Sestrin2 is a highly conserved stress-inducible protein, acting as a crucial part in regulating homeostasis in response to various stress conditions in the cell. However, the role of Sestrin2 in regulating cell apoptosis related to endoplasmic reticulum (ER) has not been fully investigated. Our study presented here aims to reveal the effect of Sestrin2 in tunicamycin (TM)-induced cell apoptosis related to ER stress and its underlying molecular mechanisms. The results demonstrated that Sestrin2 expression was significantly upregulated correlated with ER stress responses in TM treated HepG2 cells. Sestrin2 overexpression obviously alleviated ER stress with the determination of ER stress-related proteins expression. In addition, Sestrin2 overexpression inhibited cell apoptosis with the examination of apoptosis-related proteins and TUNEL assay. However, Sestrin2 knockdown further promoted the ER stress-mediated cell apoptosis. The further mechanistic study revealed that Sestrin2 overexpression inhibited TM-induced mTOR pathway activation. Taken together, our current study indicated that Sestrin2 overexpression ameliorates ER stress-induced apoptosis via inhibiting mTOR pathway in HepG2 cells.

Hair cell death induced by excessive reactive oxygen species (ROS) has been identified as the major pathogenesis of noise-induced hearing loss (NIHL). Recent studies have demonstrated that cisplatin- and neomycin-induced ototoxicity can be alleviated by ferroptosis inhibitors. However, whether ferroptosis inhibitors have a protective effect against NIHL remains unknown. We investigated the protective effect of the ferroptosis inhibitor ferrostatin-1 (Fer-1) on NIHL in vivo in CBA/J mice and investigated the protective effect of Fer-1 on tert-butyl hydroperoxide (TBHP)-induced hair cell damage in vitro in cochlear explants and HEI-OC1 cells. We observed ROS overload and lipid peroxidation, which led to outer hair cell (OHC) apoptosis and ferroptosis, in the mouse cochlea after noise exposure. The expression level of apoptosis-inducing factor mitochondria-associated 2 (AIFM2) was substantially increased following elevation of the expression of its upstream protein P53 after noise exposure. The ferroptosis inhibitor Fer-1was demonstrated to enter the inner ear after the systemic administration. Administration of Fer-1 significantly alleviated noise-induced auditory threshold elevation and reduced the loss of OHCs, inner hair cell (IHC) ribbon synapses, and auditory nerve fibers (ANFs) caused by noise. Mechanistically, Fer-1 significantly reduced noise- and TBHP-induced lipid peroxidation and iron accumulation in hair cells, alleviating ferroptosis in cochlear cells consequently. Furthermore, Fer-1 treatment decreased the levels of TfR1, P53, and AIFM2. These results suggest that Fer-1 exerted its protective effects by scavenging of ROS and inhibition of TfR1-mediated ferroptosis and P53-AIFM2 signaling pathway-mediated apoptosis. Our findings suggest that Fer-1 is a promising drug for treating NIHL because of its ability to inhibit noise-induced hair cell apoptosis and ferroptosis, opening new avenues for the treatment of NIHL.

Placenta previa greatly contributes to severe antenatal and post-partum hemorrhage. Previous studies have mainly focused on the risk factors of placenta previa, with very few studies reporting which factors may affect the potential resolution of 28th-week previa. This study aimed to investigate the impact of maternal characteristics on potential resolution of placenta previa from the 28th-to the 36th-week of pregnancy.A retrospective longitudinal sub-cohort investigation was carried out among 368 pregnant women with 28th-week previa from the Longitudinal Placenta Previa Study (LoPPS). Logistic regression analysis was used to discover the connections between maternal covariates and the placental potential resolution. Multivariable linear regression analysis was used to detect the associations between perioperative characteristics and volume of intraoperative bleeding.Among pregnant women whose placenta completely or partially covered the internal os at the 28th-week of pregnancy, 37.5% were without placenta previa at the 36th-week and 25.8% converted into marginal placenta previa. There were significant correlation between placenta previa type and GHD (Beta: 2.808, 95% CI: 1.642, 7.138; p = 0.041), type of 28th-week previa (Beta: 6.767, 95% CI: 1.592, 18.767; p 0.001), and number of prior cesarean sections (Beta: 3.326, 95% CI: 1.580, 9.081; p 0.001).62.5% of the pregnant women with 28th-week placenta previa were still with previa at the 36 weeks of gestation (25.8% with marginal and 36.7% with partial/complete placenta previa). Background Venomous arthropods have substances in their venom with antiproliferative potential for neoplastic cells. Objectives To identify a polypeptide from Myrmeleon bore (antlion) with antiproliferative activity against neoplastic cells, and to elucidate the molecular mechanism of the activity. Methods We used gel filtration and ion exchange chromatography to purify a polypeptide with antiproliferative activity against MG-63 human osteosarcoma cells from a proteinaceous extract of antlion. The polypeptide was sequenced and the stability of its antiproliferative activity was tested under a range of conditions in vitro. An 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to determine the antiproliferative activity of the polypeptide against the MG-63 osteosarcoma cells and MC3T3-E1 mouse calvarial osteoblasts, which were used as a non-neoplastic control. We used western blotting to compare the levels of expression of heat shock transcription factor 1 (HSF1), heat shock protein 90 (HSP90), cyclin-dependent kinase 4 (CDK4), and protein kinase B alpha (ATK1) in MG-63 osteosarcoma cells and their mouse homologs in MC3T3-E1 osteoblasts after their treatment with the antlion antiproliferative polypeptide (ALAPP). Results The 85-amino-acid ALAPP has a 56% sequence identity with the human heat shock factor binding protein 1 (HSBP1). The antiproliferative activity of the polypeptide is relatively insensitive to temperature, pH, and metal ions. ALAPP has a strong concentration-dependent antiproliferative activity against MG-63 osteosarcoma cells compared with its effect on MC3T3-E1 osteoblasts. ALAPP significantly upregulates the expression of HSF1 in MC3T3-EL osteoblasts, but not in MG-63 osteosarcoma. ALAPP significantly downregulated the expression of HSP90, CDK4, and AKT1 expression in MG-63 osteosarcoma, but not in the osteoblasts. Conclusions ALAPP has significant antiproliferative activity against MG-63 osteosarcoma cells, but not nonneoplastic MC3T3-E1 osteoblasts. We speculate that non-neoplastic cells may evade the antiproliferative effect of ALAPP by upregulating HSF1 to maintain their HSP90, CDK4, and AKT1 expression at a relatively constant level.

This study aimed to explore whether sex is influences tinnitus severity and whether the risk factors for tinnitus severity are the same in tinnitus patients of different sexes.This was a retrospective study of data from 1427 patients complaining of tinnitus in a local hospital otolaryngology clinic from November 2019 to January 2022. All patients were interviewed and assessed by otoscopy, pure-tone audiometry, tinnitus handicap inventory (THI), visual analogue scale (VAS), and tinnitus refinement test.THI values were higher in females than in males (P = 0.00). Types of tinnitus sounds (OR 0.667, P = 0.000) and degree of hearing loss (OR 1.318, P = 0.000) were risk factors for tinnitus severity in males. Types of tinnitus sounds (OR 0.789, P = 0.005), sensation level (OR 1.023, P = 0.037), tinnitus types (OR 1.163, P = 0.041), tinnitus location (OR 1.198, P = 0.026), and the degree of hearing loss (OR 1.303, P = 0.000) were risk factors for tinnitus severity in females. Sex was an influencing factor for tinnitus severity. There were different risk factors for the tinnitus severity in different sexes.The risk factors for tinnitus severity differed according to sex in tinnitus patients, and the risk factors for tinnitus severity were greater in women than in men. These findings add to the literature on sex differences in tinnitus and suggest that medical and psychological screening of affected individuals and customized tinnitus treatment for each individual with tinnitus are needed.ChiCTR2200057958, 2022/3/24 (retrospectively registered trials).

We address the question: What may be the conceptual sources of Chinese elementary students’ correct and incorrect solutions to multiplicative problem situations? We focus on situations for eliciting evidence about their ability to add and subtract sets of composite units—mental structures that underlie conceiving of whole numbers as a single entity composed of smaller units (e.g., “3” is composed of three “1s”). A conception postulated to underlie this ability is termed Same-Unit Coordination (SUC). We attribute student errors to reasonable-to-them spontaneous use of a previously established, repetitively practiced way of operating on 1s (“Ones”) contained within composite units—a conception we term Totaling. We analyze qualitative data to illuminate this phenomenon and quantitative data to depict its scope. These analyses support our claim that student solutions, seen by an observer as correct or erroneous, can be explained as (a) reasonable from the students’ frame of reference and (b) possibly arising from instructional focus on mastering multiplication facts to find totals of 1s in equal-size sets.

The urea oxidation reaction (UOR) has been well-acknowledged as one of the promising alternatives for hydrogen production through electrochemical water splitting system because of the more favorable thermodynamic potential. But the shortage of cost-effective electrocatalysts with high catalytic activity and durability restricts its practical development. Herein, the partially amorphous fluorine-decorated nickel iron layered double hydroxides (NiFe-F) is constructed via a low-temperature fluoridation method. Our study found that HF acid etching of NiFe LDH precursor resulted in the partially amorphous feature and abundant oxygen vacancies, providing rich reaction sites. Simultaneously, the formation of ionic metal-F bond makes it easier to form high-valence metal oxygen hydroxide active sites. Purpose The purpose of this study is to evaluate the effect of graphene, basalt flakes and their synergy on the corrosion resistance of zinc-rich coatings. As the important heavy-duty anticorrosion coatings, zinc-rich coatings provided cathodic protection for the substrate. However, to ensure cathodic protection, a large number of zinc powder made the penetration resistance known as the weakness of zinc-rich coatings. Therefore, graphene and basalt flakes were introduced into zinc-rich coatings to coordinate its cathodic protection and shielding performance. Design/methodology/approach Three kinds of coatings were prepared; they were graphene modified zinc-rich coatings, basalt flakes modified zinc-rich coatings and graphene-basalt flakes modified zinc-rich coatings. The anticorrosion behavior of painted steel was studied by using the electrochemical impedance spectroscopy (EIS) technique in chloride solutions. The equivalent circuit methods were used for EIS analysis to obtain the electrode process structure of the coated steel system. Simultaneously, the corrosion resistance of the three coatings was evaluated by water resistance test, salt water resistance test and salt spray test. Findings The study found that the addition of a small amount of graphene and basalt flakes significantly improved the anticorrosion performance of coatings by enhancing their shielding ability against corrosive media and increasing the resistance of the electrochemical reaction. The modified coatings exhibited higher water resistance, salt water resistance and salt spray resistance. The graphene-basalt flakes modified zinc-rich coatings demonstrated the best anticorrosion effect. The presence of basalt scales and graphene oxide in the coatings significantly reduced the water content and slowed down the water penetration rate in the coatings, thus prolonging the coating life and improving anticorrosion effects. The modification of zinc-rich coatings with graphene and basalt flakes improved the utilization rate of zinc powder and the shielding property of coatings against corrosive media, thus strengthening the protective effect on steel structures and prolonging the service life of anticorrosion coatings. Originality/value The significance of developing graphene-basalt flakes modified zinc-rich coatings lies in their potential to offer superior performance in corrosive environments, leading to prolonged service life of metallic structures, reduced maintenance costs and a safer working environment. Furthermore, such coatings can be used in various industrial applications, including bridges, pipelines and offshore structures, among others.
