Rationale: Saliva as a sample matrix is rapidly gaining interest for disease diagnosis and point-of-care assays because it is easy to collect (non-invasive) and contains many health-related biomarkers. However, saliva poses particular problems relative to more common urine and blood matrices, which includes low analyte concentrations, lack of understanding of biomolecule transportation and inherent viscosity variability in human samples. While several studies have sought to improve assay sensitivity, few have addressed sample viscosity specifically. The goal of this study is to minimize the effect of sample viscosity on paper-based analytical devices (PADs) for the measurement of pH and nitrite in human saliva. Methods: PADs were used to measure salivary pH from 5.0 to 10.0 with a universal indicator consisting of chlorophenol red, phenol red and phenolphthalein. Nitrite determination was performed using the Griess reaction. Artificial saliva with viscosity values between 1.54 and 5.10 mPa∙s was tested on the proposed PAD. To ensure the proposed PADs can be tailored for use in-field analysis, the devices were shipped to Australia and tested with human specimens. Results: Initial experiments showed that viscosity had a significant impact on the calibration curve for nitrite; however, a more consistent curve could be generated when buffer was added after the sample, irrespective of sample viscosity. The linear range for nitrite detection was 0.1 to 2.4 mg/dL using the improved method. The nitrite measurement in artificial saliva also showed a good correlation with the standard spectrophotometry method ( p =0.8484, paired sample t -test, n=20). Measured pH values from samples with varying viscosities correlated well with the results from our pH meter. Conclusions: The inherent variation of salivary viscosity that impacts nitrite and pH results can be addressed using a simple washing step on the PAD without the need for complex procedures., Competing Interests: Competing Interests: The authors have declared that no competing interest exists.

A new biodegradable cellulose derivative-non-isocyanate polyurethane (NIPU) was prepared, and densities, viscosities, and surface tension of NIPU were measured by density bottle method, capillary viscometer and BZY-2 type full automatic surface tension meter respectively from 298.15 to 318.15 K. The results showed that densities, viscosities and surface tension were related to temperature and concentration. The viscosity and surface tension decreased with the increase in temperature at the same concentration; at the same temperature, viscosity and surface tension were positively correlated with concentration. And the experimental data were correlated by the correlation equations of viscosity and temperature, they showed a good agreement with the fitting results of the related equations. In this study we have evaluated the suitability of laboratory testing methods to predict inkjet printing results. We have developed and used testing liquids that are spanning the operational window of industrial High Speed Inkjet (HSI) printers while still covering the maximum possible range of viscosity and surface tension. First we correlated liquid penetration measured with ultrasound (ULP) and direct absorption (ASA) to print through from HSI prints. The best correlation (R² É 0.7) was found for the sized paper. For papers with increasing liquid penetration speed we found a decreasing ability of both testing methods to predict print through, for the strong absorbing paper the correlation drops to R² É 0.2. Second we correlated contact angle and drop diameter to the dot area from HSI prints. Contact angle turned out to be a better predictor for printed dot area than drop diameter. Evaluating the change in contact angle over time we found the highest correlation to the dot area in the print when measuring the contact angle as soon as possible, in our case 1 ms after deposition of the drop on the paper. We also compared contact angle with microliter drops to picoliter drops, which are in the size scale of the actual inkjet droplet. To our great surprise correlations for microliter drops were equal or better than for picoliter drops, particularly for highly absorbing papers. Thus in order to predict dot spreading on paper our results suggest to measure the contact angle with microliter drops. Overall we found that, using laboratory testing methods, print through and dot spreading for HSI printing can be quite well predicted for slow absorbing papers but not very well for fast absorbing papers.

Deoxyribonuclease I (DNase I) is a biomarker which has important applications in various biological processes. Thus, it is highly important to develop a user-friendly method for the detection of DNase I. Here, we present a paper-based distance sensor for the rapid detection of DNase I based on changes in the viscosity of DNA mucus. The viscosity of DNA mucus varies with different concentrations of DNase I, showing different water flow lengths on the pH test papers, this makes the quantification of DNase I possible. This method has a wide linear range (0.01–10 U/mL), excellent sensitivity, remarkable specificity and excellent reproducibility. The detection limit reaches 0.003 U/mL. Additionally, it can be well applied to detection of DNase I inhibitors, assay of DNase I in human serum and quality evaluation of nucleic acid scavengers. In general, this study offers a brief, convenient, label-free, and economical method to construct paper-based distance sensors using DNA mucus, which is very promising in the detection of DNase I in various applications. Chymotrypsin (CHT) is an important proteolytic enzyme that can quickly decompose denatured proteins. Herein, we reported a new, simple, rapid method for the detection of CHT and its inhibitors by a paper-based flow sensor for the first time. The principle of the sensor is the interconversion of solidification and dissolution of gelatin. Generally, the 3D structure of gelatin can be enzymatically hydrolyzed by CHT releasing internal water, leading to a gradual increase in the water mark coverage area. Conversely, water cannot be released and the water mark coverage area is 0 in the absence of CHT, which is attributed to the water being trapped in the gelatin hydrogel. The limit of detection of CHT is determined to be 0.8 U/mL. In addition, the detection of CHT in milk, screening of CHT inhibitors and determination of Bowman-Birk inhibitors in soymilk were successfully achieved. Interestingly, the sensor only requires gelatin hydrogel, pH indicator strips, and PVC plates. In summary, a rapid, high-throughput, low-cost strategy for the detection of CHT and screening of CHT inhibitors was developed. Moreover, this method also allows the detection of CHT in non-transparent samples such as milk and soymilk. Paper-based flow sensors are highly promising for applications in food quality control, new medicine discovery and disease diagnosis, which also have a high potential for commercialization. Interest in using vegetable oils as dielectric coolants in power transformers dates back to the early studies on linseed- and castor- based fluids as possible replacements of mineral oil. However, interest in vegetable oils was lost due to their high viscosity and their tendency to oxidize. But, in the early 1990s and due to environmental concerns, the interest in dielectric fluids based on vegetable esters re-emerged. Abstract: The influence of operational variables in the pulping of vine shoots by use of ethylene glycol [viz. temperature (155–185°C), cooking time (30–90min) and ethylene glycol concentration (50–70% v/v)] on the properties of the resulting pulp (viz. yield, kappa number and viscosity) and paper sheets (breaking length, stretch, burst index, tear index and brightness) was studied. A central composite factorial design was used in conjunction with the software ANFIS Edit Matlab 6.5 to develop fuzzy neural model that reproduced the experimental results of the dependent variables with errors less than 5%. Abstract: Antimicrobial papers were prepared by coating paper with soy protein isolate (SPI) solution as inclusion matrix of carvacrol, an antimicrobial agent. Addition of carvacrol (30% w/w of SPI) to SPI solution (10% w/v) prepared at 25°C induced soy protein aggregates and viscosity decrease. Heat treatment (50, 70, 90°C) of SPI solutions and carvacrol addition improved homogeneity reduced particles size and increased viscosity of solutions. The aggregated structure of SPI in the presence of carvacrol at 25°C may play the role of a trapping structure leading to low carvacrol losses during coating and drying process of paper (9.6% against 37% after heat treatment at 90°C) and to lower release rates specially the first three days (0.04g/m2/day and 0.31g/m2/day when SPI coating solutions were prepared at 25 and 90°C, respectively). Regardless of the heat treatments received by the SPI solutions, residual carvacrol quantities in the coated papers after 50 days ranged between 0.6 and 0.7g/m2. [Copyright &y& Elsevier]
