目的：观察梗阻性黄疸（ OJ）大鼠肝组织诱导型一氧化氮合酶（ iNOS）蛋白的表达，分析其与肝细胞线粒体损伤的关系。方法将60只成年SD大鼠随机分为两组各30只，O组行OJ造模，S组仅手术不结扎。分别于术后1、3、7、10、14 d时各处死6只，检测肝组织普通病理改变及iNOS蛋白、NADH脱氢酶亚单位1（ND1） DNA表达。结果 S组术后各时点肝组织iNOS蛋白及ND1 DNA表达比较，P均＞0．05。与S组同时点比较，O组术后肝组织iNOS蛋白表达逐渐升高（P均＜0．05），14 d升至最高；ND1 DNA术后1、3 d升高，3 d后开始下降，10 d开始低于O组，14 d降至最低。结论 OJ大鼠肝组织iNOS蛋白表达增加，梗阻后肝细胞损伤可能与NO对线粒体DNA的损伤有关；梗阻初期NO促进肝细胞线粒体ND1 DNA表达，其后过量的NO可损伤线粒体ND1 DNA。 [ABSTRACT FROM AUTHOR]
