Abstract Background Chronic pain is a major health issue and rapid population ageing exacerbates the burden to health systems in countries like Germany. Nonpharmacological interventions (NPIs) are essential in pain care and the prioritization of active NPIs is emphasized in guidelines. This paper examines the utilization of NPIs for chronic pain management in community-dwelling older adults with a certified need of care in Berlin, Germany. Methods Cross-sectional data was collected through standardized face-to-face surveys with older adults (≥65 years), using validated instruments (e.g., Brief Pain Inventory), and structured lists for NPI utilization. Categorization into active and passive NPIs was performed through a literature-based, iterative process by an interdisciplinary team. For not normally distributed data, non-parametric tests were used as appropriate. Logistic regression was conducted for multivariate analysis. Results In total, 250 participants were included in this analysis (aged 65-104, x̅ = 81.8, 68.8% female). Most (92%) use NPIs for chronic pain management: 85.6% use active NPIs, 50.4% active movement and only 5.6% use solely passive approaches. Most common NPIs are distraction, thermotherapy/compresses, and physiotherapy. The odds of utilizing physiotherapy are three times higher for those with high educational status when compared to those with low education while those with low educational status had higher odds of using thermotherapy/compresses. Conclusions In our sample, most community-dwelling older adults with a certified need of care use active NPIs for chronic pain management with about half using active movement approaches. Considering the high vulnerability of this population, physiotherapy (in the form of therapeutic exercise) is a particularly appropriate intervention, and it was the third most frequent NPI in our sample. However, there is a social gradient in the utilization of physiotherapy for chronic pain management which might be rooted in issues around awareness, appeal, and access to such measures. It is important to take socioeconomic differences into account when planning the care for older chronic pain patients but also when designing research or user-friendly guidelines for this target group. Objetivo. Determinar la prevalencia, cargas parasitarias y distribución de las infecciones naturales con estróngilos digestivos dentro de rebaños bovinos de una zona ganadera del Estado Falcón en Venezuela Materiales y métodos. Se evaluaron cuatro fincas de bovinos doble propósito. Los animales son tipo “Carora” (mestizaje de razas: Holstein, Pardo Suizo, Cebuínos). Durante siete meses se analizaron (Técnica de Mc Master) un total de 1140 muestras. La muestra de cada finca se estratificó en cuatro grupo etarios (1: Becerros 0-6 meses; 2: Mautes 7-12; 3: Novillas 13-24 y 4: Adultos >25 meses). Resultados.  Las estrongilidosis digestivas, son las infecciones parasitarias de mayor impacto en los pequeños rumiantes. Son causadas por helmintos nematodos del orden Strongylida. La escala FA- MACHA®, permite identificar a los animales más sensibles a los parásitos dentro del rebaño para concentrar en ellos las medidas de control. Con el objetivo de evaluar la prevalencia de infección y contajes fecales de estrongilidos y su relación con los valores de la escala FAMACHA® en un rebaño caprino, se establecieron tres grupos (Grupo 1-G1: cabras en lactación; Grupo 2-G2: padrotes; Grupo 3-G3: cabritos de ambos sexos). Se evaluaron 62 animales, las muestras fecales se procesaron con la técnica de Mc Master y se hicieron coprocultivos; se usaron tarjetas FAMACHA® para registrar valores de coloración de conjuntiva ocular de cada animal. La prevalencia promedio de estrongilidosis fue 64,94%; los contajes fecales promedio fueron de moderados a altos (G1= 313,64 Ó 63,53; G2= 1050 Ó 549,35; G3= 345,48 Ó 275,11). De los coprocultivos se identificaron L3 de: Haemonchus spp., y Trichostrongylus spp. Los valores de FAMACHA® (2-5) mostraron diferencias entre los grupos etarios (P < 0,05) y al correlacionarlos con los contajes fecales (P = 0,0097), evidenciando la asociación entre ambas variables, indicando su utilidad en la identificación de los animales susceptibles a estrongilidos dentro de este rebaño.

Las helmintiasis del tracto gastrointestinal de los caninos domésticos constituyen un grupo de parasitosis con serias implicaciones en la salud de los animales y algunas son causantes de zoonosis de importancia en Salud Pública. El presente trabajo tuvo como objetivo evaluar la eficiencia antihelmíntica del Nitroscanato en perros naturalmente infectados con Toxocara canis y Ancylostoma spp. Se realizó un ensayo conformado por un grupo de 8 caninos tratados a la dosis de 50 mg/kg v.o. (2 aplicaciones con intervalo de 2 semanas) y un grupo control no tratado de 8 animales. El Nitroscanato demostró una alta eficiencia (100%), disminuyendo en forma significativa la carga parasitaria de Toxocara canis y Ancylostoma spp, con una reducción del 100% en el HPG de Ancylostoma spp y en la prevalencia de de Toxocara canis y Ancylostoma spp. Se recomienda utilizar este producto como una alternativa de utilidad en el tratamiento y control de las principales helmintiasis gastrointestinales que afectan a los perros en Venezuela.

Los estróngilos son los parásitos más comunes en los caballos, causando serios problemas aún en aquellos animales que reciben tratamiento antihelmíntico. Existe en el país escaso conocimiento del comportamiento de este parasitismo en caballos salvajes cuyas poblaciones de helmintos no están influenciadas por el uso de fármacos antiparasitarios, por lo que el objetivo del presente estudio fue determinar las características de la infección por nematodos Strongylidae en caballos salvajes venezolanos no sometidos a tratamiento antihelmíntico en base a examen coprológico. El estudio se realizó en Abril 2006 en el Hato El Frío, ubicado cerca de Mantecal, estado Apure, Venezuela. Se tomó una muestra de heces directamente del recto a un total de 35 caballos salvajes (21 machos, 14 hembras, representando el 15% de la población), las cuales se procesaron por la técnica de Mc Master modificada y se realizaron cultivos a partir de 4 grupos (pool) de muestras positivas para la identificación de las larvas Strongylidae de tercer estado. Se obtuvo una prevalencia general de 82,86% (29/35) con un promedio general de 818,57 huevos por gramo de heces (hpg) [0-2.300]. De los animales examinados, el 25,71% tenía entre 500 y 1.000 hpg, mientras que 37,14% tenía más de 1.000 hpg. Strongylus vulgaris fue encontrado en 3 de 4 coprocultivos (75%) en porcentajes de 2, 12 y 7%, respectivamente. S. edentatus fue encontrado en 1 de 4 coprocultivos (25%) en porcentaje del 2%. La presencia de grandes estróngilos (S. vulgaris, S. edentatus) encontrados en los coprocultivos fue bastante marcada, así como el significativo número de animales con contajes superiores a 1.000 hpg.

El presente estudio determinó la fluctuación poblacional de las fases no parasíticas de Rhipicephalus sanguineus durante octubre 2000 - septiembre 2001. Mensualmente se recolectaron todas las garrapatas presentes en los distintos ambientes identificados dentro de las instalaciones del Bioterio de la Facultad de Ciencias Veterinarias de la Universidad Central de Venezuela, Maracay, estado Aragua, Venezuela. Se recolectaron 16.065 ejemplares de R. sanguineus en diferentes fases de desarrollo: 5.981 larvas, 7.378 ninfas, 796 machos y 1.916 hembras; el mayor número se obtuvo en el interior de las jaulas (13.364), representando las ninfas la fase más abundante, tanto en el interior como en el exterior. A field experiment was carried out to quantify the effect of a native bacterial inoculant on the growth, yield, and quality of the wheat crop, under different nitrogen (N) fertilizer rates in two agricultural seasons. Wheat was sown under field conditions at the Experimental Technology Transfer Center (CETT-910), as a representative wheat crop area from the Yaqui Valley, Sonora México. The experiment was conducted using different doses of nitrogen (0, 130, and 250 kg N ha−1) and a bacterial consortium (BC) (Bacillus subtilis TSO9, B. cabrialesii subsp. tritici TSO2T, B. subtilis TSO22, B. paralicheniformis TRQ65, and Priestia megaterium TRQ8). Results showed that the agricultural season affected chlorophyll content, spike size, grains per spike, protein content, and whole meal yellowness. The highest chlorophyll and Normalized Difference Vegetation Index (NDVI) values, as well as lower canopy temperature values, were observed in treatments under the application of 130 and 250 kg N ha−1 (the conventional Nitrogen dose). Wheat quality parameters such as yellow berry, protein content, Sodium dodecyl sulfate (SDS)-Sedimentation, and whole meal yellowness were affected by the N dose. Moreover, the application of the native bacterial consortium, under 130 kg N ha−1, resulted in a higher spike length and grain number per spike, which led to a higher yield (+1.0 ton ha−1 vs. un-inoculated treatment), without compromising the quality of grains. In conclusion, the use of this bacterial consortium has the potential to significantly enhance wheat growth, yield, and quality while reducing the nitrogen fertilizer application, thereby offering a promising agro-biotechnological alternative for improving wheat production.

Indigenous peoples of Taiwan, the most disadvantaged of the minority groups, account for only about 2.4% of the total population. Within the Minnan dominant society of Taiwan, the Indigenous minority groups are confronted by critical language loss often attributed to cultural and structural assimilation factors. This study aims to investigate factors related with Austronesian language retention for indigenous peoples in Taiwan. Results reveal, Austronesian language fluency is reduced by education, but maintained by age, residing in an ethnic enclave during childhood, and practicing parental endogamy.

Abstract Despite the advance and success of precision oncology in gastrointestinal cancers, the frequency of molecular‐informed therapy decisions in pancreatic ductal adenocarcinoma (PDAC) is currently neglectable. We present a longitudinal precision oncology platform based on functional model systems, including patient‐derived organoids, to identify chemotherapy‐induced vulnerabilities. We demonstrate that treatment‐induced tumor cell plasticity in vivo distinctly changes responsiveness to targeted therapies, without the presence of a selectable genetic marker, indicating that tumor cell plasticity can be functionalized. By adding a mechanistic layer to precision oncology, adaptive processes of tumors under therapy can be exploited, particularly in highly plastic tumors, such as pancreatic cancer.

Abstract By accentuating drug efficacy and impeding resistance mechanisms, combinatorial, multi‐agent therapies have emerged as key approaches in the treatment of complex diseases, most notably cancer. Using high‐throughput drug screens, we uncovered distinct metabolic vulnerabilities and thereby identified drug combinations synergistically causing a starvation‐like lethal catabolic response in tumor cells from different cancer entities. Domperidone, a dopamine receptor antagonist, as well as several tricyclic antidepressants (TCAs), including imipramine, induced cancer cell death in combination with the mitochondrial uncoupler niclosamide ethanolamine (NEN) through activation of the integrated stress response pathway and the catabolic CLEAR network. Using transcriptome and metabolome analyses, we characterized a combinatorial response, mainly driven by the transcription factors CHOP and TFE3, which resulted in cell death through enhanced pyrimidine catabolism as well as reduced pyrimidine synthesis. Remarkably, the drug combinations sensitized human organoid cultures to the standard‐of‐care chemotherapy paclitaxel. Thus, our combinatorial approach could be clinically implemented into established treatment regimen, which would be further facilitated by the advantages of drug repurposing.

The Hakka people, the largest ethno-linguistic minority group in Taiwan, have found their ethnic language retention diminishing. Using the data collected by the Taiwan Education Panel Survey and Beyond in 2010, we are the first to study its reason for decrease. Results indicate that out-marriage amongst Hakka people and losing ethnic concentration are the major factors preventing Hakka language retention.

Esta investigación se enfoca en la caracterización de la conducta adaptativa en escolares con y sin dis - capacidad intelectual de la Región de La Araucanía, Chile. Se utilizó el Inventario para la Planificación de Servicios y Programación Individual (ICAP), aplicado a padres y/o tutores legales de 22 niños con disca - pacidad intelectual y 33 niños sin discapacidad. Las edades fluctuaron entre los 6 y 12 años, con un di - seño no probabilístico en conglomerados. El método utilizado es descriptivo-comparativo. Los resultados señalan que existen diferencias de las conductas adaptativas, donde los escolares sin discapacidad presentan un mayor nivel de desarrollo, además ambos grupos poseen un perfil heterogéneo en las destrezas que componen el funcionamiento adapta - tivo, donde las destrezas motoras son las más altas, mientras que las destrezas de la vida en comunidad son las más bajas. Se concluye que las diferencias encontradas se ven afectadas por la presencia de discapacidad intelectual y por el contexto educativo. Type 1 diabetes (T1D) is an autoimmune disease caused by the selective destruction of the pancreatic β-cells, causing inability to produce insulin. Proinflammatory cytokines such as IL-1β, IL-6, TNF-α, IFN-γ, IL-12, IL-17, and NO can be released by CD4 and CD8+ lymphocytes as well as by classically activated macrophages (CAMϕs), which are important in the development of T1D. Helminth infections have been shown to prevent T1D, mainly through Th2-biased responses and increased recruitment of regulatory cell populations. Previously, we have shown that Taenia crassiceps infection in mice significantly reduces hyperglycemia, insulitis, and the incidence of T1D. In this study, we determined whether T. crassiceps-derived products such as soluble (TcS) or excreted/secreted (TcES) antigens might have a beneficial influence on the development of experimental T1D. Treatment with different doses before or after induction of T1D was analyzed. Mice that were pretreated with TcS were unable to develop T1D, whereas those receiving TcES early after T1D induction displayed significantly reduced insulitis and hyperglycemia along with increased recruitment of alternatively activated macrophages (AAMϕs) and myeloid-derived suppressor cells (MDSCs). Finally, we examined the modulatory role of AAMϕs on T1D by depleting macrophages with clodronate-loaded liposomes, demonstrating that AAMϕs are key cells in T1D regulation.

This paper describes a student essay competition with the title "What makes a good engineering lecturer?" run by the Engineering Subject Centre of the UK Higher Education Academy. The competition was intended primarily to provide engineering lecturers with general feedback. The aim of the paper is to make the outcomes of the competition available in a convenient form to those who might be interested--particularly engineering lecturers. The paper describes the competition, sets out the approach used to analyse the essays, presents the content in terms of the main attributes identified, and compares these with published lists of recommended attributes. The paper lets the students speak for themselves through extensive quotation. (Contains 1 table.)

Rural/urban differences were studied in self-reported stress, coping, and behavioral problems in adolescents. Levels of stress and ways of coping were similar between the two groups. However, urban males reported more conflict and externalizing behaviors than females and rural males. Results suggest that adolescents may utilize many coping strategies that serve little benefit in terms of behavioral outcomes. (Contains 45 references and 4 tables.)

Examines the relationship between perceived social inadequacy and depressed mood in a sample of community adolescents. The Checklist of Adolescent Problem Situations (CAPS) and Children's Depression Inventory (CDI) were administered to 224 high-school students on two occasions 4 months apart. Results indicated that perceived social inadequacy in adolescents is stable over time, independent of its association with depressed mood. (Contains 21 references and 1 table.) (GCP)

In order to prepare protein extracts of Plasmodium falciparum blood stages for western blot analysis, infected red blood cells (iRBC) need to be separated from uninfected red blood cells (uRBC) which make up the bulk of the parasite culture. Depending on the localisation of the parasite protein of interest, different methods are available to achieve this. If the protein is present within the parasite or is attached to a cellular structure of the iRBC cell, saponin can be used. This reagent lyses the membranes of infected and uninfected erythrocytes, the Maurer´s clefts (vesicular structures in the iRBC) and the parasitophorous vacuole membrane containing the parasite but leaves the parasite plasma membrane intact, providing a convenient procedure to isolate intact parasites without uRBCs. However, this method has the disadvantage that the host cell cytosol and the parasitophorous vacuole (PV) content of iRBCs are lost. If this has to be avoided, it is possible to use a Percoll gradient to separate intact iRBCs from uRBCs. Sequential treatment with Tetanolysin and saponin can then be used to selectively release the iRBC cytosol and the PV content from the parasite. These selective lysis methods are also suitable to determine the subcellular localisation of a protein of interest.

This protocol was used to prepare pre-embedding samples of Plasmodium falciparum blood stage parasites that overexpressed the parasite protein PF13_0191 tagged with GFP. Using GFP-specific antibodies and Protein A-Gold the localisation of the overexpressed protein in the infected host cell was determined using standard transmission electron microscopy (EM). Pre-embedding EM is a common method where the antibodies are introduced before embedding and sectioning. This method avoids the problem that antigens are often difficult to detect on EM-sections after embedding. In the method described here antigens in the parasite-infected host cell are detected. Entry of the antibody is made possible through permeabilisation of the host cell with tetanolysin. In principle this method could also be used to detect antigens within the parasite if the sample is appropriately fixed and permeabilised before addition of the relevant antibody. While access of the antibody will avoid the detection problems often seen with post-embedding methods, this procedure will produce comparably poorer morphology.
