range farming by using Gompertz model to gain insights into their capabilities to face environmental and nutritional challenges. Three in vivo trials (T1: heat stress, T2: low-input diets, and T3: free-range system) were conducted, involving a total of 671 animals. Five chicken genotypes were employed in each trial: 2 Italian local breeds, Bionda Piemontese (BP) and Robusta Maculata (RM), along with their crossbreeds with Sasso hens (BP×SA and RM×SA), and a commercial hybrid (Ross 308). One-day-old male chicks were individually identified, and the 5 genotypes were randomly allocated to different challenging conditions: T1 involved 2 environmental temperatures (thermoneutral vs. high temperature); T2 involved 2 diets (standard vs. low-input); T3 involved 2 rearing systems (conventional vs. free-range). The chickens were weighed once a week from their arrival until slaughtering, and the data were used to build growth curves using the Gompertz model. Chickens from different genotypes were slaughtered at varying ages based on their maturity. In all trials, the challenging conditions significantly reduced adult body weight (A; −31.0%) and maximum growth rate (MGR; −25.6%) of Ross chickens. In contrast, in T1 and T2, no significant changes were observed in the main growth curve parameters of local breeds and crossbreeds, while under free-range conditions, there was even an increase in the A and MGR of these genotypes. The crossbreeding was effective in increasing A and MGR of BP (+30.5% in BP×SA), as well as in improving the precocity and MGR of RM (+19.5% in RM×SA). Our findings highlight the effectiveness of the Gompertz model as a tool for evaluating birds’ adaptability and confirm the greater ability of local breeds and crossbreeds to adapt to different challenges. In conclusion, our methodological approach could be used to choose the genotype most suited to the environmental context and confirm the potential advantages of crossbreeding for enhancing resilience and sustainability.

According to FAO projections, meat and fish production will have to increase by 70% by 2050 to meet the needs of the growing global population and its protein requirements. The poultry sector has been one of the most responsive industries to the growing demand for protein, with a significant increase in egg and chicken meat production. Although eggs are important foods in the human diet, environmental impacts and ethical aspects could also affect dietary choices. This study aimed to assess the environmental impacts of egg production, by using Life Cycle Assessment (LCA). More specifically, the study compared two different farming systems of conventional and organic egg production, with reference to two case studies in northern Italy. The results obtainedgenerally showed the conventional system to be more efficient compared to the organic system. For example, as regards the impact category of Fine particulate matter formation, the conventional system had a 28.6% lower impact than the organic system. Another major difference was in the Land use category, where the conventional system had an impact of 48.5% less than the organic system. The Organic Scenario showed some critical aspects, e.g. the need for a large agricultural area, low feed conversion rates and low crop yields. The two production scenarios showed that in terms of environmental impact, the main difference between organic and conventional egg production resides in feed production. I It is, therefore, essential to work on a low input formula, to substitutethe diet components with the greatest impact and adopt strategies to obtain an increase in yields and/or a reduction in resource consumption during the cultivation phase. The complete substitution of soybean with other local crops could reduce several impacts, including the one linked to transport. The further reduction of maize could also limit the expected impact.

Abstract Commercial laying hens have high frequency of damage to the keel bone (KB), which causes negative effects on health and welfare. KB damage may consist in fractures (KBF) and deviations (KBD). The aim of the present study was to compare the KB shape, by means of Geometric Morphometric, and the occurrence of fractures in different chicken genotypes reared either free-range (FR) or in enriched cages. Moreover, the relationship between KB shape, genotype and rearing system was analysed. Sixty birds/genotype (2 Italian local breeds, Bionda Piemontese and Robusta Maculata, their crossbreeds with Sasso and Lohmann Brown) were used. All the hens fed the same commercial feed throughout the trial. Body weight, egg production, feed intake and mortality were recorded from 25 to 66 weeks of age. Ca intake (IN) and output (OUT) were estimated and Ca OUT/IN was calculated. FR affected the occurrence of KB deviations but not the shape, whereas the fractures were mainly affected by genotype. Local breeds had a lower prevalence of KBF with similar level of KBD but with different shapes. Crossbreeds seemed to be a suitable compromise between egg deposition rate and occurrence of KB damages.

Abstract The use of the scan-sampling method, especially when a large amount of data is collected, has become widespread in behavioral studies. However, there are no specific guidelines regarding the choice of the sampling interval in different conditions. Thus, establishing a standard approach for video analysis represents an important step forward within the scientific community. In the present work, we hypothesized that the length of the sampling interval could influence the results of chicken behavioral study, for which we evaluated the reliability, accuracy, and validity of three different sampling intervals (10, 15 and 30 min). The Bland–Altman test was proposed as an innovative approach to compare sampling intervals and support researcher choices. Moreover, these sampling intervals were applied to compare the behavior of 4 chicken genotypes kept under free-range conditions. The Bland–Altman plots suggested that sampling intervals greater than 10 min lead to biases in the estimation of rare behaviors, such as “Attacking”. In contrast, the 30-min sampling interval was able to detect differences among genotypes in high-occurrence behaviors, such as those associated with locomotory activity. Thus, from a practical viewpoint, when a broad characterization of chicken genotypes is required, the 30-min scan-sampling interval might be suggested as a good compromise between resources and results.

In the present study, we evaluated the effect of various concentrations of date palm pollen (DPP) aqueous extract on rabbit sperm during storage at 4°C for 48 h. Semen was collected from old and young rabbit bucks using an artificial vagina and initially evaluated for sperm quality. The sperm were diluted in Tris buffer supplemented with (20, 40, 80 mg/mL) of DPP aqueous extract. The extended samples were stored at 4°C for 48 h. Sperm motility and motion kinetics were assessed after 2, 4, 24 and 48 h of storage. At each time, an aliquot was frozen for the analytical evaluation of thiobarbituric acid reactive substances (TBARS) (lipid peroxidation) and tocols (vitamin E). Proximate composition, antioxidant and sugar content were evaluated in DPP. The results showed that DPP was characterised by a high proportion of protein (27.10%) and ash (18.43%), whereas the lipid fraction was very low (0.51%) and total sugar was also high (16.25 g/100 g of fresh matter, f.m.). Regarding the antioxidants content, the sum of tocols showed a total value of 26.48 mg/g f.m. The total polyphenols content was 5.01 mg gallic acid equivalents/g f.m, and polyunsaturated fatty acids was around 30%. Date palm pollen extract had a dose-dependent effect on sperm parameters (curvilinear velocity [VCL] mainly) of old rabbit, although a null or negative effect was recorded at doses >DPP40. Conversely, in optimal conditions (i.e. young rabbit semen), the addition of pollen had no effect on sperm traits (motility and VCL). The progressive increase in DPP, despite providing sperm with an additional amount of tocols, also caused higher tocol consumption and an increase in lipid oxidation. In particular, DPP80 increased the TBARS level in sperm of both rabbit ages. The better trend was found in DPP20, where the tocol consumption figures were 10.46 % and 15.28 %, respectively, in old and young bucks, and the lipid oxidation was lower compared to the higher doses of DPP. In conclusion, the findings of this study demonstrated that supplementation of DPP extract to Tris buffer extender enhanced chilled rabbit semen traits only if administered in old rabbit semen at concentration lower than 40 mg/mL.

ABSTRACT: The majority of poultry meat used to be sourced from intensively housed birds. However, consumer preference has since demanded poultry producers develop more sustainable farming systems. Although free-range farming is considered beneficial for animal welfare, it is not as easy to standardize as an intensive system, which makes the choice of bird genotype appear crucial for alternative systems. In this study, we aimed to evaluate the effect of conventional and free-range rearing systems on the immune status, stress parameters, intestinal morphology and mortality in commercial hybrids (Ross 308) and local poultry strains, Bionda Piemontese (BP), Robusta Maculata (RM), BP x Sasso (BPxS), and RM x Sasso (RMxS). RNA was extracted from the jejunum and spleen to assess the mRNA expression of IL-2, IL-6, IL-10, IL-18, IL-1β, inducible nitric oxide synthase (iNOS), toll-like receptor (TLR)-4, and interferon gamma (IFN-γ). The heterophil:lymphocyte (H/L) ratio and intestinal histomorphometric evaluation were also calculated. We found that compared to the conventional system, the rearing system significantly affected the jejunum expression of IL-10, iNOS, IL-2, and IL-6, where these genes were upregulated in free-range system. A significant interaction between the rearing system and the genotype was also shown. More specifically, local breeds showed a significantly higher expression (P < 0.001) of IL-6 in the free-range system compared to the same genotypes in the conventional system. Moreover, IL-6 is constantly upregulated in local breeds within the free-range system compared to Ross hybrids. We also found significantly increased H/L and mortality rates in the latter, compared to the local breeds in the free-range reared system. The jejunum morphology also demonstrated a significantly higher villus height in BP and BPxS compared to the Ross hybrids. Overall, the results of our study confirm that the intense selection for growth in broiler chickens may have reduced their ability to react to the environmental stimuli related to free-range systems, resulting in a lower adaptability to a free-range environment, thus making them inappropriate for any farming system other than the conventional one. On the contrary, local chicken breeds are able to adapt and survive in the free-range system of rearing, and represent a genetic resource especially when adaptability to free-range conditions is required.

The aim of this research was to validate the effectiveness of the Healthy Fatty Index (HFI) regarding some foods of animal origin (meat, processed, fish, milk products, and eggs) typical of the Western diet and to compare these results with two consolidated indices (atherogenic—AI, and thrombogenic—TI) in the characterization of the nutritional features of their lipids. The fatty acids profile (% of total fatty acids and mg/100 g) of 60 foods, grouped in six subclasses, was used. The AI, TI, and HFI indexes were calculated, and the intraclass correlation coefficients and the degree of agreement were evaluated using different statistical approaches. The results demonstrated that HFI, with respect to AI and TI, seems better able to consider the complexity of the fatty acid profile and the different fat contents. HFI and AI are the two most diverse indices, and they can provide different food classifications. AI and IT exhibit only a fair agreement in regards to food classification, confirming that such indexes are always to be considered indissolubly and never separately, in contrast to the HFI, which can stand alone.

This research aimed to investigate, through a multifactorial approach, the relationship among some in-vivo parameters (i.e., behavior and blood traits) in broilers exposed to chronic HS, and their implications on proximate composition, technological properties, and oxidative stability of breast meat. A total of 300 Ross 308 male chickens were exposed, from 35 to 41 days of age, to either thermoneutral conditions (TNT group: 20°C; six replicates of 25 birds/each) or elevated ambient temperature (HS group: 24 h/d at 30°C; six replicates of 25 birds/each). In order to deal with thermal stress, HS chickens firstly varied the frequency of some behaviors that are normally expressed also in physiological conditions (i.e., increasing “drinking” and decreasing “feeding”) and then exhibited a behavioral pattern finalized at dissipating heat, primarily represented by “roosting,” “panting” and “elevating wings.” Such modifications become evident when the temperature reached 25°C, while the behavioral frequencies tended to stabilize at 27°C with no further substantial changes over the 6 days of thermal challenge. The multifactorial approach highlighted that these behavioral changes were associated with oxidative and inflammatory status as indicated by lower blood γ-tocopherol and higher carbonyls level (0.38 vs. 0.18 nmol/mL, and 2.39 vs. 7.19 nmol/mg proteins, respectively for TNT and HS; p < 0.001). HS affected breast meat quality by reducing the moisture:protein ratio (3.17 vs. 3.01, respectively for TNT and HS; p < 0.05) as well as the muscular acidification (ultimate pH = 5.81 vs. 6.00, respectively; p < 0.01), resulting in meat with higher holding capacity and tenderness. HS conditions reduced thiobarbituric acid reactive substances (TBARS) concentration in the breast meat while increased protein oxidation. Overall results evidenced a dynamic response of broiler chickens to HS exposure that induced behavioral and physiological modifications strictly linked to alterations of blood parameters and meat quality characteristics.

Abstract Agroforestry systems, which are based on the integration of trees and animals, represent a useful practice for implementing the “One Welfare” concept. Geese could adapt well to these systems due to their kinetic and grazing abilities. However, the lack of specific ethograms and animal-based measures have not yet allowed a deep assessment of their welfare and behavior. The aim of this study was to develop and validate a protocol to evaluate the behavior of geese reared in two agroforestry systems (i.e., apple orchard and vineyard). Thus, a behavior observation form (BOF) including a specific ethogram was proposed, and its interobserver reliability, content, criterion and construct validity were determined. Moreover, the influence of the time of day and type of agroforestry system on geese's behavior was also investigated. Agreement and principal component analyses, as well as the comparison between data collected through direct observation by the BOF and indirect observation by a computerized system, supported the reliability and validity of the proposed protocol. While the BOF also highlighted differences in the behavior expressed by the geese according to the time of day and the environmental context, both the vineyard and apple orchard systems seem to meet their biological and behavioral needs.

ABSTRACT: The present study aims to validate the Gompertz model to predict the growth performance of chicken crosses according to growth curve parameters of the parental lines and the estimated heterosis for each curve parameter. A total of 252 one-day-old chicks of both sexes belonging to 6 genotypes, including Ross 308, Sassò (SA), Bionda Piemontese (BP), and Robusta Maculata (RM), and the crosses between these local breeds and SA (BP × SA and RM × SA) were randomly allocated in 18 pens (3 pens/genotype) in mixed-sex groups (14 animals/pen; 7 females and 7 males). The individual body weight (BW) of all birds was recorded once a week from hatching until slaughtering (81 d for Ross 308; 112 d for SA, 140 d for the other genotypes). We drew up our final dataset with 240 birds (40 birds/genotype; 20 females and 20 males). The growth curve of each genotype was described using the Gompertz model, and the heterosis for each growth curve parameter was calculated as the difference between F1 crosses and the average of parental breeds. The predicted growth curve parameters were evaluated by cross-validation. The Gompertz model accurately estimated the growth curves of all the genotypes (R2 > 0.90). Heterosis was significant for almost all growth curve parameters in both crosses (P < 0.05). Heterosis ranged from −13.0 to +11.5%, depending on parameters, but varied slightly between the crossbreeds (BP × SA and RM × SA). The predicted values of adult BW, weight at the inflection point, and maximum growth rate were overestimated for BP × SA and underestimated for RM × SA, with a mean error between observed and predicted values

Nerve growth factor (NGF) signalling affects spermatogenesis and mature sperm traits. In this paper, we aimed to evaluate the distribution and the role of NGF and its receptors (p75NTR and TrKA) on the reproductive apparatus (testis and epididymis) and sperm of fertile men (F) and men with different pathologies, namely varicocele (V) and urogenital infections (UGIs). We collected semen samples from 21 individuals (31–40 years old) subdivided as follows: V (n = 7), UGIs (n = 7), and F (n = 7). We submitted the semen samples to bacteriological analysis, leucocyte identification, and analysis of sperm parameters (concentration, motility, morphology, and viability). We determined the seminal plasma levels of NGF, interleukin 1β (IL-1β), and F2-isoprostanes (F2-IsoPs), and the gene and protein expression of NGF receptors on sperm. We also used immunofluorescence to examine NGF receptors on ejaculated sperm, testis, and epididymis. As expected, fertile men showed better sperm parameters as well as lower levels of NGF, F2-IsoPs, and IL-1β compared with men with infertility. Notably, in normal sperm, p75NTR and TrKA were localised throughout the entire tail. TrKA was also found in the post-acrosomal sheath. This localisation appeared different in patients with infertility: in particular, there was a strong p75NTR signal in the midpiece and the cytoplasmic residue or coiled tails of altered ejaculated sperm. In line with these findings, NGF receptors were intensely expressed in the epididymis and interstitial tissue of the testis. These data suggest the distinctive involvement of NGF and its receptors in the physiology of sperm from fertile men and men with infertility, indicating a possible role for new targeted treatment strategies.

Many dietary factors can affect sperm traits. We compared the effect of diets rich in pro-oxidant (flaxseed oil) and pro-atherogenic (coconut oil) substances without added antioxidants on semen traits, using the rabbit as an animal model. Thirty rabbit bucks (8 months old) were fed three diets for 150 days: CNT (control) a standard diet; HA (high-atherogenic) standard diet + 3% coconut oil, and HO (high-oxidizing) standard diet + 3% flaxseed oil. Semen samples were collected weekly for the evaluation of qualitative traits (kinetics, viability) and the oxidative damage (MDA and cytokines). Blood was collected at the start (T0) and end (T8) of the experimental period for the assessment of the oxidative damage (MDA and isoprostanoids), lipid profile, and testosterone. A worsening of sperm kinetics and viability was recorded in the HA group. Lipid oxidation in seminal plasma, as well as isoprostanoids in blood (F3-IsoPs and F4-NeuroPs), increased in both the HO and HA groups. A high level of TNF-α, a marker of inflammatory status, was recorded in the seminal plasma of the HA group. The resulting outcomes were mainly attributable to the different fatty acid profiles (SFA vs. PUFA) of the diets, which modulated an inflammatory/oxidative response.

Abstract Humans and mammalian species are unable to synthesize significant amounts of polyunsaturated fatty acids (PUFA), which therefore must be introduced with the diet. In birds, lipogenesis takes place primarily in the liver, whereas adipose tissue serves as the storage site for triacylglycerols (TG, composed by 80–85% esterified fatty acids). However, both the nature (unsaturation level, n-3, or n-6 series) and the allocation (such as constituents of complexed lipids) of PUFA are very important to evaluate their function in lipid metabolism. The objective of the present investigation was to study the liver lipid metabolism, with particular attention to non-esterified fatty acids (NEFA), TG, phospholipids (PL), FADS2 gene expression, and Δ6-desaturase activity of three chicken genotypes, Leghorn (Leg), Ross 308 (Ross), and their crossbreed (LxR), by LC/MS analysis. The concentration of single fatty acids in muscle was quantified by GC-FID. The results showed that the Ross has a lipid metabolism related mainly to storage and structural roles, exhibiting higher levels of TG, phosphatidylethanolamine (PE) and phosphatidylcholine (PC) that are largely unsaturated. Meanwhile Leg showed a relevant amount of n-3 NEFA characterized by a higher phosphatidylserine (PS) unsaturation level, FADS2 gene expression and enzyme activity. The LxR seem to have a moderate trend: n-6 and n-3 NEFA showed intermediate values compared with that of the Ross and Leg and the TG trend was similar to that of the Ross, while PE and PC were largely unsaturated (mainly 6 and 7 UNS most of the metabolic energy for storage fatty acids in their tissues (TG) whereas, the Leg birds were characterized by different lipid metabolism showing in their liver a higher content of n-3 NEFA and higher unsaturation level in PS. Furthers details are needed to better attribute the lipid energy to the different metabolic portion.

Abstract The metabolism of polyunsaturated fatty acids (PUFAs) plays an important role in male reproduction. Linoleic and alpha-linolenic acids need to be provided in the diet and they are converted into long chain polyunsaturated fatty acids by steps of elongation and desaturation, exerted by elongases 2 (ELOVL2) and 5 (ELOVL5) and Δ5- (FADS1) and Δ6-desaturase (FADS2). This study aims to assess the gene expression and localization of enzymes involved in the synthesis of n-3 and n-6 long-chain PUFAs in control rabbits and those fed diets containing 10% extruded flaxseed. Enzyme and PUFA localization were assessed in the testes and epididymis by immunofluorescence. Testes showed high gene expression of FADS2, ELOVL2 and ELOVL5 and low expression of FADS1. Intermediate metabolites, enzymes and final products were differently found in Leydig, Sertoli and germinal cells. FADS2 was localized in interstitial cells and elongated spermatids; ELOVL5 in meiotic cells; FADS1 was evident in interstitial tissue, Sertoli cells and elongated spermatids; ELOVL2 in interstitial cells. Epididymal vesicles were positive for FADS1, ELOVL2 and ELOVL5 as well as docosahexaenoic, eicosapentaenoic, and arachidonic acids. This knowledge of fatty acids (FA) metabolism in spermatogenesis and the influence of diet on FA profile could help identify causes of male infertility, suggesting new personalized therapy.

ABSTRACT: The recovery, safeguarding, and valorization of autochthonous poultry breeds may contribute toward the preservation of animal biodiversity and utilization of marginal lands that otherwise offer little agricultural or industrial value. A key strategy in promoting local breeds involves the characterization of morphological traits and productive performances, which are influenced by the breed's genetic make-up as well as its environment. The Mugellese breed is an Italian local poultry breed originating in the Mugello area of north-east Tuscany. It is characterized by frugality, resilience and resistance to disease, cold, and heat stress. Moreover, these birds are particularly suitable for free-range farming. The Mugellese chicken is described as a dwarf breed with a medium neck, broad shoulders, fairly long and horizontal wings, wide, and well-developed breast (especially in the hen). Over the course of a 1-yr observation and data collection period, involving 23 breeders and 405 adult chickens, the Mugellese breed showed the following performances: 1) a hen-day egg production characterized by 2 major peaks: the first in the spring time (March–April, 65.75%), and the second in the late summer period (August–September, 51.86%); 2) high true fertility values (94.35%) throughout the entire breeding season; 3) a weight gain of 732.44 ± 117.06 g and a feed conversion ratio of 3.94 ± 2.42 at an age of 140 d; 4) a slaughter yield of 77.80% (± 3.91); v) a respective protein, fat, and mineral content in the yolk and albumen were: 27.21 ± 4.21 g, 57.77 ± 1.03 g and 3.47 ± 0.40 g per 100 g of yolk; and 82.50 ± 0.57 g, 0.12 ± 0.01 g and 5.43 ± 0.34 g per 100 g of albumen. More data are needed to validate the data obtained in this trial.

Rabbit is an induced ovulatory species, so ovulation takes place after mating. Traditionally, exogenous and synthetic hormonal factors (administered by intramuscular and intravaginal route) such as GnRH and analogues, or different physical procedures (i.e. stimulation by intravaginal cannula) have been used to induce ovulation in females when artificial insemination is applied in rabbit farms. Restriction and public rejection of the use of hormones is leading to the study of the seminal plasma components with potential action on ovulation induction. The aim of the present review is to collect and summarise the strategies used in recent years to trigger ovulation and improve rabbit fertility management with respect to more animal-friendly manipulation methods. Furthermore, special attention has been paid to the use of a semen component (as endogen molecule) such as beta nerve growth factor (β-NGF) in male and female rabbit reproductive physiology. This neurotrophin and its receptors (TrKA and p75NTR) are abundantly distributed in both male and female rabbit reproductive tracts, and it seems to have an important physiological role in sperm maturation and behaviour (velocity, apoptosis and capacitation), as well as a modulatory factor of ovulation. Endogen β-NGF is diluted in the seminal doses with the extenders; hence it could be considered an innovative and alternative strategy to avoid the current exogenous (by intramuscular route) and stressful hormonal treatments used in ovulation induction. Their addition in seminal dose could be more physiological and improve animal welfare in rabbit farms.

Apelin and other novel adipokines have been associated with normal and pathological reproductive conditions in humans and animals. In this paper, we used a rabbit model to investigate if apelin and resolvin (RvD1) in testis and sperm are associated with the oxidative status of semen and serum testosterone of rabbits fed different diets enriched with flaxseed (alpha-linolenic acid, ALA) or with fish oil (eicosapentaenoic acid, EPA, docosapentaenoic acid, DPAn-3, and docosahexaenoic acid, DHA). Apelin and RvD1 were detected by ELISA and apelin and the apelin receptor by immunofluorescence. Increased levels of apelin in testes from both enriched diets were shown, particularly in the interstitial tissue of the FLAX group. The FLAX diet enhanced serum testosterone, and both enriched diets showed higher levels of malondialdehyde and RvD1 in the testis. In ejaculated sperm, apelin and its receptor were localized in the entire tail of the control and both treated groups. The ryanodine receptor was investigated in rabbit testis; the fluorescent signal was increased in mature elongated spermatids of the FLAX group. In conclusion, this data seems to indicate that FLAX increases the amount of apelin in testis, suggesting an involvement of this adipokine in male reproduction and probably a role in the resolution of the inflammatory status.

Date palm pollen (DPP) has been widely tested in vitro in preservation of spermatozoa of different farm animals except in rabbits. The objective of this study was to evaluate the effect of various concentrations (20, 40, 80 mg/mL) of DPP aqueous extract on epididymal and ejaculated rabbit spermatozoa during in vitro incubation at 37 °C during 120 min. NaCl Pollen Extender (NPE) and TRIS Base Extender (TPE) were respectively used in epididymal and ejaculated sperm preservation. Sperm motility parameters were objectively analysed by a Computer Aided Sperm Analyser (CASA). The results revealed that in epididymal sperm at 0 minute, sperm motility showed significant higher values (p ≤ 0.05) for all doses of DPP in comparison to the control group. After 30, 60 and 120 min, the motility rate was higher (p > .05) in all the experimental groups compared to the control. Concerning velocity traits (VCL and VAP), and trajectories parameters (LIN, ALH, BCF), the doses of 40 mg/mL and 80 mg/mL showed the highest values than Control. In ejaculated samples, the motility rate at 0 minute improved only with the dose of 80 mg/mL and DPP showed protective effect at all doses (20, 40, 80 mg/mL) up to 60 min of incubation. DPP enhanced spermatozoa velocity (VCL and VAP) and the trajectory of sperm (LIN, ALH, BCF) even from 20 mg/mL at any points of storage. In conclusion, aqueous extract of date palm pollen is a suitable supplement to the extender for rabbit sperm by protecting and improving sperm motility parameters.HIGHLIGHTS The addition of DPP extract improves the quality of epididymal and ejaculated rabbit sperm. The extender supplemented with 80 mg/mL of DPP in NPE has a beneficial effect on kinetic traits of epididymal sperm. Nevertheless, in TPE extender, 20 mg/mL of DPP was enough to improve sperm motility traits in ejaculated rabbit semen.

The creation of genetic resource cryobanks provides a crucial link between in situ and ex situ techniques to improve the efficiency of conservation programs. Aim of the present review is to describe all the activities developed for the implementation of the first Italian Semen Cryobank of Autochthonous Chicken and Turkey Breeds. These activities can be classified into three main topics: (1) identification of species-specific semen freezing/thawing reference procedures; (2) drafting Standard Operative Procedures (SOP) for the implementation of the semen cryobank; (3) storage of semen doses from Italian chicken and turkey breeds to establish the cryobank. Several trials have been developed to identify a specie-specific semen cryopreservation protocol for chickens and turkeys. The major results are reviewed and a final reference protocol described. Taking into consideration the FAO guidelines for cryoconservation of animal genetic resources, SOP were drafted with the aim to provide technical guidance and logistical support on the choice of priority breeds, selection of birds for semen production, infrastructures and storage sites, birds and semen management, cryopreservation process and doses traceability. Lastly, the Italian Semen Cryobank was created. A total of 112 semen doses from 22 cockerels of three breeds, and 74 doses from 12 turkey males of three breeds were stored in the Cryobank. Breed specific semen quality parameters assessed before and after cryopreservation are reported. Nowadays most of the Italian chickens’ breeds are replaced with commercial hybrids characterised by higher productive performance. However, the safeguard of biodiversity and consequently the genetic variability are a very important issue, and for this reason, it is fundamental to include local strains in preservation programs. In Italy, the poultry Genealogic Book has been recently constituted, and includes 22 chicken breeds. The diffusion of these strains is strictly linked to the territory of origin, in particular in the Central Italy, the Ancona and Livorno chicken breeds are the mainly widespread ones. The aim of this study was to determine the consistency and the characteristics of the farms involved in the farming of Ancona and Livorno chicken breeds in Central Italy. A survey to analyse the facilities, the human resources and the chicken population consistencies was conducted in the farms of Abruzzo, Lazio, Marche, Tuscany and Umbria regions. All the investigated farms can be identified as ‘hobby-farms’ being small scale ones and leading the breeding of Ancona and Livorno chickens as a secondary business. The survey also confirmed that the presence of the strains depends on the region: 100% of Marche farms bred Ancona whereas, in Tuscany the Livorno breed is the most common.Highlights A survey in centre Italy was carried out to determine the consistency of Ancona and Livorno chicken breeds; The Ancona and Livorno chickens breed were reared in small scale farms; The widespread of the breeds depend on the region: Ancona breed is most present in the Marche region while the Livorno is mainly bred in Tuscany.

An extensive rearing system (ERS) for poultry requires an outdoor run, which enhances the foraging activity of chickens. Slow-growing (SG) strains are more adapted to ERS than fast-growing (FG); and generally, have higher levels of bioactive compounds in their meat. The aim of this paper was to assess the storage efficiency of n-3 and n-6 polyunsaturated fatty acids (PUFA), tocols and carotenes in the meat of seven commercial SG genotypes (SG1-7). One hundred SG chicks/strain of both sexes were included and their walking activity (High- or Low-W) was classified: SG1-4, HW comprised more than 10% of the time budget, and SG5-7, less than 10% (LW). Chickens were reared in pens (4 pens/strain) with indoor (0.10 m2/bird) and outdoor (4 m2/bird) areas, and they were fed the same diet ad libitum (starter feed for 1–21 d, grower feed from 22 d to slaughter at 81 d). The chickens were weighed weekly; feed consumption and grass intake were also estimated. At 81 days of age, 32 chickens/genotype were selected on the basis of the average weight (1:1, M:F) and slaughtered. The breast, thigh and drumstick meat were excised from 30 carcasses/genotype, sampled and stored at -20°C until analysis. Nutrients (e.g., n-3, n-6, carotenes and tocols) of feed, grass and meat were analyzed. The storage efficiency of nutrients was estimated as the ratio between the amount deposited in the body muscles (OUT) and the dietary intake (feed and grass, IN). The genotype affected chickens foraging behavior and the intake of nutrients. For SG1, SG2 and SG3, more than 50% of the intake of n-3 came from grass, whereas in the other genotypes, less than 20%. Accordingly, chickens that foraged more showed better meat nutritional profiles (less fat, more n-3 and antioxidants), which, in ERS, was ascribed to grass ingestion. However, the storage efficiency of nutrients into meat was inversely correlated with the grass intake: strains with higher grass intake (SG1, SG2, and SG3) had lower storage rates. Several hypotheses were proposed to explain these trends.

Eggs are a complete food with high-quality proteins; a 2:1 ratio of unsaturated to saturated fatty acid (SFA); and a good amount of minerals, as well as vitamins or antioxidant compounds. Seeds or mature plants were usually added to the feed to improve egg quality. This study aimed to evaluate the effect of alfalfa and flax freeze-dried sprouts supplementation in diets of laying hens on egg oxidative status and key bioactive compounds. Thirty Sassò hens were fed with three different diets: standard, standard + 3% freeze-dried alfalfa sprouts, or flaxseed sprouts. Ten pools of 10 egg yolks per group were collected at 0, 4, and 8 weeks and analyzed. Supplementation with sprouts enriched the phytosterols, phytoestrogens, tocols, carotenes, vitamin D, and n-3 fatty acid contents in the eggs. Cholesterol content was lower in both sprout-supplemented groups, and a decrease in its oxidative products was also observed. It was found that a 3% freeze-dried sprouts supplementation of approximately 56 days improves the egg quality. Further studies are necessary to verify higher supplementing doses and the applicability of this strategy in the commercial egg production chain.

ABSTRACT: The aim of this study was to analyze the relationship between the kinetic behavior, carcass characteristics, oxidative status (blood and meat), and meat fatty acid profiles of 6 organically reared slower growing chicken genotypes (SrG). One hundred male chickens of 6 SrG were used: Ranger Classic (RC), Ranger Gold (RG), Rowan Ranger (RR), RedJA (RJ), CY Gen 5 JA87 (CY), and M22 × JA87 (M). Twenty chickens/genotype were selected to analyze behavior, while, 15 individuals were slaughtered and different traits were analyzed in the blood and drumstick meat. The variables were grouped into different principal components: kinetic activity (PC1, with explorative attitude as the highest score), productive performance (PC2, carcass and head/feet yields), blood (PC3, carbonyls, and TBARS) and meat (PC6, thiols, and TBARS) markers, technological traits (PC4, pH, and color), proximate meat composition (PCA5, moisture, lipids, protein, and ash), fatty acid profile, and nutritional indexes (PC7, IP, and PUFAn-3). Uni- and bivariate analyses showed a strong positive association between kinetic behaviors and blood and meat oxidation and a medium positive association with fatty acid profile and nutritional indexes, whereas a negative association was found between productive performance loads and the technological traits of meat. Generalized linear models showed that all PCs were influenced by genotype. In particular, CY and M resulted as less active genotypes; conversely, RR showed more kinetic activity, whereas RJ, RG, and RC exhibited intermediate levels of activity. Cluster analysis of kinetic behavior and blood or meat oxidative status highlighted 2 groups: nonwalking (NW: CY and M) and walking (W: RC, RG, RR, and RJ) animals. However, in the W group, another was visualized, constituted by genotypes with high kinetic activity resulting in the worst oxidative balance (Walking not trained-genotypes, Wnt: RR and RJ). The present results confirmed that the kinetic behavior of SrG genotypes is negatively correlated with productive performance. Furthermore, a significant association between kinetic behavior and blood (positively correlated) or meat (negatively correlated) oxidative status was noted. Such differences are mainly due to the intrinsic response of the genotypes used (i.e., training-walking capacity).

The preservation of genetic variability of autochthonous poultry breeds is crucial in global biodiversity. A recent report revealed small breed size and potential risk of extinction of all native Italian poultry breeds; therefore, a correct assessment of their genetic diversity is necessary for a suitable management of their preservation. In this work, we provided an overview of the contribution to poultry biodiversity of some Italian autochthonous breeds reared in conservation centers devoted to local biodiversity preservation. The level of genetic diversity, molecular kinship, inbreeding, contribution to overall genetic diversity, and rate of extinction of each breed were analyzed with a set of 14 microsatellite loci in 17 autochthonous chicken breeds. To evaluate genetic variability, total number (Na), and effective number (Ne) of alleles, observed (Ho) and expected (He) heterozygosity, and F (Wright’s inbreeding coefficient) index were surveyed. The contribution of each analyzed breed to genetic diversity of the whole dataset was assessed using MolKin3.0; global genetic diversity and allelic richness contributions were evaluated. All the investigated loci were polymorphic; 209 alleles were identified (94 of which private alleles). The average number of alleles per locus was 3.62, and the effective number of alleles was 2.27. The Ne resulted lower in all breeds due to the presence of low-frequency alleles that can be easily lost by genetic drift, thus reducing the genetic variability of the breeds, and increasing their risk of extinction. The global molecular kinship was 27%, the average breed molecular kinship was 53%, and the mean inbreeding rate 43%, with a self-coancestry of 78%. Wright’s statistical analysis showed a 41% excess of homozygous due to breed genetic differences (34%) and to inbreeding within the breed (9%). Genetic variability analysis showed that 11 breeds were in endangered status. The contribution to Italian poultry genetic diversity, estimated as global genetic diversity, and ranged from 30.2 to 98.5%. In conclusion, the investigated breeds maintain a unique genetic pattern and play an important role in global Italian poultry biodiversity, providing a remarkable contribution to genetic variability.

This study compared dietary isoflavone aglycones with the glycoside conjugates in a novel model of postmenopausal status, the aging domestic hen (Gallus gallus domesticus), to determine the effects on reproductive performance, cholesterol levels, and nutritional quality of eggs laid. Hens, 18 mo old, were randomized into four groups (n = 10/group) and fed for 28 d a conventional poultry corn/soymeal diet (Control), or diets supplemented with isoflavone glycosides from soy germ (diet A), isoflavone aglycons from a soy germ pasta (diet B), or conventional pasta lacking isoflavones (diet C). The egg-laying rate was recorded daily, plasma isoflavones and cholesterol were measured, and the nutritional composition of the eggs was determined. Egg-laying declined over a 4-week period in hens in the Control group and those fed isoflavone glycosides (diets A and C), whereas hens fed isoflavone aglycons (diet B) significantly increased their egg-laying efficiency. The total egg count and egg yield were significantly higher in hens fed isoflavone aglycons, and their plasma cholesterol concentrations were lower and the eggs laid had a 30% lower yolk cholesterol content. None of these effects were observed with diets containing similar levels of isoflavone glycosides. These studies recapitulate the clinical effects of soy germ pasta enriched with isoflavone aglycons and lend support to the greater efficacy of a diet rich in isoflavone aglycons.

ABSTRACT: In organic poultry production it is important to rear animals with a dynamic attitude to take advantage of outdoor areas. Farmers are reluctant to use such strains due to their lower productivity and older slaughtering age. However, fast growing lines grown in organic system often suffer poor health and welfare conditions. The kinetic metabolism of chickens is correlated with different types of muscle fiber: type I (in red muscles or oxidative) for prolonged and moderate movement and type II (in white muscles or glycogenic) for fast movements. Red muscle metabolism produces energy mainly by β-oxidation of Highly Unsaturated n-3 Fatty Acids (HUFA). Accordingly, kinetic activity causes higher consume of HUFA in red muscles than in white muscles, so the ratio between n-3 HUFA and their precursor C18:3 n-3 (ALA) is likely to be smaller in red than in white muscles. However, these ratios are highly affected by the environment. To reduce the effect of environmental variables, we propose an “Activity index” as the difference between n-3 HUFA/ALA in white and red muscle within the same bird. This index, measured after slaughtering, should represent the activity performed by the chicken during its life. Given that birds in good health had the possibility of moving, the “Activity index” would measure the activity actually performed by the animals. Should birds of a given strain show a higher activity level, this would be an indication of the suitability of that strain to outdoor systems. This work verified the application of this “Activity index” on 90 birds from 6 genetic strains with known kinetic behavior reared in an experimental farm. The “Activity index” was also tested on chicken strains collected form commercial organic farms. The results confirmed that strains recognized for higher kinetic attitude actually walked more and their behavior was clearly detected by the “Activity index” estimated from their muscles.

Abstract Background The nerve growth factor (NGF), a member of the neurotrophins family, plays an important role not only in the nervous but also in other non-nervous systems such as the reproductive system. The aim of the paper is to study the in vitro effect of NGF on rabbit sperm functions. Methods Ten adult rabbit bucks were collected five times, and pooled semen samples have been analysed. NGF was quantified in seminal plasma, and the distribution of NGF receptors (TrKA and p75NTR) in sperm was established. Moreover, the dose-effect of NGF on motility rate and track speed was evaluated. Successively, the effect of the neutralisation of NGF receptors was assessed to verify the specific role of each receptor. Untreated sperm were used as control. Results Our study identified several interesting results: i) We detected NGF in seminal plasma and TrKA and p75NTR in sperm surface. In particular, TrKA is localised in the head and p75NTR in the midpiece and tail of rabbit sperm. ii) Once the optimal dose of NGF (100 ng/mL) was established, its addition affected both kinetics and other physiological traits (capacitation, apoptosis and necrosis) of rabbit sperm. (iii) The neutralisation of TrKA and p75NTR receptors affected sperm traits differently. In particular, sperm speed, apoptosis and capacitation seemed mainly modulated via p75NTR receptor, whereas motile, live cells, necrosis and acrosome reaction were modulated via TrKA. Conclusion For the first time, we showed the presence of p75NTR in rabbit sperm. NGF affects kinetic and other physiological traits of rabbit sperm. Most of these changes are modulated by the receptors involved (TrKA or p75NTR). Considering that some seminal disorders in human have been correlated with a lower NGF concentration and no studies have been done on the possible involvement of NGF receptors, these findings also provide new insights on human fertility.

The aim of this research was to investigate the nutritional composition of raw and fried big-scale sand smelt (Atherina boyeri) from Trasimeno Lake. Four hundred big-scale sand smelts were caught with nets and analysed immediately. We created a total of 20 batches with 20 whole fish in each batch. Ten batches were analysed as raw samples, while the other 10 batches were analysed after being fried in sunflower oil at a temperature of 190 °C for 3 min (deep fat frying). The pH, proximate composition, fatty acid profile, oxidative stability and nutritional indexes of both groups were assessed. As expected, cooking strongly influenced the characteristics of the meat, mainly in terms of lipids, which were seven times greater in the fried product due to the oil. Frying also affected the fatty acid profile of the meat because oil absorption caused a significant increase in oleic and linoleic acids. Furthermore, we found a slight reduction in long chain n-3 fatty acids (eicosapentaenoic and docosahexaenoic acids). Frying increased oxidative processes and decreased the nutritional value of sand smelt. The obtained results can be considered preliminary because the effects of the fishing season and different physiological phases of sand smelt require further analytical confirmation.Highlights The aim of the research was to investigate the nutritional composition of raw and fried big-scale sand smelt (Atherina boyeri) from Trasimeno Lake. This study aimed to assess the effect of rearing system (intensive vs. free-range under vineyard) on the qualitative characteristics of goose meat. In particular, the integration of a goose raising system with an organic grape production was evaluated. Six hundred Romagnola geese of both sexes were divided into two groups at 21 d of age: Vineyard group (V) – 480 geese (120 geese/ha) pasturing in 4 ha of vineyard, and Control Group (C) – 120 geese of the same genotype in the experimental farm of Perugia, without any access to the pasture. Live weight, feed consumption and other performance were registered weekly on 30 selected and marked animals/group. At 160 days of age, 15 geese/group were slaughtered and characteristics of carcase, breast and drumstick (physical, chemical, oxidative status, fatty acids profile) were evaluated. The chemical analysis of diet and pasture and the ingestion of crude protein, digestible energy and bioactive compounds were also estimated. Vineyard geese, showed lower productive performance than the C ones (live and carcase weight), however, due to pasture availability, the intake of bioactive compounds (vitamin E, retinol, n-3 long-chain fatty acids) was higher and positively affected the antioxidant content of breast and drumstick. The higher kinetic activity of V geese reduced the fat amount of carcase and meat, whereas increased the development of drumstick muscle (higher meat/bone ratio) and worsened the oxidative status of meat. Concluding, the free-range vineyard geese resulted in a positive payoff on the geese meat quality viewpoint.Highlights Pasture enhance the amount of VIT E in goose muscle. The present survey evaluates production, research funds and scientific activity relating to rabbits in Italy, Europe and all over the world during the last 20 years. Official statistics have several weaknesses for a small and diversified production like that of rabbits. Different methods for collecting and providing data in the different countries can be used and data for backyard production can miss. According to FAO, in 2017, China and the Democratic People’s Republic of Korea contributed 73.3% to the global volume of rabbit meat. The two main EU producing countries, Spain and France, lost 12.2 and 6.4%, respectively, of their former share of world production compared to 1998. Whilst Italy is self-sufficient in rabbit meat production, trade exchanges are low, and meat consumption is decreasing. In regards to research funding, based on collected information which can be not fully comprehensive, China stands in the top position and Italy is placed fourth after France and Germany. Italy, France and Spain are among the top-five publishing countries and ‘World Rabbit Science’ is among the top publishing scientific journal in Agricultural and Biological Sciences (ABS). In the case of Veterinary Sciences, ‘Veterinary Record’ is one of the top journals. In Italy, ABS publications mainly focussed on Meat quality, Nutrition and feeding and Ethology and welfare. Pathology and hygiene and Breeding and genetics were the most published topics in Veterinary Science. The Universities of Napoli, Milano, Bari, Padova, Pisa, Perugia and Bologna provide teaching in rabbit science and production within their educational offer.Highlights Italian funds for research in rabbit science are minimal and mainly provided by the Italian Ministry of Health; Italian researchers in rabbit science are very active in publications through peer-reviewed international scientific journals; Italian research activity is focussed on Meat quality, Nutrition and feeding, and to a lesser extent Breeding and genetics and Welfare and ethology.

Chicken meat is becoming the most consumed in the world for both economic and nutritional reasons; regarding the latter, the lipid profile may play positive or negative roles in the prevention and treatment of diseases. In this study, we define the state of the art of lipid-based nutritional indexes and used the lipid content and fatty acid profile (both qualitative and quantitative) of breast meat of two poultry genotypes with different growth rates and meat traits. Further, we summarize and review the definitions, implications, and applications of nutritional indexes used in recent years and others of our own design to provide a useful tool to researchers working in the field of meat quality (not only in poultry) to select the most appropriate index for their own scientific purposes. All indexes show advantages and disadvantages; hence, a rational choice should be applied to consider the nutritional effect of meat on human health and for a possible assessment of the most suitable rearing systems (genotype, feeding, farming system or postmortem handling).

As organic and conventional poultry production increased in the last decade, so did consumers’ concerns, sustainability requirements, and animal welfare as well as health issues. According to Reg. EU 848/2008 on organic production, poultry must be adapted to organic outdoor systems and cope with all the regulatory constraints in terms of nutrition, health, and welfare. Adaptability must take into account the above challenges, constraints, and concerns. Chicken adaptability should not only mean being able to use pasture and outdoor areas, but also mean being able to overcome, or be resilient to, the challenges of organic farming without compromising welfare, performance, and product quality. This commentary identifies solutions to the new challenges that organic poultry chains must face in future productive scenarios, detects consumer viewpoints to provide a perspective on organic poultry production, and summarizes as well as defines chicken adaptability to organic production, assessing the main factors of chicken adaptability.

The beneficial effects of n-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) on human health are widely known. Humans are rather inefficient in synthesizing n-3 LC-PUFA; thus, these compounds should be supplemented in the diet. However, most Western human diets have unbalanced n-6/n-3 ratios resulting from eating habits and the fact that fish sources (rich in n-3 LC-PUFA) are not sufficient (worldwide deficit ~347,956 t/y) to meet the world requirements. In this context, it is necessary to find new and sustainable sources of n-3 LC-PUFA. Poultry products can provide humans n-3 LC-PUFA due to physiological characteristics and the wide consumption of meat and eggs. The present work aims to provide a general overview of the main strategies that should be adopted during rearing and postproduction to enrich and preserve n-3 LC-PUFA in poultry products. The strategies include dietary supplementation of α-Linolenic acid (ALA) or n-3 LC-PUFA, or enhancing n-3 LC-PUFA by improving the LA (Linoleic acid)/ALA ratio and antioxidant concentrations. Moreover, factors such as genotype, rearing system, transport, and cooking processes can impact the n-3 LC-PUFA in poultry products. The use of a multifactorial view in the entire production chain allows the relevant enrichment and preservation of n-3 LC-PUFA in poultry products.

Defining optimal nutrition in animals and humans remains a main scientific challenge. The objective of the work was to develop a dynamic model of reactive oxygen species (ROS)–polyunsaturated fatty acid (PUFA)–antioxidant homeostasis using the rabbit as a model. The problem entity was to evaluate the main metabolites generated from interactions between traits included in the conceptual model and identified by three main sub–models: (i) ROS generation, (ii) PUFA oxidation and (iii) antioxidant defence. A mathematical model (VENSIM software) that consisted of molecular stocks (INPUTs, OUTPUTs), exchange flows (intermediate OUTPUTs) and process rates was developed. The calibration was performed by using standard experimental data (Experiment 1), whereas the validation was carried out in Experiments 2 and 3 by using supra–nutritional dietary inputs (VIT E+ and PUFA+). The accuracy of the models was measured using 95% confidence intervals. Analytical OUTPUTs (ROS, PUFA, Vit E, Ascorbic acid, Iso–/NeuroProstanes, Aldehydes) were well described by the standard model. There was also good accuracy for the VIT E+ scenario, whereas some compensatory rates (Kc1–Kc4) were added to assess body compensation when high levels of dietary PUFA were administered (Experiment 3). In conclusion, the model can be very useful for predicting the effects of dietary treatments on the redox homeostasis of rabbits.

Background: Nerve Growth Factor (NGF) plays an important role in the reproductive system through its receptor’s interaction (p75NTR). This paper aims to analyze the impact of NGF p75NTR in epididymal and ejaculated rabbit semen during in vitro sperm storage. Methods: Semen samples from 10 adult rabbit bucks were collected four times (n = 40) and analyzed. NGF was quantified in seminal plasma, and the basal expression of p75NTR in sperm was established (time 0). Moreover, we evaluated p75NTR, the apoptotic rates, and the main sperm parameters, at times 2–4 and 6 h with or without the administration of exogenous NGF. Results: Based on the level of p75NTR, we defined the threshold value (25.6%), and sperm were divided into High (H) and Normal (N). During sperm storage, p75NTR of H samples significantly modulated some relevant sperm parameters. Specifically, comparing H samples with N ones, we observed a reduction in motility and non-capacitated cell number, together with an increased percentage of dead and apoptotic cells. Notably, the N group showed a reduction in dead and apoptotic cells after NGF treatment. Conversely, the NGF administration on H sperm did not change either the percentage of dead cells or the apoptotic rate. Conclusion: The concentration of p75NTR on ejaculated sperm modulates many semen outcomes (motility, apoptosis, viability) through NGF interaction affecting the senescence of sperm.

The present study evaluated the effects of feed supplemented with two dietary sources of n-3 polyunsaturated fatty acids (PUFAs; fish oil and extruded flaxseed) on the gut microbiota, caecal fermentations, gastrointestinal histology, and histochemistry in rabbits. Fifteen male New Zealand White rabbits were divided into three groups (n = 5/group) and fed with different diets from weaning (35 days of age) until slaughtering (90 days of age): C group, fed with a commercial diet; F group, supplemented with 10% of extruded flaxseed; and O group, supplemented with 3.5% of fish oil. At slaughter, the content of the stomach, duodenum, jejunum, ileum, caecum, and colon was collected and analyzed by Next Generation 16S rRNA gene sequencing. Tissue samples of the same tracts were evaluated with histological and histochemical analysis. Ammonia and lactic acid in the caecum were also quantified. Twenty-nine operational taxonomic units (OTUs) were significantly different between groups. Groups receiving n-3 PUFAs supplementation showed an increase in Bacteroidetes and Lachnospiraceae in several gastrointestinal tracts, while Bacilli abundance, as well as Firmicutes/Bacteroidetes ratio, were reduced compared to the control group (for all p < 0.05). Caecal ammonia was lower in the F than C group (p < 0.032), whereas no difference was found for lactic acid. Finally, histological evaluations revealed a mild hemorrhagic infiltration and vessels ectasia in the stomach mucosa of both F and O groups, but no effect of nutritional treatment was evidenced by the histochemical analyses. In conclusion, n-3 PUFAs supplementation could modify the rabbit gut microbiota and fermentation. The increase in beneficial bacterial populations may, at least partially, explain the positive effects of n-3 PUFAs diet supplementation on human and animals’ health, although the appropriate dosage should be established.

The aim of the present study was to investigate the effect of transport on welfare traits, several haematological parameters and carcase hygiene in two different chicken genotypes (fast- and slow-growing strains) reared under free-range conditions. For this aim, two hundred male chicks, 100 from fast-growing (Ross 308, R) and 100 from slow-growing (Naked Neck, NN) strain were farmed. At the end of the rearing period, at 81 days of age, 56 birds/strain were randomly selected for slaughtering and submitted to two different pre-slaughter conditions: no transport (0h) or 4 hours of transport (4h). Tonic immobility (TI), blood parameters and carcase hygiene traits were determined. Strain and transport significantly affected TI of birds. Both experimental factors and their interaction significantly affected plasma creatine kinase, alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase. Cholesterol and triglycerides were not different between the experimental groups, whereas glucose decreased after 4 hours of transport in both strains. A significant difference between groups for the heterophils/lymphocytes ratio after transport was also observed, with NN being higher than Ross. Concerning the oxidative stress, we observed a higher ROS production in NN chickens. The carcase microbial characteristics showed a higher level of contamination after the transport (total viable counts), but not concerning the Enterobacteriaceae counts.

The neurotrophin β-nerve growth factor (NGF) has been described as an important mediator of seminal plasma-induced ovulation in reflexive ovulators like camelids, but this action has not been evidenced in rabbits. In this study, the mRNA expression of NGF was analysed in rabbit male organs by PCR and in a purified basic fraction of seminal plasma by liquid chromatography mass spectrometry. Besides, the ovulatory response [ovulation rate (OR), corpora lutea number, LH peak and progesterone profile] of 27 rabbit does was recorded after injection with murine NGF, gonadoreline (GnRH) or saline solution (SS). An empty catheter was introduced through the vagina (n = 4 animals per group) to simulate the nervous/mechanical stimulus of coitus. Molecular studies confirmed the expression of β-NGF mRNA in the prostate, seminal gland and testicles of male adult rabbits, and the presence of the protein in seminal plasma. For the in vivo experiment, all GnRH-group does exhibited a significantly higher OR (100%) compared to females treated with NGF (30%) and SS (25%), who showed significantly lower LH and progesterone concentrations at 120 min. However, in the non-vaginally stimulated animals, only the gonadoreline group ovulated properly, since no ovulation was recorded in the SS group, and only 17% of the NGF does ovulated; this was associated to a tendency to a higher rate of anovulatory haemorrhagic follicles. mRNA expression of NGF and its presence in seminal plasma evidence a role in rabbit reproduction. Nevertheless, murine NGF triggered some stimulatory effect in the females but not enough to elicit a significant ovulatory response.

The aim of the present study was to develop a multicriteria model for the comparison of three commercial poultry farms: organic with Ross 308 genotype (OR), organic with Naked Neck genotype (ONN) and a conventional system (C), which represents the most common commercial farming system. A model based on multicriteria decision analysis was developed, considering for the first time the One Welfare approach in an operational manner, including three dimensions: human, environmental and animal welfare. The three alternatives demonstrated different performances, according to the different dimensions considered. In particular, the two organic systems performed better for human welfare and animal welfare, with relevant differences due to the genetic strains used. Conventional rearing performed better for the environment index due to the method chosen. The multicriteria analysis showed that the organic system performed better overall than the conventional system. In particular, the use of an adapted Slow Growing (SG) strain positively affected the final rank, mainly by reducing welfare problems and producing good economic and social performance. The stability of the results was verified by performing a sensitivity analysis, specifically a weight stability analysis, which confirmed the strength of results.

This study aimed to develop an adaptability score (AS) for chicken strains, which includes behavioral, plumage conditions, and body lesion indicators through a multifactorial approach. A total of 600 male chickens from 6 poultry genotypes—Ranger Classic (R1), Ranger Gold (R2), Rowan Ranger (R3), Hubbard Red JA (A), CY Gen 5 × JA87 (CY), and M22 × JA87 (M)—were reared under organic conditions, fed ad libitum, and individually weighed weekly to calculate the daily weight gain (DWG). The behavioral observations consisted of the explorative attitude (EA), recorded at 21 days, and the behavioral patterns (BPs) recorded the week before the slaughter. The AS was established by a principal component analysis, and the AS of these genotypes was compared. Moreover, the effect of DWG and genotype on the AS was evaluated by univariable and multivariable regression models. Although the DWG and genotype were strictly dependent, genotype was the most important factor affecting the AS. In fact, its effect was significant both in univariable (p < 0.001) and multivariable models (p < 0.001). Conversely, the DWG was significant only in the univariable and lost significance when the effect of genotype was introduced in the model.

F4-neuroprostanes (F4-NeuroPs), derived from the oxidative metabolization of docosahexaenoic acid (DHA), are considered biomarkers of oxidative stress in neurodegenerative diseases. Neurons and spermatozoa display a high DHA content. NeuroPs might possess biological activities. The aim of this in vitro study was to investigate the biological effects of chemically synthetized 4-F4t-NeuroP and 10-F4t-NeuroP in human sperm. Total progressive sperm motility (p < 0.05) and linearity (p = 0.016), evaluated by a computer-assisted sperm analyzer, were significantly increased in samples incubated with 7 ng F4-NeuroPs compared to non-supplemented controls. Sperm capacitation was tested in rabbit and swim-up-selected human sperm by chlortetracycline fluorescence assay. A higher percentage of capacitated sperm (p < 0.01) was observed in samples incubated in F4-NeuroPs than in the controls. However, the percentage of capacitated sperm was not different in F4-NeuroPs and calcium ionophore treatments at 2 h incubation. The phosphorylated form of AMPKα was detected by immunofluorescence analysis; after 2 h F4-NeuroP incubation, a dotted signal appeared in the entire sperm tail, and in controls, sperm were labeled in the mid-piece. A defined level of seminal F4-NeuroPs (7 ng) showed a biological activity in sperm function; its addition in sperm suspensions stimulated capacitation, increasing the number of sperm able to fertilize.

The stomach content and samples of Longissimus dorsi muscle of 32 feral wild boars were collected in two different feeding areas (forest and farmland) of Umbria region (Italy). The animals from each feeding area were divided into two age classes: class 1 (12–24 months of age; 48 kg average weight) and class 2 (animals older than 2 years of age; 84 kg average weight). The major food categories consumed were hard mast and crops (89.02–75.98%). The L*(lightness) and a*(red to green colour) values of the meat were affected by the feeding area as well as the b*(yellow colour) value; the age significantly affected only the a* and the b* value of the meat. The α-tocopherol was the most abundant vitamin E homologue, ranged between 520.63 and 1881.33 ng/g and was higher in farmland areas. The index of lipid oxidation (TBARS) ranged from 0.093 and 0.140 mg MDA/kg and was higher in wild boars from farmland. The monounsaturated fatty acids (MUFA) ranged between 38.36 and 46.75% and were higher in wild boar of class 2. The total polyunsaturated fatty acids (PUFA) as well as PUFAn-6 were affected by age, while PUFAn-3 was only affected by feeding area and ranged from 0.91 and 1.99 in farmland and forest, respectively. The feeding area affects the intramuscular fat contents in terms of nutritional characteristics of the meat: the n-6/n-3 ratio that was lower in meat from animals hunted in the forest area (p ≤ .001), as well as the ARA/(EPA + DHA) ratio (p ≤ .01).

Polyunsaturated fatty acid (PUFA) metabolism and tissue distribution is modulated by the oxidation of these molecules. This research aimed to investigate the implication of dietary n-3 PUFA supplementation (precursor and long-chain PUFA) on the PUFA profile and oxidative status of the liver, testis, and brain of adult rabbit bucks. Twenty New Zealand White rabbit bucks were divided into four experimental groups (n = 5 per group) and were fed different diets for 110 days: control (CNT), standard diet containing 50 mg/kg alpha-tocopheryl acetate (vitamin E); CNT+, standard diet + 200 mg/kg vitamin E; FLAX, standard diet + 10% flaxseed + 200 mg/kg vitamin E; or FISH, standard diet + 3.5% fish oil + 200 mg/kg vitamin E. Antioxidants (enzymatic and non-enzymatic), oxidative status (malondialdehyde and isoprostanoids), and n-3 and n-6 PUFAs of tissues were analysed. A chain mechanism of oxidant/antioxidant molecules, which largely depended on the particular PUFA composition, was delineated in the different organs. The liver showed an oxidant/antioxidant profile and lipid pathways widely modulated by PUFA and vitamin E administration; on the other hand, the testis’ oxidative profile rather than its lipid profile seemed to be particularly affected, an outcome opposite to that of the brain (modulation operated by dietary PUFA).

The demand for poultry meat, being cheaper than red meat, will drive worldwide production of this product. Accordingly, an increase in production up to 16% is expected in 2025, most of which will occur in developing countries. Most poultry meat production is realized with intensive production systems, and extensive rearing systems (ERS) of poultry (organic, free-range, and low-input) represent only a small portion of poultry production in the EU (about 5%). However, there is an increasing interest in such rearing systems to maintain the good image of product and environmental sustainability, improved animal welfare, and meat quality with an annual trend of growth of about 10%. The aims of this work were to summarize the activities and the viewpoint of the researchers of the Department of Agricultural, Food, and Environmental Science of the University of Perugia (Italy). One of the most important goals of the research unit was the challenge of identifying the best poultry genotypes for ERS, which are important not only for the food industry but also for the improvement of human nutrition. Only the definition of the best genotypes adapted to ERS through the measurement of a wide panel of traits—genetic, physiologic, and behavior—and not only relying on daily weight gain will allow us to achieve this goal.

The most reared species of farm animal around the world is the chicken. However, the intensification of livestock systems has led to a gradual increase in the concentration of a limited number of breeds, resulting in substantial erosion to the genetic pool. The initial step of an ‘animal conservation program’ entails establishing the actual conservation statuses of the breeds concerned in a defined area; in this case, in Italy. To this end, a survey of breeds was performed by means of a census questionnaire divided into two parts. The first part collected information on breeds, breeders, housing facilities, and management aspects, the results of which are presented here. The second part of the questionnaire regarded chicken products and their markets, and these data will be reported in a second paper. The breed status of six chicken breeds was shown to be exceptionally worrying, with total numbers ranging from just 18 to 186 birds. Population sizes exceeding 1000 birds was identified for just four breeds, the maximum being 3400. Some improvements in status were noted in relation to breeds which had been the subject of conservation efforts in the past. The two most common breeds reported are the Bionda Piemontese, a double-purpose breed, and the Livorno egg-laying hen. Collo Nudo Italiano, Millefiori Piemontese, Pollo Trentino, and Tirolese chicken breeds and the Castano Precoce turkey breed were not listed by breeders at all. The most reported turkey breeds are the Bronzato Comune and the Ermellinato di Rovigo. The population sizes of native Italian poultry breeds were shown to be generally poor. Italian poultry farmers and the population at large are largely ignorant about indigenous poultry breeds. Thus, promoting the virtues of Italian breeds would help their conservation by encouraging breeders to rear these birds and consumers to buy their products. The identification of strategies to facilitate access to pure breed birds is essential, and will require the collaboration of university research centers, public entities, and breeders. The results presented in this paper constitute the initial part of a more complex conservation program.
