In this study we screened twelve newly synthesised N-(substituted phenyl)-2-chloroacetamides for antimicrobial potential relying on quantitative structure-activity relationship (QSAR) analysis based on the available cheminformatics prediction models (Molinspiration, SwissADME, PreADMET, and PkcSM) and verified it through standard antimicrobial testing against Escherichia coli, Staphylococcus aureus, methicillin-resistant S. aureus (MRSA), and Candida albicans. Our compounds met all the screening criteria of Lipinski’s rule of five (Ro5) as well as Veber’s and Egan’s methods for predicting biological activity. In antimicrobial activity tests, all chloroacetamides were effective against Gram-positive S. aureus and MRSA, less effective against the Gram-negative E. coli, and moderately effective against the yeast C. albicans. Our study confirmed that the biological activity of chloroacetamides varied with the position of substituents bound to the phenyl ring, which explains why some molecules were more effective against Gram-negative than Gram-positive bacteria or C. albicans. Bearing the halogenated p-substituted phenyl ring, N-(4-chlorophenyl), N-(4-fluorophenyl), and N-(3-bromophenyl) chloroacetamides were among the most active thanks to high lipophilicity, which allows them to pass rapidly through the phospholipid bilayer of the cell membrane. They are the most promising compounds for further investigation, particularly against Gram-positive bacteria and pathogenic yeasts.

Supercritical CO2 extraction was applied on fenugreek (Trigonella foenum-graecum L., Fabaceae) seeds with the aim to define optimal process conditions to obtain the maximal extract yields content of steroidal sapogenins. Central composite rotatable design (CCRD) combined with response surface methodology (RSM) was applied to determine optimal process conditions defined by the influence and interaction of pressure, temperature and time of extraction through consumption of SC CO2. Optimization experiments revealed that the pressure of 24.73 MPa, the temperature of 38.°C and the consumption of SC CO2 of 19.24 g/gDM were the best process conditions enabling the maximal yield of extract and gain of the highest content of sapogenins. The optimal values of pressure and temperature defined SC CO2 density of 885.47 kg/m3, which provided the maximal yield of the extract with the highest content of steroidal sapogenins. The achieved yield of extract at these conditions was 0.073g/gDM, with 0.774 mg/gDM diosgenin, 0.477 mg/gDM protodioscin, 0.713 mg/gDM sarsapogenin and 0.205 mg/gDM oleanolic and ursolic acid with a significant quantity of 2.475 mg/gDM in the obtained extract. The main goal of this study was the assessment of the N, N-disubstituted-2-chloroacetamides' biological profile by chromatographic and in silico approach. Preliminary, their drug-like as well as lead-like properties and the bioactivity scores were analyzed. By applying reversed phase thin-layer chromatography as the simulator of compound's distribution across biological membranes, the assumed bioactivity descriptors of the studied chloroacetamides were obtained. The compliance between obtained chromatographic parameters (RM,0 m) and relevant software lipophilicity/pharmacokinetic/ecotoxicity predictors of studied derivatives were examined by linear regression and additionally supported by more detailed relations established by multivariate methods. Also, it was confirmed that the studied bioactivity properties of the given chloroacetamides are conditioned by the type of the present hydrocarbon substituents (number, arrangement and binding of C atoms). Extensive evidence indicates that adults with experience of childhood trauma and disorganised attachment are at a higher risk of suffering from depressive symptoms, while at the same time dissociation and mentalisation are very important risk and protective factors in this relation. Although mentalisation and dissociation are mentioned as potential mediators of this relationship, very few studies have analysed the mutual relations between these variables. Bearing in mind the importance of creating more specific and effective ways for dealing with depression, especially within vulnerable groups such as the traumatised, the aim of this research is to examine a serial mediation model in which disorganisation is modelled as effecting a decrease in mentalisation, in turn lead to dissociation, thereby resulting in depressive symptoms. Adult respondents (N = 153) with a history of interpersonal childhood trauma were selected from a larger geographical cluster sample from the general population of Serbia. The respondents filled in a battery of instruments, including The Childhood Traumatic Events Scale, Relationship Questionnaire – CV, Patient Health Questionnaire, Short Scale for the Assessment of Stress‐Related Dissociation Symptomatology and the Mentalisation Scale. Our findings suggest that disorganisation has an effect on depression by leading to a decrease in the ability to mentalise, which further triggers dissociation, resulting in depressive symptoms as a final consequence (estimated indirect effect = 0.014, 95 percent CI = 0.005 to 0.026). Additionally, the specific indirect effect involving dissociation as the sole mediator was also significant (estimated indirect effect = 0.058, 95 percent CI = 0.030 to 0.091). Targeting dissociative symptoms in the treatment in patients struggling with depression aftermath of traumatic experience could gain a great benefit for recognition and selection of more adequate treatment strategies. Duloxetine hydrochloride (DUL) is effective in treating depression, and was examined for electroanalytical purposes. The DUL standard was investigated by cyclic voltammetry (CV) and determined using differential pulse voltammetry (DPV) via its electro-oxidation at the Au electrode in 0.05 M NaHCO3. DPV showed a linear dependency of the anodic peak currents vs DUL standard concentrations in the range from 0.1 to 3.33 μg ml−1 with the values of the limit of detection (LOD) and the limit of quantification (LOQ): 0.133 and 0.667 μg ml−1, respectively. Using the constructed and validated calibration curve, the values of unknown DUL concentrations in both Taita® tablets and in human serum spiked with the standard were determined. The number of protons and electrons transferred was calculated and possible reaction mechanisms taking place on the surface of both electrodes were proposed. The Au electrode exhibited a better sensitivity and a wider range of current vs concentration linear dependency for DUL quantitative determination than the GC electrode. The study of DUL degradation showed that at the Au and GC electrodes, after 4.5 h of potential cycling, degradation occurs, giving formaldehyde as a product, which was confirmed by high performance liquid chromatography (HPLC). The aim behind this study was to first analyze the nature and the extent of childhood trauma and existing maladaptive outcomes (internalized and externalized problems, and dissociation) among adolescents in the foster system, and then analyze the possibility of using traumatic experiences to predict the aforementioned outcomes of childhood trauma. The sample consists of 121 respondents, children and youths in the care of child protective services, without adequate parental care, residing in temporary foster care families on the territory of Serbia, aged between 11 and 18. The respondents filled out the Childhood Trauma Questionnaire -- CTQ (Bernstein & Fink, 2003), the Dissociative experience scale for adolescents, A-DES (Armstrong et al., 1997), and the Child behavior checklist -- youth self-report (Achenbach & Rescorla, 2001). The results of the analyses have indicated that physical and emotional neglect are the most frequent forms of maltreatment in early childhood, with a relatively high prevalence of the other individual forms of trauma. Early childhood trauma statistically significantly predicted all the analyzed maladaptive outcomes, explaining approximately 20% of the variance of internalized and externalized problems, and dissociation. This study aimed to test the model set up by Obradović and Čudina- Obradović, according to which marital quality is determined by certain traits of marital quality from each of these groups of factors: the personality traits of the partners, the characteristics of the marriage, the marital processes and the marital environment. We operationalized this model, by testing the effect personality traits and the tendency towards alcohol consumption of marriage partners (as a personal characteristic of the marriage partner), the duration of the marriage (as the characteristic of the marriage itself), satisfaction with the division of household responsibilities (as one of the indicators of the development of the marital processes) and doubt regarding the marriage partner's fidelity (as the feature of the wider marital environment) have on the experienced marital quality. NEO-PI-R, The Dyadic Adjustment Scale and questionnaire designed for the aims of this study were used on a sample of 199 respondents from Serbia. Our model shows an excellent fit (X2 >.05; RMSEA<.05; GFI>.90; AGFI>.90; CFI>.90; NFI>.90). It can be concluded that Neuroticism, The Duration of marriage, Doubt regarding the partner's fidelity and Dissatisfaction with the division of household responsibilities have a negative effect on marital quality. The specific objectives of the presented study were related to the optimization of the production process of N-alkyl-, N,N-dialkyl-, and N-cycloalkyl-O-isobutyl thiocarbamate; trial industrial production of N-ethyl-O-isobutyl thiocarbamate; and the evaluation of flotation efficiency of N-ethyl-O-isobutyl thiocarbamate using a real ore sample. The optimization of thiocarbamate syntheses were performed by varying the molar ratio of isobutyl alcohol, carbon disulfide, potassium hydroxide, reaction time, and reaction temperature. In the first step, one-pot reaction took place to produce alkyl xanthate and was followed with chlorination to give alkyl chloroformate (O-alkyl carbonochloridothioate); finally, thiocarbamates were obtained by the reaction with corresponding amines. N-alkyl-O-ethyl thiocarbamate was synthesized as a comparative flotation agent. The structure of the synthesized compounds was confirmed by IR, 1H and 13C NMR, and MS instrumental methods, and the purity was determined by gas chromatographic method and elemental analysis. The optimized methods gave high-purity products in a significant yield that was also confirmed by semi-industrial production of N-ethyl-O-isobutyl thiocarbamate. The optimized thiocarbamate synthesis, without isolation of intermediates, is of great importance from the aspect of green technologies. Flotation efficiency test results, using real copper and zinc ores, showed the highest activity of N-ethyl-O-isobutyl thiocarbamate. The optimal one-pot thiocarbamate synthesis provides a simple procedure with a high conversion degree, and, thus, offers valuable technology applicable at the industrial scale. An optimal laboratory two-step procedure was developed for sodium/potassium O-alkyl carbonodithioate (sodium/potassium alkyl xanthates) production in the form of aqueous solutions. Sodium isopropyl xanthate (Na-iPrX), as the most effective salt in a real ore flotation process, was also produced at an industrial level and introduced as a collector for copper ores. In order to reduce toxicity of the flotation process and improve sustainability by minimising environmental impacts, collector efficiency and selectivity in the flotation process were studied in relation to possible synergism of xanthates combined with the derived biomass and biodegradable green reagents levulinic acid, 5-hydroxymethyl-2-furanacrylic acid (HMFA), and condensation product of hydroxymethylfurfural and levulinic acid. Optimal content of 30% of HMFA in the collector pointed to the possibility of substitution of commercial xanthate collector without significantly affecting its efficiency/selectivity. The density functional theory (DFT) calculations provided insights into the interaction mechanism of Na-iPrX and HMFA with cooper. The benefits of the presented method highlight production simplicity, mild reaction conditions, high purity and yield of products, no by-products, and technological applicability on an industrial scale.
