Pelleted total mixed ration (P-TMR) feeding, which has become a common practice in providing nutrition for fattening sheep, requires careful consideration of the balance between forage neutral detergent fiber (FNDF) and rumen degradable starch (RDS) to maintain proper rumen functions. The present study aimed to investigate the effects of the dietary FNDF/RDS ratio (FRR) on chewing activity, ruminal fermentation, ruminal microbes, and nutrient digestibility in Hu sheep fed a P-TMR diet. This study utilized eight ruminally cannulated male Hu sheep, following a 4 × 4 Latin square design with 31 d each period. Diets consisted of four FRR levels: 1.0 (high FNDF/RDS ratio, HFRR), 0.8 (middle high FNDF/RDS ratio, MHFRR), 0.6 (middle low FNDF/RDS ratio, MLFRR), and 0.4 (low FNDF/RDS ratio, LFRR). Reducing the dietary FRR levels resulted in a linear decrease in ruminal minimum pH and mean pH, while linearly increasing the duration and area of pH below 5.8 and 5.6, as well as the acidosis index. Sheep in the HFRR and MHFRR groups did not experience subacute ruminal acidosis (SARA), whereas sheep in another two groups did. The concentration of total volatile fatty acid and the molar ratios of propionate and valerate, as well as the concentrate of lactate in the rumen linearly increased with reducing dietary FRR, while the molar ratio of acetate and acetate to propionate ratio linearly decreased. The degradability of NDF and ADF for alfalfa hay has a quadratic response with reducing the dietary FRR. The apparent digestibility of dry matter, organic matter, neutral detergent fiber, and acid detergent fiber linearly decreased when the dietary FRR was reduced. In addition, reducing the dietary FRR caused a linear decrease in OTUs, Chao1, and Ace index of ruminal microflora. Reducing FRR in the diet increased the percentage of reads assigned as Firmicutes , but it decreased the percentage of reads assigned as Bacteroidetes in the rumen. At genus level, the percentage of reads assigned as Prevotella , Ruminococcus , Succinivibrio , and Butyrivibrio linearly decreased when the dietary FRR was reduced. The results of this study demonstrate that the dietary FRR of 0.8 is crucial in preventing the onset of SARA and promotes an enhanced richness of ruminal microbes and also improves fiber digestibility, which is a recommended dietary FRR reference when formulating P-TMR diets for sheep. Introduction: Defined seasonality in savanna species can stimulate physiological responses that maximize photosynthetic metabolism and productivity. However, those physiological responses are also linked to the phenological status of the whole plant, including leaf phenophases. Objective: To study how physiological traits influence phenophase timing among congeneric and co-occurring savanna species. Methods: We evaluated the leaf phenology and physiological traits of populations of Byrsonima intermedia, B. coccolobifolia, and B. verbascifolia. Physiological measurements were performed at the onset of the dry and rainy seasons and again late in the season. Results: B. intermedia and B. coccolobifolia were classified as brevideciduous and B. verbascifolia as evergreen. The maximum quantum yield for B. intermedia and B. coccolobifolia were lowest during the dry season. At the onset of the dry period, the highest chloroplastidic pigment levels were observed, which decreased as the season advanced, total chlorophyll/carotenoid ratios were lowest, and carotenoid contents were highest. We detected low starch content values at the start of the rainy season, coinciding with the resumption of plant growth. Two months into this season, the leaves were at their peak structural and functional maturity, with high water-soluble polysaccharide values and photosynthetic rates, and were storing large amounts of starch. Conclusions: Physiological and leaf phenological strategies of the Byrsonima species were related to drought resistance and acclimatization to the seasonality of savanna water resources. Metamitron is an apple (Malus domestica Borkh.) fruit thinner that acts by reducing the photosynthetic capacity of trees. Relative humidity may influence thinning efficacy; however, the broadness of this effect is not yet fully understood. Trials were set in Sint-Truiden (Belgium) in 2018 and Lleida and Girona (Spain) in 2019, using 4-year-old cvs. Braeburn and Elstar trees in Belgium, and 16-year-old cv. Golden Reinders trees in Spain. Four treatments were implemented at the stage of 12-14 mm fruit diameter: (a) CTR--control, trees under natural environmental conditions; (b) HH--high humidity, trees submitted to artificially increased air relative humidity applied for 3 h prior to the beginning of the experiment; (c) MET--trees sprayed with 247.5 mg/L metamitron; (d) MET + HH--trees submitted to the combination of increased humidity (HH) and metamitron (MET) application. In Belgium, metamitron absorption by leaves was greater than in Spain. This might be related to morphological characteristics of the leaves which developed under greater natural relative humidity levels in Belgium than in Spain. Compared to MET alone, 'Braeburn' and 'Elstar' demonstrated significantly greater metamitron absorption, 59% and 84%, respectively, under MET + HH, accompanied by declines in leaf net photosynthesis (10% and 32%, respectively) and sucrose (31% and 26%, respectively). At harvest, MET + HH treatment reduced yield by 24% and 32% in 'Braeburn' and 'Elstar', respectively, when compared with MET alone. A large reduction (considered over thinning) in the yield of 'Elstar' occurred. In contrast, metamitron absorption by 'Golden Reinders' using MET alone was similar to MET + HH; however, there was a slight foliar sugar reduction in the latter treatment. In addition, both treatments enhanced shoot growth and increased fruit abscission with similar improvements in fruit weight and size. In this study, high relative humidity enhanced fruit thinning efficacy under certain circumstances, such as age or genetic predisposal, which left the tree more susceptible to a negative carbohydrate balance. For instance, 'Braeburn' and 'Elstar' were easier to thin when compared to 'Golden Reinders'. In addition, this study raises a question that requires further research regarding the impact of HH before and after spraying as well as its effect in combination with higher temperatures. Drought events impair the carbon and water balances in plants. Climate changes highlight the importance to understand the limits of woody species to reallocate carbon in different processes and the mechanisms driving the osmotic adjustments during the day under stress. In this frame, the aim of this work was to investigate the plant capability to shift energy among competing sinks and preserve the osmotic balance during the day under severe short periods of water deficit. The role of carbohydrates as osmolytes as well as energy sources was investigated in poplar plants. Results highlighted that during water deficit soluble sugars, derived both from the new synthetised carbon and starch degradation, were principally convoyed in the bark. This increase in carbohydrates allowed the maintenance of a water reserve used during the day to prevent a water decrease within the xylem. The decrease of xylem sap osmotic potential during the night, driven by an increase of K, Ca, and fructose (+0.46, 0.52, and 0.26 mg ml−1 in water limited plants after 8 days of withholding water, respectively), probably further attracted water into the xylem. This response mechanism increased at higher water deficit intensity. The little variations in carbohydrates and mineral elements within the leaves highlighted the main role of sinks rather than sources in the early response to water deficit. • DS inhibited in vivo pollen tube growth due to restricted carbon and energy supply. • Low carbon accumulation in droughted pistils was reflected as low starch deposit. • Low starch deposit in droughted pistils was due to low AGPase and SSSase activities. • High sucrose level in droughted pistils was due to decreased SuSy and acid invertase activities. • Low expression of GhSusA, GhSusB, GhSusD, GhINV1 and GhINV2 in pistil also contributed to the high sucrose level under DS. It has been reported that drought stress (DS) reduces cotton yield by negatively affecting reproductive activities. Some studies have investigated the effects of DS on pollen physiology and biochemistry, but studies exploring the impact of drought on pistil biochemistry and its relationship with pollen tube growth rates in vivo are scarce. In order to investigate these objectives, a greenhouse study was conducted with a drought sensitive cotton cultivar, Yuzaomian 9110. Two water treatments were imposed at flowering stage, 1. control, where plants were irrigated with optimum quantity of water and 2. DS treatment, where plants were irrigated with 50% of the optimum quantity of water. Results indicated that stored starch content at the early stage of pollen tube growth (12:00 h) was 31.6% lower in drought-stressed pistils than control pistils, and it was highly correlated with pollen tube growth rate. The decline in starch accumulation of drought-stressed pistils could be attributed to the impeded transport of photosynthetic carbon assimilates. Moreover, decreased ADP-glucose pyrophosphorylase and soluble starch synthase activities also resulted in curtailing starch accumulation in drought-stressed pistils. Furthermore, pistil sucrose concentration was significantly higher in droughted plants relative to control plants at 12:00 and 18:00 h (during the rapid growth period), which was due to lower activities of sucrose synthase and acid invertase, and the down-regulated expressions of sucrose synthase genes, GhSusA , GhSusB and GhSusD , and acid invertase genes, GhINV1 and GhINV2 , in drought-stressed pistils, limiting as a result the hydrolysis of sucrose into hexose. Drought-stressed pistils sampled at 18:00 h had lower α-amylase activity compared to control pistils, resulting in decreased starch decomposition, which, in conjunction with the decreased hydrolysis of sucrose, led to lower glucose and fructose contents in drought-stressed pistils at 18:00 h. Finally, lower pyruvate level in drought-stressed pistils could not produce enough acetyl-CoA in the tricarboxylic acid cycle to yield sufficient energy (ATP) for pollen tube growth. We conclude that DS disrupts the carbohydrate balance of pistil, reducing as a consequence carbon and energy supply for pollen tube elongation in the style, which will ultimately result in reproductive failure. Thinning strategies, namely shade or photosynthetic inhibitors, rely on the reduction of carbon supply to the fruit below the demand, causing fruit abscission. In order to clarify the subject, seven field trials were carried out in Lleida, Girona, and Sint-Truiden (2017 + 2018), using orchards of ‘Golden’ and ‘Gala’ apple trees. At the stage of 9–14-mm fruit diameter, four treatments were implemented: (A) CTR-control, trees under natural environmental conditions; (B) SN-shaded trees, trees above which shading nets reducing 50% of irradiance were installed 24 h after metamitron application date—without application of metamitron—and removed after five days; (C) MET-trees sprayed with 247.5 ppm of metamitron; (D) MET + SN-trees submitted to the combined exposure to metamitron application and shading nets. Low radiation significantly increased metamitron absorption (36–53% in the three locations in 2018) and reduced its degradation. Net photosynthesis and stomatal conductance were strongly reduced in all treatments, with minimum values 2 days after spraying (DAS) and incomplete recovery 10 DAS in MET + SN. All treatments resulted in leaf sucrose and sorbitol decreases, leading to a negative carbon balance. SN and MET + SN promoted the highest thinning efficacy, increasing fruit weight and size, with MET + SN causing over-thinning in some trials. Leaf antioxidant enzymes showed moderate changes in activity increases under MET or MET + SN, accompanied by a rise of glutathione content and a reduction in ascorbate, however without lipid peroxidation. This work shows that environmental conditions, such as cloudy days, must be carefully considered upon metamitron application, since the low irradiance enhances metamitron efficacy and may cause over-thinning.

Reproductive success in higher plants requires a lot of energy and substance provided by carbohydrate and protein metabolism, and potassium (K) plays an important role in carbohydrate and protein metabolism. However, it is unclear whether K deficiency limits reproductive success by disturbing carbohydrate and protein metabolism. The objectives of this study were to explore the effects of K deficiency on carbohydrate and protein metabolism in subtending leaves, phloem and pistils, and their relationship with reproductive success. A cotton cultivar DP0912 was grown in K-deficient (0 mM K + ) and K-sufficient (6 mM K + ) nutrient solution in growth chambers. Results showed that Pn of the subtending leaves was decreased under K deficiency, but sucrose, starch and free amino acid contents were markedly increased in the K-deficient leaves, because K deficiency limited the translocation of sucrose and amino acid in phloem. As a result, sucrose and free amino acid contents were reduced by 47.3% and 51.8% in the K-deficient pistils than K-sufficient pistils, which led to further decreases in starch and protein accumulation in the K-deficient pistils. Glucose content was also reduced by 53.1% in the K-deficient pistils than K-sufficient pistils, due to the decreased acid and alkaline invertase activities, since sucrose synthase activity was not affected. Lastly, soluble carbohydrate and ATP contents were lower in the K-deficient pistils than K-sufficient pistils, similarly to the changes of pollen tube growth rate and seed set efficiency. It was concluded that the lower carbohydrate and ATP contents in the K-deficient pistils could not meet the energy requirements of pollen tube growth and seed set. Energy is a concept of universal importance. In applying it to body weight regulation, the focus has been on energy balance and how this balance is affected by intakes and expenditures. However, energy is an abstract concept without biological equivalent and applying it to explain body weight regulation has led to various misconceptions and created intellectual obstacles in understanding the obesity problem. When nutrient and substrate interactions are considered, instead, a number of important issues pertaining to body weight regulation and to the obesity epidemic can be much more pertinently addressed. The present study aimed to investigate whether substrate metabolism, appetite and feeding behaviour differed between high and low energy turnover conditions. Thirteen overweight premenopausal women completed two 1 d trials: low energy turnover (LET) and high energy turnover (HET), in a randomised, cross-over design. In LET, subjects consumed a test breakfast (49 % carbohydrate, 37 % fat, 14 % protein) calculated to maintain energy balance over a 6 h observation period, during which metabolic rate and substrate utilisation were measured and blood samples taken. Immediately following this an ad libitum buffet meal was provided. HET was identical to LET, except that subjects walked on a treadmill for 60 min at 50 % VO2max before the test breakfast, which was increased in size (by about 65 %) to replace the energy expended during the walk and maintain energy balance over the observation period. Postprandial fat balance (i.e. the difference between fat intake and oxidation) was lower and carbohydrate balance higher in HET compared with LET throughout the postprandial period (P < 0·05 for both). After the buffet meal, carbohydrate balance did not differ between trials but energy and fat balances were lower (by 0·28 MJ and 11·6 g, respectively) in HET compared with LET (P < 0·001 for both). Carbohydrate balance immediately before the buffet meal correlated negatively with buffet energy intake (r − 0·49) and postprandial acylated ghrelin responses (r − 0·48), and positively with postprandial glucose responses (r 0·49). These findings demonstrate that HET resulted in a more positive carbohydrate balance than LET, which associated with lower subsequent energy intake. Energy homeostasis is critical for the survival of species. Therefore, multiple and complex mechanisms have evolved to regulate energy intake and expenditure to maintain body weight. For weight maintenance, not only does energy intake have to match energy expenditure, but also macronutrient intake must balance macronutrient oxidation. However, this equilibrium seems to be particularly difficult to achieve in individuals with low fat oxidation, low energy expenditure, low sympathetic activity or low levels of spontaneous physical activity, as in addition to excess energy intake, all of these factors explain the tendency of some people to gain weight. Additionally, large variability in weight change is observed when energy surplus is imposed experimentally or spontaneously. Clearly, the data suggest a strong genetic influence on body weight regulation implying a normal physiology in an ‘obesogenic’ environment. In this study, we also review evidence that carbohydrate balance may represent the potential signal that regulates energy homeostasis by impacting energy intake and body weight. Because of the small storage capacity for carbohydrate and its importance for metabolism in many tissues and organs, carbohydrate balance must be maintained at a given level. This drive for balance may in turn cause increased energy intake when consuming a diet high in fat and low in carbohydrate. If sustained over time, such an increase in energy intake cannot be detected by available methods, but may cause meaningful increases in body weight. Background: The prevention and treatment of obesity is a public health challenge. Objective: We investigated the effects of dietary composition, insulin sensitivity (SI), and energy balance on predicted changes in body composition. Design: In a randomized crossover design study, 39 normal-weight (n = 23), overweight (n =8), and obese (n = 8) men and women (aged 25-36 y) each followed a 15-d isocaloric high-fat (HF; 50% fat) and high-carbohydrate [HC; 55% carbohydrate (CHO)] diet with a 4-6-wk washout period during the first year. During each treatment, energy balance was measured while the subjects were inactive by using indirect calorimetry on day 15, and SI was measured by using a euglycemic clamp study (40 mU . m-2. min-1) on day 16. Weight and body composition were then measured annually for 4 y. The outcomes for fat mass, percentage body fat, and weight were measured by using a linear 2-stage mixed model. Results: CHO balance (day 15) and SI (day 16) on the HC diet were highly and significantly correlated (r=0.55, P<0.001). On theHC diet, the subjects who had a higher positive CHO balance (day 15) gained less fat mass (P <0.001), percentage body fat (P < 0.006), and weight (P = 0.024) over time. When adjusted for SI, CHO balance remained a significant predictor of changes in fat mass (P= 0.021) and percentage body fat (P = 0.025). Conclusions: On a HC diet, the subjects who had a higher positive CHObalance on day 15 while they were inactive gained less fat mass during 4 y, a predictive effect independent of SI. The measurement of fat balance (fat input minus fat output) involves the accurate estimation of both metabolizable fat intake and total fat oxidation. This is possible mostly under laboratory conditions and not yet in free-living conditions. In the latter situation, net fat retention/mobilization can be estimated based on precise and accurate sequential body composition measurements. In case of positive balance, lipids stored in adipose tissue can originate from dietary (exogenous) lipids or from nonlipid precursors, mainly from carbohydrates (CHOs) but also from ethanol, through a process known as de novo lipogenesis (DNL). Basic equations are provided in this review to facilitate the interpretation of the different subcomponents of fat balance (endogenous vs exogenous) under different nutritional circumstances. One difficulty is methodological: total DNL is difficult to measure quantitatively in man; for example, indirect calorimetry only tracks net DNL, not total DNL. Although the numerous factors (mostly exogenous) influencing DNL have been studied, in particular the effect of CHO overfeeding, there is little information on the rate of DNL in habitual conditions of life, that is, large day-to-day fluctuations of CHO intakes, different types of CHO ingested with different glycemic indexes, alcohol combined with excess CHO intakes, etc. Three issues, which are still controversial today, will be addressed: (1) Is the increase of fat mass induced by CHO overfeeding explained by DNL only, or by decreased endogenous fat oxidation, or both? (2) Is DNL different in overweight and obese individuals as compared to their lean counterparts? (3) Does DNL occur both in the liver and in adipose tissue? Recent studies have demonstrated that acute CHO overfeeding influences adipose tissue lipogenic gene expression and that CHO may stimulate DNL in skeletal muscles, at least in vitro. To clarify how both interact in apple tree physiologic mechanisms and on fruit abscission, five field trials were carried out in Lleida, Girona and Sint-Truiden (2017 + 2018), using orchards of 'Golden' apple trees. At the stage of 12–14 mm fruit diameter, four treatments were established: (A) CTR – control, trees under natural environmental conditions; (B) HNT – high nighttime temperature, trees exposed to artificially increased nighttime temperature during 5 nights after the day of spraying, without metamitron application; (C) MET - 247.5 ppm of metamitron application and (D) MET + HNT - trees submitted to the combined exposure to metamitron application (MET) and to artificially increased nighttime temperature (HNT). HNT did not affect metamitron absorption, net photosynthesis (P n) and stomatal conductance however, promoted significant reductions in leaf CH content mainly before sunrise, especially in sucrose (18–45%) and in sorbitol (19–26%). Metamitron significantly reduced P n to about 50% of CTR, which resulted in decreases in leaf sucrose and sorbitol, reaching minimum values 5 days after spraying, between 21 and 57% and 19–26%, respectively. Fruit growth rate of both treatments was retarded by 30%, 2 days after either metamitron application or HNT. Both treatments originated a similar reduction in the number of fruits and size improvement. The combined exposure (MET+HNT) promoted similar P n reductions as MET, but showed the greatest sucrose (44–60%) and sorbitol (73–84%) decreases which resulted in the strongest thinning efficacy. Lipid peroxidation was not affected by the treatments however, antioxidant enzyme activity showed moderate changes with activity increases mainly under MET and MET + HNT, accompanied by a rise in glutathione content and reduction in ascorbate. This work shows that the overlap of photosynthesis inhibition (reducing CH production) by means of metamitron spraying, and likely greater respiration (increased CH consumption), by HNT imposition, translates less CH production than the growing fruits demand (negative CH balance) leading to a metamitron thinning effect enhancement. Fructan synthesis and carbon partitioning to roots of tall fescue (Festuca arundinacea Schreb.) were studied with a 14C-labelling technique at three growth temperatures: 24/17 °C, 16/10 °C and 8/5 °C (day/night). Plants at 16/10°C and 8/5 °C had an increased proportion of assimilates allocated to the root system; 10 d after exposure at 14CO2, the relative carbon partitioning to the roots averaged 12.5, 16.3 and 26.8% respectively, at 24/17 °C, 16/10 °C and 8/5 °C. Chilling temperatures induced an increase in the amounts of sucrose and fructans in leaves. Interestingly, a similar pattern occurred in roots where low-DP fructans (DP < 8) are the predominant non- structural carbohydrate constituent. Incorporation of 14CO2 into root oligo- and polysaccharides showed a progressive movement of radioactivity from sucrose to fructans. These results show that the roots can serve as a carbon storage organ and that fructan synthesis depends on sucrose supply. The function of the fructan stored in the roots is unknown. Thinning strategies, namely shade or photosynthetic inhibitors, rely on the reduction of carbon supply to the fruit below the demand, causing fruit abscission. In order to clarify the subject, seven field trials were carried out in Lleida, Girona, and Sint-Truiden (2017 + 2018), using orchards of 'Golden' and 'Gala' apple trees. At the stage of 9–14-mm fruit diameter, four treatments were implemented: (A) CTR-control, trees under natural environmental conditions; (B) SN-shaded trees, trees above which shading nets reducing 50% of irradiance were installed 24 h after metamitron application date—without application of metamitron—and removed after five days; (C) MET-trees sprayed with 247.5 ppm of metamitron; (D) MET + SN-trees submitted to the combined exposure to metamitron application and shading nets. Low radiation significantly increased metamitron absorption (36–53% in the three locations in 2018) and reduced its degradation. Net photosynthesis and stomatal conductance were strongly reduced in all treatments, with minimum values 2 days after spraying (DAS) and incomplete recovery 10 DAS in MET + SN. All treatments resulted in leaf sucrose and sorbitol decreases, leading to a negative carbon balance. SN and MET + SN promoted the highest thinning efficacy, increasing fruit weight and size, with MET + SN causing over-thinning in some trials. Leaf antioxidant enzymes showed moderate changes in activity increases under MET or MET + SN, accompanied by a rise of glutathione content and a reduction in ascorbate, however without lipid peroxidation. This work shows that environmental conditions, such as cloudy days, must be carefully considered upon metamitron application, since the low irradiance enhances metamitron efficacy and may cause over-thinning.
