Mutation analysis of epidermal growth factor receptor (EGFR) gene is essential for treatment selection in non-small cell lung cancer (NSCLC). Analysis is usually performed in tumor samples. We evaluated the clinical utility of EGFR analysis in plasma cell-free DNA (cfDNA) from patients under treatment with EGFR inhibitors. We selected 36 patients with NSCLC and EGFR-activating mutations. Blood samples were collected at baseline and during treatment with EGFR inhibitors. Wild-type EGFR, L858R, delE746-A750, and T790M mutations were quantified in cfDNA by droplet digital PCR. Stage IV patients had higher total circulating EGFR copy levels than stage I (3523 vs. 1003 copies/mL; p < 0.01). There was high agreement for activating mutations between baseline cfDNA and tumor samples, especially for L858R mutation (kappa index = 0.679; p = 0.001). In 34 % of advanced NSCLC patients, we detected mutations in cfDNA not previously detected in tumor samples and double mutations in 17 %. Patients with baseline total EGFR copy levels above the median presented decreased overall survival (OS) (341 vs. 870 days, p < 0.05) and progression-free survival (PFS) (238 vs. 783 days; p < 0.05) compared with those with total EGFR copy levels below the median. Patients with baseline concentrations of activating mutations above the median (94 copies/mL) had lower OS (317 vs. 805 days; p < 0.05) and PFS (195 vs. 724 days; p < 0.05). During follow-up, T790M resistance mutation was detected in 53 % of patients. Total and mutated EGFR analysis in cfDNA seems a relevant tool to characterize the molecular profile and prognosis of NSCLC patients harboring EGFR mutations. [ABSTRACT FROM AUTHOR]

The aim of this paper is to estimate efficiently insert wear in metal machining and to improve tool replacement operations. Image processing and classification are used to automatize the decision making about the adequate time for tool replacement. Specifically, the shape descriptor aZIBO (absolute Zernike moments with Invariant Boundary Orientation) has been used to characterize insert wear and ensure its optimal usage. A dataset composed of 577 regions with different levels of wear has been created. Two different classification processes have been carried out: the first one using three different classes (Low, Medium and High wear -L, M and H, respectively-) and the second one with just two classes: Low (L) and High (H). Classification was carried out using on the one hand kNN with five different distances and five values of k and, on the second hand, a Support Vector Machine (SVM). aZIBO performance has been compared with classical shape descriptors such as Hu and Flusser moments. It outperforms them, obtaining success rates up to 91.33% for the L-H classification and 90.12% for the L-M-H classification. [ABSTRACT FROM AUTHOR]

In this paper, an automatic process to determine tool wear in machining procedures has been developed using computer vision and texture recognition techniques. Two different methods based on Local Binary Pattern (LBP) were evaluated combined with the LOSIB texture booster (Local Oriented Statistical Information Booster). The dataset used is composed by 577 images representing different wear of inserts. Two classifications were carried out: (i) a binary classification with Low-High discrimination and (ii) a ternary classification with Low-Medium-High discrimination. The results show that when combining LBP with LOSIB, all the other methods are outperformed featuring an 80.58% of accuracy in the binary classification and a 67.76% in the ternary classification. These results are very interesting for industry due to the possible savings in terms of cost and time if it is applied in a tool condition monitoring system (TCMS). [ABSTRACT FROM AUTHOR]

This paper presents a study about the identification of cutting tool wear state by means of vibration signal analysis in steel dry turning operations. Analyzing the RMS value evolution and FFT frequency spectra of signal an on-line tool condition monitoring system has been developed. The main purpose is to determine the instant from which tool condition is considered unacceptable affecting machining process quality. After the analysis was carried out, the main conclusion is that both the RMS and frequency amplitude ranges of certain spectrum bands are related to tool wear. [ABSTRACT FROM AUTHOR]

Thirteen dogs with nictitans plasmocytic conjunctivitis were treated with 2.0 per cent cyclosporin drops in the right eye and with 0.1 per cent dexamethasone ointment in the left eye. The response to both therapies was monitored for six weeks, repeat biopsy specimens were taken, and the time for the clinical signs to recur recorded. Conjunctival cultures were taken before and after both therapies. There were no significant differences between the treatments in the remission of clinical signs, the reduction of inflammatory infiltrate in the biopsy specimens, or the time to recurrence of the condition or its subsequent severity. However, the eyes treated with 0.1 per cent dexamethasone tended to recover more rapidly than the eyes treated with 2.0 per cent cyclosporin, and the eyes treated with 2.0 per cent cyclosporin tended to be protected from a recurrence for longer than the eyes treated with 0.1 per cent dexamethasone. The article discusses a case of mycotic blepharitis in a bubo or Eurasian eagle owl. The bird had received topical ophthalmic treatment in the right eyelid every 12 hours which was ineffective. The lesion had spread from the lower eyelid to the upper eyelid. Skin scrapings from eyelid were examined for bacterial and fungal organisms. A mycelial colony was isolated and identified as Fusarium proliferatum.

Purpose The iridocorneal endothelial (ICE) syndrome is a rare proliferative corneal endotheliopathy of unknown etiology. Common manifestations include corneal edema,iris stromal abnormalities, peripheral anterior synechiae and glaucoma, which does not usually respond to medical treatment. We want to know what is the most successful surgical practice for the control of intraocular presure (IOP) in these patients. Methods We presents a case of man of 27 years with ocular hypertension in the right eye (RE) difficult to control for 6 months. AV 8/10 in his RE. Iris atrophy with corectopia, uveal ectropion and peripheral anterior synechiae were observed in RE. Abnormalities of angle and endothelial disruption without corneal edema led to a diagnosis of ICE syndrome. We performed an Ahmed valve implant in that eye. Two months after surgery the IOP remains controlled with topical beta‐blocker. Results The ICE syndrome is usually unilateral disorder that can develop glaucoma resistant to medical treatment. There is a lack of consensus on the surgical technique of choice. It is described a higher survival rate of valve implants on trabeculectomy at 3 and 5 years as well as higher rates of failure than other glaucoma, due to endothelialization of the fistula. The use of Mitomycin would be indicated in these cases. Conclusion When is not possible to perform a classical filtering surgery due to angle morphological characteristics, the placement of a valve implant should be considered. [ABSTRACT FROM AUTHOR]

Purpose To realize a retrospective study on the glaucoma post keratoplasty glaucoma Between 20‐30% of patients undergoing a cornea transplant, require medical or surgical treatment, antihypertensive. Methods A retrospective study of 50 penetrating keratoplasty. The variables analyzed included etiology of graft, control intraocular pressure (IOP) per week, 1, 3, 6 months and one year after surgery, number and type of treatments, and patient gonioscopic study. Results The most common etiology implicated as a cause of keratoplasty was bullous keratopathy (50%), followed by herpes (15%). Triple procedure was performed in 50% (25 cases) followed by keratoplasty simple 30% (15 cases). In 60% of cases there normotensional behavior at 8 months follow‐up, showing the 12% of patients, an associated hypotensive treatment and in 6% the use of implants for blood pressure control valve. Conclusion The triple procedure is shown as the safest, with a good blood pressure control per year, using the topical betobloqueantes use as first choice therapy. In refractory cases, the use of drainage systems, achieves blood pressure control in most of our cases. [ABSTRACT FROM AUTHOR]

Selecting the optimal sperm population is essential for success with reproductive techniques. Porcicoll (formerly Androcoll-P) is a colloid formulation for selection of high-quality boar spermatozoa by single layer centrifugation (SLC). To date, most studies have been carried out with fresh semen and large volumes. We carried out 2 experiments to test the use of Porcicoll for thawed boar semen in small volumes. In Experiment 1, cryopreserved semen doses were thawed, split in 200-μL aliquots and layered on 1 mL of Porcicoll 70%, 80% or 90%, or buffer without colloid. We assessed sperm recovery (the proportion of the loading dose that appeared in the pellet, %), and the physiology of the selected spermatozoa (flow cytometry: Viability, apoptotic changes, capacitation, mitochondrial activity, intracellular reactive oxygen species). The most suitable proportion was Porcicoll 80%, allowing acceptable sperm recovery (16.9 ± 4.2%, compared to 70% (35.4% ± 3.0, p < 0.001) and 90% (8.2% ± 3.0, P = 0.001), and improved quality (mitochondrial activity: Porcicoll 80%: 77.7 ± 1% vs Control: 60.3 ± 0.7%, P < 0.05). In Experiment 2, we compared 3 supplements to Porcicoll 80%: 500 mM reduced glutathione (GSH), 20% seminal plasma (SP) and 0.5% bovine serum albumin (BSA). Supplementation with GSH or BSA did not cause relevant changes relative to Control. In contrast, SP induced membrane and acrosomal changes resembling capacitation, which might preclude its use in some applications, and decreased recovery (5.5% ± 1.9 vs. 24.3% ± 1.2 Control; P < 0.001). However, it could be useful prior to other applications such as in vitro fertilisation. Overall, Porcicoll is an effective colloid for isolating a high-quality population from thawed boar sperm, 80% being a balanced option for good recovery and high quality.
