Abstract

The stiffness coefficients of anisotropic materials were determined through a scanned point source-point receiver method, based on laser generation and detection of acoustic waves. A silicon crystal which is very sensitive to acoustic wave focusing induced by anisotropy was examined first. Four coefficients of the stiffness tensor were recovered with good reliability by scanning a symmetry plane.

Abstract

A specific ultrasonic device investigates the inelastic behavior of a ceramic matrix composite and confirms their nonlinear behavior due to microcracks. The growth of microcracks retarded the microscopic nature of matrix composites, this effect being anisotropic and disoriented. Damage involving the C55 shear modulus is as severe as that related to the stiffness of C33 in the tensile direction.

Abstract

Recently, the coherent generation of GHz acoustic waves using ultrashort laser pulses has demonstrated the ability to probe the sound velocity in vegetal cells and in cell-mimicking soft micro-objects with micrometer resolution, opening tremendous potentialities for single-cell biology. However, manipulating biological media in physiological conditions is often a technical challenge when using a laser-based setup. In this article, we present a new opto-acoustic bio-transducer composed of a thin metal film sputtered on a transparent heat sink that allows reducing importantly the laser-induced cellular stresses, and offers a wide variety of optical configurations. In particular, by exploiting the acoustic reflection coefficient at the sample-transducer interface and the photoacoustic interaction inside the transparent sample, the density and compressibility of the sample can be probed simultaneously. Using an ad hoc signal analysis based on Hilbert and wavelet transforms, these quantities are measured accurately for a reference fluid. Similar analysis performed in a single vegetal cell also suggests high sensitivity to the state of the transducer-cell interface, and notably to the presence of the plasma membrane that encloses the cell vacuole. Abstract

The acoustic displacements excited by a laser line source under either the ablation or thermoelastic regime are calculated for a two-layered cylinder made of homogeneous and isotropic materials. Two samples made by welding tin in two different copper tubes are considered. Experimental displacements are observed by the laser ultrasonic technique, and corresponding theoretical waveforms are calculated. Good agreement is found in the arrival time, shape, and relative amplitude of various longitudinal and shear bulk waves propagating in the two-layered cylinders, and the waves reflected by the interface are further identified by the ray trajectory analysis. The longitudinal wave reflected at the interface may be used for the nondestructive evaluation of the interface quality. [ABSTRACT FROM AUTHOR]

Abstract

An optical grating has been introduced in a picosecond ultrasonics experiment, in order to vary continuously the duration of the laser beam pulse from 0.1 to 150 ps. The evolution of the measured signal has been observed and analyzed through the comparison with a theoretical approach based on a two-temperature model. The latter allows matching the acoustic echoes together with the thermal background and the coincidence peak, for each pulse duration and at any time scale. The broadening of the acoustic echoes and the disappearing of its Brillouin component, along with the diminishing of the thermal coincidence peak, have been demonstrated when increasing the pulse duration. For a constant incident pulse energy, the efficiency of acoustic generation is optimum for the shortest pulses. Nevertheless, for longer pulses designed to obtain thermal conditions below the ablation threshold, acoustic generation could be enhanced. [ABSTRACT FROM AUTHOR]

Abstract

A noncontact laser–ultrasonic technique is described that allows determination of material properties of anisotropic platelike samples. The method requires access to only one side of the sample. Plane waves are synthesized by scanning the surface with a laser line source at judiciously chosen intervals. By doing so, the intricacies associated with the reflection of an acoustic ray off an interface of an anisotropic medium are avoided. The plane-wave reflections off the opposite free surface of the plate are simply governed by Snell's law. The identification of the stiffness-tensor components is based on the inversion of the phase velocities of these reflected plane waves. The procedure developed is presented and applied to both simulated and experimental signals. The method is easy to implement, and provides a means for noncontact measurement of elastic properties where there is limited access to test specimens. © 2001 American Institute of Physics. Abstract

Objective: In early multiple sclerosis, although brain T2 lesions accrual are hallmark of the disease, only weak correlations were found between T2 lesions accrual and EDSS progression, the disability scale commonly used in multiple sclerosis studies. This may be related to the very poor sensitivity of EDSS to cognitive dysfunctions that may occur and progress from the first stage of the disease. In the present study, we aimed to demonstrate that cognitive deficits progress during the first ten years of MS and are significantly impacted by new T2 lesions. Methods: EDSS and extensive neuropsychological battery (22 measures) exploring memory, attention/speed of information processing and executive functions were assessed at baseline, Year 1 and Year 10 in 26 patients enrolled after their first clinical attack. To limit the bias of test-retest effect, only measures obtained at Year 1 and Year 10 were reported in the analysis. Raw scores of patients were transformed into z-scores using published normative data when available or scores of matched controls. Lesion probability mapping was used to assess the potential relationships between T2 lesions accumulation, cognitive decline and EDSS progression (P<0.05, FWE-corrected). Results: At Year 1, 27% of patients showed attention/speed of information processing deficits, 11.5% executive dysfunction and 11.5% memory impairment. During the follow-up, frequency and severity of executive dysfunction increased (from 11.5% of patients at Year 1 to 42% at Year 10, p<0.01) while no significant changes were evidenced for the other cognitive domains. Median EDSS increased from 0.5 [range: 0–3] at Year 1 to 2.5 [range: 0–6.5] at Year 10 (p<0.001). During the ten-year follow-up, lesions accumulation in the left cerebellum and semi-ovale centers was associated with EDSS progression. In contrast, most lesions accumulation in the frontal, parietal and temporal lobes were associated with cognitive decline but had no effect on EDSS progression. Conclusion: The present study provides strong evidence that clinically silent T2 lesions impact cognition in early MS. In daily practice, early prevention of T2 lesions accrual may be useful to limit cognitive decline. [ABSTRACT FROM AUTHOR]

Abstract

Background: Despite a growing use of rituximab (RTX) in neuromyelitis optica (NMO), data are lacking in patients with refractory NMO (RNMO), defined as cases with at least one relapse during immunosuppressive therapy. Objective: The purpose of this study was to assess RTX as a maintenance therapy in RNMO. Methods: Out of a total of 305 NMO cases from a population-based cohort, 21 RNMO patients received RTX during a mean follow-up period of 31 months. Results: After RTX, 11 patients (52.3%) were relapse free, meaning that 47.7% were refractory to RTX. The mean annualized relapse rate decreased from 1.3 to 0.4 (p<0.001) and median EDSS from 5 to 3 (p=0.02). Body mass index (BMI) was predictive of EDSS worsening. Conclusions: RTX is an effective and well-tolerated treatment in RNMO. BMI could be a predictive factor for efficacy. [ABSTRACT FROM AUTHOR]

Abstract

Techniques that can probe the thermal properties of cells are used in many applications ranging from cryogenic preservation to hyperthermia therapy, and provide powerful tools to investigate diseased conditions. The structural complexity of cells, however, requires innovative modalities operating at a subcell scale. We developed a label-free, non-ionizing technique based on a thermoelastic lens. With this device, we captured images of single cells with a ~2 μm resolution based on thermal properties as the contrast mechanism. To investigate the thermorheological behaviour of cells, we present simultaneous acoustic imaging using an inverted opto-acoustic microscope. Acoustic impedances extracted from the acoustic images support the effusivity obtained from the thermal images. This technique should provide diagnostic tools at the single cell scale.
