Organs obtained from brain dead donors can have suboptimal outcomes. Activation of the innate immune system and translocation of intestinal bacteria could be causative. Thirty two pigs were assigned to control, brain death (BD), BD + luminal intestinal polyethylene glycol (PEG), and BD + luminal intestinal University of Wisconsin solution (UW) groups. Animals were observed for 360 min after BD before organ retrieval. 2,000 mL luminal intestinal preservation solution was instilled into the duodenum at the start of organ procurement. LPS binding protein increased following BD in all groups except BD + PEG, which at 480 min was significantly lower compared with all other groups. Neuromyelitis optica spectrum disorder (NMOSD) is associated with pathological aquaporin-4 immunoglobulin G (AQP4-IgG), which cause brain damage. However, the impact of AQP4-IgG on retinal tissue remains unclear. Additionally, dysregulated complement anaphylatoxins C3a and C5a, known to modulate the endothelial barrier, are implicated in NMOSD. This study evaluates the susceptibility of human brain microvascular endothelial cells (HBMEC) and human retinal endothelial cells (HREC) to C3a- and C5a-mediated stress using real-time cell barrier analysis, immunocytochemical staining, qPCR and IgG transmigration assays. The findings reveal that C3a induced a concentration-dependent paracellular barrier breakdown and increased transcellular permeability in HBMEC, while HREC maintained barrier integrity under the same conditions. C5a attenuated C3a-induced disruption in HBMEC, indicating a protective role. Anaphylatoxin treatment elevated transcript levels of complement component C3 and increased C5 gene and protein expression in HREC, with no changes observed in HBMEC. In HBMEC, C5a treatment led to a transient upregulation of C3a receptor (C3AR) mRNA and an early decrease in C5a receptor 1 (C5AR1) protein detection. Conversely, HREC exhibited a late increase in C5aR1 protein levels. These results indicate that the retinal endothelial barrier is more stable under anaphylatoxin-induced stress compared to the brain, potentially offering better protection against paracellular AQP4-IgG transport. Tricalcium aluminate (C3A) and tricalcium silicate (C3S) with gypsums play an important role in the hydration of oil well cement. This study aims to assess the influence of C3S and C3A with various gypsums on the hydration of oil well cement at 30, 50, and 80 °C through the analyses of hydration heat evolution by isothermal calorimetry analysis, setting and thickening times, strength, and hydration products by X-ray diffraction. The result reveals that C3A hydrates, in the presence of gypsums, rapidly at 80 °C to produce the C3AH6 phase leading to higher heat evolution. The conversion of AFt to AFm is observed in oil well cement paste at 50 °C. An increase in C3A leads to an increase in compressive strength and a decrease in permeability at 80 °C. An increase of C3S leads to the reduction of cumulative heat evolution and early compressive strength before 28 d. Gypsums shorten cement setting and thickening times opposite to the commonly known retardation effect due to the lower content of C3A and its acceleration on C3S hydration. Dihydrate and hemihydrate gypsums show a greater promotion effect on strength at 30 and 50 °C, while anhydrous gypsum presents a greater enhancing effect at 80 °C. In this study, the performances of methylallyl ether (HPEG) type polycarboxylate ether-based water reducing admixtures (PCE) partially substituted with phosphate and sulfonate anionic groups were investigated. The effects of synthesized PCE's on the fresh, rheological, and some hardened properties of mixtures prepared with cements having different C3A contents were examined. Increasing the phosphate and sulfonate substitution ratio of PCE up to a certain value improved the rheological properties of the mixtures. Best flow retention and rheological properties were obtained with 9% phosphate (P9) and 7% sulfonate (S7) PCEs. Due to the high PCE dosage used in the Marsh-funnel and mini slump experiments, the nonadsorbed PCE in the pore solution blocked the adsorbed polymers on the cement particles. Also, the hydrodynamic radius of the polymer had a significant effect on the adsorption and dispersion performance of PCE. Morover, with the increase of cement C3A content, the fresh properties of the mixtures were adversely affected. In the quest to address the critical shortage of donor organs for transplantation, xenotransplantation stands out as a promising solution, offering a more abundant supply of donor organs. Yet, its widespread clinical adoption remains hindered by significant challenges, chief among them being immunological rejection. Central to this issue is the role of the complement system, an essential component of innate immunity that frequently triggers acute and chronic rejection through hyperacute immune responses. Such responses can rapidly lead to transplant embolism, compromising the function of the transplanted organ and ultimately causing graft failure. This review delves into three key areas of xenotransplantation research. It begins by examining the mechanisms through which xenotransplantation activates both the classical and alternative complement pathways. It then assesses the current landscape of xenotransplantation from donor pigs, with a particular emphasis on the innovative strides made in genetically engineering pigs to evade complement system activation. These modifications are critical in mitigating the discordance between pig endogenous retroviruses and human immune molecules. Additionally, the review discusses pharmacological interventions designed to support transplantation. By exploring the intricate relationship between the complement system and xenotransplantation, this retrospective analysis not only underscores the scientific and clinical importance of this field but also sheds light on the potential pathways to overcoming one of the major barriers to the success of xenografts. Defective a-galactosidase A (AGAL/GLA) due to missense or nonsense mutations in the GLA gene results in accumulation of the glycosphingolipids globotriaosylceramide (Gb3) and its deacylated derivate globotriaosylsphingosine (lyso-Gb3) in cells and body fluids. The aberrant glycosphingolipid metabolism leads to a progressive lysosomal storage disorder, i. e. Fabry disease (FD), characterized by chronic inflammation leading to multiorgan damage. Enzyme replacement therapy (ERT) with agalsidase-alfa or -beta is one of the main treatment options facilitating cellular Gb3 clearance. Proteome studies have shown changes in complement proteins during ERT. However, the direct activation of the complement system during FD has not been explored. Here, we demonstrate strong activation of the complement system in 17 classical male FD patients with either missense or nonsense mutations before and after ERT as evidenced by high C3a and C5a serumlevels. In contrast to the strong reduction of lyso-Gb3 under ERT, C3a and C5a markedly increased in FD patients with nonsense mutations, most of whom developed anti-drug antibodies (ADA), whereas FD patients with missense mutations, which were ADA-negative, showed heterogenous C3a and C5a serum levels under treatment. In addition to the complement activation, we found increased IL-6, IL-10 and TGF-ß1 serum levels in FD patients. This increase was most prominent in patients with missense mutations under ERT, most of whom developedmild nephropathy with decreased estimated glomerular filtration rate. Together, our findings demonstrate strong complement activation in FD independent of ERT therapy, especially in males with nonsense mutations and the development of ADAs. In addition, our data suggest kidney cell-associated production of cytokines, which have a strong potential to drive renal damage. This study investigated, the effect of polycarboxylate-based water-reducing admixture (PCE) molecule chain length modification on the cementadmixture compatibility. Seven PCEs with varying chain lengths were synthesized for this aim. Paste and mortar mixtures were prepared by combining a high C3A (9.1%) CEM I-42.5 R-type cement with PCEs. The Marsh-funnel flow time, mini-flow, dynamic yield shear stress, final viscosity and setting times of the paste mixtures were analyzed. The PCE requirement for target flow, the time-dependent flow performance, and compressive strength of mortar mixtures were studied. In this way, effects of the change in only the main chain length, only the side chain length, and both main and side chain lengths of PCE molecule on the Portland cement-admixture compatibility were comparatively researched. Consequently, the PCE with medium main and side chain lengths exhibited the best performance and was the most compatible with high C3A cement in terms of the examined properties. The effect of PCE chain length change on cement-admixture compatibility was investigated. PCE with medium main and side chain lengths was the most compatible with high C3A cement. BackgroundSilica nanoparticles (SiNPs) enter the human body through respiratory tract, digestive tract, and skin, causing body damage. Lung is one of the main damaged organs. ObjectiveTo observe the expressions of complement activated fragment C3a and its receptor C3aR in the lungs of mice exposed to SiNPs through respiratory tract, and to explore the involvement of C3a/C3aR in lung injury induced by SiNPs exposure. MethodsThe ultrastructure of SiNPs (particle size 5-20 nm) was determined under a transmission electron microscope, and the hydrodynamic diameter and surface Zeta potential of SiNPs were determined using a nanoparticle size analyzer. A total of 88 SPF C57BL/6J mice were randomly divided into five groups: a blank control group without any treatment (14 mice), a vehicle control group treated with 50 μL stroke-physiological saline solution by intratracheal instillation (14 mice), and three SiNPs exposure groups (low-dose group, medium-dose group, and high-dose group with 20 mice in each group, who were given 50 μL SiNPs suspension of 7, 21, and 35 mg·kg−1 respectively and exposed once every 3 days for 5 times). The mice were anesthetized on day 1 (1-day model group) and day 15 (15-day model group) after exposure, then sacrificed after extraction of bronchoalveolar lavage fluid (BALF), and lung tissues were retained. The morphological changes of lung tissues were observed by HE staining, the expression level of C3a in BALF was detected by enzyme-linked immunosorbent assay, the deposition of C3a and C3aR in lung tissues were observed by immunohistochemistry, the protein expression level of C3aR was determined by Western blotting, and the localization and semi-quantitative detection of C3a and C3aR in lung tissues was observed by immunofluorescence. ResultsSiNPs agglomerated in stroke-physiological saline solution. The average hydrodynamic diameter was (185.60±7.39) nm and the absolute value of Zeta potential was (43.33±0.76) mV. The condition of mice in the 1-day model group and the 15-day model group was good, while 2 mice died in the medium-dose group of the 1-day model group due to misoperation. The autopsy results of the two mice showed congestion of the lung tissue, emphysema, and no imperfection of trachea integrity. No death was observed in other dose groups. The HE staining results showed pathological damage to the mouse lung, including alveolar wall thickening and inflammatory cell infiltration after SiNPs exposure. The pathological damage became more serious with the increase of dose. Regarding pathological changes, the 15-day model group was slightly relieved compared with the 1-day model group, but there were still pathological changes. The dysregulation of complement system activation usually results in acute or chronic inflammation and can contribute to the development of various diseases. Although the activation of complement pathways is essential for innate defense, exacerbated activity of this system may be harmful to the host. Thus, drugs with the potential to inhibit the activation of the complement system may be important tools in therapy for diseases associated with complement system activation. The synthetic peptides Cp40 and PMX205 can be highlighted in this regard, given that they selectively inhibit the C3 and block the C5a receptor (C5aR1), respectively. The zebrafish (Danio rerio) is a robust model for studying the complement system. The aim of the present study was to use in silico computational modeling to investigate the hypothesis that these complement system inhibitor peptides interact with their target molecules in zebrafish, for subsequent in vivo validation. For this, we analyzed molecular docking interactions between peptides and target molecules. Our study demonstrated that Cp40 and the cyclic peptide PMX205 have positive interactions with their respective zebrafish targets, thus suggesting that zebrafish can be used as an animal model for therapeutic studies on these inhibitors. Abstract Background Dysregulated complement system is linked to pathophysiology of major depressive disorder (MDD). Childhood trauma has been associated with an increased incidence of adult depression via a putative mechanism of immune activation. This study aimed to measure and compare peripheral levels of complement C3, C3a, C1q and C-reactive protein (CRP) in MDD patients and healthy controls and explore the relationship between these molecule levels and childhood trauma history in the participants. Methods The participants were 49 medication-free MDD patients and 45 healthy controls. All participants were asked to finish the Childhood Trauma Questionnaire, followed by blood sampling for measurement of plasma complement C3, C3a, C1q and CRP by means of enzyme-linked immunosorbent assay. Results Peripheral plasma concentration of C3 and C3a in medication-free MDD group was significantly higher than that in the healthy controls; whereas the concentration of plasma C1q and CRP in depressed patients was comparable to that in healthy controls. All these inflammatory factors were not associated to childhood trauma experience in patients with MDD. Conclusion Our data suggest that complement C3 and C3a may be implicated in the pathophysiology of MDD, although traumatic childhood experiences were not associated with the circulating levels of complement C3, C3a, C1q and CRP.

Aristolochic acid I (AAI) can cause nephrotoxicity and is characterized by interstitial fibrosis. The C3a/C3aR axis of macrophages and matrix metalloproteinase-9 (MMP-9) play important roles in fibrosis, but whether they are involved in AAI-induced renal interstitial fibrosis and are related remains to be elucidated. In this study, we investigated whether C3a/C3aR axis of macrophages promotes renal interstitial fibrosis by regulating MMP-9 in aristolochic acid nephropathy (AAN). Intraperitoneal injection of AAI for 28 days successfully induced AAN in C57bl/6 mice. The content of C3a in the kidney of AAN mice was increased, and there was a significant distribution of macrophages in the renal tubules. The same results were observed in the in vitro experiment. We also explored the role and mechanism of macrophages after AAI administration in the epithelial–mesenchymal transformation (EMT) of renal tubular epithelial cells (RTECs) and found that AAI could activate the C3a/C3aR axis of macrophages to upregulate p65 expression in macrophages. p65 upregulated MMP-9 expression in macrophages not only directly but also by promoting the secretion if interleukin-6 by macrophages and then activating STAT3 in RTECs. The upregulation of MMP-9 expression could promote the EMT of RTECs. Taken together, our study demonstrated that the AAI-activated the C3a/C3aR axis of macrophages, which induced MMP-9 production, was one of the causes of renal interstitial fibrosis. Therefore, targeting the C3a/C3aR axis of macrophages is an effective therapeutic strategy for the prevention and treatment of renal interstitial fibrosis in AAN. Here, we focused on the relationship between AA I-induced migration, invasion and epithelial-mesenchymal transition in HCC cells, as well as the possible role of the C3a/C3AR axis in these effects. HCC cells were exposed to different concentrations of AA I for 24 h. Cell migration and invasion abilities were evaluated with wound healing assays and Transwell invasion assays. The protein and mRNA expression levels were detected by western blot, immunofluorescence and quantitative real-time polymerase chain reaction (qRT-PCR) assays. Furthermore, the level of complement component C3a in the cell supernatant was determined by enzyme-linked immunosorbent assay. C3aRA, a C3a receptor antagonist, was used to block the C3a-C3aR axis. The results showed that aristolochic acid I promoted HCC cell invasion and migration. AAI exposure also induced EMT in HCC cells through E-cadherin downregulation and Snail, N-cadherin, and vimentin upregulation. AAI exposure increased the levels of secreted C3a and the expression of C3aR protein and mRNA in HCC cells. We further found that AA I-induced C3a/C3AR activation was involved in these effects. AA I-induced epithelial-to-mesenchymal transition (EMT), cell migration, and invasion were decreased by C3aR inhibition. This paper presents a study related to the biological degradation of a tricalcium aluminate (C3A) phase treated with reactive media from the agricultural industry. During one month of setting and hardening, synthetic C3A was subjected to corrosion in corn silage, pig slurry and chicken manure. The hardening process of the C3A phase in water was used as a reference sample. The phase composition and microstructure of the hydrating tricalcium aluminate slurries were characterised by X-ray diffraction (XRD), thermal analysis (DTA/TG/DTG/EGA), scanning microscopy (SEM, EDS) and infrared spectroscopy (FT-IR). In the samples studied, it was observed that the qualitative and quantitative phase composition of the synthetic tricalcium aluminate preparations changed depending on the corrosion exposure conditions. The main crystalline phases formed by the hydration of the examined samples in water as well as in corrosive media were the catoite (Ca3Al2(OH)12) and hydrocalumite (Ca2Al(OH)7·3H2O) phases. Detailed analysis showed the occurrence of secondary crystallisation in hydrating samples and the phases were mainly calcium carbonates (CaCO3) with different crystallite sizes. In the phase composition of the C3A pastes, varying amounts of aluminium hydroxides (Al(OH)3) were also present. The crystalline phases formed as a result of secondary crystallisation represented biological corrosion products, probably resulting from the reaction of hydrates with secondary products resulting from the metabolic processes of anaerobic bacterial respiration (from living matter) associated with the presence of bacteria in the reaction medium. Evidence suggests a role of the immune system in the pathogenesis of a number of mental conditions, including schizophrenia (SCH). In terms of physiology, aside from its crucial protective function, the complement cascade (CC) is a critical element of the regeneration processes, including neurogenesis. Few studies have attempted to define the function of the CC components in SCH. To shed more light on this topic, we compared the levels of complement activation products (CAP) (C3a, C5a and C5b-9) in the peripheral blood of 62 patients with chronic SCH and disease duration of ≥ 10 years with 25 healthy controls matched for age, sex, BMI and smoking status. Concentrations of all the investigated CAP were elevated in SCH patients. However, after controlling for potential confounding factors, significant correlations were observed between SCH and C3a (M = 724.98 ng/mL) and C5a (M = 6.06 ng/mL) levels. In addition, multivariate logistic regression showed that C3a and C5b-9 were significant predictors of SCH. There were no significant correlations between any CAP and SCH symptom severity or general psychopathology in SCH patients. However, two significant links emerged between C3a and C5b-9 and global functioning. Increased levels of both complement activation products in the patient group as compared to healthy controls raise questions concerning the role of the CC in the etiology of SCH and further demonstrate dysregulation of the immune system in SCH patients. Scanning transmission X-ray microscopy (STXM) and calcium L2,3-edge near-edge X-ray absorption fine structure (Nexafs) spectra were used to investigate the morphology and chemical information of cement-based materials. The interaction between calcium chloride (CaCl2) and monosulfoaluminate (Ms) was investigated and the intermediate phases were identified. With an increase in the concentration of the calcium chloride solution, calcium Ms was converted into ettringite, Kuzel's salt (Ks) and Friedel's salt (Fs) to varying degrees, while SO42− ions in the ettringite and Ks were continuously replaced by Cl− ions in the solution, eventually forming Fs and gypsum. Based on the calcium L2,3-edge Nexafs absorption spectra, ettringite, Ms, Ks and Ms were greatly distinguished by the number and shape of the leading peaks in the spectra due to the difference in SO42− content among the interlayer structures of Ms, Ks and Fs. The present work is based on the references for the STXM study of the calcium (sulfo)aluminates, for demonstrating a more complete understanding of the phase evolution under chloride environments. Abstract Background Medulloblastoma (MB) is the most common malignant brain tumor in children. Approximately one-third of MB patients remain incurable. Understanding the molecular mechanism of MB tumorigenesis is, therefore, critical for developing specific and effective treatment strategies. Our previous work demonstrated that astrocytes constitute the tumor microenvironment (TME) of MB and play an indispensable role in MB progression. However, the underlying mechanisms by which astrocytes are regulated and activated to promote MB remain elusive. Methods By taking advantage of Math1-Cre/Ptch1 loxp/loxp mice, which spontaneously develop MB, primary MB cells and astrocytes were isolated and then subjected to administration and coculture in vitro. Immunohistochemistry was utilized to determine the presence of C3a in MB sections. MB cell proliferation was evaluated by immunofluorescent staining. GFAP and cytokine expression levels in C3a-stimulated astrocytes were assessed by immunofluorescent staining, western blotting, q-PCR and ELISA. C3a receptor and TNF-α receptor expression was determined by PCR and immunofluorescent staining. p38 MAPK pathway activation was detected by western blotting. Transplanted MB mice were treated with a C3a receptor antagonist or TNF-α receptor antagonist to investigate their role in MB progression in vivo. Results We found that complement C3a, a fragment released from intact complement C3 following complement activation, was enriched in both human and murine MB tumor tissue, and its receptor was highly expressed on tumor-associated astrocytes (TAAs). We demonstrated that C3a activated astrocytes and promoted MB cell proliferation via the p38 MAPK pathway. Moreover, we discovered that C3a upregulated the production of proinflammatory cytokines, such as IL-6 and TNF-α in astrocytes. Application of the conditioned medium of C3a-stimulated astrocytes promoted MB cell proliferation, which was abolished by preincubation with a TNF-α receptor antagonist, indicating a TNF-α-dependent event. Indeed, we further demonstrated that administration of a selective C3a receptor or TNF-α receptor antagonist to mice subcutaneously transplanted with MB suppressed tumor progression in vivo. Conclusions C3a was released during MB development. C3a triggered astrocyte activation and TNF-α production via the p38 pathway, which promoted MB cell proliferation. Our findings revealed the novel role of C3a-mediated TNF-α production by astrocytes in MB progression. These findings imply that targeting C3a and TNF-α may represent a potential novel therapeutic approach for human MB.

Background: Renin-dependent hypertension with tubulointerstitial injury remains a problem with high prevalence in the clinic. However, whether and how renin participates in tubulointerstitial injury remains incompletely understood. New evidence suggests that renin cleaves C3 into C3a and C3b. In the present study, we aimed to explore the role of renin-mediated C3a/C3a receptor (C3aR) signaling in renin-dependent hypertension-induced kidney injury and illustrate the detailed mechanisms. Methods: C3a concentration changes in serum from healthy volunteers incubated with recombinant renin were detected by ELISA. C3aR expression in human tubular epithelial cells was evaluated in renal biopsy sections from malignant arteriolonephrosclerosis and benign arteriolonephrosclerosis patients. C3aR changes in human kidney 2 (HK2) cells were detected after the cells were treated with human serum, renin and aliskiren. The C3a analogue and C3aR antagonist SB290157 were used to stimulate HK2 cells to explore the downstream signaling of C3a/C3aR activation. For in vivo studies, two-kidney, one-clipped (2K1C) hypertensive rat model was established to simulate renin-dependent hypertension conditions. C3a and C3aR expression was detected in the clipped kidneys. SB290157 was injected intraperitoneally to block C3a/C3aR signaling in 2K1C rats. Results: The results showed that renin cleaved C3 into C3a and activated C3a/C3aR signaling in tubular epithelial cells (TECs) from both humans and rats. In vitro results demonstrated that C3a/C3aR activation impaired peroxisome proliferator-activated receptor alpha (PPARα)/carnitine palmitoyltransterase-1alpha (CPT-1α)-mediated mitochondrial fatty acid oxidation (Mito FAO) in HK2 cells and induced HK2 cell transition to a profibrotic phenotype, which was inhibited by treatment with the C3aR antagonist SB290157. In vivo results showed that renin mRNA levels, C3a concentrations, C3aR levels and tubulointerstitial fibrosis increased concurrently in the clipped kidney cortex of 2K1C rats. Treatment with the C3aR antagonist SB290157 significantly mitigated the effect of renin induction of C3aR expression and alleviated renin-dependent hypertension-induced tubulointerstitial fibrosis by improving PPARα/CPT-1α-mediated Mito FAO in TECs, as well as inhibiting tubular profibrotic phenotype transition. Conclusions: Our results prove that renin activates C3a/C3aR signaling to promote renal tubulointerstitial fibrosis by impairing PPARα/CPT-1α-mediated tubular Mito FAO. SB290157 confers a potential therapeutic approach for renin-dependent hypertension-induced kidney injury.

Acute lung injury (ALI) occurs in patients with severe sepsis and has a mortality rate of 40%–60%. Severe sepsis promotes the release of histones from dying cells, which can induce platelet aggregation, activate coagulation and cause endothelial cell (EC) death. We previously reported that the expression of membrane complement receptor type 1‐related gene Y (Crry)/p65, which plays a principal role in defence against abnormal activation of complement in the blood, is reduced in response to peritoneal mesothelial cell injury, and we hence hypothesized that a similar mechanism occurs in pulmonary ECs. In this study, we examined the role of Crry/p65 in histone‐mediated ALI using an experimental animal model. In ALI model mice, exposure to extracellular histones induces lung injury and results in a decrease in Crry/p65 expression. The levels of lactic acid dehydrogenase (LDH), a marker of cell damage, were significantly increased in the serum of ALI model compared with vehicle mice. The significant inverse correlation between the expression of Crry/p65 and LDH levels in plasma revealed an association between Crry/p65 expression and cell damage. The levels of complement component 3a (C3a) were also significantly increased in the serum of the ALI model compared with vehicle mice. Notably, a C3a receptor antagonist ameliorated lung injury induced by histones. We hypothesize that extracellular histones induce complement activation via down‐regulation of Crry/p65 and that C3a might serve as a therapeutic target for the treatment of ALI.

Background: Dysregulated complement system is linked to pathophysiology of major depressive disorder (MDD). Childhood trauma has been associated with an increased incidence of adult depression via a putative mechanism of immune activation. This study aimed to measure and compare peripheral levels of complement C3, C3a, C1q and C-reactive protein (CRP) in MDD patients and healthy controls and explore the relationship between these molecule levels and childhood trauma history in the participants. Methods: The participants were 49 medication-free MDD patients and 45 healthy controls. All participants were asked to finish the Childhood Trauma Questionnaire, followed by blood sampling for measurement of plasma complement C3, C3a, C1q and CRP by means of enzyme-linked immunosorbent assay. Results: Peripheral plasma concentration of C3 and C3a in medication-free MDD group was significantly higher than that in the healthy controls; whereas the concentration of plasma C1q and CRP in depressed patients was comparable to that in healthy controls. All these inflammatory factors were not associated to childhood trauma experience in patients with MDD. Conclusion: Our data suggest that complement C3 and C3a may be implicated in the pathophysiology of MDD, although traumatic childhood experiences were not associated with the circulating levels of complement C3, C3a, C1q and CRP. Les phénomènes d'hypersensibilité aux vaccins à ARNm dirigés contre le SARS-CoV-2 sont très rares. Ils peuvent être liés à un mécanisme IgE-dépendant impliquant les PEG contenus dans les vaccins sous forme de liposomes. Une activation directe de la voie classique du complément (CARPA) a été également fortement suspectée. En complément des tests cutanés, des biomarqueurs ont été proposés, en particulier la recherche d'anticorps anti-PEG, le dosage des anaphylatoxines C5a et C3a ou du complexe soluble C5b-9. Les anticorps anti-PEG sont dosables par quelques méthodes non standardisées mais leur présence dans ces réactions n'a pas été confirmée. Il en est de même pour les protéines du complément. L'histamine et la tryptase ont rarement pu être dosées au moment de la réaction et leur augmentation est inconstante selon les études. Une tryptase basale un peu élevée chez certains patients suggère qu'une hyper-alpha-tryptasémie pourrait être impliquée. Le test d'activation des basophiles s'est imposé comme un examen pouvant être utile mais les résultats sont encore difficiles à interpréter en raison de la variabilité des allergènes utilisés : PEG, PEG sous forme de liposomes ou vaccin lui-même. L'absence de positivité des tests cutanés alors que les basophiles sont capables de s'activer en présence du même allergène ex vivo chez certains patients est en faveur, dans ce cas, d'un phénomène non IgE-dépendant. En conclusion, dans l'exploration des réactions immédiates aux vaccins à ARNm contre le SARS-CoV-2, l'importance de la place de marqueurs biologiques nécessite des études complémentaires afin de mieux identifier les acteurs et les mécanismes impliqués. Hypersensitivity to mRNA vaccines directed against SARS-CoV-2 are rare. They may be related to an IgE-dependent mechanism involving PEG contained in vaccines in the form of liposomes. Direct activation of the classical complement pathway (CARPA) has also been strongly suspected. In addition to skin tests, biomarkers have been proposed: anti-PEG antibodies, determination of anaphylatoxins C5a and C3a, or soluble complex C5b-9. Anti-PEG antibodies can be measured by non-standardized in house methods; their presence in post-vaccination reactions against SARS-CoV2 has not been confirmed by all the studies as well as for complement proteins. Mast cell mediators (histamine and tryptase) could rarely be assayed at the time of reaction and their increase is inconstant depending on studies. A slightly elevated baseline tryptase level in some patients suggests that hyper-alphatryptasemia might be involved. A basophil activation test (BAT) can be performed but the results are still preliminary depending on the allergens used: PEG, PEG in the form of liposomes or vaccine itself. The lack of positivity of skin tests in some patients even though basophils are able to be activated in the presence of the same allergen confirms the hypothesis in this case of a possible non-IgE-dependent phenomenon. In conclusion, concerning the exploration of immediate reactions to mRNA vaccines directed against SARS-CoV-2, the place of biological markers requires additional studies in order to better identify the actors and mechanisms involved. The objective of this study was to explore the influence of an alkali liquid accelerator based on sodium aluminate (ASa) on the early hydration behaviour of cement paste, tricalcium aluminate (C3A) and tricalcium silicate (C3S) and identify its accelerating mechanism. The setting times and mechanical properties of cement paste and mortar mixed with different amounts of ASa were determined. Isothermal calorimetry showed that the addition of ASa increased the first exothermic peak of cement hydration and advanced the second exothermic peak. The morphology of hydration products was analysed by scanning electron microscopy (SEM) and a model for the early hydration of cement paste was established. X-ray diffraction and SEM were also used to analyse the phase composition and morphology changes of C3A and C3S with or without ASa and early hydration models were established. Analysis of the experimental results revealed that the sodium hydroxide produced by ASa hydrolysis had the function of consuming gypsum, breaking calcium silicate hydrate gel film and increasing the calcium hydroxide concentration in the cement paste. The hydration of C3A-gypsum systems was studied in the presence of various types of sulfates such as gypsum, hemihydrate and Na2SO4 in the first hour. The BET method combined with a DSC analysis enabled us to quantitatively characterize the amount of precipitated ettringite and its specific surface area along the hydration. It was found that sulfates not only affected the formation rate of ettringite, but also had a significant impact on the morphology of ettringite. For all the C3A-gypsum systems, a large part of the ettringite precipitated within the first 20 min and the specific surface area of the hydrated sample strongly increased within the first 5 min, whereas the specific surface area of ettringite gradually decreased along the C3A hydration reaction. Incorporating a small amount of Na2SO4 in the C3A-gypsum system could greatly promote the formation rate of ettringite in the first 20 min, and significantly decrease the specific surface area of ettringite. As hemihydrate was added to the C3A-gypsum system, two processes of ettringite precipitation and gypsum precipitation occurred. The nucleation and growth process of ettringite and gypsum resulted in the complex changes in the specific surface area of the hydrated sample, which first increased at the very beginning, then decreased and, finally, increased. The role of the complement system in pain syndromes has garnered attention on the back of preclinical and clinical evidence supporting its potential as a target for new analgesic pharmacotherapies. Of the components that make up the complement system, component 5a (C5a) and component 3a (C3a) are most strongly and consistently associated with pain. Receptors for C5a are widely found in immune resident cells (microglia, astrocytes, sensory neuron-associated macrophages (sNAMs)) in the central nervous system (CNS) as well as hematogenous immune cells (mast cells, macrophages, T-lymphocytes, etc.). When active, as is often observed in chronic pain conditions, these cells produce various inflammatory mediators including pro-inflammatory cytokines. These events can trigger nervous tissue inflammation (neuroinflammation) which coexists with and potentially maintains peripheral and central sensitization. C5a has a likely critical role in initiating this process highlighting its potential as a promising non-opioid target for treating pain. This review summarises the most up-to-date research on the role of the complement system in pain with emphasis on the C5 pathway in peripheral tissue, dorsal root ganglia (DRG) and the CNS, and explores advances in complement-targeted drug development and sex differences. A perspective on the optimal application of different C5a inhibitors for different types (e.g., neuropathic, post-surgical and chemotherapy-induced pain, osteoarthritis pain) and stages (e.g., acute, subacute, chronic) of pain is also provided to help guide future clinical trials. PERSPECTIVE: This review highlights the role and mechanisms of complement components and their receptors in physiological and pathological pain. The potential of complement-targeted therapeutics for the treatment of chronic pain is also explored with a focus on C5a inhibitors to help guide future clinical trials., (Copyright © 2024. This work examined the effects of seawater (SW) on the hydration of tricalcium aluminate (C3A) in C3A–gypsum and C3A–gypsum–Ca(OH)2 systems through the characterization of hydration heat release, the evolution of aqueous phase composition and hydration products with the hydration time. It was found that SW increased the dissolution driving force of C3A and solubility of gypsum, which accelerated the early hydration of C3A and the formation of ettringite (AFt), leading to a higher hydration degree of C3A at an early age compared with the deionized (DI) water–mixed pastes. After gypsum depletion to form AFt, and in the absence of Ca(OH)2, the formation of chloroaluminate hydrates was slower due to the insufficient Ca resulted in an accumulation of Al in solution. This would delay the subsequent transformation of AFt to monosulfate (SO4–AFm) and the formation of hydrogarnet (C3AH6), which would further reduce the hydration degree of the C3A at the later ages. However, in the presence of Ca(OH)2, the hydration degree of C3A–gypsum–Ca(OH)2 at later ages was increased, which was similar to that of the corresponding DI pastes. Breast cancer is a common women malignancy worldwide. Emerging evidence suggests that various complement molecules function as an immune suppressors in the tumor microenvironment (TME). Therefore this study aimed to investigate the role of the anaphylatoxins C3a and C5a in the propagation of breast cancer and their relationships with clinicopathological factors in Iraqi women suffering from breast tumors. ELISA technique was used to determine the serum anaphylatoxins C3a and C4a levels in 60 Iraqi women with breast tumors (30 with primary breast cancer and 30 with benign breast tumor) before surgery and treatment as 30 healthy controls. In addition, the clinicopathological factors of all the patients were collected. The result showed a significantly increased level of anaphylatoxins C3a and C5a (P<0.001) in primary breast cancer patients than in benign breast tumor and healthy control women. The area under the curve (AUC) of C3a and C5a for distinguishing patients with primary breast cancer and healthy control subjects was 0.878 (95% CI: 0.768 to 0.948) and 0.953 (95% CI: 0.865 to 0.991) respectively. C5a correlated with larger tumor size, lymph node metastasis, and histologic grade III, whereas C3a only with lymph node metastasis. In conclusion, preoperative levels of C5a exhibit high accuracy for primary breast cancer than C3a. C5a correlated with tumor burden more than C3a and suggested an enhancing effect of C5a on BC proliferation. Thus, suggesting prognostic value for C5a in primary BC. [ABSTRACT FROM AUTHOR]

The complement system (CS) is an ancient and highly conserved part of the innate immune system with important functions in immune defense. The multiple fragments bind to specific receptors on innate and adaptive immune cells, the activation of which translates the initial humoral innate immune response (IR) into cellular innate and adaptive immunity. Dysregulation of the CS has been associated with the development of several autoimmune disorders such as systemic lupus erythematosus (SLE), rheumatoid arthritis (RA), ANCA-associated vasculitis, and autoimmune bullous dermatoses (AIBDs), where complement drives the inflammatory response in the effector phase. The role of the CS in autoimmunity is complex. On the one hand, complement deficiencies were identified as risk factors to develop autoimmune disorders. On the other hand, activation of complement can drive autoimmune responses. The anaphylatoxins C3a and C5a are potent mediators and regulators of inflammation during the effector phase of autoimmunity through engagement of specific anaphylatoxin receptors, i.e., C3aR, C5aR1, and C5aR2 either on or in immune cells. In addition to their role in innate IRs, anaphylatoxins regulate humoral and cellular adaptive IRs including B-cell and T-cell activation, differentiation, and survival. They regulate B- and T-lymphocyte responses either directly or indirectly through the activation of anaphylatoxin receptors via dendritic cells that modulate lymphocyte function. Here, we will briefly review our current understanding of the complex roles of anaphylatoxins in the regulation of immunologic tolerance and the early events driving autoimmunity and the implications of such regulation for therapeutic approaches that target the CS. Background: We previously reported higher plasma levels of complement fragments C3a and C5a in neovascular Age-related Macular Degeneration (nAMD) patients with macular fibrosis. This study aimed to understand whether complement activation contributes to the development of macular fibrosis and the underlying mechanisms involved. Methods: Complement activation was blocked using a C5 neutralizing antibody (BB5.1) in C57BL/6J mice after induction of subretinal fibrosis using the two-stage laser protocol. Fibrotic lesions were examined 10 days after the 2nd laser through fundus examination and immunohistochemistry. The expression of C5aR in fibrotic lesions and retinal pigment epithelial (RPE) cultures were examined by confocal microscopy. Primary murine RPE cells were treated with C3a or C5a (10–100 ng/mL) or TGF-β2 (10 ng/mL). Epithelial-to-mesenchymal transition (EMT) was assessed through various readouts. The expression of E-cadherin, vimentin, fibronectin, α-SMA, Slug, ERK/AKT and pSMAD2/3 were determined by Western blot and immunocytochemistry. Collagen contraction and wound-healing assays were used as functional readouts of EMT. The production of IL-6, TGF-β1, TGF-β2 and VEGF by RPE cells were determined by ELISA. PMX53 was used to block C5aR in RPE cultures and in vivo in mice with subretinal fibrosis. Results: Extensive C5b-9 deposition was detected at the site of subretinal fibrosis. BB5.1 treatment completely abrogated complement activation and significantly reduced subretinal fibrosis. C5aR was detected in RPE and infiltrating MHC-II+ cells in subretinal fibrosis. In vitro, RPE cells constitutively express C5/C5a and C5aR, and their expression was increased by TGF-β2 treatment. C5a but not C3a increased fibronectin, α-SMA, vimentin and Slug expression, and decreased E-cadherin expression in RPE cells. C5a treatment also increased the contractility and migration of RPE cells and enhanced the production of VEGF and TGF-β1/2. C5a treatment induced pSmad2/3 and pERK1/2 expression in RPE cells and this was blocked by PMX53. PMX53 treatment significantly reduced sodium fluorescein leakage in the subretinal fibrosis model, while collagen-I+ lesions only mildly reduced. Conclusions: Complement activation is critically involved in the development of subretinal fibrosis, partially through C5a–C5aR-mediated EMT in RPE cells. Calcium aluminate phases have a particular effect on the early heat release during setting initiation and have a substantial influence on the further workability of ordinary Portland cement. The nature of the calcium aluminate hydration products and its kinetics strongly depends on sulfate content and humidity. The effect of mineralisers on melt formation and viscosity is well described for calcium silicate systems, but information is still lacking for calcium aluminates. Therefore, the synergistic effect on the crystal structure and hydration mechanism of the tricalcium aluminate phase of the addition of mineralizers, i.e. Li2O, CuO, SO3 to the raw meal is here investigated. Co-doped calcium aluminate structures were formed during high-temperature treatment. Thermal analysis (TG–DTA and heating microscopy) was used to describe the ongoing high-temperature reaction. Resulting phase composition was dependent on the concentration of the mineralizer. While phase pure system was prepared with low mineralizer concentrations, with increasing mineralizer content the secondary phases were formed. Raman spectroscopy and XPS analysis were used to investigate the cation substitution and to help describe the cations bonding in co-doped calcium aluminate system. Prepared powders have been hydrated in a controlled manner at different temperatures (288, 298, 308 K). The resulting calorimetric data have been used to investigate the hydration kinetics and determine the rate constant of hydration reaction. First-order reaction (FOR) model was here applied for the activation energy and frequency factor calculations. The metastable and stable calcium aluminate hydrates were formed according to initial phase composition. In phase pure systems with low S content, the formation of stable and metastable hydrates was depended on the reaction temperature. Conversely, in systems with secondary phases and higher S content, the hydration mechanism resembled that which appears in calcium sulfoaluminates. [ABSTRACT FROM AUTHOR]

Background: Medulloblastoma (MB) is the most common malignant brain tumor in children. Approximately one-third of MB patients remain incurable. Understanding the molecular mechanism of MB tumorigenesis is, therefore, critical for developing specific and effective treatment strategies. Our previous work demonstrated that astrocytes constitute the tumor microenvironment (TME) of MB and play an indispensable role in MB progression. However, the underlying mechanisms by which astrocytes are regulated and activated to promote MB remain elusive.Methods: By taking advantage of Math1-Cre/Ptch1loxp/loxp mice, which spontaneously develop MB, primary MB cells and astrocytes were isolated and then subjected to administration and coculture in vitro. Immunohistochemistry was utilized to determine the presence of C3a in MB sections. MB cell proliferation was evaluated by immunofluorescent staining. GFAP and cytokine expression levels in C3a-stimulated astrocytes were assessed by immunofluorescent staining, western blotting, q-PCR and ELISA. C3a receptor and TNF-α receptor expression was determined by PCR and immunofluorescent staining. p38 MAPK pathway activation was detected by western blotting. Strict environmental norms and increasing awareness of the need to recycle solid wastes generated during ironmaking and steelmaking processes are the key driving force for the development of various new materials and manufacturing technologies. In this study, a calcium aluminate clinker was prepared from steel ladle slag by modifying its mineral composition. The slag paste, prepared by mixing slag with water, exhibited flash setting behaviour due to the presence of C12A7 and C3A phases. In contrast, the slag clinker, developed by sintering a mixture of a pre-determined quantity of slag and aluminium oxide at 1400°C for 2 h and 4 h, contained CA, CA2, gehlenite and 'Q' phases. Hydrated slag clinker contained stable C3AH6, AH3 and stratlingite with preferential growth of calcium aluminate hydrate prisms along the c-axis, which provided a well-defined raceme-like morphology with an interlinked structure. This improved the setting time and crushing strength of the clinkers after 6 h and 24 h of curing at room temperature. Additionally, the presence of the Q phase, with lamellar prismatic crystals, also helped to enhance strength. The developed clinker also exhibited superior crushing strength as compared with commercially available calcium aluminate cement of medium purity. The slag, used as a source of calcium oxide, could replace calcium carbonate completely and thus contribute to reductions in carbon dioxide emissions during the clinker-making process. Mast cells (MCs) are distributed in tissues throughout the body and are highly involved in many physiological and pathophysiological processes. The potential and involvement of different MC phenotypes are still not well understood. MCs are present in blood vessel walls, but their specific phenotypic features are unknown. We aimed at characterizing MCs from human saphenous veins for localization, mediator content, and receptor expression. This was done in MCs from both healthy and varicose human saphenous veins (hSV and vSV, respectively). For both vSV and hSV, we found that vein MCs are mainly present in the tunica adventitia (99% MCs in adventitia) and that the population consists of both MCT and MCTC phenotypes (vSV: 55% MCT, hSV: 64% MCT). The vein MCs contained high levels of histamine (vSV: 27 pg/MC, hSV: 55 pg/MC) and tryptase (vSV: 98 pg/MC, hSV: 111 pg/MC), indicating a strong potential for regulatory effects on blood vessels. The receptor expression of FcεRI, MRGPRX2, PTAFR, C3aR, and C5aR was found, even though the percentage of positive cells differed between vSV and hSV MCs. Purpose: To investigate the association of risk alleles in complement factor H (CFH) and age-related maculopathy susceptibility 2 (ARMS2) with complement activation products in the aqueous humor in eyes with neovascular age-related macular degeneration (nAMD) including polypoidal choroidal vasculopathy (PCV), retinal angiomatous proliferation (RAP), and pachychoroid neovasculopathy (PNV). Design: Prospective, comparative, observational study. Participants: Treatment-naïve patients with nAMD and cataract patients as controls. Methods: The study included 236 eyes of 236 patients with nAMD and 49 control eyes of 49 patients. Aqueous humor samples were collected from 67 eyes with drusen-associated nAMD, 72 eyes with PCV, 26 eyes with RAP, and 71 eyes with PNV before intravitreal anti-VEGF injection and cataract surgery in the 49 control eyes. Clinical samples were measured for complement component 3a (C3a), C4a, and C5a using a bead-based immunoassay. Genotyping of the ARMS2 A69S (rs10490924), CFH I62V (rs800292), and CFH Y402H (rs1061170) was performed using TaqMan genotyping. Main Outcome Measures: The levels of complement activation products (C3a, C4a, and C5a) in the aqueous humor in each genotype of ARMS2 and CFH. Results: The C3a level in the aqueous humor was significantly elevated (P = 0.006) in patients with nAMD and the ARMS2 A69S risk allele, whereas the levels of the complement activation products were not associated with CFH I62V and Y402H genotypes. Among the control eyes, no significant differences were seen in any complement activation products for all genetic polymorphisms. The levels of the complement activation products in the aqueous humor of eyes with the nAMD subtypes for each genetic polymorphism did not show significant differences. Conclusions: The C3a concentration in the aqueous humor was significantly higher in Japanese nAMD patients with the ARMS2 A69S risk allele, whereas it was not elevated in the patients with CFH I62V. Age-related maculopathy susceptibility 2 A69S polymorphism is strongly associated with local complement activation in nAMD patients.

The complement system is an important part of the immune system of teleost fish. Besides, teleost B cells possess both phagocytic activity and adaptive humoral immune function, unlike mammalian B1 cells with phagocytic activity and B2 cells specific to adaptive humoral immunity. However, the cross talk between complement system and phagocytic B cells in teleost fish still requires elucidation. Here, we show that, unlike tetrapods with a single C3 gene, nine C3 genes were identified from the grass carp (Ctenopharyngodon idella) genome, named C3.1 - C3.9. Expression analysis revealed that C3.1 is the dominant C3 molecule in grass carp, for its expression was significantly higher than that of the other C3 molecules both at the mRNA and protein levels. The C3a fragment of C3.1 (C3a.1) was determined after the conserved C3 convertase cleavage site. Structural analysis revealed that C3a.1 consists of four α-helixes, with the C-terminal region forming a long α-helix, which is the potential functional region. Interestingly, we found that the recombinant GST-C3a.1 protein and the C-terminal α-helix peptide of C3a.1 both could significantly enhance the phagocytic activity of IgM+ B cells. Further study revealed that the C3a receptor (C3aR) was highly expressed in grass carp IgM+ B cells, and the phagocytosis-stimulating activity of C3a.1 could be dramatically inhibited by the anti-C3aR antibodies, indicating that C3a.1 performed the stimulating function through C3aR on IgM+ B cells. Taken together, our study not only uncovered the novel phagocytosis-stimulating activity of C3a, but also increased our knowledge of the cross talk between complement system and phagocytic B cells in teleost fish. In all pathways of complement activation, the pivotal step is conversion of the component C3 to C3b and C3a, which is responsible to eliminate the pathogen and opsonization. In this study, we examined the immunological properties of C3 and its activated fragment C3a from Japanese flounder (Paralichthys olivaceus) (PoC3 and PoC3a), a teleost species with important economic value. PoC3 is composed of 1655 amino acid residues, contains the six domains and highly conserved GCGEQ sequence of the C3 family. We found that PoC3 expression occurred in nine different tissues and was upregulated by bacterial challenge. In serum, PoC3 was able to bind to a broad-spectrum of bacteria, and purified native PoC3 could directly kill specific pathogen. When PoC3 expression in Japanese flounder was knocked down by siRNA, serum complement activity was significantly decreased, and bacterial replication in fish tissues was significantly increased. Recombinant PoC3a (rPoC3a) exhibited apparent binding capacities to bacteria and Japanese flounder peripheral blood leukocytes (PBL) and induce chemotaxis of PBL. Japanese flounder administered rPoC3a exhibited enhanced resistance against bacterial infection. Taken together, these results indicate that PoC3 is likely a key factor of complement activation, and PoC3 and PoC3a are required for optimal defense against bacterial infection in teleost. Acute lung injury (ALI) occurs in patients with severe sepsis and has a mortality rate of 40%–60%. Severe sepsis promotes the release of histones from dying cells, which can induce platelet aggregation, activate coagulation and cause endothelial cell (EC) death. We previously reported that the expression of membrane complement receptor type 1‐related gene Y (Crry)/p65, which plays a principal role in defence against abnormal activation of complement in the blood, is reduced in response to peritoneal mesothelial cell injury, and we hence hypothesized that a similar mechanism occurs in pulmonary ECs. In this study, we examined the role of Crry/p65 in histone‐mediated ALI using an experimental animal model. In ALI model mice, exposure to extracellular histones induces lung injury and results in a decrease in Crry/p65 expression. The levels of lactic acid dehydrogenase (LDH), a marker of cell damage, were significantly increased in the serum of ALI model compared with vehicle mice. The significant inverse correlation between the expression of Crry/p65 and LDH levels in plasma revealed an association between Crry/p65 expression and cell damage. The levels of complement component 3a (C3a) were also significantly increased in the serum of the ALI model compared with vehicle mice. Notably, a C3a receptor antagonist ameliorated lung injury induced by histones. We hypothesize that extracellular histones induce complement activation via down‐regulation of Crry/p65 and that C3a might serve as a therapeutic target for the treatment of ALI. In this study, the effect of the PCE main chain length variation on the admixture demand to provide the desired slump value (27±2 cm) and the consistency retention performance of mortar mixtures having different C3A content was investigated. For this purpose, a total of 12 mortar mixtures were prepared by using CEM I 42.5R type cement having four different C3A content (2%, 3%, 6%, 9%) and using PCEs having three different main chain lengths (27000 g/mol, 55500 g/mol, 78000 g/mol). According to the results, irrespective of the PCE type, the PCE requirement increased by an increment of the C3A ratio. This behavior was more pronounced in the mixture containing the cement having the highest C3A ratio (9%). Compared to the mixture containing PCE having medium main chain length, the admixture demand to provide desired slump value of the mixture containing PCEs having short and long main chains was obtained at 3.9-4.5 and 4.7-6.5 times higher, respectively. The shortness of the PCE's main chain reduces the adsorption and electrostatic effect of the admixture. The poor performance of the PCE having a long main chain may be due to the bridging effect and the intertwining of the long polymers. PCE having a medium main chain showed almost %21 lower consistency retention performance than the others. This is due to the greater admixture requirement to achieve target flow value in the mixtures containing admixture with short and long main chains compared to admixtures with medium chains. Abstract Background Mounting evidence suggests that complement components promote tumor progression via modulating immune suppression, angiogenesis, or tumor cell proliferation. However, the role of C3a-C3aR signaling in regulating lung metastasis of breast cancer remains unknown. Methods We performed various ex-vivo and in-vivo assays. Genetic and pharmacological C3aR blockade models were applied to investigate the role of C3a-C3aR in metastasis of breast cancer. Results C3a-C3aR signaling in CAFs facilitates the metastasis of breast cancer. Mechanically, C3a-C3aR signaling augments pro-metastatic cytokine secretion and extracellular matrix components expression of CAFs via the activation of PI3K-AKT signaling. Genetic or pharmacological blockade of C3aR signaling effectively inhibited lung metastasis of breast cancer in mouse models. Conclusions C3a-C3aR signaling in CAFs facilitates the metastasis of breast cancer. Targeting C3aR signaling is a potential anti-metastasis strategy for breast cancer therapy.

The TLQP-21 neuropeptide has been implicated in functions as diverse as lipolysis, neurodegeneration and metabolism, thus suggesting an important role in several human diseases. Three binding targets have been proposed for TLQP-21: C3aR1, gC1qR and HSPA8. The aim of this review is to critically evaluate the molecular identity of the TLQP-21 receptor and the proposed multi-receptor mechanism of action. Several studies confirm a critical role for C3aR1 in TLQP-21 biological activity and a largely conserved mode of binding, receptor activation and signaling with C3a, its first-identified endogenous ligand. Conversely, data supporting a role of gC1qR and HSPA8 in TLQP-21 activity remain limited, with no signal transduction pathways being described. Overall, C3aR1 is the only receptor for which a necessary and sufficient role in TLQP-21 activity has been confirmed thus far. BACKGROUND: During pulp-dentin regeneration, other types of cells in dental pulp, such as dentin cells and stem cells, have been widely concerned and studied by scholars, while fibroblasts have been rarely studied. The study on dental pulp fibroblasts may provide a new source of cells for future pulp treatment. OBJECTIVE: To review the role of dental pulp fibroblasts in pulp-dentin regeneration. METHODS: The computer was applied to search articles on dental pulp fibroblasts and dental pulp dentine regeneration published from 2000 to 2020 in PubMed and CNKI databases. The English search terms were “pulp, fibroblast, dentin-pulp, regeneration, complement, C5a, C3a”. The Chinese search terms were “dental pulp, fibroblasts, dentin-pulp, complement, C3a, C5a”. Finally, 56 articles were summarized. RESULTS AND CONCLUSION: Fibroblasts are the most abundant cells in dental pulp. Fibroblasts play an important role in regulating the immune response and function of dental pulp under normal and pathological conditions. They enable complement proteins to produce bioactive fragments, such as C3a, C5a and membrane-attack complexes, and simultaneously, through the secretion of growth factors, fine-regulate the synthesis of restorative dentin, angiogenesis and the distribution of nerve growth, which play an important role in the pulp and dentin regeneration. Therefore, in addition to odontoblasts and pulp stem cells, dental pulp fibroblasts should also be considered as an important cell in the strategy of inducing dentin-pulp regeneration. Ischemic stroke is a major cause of disability. No efficient therapy is currently available, except for the removal of the occluding blood clot during the first hours after symptom onset. Loss of function after stroke is due to cell death in the infarcted tissue, cell dysfunction in the peri-infarct region, as well as dysfunction and neurodegeneration in remote brain areas. Plasticity responses in spared brain regions are a major contributor to functional recovery, while secondary neurodegeneration in remote regions is associated with depression and impedes the long-term outcome after stroke. Hypoxic-ischemic encephalopathy due to birth asphyxia is the leading cause of neurological disability resulting from birth complications. Despite major progress in neonatal care, approximately 50% of survivors develop complications such as mental retardation, cerebral palsy or epilepsy. The C3a receptor (C3aR) is expressed by many cell types including neurons and glia. While there is a body of evidence for its deleterious effects in the acute phase after ischemic injury to the adult brain, C3aR signaling contributes to better outcome in the post-acute and chronic phase after ischemic stroke in adults and in the ischemic immature brain. Here we discuss recent insights into the novel roles of C3aR signaling in the ischemic brain with focus on the therapeutic opportunities of modulating C3aR activity to improve the outcome after ischemic stroke and birth asphyxia. Different liquid accelerators including sodium aluminate liquid accelerator (SAa), fluoaluminate liquid accelerator (Fa) and aluminium sulfate liquid accelerator (ASa) were selected to investigate their influences on the carbonation properties and morphology of tricalcium aluminate (C3A) and tricalcium silicate (C3S) hydration products. After an accelerated carbonation test, the relative contents of ettringite (AFt), calcium hydroxide (CH) and calcium carbonate (CaCO3) in the hydration products of C3A and C3S samples were analysed by thermogravimetric analysis. The results indicated that the hydration products of C3A and C3S with liquid accelerators were easier to carbonate, and the promotion effect of SAa was the most obvious. The morphology of hydrated products of C3A and C3S was observed by scanning electron microscopy analysis. It was observed that different liquid accelerators had a different influence on the morphology of C3A and C3S hydration products. Among them, ASa had the greatest influence on the morphology of C3A and C3S hydration products, and experiments proved that ASa could promote the transformation of AFt into AFm and make the CH crystal structure become thicker and larger.
