BackgroundFoam sclerotherapy is currently the first-line treatment for venous malformations (VMs). Hyaluronic acid-polidocanol (HA-POL) foam has been used in the treatment of head and neck VMs recently; however, its clinical efficacy and safety have yet to be further evaluated, and the impact of age and other related factors on its safety is still unclear.ObjectiveTo assess the efficacy and safety of HA-POL foam in the treatment of head and neck VMs.Methods and materialsWe performed a single-center retrospective review of all patients with VMs involving the head and neck region undergoing HA-POL foam sclerotherapy from February 2015 to February 2022 in the Oral and Maxillofacial Surgery Department of Qilu Hospital Shandong University. Patients’ medical records were collected and all patients enrolled were followed up for 1–6 months (group 1), part of them were followed up for 3–9 years (group 2).ResultsA total of 223 patients with head and neck VMs were enrolled in the study, with 36 patients who were followed for 3–9 years. Total response rate in group 1 was 96.41% (n = 215), of which 30.94% (n = 69) of the patients met the criteria of “resolution,” and 65.47% (n = 146) of the patients had “significant improvement.” In group 2, the total response rate was 72.22% (n = 26), of which the rates of the patients met the criteria of “resolution” and patients had “significant improvement” were all 36.11% (n = 13)0.144 (64.57%) patients experienced complications like localized swelling, pain and fever, and no serious complications occurred. The risk of developing complications after treatment was independent of age, and was weakly associated with the dose of HA-POL foam.ConclusionThe HA-POL foam sclerotherapy is safe and effective in the treatment of head and neck VMs.

IntroductionAn effective tool is needed to predict the prognosis of head and neck squamous cell carcinoma (HNSCC). Human papillomavirus (HPV) positive HNSCC patients generally have a favorable survival and a promising responsiveness to radiotherapy, chemoradiotherapy and checkpoint blockades. However, HPV negative patients, the majority of HNSCC patients, have been largely overlooked. Cell death has been involved in the therapeutic resistance of cancers. To this end, we aimed to identify the association of autophagy, apoptosis and pyroptosis-related genes with the prognosis of HNSCC, and construct a prognostic signature to predict the prognosis for HNSCC, especially for HPV negative HNSCC.MethodsAutophagy and apoptosis-related genes were obtained from Gene Set Enrichment Analysis (GSEA) website, and pyroptosis-related genes were obtained from GSEA and Gene Ontology (GO) database. We established the cell death index (CDI) based on RNA sequencing (RNA-seq) data and clinicopathological information from The Cancer Genome Atlas (TCGA) dataset. The prognostic value of CDI was verified by Kaplan-Meier, receiver operating characteristic (ROC) and univariate and multivariate Cox regression analyses in TCGA dataset, and validated with the datasets from Gene Expression Omnibus (GEO) and Qilu Hospital of Shandong University. We further assessed the immune microenvironment of patients with high and low CDI scores. Moreover, the expression of the signature genes in HNSCC cell lines were explored.ResultsWe found that CDI was an independent prognostic indicator for overall survival (hazard ratio 3.80, 95% confidential interval: 2.70-5.40, P < 0.001). Furthermore, HNSCC patients with high CDI scores obtained increased overall survival post radiation indicating benefits from radiotherapy of this subgroup. On the other hand, HPV negative HNSCC patients with low CDI exhibited increased checkpoint gene expressions, an inflamed tumor microenvironment and an enriched immune response-related functions, suggesting the potential benefits from checkpoint immunotherapies of this subgroup. Moreover, we validated the baseline and induced expressions of above 16 genes in two HPV negative HNSCC cell lines, CAL27 and SCC-15.DiscussionWe established a prognostic signature and emphasized its implements in the therapeutic choices of HPV negative HNSCC patients, the majority and the poor outcome population of HNSCC.

Abstract Background AKT2 is one of the key molecules that involves in the insulin-induced signaling and the development of cancer. In B cells, the function of AKT2 is unclear. Methods In this study, we used AKT2 knockout mice model to study the role of AKT2 in BCR signaling and B cell differentiation. Results AKT2 promotes the early activation of B cells by enhancing the BCR signaling and actin remodeling. B cells from AKT2 KO mice exhibited defective spreading and BCR clustering upon stimulation in vitro. Disruption of Btk-mediated signaling caused the impaired differentiation of germinal center B cells, and the serum levels of both sepecific IgM and IgG were decreased in the immunized AKT2 KO mice. In addition, the actin remodeling was affected due to the decreased level of the activation of WASP, the actin polymerization regulator, in AKT2 KO mice as well. As a crucial regulator of both BCR signaling and actin remodeling during early activation of B cells, the phosphorylation of CD19 was decreased in the AKT2 absent B cells, while the transcription level was normal. Conclusions AKT2 involves in the humoral responses, and promotes the BCR signaling and actin remodeling to enhance the activation of B cells via regulating CD19 phosphorylation. Video Abstract

Due to the biomagnifying effect in the food chains, heavy metals will cause serious harm to the food produced in paddy soil, and then threaten human health. The remediation of soil heavy metals by the addition of amendments is a common method. However, the combination of the two amendments has been less studied and its effect is unknown. In this study, we investigated the effects of different concentrations of a lime and calcium-magnesium phosphate (CMP) amendments metal availability and paddy soil bacteria biodiversity. The experiment proves that the addition of 0.5 and 1.0‰ amendment can effectively reduce cadmium (Cd) availability and the cadmium content in rice to be below 0.2 mg/kg, meeting the national food safety level. The results demonstrate that increasing pH and phosphorous (P) in soil were two important factors decreasing available cadmium. Furthermore, biodiversity analysis of the treated soil showed that the amendment increased biodiversity. Proteobacteria and Chloroflex were the most abundant bacteria at the phylum level, followed by Acidobacterium and Nitrospirae. The abundance of Bacterodietes-vadinHA17, Syntrophaceae, and Thiobacillus increased as phosphorous increased. Cadmium passivation might induce those species.

With the rapid development of AC/DC hybrid microgrids and the widespread use of distributed power resources, planning strategies for microgrids with high-density distributed power generation have become an urgent problem. Because current research on microgrid planning has not considered line factors, this paper analyses the planning of an AC/DC hybrid microgrid based on an AC microgrid. The capacity and siting of the distributed power resources are optimized, taking into account the influence of the line investment cost and the interactive power upper limit on the planning results. In the proposed model, the objective is aimed at minimizing the sum of investment cost, load-loss economic cost, and system losses, taking into consideration power balance constraints and feeder number constraints. The commercial solver CPLEX is applied to attain the optimal distributed power capacity and site. The theoretical results are verified by an actual system.

Phenolics were extracted from grape peels, and the compounds of grape peel phenolics (GPP) were identified by LC-MS/MS. From a perspective of dietary nutrition, this study aimed to evaluate the effect of GPP on lipid accumulation in sodium palmitate (PA)-treated HepG2 cells, and further to explore its mechanisms. HepG2 cells were induced by 0.4 mmol/L PA as an in vitro model of lipid accumulation. The results showed that GPP could significantly reduce the TG level and ROS production in PA-treated HepG2 cells. RT-qPCR and Western blotting analysis indicated that GPP supplementation can downregulate the mRNA and protein expression of lipogenesis factors and upregulate the expression of lipolysis factors in PA-treated cells. GPP reduced lipid accumulation and oxidative stress by activating the AMPK/mTOR/AKT signaling pathways, and the most significant effect was detected at 250 μg/mL GPP. Thus, GPP as a dietary phenolic, have the function to regulate lipid metabolism.

Steam explosion, as a typical physical-chemical pretreatment technology, destroys the cell wall structure by instantaneous release of high-temperature and high-pressure steam, which promotes the release of plant active ingredients. Appropriate steam explosion pretreatment can significantly improve the extraction rate of free polyphenols, effectively promote the release of polyphenols, and improve the antioxidant capacity. In recent years, steam explosion was applied into the pretreatment of plants and their by-products, and it can significantly improve the extraction of natural active ingredients and enhance functional activity. The influence of steam explosion technology on the content and composition of bound polyphenols and free polyphenols in plants, and the influence on their antioxidant capacity in vitro are summarized in this paper. This would provide a guidance for the use of steam explosion in the food processing industry.

Living Phanerochaete chrysosporium mycelia were used to remove heavy metals of Cd(II), Cu(II) and Zn(II) in auqeous solution. The uptake of heavy metal by the mycelia was dependent on the environmental conditions. The optimum biosorption conditions of Cd(II), Cu(II), and Zn(II) were pH 5.5–6.5 at 37◦C, and 6 h. Under these conditions, the fungal biosorbent removed Cd(II), Cu(II), and Zn(II) rapidly and efficiently with maximum metal removal capacities of 59.77 mg/g, 74.78 mg/g, and 54.12 mg/g, respectively. The pseudo second-order kinetic model was superior to the pseudo first-order kinetic model, which indicated that the change of the surface sorption sites number was proportional to the square of the remaining unoccupied surface sites number. Fourier transform infrared (FT-IR) spectra indicated that hydroxyl and carboxyl groups were relevant to biosorption. Some crystal particles were found on the surface of the P. chrysosporium under scanning electron microscopy. Energy dispersive X-ray analysis and FT-IR revealed that amino acids and proteins were involved in binding metal ions. The results demonstrated that P. chrysosporium was a good potential biosorbent for adsorbing heavy metals. [ABSTRACT FROM AUTHOR]

Phanerochaete chrysosporium has been recognised as an effective bioremediation agent due to its unique degradation to xenobiotic and biosorption ability to heavy metals. However, few studies have focused on the simultaneous removal of heavy metals and organic pollutants. The aim of this work was to study the feasibility of simultaneous cadmium removal and 2,4-dichlorophenol (2,4-DCP) degradation in P. chrysosporium liquid cultures. The removal efficiencies were pH dependent and the maximum removal efficiencies were observed at pH 6.5 under an initial cadmium concentration of 5 mg/L and an initial 2,4-DCP concentration of 20 mg/L. The removal efficiencies for cadmium and 2,4-DCP reached 63.62% and 83.90%, respectively, under the optimum conditions. The high production levels of lignin peroxidase (7.35 U/mL) and manganese peroxidase (8.30 U/mL) resulted in an increase in 2,4-DCP degradation. The protein content decreased with increasing cadmium concentration. The surface characteristics and functional groups of the biomass were studied by scanning electron microscopy and a Fourier-transformed infrared spectrometer. Borago officinalis is a kind of vegetable and traditional medicinal herb in the Mediterranean region, and now it has spread many areas around the world. In this study, we sequenced a complete chloroplast (cp) genome sequence of B. officinalis to investigate its phylogenetic relationship with other species. The whole cp genome of B. officinalis is 149,835 bp in length with 37.82% overall GC content, including a large single-copy (LSC) region of 78,840 bp and a small single-copy (SSC) region of 16,967 bp, which are separated by a pair of inverted repeats (IRs) of 27,014 bp. The cp genome contains 110 genes, including 76 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. The phylogenetic analysis based on cp genome sequences showed that B. officinalis is closely related to Antirhea chinensis, Mitragyna speciosa and Neolamarckia cadamba.

Polyphenols, including extractable polyphenols (EPP) and non-extractable polyphenols (NEPP), are natural and secondary metabolic substances in plants that have beneficial properties to human health. However, NEPP associated with dietary fiber and protein are not taken into account in most literature data. In this paper, NEPP were released from blueberries with acid or alkaline hydrolysis methods, and the related extraction conditions were determined and optimised by response surface methodology (RSM). The results showed that NEPP yield obtained with alkaline hydrolysis was much higher than that obtained with acid treatment. The NEPP yield in alkaline hydrolysis process was significantly affected by the NaOH concentration and liquid/solid ratio, while in the acid hydrolysis process, the NEPP yield was significantly affected by the temperature, time and liquid/solid ratio. The second order polynomial models were developed for predicting NEPP content in blueberries. The optimisation of the extraction process of NEPP in blueberries would provide a good idea and basis for the application of non-extractable fractions.

The total polyphenol (TP), flavonoid, proanthocyanidin (PAC) content, and antioxidant capacity of both onion varieties (red and yellow) were compared. The content of TP, flavonoids, and PAC was determined by Folin-Ciocalteu colorimetric method, AlCl3, and by DMAC colorimetric method, respectively. The results showed that the contents of TP and flavonoids decreased from the outer to the inner layers in both onions, but there was no significant difference in PAC content. The outer layers had the highest antioxidant activity of extracts followed by a continuous decrease towards the inner layers in both varieties. The contents of phenolic acids and flavonoids were quantified by HPLC. Gallic acid, ferulic acid, and quercetin, as the main compounds in polyphenols, were detected in each layer of both onions. The red variety showed better antioxidant activity than yellow onion according to the linoleic acid system and DPPH assay. The higher contents of TP and flavonoids were associated with higher antioxidant activity.

The contents of extractable polyphenols (EPP), non-extractable polyphenols (NEPP), extractable proanthocyanidins (EPAC), non-extractable proanthocyanidins (NEPAC) and extractable anthocyanidins (EAC) in blueberry leaves were investigated. An experiment was conducted to analyse the effect of solvent types (methanol and ethanol), concentration (50 and 70%) and pH (2 and 6) on the extraction of bioactive compounds. Total extractable polyphenols (TEPP), total extractable proanthocyanidins (TEPAC), and total extractable anthocyanidins (TEAC) were analysed in methanol/ethanol/water extracts, NEPP were determined in acidic hydrolysates and NEPAC were quantified by depolymerisation in HCl/butanol. The results showed that ethanol and methanol did not affect the extraction of TEPP, while methanol was better for TEPAC and TEAC. The contents of TEPP and TEAC in 50% solvent were higher than those in 70% solvent. When solvent pH dropped to 2, the contents of EPP and EPAC were increased, while the EAC content was decreased.

Traditional three-domain fungal and bacterial laccases have been extensively studied for their significance in various biotechno-logical applications. Growing molecular evidence points to a wide occurrence of more recently recognized two-domain laccase-like multicopper oxidase (LMCO) genes in Streptomyces spp. However, the current knowledge about their ecological role and distribution in natural or artificial ecosystems is insufficient. The aim of this study was to investigate the diversity and composition of Streptomyces two-domain LMCO genes in agricultural waste composting, which will contribute to the understanding of the ecological function of Streptomyces two-domain LMCOs with potential extracellular activity and ligninolytic capacity. A new specific PCR primer pair was designed to target the two conserved copper binding regions of Streptomyces two-domain LMCO genes. The obtained sequences mainly clustered with Streptomyces coelicolor, Streptomyces violaceusniger, and Streptomyces griseus. Gene libraries retrieved from six composting samples revealed high diversity and a rapid succession of Streptomyces two-domain LMCO genes during composting. The obtained sequence types cluster in 8 distinct clades, most of which are homologous with Streptomyces two-domain LMCO genes, but the sequences of clades III and VIII do not match with any reference sequence of known streptomycetes. Both lignocellulose degradation rates and phenol oxidase activity at pH 8.0 in the composting process were found to be positively associated with the abundance of Streptomyces two-domain LMCO genes. These observations provide important clues that Streptomyces two-domain LMCOs are potentially involved in bacterial extracellular phenol oxidase activities and lignocellulose breakdown during agricultural waste composting.
