This paper reported a headspace gas chromatography (HS-GC) for the determination of denitrifying enzyme activity in soil samples. It was based on measuring the NO 2 signal in a set of closed/air-free vials containing soil samples that incubated at the designated conditions. The results showed that the method has a good measurement precision (RSD < 5.0%) and reasonable accuracy for determining the denitrifying enzyme activity in soil samples. The heterogeneity of lignin chemical structure and molecular weight results in the lignin inhomogeneous properties which also covers the antioxidant performance. In order to evaluate the effects of lignin heterogeneity on its antioxidant activity, four lignin fractions from enzymatic hydrolysis lignin were classified by sequential organic solvent extraction and further evaluated by DPPH (1,1-Diphenyl-2-Picrylhydrazyl) free radical scavenging capacity and reducing power analysis. The characterization including FTIR, 1 H NMR and GPC showed that the fractionation process could effectively separate lignin fractions with distinctly different molecular weight and weaken the heterogeneity of unfractionated lignin. The antioxidant performance comparison of lignin fractions indicated that the dichloromethane fraction (F1) with lowest molecular weight (4585g/mol) and highest total phenolics content (246.13mg GAE/g) exhibited the highest antioxidant activity whose value was close to commercial antioxidant BHT (butylated hydroxytoluene). Moreover, the relationship between the antioxidant activity and the structure of lignin was further discussed to elucidate the mechanism of antioxidant activity improvement of lignin fractionation. Lignin is the second most abundant bio-resource in nature. It is increasingly important to convert lignin into high value-added chemicals to accelerate the development of the lignocellulose biorefinery. Over the past several decades, physical and chemical methods have been widely explored to degrade lignin and convert it into valuable chemicals. Unfortunately, these developments have lagged because of several difficulties, of which high energy consumption and non-specific cleavage of chemical bonds in lignin remain the greatest challenges. A large number of enzymes have been discovered for lignin degradation and these are classified as radical lignolytic enzymes and non-radical lignolytic enzymes. Radical lignolytic enzymes, including laccases, lignin peroxidases, manganese peroxidases and versatile peroxidases, are radical-based bio-catalysts, which degrade lignins through non-specific cleavage of chemical bonds but can also catalyze the radical-based re-polymerization of lignin fragments. In contrast, non-radical lignolytic enzymes selectively cleave chemical bonds in lignin and lignin model compounds and, thus, show promise for use in the preparation of high value-added chemicals. In this mini-review, recent developments on non-radical lignolytic enzymes are discussed. These include recently discovered non-radical lignolytic enzymes, their metabolic pathways for lignin conversion, their recent application in the lignin biorefinery, and the combination of bio-catalysts with physical/chemical methods for industrial development of the lignin refinery.

A modified process consisting of an initial mechanical refining (R) followed by a low-alkali (5.5% NaOH) cold caustic extraction (CCE) and finally an endoglucanase (EG) treatment (R-5.5%CCE-EG) was investigated for upgrading paper-grade pulp to dissolving pulp. Results showed that compared to the conventional process (9%CCE-EG), the modified process can decrease the alkali concentration (from 9% to 5.5%) to achieve a similar hemicelluloses removal while simultaneously enhancing the Fock reactivity (from 62.2% to 81.0%). The improved results were due to the fact that the mechanical refining resulted in favorable fiber morphological changes, including increased pore volume/size, water retention value and specific surface area. Consequently, the hemicelluloses removal was enhanced even under the subsequent low-alkali CCE condition. Green liquor (Na2S+Na2CO3, GL) pretreatment is a proven pathway to improve the enzymatic saccharification for the production of bioethanol. In this work, the effects of GL pretreatment on the chemical composition and enzymatic digestibility of rice straw at various total titratable alkali (TTA) charge and temperature were investigated. The GL pretreatment showed excellent performance in high polysaccharides retention and delignification selectivity. Under the optimized GL pretreatment condition (4% TTA charge, 20% sulfidity and 140°C), 92.5% of glucan, 82.4% of xylan and 81.6% of arabinan in rice straw were recovered with a delignification of 39.4%. The maximum sugar yields of 83.9%, 69.6% and 78.0%, respectively for glucan, xylan and total sugar, were achieved at the same GL pretreatment condition with an enzyme loading of 40 FPU/g-substrate. Lignocellulosic biomass can be utilized to produce promising biofuels. In this study, liquid hot water pretreatments were performed to break the intricate structure of sugarcane bagasse, which resists the enzyme accessibility to cellulose. The effects of temperatures and times on the hemicellulose degradation (including the yields of pentoses and hexoses, the proportion of monomers and oligosaccharides, as well as limited inhibitors) and cellulose enzymatic digestibility were evaluated. The results indicated that the maximum xylose yields (combined 3.85 g xylose and 13.21 g xylo-oligosaccharides per 100 g raw material) in prehydrolyzate liquid were obtained at 180 °C and 30 min. Due to the effective removal of hemicellulose, the maximum glucose yield in enzyme hydrolyzate reached 37.27 g per 100 g raw material, representing 90.13% of glucose in the sugarcane bagasse. The enzymatic hydrolysis of the bagasse pulp prepared from the treatment process with active oxygen and MgO-based solid alkali was studied. The hydrolysates were tested by IC (ionic chromatography) for the analysis of monosaccharide. Additionally, the changes of pulp before and after hydrolysis were characterized with Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), X-ray diffraction (XRD), Kajaani cellulose automatic analyzer and atomic force microscopy (AFM) techniques. The results showed that an optimized sugar yield of 82.38% was obtained at the substrate concentration of 5% for 72h with the enzyme dosage of 15IU/g. Furthermore, as the length of the cellulose fiber decreased, the crystallinity of cellulose increased, and more depressions appeared on the surface of pulp after enzymatic hydrolysis., (Copyright © 2013 Elsevier Ltd. Green liquor consists of sodium carbonate and sodium sulfide and is readily available in any kraft mills. The green liquor pretreatment process for bioethanol production was developed for wood chips. This process uses only proven technology and equipment currently used in a kraft pulp mill and has several additional advantages such as high sugar recovery and concentration, no inhibitive substances produced, as compared to acid-based pretreatment methods. The liquor was used to pretreat corn stover for enhancing enzymatic hydrolysis in bioethanol production. Pulp yield of 70% with 45% lignin removal was achieved under optimized conditions (8% total titratable alkali, 40% sulfidity and 140°C). About 70% of the original polysaccharides were converted into fermentable sugars, using 20 FPU/g-pulp of enzyme in the subsequent enzymatic hydrolysis. The effect of polymer on the retention and the thermal stability of bioactive enzymatic papers was measured using a colorimetric technique quantifying the intensity of the enzyme-substrate product complex. Alkaline phosphatase (ALP) was used as model enzyme. Three water soluble polymers: a cationic polyacrylamide (CPAM), an anionic polyacrylic acid (PAA) and a neutral polyethylene oxide (PEO) were selected as retention aids. The model polymers increased the enzyme adsorption on paper by around 50% and prevented enzyme desorption upon rewetting of the papers. The thermal deactivation of ALP retained on paper with polymers follows two sequential first order reactions. This was also observed for ALP simply physisorbed on paper. The retention aid polymers instigated a rapid initial deactivation which significantly decreased the longevity of the enzymatic papers. This suggests some enzyme-polymer interaction probably affecting the enzyme tertiary structure. Wood fibres have been utilized by our society as an important component of paper products and are presently gaining more interest as reinforcement in composite materials. During the last decades biochemical treatments have also found applications in the processing of wood fibres. The chemical, mechanical and biochemical treatments affect the morphology of the fibre wall structure at the micro- and nano-level. In this study, we present a modern approach where field-emission SEM (FE-SEM) and relevant computerized image analysis are applied to quantify the fibre wall characteristics. Details such as surface roughness and texture of the fibre walls are quantified objectively. Global polar plots are generated, which are considered to represent the fingerprint of a given pulp. The approach offers a novel perspective in the characterisation of surface structures, moving forward from performing subjective evaluations to performing objective quantifications of wood pulp fibre surfaces., ((c) 2010 Elsevier Ltd. Regioselective enzymatic acylations of 1-beta-D-arabinofuranosylcytosine (ara-C) with vinyl laurate (VL) in binary organic solvents were explored for the preparation of 5'-O-laurate of ara-C. Among the nine kinds of enzymes, Novozym 435 showed the highest regioselectivity (>99.9%) towards the 5'-OH of ara-C. This lipase showed higher catalytic activity in hexane-pyridine than in other tested solvent mixtures. The most suitable VL to ara-C molar ratio, initial water activity, and reaction temperature were shown to be 15:1, 0.07, and 50 degrees C, respectively, under which the initial reaction rate and the maximum substrate conversion were as high as 84.0 mmol L(-1) h(-1) and 98.1%, respectively. Coastal Bermuda grass (GBG) was pretreated using an autohydrolysis process with different temperatures and times, and the pretreated materials were enzymatically hydrolyzed using a mixture of cellulase, xylanase and beta-glucosidase with different enzyme loadings to evaluate sugar yields. Compared with untreated CBG, autohydrolysis pretreatments at all elevated temperatures and residence times tested enhanced enzymatic digestibility of both cellulose and hemicellulose. Increasing the temperature and residence time also helps to solubilize hemicelluloses, with 83.3% of the hemicelluloses solubilized at 170 degrees C for 60 min treatment. However, higher temperatures and longer times resulted in an overall lower sugar recovery when considering monosaccharides in the prehydrolyzate combined with the enzyme hydrolyzate. Autohydrolysis at 150 degrees C for 60 min provided the highest overall sugar yield for the entire process. A total of 43.3 g of sugars, 70% of the theoretical sugar yield, can be generated from 100g CBG, 15.0 g of monosaccharide in the prehydrolyzate and 28.3 g in the enzyme hydrolyzate. The conversion efficiency could be further improved by optimizing enzyme dosages and xylanases:cellulases ratio and pretreatment conditions to minimize sugar degradation.

Lignin is one of the most abundant biopolymers, and it has a complex racemic structure. It may be formed by a radical polymerization initiated by redox enzymes, but much remains unknown about the process, such as how molecules as large as enzymes can generate the compact structure of the lignified plant cell wall. We have synthesized lignin oligomers according to a new concept, in which peroxidase is never in direct contact with the lignin monomers coniferaldehyde and coniferyl alcohol. Instead, manganese oxalate worked as a diffusible redox shuttle, first being oxidized from Mn(II) to Mn(III) by a peroxidase and then being reduced to Mn(II) by a simultaneous oxidation of the lignin monomers to radicals that formed covalent linkages of the lignin type. Furthermore, a high molecular mass polymer was generated by oxidation of coniferyl alcohol by Mn(III) acetate in a dioxane and water mixture. This polymer was very similar to natural spruce wood lignin, according to its NMR spectrum. The possible involvement of a redox shuttle/peroxidase system in lignin biosynthesis is discussed.
