Summary: Cardiac surgery requiring cardiopulmonary bypass is associated with postoperative acute kidney injury and neurocognitive disorders, including delirium. Intra‐operative inflammation and/or impaired tissue perfusion/oxygenation are thought to be contributors to these outcomes. It has been hypothesised that these problems may be ameliorated by the highly selective α2‐agonist, dexmedetomidine. We tested the effects of dexmedetomidine on renal and cerebral microcirculatory tissue perfusion, oxygenation and histology in a clinically relevant ovine model. Sixteen sheep were studied while conscious, after induction of anaesthesia and during 2 h of cardiopulmonary bypass. Eight sheep were allocated randomly to receive an intravenous infusion of dexmedetomidine (0.4–0.8 μg.kg‐1.h‐1) from induction of anaesthesia to the end of cardiopulmonary bypass, and eight to receive an equivalent volume of matched placebo (0.9% sodium chloride). Commencement of cardiopulmonary bypass decreased renal medullary tissue oxygenation in the placebo group (mean (95%CI) 5.96 (4.24–7.23) to 1.56 (0.84–2.09) kPa, p = 0.001), with similar hypoxic levels observed in the dexmedetomidine group (6.33 (5.33–7.07) to 1.51 (0.33–2.39) kPa, p = 0.002). While no differences in kidney function (i.e. reduced creatinine clearance) were evident, a greater incidence of histological renal tubular injury was observed in sheep receiving dexmedetomidine (7/8 sheep) compared with placebo (2/8 sheep), p = 0.041. Graded on a semi‐quantitative scale (0–3), median (IQR [range]) severity of histological renal tubular injury was higher in the dexmedetomidine group compared with placebo (1.5 (1–2 [0–3]) vs. 0 (0–0.3 [0–1]) respectively, p = 0.013). There was no difference in cerebral tissue microglial activation (neuroinflammation) between the groups. Dexmedetomidine did not reduce renal medullary hypoxia or cerebral neuroinflammation in sheep undergoing cardiopulmonary bypass. [ABSTRACT FROM AUTHOR]

Much of the anthropological scholarship on women can be divided between those who emphasize the relative importance of capitalism (for example, Leacock 1981; Nash and Fernandez-Kelly 1983; Boserup 1970) and those who emphasize culture (for example, Ortner and Whitehead 1981; Schlegel 1990; Rosaldo 1974) as determinants of gender roles and relations.

Molecular-based analysis has become a fundamental tool to understand the role of Quaternary glacial episodes. In the Magellan Province in southern South America, ice covering during the last glacial maximum (20 ka) radically altered the landscape/seascape, speciation rates and distribution of species. For the notothenioid fishes of the genus Harpagifer, in the area are described two nominal species. Nevertheless, this genus recently colonized South America from Antarctica, providing a short time for speciation processes. Combining DNA sequences and genotyping-by-sequencing SNPs, we evaluated the role of Quaternary glaciations over the patterns of genetic structure in Harpagifer across its distribution in the Magellan Province. DNA sequences showed low phylogeographic structure, with shared and dominant haplotypes between nominal species, suggesting a single evolutionary unit. SNPs identified contrastingly two groups in Patagonia and a third well-differentiated group in the Falkland/Malvinas Islands with limited and asymmetric gene flow. Linking the information of different markers allowed us to infer the relevance of postglacial colonization mediated by the general oceanographic circulation patterns. Contrasting rough- and fine-scale genetic patterns highlights the relevance of combined methodologies for species delimitation, which, depending on the question to be addressed, allows discrimination among phylogeographic structure, discarding incipient speciation, and contemporary spatial differentiation processes. Abstract: A combination of state-of-the-art instrumentation and optimized data processing has enabled for the first time the chemical mapping of sub-nanometre particles using energy-filtered transmission electron microscopy (EFTEM). Multivariate statistical analysis (MSA) generated reconstructed datasets where the signal from particles smaller than 1nm in diameter was successfully isolated from the original noisy background. The technique has been applied to the characterization of oxide dispersion strengthened (ODS) reduced activation FeCr alloys, due to their relevance as structural materials for future fusion reactors. Results revealed that most nanometer-sized particles had a core–shell structure, with an Yttrium–Chromium–Oxygen-rich core and a nano-scaled Chromium–Oxygen-rich shell. This segregation to the nanoparticles caused a decrease of the Chromium dissolved in the matrix, compromising the corrosion resistance of the alloy. [Copyright &y& Elsevier]

Aims:  Characterization of the role of CaiC in the biotransformation of trimethylammonium compounds intol(−)-carnitine in Escherichia coli. Methods and Results:  The caiC gene was cloned and overexpressed in E. coli and its effect on the production ofl(−)-carnitine was analysed. Betaine:CoA ligase and CoA transferase activities were analysed in cell free extracts and products were studied by electrospray mass spectrometry (ESI-MS). Substrate specificity of the caiC gene product was high, reflecting the high specialization of the carnitine pathway. Although CoA-transferase activity was also detected in vitro, the main in vivo role of CaiC was found to be the synthesis of betainyl-CoAs. Overexpression of CaiC allowed the biotransformation of crotonobetaine tol(−)-carnitine to be enhanced nearly 20-fold, the yield reaching up to 30% (with growing cells). Higher yields were obtained using resting cells (up to 60%), even whend(+)-carnitine was used as substrate. Conclusions:  The expression of CaiC is a control step in the biotransformation of trimethylammonium compounds in E. coli. Significance and Impact of the Study:  A bacterial betaine:CoA ligase has been characterized for the first time, underlining its important role for the production ofl-carnitine with Escherichia coli. Abstract: Background: The diagnosis of renal failure is important in cirrhosis. Cystatin C (Cys) has been suggested not only to be a sensitive marker of renal function, but also a stronger predictor of the risk of death and cardiovascular events in heart failure. Our aims were to investigate plasma Cys concentrations for the detection of moderately impaired renal function and its association with mortality and cardiovascular events among cirrhotic patients after liver transplantation (OLT). Methods: Clinical and biochemical data, including Cys levels, were analyzed in 99 cirrhotic candidates for OLT. We recorded cardiovascular events. Results: Receiver-operator characteristics curves showed a similar efficiency to detect a creatinine clearance <60 mL/min per 1.73m2 (Cys = 0.753; creatinine [Cr] = 0.799; glomerular filtration rate [GFR, Cockcroft–Gault formula] = 0.842; urea = 0.823; P = .001). However, at cutoff concentrations of 1.3 mg/dL, Cr showed great specificity (96%) but poor sensitivity (13%), while the sensitivity of Cys was superior (83%) with moderate specificity (55%) at a cutoff of 1400 ng/mL. Over a median follow-up of 2.7 years, 14 patients developed a cardiovascular event, including, 11 who displayed Cys levels >1400 ng/mL before OLT, showing a significant difference (P < .05) compared to patients who showed no cardiovascular event. Kaplan–Meier analysis Cys discriminated significantly better than the Model for End-Stage Liver Disease score between survivors and nonsurvivors (P < .05). Conclusion: Cys determinations could be a valuable tool for early diagnosis of renal dysfunction among cirrhotic patients. Los restos óseos del sitio Mata Molle (Provincia de Neuquén) han constituido una importante referencia en los estudios sobre el poblamiento de la región Patagónica debido a la elevada antigüedad y las características morfológicas singulares que le fueron asignadas. En este trabajo se presentan los resultados del análisis de la morfología craneofacial en el contexto de los restos humanos tempranos de Sudamérica, junto con la primera datación radiocarbónica obtenida directamente sobre material óseo. Por medio del análisis osteológico se determinó que el entierro corresponde a dos individuos adultos, uno masculino (Ind. 1) y otro femenino (Ind. 2). El Ind. 1 presentó un elevado grado de desgaste dental y marcadores de estrés músculo esqueletal en el radio derecho y huesos de los miembros inferiores, compatibles con elevados niveles de actividad física. La estatura y masa corporal estimadas se encuentran dentro del rango observado en muestras del Norte de Patagonia de similar cronología. La morfología craneana del Ind. 1 exhibe gran similitud con la de las muestras más tempranas de Sudamérica y de la región Pampeana y se caracteriza por una mayor longitud anteroposterior y una reducción en el ancho y la altura. Sin embargo, la datación radiocarbónica indica que el entierro de Mata Molle corresponde al Holoceno tardío inicial (3270±46 años AP) y no al Holoceno medio o temprano como había sido señalado previamente. Los valores de d13C y de d15N se encuentran dentro de los rangos esperados para dietas terrestres.

La variación morfológica de las poblaciones humanas que habitaron el actual territorio de la provincia del Neuquén durante el Holoceno tardío, constituye un tema de investigación escasamente explorado hasta el presente. El objeto de éste trabajo es cuantificar el patrón de variación y el grado de disparidad en forma y tamaño facial de las poblaciones humanas del Noroeste de Patagonia durante el Holoceno tardío. Con fines comparativos, se analizaron dos muestras procedentes del Nordeste (Río Negro) y Centro-este (Chubut) de Patagonia, áreas para las que se cuenta con información bioarqueológica y arqueológica detallada. Los resultados obtenidos indican que la muestra de Neuquén presenta diferencias significativas en el patrón de variación en tamaño y forma del esqueleto facial con respecto a las otras dos muestras analizadas. Asimismo, la magnitud de la variación en el tamaño y la forma observada para la muestra de Neuquén resultó particularmente elevada, especialmente al comparar el área de estudio con el Nordeste de Patagonia, una región previamente caracterizada por elevados grados de variación morfológica. La evaluación de los procesos y eventos evolutivos y/o ecológicos que dieron origen a los patrones de variación aquí presentados requiere futuros estudios.

Durante las recientes excavaciones efectuadas en los restos de la corbeta británica HMS Swift, naufragada en 1770 en Puerto Deseado (Provincia de Santa Cruz), se encontró un esqueleto humano completo. La documentación histórica oficial señala que durante el naufragio desaparecieron dos infantes de marina, de 21 y 23 años de edad, cuyos nombres y lugar de procedencia son conocidos. Para determinar si el esqueleto corresponde a uno de estos dos individuos, se están desarrollando investigaciones centradas en obtener información relevante mediante el estudio del sexo, edad, estatura, patologías, patrones de actividad, dieta (isótopos estables del C y N) y características genéticas (rasgos no métricos, ADNmt). Los resultados de las estimaciones de sexo, edad y estatura indican que se trataría de un individuo masculino, adulto joven, de alrededor de 25 años al momento de morir (rango de edad calculada entre 17 y 34 años) y de alrededor de 1,67 m de estatura. La evidencia obtenida hasta el presente indica que, con una alta probabilidad, el esqueleto corresponde a uno de los dos tripulantes desaparecidos, aunque aún no puede asignarse a uno de ellos en particular, por lo que se espera que la información genética pueda aportar a su potencial identificación.

El objetivo del presente trabajo es analizar la variabilidad en la morfometría dental en muestras de restos óseos humanos del SE de la Región Pampeana asignables a distintos momentos del Holoceno tardío y comparar, desde un punto de vista espacial, el grado de similitud morfológica con muestras procedentes del NE de Patagonia (valles de Río Negro y Chubut, San Blas, Isla Gama). Se relevaron los diámetros mesiodistal y bucolingual medidos a nivel del margen cervical en la totalidad de las piezas dentales permanentes correspondientes a 130 individuos de ambos sexos. Las medidas obtenidas fueron empleadas en análisis estadísticos multivariados con el fin de evaluar el grado similitud morfológica entre las muestras. Los resultados obtenidos indican que: 1- hay diferencias entre las muestras procedentes del SE de la Región Pampeana correspondientes al Holoceno tardío inicial y final, caracterizándose estas últimas por un mayor tamaño dental; 2- la muestra del SE de la Región Pampeana correspondiente al Holoceno tardío final presenta mayor similitud morfológica con la muestra más tardía del valle de Río Negro; 3- existen variaciones cronológicas en el tamaño dental entre las muestras procedentes del NE de Patagonia.

En el presente, se encuentra en desarrollo un proyecto orientado a analizar la dinámica evolutiva de las poblaciones humanas de la porción central de Argentina y de Patagonia Continental durante el Holoceno, mediante la integración de diferentes líneas de evidencia morfológica (i.e. rasgos de variación continua y discontinua a nivel craneofacial, dental y postcraneal). El objetivo de este trabajo es presentar resultados parciales de esta investigación, obtenidos a partir del análisis del esqueleto craneofacial (morfometría geométrica) y de piezas dentales permanentes (morfometría tradicional) de 50 individuos adultos de ambos sexos, procedentes de sitios arqueológicos del Sur de Mendoza asignables a diferentes momentos del Holoceno tardío. Se registraron coordenadas de landmarks y semilandmarks sobre el esqueleto craneofacial en norma frontal y basilar y se relevaron los diámetros mesiodistal y bucolingual a nivel del cuello dental. Estas variables fueron empleadas en análisis de componentes principales. Los resultados obtenidos indican que, a nivel dental, se observó una diferenciación entre las muestras del Holoceno tardío inicial y final, caracterizándose las primeras por presentar mayores tamaños dentales. A nivel craneofacial también se observaron diferencias temporales, aunque con un patrón diferencial entre y dentro de cada sexo.
