Enzymes derived from microbial sources have gained increasing popularity in industrial applications over the past decades. Despite the high production cost, alkaline proteases have wide applications in industries such as tanneries, food production, and detergents. In recent years, there has been a shift towards utilizing natural carbon sources for cultivating microorganisms and extracting proteases in order to reduce production costs. This study aimed to investigate the biochemical and kinetic properties of protease enzymes obtained from Aspergillus niger cultivated in a paper waste medium and compare with the enzyme produced in a basal medium. Glucose is a more favorable carbon source compared to cellulose, so paper waste was pretreated with cellulose-degrading bacteria to convert cellulose into smaller carbohydrates. After the growth of A. niger in basal and combinational media, the enzymatic properties were compared between the extracted enzymes by using casein as substrate. The results demonstrated that A. niger could produce protease enzymes in the paper waste medium similar to the basal medium with more than 5-fold cost saving. The specific activity of the enzymes isolated from the basal and paper waste media was calculated to be 184.95 ± 10.56 U ml -1 and 169.88 ± 11.05 U ml -1 , respectively. Carbon sources did not affect the optimum pH and temperature of the protease enzyme, which were found to be 8 and 37 °C, respectively. Melanins play a fundamental role in the biology and ecology of several fungal species. Unfortunately, this group of amorphous macromolecules also severely (and most times irreversibly) stains cultural heritage objects. Despite efforts made throughout the years, knowledge of the chemical composition and structure of melanins is still insufficient, which hampers the task of safely cleaning these colourants from cultural heritage materials in a targeted way without causing further deterioration. This work aimed therefore to contribute towards enlightening the characteristics of fungal melanins from three fungi that are common paper colonizers: Aspergillus niger, Chaetomium globosum and Cladosporium cladosporioides. The extracted melanins were characterized by FTIR, Raman, UV-vis, Solid-State NMR and MALDI-TOF MS spectroscopies and the effect of inhibitors of DHN-melanin and DOPA-melanin pathways on colony pigmentation and growth was evaluated. Although all the extracted colourants show a predominantly aromatic structure with carbonyl and phenolic groups, some differences between the melanins can be highlighted. Melanins obtained from Ch. globosum and Cl. cladosporioides exhibited similar structures and composition and both presented DHN-melanin characteristics, while A. niger’s melanins revealed a more complex and ordered structure, with a higher prevalence of highly conjugated carbonyls than the others, besides the additional presence of a yellow/green component. These conclusions cannot be overlooked while selecting targeted cleaning methodologies for melanin stains on cultural heritage materials.

This study aims to investigate, analyze and conserve the general arrangement plan of the Egyptian royal yacht “Safra El-Bahr” from the National Archives of Egypt’s tracing paper collection. The yacht was specifically designed for Abbas II, the khedive of Egypt in 1894 A.D. The following analytical techniques were used for this study: pH value measurement, fungal testing, XRD, ATR-FTIR and SEM-EDX. Results revealed that the pH value of the plan paper is 6.5. Fungal testing proved the presence of Aspergillus Niger and Penicillium sp. The crystallinity index of cellulose in the historical paper increased compared to modern tracing paper indicating the occurrence of deterioration. The ATR-FTIR spectrum of the studied tracing paper revealed the presence of gelatin as a sizing material. The SEM investigation showed the effect of the natural ageing and surrounding environmental conditions on the surface morphology of the fibers. The EDX analysis revealed the use of kaolinite Al2Si2O3(OH)4, Cl, NaOH and CaCo3 in paper manufacture. Copper carbonate (CuCO3) was used for the blue color. Conservation treatments conducted on the object included cleaning, removal of pressure sensitive tape, removal of adhesive residues, tear mending and lining. In this study, extractives from Pinus rigida heartwood (PRW), Eucalyptus camaldulensis var. obtusa aerial parts (ECL), and Eucalyptus flower buds (ECF) were used as additives in paper sheets produced from the pulp of linen fibers, and their effects on the mechanical and antifungal properties of the paper sheets were studied. The highest tensile and tear indices were 31.5 Nm/g, and 17.3 mNm2/g as pulp treated with PRW (4%), and ECL (1%), respectively. All the pulp additives yielded lower burst index values compared to the control (2.24 KPa.m2/g); the nearest value was 2.23 KPa.m2/g (ECL 4% pulp additive). The brightness percentages (ISO%) ranged from 65.7% to 70.2%, which were lower than the control treatment (70.3%). The paper sheets produced from pulp treated with 2% or 4% PRW, and with 4% ECL suppressed Aspergillus niger growth on the paper disc; pulp treated with 2% or 4% PRW, 4% ECL, and with 2% or 4% ECF completely suppressed A. terreus growth. Bioethanol and enzymes were produced from fiber sludges through sequential microbial cultivations. After a first simultaneous saccharification and fermentation (SSF) with yeast, the bioethanol concentrations of sulfate and sulfite fiber sludges were 45.6 and 64.7 g/L, respectively. The second SSF, which included fresh fiber sludges and recycled yeast and enzymes from the first SSF, resulted in ethanol concentrations of 38.3 g/L for sulfate fiber sludge and 24.4 g/L for sulfite fiber sludge. Aspergillus niger carrying the endoglucanase-encoding Cel7B gene of Trichoderma reesei was grown in the spent fiber sludge hydrolysates. The cellulase activities obtained with spent hydrolysates of sulfate and sulfite fiber sludges were 2,700 and 2,900 nkat/mL, respectively. The high cellulase activities produced by using stillage and the significant ethanol concentrations produced in the second SSF suggest that onsite enzyme production and recycling of enzyme are realistic concepts that warrant further attention.

The deinking in the paper industry is in great demand, and microbial enzymes are key players. In the present study, the endoglucanase production from newly isolated fungi Aspergillus niger MPS25 is reported. The optimization of endoglucanase production was carried out using one factor at a time approach resulting in endoglucanase activity (2.37 IU/ml) at 45 °C and pH 8 in submerged fermentation (SmF), which shows optimum enzyme activity at 60 °C. Interestingly, the metal ions viz. Co2+ stimulated the endoglucanase activity, whereas Mn2+ reduced the enzyme activity, which shows that this enzyme can be used for effluent treatment released through deinking. The enzymatic hydrolysis of wheat straw produced 26.96 ± 0.108 mg/g of reducing sugars, indicating its potential in saccharification and the biofuel industry. Furthermore, the validation of the deinking efficiency of this enzyme resulted in improved deinking of mixed office waste and old newspapers by 31.5% and 20.4%, respectively. The strength properties, viz. burst factor and tear index, breaking length, and tensile index of the handmade paper sheets, were also improved which were analyzed by the scanning electron micrographs. The FTIR and XRD analysis of pulp provided insights into the changes in functional groups and cellulose crystallinity, respectively. These results indicate that multi-efficient endoglucanase from Aspergillus niger MPS25 is suitable for enzyme-based eco-friendly deinking for waste paper recycling and lignocellulosic biomass saccharification. Pycnoporus cinnabarinus laccase was fused to the C-terminal linker and carbohydrate binding module (CBM) of Aspergillus niger cellobiohydrolase B (CBHB). The chimeric enzyme of molecular mass 100 kDa was successfully produced in A. niger. Laccase-CBM was further purified to determine its main biochemical properties. The Michaelis-Menten constant and pH activity profile were not modified, but the chimeric enzyme was less thermostable than either the P. cinnabarinus laccase or the recombinant laccase produced in the same strain. Laccase-CBM was able to bind to a cellulosic substrate and, to a greater extent, to softwood kraft pulp. Binding to the pulp was shown to be mainly time and temperature-dependent. Laccase-CBM was further investigated for its softwood kraft pulp biobleaching potential and compared with the P. cinnabarinus laccase. Addition of a CBM was shown to greatly improve the delignification capabilities of the laccase in the presence of 1-hydroxybenzotriazole (HBT). In addition, ClO(2) reduction using 5 U of chimeric enzyme per gram of pulp was almost double than that observed using 20 U of P. cinnabarinus laccase per gram of pulp. We demonstrated that conferring a carbohydrate binding capability to the laccase could significantly enhance its biobleaching properties.

Xylanase is a hemicellulase enzyme that catalyses the hydrolysis of xylan to xylose which is widely used in processing of feed, pulp and paper. It is produced by many microorganisms especially filamentous fungi like Trichoderma and Aspergillus. A potential xylanolytic fungal isolate Aspergillus niger was isolated from forest soils of Tirumala, AP, India, and its crude enzyme was checked for its potential in paper bleaching. Under submerged fermentation, production of xylanase, cellulase, biomass, total protein and sugar released were analysed after 7 days of incubation at room temperature. Maximum enzyme activity was recorded on the fifth day of incubation, biomass after the seventh day, total protein and sugar released on the sixth day of incubation. Enzyme pretreatment of paper reduced 3.5 points in kappa number, 3.1 points increase in brightness and removal of chromophores and hydrophobic compounds. The FTIR and SEM analysis of enzyme-treated sample had shown modification in surface morphology and functional groups. These results clearly demonstrated that the xylanase produced by A. niger was effective as a pulp biobleaching agent which can be used on an industrial scale. [ABSTRACT FROM AUTHOR]

The filamentous fungus Aspergillus niger is well known for its high protein secretion capacity and a preferred host for homologous and heterologous protein production. To improve the protein production capacity of A. niger even further, a set of dedicated protein production strains was made containing up to 10 glucoamylase landing sites (GLSs) at predetermined sites in the genome. These GLSs replace genes encoding enzymes abundantly present or encoding unwanted functions. Each GLS contains the promotor and terminator region of the glucoamylase gene (glaA), one of the highest expressed genes in A. niger. Integrating multiple gene copies, often realized by random integration, is known to boost protein production yields. In our approach the GLSs allow for rapid targeted gene replacement using CRISPR/Cas9-mediated genome editing. By introducing the same or different unique DNA sequences (dubbed KORE sequences) in each GLS and designing Cas9-compatible single guide RNAs, one is able to select at which GLS integration of a target gene occurs. In this way a set of identical strains with different copy numbers of the gene of interest can be easily and rapidly made to compare protein production levels. As an illustration of its potential, we successfully used the expression platform to generate multicopy A. niger strains producing the Penicillium expansum PatE::6xHis protein catalysing the final step in patulin biosynthesis. The synthesis of xylosyloligosaccharides from beta-(1----4)-xylobiose in the presence of D-mannose by transxylosylation with beta-xylosidase from Aspergillus niger IFO 6662 was studied. At first, the transxylosylation products were separated by charcoal column chromatography in 5% ethanol elution into three peaks, P-1, P-2, and P-3. They were further separated on thin layer chromatography, and their three products, saccharides O-4, O-5, and O-N were purified by preparative paper chromatography. Saccharides O-4 and O-5 were identified as O-beta-D-xylopyranosyl-(1----4)-D- mannopyranose and O-beta-D-xylopyranosyl-(1----6)-D-mannopyranose, respectively, by measurement of the degree of polymerization, specific rotation, acid hydrolysis, and methylation analysis. By 1H-NMR spectroscopy in addition to these analyses, saccharide O-N was identified as O-beta-D-xylopyranosyl-(1----1')-beta-D-xylopyranose, a new nonreducing xylobiose.

Objective: To develop an endo-β-1,4-xylanase with high specificity for production of prebiotic xylooligosaccharides that optimally works at moderate temperature desirable to reduce the energy cost in the production process., Results: The xylB gene, encoding for a glycosyl hydrolase family 11 xylanase from a thermoresistant fungus, Aspergillus niger BCC14405 was expressed in a methylotrophic yeast P. pastoris KM71 in a secreted form. The recombinant XylB showed a high specific activity of 3852 and 169 U mg -1 protein on beechwood xylan and arabinoxylan, respectively with no detectable side activities against different forms of cellulose (Avicel Ò PH101 microcrystalline cellulose, phosphoric acid swollen cellulose and carboxymethylcellulose). The enzyme worked optimally at 45 °C, pH 6.0. It showed a specific cleavage pattern by releasing xylobiose (X2) as the major product from xylooligosaccharides (X3 to X6) substrates. The highest XOS yield of 708 mg g -1 substrate comprising X2, X3 and X6 was obtained from beechwood xylan hydrolysis., Conclusion: The enzyme is potent for XOS production and for saccharification of lignocellulosic biomass., (© 2021. Herein, the deteriorating aspects of a historical manuscript (papers and leather bookbinding) dated back to the 17th century were evaluated with reference to the role of the associated fungal communities. The deterioration signs were investigated using visual assessment, SEM, ATR-FTIR, XRD, color changes, and pH values compared with control. Data showed that the most deterioration aspects were represented by the dust, dirt, erosion, stains, bores, weakness, missed parts, decreasing in paper crystallinity, shifting of wavenumbers of cellulosic band, and changing of color and pH. The dependent-culturable technique showed that thirteen fungal strains were associated with historical manuscript and identified using traditional and molecular methods as Aspergillus niger (three isolates), A. fumigatus (two isolates), A. quadrilineatus (three isolates), Penicillium citrinum (two isolates), and P. chrysogenium (three isolates). These fungal strains showed high efficacy to secretion various hydrolytic enzymes including cellulase, amylase, gelatinase, and pectinase which play a critical role in biodeterioration. Cellulase enzyme was produced by a selected strain of Aspergillus niger isolated from deteriorated wood and grown on different carbon sources. Filter paper gave the highest yield, followed by carboxymethyl cellulose (CMC). Cellobiose as well as glucose gave a low yield, while the yield from lactose was negligible. The concentration of filter paper cellulose that induced the maximum yield of the enzyme was 1%. Both soluble cellulose (CMC) and cotton cellulose treated with phosphoric acid (swollen) were easily hydrolyzed by cellulase; an increase in cellulase concentration lead to more hydrolysis of CMC and gave linearity in the reaction velocity. At certain concentrations of the enzyme, increase in CMC concentration, (up to 1%) resulted in more reducing sugar. Beyond this point no more hydrolysis occur.

Cellulases are commercially important enzymes with application in various industries such as biofuel, detergent, food processing, brewery, pulp and paper. To make its production cost-effective, a preferred method is to use solid-state fermentation and with use of inexpensive substrates. Solid-state fermentation is an alternative culturing method and yields higher enzymes compared to submerged fermentation. In the current study, Aspergillus niger was isolated and further developed as inoculum for solid-state fermentation. Agroindustrial discards like banana pseudostem, jackfruit waste were used as the substrates. The substrates were pretreated by acid and were characterized by FTIR analysis to confirm the presence of cellulosic content. Different concentrations of the substrates were attempted for fermentation and the yield of the enzyme was compared. The solid-state fermentation was stable for enzyme production as well as microbial growth. The cellulase activity per gram of the substrate (U/g) was obtained maximum for jackfruit waste-based media (17±1.1 U/g). Both the lignocellulosic substrates were potential substrates for the production of cellulase enzyme. With further optimization and scale-up, this could be a cheap and sustainable process. To prepare and characterize silver nanoparticles from Guava leaves (Psidium guajava), extracts (chloroform and ethanol). The fabricated silver nanoparticles were tested on resistant fungal strains, Aspergillus niger and Candida glabrata. Psidium guajava leaf extracts were obtained by cold maceration method, by using Chloroform and Ethanol. To one mL of extract 2mM silver nitrate solution was added. The mixture was kept overnight in a dark chamber at room temperature. The solution was centrifuged at 10,000 rpm for 15 minutes and filtered by wattman filter paper after centrifugation. Psidium guajava leaf extract based silver nanoparticles were prepared and evaluated for anti-fungal activity by microtitre well plate method on resistant strains Aspergillus niger and Candida glabrata. This study investigates the potential of bioethanol production from agro wastes. Agro waste from rice husk was subjected to a pretreatment process using acid hydrolysis. 20g of rice husk was soaked in 2%, 6% and 10% H2SO4 acid concentration for 2 hours, followed by filtration using 1 Whatman filter paper. The pH of the filtrate sample was adjusted to pH of 4.5 using 10% NaOH, and co-culture of Saccharomyces cerevisiae and A. Niger was introduced into the filtrate and stored for 5 days. Distillation was carried out at 78 °C. The ethanol yield, sugar content and ethanol content of all the samples obtained was analysed. The results obtained shows that 10% H2SO4 pretreated sample resulted into maximum ethanol yield (32.13 g/ml) and sugar content (13.4%). Objectives: The cellulase enzyme is used for various industrial applications such as textile, paper, food and biofuel industries. Industrially, the fungal strain, Aspergillus niger, is widely used to produce cellulase enzymes. Emerging evidence has indicated the possible role of magnetic nanocomposites in enhancing cellulase production by A. niger. The cellulase/iron oxide nanocomposites are already reported to be an eco-friendly method for modulating microbial biochemical characteristics. Methods: The present study was used to assess the efficacy of cellulase/iron oxide magnetic composites (CMNPS) for the enhanced production of cellulase enzyme. The fungal strain Aspergillus niger was isolated from the soil samples by using standard techniques. The fungi were then transfected with CMNPS and further evaluated for the production of cellulase enzyme. Results: Synthesis of the cellulase/iron oxide magnetic composites was characterized by UV, FTIR, and XRD. The produced CMNPS was used as substrate and enzyme production. The cellulase enzyme production by Aspergillus niger was analyzed by CMC (0.82 IU/ml enzyme activity) and FPA assay (0.039 IU/ml enzyme activity). CMNPS was also analyzed by CMC (0.74 IU/ml enzyme activity) and FPA assay (0.039 IU/ml enzyme activity). Reuse of CMNPS is the ultimate application and ecofriendly approach for the synthesis of cellulase enzymes. Conclusion: The study concludes that the cellulase/ iron oxide nanocomposites may be a useful tool for the enhanced production of cellulase enzyme from the soil fungus A. niger.

To improve the quality of Ginkgo biloba leaves as biological feed additives, twelve Aspergillus niger strains were evaluated for their growth in the moisture ginkgo leaf meal media through solid-state fermentation . The results relating to flavor, flavonoids, enzymes, crude protein, and reducing sugars showed A. niger Gyx086 strain was capable of efficiently fermenting ginkgo leaves. The optimal cultural conditions were three loops of spores inoculation to every 75 g medium containing 60 % water, grew at 28˚C for 48 h. The Gyx086 grew well in the medium . The fermented leaves generated a strong sweet-smelling odor, could be identified by electronic nose equipment using a cluster analysis, other than the original offensive smell from non-fermented ginkgo leaves. Each gram dried culture with Gyx086 showed 2.83 × 10 9 CFU of A. niger ; 3.19 ± 0.37 FPU of acid-resistant filter paper activity. Its total contents of flavonoids, reducing sugars, and crude proteins were 19.95 ± 0.23 mg, 24.28 ± 2.35 mg, and 162.81 ± 3.46 mg in each gram of leaves, 26.03 %, 62.73 %, and 14.58 % higher than the controls, respectively. In this study, the family GH10 xylanase AnXylA10 derived from Aspergillus niger JL15 strain was expressed in Pichia pastoris X33. The recombinant xylanase, reAnXylA10 exhibited optimal activity at 40 ℃ and pH 5.0. The hydrolysates generated from beechwood xylan using reAnXylA10 primarily consisted of xylobiose (X2) to xylohexaose (X6) and demonstrated remarkable antioxidant capacity. Furthermore, the rice xylanase inhibitory protein (riceXIP) was observed to competitively inhibit reAnXylA10, exhibiting an inhibition constant (K i ) of 140.6 nM. Molecular dynamics (MD) simulations of AnXylA10-riceXIP complex revealed that the α-7 helix (Q225-S238) of riceXIP intruded into the catalytic pocket of AnXylA10, thereby obstructing substrate access to the active site. Specifically, residue K226 of riceXIP formed robust interactions with E136 and E242, the two catalytic sites of AnXylA10, predominantly through high-occupied hydrogen bonds. Based on QTAIM, electron densities for the atom pairs K226 riceXIP @HZ1-E136 AnXylA10 @OE2 and K226 riceXIP @HZ3-E242 AnXylA10 @OE1 were determined to be 0.04628 and 0.02914 a.u., respectively. Binding free energy of AnXylA10-riceXIP complex was -59.0±7.6 kcal/mol, significantly driven by electrostatic and van der Waals forces. The present study was carried out to evaluate the effects of different fungal species on the metal uptake from paper sludge by Hemarthria compressa L. Paper sludge (PS) in various concentrations (0, 30% and 60%) were used with four treatments of two fungal species (F0 = Control, F1 = Aspergillus niger, F2 = Trichoderma pseudokoningii, F3 = A. niger + T. pseudokoningii). Paper sludge as rooting medium steadily influenced growth and physio-biochemical attributes of H. compressa in F0. Results revealed discrete variations in growth attributes with different PS concentrations and each fungal treatment. The maximum damages in ionic homeostasis (Na+, K+, Ca2+, Mg2+) due to PS toxicity were evident in parallel with declined chlorophyll concentration. The highest growth, total chlorophyll and biomass of the plants were observed when PS was treated with combined fungal strains (F3). Translocation factor decreased with F3 that is indicative of changes in Cd, Pb, and Cu movement to shoot from root. The plants with combined fungal treatment also showed greater catalase (CAT), superoxide dismutase (SOD), and peroxidase (POD) activity. Ions, such as Ca, Mg, and Na, also increased gradually with combined fungal treatment. The correlation matrix displayed a close association among diverse fungal and PS levels with multiple plant attributes. PCA-Biplot confirmed the outcome of correlational analyses among different characters of H. compressa. The combined fungal mediate the extraction of heavy metals from the paper sludge by H. compressa recommends the high accumulation of heavy metal (HM) and possible reutilization of metal free sludge as a fertilizer, for application in cultivated fields. Furthermore, the effects of fungal species on HM attenuation in PS are linear and can be used in different habitats. Black mold of onion caused by Aspergillus niger is a serious and prevalent postharvest disease of onion. This problem drastically reduces the quality and marketability of onion bulbs. Conventional fungicides employed to control A. niger contamination posed negative impact on human health and environment and highlighted the need to discover ecofriendly management practices. In the present study we attempted to explore the use of eugenol for the management of black mold of onion. Eugenol may exhibit differences in antifungal activity in liquid state and in volatile phase. Hence, we developed and validated a new method, conical flask-paper cone method to study the antifungal activity of vapour phase disinfectants. This method was found to be precise, reproducible, sensitive and accurate with inter-assay R.S.D < 10%, intra-assay R.S.D <5% and R2>0.99. Conical flask-paper cone method was further utilized to determine the minimum inhibitory concentration of eugenol against A. niger. We observed that 0.03355 µl / cm3 eugenol caused invitro and invivo inhibition of A. niger. Onion bulbs that were stored by eugenol fumigation, prior inoculated by A. niger, did not develop black mold symptoms and maintained high sensory acceptability. Microscopic observations revealed that eugenol fumigation resulted in inhibition of spore germination, coagulation of cytoplasm, formation of large vesicles and shrinkage of A. niger mycelium. In conclusion, our study showed for the first time that eugenol fumigation significantly controlled black mold incidence during storage and maintained post harvest quality and sensory acceptance. Enzymatic hydrolysis of cellulose-containing raw materials, using Aspergillus niger, were studied. Filter paper, secondary cellulose-containing or starch-containing raw materials, miscanthus cellulose after alkaline or acid pretreatment, and wood chip cellulose, were used as substrates. The study focused on a wild A. niger strain, treated, or not (control), by ultraviolet (UV) irradiations for 45, 60, or 120 min (UV45, UV60, or UV120), or by UV irradiation for 120 min followed by a chemical treatment with NaN3 + ItBr for 30 min or 80 min (UV120 + CH30 or UV120 + CH80). A mixture of all the A. niger strains (MIX) was also tested. A citrate buffer, at 50 mM, wasthe most suitable for enzymatic hydrolysis. As the UV exposure time increased to 2 h, the cellulase activity of the surviving culturewas increased (r = 0.706; p < 0.05). The enzymatic activities of the obtained strains, towards miscanthus cellulose, wood chips, and filter paper, were inferior to those obtained with commercial enzymes (8.6 versus 9.1 IU), in some cases. Under stationary hydrolysis at 37 °C, pH = 4.7, the enzymatic activity of A. niger UV120 + CH30 was 24.9 IU. The enzymatic hydrolysis of secondary raw materials, using treated A. niger strains, was themost effective at 37 °C. Similarly, the most effective treatment of miscanthus cellulose and wood chips occurred at 50 °C. The maximum conversion of cellulose to glucose was observed using miscanthus cellulose (with alkaline pretreatment), and the minimum conversion was observed when using wood chips. The greatest value of cellulase activity was evidenced in the starch-containing raw materials, indicating that A. niger can ferment not only through cellulase activity, but also via an amylolytic one. Disposal and recycling of heavy metal-enriched biomass is the key to measure the success of phytoremediation. This study employed innovative approach to use Aspergillus niger (A. niger) for the treatment of Cd-contaminated Helianthus annuus L. (sunflower) stalk after phytoremediation. Single-factor results showed that the removal of Cd at an initial pH of 3 was superior to sucrose and inoculation amount. 67.67% of Cd was removed by A. niger leaching system after 11 days based on response surface methodology optimum conditions (sucrose: 76.266 g L -1 ; inoculation amount: 10%; initial pH: 3), while the concentrations of nitrogen, phosphorus and potassium (N, P and K) of sunflower stalk were unaffected. While physicochemical pretreatment effectively enhanced the bioleaching efficiency, it also resulted in significant loss of P and K elements, thereby reducing the value of biomass for recycling and utilization. Glue laminated bamboo (glubam) is a type of engineered bamboo material developed for applications in building structures and interiors. This paper focuses on the fungal (Aspergillus niger) colonization from 14 to 56 days in thick- and thin-strip glubam board with the investigation of physical, mechanical (compression), and microcosmic properties. Two-degree of carbonization treatment was employed to improve the antifungal property of the thick-strip glubam. After 56 days of infection, the deep-degree carbonized thick-strip glubam presents better anti-mold properties than medium and non-carbonized specimens. For thin-strip glubam, both parallel and perpendicular to the main bamboo fiber direction were considered. The longitudinal thin-strip glubam retains decent compressive properties, while the transverse specimens stay a stable compressive strength along all fungal tests. The paper reports the experimental values of mass loss, color changes, compressive strengths, modulus of elasticity in compression, and microstructure observations from optical and SEM microscopy at different fungal exposure timespans. Phosphorus (P) is an essential nutrient for living organisms. Under P-limiting conditions plants and microorganisms can exude extracellular phosphatases that release inorganic phosphate (Pi) from organic phosphorus compounds (Porg). Phytic acid (IP6) is an important form of Porg in many soils. The enzymatic hydrolysis of IP6 by phytase yields plant available inorganic phosphate (Pi) and less phosphorylated inositol derivates as products. The hydrolysis of organic P-compounds by phosphatases leaves an isotopic imprint on the oxygen isotope composition (δ18O) of released Pi, which might be used to trace P in the environment. This study aims at determining the effect of phytase on the oxygen isotope composition of released Pi. For this purpose, enzymatic assays with histidine acid phytases from wheat and Aspergillus niger were prepared using IP6, adenosine 5'monophosphate (AMP) and glycerophosphate (GPO4) as substrates. For a comparison to the δ18O of Pi released by other extracellular enzymes, enzymatic assays with acid phosphatases from potato and wheat germ with IP6 as substrate were prepared. During the hydrolysis of IP6 by phytase, four Pi are released, and one oxygen atom from water is incorporated into each Pi. This incorporation of oxygen from water into Pi is subject to an apparent inverse isotopic fractionation (ε ~ 6 to 10 ‰), which is similar to that imparted by acid phosphatase from potato during the hydrolysis of IP6 (ε ~ 7 ‰) where less than three Pi are released. The incorporation of oxygen from water into Pi during the hydrolysis of AMP and GPO4 by phytase yielded a normal isotopic fractionation (ε ~ -12 ‰), again similar to values reported for acid phosphatases from potato and wheat germ. We attribute this similarity in ε to the same amino acid sequence motif (RHGXRXP) at the active site of these enzymes, which leads to similar reaction mechanisms. We suggest that the striking substrate-dependency of the isotopic fractionation could be attributed to a difference in the δ18O-values of the C-O-P bridging and non-bridging oxygen atoms in organic phosphate compounds. Aspergillus niger is widely applied in the fermentation industry, but produce abundant mycelium residues every year. As a kind of solid waste, mycelium residues seriously affect the environment. How to manage and utilize this solid waste is a problem for the fermentation industry. It was reported that many kinds of biomass could be utilized to produce carbon materials, which would be further used to produce lithium-ion rechargeable batteries (LIBs). Here, porous biochar was prepared from A. niger mycelial residues and further used as an anode for LIBs. Since the A. niger mycelium contains abundant nitrogen (5.29%) from its chitosan-dominated cell wall, and silicon (9.63%) from perlite filter aid, respectively, the biochar presented an excellent cycle stability and rate performance when applied as the anode of LIBs. The conclusion of this research shows the wide application prospect of fungal fermentation residues as carbon precursors in energy storage devices. In this paper, for the first time, photopolymers were synthesized from glycerol acrylates with different numbers of functional groups, 2-hydroxy-3-phenoxypropyl acrylate, glycerol dimethacrylate or glycerol trimethacrylate, without and with the addition of vanillin styrene. The photocuring kinetics were monitored by real-time photorheometry. The mechanical, rheological, thermal, antimicrobial and shape-memory properties of the photopolymers were investigated. All polymers synthesized demonstrated antibacterial activity against Escherichia coli and Staphylococcus aureus, as well as antifungal activity against Aspergillus flavus and Aspergillus niger. 2-Hydroxy-3-phenoxypropyl acrylate-based polymers showed thermoresponsive shape-memory behavior. They were able to maintain their temporary shape below the glass transition temperature and return to their permanent shape above the glass transition temperature. The paper deals with the possible use of fungi to decontaminate polluted waters. Specifically, the focus is the selection of the most promising fungal strain capable of bioaccumulating arsenic, which is a globally widespread environmental contaminant. To this aim, allochthonous fungal strains from the ColD UNIGE JRU MIRRI strains collection were selected. Their capability to survive and accumulate this kind of pollutant was evaluated and compared with that of an autochthonous fungi set directly isolated from the arsenic polluted water. A preliminary screening at various concentrations of arsenic (0, 200, 400, 800, 1600 μg L−1) revealed that the best performing strains were Aspergillus niger and Penicillium expansum among the autochthonous strains and Aspergillus niger and Penicillium expansum among the allochthonous strains. Moreover, all the strains were subjected to bioaccumulation tests at a 1600 μg L−1 concentration. Local and allochthonous fungal strains showed different behaviors: the allochthonous strains grew rapidly and in a sustained way but without expressing any bioaccumulation activity. On the contrary, the indigenous fungi, despite a moderate growth, showed a good bioaccumulation capacity (in particular, Aspergillus niger). Untreated tannery effluent discharge, which causes severe environmental pollution. This research was performed to assess the bioremediation (multi-pollutant adsorption) potential of pre-identified and multi metal tolerant Aspergillus niger and Aspergillus tubigenesis through a stirred tank bioreactor in free and immobilized form. Physicochemical property analysis results showed that most of the tannery effluent properties were beyond the permissible limits. These A. niger and A. tubigenesis effectively immobilized on corncob and coir solid support material. The stirred tank bioreactor based bioremediation study revealed that the fungal biomass (Aspergillus niger and Aspergillus tubigenesis) immobilized coir and corncob material demonstrated remarkable multi-pollutant (TSS: 22.5% & 13.5%, TS: 29% & 22%, BOD: 21% & 10%, TDS: 28% & 19%, COD: 30% & 22%, Cr: 27% & 19%, Cu: 28% & 12%, and Pb: 48% & 29% respectively) adsorption potential in a week of treatment. Moreover, it reduced the toxicity of tannery effluent and promotes the sprouting of Oriza sativa seeds, as demonstrated by petri plate bioassay. These finding suggest that the metal-tolerant fungal isolates A. niger and A. tubigenesis demonstrated impressive bioremediation proficiencies in an immobilized state. Nowadays, the increased number of multidrug-resistant strains among pathogens is a severe public health concern and cancer is posing a great threat for humans. These problems should be tackled with the development of novel and broad-spectrum antimicrobials from microbial origin. During the present study, the bioactive secondary metabolites from Aspergillus niger CJ6 were extracted, characterized; their biological properties were evaluated by subjecting them for antimicrobial, antifungal and anticancer activities. The potent isolate Aspergillus niger CJ6 with nucleotide sequence of 959 base pairs showed antagonistic activity against fungal pathogens in dual culture. The chemical profiling of crude ethyl acetate extract indicated the presence of various bioactive molecules belonging to phenolic, hydrocarbons, and phthalate derivative classes. In antimicrobial activity, the crude extract displayed increasing activity with increased concentration; the highest activity observed against Shigella flexneri with 15 ± 1.0, 19 ± 0.5, 20 ± 1.0 and 24 ± 1.0 mm zones of inhibition at 25, 50, 75 and 100 μl concentrations. The MTT assay illustrated deformed cells of MIA PaCa-2 cell line in in-vitro cytotoxic activity; outflow of cell matrix and membrane rupture; the IC 50 of 90.78 μg/ml suggested moderate potential of extract to prevent cancer cell growth. The apoptosis/necrosis study by flow cytometer exhibited 8.98 ± 0.85% early and 73 ± 0.7% of late apoptotic population with 3.8 ± 1.1% necrotic cells; only 14.22 ± 0.6% of healthy cells suggested the increased apoptosis inducing capacity of Aspergillus niger CJ6 crude extract. Infections due to the Aspergillus species constitute an important challenge for human health. Invasive aspergillosis represents a life-threatening disease, mostly in patients with immune defects. Drugs used for fungal infections comprise amphotericin B, triazoles, and echinocandins. However, in the last decade, an increased emergence of azole-resistant Aspergillus strains has been reported, principally belonging to Aspergillus fumigatus species. Therefore, both the early diagnosis of aspergillosis and its epidemiological surveillance are very important to establish the correct antifungal therapy and to ensure a successful patient outcome. In this paper, a literature review is performed to analyze the prevalence of Aspergillus antifungal resistance in European countries. Amphotericin B resistance is observed in 2.6% and 10.8% of Aspergillus fumigatus isolates in Denmark and Greece, respectively. A prevalence of 84% of amphotericin B-resistant Aspergillus flavus isolates is reported in France, followed by 49.4%, 35.1%, 21.7%, and 20% in Spain, Portugal, Greece, and amphotericin B resistance of Aspergillus niger isolates is observed in Greece and Belgium with a prevalence of 75% and 12.8%, respectively. The prevalence of triazole resistance of Aspergillus fumigatus isolates, the most studied mold obtained from the included studies, is 0.3% in Austria, 1% in Greece, 1.2% in Switzerland, 2.1% in France, 3.9% in Portugal, 4.9% in Italy, 5.3% in Germany, 6.1% in Denmark, 7.4% in Spain, 8.3% in Belgium, 11% in the Netherlands, and 13.2% in the United Kingdom. The mechanism of resistance is mainly driven by the TR34/L98H mutation. In Europe, no in vivo resistance is reported for echinocandins. This study examined the fermentation duration in relation to the effectiveness of tannin levels in mangrove (Sonneratia alba) fruit during the fermentation process using Aspergillus niger. The tested durations were 7, 10, 13, and 16 days in anaerobic conditions. The outcomes measured included crude fat, crude fibre, and nitrogenfree extract digestibility. Microbial biomass, protozoa population, and methane gas production were also recorded as responses to the rumen microbial activity. The results showed that 16 days of fermentation gave the highest average for all responses. The protozoa population reached around 4.07 × 105 cells/ml and methane gas amounted to 33.9 ml/g of dry matter (DM). This is caused by a decrease in the anti-nutrient tannin content in mangrove (S. alba) fruit due to fermentation by A. niger according to treatment. The conclusion of this research is that the 16-day fermentation treatment (T4) of mangrove fruit is the optimal time to be used as a source of concentrate feed for livestock in terms of the increase in nutritional value and gastrointestinal microbes represented by total protozoa population, crude fibre digestibility, crude fat digestibility, and the highest nitrogen-free extract digestibility. In the starch processing industry including the food and pharmaceutical industries, α-amylase is an important enzyme that hydrolyses the α-1,4 glycosidic bonds in starch, producing shorter maltooligosaccharides. In plants, starch molecules are organised in granules that are very compact and rigid. The level of starch granule rigidity affects resistance towards enzymatic hydrolysis, resulting in inefficient starch degradation by industrially available α-amylases. In an approach to enhance starch hydrolysis, the domain architecture of a Glycoside Hydrolase (GH) family 13 α-amylase from Aspergillus niger was engineered. In all fungal GH13 α-amylases that carry a carbohydrate binding domain (CBM), these modules are of the CBM20 family and are located at the C-terminus of the α-amylase domain. To explore the role of the domain order, a new GH13 gene encoding an N-terminal CBM20 domain was designed and found to be fully functional. The starch binding capacity and enzymatic activity of N-terminal CBM20 α-amylase was found to be superior to that of native GH13 without CBM20. Benth endophytes. Endophytic fungi reside inside plants in a mutualistic relationship with the host and produce numerous bioactive compounds, including extracellular enzymes. A rich diversity of endophytic fungi associated with the medicinal plant Albizia lebbeck (L.) Benth was bio-prospected in the present study. Thirty-three isolates obtained from the plant were assessed for secretion of six different extracellular hydrolytic enzymes. The results showed that 38%, 9%, 69%, 16%, 38%, and 13% of the endophytic isolates could produce amylase, protease, lipase, laccase, cellulase, and gelatinase enzymes, respectively. Overall, 78% of the isolates showed positive results for one or the other enzyme. Aspergillus niger (Isolate 8) isolated from the bark proved to be most efficient in producing all the enzymes and was further exploited for various applications. The crude fungal amylase of the isolate helped in 39.08% reduction in biochemical oxidation demand (BOD) of municipal wastewater. Effective blood stain removal from the white cotton cloth was achieved when crude protease was used in combination with detergent. 13.29% biodegradation of vegetable oil was observed using fungal lipase. Dye decolorization of methylene blue (71.65%) and methyl orange (56.53%) was observed by fungal laccase. Saccharification of bagasse (83.94%) and paper (64.93%) was achieved by using fungal cellulase. Complete extraction of silver from waste X-ray films was achieved by crude gelatinase. In this paper, the inhibitory effect of various nanomaterials on the growth of Aspergillus niger was studied. Among them, copper nanorods had the most obvious inhibitory effect on the growth of Aspergillus niger. The phase of copper nanorods was modified by chitosan, and its inhibitory effect on the expansion of Aspergillus niger was measured. 1. Preparation of copper nanorods and chitosan@copper nanorods: Copper nanorods with a diameter of about 300–350 nm and a length of about 100–800 nm were prepared by the liquid-phase reduction method. The chitosan solution was prepared by using the characteristics of chitosan dissolved in dilute acid to prepare chitosan@copper nanorods and modify the phase of copper nanorods. 2. Determination of the inhibitory effect of various copper nanomaterials on the growth of Aspergillus niger, including Cuprous Oxide nanoparticles, copper nanorods, nano copper oxide, and copper hydroxide, which have certain inhibitory effects on the growth of Aspergillus niger. Among them, copper nanorods have a better effect. On this basis, chitosan@copper nanorods are obtained by modifying the phase of copper nanorods with chitosan. Pepsinogen A (PGA) plays an important role in the treatment of human gastrointestinal diseases. At present, PGA is mainly extracted from pig stomach, so its source is very limited and its price is very expensive. Production of PGA by microbial fermentation using an engineered strain with high PGA yield would be an ideal solution. This paper presents a new system for the high-level expression of PGA from Homo sapiens (hPGA) in Aspergillus niger. The hPGA5 gene codon was optimized according to the codon bias of A. niger and then connected to a strong promoter and signal peptide to construct an hPGA5 expression cassette. An ingenious multi-copy knock-in expression strategy mediated by the CRISPR/Cas9 tool was used to improve the production of hPGA in A. niger. By optimizing the copy number and integration sites of the hPGA5 gene, an engineering strain with a high yield of hPGA was constructed. After shake-flask fermentation for 7 d, the enzyme activity of recombinant hPGA reached 542.3 U/mL, which is the highest known activity. Fungal spore incidence inside Gandhi Gyan library of Wardha city was recorded by exposing potato dextrose agar and Czapek Dox agar culture media petri plates for 10 minutes and then incubated at 28±1° for 4-5 days. The plates were regularly examined. Number of fungal colonies was recorded from the library for 6 months in the year 2011. Among the various species encountered Aspergillus niger, Asparagus fumigatus were the predominant fungi in the library. Investigations by the petri plate exposure culture plate method helped us in determining the occurrence of fungal species in the air inside the selected library which also causes allergic diseases to human beings working therein. The fungi were also isolated from highly damaged, old and unreadable books. The cellulolytic activity and damage of library materials by some common airborne fungi were studied. The present investigation dealt with the isolation of fungal species from indoor environment of library, identification of cellulose-degrading fungal species from library books and evaluation of cellulolytic capabilities, based on loss in weight of newspaper and book paper discs. The optimization of extraction, chemical characterization, and the evaluation of antioxidant activity and α-amylase inhibition capacities of the cell wall polysaccharides extracted from Aspergillus niger ATCC 1004 were studied in this paper. The response surface methodology through a factorial design of three levels indicated the optimal conditions for extraction: pH 13 and 180 min. Characterization results showed that the polysaccharide is glucogalactan, consisting of β-D-galactose-linked units (1 → 6) and β-D-linked glucose (1 → 3). The antioxidant activity was evaluated through three in vitro assays. It could effectively scavenge DPPH, ABTS and hydroxyl radicals with inhibition rates of 82.12%, 75.87% and 79.24, respectively, at 6.4 mg/mL, which were higher than those of the other polysaccharides. For inhibitory activity against α-amylase, a blocking effect of 53.7% was observed at a concentration of 2 mg/mL. Therefore, the cell wall polysaccharides of Aspergillus niger, (1 → 3)(1 → 6)-β-D-glucogalactan, seem to be a promising source for use as an antioxidant, in addition to holding an in vitro hypoglycemic potential. The removal of microalgae represents a problematic part of the water decontamination process, in which most techniques are expensive and non-ecological. In the paper, we focus on the synergistic relationship between microscopic filamentous fungi and algal culture. In the process of decontamination of a model sample containing ammonium ions, efficient biocoagulation, resp. co-pelletization of dried algae Chlorella sp. and Aspergillus niger sensu stricto are shown. The microscopic filamentous fungus species A. niger was added to a culture of an algal suspension of Chlorella sp., where the adhesion of the algal cells to the fungi subsequently occurred due to the electrostatic effect of the interaction, while the flocculation activity was approximately 70 to 80%. The algal cells adhered to the surface of the A. niger pellets, making them easily removable from the solution. The ability of filamentous fungi to capture organisms represents a great potential for the biological isolation of microalgae (biocoagulation) from production solutions because microalgae are considered to be a promising renewable source of oil and fermentables for bioenergy. This form of algae removal, or its harvesting, also represents a great low-cost method for collecting algae not only as a way of removing unnecessary material but also for the purpose of producing biofuels. Algae are a robust bioabsorbent for absorbing lipids from the environment, which after treatment can be used as a component of biodiesel. Chemical analyses also presented potential ecological innovation in the area of biofuel production. Previous studies have shown that the soil microbial population and soil enzyme activity are seriously affected by fracturing flow-back fluid (FFBF) from the shale gas mining process. However, the toxic effect of FFBF on specific bacteria, fungi, and plankton has not been systematically confirmed in detail. In this paper, a toxic effect evaluation of FFBF was conducted using the representative toxicity test organisms Vibrio fischeri, Daphnia, Aspergillus niger, and S. cerevisiae, indicating that FFBF can significantly decrease the survival rate of these species. The results also showed that there was a significant negative correlation between the concentration of some inorganic toxicity factors and the survival rate when Daphnia was used as the test organism, indicating that the toxicity degree order for these inorganic toxicity factors is Ba2+ > Li+ > As3+ > Cl— > Cu2+ > Rb2+ > Ga2+ > V2+ > Na+. In addition, other toxic factors, including polycyclic aromatic hydrocarbons (PAHs), were also determined, and the order of toxic effects with a negative correlation to the Daphnia survival rate was confirmed. These results showed that the biological toxicity of FFBF was caused not only by inorganic toxicity factors such as heavy metals but also by organic compounds such as PAHs. The results not only provide a significant reference value for the systematic assessment of biological toxicity by FFBF, but they also have great significance for developing approaches to appropriate FFBF treatment. Theabrownins are macromolecular compounds with many hydroxyl, carboxyl, and phenolic functional groups. They are usually extracted from dark tea, in which they are present at low concentrations. In this paper, a low-cost microbial liquid fermentation method was established by inoculating Aspergillus niger with minced tea extract as the raw material. After applying the Box–Behnken and response surface approach design, the optimum fermentation conditions in the fermentor were determined to be an 8% (80 g/L) sucrose concentration, 1:31 (0.03226 g/mL) solid–liquid ratio, 4.141 × 106 CFU/mL bacterial liquid concentration, 5 d fermentation time, 28 °C fermentation temperature, 187 r/min (rpm) rotation speed, and an oxygenation of 0.5 V/V·min (V). After fermenting about 168 h, the theabrownins content reached the maximum of 28.34%. The total phenolic content of the liquid-fermented theabrownins was 25.74% higher than that of solid fermentation. Acidic functional groups were determined, indicating that the phenolic hydroxyl groups were the main acidic groups of the theabrownins. The antioxidant activity of theabrownins was verified by measuring the potassium ferricyanide reducing power, hydroxyl radical scavenging rate, superoxide anion radical scavenging rate, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging rate of solid- and liquid-fermented chabein. The results of this study show that the production of theabrownins by the liquid fermentation of Aspergillus niger is fast, high in yield, and has antioxidant activity, which provides a basis for industrial production of theabrownins. The spread of microorganisms in the air, especially pathogenic microorganisms, seriously affects people's normal life. Therefore, the analysis and detection of airborne microorganisms is of great importance in environmental detection, disease prevention and biosafety. As an emerging technology with the advantages of integration, miniaturization and high efficiency, microfluidic chips are widely used in the detection of microorganisms in the environment, bringing development vitality to the detection of airborne microorganisms, and they have become a research highlight in the prevention and control of infectious diseases. Microfluidic chips can be used for the detection and analysis of bacteria, viruses and fungi in the air, mainly for the detection of Escherichia coli, Staphylococcus aureus, H1N1 virus, SARS-CoV-2 virus, Aspergillus niger, etc. The high sensitivity has great potential in practical detection. Here, we summarize the advances in the collection and detection of airborne microorganisms by microfluidic chips. The challenges and trends for the detection of airborne microorganisms by microfluidic chips was also discussed. It is well known that Trichoderma reesei 's cellulase is deficient in β-glucosidase and cellobiohydrolase II, hampering the synergism of cellulase components and attenuating the cellulose-degrading capability. T. reesei was engineered to enhance its cellobiohydrolase II and cellulase production. However, it was still deficient in β-glucosidase. Therefore, the mixed culture of the recombinant T. reesei and Aspergillus niger was introduced to further improve cellulase production and the performance in enzymatic hydrolysis. It was found to produce the most robust cellulase, whose filter paper activity was 12.17 ± 1.18 FPIU/mL, cellobiohydrolase activity 53.10 ± 2.95 U/mL, endoglucanase activity 716.11 ± 41.16 U/mL, and β-glucosidase activity 1.93 ± 0.28 IU/mL. It had the highest enzymatic hydrolysis yield 89.35%, the lowest cellulase input 25.18 FPIU for 1 g glucose. It just needed 1.96 g SECS (dry material) for producing 1 g glucose.
