This research explores the dynamics of a fluidically loaded Bi-Material cantilever (B-MaC), a critical component of μPADs (microfluidic paper-based analytical devices) used in point-of-care diagnostics. Constructed from Scotch Tape and Whatman Grade 41 filter paper strips, the B-MaC’s behavior under fluid imbibition is examined. A capillary fluid flow model is formulated for the B-MaC, adhering to the Lucas–Washburn (LW) equation, and supported by empirical data. This paper further investigates the stress–strain relationship to estimate the modulus of the B-MaC at various saturation levels and to predict the behavior of the fluidically loaded cantilever. The study shows that the Young’s modulus of Whatman Grade 41 filter paper drastically decreases to approximately 20 MPa (about 7% of its dry-state value) upon full saturation. This significant decrease in flexural rigidity, in conjunction with the hygroexpansive strain and coefficient of hygroexpansion (empirically deduced to be 0.008), is essential in determining the B-MaC’s deflection. The proposed moderate deflection formulation effectively predicts the B-MaC’s behavior under fluidic loading, emphasizing the measurement of maximum (tip) deflection using interfacial boundary conditions for the B-MaC’s wet and dry regions. This knowledge of tip deflection will prove instrumental in optimizing the design parameters of B-MaCs.

Laser-induced graphene (LIG) has been applied in many different sensing devices, from mechanical sensors to biochemical sensors. In particular, LIG fabricated on paper (PaperLIG) shows great promise for preparing cheap, flexible, and disposable biosensors. Distinct from the fabrication of LIG on polyimide, a two-step process is used for the fabrication of PaperLIG. In this study, firstly, a highly conductive PaperLIG is fabricated. Further characterization of PaperLIG confirmed that it was suitable for developing biosensors. Subsequently, the PaperLIG was used to construct a biosensor by immobilizing glucose oxidase, aminoferrocene, and Nafion on the surface. The developed glucose biosensor could be operated at a low applied potential (−90 mV) for amperometric measurements. The as-prepared biosensor demonstrated a limit of detection of (50–75 µM) and a linear range from 100 µM to 3 mM. The influence of the concentration of the Nafion casting solution on the performance of the developed biosensor was also investigated. Potential interfering species in saliva did not have a noticeable effect on the detection of glucose. Based on the experimental results, the simple-to-prepare PaperLIG-based saliva glucose biosensor shows great promise for application in future diabetes management.

The novel paper-based Bi-Material Cantilever (B-MaC) valve allows the autonomous loading and control of multiple fluid reagents which contributes to the accurate operation of paper-based microfluidic devices utilized for biological and chemical sensing applications. In this paper, an extensive parametric study is presented to evaluate the effects of key geometric parameters of the valve, such as paper direction, cantilever width, paper type, tape type, and sample volume, in addition to the effects of relative humidity and temperature on the functionality of the B-MaC and to provide a better understanding of the rate of fluid flow and resulting deflection of the cantilever. Machine direction, cantilever width, paper type, and tape type were found to be important parameters that affect the B-MAC’s activation time. It was also observed that the rate of fluid imbibition in the B-MaC is considerably affected by change in humidity for high (55 °C) and low (25 °C) temperatures, while humidity levels have no significant effect during imbibition in the B-MaC at an ambient temperature of 45 °C. It was also found that a minimum distance of 4 mm is required between the B-MaC and the stationary component to prevent accidental activation of the B-MaC prior to sample insertion when relative humidity is higher than 90% and temperature is lower than 35 °C. The rate of fluid imbibition that determines the wetted length of the B-MaC and the final deflection of the cantilever are critical in designing and fabricating point-of-care microfluidic paper-based devices. The B-MaC valve can be utilized in a fluidic circuit to sequentially load several reagents, in addition to the sample to the detection area.

New technological and scientific advances in the development of sensors and actuators demand the development of new devices to deal with recent problems and challenges in these new and emerging processes. Moreover, paper-based devices have tremendous potential for developing actuators as paper exhibits capillary transport and hygroexpansion due to swelling of the fibers when absorbing water. Therefore, this paper proposes a mini actuator that is based on a hygro-thermal-paper-based cantilever beam that is activated by means of a droplet of an aqueous solution in combination with a circulating electrical current to analyze its response. The contribution of this proposal includes the analysis of the flexural response of the mini actuator when it is tested by using two different solutions: distilled water and a water/alcohol solution. Additionally, four cases related to the droplet volume are studied and a statistical analysis of the bending responses is presented. The results achieved show that that water-alcohol solutions have a lower deviation in comparison with water only. Moreover, it is demonstrated that a specific change in the maximum displacement is obtained according to the volume and the type of solution. Thus, it is suggested that the response of the mini actuator can be tuned using different aqueous solutions.

We report on a novel colorimetric sensor system for highly sensitive detection of formaldehyde (FA) in the gas phase. The sensor is constructed with paper towel as a substrate coated with the sulfuric acid salt of hydroxylamine ((NH2OH)2·H2SO4) together with two pH indicators, bromophenol blue and thymol blue. Upon exposure to FA, the hydroxylamine will react with the absorbed FA to form a Schiff base (H2C=N-OH), thus releasing a stoichiometric amount of sulfuric acid, which in turn induces a color change of the pH indicator. Such a color change was significantly enriched by incorporating two pH indicators in the system. With the optimized molar ratio of the two pH indicators, the color change (from brown to yellow, and to red) could become so dramatic as to be visible to the eye depending on the concentration of FA. In particular, under 80 ppb of FA (the air quality threshold set by WHO) the color of the sensor substrate changes from brown to yellow, which can even be envisioned clearly by the naked eyes. By using a color reader, the observed color change can be measured quantitatively as a function of the vapor concentration of FA, which produces a linear relationship as fitted with the data points. This helps estimate the limit of detection (LOD), to be 10 ppb under an exposure time of 10 min, which is much lower than the air quality threshold set by WHO. The reported sensor also demonstrates high selectivity towards FA with no color change observed when exposed to other common chemicals, including solvents and volatile organic compounds. With its high sensitivity and selectivity, the proposed paper-based colorimetric sensor thus developed can potentially be employed as a low-cost and disposable detection kit that may find broad application in detecting FA in indoor air and many other environments.

A planar solid-state ammonium-selective electrode, employing a composite mediator layer of graphite particles embedded in a polyvinyl butyral matrix on top of an inkjet-printed silver electrode, is presented in this paper. The effect of graphite powder mass fraction on the magnitude of the potentiometric response of the sensor was systematically verified using a batch-mode and a flow injection measurement setup. Under steady-state conditions, the paper electrode provided a Nernstian response of 57.30 mV/pNH4 over the concentration range of 10−5 M to 10−1 M with a detection limit of 4.8 × 10−6 M, while the analytical performance of the array in flow mode showed a narrower linear range (10−4 M to 10−1 M; 60.91 mV/pNH4 slope) with a LOD value of 5.6 × 10−5 M. The experimental results indicate that the prepared electrode exhibited high stability and fast response to different molar concentrations of ammonium chloride solutions. The pH-response of the paper NH4-ISE was also investigated, and the sensor remained stable in the pH range of 2.5–8.5. The potentiometric sensor presented here is simple, lightweight and inexpensive, with a potential application for in-situ analysis of environmental water samples.

A novel paper-based potentiometric sensor with an enhanced response for the detection of glucose in biological fluids is presented. The electrode consists on platinum sputtered on a filter paper and a Nafion membrane to immobilize the enzyme glucose oxidase. The response obtained is proportional to the logarithm of the concentration of glucose, with a sensitivity of -119 +/- 8 mV.decade(-1), a linear range that spans from 10(-4)M to 10(-2.5) M and a limit of detection of 10(-4.5) M of glucose. It is shown that Nafion increases the sensitivity of the technique while minimizing interferences. Validation with human serum samples shows an excellent agreement when compared to standard methods. This approach can become an interesting alternative for the development of simple and affordable devices for point of care and home-based diagnostics.

We present a novel approach to measure ionic conductivity with a self-powered strategy. In particular, we propose the use of a paper-based battery as a sensor. The battery sensor unit consists of two electrodes placed side-by-side and covered by a piece of hydrophilic paper strip. The electrodes are externally connected to a resistive element. The addition of the fluid to be sensed - which acts as the electrolyte - activates the battery, which generates an output voltage that is dependent on the conductivity of the liquid sample. The device, which is conceived as a single-use disposable sensor, has been tested with different synthetic and biological liquid samples. The battery sensor effectiveness has been assessed by comparing its performance with a commercial laboratory conductometer. The device opens new avenues for conductivity monitoring in small portable and wearable devices, as it simplifies the number of electronic components and the need of additional power sources.

The smart, rapid, and customizable detection of chemical warfare agents is a huge issue for taking the proper countermeasures in a timely fashion. The printing techniques have established the main pillar to develop miniaturized electrochemical biosensors for onsite and fast detection of nerve and mustard agents, allowing for a lab on a chip in the chemical warfare agent sector. In the fast growth of novel technologies, the combination of miniaturized electrochemical biosensors with flexible electronics allowed for the delivery of useful wearable sensors capable of fast detection of chemical warfare agents. The wearable microneedle sensor array for minimally invasive continuous electrochemical detection of organophosphorus nerve agents, as well as the wearable paper-based origami functionalized with nanomaterials for mustard agents in the gas phase, represent two examples of the forefront devices developed in the chemical warfare agent detection field. This review will highlight the most promising electrochemical biosensors developed by exploiting nanomaterials and cross-cutting technologies for the fabrication of smart and sensitive electrochemical biosensors for the detection of chemical warfare agents.

Conjugated polyelectrolytes (CPEs) have been widely used as reporters in colorimetric assays targeting nucleic acids. CPEs provide naked eye detection possibility by their superior optical properties however, as concentration of target analytes decrease, trace amounts of nucleic acid typically yield colorimetric responses that are not readily perceivable by naked eye. Herein, we report a pixelated analysis approach for correlating colorimetric responses of CPE with nucleic acid concentrations down to 1 nM, in plasma samples, utilizing a smart phone with an algorithm that can perform analytical testing and data processing. The detection strategy employed relies on conformational transitions between single stranded nucleic acid-cationic CPE duplexes and double stranded nucleic acid-CPE triplexes that yield distinct colorimetric responses for enabling naked eye detection of nucleic acids. Cationic poly[N,N,N-triethyl-3-((4-methylthiophen-3-yl)oxy)propan-1-aminium bromide] is utilized as the CPE reporter deposited on a polyvinylidene fluoride (PVDF) membrane for nucleic acid assay. A smart phone application is developed to capture and digitize the colorimetric response of the individual pixels of the digital images of CPE on the PVDF membrane, followed by an analysis using the algorithm. The proposed pixelated approach enables precise quantification of nucleic acid assay concentrations, thereby eliminating the margin of error involved in conventional methodologies adopted for interpretation of colorimetric responses, for instance, RGB analysis. The obtained results illustrate that a ubiquitous smart phone could be utilized for point of care colorimetric nucleic acids assays in complex matrices without requiring sophisticated software or instrumentation., Graphical abstract Image 1, Highlights • A cost effective sensory cartridge and an algorithm compatible with smart phone are developed. • Conjugated polyelectrolyte impregnated paper for detection of nucleic acid. • A pixelated assay to visualize nucleic acid down to concentration of 1 nM.

Chemical analysis of paper-based electronic sensors is frequently interpreted using complex software and electronic displays which compromises the advantages of using paper. The ultimate aim of this PhD project is to develop simple and versatile paper-based readout systems that could directly convert the electrical signal into colour visualisation and later progress towards an integrated sensing device combining a readout system, a sensor and battery. The research starts with developing two semi-quantitative paper-based readout systems that can visually measure a change in resistance of a resistive-based sensor. The readout system exploits the electrochromic concept and also the effect of voltage gradient generation along a low-resistive gold nanoparticle film upon application of voltage. Due to this phenomenon, the electrochromic material changes colour along the gold nanoparticle film where the voltage is at the redox potential of the electrochromic materials. Hence, the resistance of the sensor can be semi-quantified by measuring the length of the transparent film (analog readout) or by counting the number of transparent gold segments (digital readout). The concept of displaying an electrical signal colorimetrically is validated by integrating an improved type of digital readout system, a piezoresistive pressure sensor and battery for developing a lightweight device for measuring bandage compression. A new type of piezoresistive pressure sensor is fabricated using an optimised mixture of irregular-shaped copper particles, polydimethylsiloxane and carbon paste. Through this integration, the pressure applied to the sensor can be visualised by counting the number of transparent gold nanoparticle segments in the readout system. The integration of a piezoresistive pressure sensor and readout system is extended for developing “balance-in-a-box” to determine the weight of a newborn baby. A new concept of tuning the resistive properties of the readout is introduced to change the critical weight value in which the gold segment changes colour. The significance of this work is that various desired reading ranges can be obtained while using the same type of pressure sensor. The last part of the study addresses the adaptability of the readout system for a chemical sensor, in particular a pH potentiometric paper-based sensor. Overall, these integrated sensing devices have paved the way for use in situations where weight is a premium or a simple colour reading will suffice.

A paramount research is focused nowadays on agrifood analysis, to ensure the commercialisation of safe products that meet the requirements of consumer perception. Food manufacturing processes might be threatened by contamination or lack of quality at each stage of the food supply chain while growing, processing, packaging, or distributing. Therefore, there is an increasing demand for fast and automatic analytical tools suitable for real-time control of food quality and safety through the whole chain. (Bio)sensors have high potential to this concern, being fast and sensitive analytical methods for the analysis of food composition and contamination, as well as for online process control. In this review, commercial (bio)sensors available for the agrifood sector are reported and compared in terms of their crucial features tailored for the different agrifood sector segments, including food analysis, process control, intelligent packaging, and smart agriculture. Moreover, an overview on the last trends in biosensor technologies is reported with the aim to enhance the biosensor performance and to develop newer sensor design and construction, fostering food production processes at high-quality standards.

In this work, we demonstrated that enhancement of electrochemical current due to redox cycling could be accomplished by paper-based biosensor without any expensive microfabrication process. The paper-based sensor had layered structure to generate higher current than a conventional one. We took advantage of the fact that the paper thickness was micrometer-sized (180um), and it defined the distance between two electrochemical electrodes on both sides of the paper. Experimental results showed signatures of the redox cycling, where the electrochemical current from low concentration molecules could be arbitrarily increased by decreasing the distance between electrodes. Such a structure was advantageous for detecting target molecules at very low concentration, proposing a low-cost highly-sensitive biochemcal sensor.

Research in paper-based analytical devices has been increasing in recent years. Before technology transfer and market acceptance, these paper-based sensors have to be validated with field samples. In this study, we have made an attempt to evaluate the effectiveness of paper-based sensors to detect pesticides in real world samples. Generation 1 biosensor has been modified to be user friendly. There is no difference in the performance of generation 2 sensors; they detect pesticides based on colorimetric assay. The assay protocol involves first introducing the sample to the sensing zone by pipetting the water sample. Following an incubation period of 15min, the substrate end of the sensor is dipped into the sample to move paper bound indoxyl acetate (IDA) to the sensing region to initiate the enzyme-catalyzed hydrolysis of the substrate, resulting in the development of blue color. The presence of pesticide is indicated by either a decrease in color intensity or with no color development at all. To evaluate the effectiveness of biosensor in detecting pesticides in real world samples, a field study was conducted in four villages of southern India. Water samples from different aquatic environment including both surface water and ground water, were tested using generation 2 paper-based sensors. The paper-based sensors were capable of detecting organophosphorus pesticides in real world samples. The results were confirmed using GC-MS. The presence of higher concentration of dibutyl phthalate (in the range of 100uM to 10mM) in water can be a potential interference for the paper-based assay for the detection of pesticides in water. The paper-based biosensor assay platform can detect pesticides in the environmental samples and results have been validated by GC-MS. But for transfer of technology to the industry, further optimization is required to improve the stability of substrate to withstand atmospheric temperature fluctuations thus allowing the storage and shipment of the biosensor strips. Additionally to conduct reliable assays and obtain consistent results, the fabrication of biosensor strips needs to be improved to maintain the consistent volumes of bioinks impregnated on paper support.
