The use of a l[micro]m ion beam as a tomographic probe could constitute a powerful tool for displaying the 3D structure of samples a few tens or hundreds of micrometers thick in a nondestructive way. At the Centre d'Etudes Nucleaires de BordeauxGradignan (CENBG), Gradignan, France, ion beam microtomography has been developed for biomedical applications at the cell level. The internal structure of cancer cells has been explored using scanning transmission ion microscopy (STIM) tomography, giving access to the 3D distribution of mass density (in g/c[m.sup.3] within the analyzed volume. The sole sample preparation required is cryofixation and freeze-drying, permitting under vacuum analysis. The combination of STIM and particle induced X-ray emission (PIXE) tomography is under progress with the aim of revealing the distributions and local concentrations of elements at the cell level. Microbeam facilities provide a unique opportunity to investigate the effects of ionising radiation on living biological cells with a precise control of the delivered dose. This paper describes dosimetry calculations performed at the single-cell level in the microbeam irradiation facility available at the Centre d'Etudes Nucléaires de Bordeaux-Cradignan in France, using (he object-oriented Geant4 Monte Carlo simulation toolkit. The cell geometry model is based on high-resolution three-dimensional voxelised phantoms of a human keratinocyte (HaCaT) cell line. Such phantoms are built from confocal microscopy imaging and from ion beam chemical elemental analysis. Results are presented for single-cell irradiation with 3 MeV incident alpha particles. An irradiation facility, able to expose cellular and subcellular targets to a precise number of particles, has been developed at CENBG for applications in radiobiology. The development of this facility was based on an existing horizontal focused microbeam developed in the early 90’s for material analysis. The focusing properties of the line allow the delivering of proton or alpha particle beams in the 1–3.5 MeV energy range with a spatial resolution down to about 1 μm under vacuum. For irradiation of living cells, a removable stage has been developed to extract the beam into air while preserving the analytical capabilities of the microbeam line under vacuum. This stage includes a high resolution epifluorescence microscope for online visualization of the cells and a motorized stage for cell positioning. Single particle control is ensured by a fast electrostatic deflector triggered by the signal induced by the particles through a transmission detector just before reaching the target. A dedicated software, based on an object-oriented architecture, has been designed to control the entire experiment. This includes semiautomatic calibration procedures (necessary to achieve the micron precision) and semiautomatic irradiation procedures used for targeting a large number of individual cells. In air irradiation of solid track detectors has permitted us to estimate that 99.5% of the particles are delivered on the target at a distance lower than 5 μm from the beam center when an alpha particles beam is used. The targeting precision of the overall irradiation procedure, which reflects the alignment precision of the beam center with the target center, has been estimated to be within ±2 μm. First experiments involving cells in culture have permitted to estimate an irradiation rate of 2000 cells per hour. Abstract: The use of ion microbeams as probes for computed tomography has proven to be a powerful tool for the three-dimensional characterization of specimens a few tens of micrometers in size. Compared to other types of probes, the main advantage is that quantitative information about mass density and composition can be obtained directly, using specific reconstruction codes. At the Centre d’Etudes Nucléaires de Bordeaux Gradignan (CENBG), this technique was initially developed for applications in cellular biology. However, the observation of the cell ultrastructure requires a sub-micron resolution. The construction of the nanobeam line at the Applications Interdisciplinaires des Faisceaux d’Ions en Region Aquitaine (AIFIRA) irradiation facility has opened new perspectives for such applications. The implementation of computed tomography on the nanobeam line of CENBG has required a careful design of the analysis chamber, especially microscopes for precise sample visualization, and detectors for scanning transmission ion microscopy (STIM) and for particle induced X-ray emission (PIXE). The sample can be precisely positioned in the three directions X, Y, Z and a stepper motor coupled to a goniometer ensures the rotational motion. First images of 3D tomography were obtained on a reference sample containing microspheres of certified diameter, showing the good stability of the beam and the sample stage, and the precision of the motion. The energy and specific energy absorbed in the main cell compartments (nucleus and cytoplasm) in typical radiobiology experiments are usually estimated by calculations as they are not accessible for a direct measurement. In most of the work, the cell geometry is modelled using the combination of simple mathematical volumes. We propose a method based on high resolution confocal imaging and ion beam analysis (IBA) in order to import realistic cell nuclei geometries in Monte-Carlo simulations and thus take into account the variety of different geometries encountered in a typical cell population. Seventy-six cell nuclei have been imaged using confocal microscopy and their chemical composition has been measured using IBA. A cellular phantom was created from these data using the ImageJ image analysis software and imported in the Geant4 Monte-Carlo simulation toolkit. Total energy and specific energy distributions in the 76 cell nuclei have been calculated for two types of irradiation protocols: a 3 MeV alpha particle microbeam used for targeted irradiation and a ²³⁹Pu alpha source used for large angle random irradiation. Qualitative images of the energy deposited along the particle tracks have been produced and show good agreement with images of DNA double strand break signalling proteins obtained experimentally. The methodology presented in this paper provides microdosimetric quantities calculated from realistic cellular volumes. It is based on open-source oriented software that is publicly available.

The ionic channels (particularly, K+ and Ca2+ channels) regulate, via the membrane potential, the ionic distribution into the vascular cells. Micro-particule induced X-ray emission (PIXE) analysis was applied to determine the ionic composition of vascular smooth muscle cells (VSMCs) and of vascular endothelial cells (VECs) in the placental human allantochorial vessels in a physiological medium (Hanks' solution) modified by the addition of a NO donor (sodium nitroprusside, SNP) and of a beta-adrenergic stimulator (isoproterenol, ISO). The addition of SNP or ISO induced no modification of the Na, K, Cl, P, S, Mg and Ca concentrations in VSMCs. In VECs, a same effect was observed except an increase of the Mg concentration with ISO. Theses results indicated a retroactive control (active feedback) of the internal ionic distribution by endothelial factors, ionic channels and exchangers.

The membrane potential, a regulator of vascular tone, is a function of the physiological activities of ionic channels (particularly, K+ and Ca2+ channels in these cells). These channels regulate the ionic distribution into these cells. Micro-particule induced X-ray emission (PIXE) analysis was applied to determine the ionic composition of vascular smooth muscle cells (VSMCs) and of vascular endothelial cells (VECs) in the placental human allantochorial vessels in a physiological medium (Hanks'solution) modified by the addition of a chemical stimulus: 5-hydroxytryptamine (5-HT), an activator of the voltage-sensitive Ca2+ channels. In VSMCs (media layer), the addition of 5-HT induced no modification of the Na, K, Cl, P, S and Ca concentrations but increased Mg concentration. In endothelium (VECs) 5-HT addition implicated an increase of the K, S, Ca concentrations, the concentration of the other ions remained constant. In VECs, Ca and K increase is due to open of L-type voltage-dependent Ca2+ channels and of K(Ca) channels. 5-HT induces also a secretion of endothelium hyperpolarizing factors which implicate decrease of [Ca2+]i in VSMCs opposite to a direct increase by 5-HT. Increase in [Mg2+]i may be due to activation of the Ca/Mg exchanger.
