Les inhibiteurs de JAK (JAKi) sont utilisés dans la PR en pratique courante depuis maintenant plusieurs années, avec une arrivée progressive sur le marché de 4 molécules. L'utilisation des JAKi a été également freinée par différentes alertes successives de tolérance ayant restreint leur utilisation chez des patients âgés ou porteurs de facteurs de risque. L'objectif de ce travail était d'étudier le maintien thérapeutique des 4 JAKI disponibles ainsi que leur efficacité et tolérance en pratique courante dans la PR. Étude rétrospective monocentrique ayant inclus l'ensemble des patients atteints de PR ayant reçu un premier JAKI entre avril 2018 et juin 2022. Le critère de jugement principal était l'analyse du maintien thérapeutique à 12 et 24 mois (courbe de survie de Kaplan-Meyer). Les critères secondaires étaient 1/l'efficacité des JAKi et de chacune des molécules à 6 mois et lors de la dernière visite disponible (jusqu'en juin 2024) évaluée sur le score DAS28-CRP (test des rangs signés de Wilcoxon) et 2/le recueil des arrêts de traitement pour inefficacité et intolérance. Nous avons inclus 108 patients (91 femmes, 84 %) traités par JAKi avec un âge moyen de 57 ± 14 ans et une durée de la maladie de 19 ± 12 ans ; les facteurs rhumatoïdes et les ACPA étaient positifs chez 91 (84 %) et 95 (88 %) patients respectivement, 83 (77 %) présentaient des érosions et 37 (34 %) des atteintes extra-articulaires (Tableau 1). Les JAKi ont été majoritairement utilisés en association à un csDMARD (83/108, 77 %) et au-delà de la 3e ligne de traitement ciblé (Tableau 1). Les caractéristiques des patients traités par les différents JAKi étaient globalement comparables et sont présentées dans le Tableau 1. Le maintien thérapeutique des JAKi était de 72 % à 12 mois et de 61 % à 24 mois. La durée médiane de survie du traitement était de 47 mois (Fig. 1). Le DAS28-CRP a été significativement réduit à la première visite (−1,46 ± 0,28 par rapport à la visite d'inclusion, p < 0,001) et à la dernière visite ayant eu lieu en moyenne 21 ± 15 mois après l'initiation du JAKi (−1,05 ± 1,67 par rapport à la visite d'inclusion, p < 0,001) (Tableau 2). Cette diminution du DAS28-CRP semble être numériquement plus importante avec les JAKi plus sélectifs (upadacitinib, filgotinib) qu'avec le baricitinib et le tofacitinib. Quarante-cinq patients (42 %) ont interrompu le traitement par JAKi au cours du suivi (16 baricitinib, 53 %, 6 tofacitinib, 60 %, 21 upadacitinib, 44 %, 2 filgotinib, 10 %), dont 26 par manque d'efficacité et 15 pour intolérance. Parmi les effets secondaires d'intérêt, on notait 1 infarctus du myocarde, 3 embolies pulmonaires et la découverte d'un cancer du sein. Le maintien thérapeutique des JAKi dans la PR à 12 et 24 mois dans cette population était respectivement de 72 % et 61 %. Utilisés majoritairement au-delà de la 3e ligne de traitement ciblé, les JAKi permettent d'améliorer significativement l'activité de la PR. Néanmoins, une proportion significative de patients a interrompu le traitement en raison d'un manque d'efficacité ou d'effets secondaires, soulignant l'importance d'une évaluation continue et personnalisée de ces thérapies en pratique clinique courante. L'ochronose ou l'alcaptonurie est une maladie autosomique récessive rare. Elle est due à un déficit de l'acide homogentisique-oxydase conduisant à l'accumulation d'acide homogentisique dans les tissus. Nous rapportons l'observation exceptionnelle de 3 patients issus d'une même famille de parents cosanguins ayant cette maladie héréditaire. Observation 1 : patiente O.F. âgée de 66 ans a présenté une raideur rachidienne lombaire, une limitation de la mobilité des ceintures scapulaire et pelvienne avec d'une boiterie. Une coloration bleuâtre des pavillons des oreilles, des angles internes, externe des yeux et des ongles a été constaté durant l'hospitalisation. L'imagerie a mis en évidence un « vide » et calcifications discales lombaires étagées (Fig. 1), des ponts ostéophytiques rachidien exubérants, une coxite bilatérale protrusive non opérable et une omarthrose avancée à droite. Le diagnostic d'ochronose a été posé sur des critères clinico-radiologiques. Durant l'hospitalisation et voyant l'intérêt accordé à son cas la malade rapporte que sa frangine a la même symptomatologie aux hanches et qu'elle a des difficultés à marcher, la sœur est convoquée pour hospitalisation... Observation 2. Patiente O.S. âgée de 62 ans, a présenté un tableau similaire avec une limitation de tous les mouvements des hanches, un dépôt de pigment bleuâtre sclérotique amarqué l'investigation diagnostique. Le test de coloration urinaire à la lumière s'est avéré positif, et la radiographie a montré une coxite bilatérale traitée par prothèse totale de hanche. Après quelques jours d'hospitalisation, la maman des 2 sœurs nous rapporte qu'un de ces fils présentait une coloration noir-bleuâtre de ses urines au moment du change lorsqu'il était enfant, le frère est convoqué en consultation pour investigation.... Observation 3. Dans un cadre de dépistage de cette famille particulière, le patient O.R. âgé de 50 ans totalement asymptomatique sur le plan rhumatologique a bénéficié d'une investigation radiologique révélant une calcification discale pathognomonique confortant le diagnostic de ce rhumatisme métabolique. Dans notre observation, les dommages structuraux été à un stade avancé, les sœurs étaient symptomatiques avec limitation de la mobilité des hanches, leur frère quant à lui est resté longtemps asymtomatique. L'ochronose est souvent révélée à l'âge adulte, principalement après la quatrième décennie. L'assombrissement urinaire est un signe annonciateur de la maladie, mais il est rarement révélateur de la maladie. Les manifestations cutanées représentent des signes d'alerte. L'arthropathie ochronotique est insidieuse mais peut être handicapante. Il n'existe pas de traitement médical spécifique de cette pathologie. Cette observation est originale de part le mode de révélation, du nombre de membres atteints de la même famille et surtout de part le retard diagnostic de l'atteinte rhumatismale révélée à un stade tardive. L'alcaptonurie est à l'origine d'une arthropathie dégénérative pouvant mettre en jeu le pronostic fonctionnel. Le dépistage par analyse moléculaire et l'étude génétique prennent un grand intérêt dans le diagnostic de certitude de cette maladie. Cette observation nous a permis de voir que même dans le cas d'une ochronose familiale, le pronostic de chaque membre reste différent et non prévisible. [ABSTRACT FROM AUTHOR]

Les maladies inflammatoires chroniques intestinales (MICI) sont associées à un surrisque d'ostéopathie fragilisante (OF) dont le diagnostic est basé sur la densitométrie osseuse (DMO). Afin de s'affranchir des contraintes de la DMO, plusieurs techniques de mesure opportuniste sont à l'étude, notamment, le Vertebral Quality Score (VBQ) développé à partir d'IRM rachidiennes. Le but de notre étude est de mesurer le VBQ afin de dépister une OF chez des patients souffrant de MICI. Ont été inclus les patients consécutifs atteints de MICI ayant eu une DMO et une IRM du rachis lombaire entre 2017 et 2023. Les mesures des scores VBQ étaient réalisées sur la séquence T1. Le critère de jugement principal était la corrélation entre la valeur du VBQ médian et le diagnostic d'une OF définie par la présence d'une ostéopénie ou d'une ostéoporose. Les critères secondaires évaluaient la corrélation entre le VBQ-L3 et le statut osseux en DMO. Les valeurs d'ostéodensitométrie et les scores VBQ ont aussi été analysés comme variables quantitatives et leur corrélation calculée par le test de Pearson. La performance du VBQ était représentée par courbes ROC, sa reproductibilité par le coefficient de corrélation intra-classe. Au total, 302 patients (65 % de femmes, âge moyen de 47 ans) ont été inclus : 121 (40 %) avaient une ostéopénie et 32 (11 %) une ostéoporose. Les comorbidités et traitements concomitants sont présentés dans le Tableau 1. On observait une augmentation significative du VBQ médian dans le groupe ostéopathie fragilisante comparé aux patients sans OP : 2,47 vs 2,19, respectivement ; p < 0,001. Le VBQ médian était significativement plus élevé chez les patients sans ostéopathie comparé à ceux ayant une ostéoporose et une ostéopénie : 2,67 et 2,42, respectivement ; p < 0,001 (Tableau 2). L'analyse des courbes ROC (AUC : 0,63 [0,55–0,68]) a montré qu'un VBQ médian > 3,1 avait une spécificité de 95 % pour le diagnostic d'ostéopathie fragilisante et une sensibilité faible (12 %) alors qu'une valeur < 2 avait une sensibilité à 77 %. Il existait une corrélation négative entre les valeurs de VBQ et les valeurs densitométriques en g/cm2, modérée au col fémoral (r = −0,38, p < 0,001), et faible au rachis lombaire (r = −0,24, p < 0,001). Enfin, le coefficient de corrélation intra-classe était bon à 0,82 (p < 0,0001). La mesure du VBQ pourrait avoir un intérêt dans le dépistage opportuniste de l'OP chez les patients atteints de MICI et guider la prescription d'une DMO, gold standard pour le diagnostic d'OP. La polyarthrite rhumatoïde (PR) est caractérisée par un paradoxe lipidique induit par l'inflammation systémique. Les inhibiteurs de JAK (JAKi) sont associés à une augmentation à court terme du cholestérol total (CT) et des fractions LDL et HDL qui pourrait être un effet direct du médicament ou indirect, secondaire à la réduction de l'inflammation systémique induite par le traitement. L'objectif de ce travail était d'étudier l'évolution des taux de CT et de ses fractions après introduction d'un JAKi. Étude rétrospective monocentrique incluant l'ensemble des patients atteints de PR mis sous JAKi entre 2019 et 2023, ayant au moins deux visites de suivi et un bilan lipidique disponible à l'inclusion, et non traités par statines lors de l'inclusion. Le critère de jugement principal était l'évolution du CT et de ses fractions à 6 mois du début des JAKi. Les critères secondaires comprenaient l'étude des facteurs associés aux modifications du bilan lipidique à 6 mois et l'évolution du bilan lipidique entre la première et la dernière visite disponible. Au total, 85 patients atteints de PR (68 femmes, 80 %) répondaient aux critères d'inclusion, avec un âge moyen de 59 ± 13 ans et une durée moyenne de la maladie de 19,5 ± 12,4 ans ; 73 (86 %) et 76 (89 %) patients avaient des facteurs rhumatoïdes et des ACPA positifs respectivement, et 66 (78 %) présentaient des érosions osseuses. 22 patients recevaient du baricitinib, 7 du tofacitinib, 38 de l'upadacitinib et 18 du filgotinib. Le DAS28-CRP à l'inclusion était de 4,09 ± 1,26 avec une CRP moyenne de 10 ± 14 mg/L. Les comorbidités, les traitements concomitants et le bilan lipidique à l'inclusion sont présentés dans les Tableaux 1 et 2. À l'inclusion, les taux de CT et de HDL cholestérol (HDL-C) corrélaient négativement avec la CRP (respectivement r = –0,23, p = 0,042 et r = –0,25, p = 0,040), mais pas avec le DAS28 ou le DAS28-CRP. Cette corrélation n'était pas mise en évidence pour le LDL et les triglycérides. À 6 mois, on notait une augmentation significative du CT (+0,17 ± 0,37 g/L, p = 0,016) du HDL-C (+0,09 ± 0,19 g/L, p = 0,017) mais pas du LDL-C (+0,02 ± 0,31, p = 0,68) (Tableau 2). Cette augmentation était inversement corrélée à la réduction des concentrations de CRP (–6 ± 10 mg/L) sous JAKi (CT : r = –0,31, p = 0,033 et HDL-C : r = –0,32, p = 0,042). Cette augmentation n'était ni associée à la prise de corticoïdes ou de méthotrexate, ni à l'évolution de l'activité articulaire de la maladie (nombre d'articulations douloureuses et gonflées). Lors de la dernière visite disponible (en moyenne 20 ± 15 mois après le début du JAKi), il existait une baisse du CT et notamment de sa fraction HDL par rapport à la première visite, avec des valeurs proches de celles de la visite d'inclusion (Tableau 2). Des statines ont été prescrites pour 9 patients au cours de la période de suivi, qui présentaient tous une dyslipidémie connue à l'inclusion (10,6 %). Cette étude confirme une augmentation transitoire du CT et de ses fractions sous JAKi liée à la réduction de l'inflammation systémique induite par le traitement. Ces anomalies lipidiques ont été régressives chez la majorité des patients. Trachurus trachurus (Linnaeus, 1758) is a semipelagic fish common in the waters of the Moroccan Mediterranean and is of major economic importance because it occupies an important place in fish landings. The population of the eastern coast was monitored during the period from October 2017 to September 2018 using monthly sampling. This work focused on the study of the growth biology of this species. In total, 390 specimens were sampled. The total lengths of the individuals collected varied between 7.8 cm and 33.8 cm for a weight that varied from 3.78 g to 310.52 g. The study of the growth of this Carangidae and the estimation of its age are based on the reading of the otoliths. The increase in length follows the linear Von Bertalanffy model; thus, it is rapid for juveniles and decreases with age. Similar growth in length and differential growth in weight were demonstrated between the two sexes. This difference is apparent only from the fourth year onwards and remains in favour of females. The results obtained constitute a valuable contribution to a better understanding of Trachurus trachurus biology and to the implementation of stock assessments to ensure the sustainability of catches of this species.

The electrografting of aryl-diazonium was proven to be an efficient way to immobilize antibodies, oligonucleotides and enzymes onto conductive supports. Biomolecules chemically functionalized with aniline derivates or diazotated derivates could be used to build complex architectures acting as sensing layers for biosensors, biochips or others bioelectronic devices. Additionally, the use of SPR offers new opportunities to characterize the grafted surfaces as well as to develop label-free assays. Here, we give an overview of our group achievements in the field during the last 7 years. We highlight the applications of these functionalized surfaces in multiparametric sandwich assays, in the label free detection and imaging of macromolecular interactions by SPRi and in the immobilization of dehydrogenase via its cofactor. Ce travail a pour objectif d'analyser les facteurs qui influent sur le comportement d'achat en ligne des consommateurs tunisiens et qui pourraient être l'un des plus importants enjeux du commerce électronique et du marketing. Pour réaliser cet objectif, nous nous sommes basées sur un modèle examinant l'impact des risques perçus, l’utilité attendue, l’expérience d’achat et la confiance envers les boutiques en ligne sur l’attitude envers l'achat en ligne ; et la relation entre cette attitude et l’intention de commander par Internet. Partant d’une riche revue de littérature, nous avons fixés 7 hypothèses de recherche dans le but d’examiner le lien entre l’intention d’achat en ligne et les facteurs explicatifs. Near-infrared (NIR) luminescence and photoacoustic (PA) imaging have attracted increasing attention for the real-time monitoring of biological samples due to high sensitivity, resolution, and pronounced signal detection depth, respectively. For improved contrast, both techniques require imaging agents possessing high absorption in the red-NIR range. Herein, we took advantage of a ternary complex formed with the anionic ytterbium(III) tetrakis (2-thenoyltrifluoroacetonate) ([Yb(tta) 4 ] - ) and the cationic NIR-absorbing chromophore, 1,1'-diethyl-2,2'-dicarbocyanine (Cy + ), to evaluate its potential to act as a dual-mode NIR luminescence and PA imaging agent. We demonstrated that, upon excitation with red-NIR light, Cy[Yb(tta) 4 ] encapsulated into polystyrene nanoparticles is able to generate both NIR Yb 3+ emission and a PA signal in an imaging experiment performed in a tissue-mimicking phantom.

Lignin is the most important natural source of aromatic compounds. The valorisation of lignin into aromatics requires fractionation steps that can be catalysed by ligninolytic enzymes. However, one of the main limitations of biological lignin fractionation is the low efficiency of biocatalysts; it is therefore crucial to enhance or to identify new ligninolytic enzymes. Currently, the screening of ligninolytic activities on lignin polymers represents a technological bottenleck and hinders the characterization and the discovery of efficient ligninolytic biocatalysts. An efficient and fast method for the measurement of such enzymatic activities is therefore required. In this work, we present a new electrochemical tool based on lignin-coated paper electrodes for the detection and the characterization of ligninolytic activity. The European hake, Merluccius merluccius (Linnaeus, 1758), is one of the most important resources for Mediterranean fisheries. Due to its pivotal role in energy transfer from lower to higher trophic levels, this species is a crucial component of the ecosystem's functioning. The ecological role of Merluccius merluccius , off the Moroccan Mediterranean Sea (southern Alboran Sea), was investigated, exploring seasonal and ontogenetic shifts, geographical variations in prey composition, and feeding strategy. Between November 2020 and July 2022, a total of 402 hake specimens were collected by oceanographic bottom trawl surveys (MEDITS) that were carried out during warm and cold seasons to assess their diet and feeding habits. The sample was analyzed according to fish sizes and seasons, and qualitative/quantitative feeding indices were calculated. The trophic spectrum of Merluccius merluccius included 24 prey items in total, mainly belonging to Osteichthyes (12), Crustacea (10), Cephalopoda (1), and Polychaeta (1), suggesting a generalist behavior of this predator as in numerous regions of the Mediterranean Sea, with several species that occasionally occurs in its diet. In the Moroccan Mediterranean Sea, Osteichthyes proved to be the most important prey item (%IRI = 78.56) among the different zoological groups, followed by Crustacea (%IRI = 16.22). The other food items were occasionally and randomly consumed, and cannibalism was low (0.8%). Hierarchical cluster analysis and nonmetric multidimensional scaling (nMDS) showed different feeding habits of two main groups separated at 60% similarity: small specimens <10 cm TL, primarily feed on zooplanktonic prey, while medium and large specimens hold a diet based on Osteichthyes with crustaceans. Furthermore, a significant positive relationship between hake and fish prey size was confirmed. Seasonally, mesopelagic Osteichthyes were the main food prey in the summer season, while pelagic species were predominant during the autumn. SIMPER analysis revealed that the prey items contributing the most to the differences between seasons and length classes were Engraulis encrasicolus , Micromesistius poutassou , Boops boops , Macroramphosus scolopax , gobids, Gadiculus argenteus , and most of Crustacea . The diet does not appear to be influenced by sex (>0.05). A trophic level (TROPH) of 4.1 was calculated, indicating that the species is a top predator (quaternary consumers). The TROPH values ranged between 2.58 and 4.38 from juveniles to adults, increasing asymptotically with the size of specimens. In contrast to what has previously been found in other Mediterranean regions, where ichthyophagous hake feed mostly on pelagic Osteichthyes , such as Engraulis encrasicolus, Sardina pilchardus , and Micromesistius poutassou , the study points up the vital role played by Atlantic horse mackerel Trachurus trachurus for hake diet in the Moroccan Mediterranean Sea. Background: Satisfaction of fundamental needs is an important concept in sport, but currently there is no tool in Arabic to measure this construct. Basic needs are often linked to high rates of motivation and performance. It is necessary to develop tools to assess psychological needs in the sport context., Aim: This study aimed to validate the Basic Needs Satisfaction in Sport Scale (BNSSS) in Arabic language across Tunisian athletes, and to test its psychometric properties (factorial structure, internal reliability, construct validity, and sensitivity)., Methods: Athletes in various sports participated in this study (370 men, 146 women; mean age 18.35) and voluntarily completed the Arabic version of the BNSSS-20. Both exploratory (EFA, N = 294; males: 68%; females: 32%; [14-18] = 182; [19-28] = 112) and confirmatory (CFA; N = 222; males: 76.6%; females: 23.4%; [14-18] = 103; [19-28] = 119) factor analyses were examined., Results: Results from the EFA suggest that the BNSSS scale reflects the theoretical model well, with good internal consistency for all factors. All 20 items of BNSSS revealed excellent reliability (McDonald's omega = 0.773, Cronbach's α = 0.886, Gutmann's λ6 = 0.970) and good temporal stability (ICC = 0.84, 95% CI = 0.55-0.93) over a 4-week period. Background: Pectus arcuatum is often mistaken for a type of pectus carinatum. However, pectus arcuatum is a unique clinical form of pectus caused by premature obliteration of the sternal sutures (manubrial sternum, four sternebrae and xiphoïd process), whereas pectus carinatum is due to abnormal growth of the costal cartilage. In order to better describe pectus arcuatum, we analysed the files of patients with pectus arcuatum followed in our centers., Methods: Multicenter retrospective study of young patients' files diagnosed with pectus arcuatum., Results: The clinical diagnosis of pectus arcuatum was made in 34 patients with a mean age at diagnosis of 10.3 years (4-23 years). A chest profile X-ray or a CT scan was performed in 16 patients (47%) and confirmed the diagnosis of PA by the presence of a sternal fusion. It was complete in 12 patients. A malformation was associated in 35% of cases (Noonan syndrome 33%, scoliosis 25% or cardiopathy 16%). 11 patients (32%) had a family history of skeletal malformation. Orthopedic treatment was initiated in 3 patients without any success. 11 patients underwent surgical correction, which was completed in 7 of them., Conclusion: The diagnosis of pectus arcuatum is based on clinical experience and if necessary, on a profile chest X-ray showing the fusion of the sternal pieces. It implies the search for any associated malformations (musculoskeletal, cardiac, syndromic). Bracing treatment is useless for pectus arcuatum. Background: Becker muscular dystrophy (BMD) is an X-linked disorder characterized by slow, progressive muscle damage and muscle weakness. Hallmarks include fibre-size variation and replacement of skeletal muscle with fibrous and adipose tissues, after repeated cycles of regeneration. Muscle histology can detect these features, but the required biopsies are invasive, are difficult to repeat and capture only small muscle volumes. Diffusion-tensor magnetic resonance imaging (DT-MRI) is a potential non-invasive alternative that can calculate muscle fibre diameters when applied with the novel random permeable barrier model (RPBM). In this study, we assessed muscle fibre diameters using DT-MRI in BMD patients and healthy controls and compared these with histology., Methods: We included 13 BMD patients and 9 age-matched controls, who underwent water-fat MRI and DT-MRI at multiple diffusion times, allowing RPBM parameter estimation in the lower leg muscles. Tibialis anterior muscle biopsies were taken from the contralateral leg in 6 BMD patients who underwent DT-MRI and from an additional 32 BMD patients and 15 healthy controls. Laminin and Sirius-red stainings were performed to evaluate muscle fibre morphology and fibrosis. Twelve ambulant patients from the MRI cohort underwent the North Star ambulatory assessment, and 6-min walk, rise-from-floor and 10-m run/walk functional tests., Results: RPBM fibre diameter was significantly larger in BMD patients (P = 0.015): mean (SD) = 68.0 (25.3) μm versus 59.4 (19.2) μm in controls. Inter-muscle differences were also observed (P ≤ 0.002). Both inter- and intra-individual RPBM fibre diameter variability were similar between groups. Laminin staining agreed with the RPBM, showing larger median fibre diameters in patients than in controls: 72.5 (7.9) versus 63.2 (6.9) μm, P = 0.006. However, despite showing similar inter-individual variation, patients showed more intra-individual fibre diameter variability than controls-mean variance (SD) = 34.2 (7.9) versus 21.4 (6.9) μm, P < 0.001-and larger fibrosis areas: median (interquartile range) = 21.7 (5.6)% versus 14.9 (3.4)%, P < 0.001. Despite good overall agreement of RPBM and laminin fibre diameters, they were not associated in patients who underwent DT-MRI and muscle biopsy, perhaps due to lack of colocalization of DT-MRI with biopsy samples., Conclusions: DT-MRI RPBM metrics agree with histology and can quantify changes in muscle fibre size that are associated with regeneration without the need for biopsies. Skeletal muscles are composed of different myofiber types characterized by the expression of myosin heavy chain isoforms, which can be affected by physical activity, aging, and pathological conditions. Here, we present a step-by-step high-throughput semi-automated approach for performing myofiber type quantification of entire human or mouse muscle tissue sections, including immunofluorescence staining, image acquisition, processing, and quantification. Skeletal muscles support the stability and mobility of the skeleton but differ in biomechanical properties and physiological functions. The intrinsic factors that regulate muscle-specific characteristics are poorly understood. To study these, we constructed a large atlas of RNA-seq profiles from six leg muscles and two locations from one muscle, using biopsies from 20 healthy young males. We identified differential expression patterns and cellular composition across the seven tissues using three bioinformatics approaches confirmed by large-scale newly developed quantitative immune-histology procedures. With all three procedures, the muscle samples clustered into three groups congruent with their anatomical location. Concomitant with genes marking oxidative metabolism, genes marking fast- or slow-twitch myofibers differed between the three groups. The groups of muscles with higher expression of slow-twitch genes were enriched in endothelial cells and showed higher capillary content. In addition, expression profiles of Homeobox ( HOX ) transcription factors differed between the three groups and were confirmed by spatial RNA hybridization. Cognitive decline and mood disorders increase in frequency with age. Many efforts are focused on the identification of molecules and pathways to treat these conditions. Here, we demonstrate that systemic administration of growth differentiation factor 11 (GDF11) in aged mice improves memory and alleviates senescence and depression-like symptoms in a neurogenesis-independent manner. Mechanistically, GDF11 acts directly on hippocampal neurons to enhance neuronal activity via stimulation of autophagy. Transcriptomic and biochemical analyses of these neurons reveal that GDF11 reduces the activity of mammalian target of rapamycin (mTOR), a master regulator of autophagy. Using a murine model of corticosterone-induced depression-like phenotype, we also show that GDF11 attenuates the depressive-like behavior of young mice. Analysis of sera from young adults with major depressive disorder (MDD) reveals reduced GDF11 levels. Lignocellulosic biomass is rich in lignins, which represent a bottomless natural source of aromatic compounds. Due to the high chemical complexity of these aromatic polymers, their biological fractionation remains challenging for biorefinery. The production of aromatics from the biological valorization of lignins requires the action of ligninolytic peroxidases and laccases produced by fungi and bacteria. Therefore, identification of efficient ligninolytic enzymes with high stability represents a promising route for lignins biorefining. Our strategy consists in exploiting the enzymatic potential of the thermophilic bacterium Thermobacillus xylanilyticus to produce robust and thermostable ligninolytic enzymes. In this context, a gene encoding a putative catalase-peroxidase was identified from the bacterial genome. The present work describes the production of the recombinant protein, its biochemical characterization, and ligninolytic potential. Our results show that the catalase-peroxidase from T. xylanilyticus is thermostable and exhibits catalase-peroxidase and manganese peroxidase activities. The electrochemical characterization using intermittent pulse amperometry showed the ability of the enzyme to oxidize small aromatic compounds derived from lignins. This promising methodology allows the fast screening of the catalase-peroxidase activity towards small phenolic molecules, suggesting its potential role in lignin transformation. Aging-associated muscle wasting is regulated by multiple molecular processes, whereby aberrant mRNA processing regulation induces muscle wasting. The poly(A)-binding protein nuclear 1 (PABPN1) regulates polyadenylation site (PAS) utilization, in the absence of PABPN1 the alternative polyadenylation (APA) is utilized. Reduced PABPN1 levels induce muscle wasting where the expression of cellular processes regulating protein homeostasis, the ubiquitin-proteasome system, and translation, are robustly dysregulated. Translation is affected by mRNA levels, but PABPN1 impact on translation is not fully understood. Here we show that a persistent reduction in PABPN1 levels led to a significant loss of translation efficiency. RNA-sequencing of rRNA-depleted libraries from polysome traces revealed reduced mRNA abundance across ribosomal fractions, as well as reduced levels of small RNAs. We show that the abundance of translated mRNAs in the polysomes correlated with PAS switches at the 3'-UTR. Those mRNAs are enriched in cellular processes that are essential for proper muscle function.
