Yam (Dioscorea opposita) is a kind of vegetables with important nutritional, medicinal and economic value. To reveal the relationship between starch synthesis and gene expression in yam tubers at gene transcription level, transcriptome profiling was conducted by RNA-Seq in Bikeqi yam (Dioscorea opposita Thunb.) tubers at five key developmental stages (105, 120, 135, 150, and 165 days after sowing, DAS). Based on transcriptome sequencing data, a total of 45,867 unigenes were obtained. The results showed that 135 days after sowing are the key period of starch accumulation. During yam tuber development, 1,941 candidate differentially expressed genes (DEGs) were successfully classified into three GO categories, respectively, and there were 292, 267 and 478 unigenes in cellular component, molecular function and biological process. There were 767, 90 and 73 DEGs enriched in metabolic, plant hormone signal transduction and Plant-pathogen interaction pathway by Kyoto Encyclopedia of Genes and Genomes (KEGG), individually. Especially 72 DEGs were enriched in starch and sucrose metabolism pathway. In this pathway, the metabolic process was mainly positive regulated by genes encoding sucrose synthase, glucose-1-phosphate adenylyltransferase, alpha-trehalase, and so on. There was negative regulated by genes encoding beta-glucosidase. 10 DEGs involved in starch synthesis were selected to prove the accuracy of the RNA-Seq data by qPCR, 85% (34/40) of the results were consistent. The results lay a theoretical foundation be used for further understanding the starch synthesis mechanism of yam tubers development and accelerating breeding progress.

Low light stress increases the chalkiness of rice; however, this effect has not been fully characterized. In this study, we demonstrated that low light resulted in markedly decreased activity of ADP-glucose pyrophosphorylase in the grains and those of sucrose synthase and soluble starch synthase in the early period of grain filling. Furthermore, low light also resulted in decreased activities of granule-bound starch synthase and starch branching enzyme in the late period of grain filling. Therefore, the maximum and mean grain filling rates were reduced but the time to reach the maximum grain filling rates and effective grain filling period were increased by low light. Thus, it significantly decreased the grain weight at the maximum grain filling rate and grain weight and retarded the endosperm growth and development, leading to a loose arrangement of the amyloplasts and an increase in the chalkiness of the rice grains. Compared to the grains at the top panicle part, low light led to a greater decrease in the grain weight at the maximum grain filling rate and time to reach the grain weight at the maximum grain filling rate at the bottom panicle part, which contributed to an increase in chalkiness by increasing the rates of different chalky types at the bottom panicle part. In conclusion, low light disturbed starch synthesis in grains, thereby impeding the grain filling progress and increasing chalkiness, particularly for grains at the bottom panicle part.

Improving grain filling in the presernt farming systems is crucial where grain filling is a concern due to the extreme use of chemical fertilizers (CF). A field experiment was conducted at the experimental station of Guangxi University, China in 2019 to test the hypothesis that cattle manure (CM) and poultry manure (PM) combined with CF could improve rice grain filling rate, yield, biochemical and qualitative attributes. A total of six treatments, i.e., no fertilizer (T1), 100% CF (T2), 60% CM + 40% CF (T3), 30% CM + 70% CF (T4), 60% PM + 40% CF (T5), and 30% PM + 70% CF (T6) were used in this study. Results showed that the combined treatment T6increased starch metabolizing enzymes activity (SMEs), such as ADP-glucose phosphorylase (ADGPase) by 8 and 12%, soluble starch synthase (SSS) by 7 and 10%, granule bound starch synthesis (GBSS) by 7 and 9%, and starch branching enzyme (SBE) by 14 and 21% in the early and late seasons, respectively, compared with T2. Similarly, higher rice grain yield, grain filling rate, starch, and amylose content were also recorded in combined treatments. In terms of seasons, higher activity of SMEs , grain starch, and amylose content was noted in the late-season compared to the early season. The increment in these traits was mainly attributed to a lower temperature in the late season during the grain filling period. Furthermore, our results suggested that an increment in starch accumulation and grain filling rate were mainly associated with the enhanced sink capacity by regulating key enzyme activities involved in Suc-to-starch conversion. In-addition, RT-qPCR analysis showed higher expression levels of AGPS2b, SSS1, GBSS1, and GBSE11b genes, which resultantly increased the activities of SMEs during the grain filling period under combined treatments. Linear regression analysis revealed that the activity of ADGPase, SSS, GBSS, and SBE were highly positively correlated with starch and amylose accumulation. Thus, we concluded that a combination of 30% N from PM or CM with 70% N from CF is a promising option in terms of improving rice grain yield and quality. Our study provides a sustainable fertilizer management strategy to enhance rice grain yield and quality at the lowest environmental cost.

Maize (Zea mays L.) is one of the most important food crops in the world, and starch is the main component of its endosperm. Transcriptional regulation plays a vital role in starch biosynthesis. However, it is not well understood in maize. We report the identification of the transcription factor ZmNAC126 and its role in regulation of starch synthesis in maize. Transcriptional expression of ZmNAC126 was higher in maize endosperm and kernels than in roots or stems. ZmNAC126 shared a similar expression pattern with starch synthesis genes during seed development, and its expression pattern was also consistent with the accumulation of starch. ZmNAC126 is a typical transcription factor with a transactivation domain between positions 201 and 227 of the amino acid sequence, is located in the nucleus, and binds to CACG repeats in vitro. Yeast one-hybrid assay revealed that ZmNAC126 bound the promoters of ZmGBSSI, ZmSSIIa, ZmSSIV, ZmISA1, and ZmISA2. Transient overexpression of ZmNAC126 in maize endosperm increased the activities of promoters pZmSh2, pZmBt2, pZmGBSSI, pZmSSIIIa, and pZmBT1 but inhibited the activities of pZmISA1 and pZmISA2. ZmNAC126 thus acts in starch synthesis by transcriptionally regulating targeted starch synthesis-related genes in maize kernels.

Starch directly determines the grain yield and quality. The key enzymes participating in the process of starch synthesis have been cloned and characterized. Nevertheless, the regulatory mechanisms of starch synthesis remain unclear. In this study, we identified a novel starch regulatory gene, ZmCBM48-1, which contained a carbohydrate-binding module 48 (CBM48) domain. ZmCBM48-1 was highly expressed in the maize endosperm and was localized in the plastids. Compared with the wild type lines, the overexpression of ZmCBM48-1 in rice altered the grain size and 1000-grain weight, increased the starch content, and decreased the soluble sugar content. Additionally, the transgenic rice seeds exhibited an alterant endosperm cell shape and starch structure. Meanwhile, the physicochemical characteristics (gelatinization properties) of starch were influenced in the transgenic lines of the endosperm compared with the wild type seeds. Furthermore, ZmCBM48-1 played a positive regulatory role in the starch synthesis pathway by up-regulating several starch synthesis-related genes. Collectively, the results presented here suggest that ZmCBM48-1 acts as a key regulatory factor in starch synthesis, and could be helpful for devising strategies for modulating starch production for a high yield and good quality in maize endosperm.

The genetic diversity of 36 rice landraces and 43 breeding materials in the upper reaches of the Yangtze River in China was studied by intragenic molecular markers of 26 starch synthesis-related loci. And research on quality traits such as the amylose content (AC), gel consistency (GC) and alkali spreading value (ASV) to analyze genetic differences in quality traits. The results showed that the number of alleles, average gene diversity and polymorphism information content values of landraces were higher than those of breeding materials. The genetic similarity coefficient (GS) of 79 rice materials ranged from 0.392 to 1, with an average of 0.757.There were significant variations in the quality traits of rice landraces and breeding materials, and the high-quality compliance rates were low, only 6.3% of the varieties have an amylose content that reached grade 1. The results of cluster analysis and population structure analysis are generally consistent; that is, the two resource types are closely related and cannot be clustered independently. This study can provide a basis for genetic improvement of rice starch quality. Make full use of the quality genetic diversity of landraces in modern breeding work, further broaden the genetic base of rice and improve rice quality.

Starch is a major component of wheat (Triticum aestivum L.) endosperm and is an important part of the human diet. The functions of many starch synthesis genes have been elucidated. However, little is known about their regulatory mechanisms in wheat. Here, we identified a novel NAC transcription factor, TaNAC019-A1 (TraesCS3A02G077900), that negatively regulates starch synthesis in wheat and rice (Oryza sativa L.) endosperms. TaNAC019-A1 was highly expressed in the endosperm of developing grains and encoded a nucleus-localized transcriptional repressor. Overexpression of TaNAC019-A1 in rice and wheat led to significantly reduced starch content, kernel weight, and kernel width. The TaNAC019-A1-overexpression wheat lines had smaller A-type starch granules and fewer B-type starch granules than wild-type. Moreover, TaNAC019-A1 could directly bind to the ‘ACGCAG’ motif in the promoter regions of ADP-glucose pyrophosphorylase small subunit 1 (TaAGPS1-A1, TraesCS7A02G287400) and TaAGPS1-B1 (TraesCS7B02G183300) and repress their expression, thereby inhibiting starch synthesis in wheat endosperm. One haplotype of TaNAC019-B1 (TaNAC019-B1-Hap2, TraesCS3B02G092800) was positively associated with thousand-kernel weight and underwent positive selection during the Chinese wheat breeding process. Our data demonstrate that TaNAC019-A1 is a negative regulator of starch synthesis in wheat endosperm and provide novel insight into wheat yield improvement.

The kernel serves as a storage organ for various nutrients and determines the yield and quality of maize. Understanding the mechanisms regulating kernel development is important for maize production. In this study, a small-kernel mutant smk7a of maize was characterized. Cytological observation suggested that the development of the endosperm and embryo was arrested in smk7a in the early development stage. Biochemical tests revealed that the starch, zein protein, and indole-3-acetic acid (IAA) contents were significantly lower in smk7a compared with wild-type (WT). Consistent with the defective development phenotype, transcriptome analysis of the kernels 12 and 20 days after pollination (DAP) revealed that the starch, zein, and auxin biosynthesis-related genes were dramatically downregulated in smk7a. Genetic mapping indicated that the mutant was controlled by a recessive gene located on chromosome 2. Our results suggest that disrupted nutrition accumulation and auxin synthesis cause the defective endosperm and embryo development of smk7a.

The sago palm (Metroxylon sagu Rottboll) is a tropical halophytic starch-producing, economically important crop palm mainly located in Southeast Asian countries. Recently, a genome survey was conducted on this palm using the Illumina sequencing platform, with a very low (21.5%) BUSCO genome completeness score, and most of them (∼78%) are either fragmented or missing. Thus, in this study, the sago palm genome completeness was further improved with the utilization of the Nanopore sequencing platform that produced longer reads. A hybrid genome assembly was conducted, and the outcome was a much complete sago palm genome with BUSCO completeness achieved at as high as 97.9%, with only ∼2% of them either fragmented or missing. The estimated genome size of the sago palm is 509,812,790 bp in this study. A sum of 33,242 protein-coding genes was revealed from the sago palm genome and around 96.39% of them had been functionally annotated. An investigation on the carbohydrate metabolism KEGG pathways also unearthed that starch synthesis was one of the major sago palm activities. The genome data obtained from this work is indispensable for future molecular evolutionary and genome-wide association studies on the economically important sago palm.

From carbon dioxide to starch: no plants required Many plants turn glucose from photosynthesis into polymers that form insoluble starch granules ideal for long-term energy storage in roots and seeds. Cai et al . developed a hybrid system in which carbon dioxide is reduced to methanol by an inorganic catalyst and then converted by enzymes first to three and six carbon sugar units and then to polymeric starch. This artificial starch anabolic pathway relies on engineered recombinant enzymes from many different source organisms and can be tuned to produce amylose or amylopectin at excellent rates and efficiencies relative to other synthetic carbon fixation systems—and, depending on the metric used, even to field crops. —MAF

WRINKLED1 (WRI1) belongs to AP2/EREBP transcription factor. Its function in dicots for fatty acids synthesis has been deeply studied, but its role in monocot, especially in rice, is still poorly understood. Here, with the overexpression of AtWRI1 in rice, we found its overexpression increased fatty acids content in vegetative organs and seed coat including aleurone layer (SCAL) but decreased fatty acids content in endosperm. Meanwhile, the overexpression of AtWRI1 increased starch content in endosperm. These results provide a new insight into the function of AtWRI1in monocot and make a previous basement for the study of the connection of fatty acids and starch synthesis in rice. The overexpression of AtWRI1 in rice enhances fatty acids content in vegetative tissues, seed coat including aleurone layer of rice but decreases fatty acids content in the endosperm. Meanwhile, the starch content in the endosperm of WRI1 rice is increased.

An experiment on an early indica rice cv. ‘Shenyou9576’ was conducted in the Key Laboratory of Phytohormones and Growth Development of Hunan Agricultural University, Changsha, Hunan, PR China in 2014 to investigate the influence of varying post-anthesis temperatures on chalkiness rate, head rice rate, and on major 6 starch synthesis enzymes i.e., SuSy (EC 1.9.3.1), ADPG-Ppas (EC 2.7.7.27), SSS (EC 2.4.1.21) and GBSS, (EC 2.4.1.21), SBE (EC 2.4.1.18) and SDBE (EC 3.2.1.70). The treatments comprised of three temperature regimes which are designated as the high (35/28oC- day/night), low (25/20oC- day/night) and natural condition (35/25oC-day/night) as the control. Under high temperature maximum chalkiness rate was 61.11% and minimum was 22.59% under low temperature treatment. The lowest head rice rate was 42.76% under high temperature treatment followed by 49.91% in the control, while the highest rate was 62.33% under low temperature treatment. Maximum grain filling rate (Gmax) was found highest (1.69 mg/day) in the high temperature and average grain filling rate (Gavg) was found highest (1.36 mg/day) under the control. The activity of SuSy, ADPG-Ppase, SSS and GBSS were decreased gradually from 14 to 35 days after flowering (DAF). Irrespective of the treatments, an increasing trend of ADPG-Ppase activity was observed from 7 to 14 DAF and then declined. Correlation between the chalkiness and the enzymes activity of SuSy, ADPG-Ppase and SSS were significantly negative at 21, 28 and 35 DAFs, i.e., higher activity of SuSy, ADPG-Ppase and SSS at the mid-late to the late caryopsis development stage mediated by low temperature treatment played an important role for the reduction of chalkiness. The correlation between GBSS activity and chalkiness was significantly negative and stronger at 14, 21 and 28 DAF indicating that GBSS played a cardinal role to reduce chalkiness in the mid to mid-late stage of rice grain development. Significantly negative correlation was found between starch branching enzyme (SBE) and chalkiness at 21, 28 and 35 DAF, i.e., the higher SBE activity under low temperature treatment at the later grain filling stage also had a positive role in reduction of chalkiness. Significance Most forms of life on Earth cannot exist without photosynthesis. Our food and atmosphere depend on it. To obtain high photosynthetic yields, light energy must be efficiently coupled to the fixation of CO2 into organic molecules. Suboptimal environmental conditions can severely impact the conversion of light energy to biomass and lead to reactive oxygen production, which in turn can cause cellular damage and loss of productivity. Hence, plants, algae, and photosynthetic bacteria have evolved a network of alternative outlets to sustain the flow of photosynthetically derived electrons. Our work is focused on the nature and integration of these outlets, which will inform a rational engineering of crop plants and algae to optimize photosynthesis and meet the increased global demand for food., Environmental stresses dramatically impact the balance between the production of photosynthetically derived energetic electrons and Calvin–Benson–Bassham cycle (CBBC) activity; an imbalance promotes accumulation of reactive oxygen species and causes cell damage. Hence, photosynthetic organisms have developed several strategies to route electrons toward alternative outlets that allow for storage or harmless dissipation of their energy. In this work, we explore the activities of three essential outlets associated with Chlamydomonas reinhardtii photosynthetic electron transport: (i) reduction of O2 to H2O through flavodiiron proteins (FLVs) and (ii) plastid terminal oxidases (PTOX) and (iii) the synthesis of starch. Real-time measurements of O2 exchange have demonstrated that FLVs immediately engage during dark-to-light transitions, allowing electron transport when the CBBC is not fully activated. Under these conditions, we quantified maximal FLV activity and its overall capacity to direct photosynthetic electrons toward O2 reduction. However, when starch synthesis is compromised, a greater proportion of the electrons is directed toward O2 reduction through both the FLVs and PTOX, suggesting an important role for starch synthesis in priming/regulating CBBC and electron transport. Moreover, partitioning energized electrons between sustainable (starch; energetic electrons are recaptured) and nonsustainable (H2O; energetic electrons are not recaptured) outlets is part of the energy management strategy of photosynthetic organisms that allows them to cope with the fluctuating conditions encountered in nature. Finally, unmasking the repertoire and control of such energetic reactions offers new directions for rational redesign and optimization of photosynthesis to satisfy global demands for food and other resources.

Summary Starch is the most important form of energy storage in cereal crops. Many key enzymes involved in starch biosynthesis have been identified. However, the molecular mechanisms underlying the regulation of starch biosynthesis are largely unknown. In this study, we isolated a novel floury endosperm rice (Oryza sativa) mutant flo16 with defective starch grain (SG) formation. The amylose content and amylopectin structure were both altered in the flo16 mutant. Map‐based cloning and complementation tests demonstrated that FLO16 encodes a NAD‐dependent cytosolic malate dehydrogenase (CMDH). The ATP contents were decreased in the mutant, resulting in significant reductions in the activity of starch synthesis‐related enzymes. Our results indicated that FLO16 plays a critical role in redox homeostasis that is important for compound SG formation and subsequent starch biosynthesis in rice endosperm. Overexpression of FLO16 significantly improved grain weight, suggesting a possible application of FLO16 in rice breeding. These findings provide a novel insight into the regulation of starch synthesis and seed development in rice.

The dynamics of starch accumulation during the transition of plants from vegetation to the dormant period in the annual shoots of various groups of garden roses grafted on a Centifolia rose or on a Rosa canina, and on their own roots was studied. It was established that in ungrafted groud cover roses, after the maximum accumulation of starch in September, its hydrolysis is observed in October. Grafting on a Centifolia rose significantly speeds up this process. In roots of polyanthus and climbing roses, starch synthesis continues in October. In these roses grafted onto a Centifolia rose or on a Rosa canina, the dynamics of starch accumulation changes, which is expressed either in the suspension of its synthesis or in the starting the hydrolysis. In general, both in ground cover and in polyanthus and climbing roses, grafting provided better preparation of grafted material for winter than was observed in roses on their own roots. The length of the day (photoperiod) is a robust seasonal signal originated by earth orbital and translational movements, a resilient external cue to the global climate change, and a predictable hint to initiate or complete different developmental programs. In eukaryotic algae, the gene expression network that controls the cellular response to photoperiod also regulates other basic physiological functions such as starch synthesis or redox homeostasis. Land plants, evolving in a novel and demanding environment, imbued these external signals within the regulatory networks controlling organogenesis and developmental programs. Unlike algae that largely have to deal with cellular physical cues, within the course of evolution land plants had to transfer this external information from the receiving organs to the target tissues, and mobile signals such as hormones were recruited and incorporated in the regulomes. Control of senescence by photoperiod, as suggested in this perspective, would be an accurate way to feed seasonal information into a newly developed function (senescence) using an ancient route (photoperiodic signaling). This way, the plant would assure that two coordinated aspects of development such as flowering and organ senescence were sequentially controlled. As in the case of senescence, there is growing evidence to support the idea that harnessing the reliability of photoperiod regulation over other, more labile signaling pathways could be used as a robust breeding tool to enhance plants against the harmful effects of climate change.

Background Chestnut seeds are important kinds of edible nuts rich in starch and protein. The characteristics and nutrient contents of chestnut have been found to show obvious metaxenia effects in previous studies. To improve the understanding of the effect of metaxenia on chestnut starch and sucrose metabolism, this study used three varieties of chestnut, ‘Yongfeng 1’, ‘YongRen Zao’ and ‘Yimen 1’, as male parents to pollinate the female parent, ‘Yongfeng 1’, and investigated the mechanisms of starch and sucrose metabolism in three starch accumulation stages (70 (S1), 82 (S2), and 94 (S3) days after pollination, DAP) in chestnut seed kernels. Result Most carbohydrate metabolism genes were highly expressed in YFF (self-pollinated ‘Yongfeng 1’) in stage S2 and in YFR (‘Yongfeng 1’ × ‘Yongren Zao’) and YFM (‘Yongfeng 1’ × ‘Yimen 1’) in stage S3. In stage S3, hub genes encoding HSF_DNA-binding, ACT, Pkinase, and LIM proteins and four transcription factors were highly expressed, with YFF showing the highest expression, followed by YFR and YFM. In addition, transcriptome analysis of the kernels at 70, 82 and 94 DAP showed that the starch granule-bound starch synthase (EC 2.4.1.242) and ADP-glucose pyrophosphorylase (EC 2.7 .7.27) genes were actively expressed at 94 DAF. Chestnut seeds regulate the accumulation of soluble sugars, reducing sugars and starch by controlling glycosyl transferase and hydrolysis activity during development. Conclusion These results and resources have important guiding significance for further research on starch and sucrose metabolism and other types of metabolism related to chestnut metaxenia.

Background As the main form of photoassimilates transported from vegetative tissues to the reproductive organs, sucrose and its degradation products are crucial for cell fate determination and development of maize kernels. Despite the relevance of sucrose synthase SH1 (shrunken 1)-mediated release of hexoses for kernel development, the underlying physiological and molecular mechanisms are not yet well understood in maize (Zea mays). Results Here, we identified a new allelic mutant of SH1 generated by EMS mutagenesis, designated as sh1*. The mutation of SH1 caused more than 90% loss of sucrose synthase activity in sh1* endosperm, which resulted in a significant reduction in starch contents while a dramatic increase in soluble sugars. As a result, an extremely high osmolality in endosperm cells of sh1* was generated, which caused kernel swelling and affected the seed development. Quantitative measurement of phosphorylated sugars showed that Glc-1-P in endosperm of sh1* (17 μg g− 1 FW) was only 5.2% of that of wild-type (326 μg g− 1 FW). As a direct source of starch synthesis, the decrease of Glc-1-P may cause a significant reduction in carbohydrates that flow to starch synthesis, ultimately contributing to the defects in starch granule development and reduction of starch content. Conclusions Our results demonstrated that SH1-mediated sucrose degradation is critical for maize kernel development and starch synthesis by regulating the flow of carbohydrates and maintaining the balance of osmotic potential.

Background: Euryale ferox Salisb. is an annual aquatic herb and the only species belonging to the genus Euryale in the Nymphaeaceae family. E. ferox seeds are used in medicine and diets. Starch is the main factor affecting E. ferox seed quality, but its regulatory mechanism has not been elucidated. Results: Herein, four time points of seed development, including after flowering T1 (10 days), T2 (20 days), T3 (30 days) and T4 (40 days), were investigated by using RNA-Seq and iTRAQ technology. Using weighted gene co-expression network analyses (WGCNAs), co-expressed genes and hub genes were identified for each module. Of particular importance are the discoveries of specific modules for seed starch during the seed developmental stages. The candidate regulators of seed starch are involved in the hormonal signaling pathways. Three ABA signaling receptor kinases, EfPYR1, EfSnRK2.1 and EfSnRK2.2, were identified as hub genes functioning in starch synthesis during the seed maturation process. The changes in expression pattern, ABA and starch content also indicated that ABA is positively correlated with starch. Conclusions: Together, these results indicate that E. ferox seed accumulation of starch is promoted by ABA, providing new insights into the regulatory mechanism of starch synthesis in E. ferox seeds.

Research in the past decade has uncovered new and surprising information about the pathways of starch synthesis and degradation. This includes the discovery of previously unsuspected protein families required both for processes and for the long-sought mechanism of initiation of starch granules. There is also growing recognition of the central role of leaf starch turnover in making carbon available for growth across the day-night cycle. Sophisticated systems-level control mechanisms involving the circadian clock set rates of nighttime starch mobilization that maintain a steady supply of carbon until dawn and modulate partitioning of photosynthate into starch in the light, optimizing the fraction of assimilated carbon that can be used for growth. These discoveries also uncover complexities: Results from experiments with Arabidopsis leaves in conventional controlled environments are not necessarily applicable to other organs or species or to growth in natural, fluctuating environments.

has attracted tremendous interest because of its high contents of glucomannan and starch. Very few genes regulating glucomannan and starch were reported in Amorphophallus. In this study, an ADP-glucose pyrophosphorylase (AGP) gene that plays a significant role in plant starch synthesis was cloned from Amorphophallus muelleri. It was shown that it encoded a predicted protein containing a conserved plant ADP-Glucose-PP repeat domain and seven potential ligand-binding sites. The real-time quantitative PCR showed that AmAGP was most abundant in tubers, and it was positively correlated with starch content. Additionally, its influencers about temperature and exogenous plant hormone were also discussed, showing that AmAGP expressed highly in tubers under treatments using 25°C and IAA. Furthermore, starch content was closely related to AmAGP expression level, suggesting that AmAGP was involved in the regulation of starch synthesis in A. muelleri. Therefore, identifying the sequence of AmAGP and its expression pattern during tuber enlarging and the changes of its transcript levels in response to temperature and plant hormones would contribute to a better understanding of starch synthesis, and also providing a reference information for future preferable breeding for obtaining more starch or more glucomannan in Amorphophallus.

The pyrenoid is an evolutionary adaptation that enables algae to fix inorganic carbon more efficiently in CO2-limited environments. The genesis of this contemporary definition is nearly two and half centuries old. One of the earliest records of pyrenoids are un-annotated puncta in a drawing of Conferva jugalis (now Spirogyra) in Flora Danica (Muller 1782). The word pyrenoid, from the Greek πυρην (pyren, kernel), was coined by Schmitz a century later (1882) in a remarkable monograph on algal chloroplasts. Schmitz observed that pyrenoids are chloroplast inclusions, that divide as chloroplasts and cells divide. He also diagnosed pyrenoid presence/absence through numerous algal lineages and astutely recorded that carbohydrate deposits around pyrenoids are highly variable, both across species and as plastic response to changes in growth conditions. Most of his observations have withstood the test of time, although the speculation that pyrenoids are akin to nucleoli was later proven unfounded (Martin and Kowallik 1999). It was the definition as a site of starch synthesis (although not all algae synthesise starch-defining α-1,4-glycosidic bonds) however that came to dominate the literature until the early 1970s and survives to these days in some textbooks and refereed papers.

Free (soluble, non-protein) asparagine concentration can increase many-fold in wheat grain in response to sulphur deficiency. This exacerbates a major food safety and regulatory compliance problem for the food industry because free asparagine may be converted to the carcinogenic contaminant, acrylamide, during baking and processing. Here, we describe the predominant route for the conversion of asparagine to acrylamide in the Maillard reaction. The effect of sulphur deficiency and its interaction with nitrogen availability is reviewed, and we reiterate our advice that sulphur should be applied to wheat being grown for human consumption at a rate of 20 kg per hectare. We describe the genetic control of free asparagine accumulation, including genes that encode metabolic enzymes (asparagine synthetase, glutamine synthetase, glutamate synthetase, and asparaginase), regulatory protein kinases (sucrose nonfermenting-1 (SNF1)-related protein kinase-1 (SnRK1) and general control nonderepressible-2 (GCN2)), and basic leucine zipper (bZIP) transcription factors, and how this genetic control responds to sulphur, highlighting the importance of asparagine synthetase-2 (ASN2) expression in the embryo. We show that expression of glutamate-cysteine ligase is reduced in response to sulphur deficiency, probably compromising glutathione synthesis. Finally, we describe unexpected effects of sulphur deficiency on carbon metabolism in the endosperm, with large increases in expression of sucrose synthase-2 (SuSy2) and starch synthases.

Leaf veins play a critical role in resource supplication and photosynthate translocation; thus, it is considered as an important agricultural trait for crop breeding. The rice minor veins are parallelly grown along all the parts of the leaf from base to tip. To understand the process of minor vein development, anatomy analysis was performed to reveal the initiation and development of minor veins in rice leaf. The frequency of minor vein initiation follows a decreased tendency from leaf base to tip. An iTRAQ-based proteomics analysis was performed in rice leaf sections. Photosynthesis- and carbon fixation-related proteins accumulated a high level in the middle part of leaves. Furthermore, marker proteins involved in sucrose degradation and starch synthesis were accumulated into initiation and mature parts of minor veins, respectively. It suggests a different source-sink activity in the initiation and mature parts of minor veins in terms of photosynthate translocation. The identified proteins are candidate markers for small vein initiation in rice leaves.

Wheat (Triticum aestivum L.) is a temperate cereal with an optimum temperature range of 15–22°C during the grain filling stage. Heat stress is one of the major environmental constraints for wheat production worldwide. Temperatures above 25°C during the grain filling stage significantly reduced wheat yield and quality. This reduction was reported due to the inactivation of the soluble starch synthase, a key heat-labile enzyme in starch transformation of wheat endosperm. To improve wheat productivity under heat stress, the rice soluble starch synthase I, under the control of either a constitutive promoter or an endosperm-specific promoter, was expressed in wheat and the transgenic lines were monitored for expression and the effects on yield-related traits. The results showed that the transgenic wheat events expressed rice soluble starch synthase I at a high level after four generations, and transgenic plants produced grains of greater weight during heat stress. Under heat stress conditions, the thousand kernel weight increased 21–34% in T2 and T3 transgenic plants compared to the non-transgenic control plants. In addition, the photosynthetic duration of transgenic wheat was longer than in non-transgenic controls. This study demonstrated that the engineering of a heat tolerant soluble starch synthase gene can be a potential strategy to improve wheat yield under heat stress conditions.

Nitrogen fertilizer is a crucial factor affecting the growth and grain quality of Tartary buckwheat. This study was to investigate the synthesis, accumulation, and physicochemical properties of Tartary buckwheat starches under four nitrogen levels (0, 90, 180, 270 kg N ha−1). The results showed that activities of four key enzymes, starch contents all first increased and then decreased with increasing nitrogen levels, and peaked at 180 kg N ha−1. All the starches showed typical A-type, while higher nitrogen levels significantly increased the relative crystallinity. The viscosities significantly decreased, onset, peak, and conclusion first decreased and then increased, while pasting temperature and gelatinization enthalpy increased with increasing nitrogen levels. Nitrogen fertilizer and year had significant effects on the synthesis, accumulation and physicochemical properties of Tartary buckwheat starch, and the nitrogen level of 180 kg N ha−1 was more suitable for planting in the northern area of the Loess Plateau.

RNAi-repression of vacuolar HvSUT2 in transgenic barley demonstrates its indispensable role for proper grain filling and for the control of sucrose homeostasis in concert with the plasma-membrane localised HvSUT1., Sucrose transport and partitioning are crucial for seed filling. While many plasma-membrane-localised sucrose transporters (SUT1 family members) have been analysed in seeds, the functions of vacuolar SUT2 members are still obscure. In barley grains, expression of HvSUT1 and HvSUT2 overlap temporally and spatially, suggesting concerted functions to regulate sucrose homeostasis. Using HvSUT2-RNAi plants, we found that grains were also deficient in HvSUT1 expression and seemingly sucrose-limited during mid-to-late grain filling. Transgenic endosperms accumulated less starch and dry weight, although overall sucrose and hexose contents were higher. Comprehensive transcript and metabolite profiling revealed that genes related to glycolysis, the tricarboxylic acid cycle, starch and amino acid synthesis, grain maturation, and abscisic acid signalling were down-regulated together with most glycolytic intermediates and amino acids. Sucrose was increased along the sucrose delivery route in the nucellar projection, the endosperm transfer cells, and the starchy endosperm, indicating that suppressed transporter activity diminished sucrose efflux from vacuoles, which generated sugar deficiency in the cytoplasm. Thus, endosperm vacuoles may buffer sucrose concentrations to regulate homeostasis at grain filling. Transcriptional changes revealed that limited endosperm sucrose initiated sugar starvation responses, such as sugar recycling from starch, hemicelluloses and celluloses together with vacuolar protein degradation, thereby supporting formation of nucleotide sugars. Barley endosperm cells can thus suppress certain pathways to retrieve resources to maintain essential cell functions.

The common buckwheat ‘cv. Fengtian 1’ (FT1), ‘cv. Yuqiao 4’ (YQ4), ‘cv. Dingtian 2 (DT2)’, and ‘cv. Tongliao (TL)’ were selected to investigate the characteristics of starch synthesis and grain filling. The chlorophyll and chlorophyll a / b contents of leaves in the third stem showed a steady downward trend with the growth of common buckwheat. Whereas, FT1, YQ4 and DT2 showed higher chlorophyll content than that of TL at 7d and 14d. The activity of soluble starch synthase(SSS) and adenosine diphosphate glucose pyrophosphorylase (ADPGP) in the four tested varieties showed a rapid upward trend until 14 days after heading but then rapidly decreased until 21 days after heading. The average adenosine diphosphate glucose pyrophosphorylase activities of FT1, YQ4 and DT2 were higher than TL, while the average soluble starch synthase activities were similar among the four tested varieties. Consistently, the four tested varieties showed rapid accumulation of starch, amylose, and amylopectin during the prefilling stage, but no difference of starch content was found at maturity. The filling process of the four common buckwheat varieties exhibited an “S” curve. The Richards equation was utilized to evaluate the grain-filling process of common buckwheat. YQ4 showed the largest values of initial growth power ( R 0 ). The time reaching the maximum grain-filling rate ( t max.G ) of FT1 was the longest, and the maximum grain-filling rate ( G max ) of DT2 was the fastest.

Two of the most economically important plants in the Artocarpus genus are jackfruit (A. heterophyllus Lam.) and breadfruit (A. altilis (Parkinson) Fosberg). Both species are long-lived trees that have been cultivated for thousands of years in their native regions. Today they are grown throughout tropical to subtropical areas as an important source of starch and other valuable nutrients. There are hundreds of breadfruit varieties that are native to Oceania, of which the most commonly distributed types are seedless triploids. Jackfruit is likely native to the Western Ghats of India and produces one of the largest tree-borne fruit structures (reaching up to 45 kg). To-date, there is limited genomic information for these two economically important species. Here, we generated 273 Gb and 227 Gb of raw data from jackfruit and breadfruit, respectively. The high-quality reads from jackfruit were assembled into 162,440 scaffolds totaling 982 Mb with 35,858 genes. Similarly, the breadfruit reads were assembled into 180,971 scaffolds totaling 833 Mb with 34,010 genes. A total of 2822 and 2034 expanded gene families were found in jackfruit and breadfruit, respectively, enriched in pathways including starch and sucrose metabolism, photosynthesis, and others. The copy number of several starch synthesis-related genes were found to be increased in jackfruit and breadfruit compared to closely-related species, and the tissue-specific expression might imply their sugar-rich and starch-rich characteristics. Overall, the publication of high-quality genomes for jackfruit and breadfruit provides information about their specific composition and the underlying genes involved in sugar and starch metabolism.

Prod? EA GEAPSI INRA; Trees in seasonal climates gauge winter progression to assure vital and productive blooming. However, how dormant plants asses environmental conditions remains obscure. We postulated that it involves the energetic reserves required for bloom, and therefore studied winter carbohydrate metabolism in deciduous trees. We quantified non-structural carbohydrates throughout winter in almond, peach, and pistachio trees in California and Israel and characterized winter metabolism. We constructed a carbohydrate-temperature (C–T) model that projects changes in starch and soluble carbohydrate concentrations by temperature mediated kinetics. Then, we tested the C–T model projections of bloom times by 20 years of temperature and phenology records from California. The C–T model attributes winter carbohydrate regulation in dormant trees to continuous updates of metabolic pathways. The model projects a surge in starch synthesis at the end of winter, and critically low concentrations of soluble carbohydrates, that trigger bloom. This is supported by field measurements of starch accumulation at the end of winter (˜50 mg g−1 DW in almonds) that preceded bloom by ˜10 days. The C–T model provides a physiological framework for bloom forecasts in deciduous orchards. It integrates contrasting notions of chill and heat and elucidates why abnormal winter temperatures may compromise bloom in deciduous orchards.

Developing plants from in vitro culture of microspores or immature pollen grains (androgenesis) is a highly genotype-dependent process whose effectiveness in cereals is significantly reduced by occurrence of albino regenerants. Here, we examined a hypothesis that the molecular differentiation of plastids in barley microspores prior to in vitro culture affects the genotype ability to regenerate green plants in culture. At the mid-to-late uninucleate (ML) stage, routinely used to initiate microspore culture, the expression of most genes involved in plastid transcription, translation and starch synthesis was significantly higher in microspores of barley cv. ‘Mercada’ producing 90% albino regenerants, than in cv. ‘Jersey’ that developed 90% green regenerants. The ML microspores of cv. ‘Mercada’ contained a large proportion of amyloplasts filled with starch, while in cv. ‘Jersey’ there were only proplastids. Using additional spring barley genotypes that differed in their ability to regenerate green plants we confirmed the correlation between plastid differentiation prior to culture and albino regeneration in culture. The expression of GBSSI gene (Granule-bound starch synthaseI) in early-mid (EM) microspores was a good marker of a genotype potential to produce green regenerants during androgenesis. Initiating culture from EM microspores that significantly improved regeneration of green plants may overcome the problem of albinism.

Hierarchical agglomerative clustering of diel metabolite data from photosynthetic and central C-metabolism in the CAM plant Phalaenopsis ‘Edessa’ reveals consistencies and marked differences in carbon and energy homeostasis in CAM and C3 plants., Crassulacean acid metabolism (CAM) is a major adaptation of photosynthesis that involves temporally separated phases of CO2 fixation and accumulation of organic acids at night, followed by decarboxylation and refixation of CO2 by the classical C3 pathway during the day. Transitory reserves such as soluble sugars or starch are degraded at night to provide the phosphoenolpyruvate (PEP) and energy needed for initial carboxylation by PEP carboxylase. The primary photosynthetic pathways in CAM species are well known, but their integration with other pathways of central C metabolism during different phases of the diel light–dark cycle is poorly understood. Gas exchange was measured in leaves of the CAM orchid Phalaenopsis ‘Edessa’ and leaves were sampled every 2 h during a complete 12-h light–12-h dark cycle for metabolite analysis. A hierarchical agglomerative clustering approach was employed to explore the diel dynamics and relationships of metabolites in this CAM species, and compare these with those in model C3 species. High levels of 3-phosphoglycerate (3PGA) in the light activated ADP-glucose pyrophosphorylase, thereby enhancing production of ADP-glucose, the substrate for starch synthesis. Trehalose 6-phosphate (T6P), a sugar signalling metabolite, was also correlated with ADP-glucose, 3PGA and PEP, but not sucrose, over the diel cycle. Whether or not this indicates a different function of T6P in CAM plants is discussed. T6P levels were low at night, suggesting that starch degradation is regulated primarily by circadian clock-dependent mechanisms. During the lag in starch degradation at dusk, carbon and energy could be supplied by rapid consumption of a large pool of aconitate that accumulates in the light. Our study showed similarities in the diel dynamics and relationships between many photosynthetic metabolites in CAM and C3 plants, but also revealed some major differences reflecting the specialized metabolic fluxes in CAM plants, especially during light–dark transitions and at night.

Rice grains accumulate starch as their major storage reserve whose biosynthesis is sensitive to heat. ADP-glucose pyrophosphorylase (AGPase) is among the starch biosynthetic enzymes severely affected by heat stress during seed maturation. To increase the heat tolerance of the rice enzyme, we engineered two dominant AGPase subunits expressed in developing endosperm, the large (L2) and small (S2b) subunits of the cytosol-specific AGPase. Bacterial expression of the rice S2b with the rice L2, potato tuber LS (pLS), or with the mosaic rice-potato large subunits, L2-pLS and pLS-L2, produced heat-sensitive recombinant enzymes, which retained less than 10% of their enzyme activities after 5 min incubation at 55°C. However, assembly of the rice L2 with the potato tuber SS (pSS) showed significantly increased heat stability comparable to the heat-stable potato pLS/pSS. The S2b assembled with the mosaic L2-pLS subunit showed 3-fold higher sensitivity to 3-PGA than L2/S2b, whereas the counterpart mosaic pLS-L2/S2b showed 225-fold lower sensitivity. Introduction of a QTC motif into S2b created an N-terminal disulfide linkage that was cleaved by dithiothreitol reduction. The QTC enzyme showed moderate heat stability but was not as stable as the potato AGPase. While the QTC AGPase exhibited approximately fourfold increase in 3-PGA sensitivity, its substrate affinities were largely unchanged. Random mutagenesis of S2bQTC produced six mutant lines with elevated production of glycogen in bacteria. All six lines contained a L379F substitution, which conferred enhanced glycogen production in bacteria and increased heat stability. Modeled structure of this mutant enzyme revealed that this highly conserved leucine residue is located in the enzyme’s regulatory pocket that provides interaction sites for activators and inhibitors. Our molecular dynamic simulation analysis suggests that introduction of the QTC motif and the L379F mutation improves enzyme heat stability by stabilizing their backbone structures possibly due to the increased number of H-bonds between the small subunits and increased intermolecular interactions between the two SSs and two LSs at elevated temperature.

Starch synthesis is a complex process that influences crop yield and grain quality in maize. Many key enzymes have been identified in starch biosynthesis; however, the regulatory mechanisms have not been fully elucidated. In this study, we identified a DOF family gene, ZmDOF36, through transcriptome sequencing analysis. Real-time PCR indicated that ZmDOF36 was highly expressed in maize endosperm, with lower expression in leaves and tassels. ZmDOF36 is a typical DOF transcription factor (TF) that is localized to the nucleus and possesses transcriptional activation activity, and its transactivation domain is located in the C-terminus (amino acids 227-351). Overexpression of ZmDOF36 can increase starch content and decrease the contents of soluble sugars and reducing sugars. In addition, abnormal starch structure in transgenic maize was also observed by scanning electron microscopy (SEM). Furthermore, the expression levels of starch synthesis-related genes were up-regulated in ZmDOF36-expressing transgenic maize. ZmDOF36 was also shown to bind directly to the promoters of six starch biosynthesis genes, ZmAGPS1a, ZmAGPL1, ZmGBSSI, ZmSSIIa, ZmISA1, and ZmISA3 in yeast one-hybrid assays. Transient expression assays showed that ZmDOF36 can activate the expression of ZmGBSSI and ZmISA1 in tobacco leaves. Collectively, the results presented here suggest that ZmDOF36 acts as an important regulatory factor in starch synthesis, and could be helpful in devising strategies for modulating starch production in maize endosperm.

Cooking quality is an important attribute in Common/Asian rice (Oryzasativa L.) varieties, being highly dependent on grain starch composition. This composition is known to be highly dependent on a cultivar’s genetics, but the way in which their genes express different phenotypes is not well understood. Further analysis of variation of grain quality genes using new information obtained from the wild relatives of rice should provide important insights into the evolution and potential use of these genetic resources. All analyses were conducted using bioinformatics approaches. The analysis of the protein sequences of grain quality genes across the Oryza suggest that the deletion/mutation of amino acids in active sites result in variations that can negatively affect specific steps of starch biosynthesis in the endosperm. On the other hand, the complete deletion of some genes in the wild species may not affect the amylose content. Here we present new insights for Starch Synthesis-Related Genes (SSRGs) evolution from starch-specific rice phenotypes.

Ulmus pumila L. is an excellent afforestation and biofuel tree that produces high-quality wood, rich in starch. In addition, U. pumila is highly adaptable to adverse environmental conditions, which is conducive to its utilization for vegetating saline soils. However, little is known about the physiological responses and transcriptional regulatory network of U. pumila under salt stress. In this study, we exposed five main cultivars in saline–alkali land (Upu2, 5, 8, 11, and 12) to NaCl stress. Of the five cultivars assessed, Upu11 exhibited the highest salt resistance. Growth and biomass accumulation in Upu11 were promoted under low salt concentrations (&lt, 150 mM). However, after 3 months of continuous treatment with 150 mM NaCl, growth was inhibited, and photosynthesis declined. A transcriptome analysis conducted after 3 months of treatment detected 7009 differentially expressed unigenes (DEGs). The gene annotation indicated that these DEGs were mainly related to photosynthesis and carbon metabolism. Furthermore, PHOTOSYNTHETIC ELECTRON TRANSFERH (UpPETH), an important electron transporter in the photosynthetic electron transport chain, and UpWAXY, a key gene controlling amylose synthesis in the starch synthesis pathway, were identified as hub genes in the gene coexpression network. We identified 25 and 62 unigenes that may interact with PETH and WAXY, respectively. Overexpression of UpPETH and UpWAXY significantly increased the survival rates, net photosynthetic rates, biomass, and starch content of transgenic Arabidopsis plants under salt stress. Our findings clarify the physiological and transcriptional regulators that promote or inhibit growth under environmental stress. The identification of salt-responsive hub genes directly responsible for photosynthesis and starch synthesis or metabolism will provide targets for future genetic improvements.

Commonly, cassava could survive for a moment under drought stress. However, there are limitations to face the drought stress for the growth and productivity then affect starch synthesis. Gunungkidul is one of the biggest cassava producers in Indonesia, in meanwhile cassava production is not able to increase due to the characteristics of karst land topography and rainfed area. Tolerance of drought stress is a complex process relates to physiological and molecular responses. In molecular level, many genes revealed multi-complex responses to drought stress. One of the responsible gene assumed to correlate to drought stress is G-Box Binding Factor (GBF3). The study aims to detect the gene expression of GBF3 in local cassava cultivars of Gunungkidul under drought stress. The result revealed that at moisture levels of 50 percent and 25 percent, GBF3 (Gene Expression of G-Box Binding Factor 3) expressions are identified in the cassava cultivar Gatotkaca, which differed from the detection result of the cassava cultivar Pandesi hijau at all moisture levels. Based on the presence of phenotypic characters and gene expression, the local cultivar of Gatotkaca is assumably to be more drought tolerant than Pandesi Hijau.

Carboxymethyl jackfruit seed starch (CMS) was synthesized under different reaction conditions. The influence of sodium hydroxide concentration, monochloroacetic acid (MCA) concentration, IPA-water ratio, solvent-starch ratio, reaction time, and temperature were evaluated for degree of substitution (DS). Results have shown that the optimal DS of 0.68 was obtained at 50 °C, 90 minutes in solvent-starch ratio and isopropanol-water were 8:1 and 10:0. The ratio of sodium hydroxide and monochloroacetate acid moles to anhydroglucose unit (AGU) moles for the optimal DS were 1 and 1. Scanning electron microscope (SEM) of CMS particles showed the starch grain structure remains the same but the surface appeared many alveolar holes and no longer smooth as MS. Fourier transform infrared spectra (FTIR) of CMS and MS confirmed that carboxymethylation takes place on native starch molecules when the absorption band appears at a wavenumber of 1643 cm−1 corresponding to the vibrations of featured functional C=O group in CMS structure.

This paper analyzes a mathematical model of the photosynthetic carbon metabolism, which incorporates not only the Calvin-Benson cycle, but also another two important metabolic pathways: starch synthesis and photorespiratory pathway. Theoretically, the paper shows the existence of steady states, stability and instability of the steady states, the effects of CO 2 concentration on steady states. Especially, a critical point is found, the system has only one steady state with CO 2 concentration in the left neighborhood of the critical point, but has two with CO 2 concentration in the right neighborhood. In addition, the paper also explores the influence of CO 2 concentration on the efficiency of photosynthesis. These theoretical results not only provide insight to the kinetic behaviors of the photosynthetic carbon metabolism, but also can be used to help improving the efficiency of photosynthesis in plants.

When bean fruits are detached from a plant at 20 days after anthesis (DAA), the material accumulating in the pod is relocalized to the seeds. This mobilization is more active during the first five days after the fruits are removed, which allows some seeds to continue their development. In freshly removed fruits, (14)C-sucrose was evenly distributed among seeds; however, in fruits that were removed three days before, the labeled sugar was concentrated in 1-2 seeds. In addition, in removed pods, embryos dissected from seeds that no longer continue development can assimilate and efficiently use sucrose for protein and starch synthesis. Our results support the hypothesis that most embryos in removed fruits are forced to stop developing by removal of the nutrient supply. We also observed that SnRK1 activity increased in embryos that were subjected to nutrient deprivation, supporting the role of SnRK1 in the metabolic adaptation to stress conditions.

Starch is a water-insoluble polymer of glucose synthesized as discrete granules inside the stroma of plastids in plant cells. Starch reserves provide a source of carbohydrate for immediate growth and development, and act as long term carbon stores in endosperms and seed tissues for growth of the next generation, making starch of huge agricultural importance. The starch granule has a highly complex hierarchical structure arising from the combined actions of a large array of enzymes as well as physicochemical self-assembly mechanisms. Understanding the precise nature of granule architecture, and how both biological and abiotic factors determine this structure is of both fundamental and practical importance. This review outlines current knowledge of granule architecture and the starch biosynthesis pathway in relation to the building block-backbone model of starch structure. We highlight the gaps in our knowledge in relation to our understanding of the structure and synthesis of starch, and argue that the building block-backbone model takes accurate account of both structural and biochemical data.

Increased seed size has greater contribution in yield improvement in recent naked oat (Avena sativa L. subsp. nudisativa) cultivars than cultivars derived from landraces. However, the underlying grain-filling mechanisms associated with seed size and grain yield in naked oat have received limited study. Two field experiments and a pot experiment compared grain-filling mechanisms, seed size and grain yield of ten naked oat cultivars, including old (landraces, released before 1950s) and new (released since 2008) cultivars. In both well-watered and water-deficit conditions, the new cultivars had higher grain yields, higher thousand kernel weights (TKW) and higher grain numbers per spike, but fewer fertile spikes per unit area or per plant, and no significant differences in grain number per unit area or per plant, than old cultivars. The findings in both the field and pot experiments demonstrated that increased grain-filling rate, rather than duration, enhanced seed size (TKW) and grain yield in the new cultivars. The increased grain-filling rate in a new cultivar was associated with higher rates of endosperm cell division and higher number of endosperm cells, compared to that in an old cultivar, but there were no significant differences in starch synthesis enzyme activity and ABA concentration between the old and new cultivars. As a result of current breeding and selection, the increased the grain yield of new cultivars of naked oat is primarily due to increased seed size (increased sink size) arising from a greater number of endosperm cells per grain, and not because of increased enzyme or phytohormonal activity. The larger seed size is important in the production of oats for food rather than for forage.
