6. Metal-catalyzed oxidation of phenylalanine-sensitive 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase from Escherichia coli: inactivation and destabilization by oxidation of active-site cysteines

Abstract

Research was conducted to examine the metal-catalyzed oxidation mechanism that causes the in vitro instability of the phenylalanine-sensitive 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase (DAHPS) from Escherichia coli. A disulfide linkage between the two cysteines, namely, Cys61 and Cys328, was observed from peptide mapping of the Cu2+-inactivated enzyme. A proposed inactivation mechanism of DAHPS features a metal redox cycle that needs a sequential oxidation of its two active-site cysteines.

Abstract

The tryptophan-inhibited 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase (DAHPS-(Trp)) of Escherichia coli was examined with regards to steady-state kinetics and tryptophan binding. Findings revealed two independent inhibitor binding sites per enzyme dimer. L-tryptophan binding decreased k(sub cat), increased affinity for both substrates, decreased positive homotropic cooperativity for both substrates and activated the enzyme at low concentrations of erythrose-4-phosphate.

Abstract

Crystallographic analysis shows that the plane of phosphoenolpyruvate bound at the active site of the mutant 3-deoxy-D-arabino-heptulosonate-7-phosphate synthase is twisted 30 degrees about its C--C single bond. Data indicate that the twist is at the carboxylate plane relative to the enol plane.

Abstract

The isolation of an aminated intermediate as a product of an enzymatic activity of the Salmonella typhimurium AS is reported. Hydrogen nuclear magnetic resonance spectrum analysis is employed to determine the proton-decoupled natural abundance of an aminochorismate compound. Results reveal that the aminochorismate compound used in the study is an intermediate in the anthranilate synthase reaction.
