Fifty years ago, it was reported that individuals of the Pacific boreal limpet Problacmaea (= Erginus) bear a penis for copulation and brood their young in the pallial cavity. These observations were based on light microscopy, but now we reveal new details of the reproductive biology of Erginus (Problacmaea) puniceus with electron microscopy. Gametogenesis is fundamentally similar to other Patellogastropoda, but there are some key differences. Oocytes develop in the ovary, dorsal to the foot in the posterior half of the body, before passing through the oviduct to the pallial cavity, where they are fertilized and brooded. Development is direct with embryos being brooded to the crawl-away juvenile stage. However, sometimes fertilization occurs internally with embryos beginning development in or near the gonad. Free sperm, which were likely autosperm (self) but could have been allosperm (cross), were found near developing oocytes in the gonad. Spermiogenesis results in the formation of an enta-quasperm with a cap-like acrosome, a bullet-shaped nucleus and spherical mitochondria in the midpiece. Copulation was not observed, but we confirm the presence of a penis with a dorsal penial groove that lies below the right cephalic tentacle in males, which is typical of functional penises of some molluscs. Males were always smaller, but intermediate-sized individuals were hermaphroditic to different extents. The largest individuals were entirely female, and their penises were smaller and often lacked the penial groove. Thus, E. (Problacmaea) puniceus appears to be a protandrous hermaphrodite, passing from male to female phase. Remnants of the testis persist into the female phase, suggesting that they are protandrous hermaphrodites with overlap, possibly allowing for self-fertilization to occur on occasion. Adipose fins of teleost fishes have been shown to function as mechanosensory organs that respond to minute bending forces created by turbulence in fast‐flowing streams. Nonetheless, adipose fins also exist in some fishes that occupy still waters, including lanternfish (Myctophidae) in the deep sea. The authors examined adipose fin structure in northern lampfish, Stenobrachius leucopsarus, from coastal British Columbia. After fixation, embedding and sectioning of the adipose and supporting tissue, it was evident that lanternfish adipose fins are stiffened by compound actinotrichia, acting like fin rays, that would create a higher aspect ratio. The actinotrichia converge at the base of the fin in a hinge point complex that anteriorly interacts with a cartilaginous endoskeletal rod, controlled by skeletal muscles. Afferent nerves enter the fin at this point and form fine branches as they track deeper alongside actinotrichia. The authors propose that the vertical nightly migration to surface waters, as well as predator evasion within large schools, results in microturbulence. An extensive community of organisms inhabits the common periwinkle, Littorina littorea (L., 1758), along the waveswept rocky shores of Canada's East coast. This community, which includes both facultative and obligate endosymbionts, comprises a diverse array of species from seven animal phyla, including Annelida, Arthropoda, Gnathostomulida, Nematoda, Nemertea, and Platyhelminthes, as well as ciliates and algae. The presence of larger numbers of endosymbionts was found to correlate with specific shell characteristics of the snail host, including a wider aperture and columella, suggesting that these individuals have a larger mantle cavity relative to snails housing a small community of endosymbionts. Snails with large communities of endosymbionts were usually encrusted with coralline algae and often had trematode infections. Although L. littorea has been extensively studied since the last century, the existence of this community of organisms has passed unnoticed. The large diversity of organisms in this community suggests that these snails may provide refugia for a wide range of smaller taxa. Eusperm and parasperm ultrastructure have been studied for the first time in the neogastropod Plicopurpura patula. Eusperm structure is typical of many Neogastropoda, in which an apical bleb characterizes the conical acrosome with subacrosomal granule; the nucleus is tubular and penetrated to its tip by basal body and axoneme; the mid-piece comprises spiral mitochondria that ensheath the axoneme but terminate at the annulus, defining the junction with the end-piece. In the proximal portion of the end-piece dense tracts of glycogen match the axoneme but these disappear distally. The parasperm is unusual in having no axonemes and no motility was observed. Eusperm do not attach to parasperm, as occurs in the similar nurse cells of Littorinimorpha, thus potential functions may be limited to some form of paternity assurance as occurs in some insects. The nucleus of the parasperm is eliminated during paraspermiogenesis, but there is considerable post-transcriptional secretion activity resulting in the production of a variety of granules, some rich in glycoproteins. A prominent paracrystalline rod accumulates in the centre of the parasperm also from Golgi-derived secretions. Abstract: The dinoflagellate Haidadinium ichthyophilum Buckland‐Nicks, Reimchen and Garbary 1997 is an ectoparasite of the spine‐deficient, three‐spine stickleback Gasterosteus aculeatus L. Reimchen 1984, a fish endemic to Rouge Lake, Haida Gwaii. Haidadinium ichthyophilum proved difficult to assign taxonomically because its morphology and complex life cycle exhibited defining characteristics of both autotrophic and heterotrophic dinoflagellates, and was tentatively assigned to the Phytodiniales. Here, we characterized a 492 bp fragment of the small subunit ribosomal RNA (SSU rRNA) from preserved H. ichthyophilum cysts. In SSU phylogeny, H. ichthyophilum branches with the fish parasites, Piscinoodinium sp., strongly supporting the inclusion of H. ichthyophilum within the Suessiales. [ABSTRACT FROM AUTHOR]

Gametogenesis in Chitonida is highly unusual, giving rise to eggs with elaborate, often spiny hulls and sperm with needle-like nuclear extensions and tiny acrosomes. My studies of oogenesis in a variety of chitonid families have revealed unique mechanisms of egg hull formation. Three-dimensional reconstruction of serial sections of oogenesis in the chitonidAcanthochitona garnoti(Blainville) confirms that the egg hull is formed largely by microapocrine secretions, rather than by exocytosis of Golgi vesicles. This novel process, detailed for the first time in Mollusca, bypasses the endomembrane system and releases compounds formed in the cytoplasm directly from the cell surface via budding of ‘microaposomes’. Fertilization is no less unusual. In virtually all Metazoa, except chitonids, the entire sperm enters the egg, and sperm mitochondria are digested. Typically, the centrosome that derives from the sperm centriole acts as a microtubule organizing centre, moving the two pronuclei together to form the zygote nucleus and later initiating cleavage of the embryo. However, in chitonids the sperm digest a minute pore in the egg hull that allows injection of DNA but blocks entry of sperm organelles, including the nuclear membrane. Serial sections of fertilized eggs ofMopalia muscosa(Gould) confirm that a membrane bag, abandoned on the egg surface, contains the sperm organelles. Thus, not only is there maternal inheritance of mitochondria and centrioles in Chitonida but also other sperm proteins normally associated with fertilization may be blocked from entering the egg. The functional morphology of the mammiliform penial glands of Littorina saxatilis has been investigated with both light and electron microscopy. These penial glands line the ventral edge of the penis and orient with the female mantle during copulation. Secretions are released from the penial glands to this interface where they probably function in adhesion. The penial gland secretions comprise heterogeneous granules as well as apocrine and mucous secretions. The heterogeneous granules are produced in separate multicellular glands arranged in a series of lobes that lie outside a thick smooth muscle layer enclosing the lumen. Each glandular lobe is surrounded by a thin layer of smooth muscle. Secretions are transported in individual cellular processes that pass through the thick smooth muscle layer and empty into the lumen. Surrounding the lumen is an epithelium containing apocrine secretory cells as well as occasional goblet-type, mucous cells. The combined action of the muscles forces secretions out of the lumen through the penial papilla, onto the external surface of the mammiliform penial gland. Longitudinal muscles extend into the penial papilla enabling its protrusion or retraction. Retraction of the penial papilla following secretion release is thought to create negative pressure beneath the penial gland producing suction adhesion. Scanning electron microscopy (SEM) enables a close-up investigation of topographical features at the cellular level down to a resolution of about 3 nm. In terms of gametes this allows for examination of minute surface details and changes that occur during fertilization. Sometimes these features are hidden from view and must be carefully exposed during preparation in order to be made visible in the SEM. This chapter describes methods that can reveal surface details of gametes for observation, as well as fix them permanently, while keeping fixation artifacts to a minimum.

Sperm dimorphism, where ejaculates contain both fertile "eusperm" and sterile "parasperm", has been implicated in sperm competition in animals. Most parasperm lack an acrosome, eliminate the nucleus and develop a complex cytoplasm, swollen with secretory vesicles. This study of parasperm in the foliate whelk Ceratostoma foliatum, focuses on characterizing the process of nuclear breakdown and the nature of cytoplasmic secretions. Testis squash preparations were treated with a Promega apoptosis detection [terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick-end labeling-TUNEL] assay (Cedar Lane) to detect nuclear apoptosis; acridine orange was used to localize lysosomes. Parasperm were isolated on Percoll gradients and their proteins characterized by running electrophoresis of lysed, purified fractions on polyacrylamide gels. Results show that the process of nuclear breakdown follows the hallmarks of apoptosis in ultrastructure, which is confirmed by the presence of 3'-OH overhangs of DNA fragments. Paraspermatozoa of Cerithioidea are oligopyrenic with a central nuclear remnant surrounded by glycoprotein bodies and an anterior acrosome complex. Posteriorly, the parasperm have one long and several small flagella. Biosynthesis of proteins begins in the rough endoplasmic reticulum (RER), early in paraspermatogenesis, prior to the degradation of the nucleus. The breakdown of the nucleus has features characteristic of apoptosis but a nuclear remnant remains that is composed of intact DNA. The acrosome complex of parasperm is Golgi-derived, forming posteriorly and migrating anteriorly along the plasma membrane to the apex of the nuclear remnant as the paraspermatid matures. This mechanism of acrosome formation is similar to that in euspermiogenesis in neomenioid aplacophorans and neritid snails and is plesiomorphic to mollusks. However, eusperm acrosomes of Cerithioidea form differently. In the paraspermatid, small, dense granules secreted by the Golgi body fuse to form larger granular vesicles, many of which are released by exocytosis into the seminal fluid. These granular vesicles stain red with acridine orange at pH 7 under 489-nm light, indicating that they are lysosomes. The retention of a nuclear remnant and development of an acrosome complex in the parasperm of cerithioideans, as well as the secretion of lysosomes into the seminal fluid, suggests novel functions for these unusual germ cells., (2005 Wiley-Liss, Inc.)

The fine structure of the male reproductive system of the hermaphroditic brittle-star, Amphipholis squamata, has been studied in specimens from both the Pacific coast (Washington) and the Atlantic coast (New Hampshire). Each testis is a small (100-μm) sphere and is attached to the internal wall of the bursa by peritoneal suspensor cells. Occasional flagellated cells are found on the external surface of the testis. The testicular wall of A. squamata is a multilayered structure, similar to that of other ophiuroids, but the hemal sinus is PAS negative in this species. Germinal cells are surrounded throughout their development by the filopodia of interstitial cells. Adjacent interstitial cells are interconnected, and thus form a structural network within the testis. Positionally and functionally, the interstitial cells resemble Sertoli cells; however, their origin, behavior and ultrastructure are different in many ways. Spermatogenesis includes a series of cyclical changes (aspermatogenic phase, proliferative phase, differentiative phase, and evacuative phase). Within a single testis, the resulting production of sperm is in short pulses, but if all 10 testes are taken together, sperm are produced continuously throughout the year. The events of spermiogenesis closely follow those that have been described in other echinoderms. However, we have provided new information on the release of excess cell membranes and the fusion process of mitochondria. The mature spermatozoa of A. squamata are flagellated and motile, and have "primitive" structural features, in spite of the fact that they fertilize the eggs inside the genital bursae. The spermatozoa do not, as was previously thought, enter the bursa by rupture of the adjacent walls. Instead, they are ejaculated through a gonoduct into the rapid incurrent flow of water entering the bursa.
