Lipids from marine organisms are a source of molecules of high nutritional significance, like polyunsaturated fatty acids (PUFAs) and antioxidants. The incorporation of these molecules into the human body is possible mainly through the intake of fish and fish oil-based food. This review covers some of the health benefits and biochemical aspects of nutritional lipids, the available marine resources for the production of PUFAs and antioxidants, and the most used methods for the extraction and identification of these natural lipid molecules in the lab and in the industry. Emphasis is put on the use of residual biomass from fisheries to obtain these interesting products and the consequent improvement of the sustainability of the fish industry. [ABSTRACT FROM AUTHOR]

Phytogenic additives such as thymol are encountering growing interest in the poultry industry. However, there are still questions concerning dynamics of their bioavailability, biological action, optimal dosage and duration of supplementation needed to achieve meaningful effects, as well as persistence of induced changes after supplement withdrawal. We studied the link between the dynamics of free thymol concentration and the changes in fatty acids composition in quail egg yolk, both during a month-long chronic dietary supplementation and after 3 weeks of supplement withdrawal (post-supplementation). Fifty, 85 days-old, female quail of homogeneous body weights (251±1g) in egg-laying peak were used. To evaluate potential dose-dependent effects, three increasing doses 2, 4, and 6.25 g of thymol/kg of feed (THY2, THY4 and THY6, respectively) and two controls were evaluated (n = 10). In parallel, we assessed the concomitant changes in free thymol excretion, potential liver histopathological changes, and birds´ performance traits. Egg yolk and droppings show a dose-dependent increase in THY concentration after 9 days of supplementation and a decrease after post-supplementation. In egg yolk, these changes were accompanied by reduced saturated fatty acid concentrations achieved by 28 days of supplementation in THY2 and 14 days of supplementation in THY4 and THY6. However, after post-supplementation the aforementioned effect disappeared in THY2 but not in THY4 and THY6. While THY2 failed to increase polyunsaturated fatty acids, THY4 and THY6 increased polyunsaturated fatty acids by day 14 of supplementation and remained increased after post-supplementation. Fatty acids changes induced by thymol are consistent with improved nutritional quality of eggs. No treatment effects were observed in liver histopathology and female performance. Findings suggest that both dose of thymol and duration of supplementation modulate thymol and fatty acids concentrations in egg yolk and thymol concentration in droppings. Isochrysis galbana is a marine microalga which highlights by containing a great diversity of antioxidant biomolecules. It shows a high content of lipids that can be used in aquaculture and biofuel, its high polyunsaturated fatty acid profile such as DHA (docosahexaenoic acid) or for having high fucoxanthin content (carotenoid group). For a century, this microalga is very well known as feed for bivalves, larva of fish, crustaceans, and mollusks. Transesterification is the necessary reaction to be able to derivative the fatty acids in methylated fatty acids (AGM) and thus be able to identify and quantify them. It is a key step to optimize and therefore know the improvement of the obtained fatty acid profile of study biomass. Hence, this work presents the significant difference of eight methods for obtaining fatty acids from the direct and indirect transesterification reaction (MTD and MTI, respectively). Moreover, acidic and alkaline catalysts were also used (AC1, AC2 and AL1 and AL2). The results presented better contents of AGMs (methylated fatty acids) respecting dry biomass in the MTD-AL1 method with ~ 6% and a relative abundance of DHA of ~ 12 % in MTI-AL2. In addition, the most abundant fatty acid profile was MTD-AL2 with 57.66 % in polyunsaturated. On the other hand, thanks to the addition of an internal standard in the experiences, it was possible to identify that the MTD-AL1 and MTI-AC1 methods were the most efficient in the transesterification obtained with ~ 93 % and ~ 87 %, respectively. Therefore, the method that is selected for the correct reading of fatty acids present in any biomass is relevant to detect a profile with more unsaturation as has been purchased with the microalga I. galbana. The increasing incidence of diabetes mellitus is becoming a serious threat to human health in various parts of the world. Studies with dairy products have shown a potential beneficial effect against diabetes. This experiment evaluated the supplementation of milk naturally enriched with polyunsaturated fatty acids (PUFA) and polyphenols in rats with streptozotocin-induced diabetes. Forty male 28-day-old Wistar rats were distributed in four experimental treatments of diabetic animals (streptozotocin induction) and a normal group (non-induced). Experimental treatments were: control (water), whole common milk (COM-M), milk enriched with PUFA (PUFA-M), milk enriched with PUFA and polyphenols (PUFA/P-M) through a special diet offered to dairy cows. Milk supplementation at a dose 5 mL/kg body weight was performed for 77 days, 42 days before and 35 days after diabetes induction. The COM-M supplementation increased brown fat deposits, reduced post-induction glucose levels, reduced blood fructosamine levels, and improved glucose tolerance. Milk enriched with PUFA reduced final fasting glucose, LDL levels, and improved blood antioxidant capacity. Milk enriched with PUFA and polyphenols promoted an increase in gastrocnemius muscle mass, and a reduction in mesenteric fat and LDL levels. Milk intake, with an emphasis on milk enriched with PUFA and polyphenols, attenuated the metabolic disorders of streptozotocin-induced diabetes in rats. [ABSTRACT FROM AUTHOR]

This study investigated whether intake of cow milk, naturally enriched with polyunsaturated fatty acids (PUFA, omega-3) and polyphenols (from propolis extract and vitamin E), from manipulation of cow’s diet, would result in positive metabolic effects in rats from weaning until adulthood. Male Wistar rats were fed a standard chow diet or a hypercaloric diet (metabolically disturbed rats, obese) which was supplemented with either whole common milk, milk enriched with PUFA (PUFA-M) or milk enriched with PUFA and polyphenols (PUFA/P-M), at 5mL/kg body weight,having water as control. Whole milk supplementation increased initial weight gain and reduced gain in the adulthood of rats. Intake of common milk reduced cholesterol levels in non-obese rats and reduced insulin resistance in obese rats. PUFA-milk showed a decreasing effect on plasma triacylglycerol and VLDL concentrations, increasing plasma HDL concentration and reducing adipocyte size of non-obese rats, but no effect was observed in obese rats. PUFA/P-milk in obese rats resulted in greater deposition of muscle mass and mesenteric fat, with a tendency to lower LDL levels, and resulted a visceral fat accumulation in non-obese rats. Thus, whole common milk and PUFA-rich milk have shown to be beneficial in a normal metabolic condition, whereas common milk and milk enriched with PUFA and polyphenols improve metabolic effects of obesity. The aim of this study was to determine the efficacy of different dietary fruit pomaces in reducing lipid oxidation in the meat of turkeys fed diets with a high content of n-3 polyunsaturated fatty acids (PUFAs). Over a period of 4 weeks before slaughter, turkeys were fed diets with the addition of 5% dried apple, blackcurrant, strawberry and seedless strawberry pomaces (groups AP, BP, SP and SSP, respectively) and 2.5% linseed oil. Pomaces differed in the content (from 5.5 in AP to 43.1 mg/g in SSP) and composition of polyphenols Proanthocyanidins were the main polyphenolic fraction in all pomaces, AP contained flavone glycosides and dihydrochalcones, BP contained anthocyanins, and SP and SSP—ellagitannins. The n-6/n-3 PUFA ratio in all diets was comparable and lower than 2:1. In comparison with groups C and AP, the percentage of n-3 PUFAs in the total fatty acid pool of white meat from the breast muscles of turkeys in groups BP, SP and SSP was significantly higher, proportionally to the higher content of α-linolenic acid in berry pomaces. The fatty acid profile of dark meat from thigh muscles, including the n-6/n-3 PUFA ratio, was similar and lower than 3:1 in all groups. Vitamin A levels in raw breast muscles were higher in group AP than in groups C and BP (P<0.05). The addition of fruit pomaces to turkey diets lowered vitamin E concentrations (P = 0.001) in raw breast muscles relative to group C. Diets supplemented with fruit pomaces significantly lowered the concentration of thiobarbituric acid reactive substances (TBARS) in raw, frozen and cooked meat. Our results indicate that the dietary application of dried fruit pomaces increases the oxidative stability of meat from turkeys fed linseed oil, and strawberry pomace exerted the most desirable effects due to its highest polyphenol content and antioxidant potential. Insect consumption as food is culturally practiced in various regions of the world. In Brazil, there are more than 130 species of edible insects registered, from nine orders, among which stands out the Coleoptera. The larva of the beetle Pachymerus nucleorum Fabricius, 1792, grows into the bocaiuva fruit (Acrocomia aculeata (Jacq.) Lodd. Ex Mart., 1845), which has proven nutritional quality. The aim of this work was to evaluate the nutritional potential of P. nucleorum larvae compared to bocaiuva kernels for human consumption. Proteins were the second largest portion of the larvae nutritional composition (33.13%), with percentage higher than the bocaiuva kernels (14.21%). The larval lipid content (37.87%) was also high, very close to the kernels (44.96%). The fraction corresponding to fatty acids in the oil extracted from the larvae was 40.17% for the saturated and 46.52% for the unsaturated. The antioxidant activity value was 24.3 uM trolox/g of oil extracted from larvae. The larvae tryptic activity was 0.032±0.006 nmol BAPNA/min. Both the larvae and the bocaiuva kernel presented absence of anti-nutritional factors. These results favor the use of P. nucleorum larvae as food, which are a great protein and lipid sources with considerable concentrations of unsaturated fatty acids compared to the bocaiuva kernel. Recent studies have proposed that n-3 polyunsaturated fatty acids (n-3 PUFAs) have direct antioxidant and anti-inflammatory effects in vascular tissue, explaining their cardioprotective effects. However, the molecular mechanisms are not yet fully understood. We tested whether n-3 PUFAs showed antioxidant activity through the activation of nuclear factor erythroid 2-related factor 2 (Nrf2), a master transcriptional factor for antioxidant genes. C57BL/6 or Nrf2−/− mice were fed a fish-oil diet for 3 weeks. Fish-oil diet significantly increased the expression of heme oxygenase-1 (HO-1), and endothelium-dependent vasodilation in the aorta of C57BL/6 mice, but not in the Nrf2−/− mice. Furthermore, we observed that 4-hydroxy hexenal (4-HHE), an end-product of n-3 PUFA peroxidation, was significantly increased in the aorta of C57BL/6 mice, accompanied by intra-aortic predominant increase in docosahexaenoic acid (DHA) rather than that in eicosapentaenoic acid (EPA). Human umbilical vein endothelial cells were incubated with DHA or EPA. We found that DHA, but not EPA, markedly increased intracellular 4-HHE, and nuclear expression and DNA binding of Nrf2. Both DHA and 4-HHE also increased the expressions of Nrf2 target genes including HO-1, and the siRNA of Nrf2 abolished these effects. Furthermore, DHA prevented oxidant-induced cellular damage or reactive oxygen species production, and these effects were disappeared by an HO-1 inhibitor or the siRNA of Nrf2. Thus, we found protective effects of DHA through Nrf2 activation in vascular tissue, accompanied by intra-vascular increases in 4-HHE, which may explain the mechanism of the cardioprotective effects of DHA. This research was carried out to compare the effects of two industrial scale processes (super press and triple phase systems) and a traditional Tunisian family-sized operation with laboratory milling for two varieties of olives: Chemlali and Chemchali. Analyses of the effect of the extraction system, on the oils’ characteristics, revealed statistically significant differences in some parameters, mainly in antioxidant compounds ( P < 0.05). Major fatty acids presented significant differences ( P < 0.05) with respect to the cultivar kind (55.93% vs. 72.21%). Oxidative stability of Chemchali was nearly two times higher than Chemlali (94.58 vs. 50.37 day kg−1). Results appear to confirm the general consensus that Chemchali oils are of much higher quality compared with the tolerable quality of Chemlali. The traditional method is a valid form of producing high quality oil with very high stability (48.87 and 109.07 day kg−1 for Chemlali and Chemchali oil, respectively). However, industrial mills showed that triple-phase scale could cause considerable loss in polyphenols. Diets supplemented with n-3 polyunsaturated fatty acids can promote lipid peroxidation and the propagation of oxygen radicals. These effects can be prevented by taurine, a functional ingredient with antioxidant properties. Here, we examined whether there is a correlation between transepithelial taurine transport, on the one hand, and membrane fatty acid composition and peroxidation in intestinal Caco-2 cells, on the other. Differentiated Caco-2 cells were maintained for 10 days, from the day of confluence, in control conditions or in a medium enriched with docosahexaenoic acid (DHA, 100 micromol/l), taurine (10 mmol/l) or DHA plus taurine. Incubation of the monolayers in a medium enriched with DHA increased the incorporation of this fatty acid into the brush-border membrane, at the expense of total n-6 fatty acids (C20:2n-6, C20:3n-6 and C22:4n-6). This was paralleled by increased membrane lipid peroxidation, which was partially limited by the addition of taurine. Transepithelial taurine transport was estimated from taurine uptake and efflux kinetic parameters at apical and basolateral domains. Cell incubation with DHA increased basolateral taurine uptake through an increase in V (max), whereas incubation with taurine downregulated basolateral uptake as occurred for apical taurine transporter. Moreover, addition of DHA reduced the apical downregulation effect exerted on taurine transport by taurine incubation. Our results suggest that the oxidative status of epithelial cells regulates taurine transport, thus satisfying antioxidant cellular requirements. [ABSTRACT FROM AUTHOR]

Aims: To investigate the effects of the polyunsaturated/saturated fatty acid (P/S) ratio and antioxidant supplementation on hepatic lipid peroxidation and enzyme activities under the control of the dietary peroxidizability index (PI) value in young and adult rats. Methods: We measured thiobarbituric acid-reactive substance (TBARS) concentration and enzyme activities in liver tissue. Female Sprague-Dawley rats aged 3 and 16 weeks were used. Dietary P/S ratios were 0.38 and 4.81 (LP and HP) when the PI value was 81.22. The diets were supplemented with vitamin E 1,000 mg/kg diet and selenium 2.5 mg/kg diet (LPS and HPS). Results: The hepatic TBARS concentration was higher in LP and LPS than in HP and HPS. In reducing TBARS concentration, antioxidant supplementation was more effective in adult rats than in young rats. Glutathione peroxidase and glutathione-S-transferase activities were lower in young rats than in adult rats. Conclusion: We conclude that dietary fats and antioxidant supplementation play an important role in the change of hepatic TBARS concentration when the dietary PI value is maintained at the same level. However, with the exception of glutathione reductase, no effects of the P/S ratio and antioxidant supplementation on hepatic enzyme activities were detected.
