Amaç: Öğrencilerin sağlık bilimleri müfredatında, temel bilimleri öğrenmeye yönelik motivasyonlarına az odaklanılmıştır. Bu çalışmanın amacı, sağlık bilimleri müfredatında insan histolojisi öğrenen öğrencilerde, motivasyonun farklı bileşenlerinin etkisini araştırmaktır. Gereç ve Yöntemler: Histoloji Motivasyon Ölçeği, tıp ve diş hekimliği ikinci sınıf lisans programlarına kayıtlı 235 öğrenciye (147 kadın ve 88 erkek), histoloji öğrenimine yönelik motivasyon bileşenlerini karşılaştırmak için kullanılmıştır. Bulgular: Bu çalışmada histoloji öğrenen tıp fakültesi öğrencilerinin, diş hekimliği öğrencilerine göre, kariyer motivasyonu, öz yeterlilik ve öz belirleme bileşenlerinde daha fazla motivasyona sahip oldukları saptanmıştır. Cinsiyetin ise histoloji öğrenme motivasyonunda farklılık göstermediği görülmüştür. Sonuç: Araştırma sonuçları, histoloji öğrenme motivasyonunda tıp ve diş hekimliği öğrencilerinde farklılık olduğunu ortaya çıkarmıştır. Bu sonuçlar öğrenme süreçlerini desteklemede, müfredatın gelişiminde ve eğitimin planlanmasında faydalı olacaktır.

Patoloji ve Histoloji Laboratuvarı Uygulama Kitabı, temelde üniversitelerin Patoloji Laboratuvar Teknikleri programında okuyan öğrenciler için hazırlanmıştır. Patoloji laboratuvarı uygulamalarında sık kullanılan takip ve boyama yöntemleri, takip aşamalarında ortaya çıkan hatalar ve bu hataların giderilmesine yönelik pratik çözümlerin yanı sıra pek çok farklı uygulamanın prosedürünü bir arada bulabileceğiniz bu kitap, aynı zamanda halen mesleğini icra etmekte olan tüm patoloji teknikerlerinin faydalanabileceği bir kaynak niteliğindedir. Kitap içerisinde rutin doku takip prosedürü, 7 farklı fiksatif solüsyonu hazırlama prosedürü, 4 farklı dekalsifikasyon solüsyonu hazırlama prosedürü ve 10 farklı doku boyama prosedürüne ek olarak histolojik dokulara ait çizim alanları ve laboratuvar uygulama çalışmaları için not alma alanları hazırlanmıştır. Prosedür uygulamalarının takibi ile teknikerlerin patoloji ya da histoloji laboratuvarlarında gereksinim duyacağı temel uygulama bilgileri kazandırılacaktır.

Beyin qabığı damarlarının struktur xüsusiyyətlərinin öyrənilməsi məqsədi ilə ağ siçovullarun quyruq venasına təmizlənmiş lipopolisaxarid (1 mq/kq) yeridilərək kəskin iltihab modeli yaradılmışdır. Beyin qabığından əldə edilmiş kriostat, eləcə də yarımvə ultranazik kəsiklər müvafiq olaraq işıq və elektron mikroskopları vasitəsilə tədqiq olunmuşdur. Eksperimental heyvanlarda subaraxnoidal və subpial sahələrdə yerləşən damarların mənfəzinin daralması, kortikal arteriolaların ətrafında perivaskulyar sahələrin genişlənməsi, mikrosirkulyasiya damarlarında hematoensefalik baryerin tamlığının pozulması əlamətləri aşkar edilmişdir.

Akut böbrek hasarı, böbrek fonksiyonlarının saatler ya da günler içinde bozulması sonucu ortaya çıkanüre ve kreatinin gibi nitrojen artık ürünlerinin birikmesi, sıvı-elektrolit ve asit-baz dengesinin bozulması ile seyreden klinik bir sendromdur. ABH prerenal, renal ve postrenal olarak üç gruba ayrılır. Renal ABH‟nın yaklaşık %85‟lik kısmını akut tübüler nekroz (ATN) oluşturur. ATN iskemi ya da nefotoksin maruziyeti sonucu oluşur. Nefrotoksisite şu an klinikte kullanılan ve geliştirilmekte olan ilaçların kullanımını sınırlamaktadır. Bu yüzden nefrotoksisiteyi önleyici ajanlara ihtiyaç duyulmaktadır. Silimarin deve dikeninden elde edilen antioksidan özellikleri olan bir üründür.Hepatoprotektif etkileri bir çok çalışmada gösterilmiştir. Bu rat çalışmasında silimarinin renal toksisitesinin olmadığını renal fonksiyonlar ve histopatoloji ile gösterilmesi hedeflendi. Sprague dawley ratlar altışar rattan oluşan iki gruba ayrıldı. Ratlar çevresel olarak kontrollü ortamda barındırıldı. Grup I‟e (kontrol) günde iki kere 0,4 cc musluk suyu gavajla verildi. Grup II‟e (silimarin) günlük tartılarına göre 100 mg/kg iki bölünmüş dozda silimarin yedi gün verildi. Sekizinci gün anestezi sonrası intrakardiak kan alımı ve nefrektomi yapıldı. Serum örneklerinde üre, kreatinin, , Sistatin C, süperoksit dismutaz (SOD), glutatyon peroksidaz (GPX), malondialdehit(MDA), total antioksidan kapasitesi(TAS) ve total oksidan kapasitesi (TOS) çalışıldı ve histopatolojik inceleme yapıldı.İki grup arasında üre, kreatinin, sistatin C, GPX, MDA, SOD ve TAS değerleri arasında anlamlı fark saptanmadı (p>0,05) fakat TOS değeri silimarin grubunda anlamlı olarak düşük saptandı (p=0,01). Histopatolojik olarak gruplarda glomerül ya da tübül hasarı izlenmedi. Bu bulgular silimarinin nefrotoksisitesinin olmadığını bilakis oksidan status üzerine olumlu etkileriyle renoprotektif olabileceğini düşündürdü. Acute kidney injury (AKI) is a clinical syndrome which is defined as a sudden decrease in kidney function, resulting in an inabilitiy to maintaine acid base, fluid and electrolyte balance and to excrete nitrogenous wastes. AKI is diveded three groups; prerenal, renal and postrenal causes. Acute tubuler necrosis (ATN) accounts approximately 85% of renal causes. ATN results from ischemia and nephrotoxin exposure. Nephrotoxicity limits the use of the drugs which are currently being used and which are developing. Due to we need some agents to prevent nephrotoxicity. Silymarin is a product obtained from milk thistle has an antioxidant properties. Hepatoprotective effect has been shown in many studies. In this study, it is aimed to evaluate silymarin affects on kidney function and the findings of kidney histology. Sprague Dawley rats were divided into two groups consisting of six rats. Rats were kept in controlled environment. Group I (control) were given 0,4 cc oral water, group II (silymarin) were given 100 mg/kg silymarin orally in two divided doses. Slymarin were given seven days according to daily weights. On the eighth day after anesthesia, intracardiac blood collection and nephrectomy was performed. In serum samples urea , creatinine, , Cystatin C , superoxide dismutase ( SOD), glutathione peroxidase (GPX ) , malondialdehyde ( MDA), total antioxidant status (TAS) and total oxidant capacity (TOS ) were studied and histopathologic examinations were performed. There was no significant difference between two groups in term of urea , creatinine, , Cystatin C , SOD, GPX , MDA and TAS (p>0,05) but TOS values were significantly lower in silymarin group (p = 0.01 ) . There were no tubuler and glomerular damage in all two groups. These findings demostrate that silymarin is not nephrotoxic drug and also has potential renoprotective effect related to possible effect on oxidant status.

Sanat, bilim ve tıp ilkçağlardan günümüze kadar varlığını sürdüren ve birbirleriyle etkileşim içinde olan üç olgudur. Estetik değerlerin dışavurumu olan sanat ile birlikte tıp, bilim ve teknoloji, insanın yaratıcı gücünü bünyesinde barındıran ortak özelliklere sahip kültür elemanlarıdır. Tıbbın babası olarak kabul edilen Hipokrat, tıbbın sanat olduğunu söyleyen ilk kişidir. Leonardo da Vinci ise hem sanat hem bilim yönü olan bir Rönesans ustası olarak kabul edilmektedir. 20. yüzyılda tıbbın hiç olmadığı kadar insan yaşamına eklemlendiği ve bu yüzyılda başat olgunun "insan sağlığı ve tıbbi uygulamalar" olduğu görülmektedir. İnsan yaşamının kendisi olarak görülen sanat ise, tıp ile yakın bir bağ içerisinde bulunmaktadır. Bu noktada Batı'da 1900-1950 arası dönemde hekim ya da cerrah olan aynı zamanda sanata ilgi duyan ve resim yapan kişilerin varlığı dikkat çekmektedir. Ayrıca bu dönemde pek çok sanatçının eserlerinde tıbbi ögelere yer verdiği görülmektedir. 1590 yılında Hollandalı bilim adamı Zacharias Janssen'in iki mercekten oluşan basit bir mikroskop yaptığı bilinmektedir. Mikroskobun icadı ve x-ışınlarının keşfi gibi tıbbi gelişmelerin sanata da yansıdığı ve Bauhaus okulunda mikroskop ve x-ışını fotoğraflarının öğretim unsuru olarak kullanılmaya başlandığı görülmektedir. Kandinsky ve Klee gibi pek çok sanatçı da bu görüntülerden etkilenerek, eserlerinde insan bedeninin derinini resmetmeye çalışmışlardır. Bu çalışmada sanatın bilimsel düşünce ve malzemelerden nasıl yararlandığı irdelenmeye çalışılmıştır. Mikroskobik anatominin, mikroorganizmaların, embriyolojik ve zoolojik formların sanatçıların eserlerinde nasıl bir dönüşüme uğradığı araştırılmıştır.

Objective: The purpose of this study was to examine the effects of apocynin (APC), an inhibitor of NADPH oxidase (NOX), on high-fat diet (HF)-induced testis cytotoxicity. Methods: Wistar albino rats were divided into three groups as control, HF and HF+APC groups. Rats in HF and HF+APC groups were fed using HF for 16 weeks and in the last four weeks of this period vehicle solution or APC (25 mg/kg) was administered orally five days a week, respectively. Control group was fed with standart lab chow for 16 weeks. Cholesterol, triglyceride, high-density lipoproteins, leptin, estrogen, testosterone, LH and FSH were estimated in blood serum. Sperm parameters were analysed from the epididymis. Testicular malondialdehyde, 8‐hydroxy‐2‐deoxyguanosine, glutathione, superoxide dismutase and myeloperoxidase levels were estimated biochemically. Testicular morphology, proliferative, apoptotic and NOX2-positive cells were analysed histologically. HF-induced obesity caused significant alterations in serum lipid and hormone profiles. Testicular malondialdehyde, 8‐hydroxy‐2‐deoxyguanosine, and myeloperoxidase levels increased, glutathione and superoxide dismutase levels decreased in this group. Moreover, altered sperm parameters, increased degenerated seminiferous tubules, apoptotic and NOX2- positive cells and decreased proliferative cells were observed in the HF group. All these biochemical and histological alterations improved in the HF+APC group. Conclusion: HF-induced obesity causes altreations in lipid values, sperm parameters and testicular morphology by increasing oxidative stress through NOX2 activity. Apocynin might prevent testis damage via regulating oxidant/antioxidant balance.

Plastination is a laboratory process to obtain permanent dry tissue and organ sample. That can be used in the future for investigation and educational purposes. This methodology is based on dehydration and penetration of synthetic substances such as silicon into tissue. In this study, it was aimed to deplastinate previously plastinated kidneys in order to examine them under the light microscope. In this study, 14 sheep kidneys were used, seven samples of control and seven samples of plastination-deplastination (p / d) group. Kidneys in both control and p/d groups were fixed in 10% formalin. The samples in the control group were embedded in paraffin following routine tissue processing protocol. However, the samples in the p/d group were deplastinated in alcohol and methylbenzene and embedded into paraffin. 5 μm thick sections obtained from paraffin blocks were stained with hematoxylin and eosin (H&E), periodic acid-shiff (PAS) and then examined under the light microscope. Typical histological structures were observed in the control group. Small fragments were obtained as it was challenging to obtain sections from the P/d group blocks. Morphological structures were visible with some pseudo degenerations and wrong staining. This study is the first study that demonstrates alcohol and methylbenzene deplastination can be partially successful for evaluating plastinated kidney samples under a light microscope. However, we believe that the kidney may have limitations due to its wide parenchyma compared with literature conclusions. Nevertheless, more studies are required to develop the optimum protocols., Plastinasyon, gelecekte araştırma ve eğitim amaçlı kullanılabilecek kalıcı kuru doku ve organ örnekleri elde etmeye yönelik bir laboratuvar işlemidir. Kısaca bu metodoloji silikon gibi sentetik maddelerin dehidrasyona ve dokuya nüfuz etmesine dayanmaktadır. Bu çalışmada ışık mikroskobu altında incelemek için önceden plastine edilmiş böbrekleri deplastine etmeyi amaçladık. Bu çalışmada 7 örnek kontrol ve 7 örnek plastinasyon-deplastinasyon (p/d) grubu olmak üzere, 14 koyun böbreği kullanıldı. Hem kontrol hem de p/d gruplarındaki böbrekler %10 formalin içinde sabitlendi. Kontrol grubundaki örnekler rutin doku işleme protokolü izlenerek parafine gömüldü, ancak p/d grubundaki örnekler alkol ve metil benzen içinde deplastine edildikten sonra parafine gömüldü. Parafin bloklarından elde edilen 5 μm kalınlığındaki kesitler Hematoksilen ve eozin (H&E) ile boyanarak ardından periyodik asit-schiff (PAS) ile ışık mikroskobu altında incelendi. Kontrolgrubunda normal histolojik yapılar gözlendi. P/d grup bloklarından kesitler elde etmek zor olduğu için küçük parçalar elde edildi. Morfolojik yapılarda bazı dejenarasyonlar gözlemlendi. Bu, plastine böbrek örneklerinin alkol ve metil benzen ile deplastinasyonunun ışık mikroskobu altında değerlendirilmesinde kısmen başarılı olabileceğini gösteren ilk çalışmadır. Ancak literatürdeki sonuçlarla karşılaştırıldığında böbreğin geniş parankiminden dolayı bazı kısıtlılıklarının olabileceğini düşünmekteyiz, ancak optimum protokolleri geliştirmek için daha fazla çalışmaya ihtiyaç vardır.

Human growth hormone (GH) is crucial modulator of cellular metabolisms, including cell proliferation and organ development, by stimulating insulin-like growth factor-1 (IGF-1), which has various functions such as cell proliferation, tissue growth, survival, or neuroprotection. Therefore, GH is implicated as a critical player in the cell and can enhance neurogenesis and provide neuroprotection during the treatment of neurological diseases such as Parkinson’s disease (PD). In this study, the neuroprotective role of GH was investigated in rotenone-induced PD models for the first time. Both SH-SY5Y and SK-N-AS neuroblastoma cells were exposed to rotenone to mimic PD pathogenesis as stated in previous studies. Our data demonstrated that overexpression of GH led to the resistance of the SH-SY5Y and SK-N-AS cell lines to rotenone treatment. The levels of ER stress markers, CHOP, PERK, XBP-1, and ATF6, were higher in wt cells than GH+ SH-SY5Y cells. However, the level of autophagy markers LC3 increased and the levels of reactive oxygen species (ROS) decreased with the overexpression of GH. Furthermore, while rotenone significantly increased the SubG1 population in the cell cycle of SH-SY5Y wt cells, there was a minor alteration in GH+ cell population. Concomitantly, the levels of the proapoptotic marker, cleaved-PARP, and positive staining of Annexin V in SH-SY5Y wt cells were higher after rotenone treatment. Together, these results revealed that overexpression of GH enhanced the autophagy response by triggering the ER stress of SH-SY5Y cells to rotenone exposure and showed a neuroprotective effect in vitro PD models.

Objective: The aim of this study was to evaluate the ameliorative effects of swimming training on renal and bladder damage caused by a highfat diet (HFD) using morphological and biochemical measurements. Methods: Sprague Dawley rats were fed either standard chow (CONT, 6% fat) or HFD (45% fat) for 18 weeks, these rats were divided into two subgroups at the last 6 weeks of the experiment. The exercise groups (CONT+EXC, HFD+EXC) were trained daily swimming sessions (1 h per day for 5 days/week) during the last 6 weeks. Kidney and bladder samples were prepared for light and electron microscopic examination at the end of experiment. Malondialdehyde, glutathione, interleukin-6, and tumor necrosis factor-α were measured by biochemically. Results: Regular morphology of the renal cortex and bladder mucosa was observed in the CONT and CONT +EXC groups. Degenerated renal corpuscles and proximal tubules in the kidney and degenerated urothelium with leaky tight junctions and mast cell increase in the bladder mucosa were observed in the HFD group. Ameliorated renal cortex and bladder mucosa were observed in the HFD+EXC group. In addition, malondialdehyde, glutathione, interleukin-6, and tumor necrosis factor-α levels were also consistent with the histological findings. Conclusion: HFD-induced renal and bladder damage may be related to increased oxidative damage. It was observed that the histological damage and altered oxidative stress parameters could be reversed by swimming training, and it is thought that moderate swimming exercise may play a role in regulating oxidative stress.

Neoadjuvant chemotherapy (NACT) in gastroesophageal junction (GEJ) and gastric cancer (GC) was shown to improve survival in recent studies. We aimed to share our real-life experience of patients who received NACT to compare the efficacy and toxicity profile of different chemotherapy regimens in our country. This retrospective multicentre study included locally advanced GC and GEJ cancer patients who received NACT between 2007 and 2021. Relation between CT regimens and pathological evaluation were analysed. A total of 794 patients from 45 oncology centers in Turkey were included. Median age at the time of diagnosis was 60 (range: 18-86). Most frequent NACT regimens used were FLOT (65.4\%), DCF (17.4\%) and ECF (8.1\%), respectively. In the total study group, pathological complete remission (pCR) rate was 7.2\%, R0 resection rate 86.4\%, and D2 dissection rate was 66.8\%. Rate of pCR and near-CR (24\%), and R0 resection (84\%) were numerically higher in FLOT arm (p > 0.05). Patients who received FLOT had also higher chemotherapy-related toxicity rate compared to patients who received other regimens (p > 0.05). Median follow-up time was 16 months (range: 1-154 months). Estimated median overall survival (OS) was 58.4months (95\% CI: 35.2-85.7) and disease-free survival (DFS) was 50.7 months (95\% CI: 25.4-75.9). The highest 3-year estimated OS rate was also shown in FLOT arm (68\%). We still do not know which NACT regimen is the best choice for daily practice. Clinicians should tailor treatment regimens according to patients' multifactorial status and comorbidities for to obtain best outcomes. Longer follow-up period needs to validate our results.

Kuşlarda gözün en göze çarpan ve ilginç özelliği pecten oculi’dir. Bu çalışmada Aseel tavukların gözünde bulunan pecten oculi’nin anatomik ve histolojik özellikleri ışık ve elektron mikroskopik uygulamalar yardımıyla tespit edildi. Çalışma için dört adet yetişkin Aseel ırkı tavuk kullanıldı. Enükleasyon işlemi sonrası gözler %10’luk tamponlu formalin solüsyonunda tespit edildi. Morfolojik yapıları stereomikroskop altında incelenen gözler daha sonra elektron mikroskop ve histolojik incelemeler için hazırlandı. Yapılan incelemede pecten oculi’nin gözün arka yarımında, retina tabakası üzerinde bulunduğu belirlendi. Sağ ve sol pecten oculi’lerin ortalama 20-22 adet piliden (kıvrımdan) oluştuğu tespit edildi. Pectenlerin ortalama bazal uzunluğu 7,92 mm, apikal uzunluğu 4,45 mm, yüksekliği ise 3 mm olarak belirlendi. Pektineal kıvrımların kesit yüzeyi incelendiğinde kıvrımların merkezine yerleşmiş büyük çaplı ve oval şekilli damarlar ile bu damarların çevresinde seyreden çok sayıda kılcal kan damarları tespit edildi. Pili kesitlerinin hem perifer hem de merkez bölgeleri üzerinde yaygın ve çok sayıda melanosit varlığı tespit edildi. Sonuç olarak, Aseel ırkı tavuklarda pekten oculi’nin morfolojik ve histolojik yapısının diğer gündüz aktif kuş türlerinde bildirilen özelliklerle genel olarak uyumlu olduğu belirlenmiştir., The most striking and interesting feature of the eye in birds is the pecten oculi. In this study, the anatomical and histological features of pecten oculi in the eyes of Aseel chickens were determined by the light and electron microscopic applications. For the study were used four adult Aseel chickens. After the enucleation procedure, eyes were fixed in 10% buffered formalin solution. Eyes whose morphological structures were examined under a stereomicroscope were then prepared for electron microscopy and histological examinations. In the examination, it was determined that the pecten oculi was located on the retina layer in the posterior half of the eye. It was determined that the right and left pecten oculi consisted of an average of 20-22 pili. The mean basal length of pectenes was 7.92 mm, apical length was 4.45 mm, and height was 3 mm. When the cross-sectional surface of the pectineal folds was examined, large diameter and oval-shaped vessels located in the center of the folds and many capillary blood vessels coursing around these vessels were detected. Numerous and diffuse melanocytes were detected on both the peripheral and central regions of pili sections. As a result, it was determined that pecten oculi in Aseel chickens was generally compatible with the characteristics reported in other diurnal bird species in terms of morphological and histological features.

Furosemide is a diuretic and is used for the treatment of patients with heart failure (HF). It has been found that in some HF patients, the drug does not treat patients efficiently. This condition is named as furosemide resistance. In this study, it is aimed to investigate the relationship between UDP-glucuronosyltransferase 1 (UGT1A1) and interleukine-6 (IL-6) variations with furosemide resistance in HF patients. Sixty HF patients using furosemide (patient group) and 30 healthy individuals (control group) were enrolled in this study. Patients were divided into two subgroups as non-responders (furosemide resistant) group (n = 30) and the responders (non-resistant) group (n = 30) according to the presence of furosemide resistance (n = 30). Variations in the first exon of UGT1A1 and rs1800795 and rs1800796 variations in IL-6 were analyzed by direct sequencing and real-time polymerase chain reaction (RT-PCR), respectively. The effects of newly detected mutations on 3-D protein structure were analyzed by in silico analysis. At the end of the study, 11 variations were detected in UGT1A1, of which nine of them are novel and eight of them cause amino acid change. Also, rs1800795 and rs1800796 variations were detected in all the groups. When patient and control groups were compared with each other, rs1800796 mutation in IL-6 was found statistically high in the patient group (p = 0.027). When the three groups were compared with each other, similarly, rs1800796 mutation in IL-6 was found statistically high in the non-responders group (p = 0.043). When allele distributions were compared between the patient and control groups, the C allele of rs1800795 mutation in IL-6 was found statistically high in the patient group (p = 0.032). When allele distributions were compared between the three groups, 55T-insertion in UGT1A1 was found statistically high in the non-responders group (p = 0.017). According to in silico analysis results, two variations were found deleterious and six variations were detected as probably damaging to protein functions. Our study may contribute to the elucidation of pharmacogenetic features (drug response–gene relationship) and the development of individual-specific treatment strategies in HF patients using furosemide.

Objective: Obesity is associated with many diseases, including urinary system disorders such as chronic kidney disease and overactive bladder syndrome. Myrtus communis L. (MC) extract has been reported to have antioxidant and anti-inflammatory effects. The aim of this study was to investigate the protective effects of MC extract on high-fat diet (HFD)-induced kidney and bladder damage. Materials and Methods: Wistar albino male rats were divided into three experimental groups: control, HFD and HFD+MC. Experimental groups were fed a standard diet (control group) or HFD (HFD and HFD+MC groups) for 16 weeks. MC extract (100 mg/kg) was administered to the HFD+MC group orally during the last 4 weeks (5 days/week) of the experiment. Highdensity lipoprotein, total cholesterol, triglyceride and leptin levels were measured in blood serum. Tissue malondialdehyde (MDA), glutathione (GSH), 8-hydroxy-2'-deoxyguanosine (8-OHdG) and myeloperoxidase (MPO) levels were evaluated biochemically. Kidney and bladder morphology, NADPH oxidase-2 (NOX-2) and nuclear factor-kappa B (NF-ҡB)-positive and apoptotic cells were evaluated histologically. Results: Lipid profiles altered and leptin levels increased in blood serum. MDA, 8-OHdG and MPO levels increased and GSH level decreased in kidney and bladder in the HFD group. Moreover, degenerated kidney and bladder morphology, increased NOX-2 and NF-ҡB-positive and apoptotic cells were observed in this group. All of these biochemical and histological parameters were ameliorated in the HFD+MC group. Conclusion: HFD-induced obesity causes kidney and bladder damage by oxidative and inflammatory processes. ÖZETGiriş: Çoğunluğu rezeksiyon olan splenik biyopsilerpatoloji rutininde seyrek karşılaşılan spesmenlerdir. Splenektomisebepleri çoğunlukla travma, daha sonra lenfoid/myeloid neoplaziler, vasküler ve kistik lezyonlar olarak sayılabilir.Ekstramedüller hematopoez ve hemolitik anemi gibinon-neoplastik hematolojik antiteler de splenik biyopsilerdekitanı çeşitliliğini arttırmaktadır. Kor biyopsi örnekleme yöntemi,splenik lezyonlara yaklaşımda majör komplikasyonlar veteknik zorluklar sebebiyle girişimsel radyologlar tarafındangenellikle tercih edilmemektedir.Olgu: Bilinen serebrovasküler atak ve asetilsalisilik asit kullanımöyküsü bulunan, halsizlik şikayetiyle hastanemizebaşvuran 71 yaşındaki erkek hastanın tetkiklerinde anemi(Hb:10,52 g/dL) ve LDH (592 U/L) yüksekliği saptanmıştır.Üst batın tomografisinde, dalak anterior posterior çapı 153mm ölçülmüş olup artmış, parankim dansitesi doğal görülmüştür.Pozitron emisyon tomografisinde (PET-CT), splenikFDG tutulumu karaciğer parankimine denk düzeydedir (SUVmax:3.8). Tariflenen bu nonspesifik bulgu düşük metabolikaktiviteli lenfoproliferatif neoplaziler yönünden şüpheli bulunmuştur.Radyolojik olarak görüntüleme eşliğinde 18Gtru-cut iğne ile kor biyopsi örneği alınmıştır. Hematoksilen-Eozinkesitlerde, splenik doğal çatının net seçilemediği ödemlidokuda difüz, polimorfik hücresel infiltrasyon izlenmiştir. Popülasyonunbüyük kısmını küçük boyutlu lenfoid hücreleroluşturmaktadır. Arada kümeleşme gösteren nötrofil ve eozinofilinfiltrasyonu mevcuttur. İmmünohistokimyasal CD61ile pozitif, bir kısmı hiperkromatik, hiperlobe nükleuslu, getoplazmalıizlennişsitoplazmalı, dismorfik özellikler gösteren megakaryositformları dikkati çekmiştir. E-cadherin ile eritroid öncü hücregrupları da tespit edilen olguya ekstramedüller hematopoeztanısı verilmiştir. İşlem sonrası komplikasyon gelişmeyen hastahalen takipte olup, klinik açıdan stabildir.Sonuç: Özgün vasküler yapılanmaya sahip dalak dokusundayüksek kanama riski sebebiyle kor biyopsi örnekleme yöntemirutin olarak tercih edilmemektedir. Ancak seyrek de olsasplenektomi için yüksek komorbidite taşıyan hastalarda dahaaz invaziv olan bu yönteme şans verilmektedir. Histopatolojikdeğerlendirmesi patologlar için zorlayıcı olabilen bu örneklerde,ayırıcı tanıda ekstramedüller hematopoezin de akıldabulundurulmasının gerektiğini düşünmekteyiz.Anahtar Kelimeler: Dalak, Ekstramedüller hematopoez, Korbiyopsi

Gold nanoparticles (GNPs) have been widely used in medicine such as imaging, drug delivery and therapeutics due to their multifunctional properties. Alterations in neuronal function may contribute to various neurological diseases. Transferrin plays a primary role in iron transportation and delivery and has recently been utilized for drug delivery to the brain. We have investigated effects of transferrin-conjugated GNPs (Tf-GNPs) on anxiety and locomotor behavior in vivo and also hippocampal neuronal activity ex vivo. Electrophysiological effects of Tf-GNP on hippocampal neurons were determined by patch clamp method. Fifteen male young adult C57BL/6 mice were randomly divided into three groups as control (200 µL PBS), GNP (bare GNP; 2.2 μg/g in PBS) and Tf-GNPs (2.2 μg/g Tf-GNP). Animals intraperitoneally received the respective treatments for seven consecutive days and were subjected to elevated plus maze (EPM) and open field tests (OFT). Ex vivo, firing frequency of the neurons significantly increased by GNP treatment (p < 0.001). In vivo, animals in Tf-GNP group showed significantly longer distance in open arms but significantly lower number of entries to the open arms in EPM (p < 0.05). Mice received bare GNPs had significantly higher locomotor activity in OFT (p < 0.05), while Tf-GNP did not alter the locomotor activity significantly (p = 0.051). Animals in Tf-GNP group spent significantly longer time in the peripheral zone in OFT (p < 0.05). The present findings have shown that Tf-GNP induces anxiety-like behavior without altering spontaneous firing rate of hippocampal neurons. We suggest that neurobiological effects of Tf-GNP should be pre-determined before using in medical applications.

Case Description: A 31-year-old female patient from Djibouti/North Africa had a complaint of neck swelling for one year. No other symptoms were described. It was reported that she was diagnosed with lymphoma (Hodgkin? Non-Hodgkin?, pathology report not available) in her country but was not treated. In PET-CT images, lymph nodes with 17-27 mm diameters (SUVmax: 2.4-4.0) were detected in the left cervical lymphatic stations, bilateral axilla, mediastinum, paraaortocaval regions, bilateral iliac chain and inguinal fossa. Spleen size increased and intraparenchymal FDG uptake was slightly increased homogeneously. A few millimetric nodular areas of density increase were observed in both lung parenchyma. In biochemistry and hemogram tests; Albumin:3.8, Total Protein:5.6, Urea:23, Urate:6.8, Creatinine:0.56, LDH:194, WBC:2430, Neutrophil:1800, Lymphocyte:410, Hb:7.48, Hct:22.87, Plt:121000. Beta-2 microglobulin: 3.26, Kappa(total, serum):0.9, Lambda(total, serum):0.5, IgA:48(65-421), IgG:1586(552-1631), IgM:21(33-293). No kappa or lambda light chain detected in urine immunofixation electrophoresis. IgG monoclonal band observed in serum immunofixation electrophoresis. Hepatic viral markers and HIV were negative. Left supraclavicular lymph node excision and bone marrow biopsy were evaluated in our department.Biopsy Fixation Details: 10% neutral buffered formalin. Bone marrow biopsy was decalcified in 10% formic acid for 8 hours.Frozen Tissue Available: -Details of Microscopic Findings:Lymph node: The architecture was totally effaced. There was atypical infiltration in diffuse and coarse nodular pattern consisting of polymorphic cells. Most of the atypical cells had a plasmacytoid morphology. Small-medium sized lymphoid cells with coarse chromatin were accompanying the plasmacytoid cells. Scattered atypical lymphocytes were observed, some resembling Reed-Sternberg cells, with large round/lobulated nuclei, vesicular chromatin, and prominent single or few small nucleoli.Bone marrow: The biopsy with six intertrabecular field was hypercellular. Alternating areas with light/dark staining and vague nodulation foci were seen. Hematopoietic cells were pushed to the paratrabecular zone by atypical infiltration. The atypical infiltration was composed of a polymorphic cellular population in this biopsy as well. Numerous plasma cells were observed in the periphery of the nodular areas. In the center of the nodules, small-medium sized lymphoid cells with coarse chromatin and scant cytoplasm were present. In a focal area atypical lymphoid cells were mixed with epithelioid histiocytes. Clusters of medium-sized lymphoid cells with vesicular chromatin were seen in some areas. Epibrassinolide is a member of brassinosteroids with a polyhydroxysteroid structure similar to steroid hormones of vertebrates. It was shown that EBR decreased cell proliferation and induced apoptosis in different colon cancer cell lines without exerting a cytotoxic effect in epithelial fetal human colon cells. This finding highlighted the potential of epibrassinolide in clinical therapeutic setup. In our previous studies, we showed that epibrassinolide was able to induce apoptosis via endoplasmic reticulum stress. Recently, we also showed that endoplasmic reticulum and apoptotic stresses can be prevented via autophagic induction in non-cancerous epithelial or aggressive forms of cancer cells. Therefore, here in this study, we evaluated the anti-tumoral effect of epibrassinolide as well as the autophagy involvement in the aggressive forms of colon cancer cell lines as well as in vivo SCID mouse xenograft colon cancer model for the first time. For this purpose, SCID mouse model was used for subcutaneous injection of colon cancer cells in matrigel formulation. We found that autophagy is induced in both in vitro and in vivo models. Following tumor formation, SCID mice were treated daily with increasing concentrations of epibrassinolide for two weeks. Our findings showed that EBR inhibited the volume and diameter of the tumor in a dose-dependent manner by causing cell cycle arrest. Therefore our data suggest that epibrassinolide exerts a cytostatic effect on the agrressive form of colon cancer model in vivo, without affecting endoplasmic reticulum stress and the induction of autophagy might have role in this effect of epibrassinolide. © 2023 Elsevier Ltd 36587800

Giriş: Gastrointestinal sistem, hematolenfoid neoplazilerin en sık görüldüğü ekstranodal bölgedir. Bununla birlikte inflamatuar lezyonlar ve epiteliyal neoplazilere kıyasla oldukça nadir gözlenen bu grup neoplazilerde tanı zorlukları yaşanabilmektedir. Bu çalışmada gastrointestinal sistemin sık görülen hematolenfoid neoplazilerinin klinikopatolojik özelliklerini sunmayı, nadir görülen antitelere ise dikkat çekmeyi amaçladık. Gereç ve yöntemler: Patoloji arşivimizden, 2014-2021 yılları arasında, gastrointestinal sistem infiltrasyonu ile hematolenfoid neoplazi tanısı alan 46 hasta retrospektif olarak tespit edilmiştir. Patoloji raporları yanı sıra demografik ve klinik verilere, endoskopi ve görüntüleme bulgularına hastane bilgi sisteminden ulaşılmıştır. Bulgular: Otuzaltı hastada primer odak gastrointestinal sistemdi. Dokuz hastada, gastrointestinal infiltrasyon sistemik hastalığın sekonder yayılımı olarak kabul edildi. Hastalardan birinde, mevcut verilerle primer veya sekonder hastalık ayrımı yapılamadı. Ortalama yaş 56 idi. Olguların yaklaşık dörtteüçü (%73,9) endoskopik biyopsi ile tanı aldı. Hastaların sıklıkla başvuru sebebi nonspesifik gastrointestinal semptomlardı (%73,9). Barsak yerleşimli orgularda ise ileus tablosu ile karşılaşıldı. En sık tanı diffüz büyük B hücreli lenfoma (n:26), ardından MALT lenfomaydı (n:6). Duodenal tip foliküler lenfoma tanısı alan dört hastamız mevcuttu. Nadir vakalarımız; IRF4 ile ilişkili büyük B hücreli lenfoma (n:1), EBV pozitif büyük B hücreli lenfoma (n:1), ekstrakaviter/solid varyant primer efüzyon lenfoma (n:1), myeloid sarkomdu (n:1)., Tartışma: Difüz büyük B hücreli lenfoma hasta sayısının MALT lenfomanın dört katından fazla olması beklenmeyen bir farktır. Özellikle nadir görülen yüksek dereceli lenfomalar ve myeloid neoplazilerin, az diferansiye karsinomlarla karıştırılabilme olasılığı önemli bir handikaptır. PORCUPINE İNHİBİTÖRÜ UYGULANMASI SONRASINDA FETUS BEYİNLERİNDE SİNİR VE KÖK HÜCRE BELİRTEÇLERİNİN ARAŞTIRILMASI. YEŞİLYURT A.G. Adnan Menderes Üniversitesi Sağlık Bilimleri Enstitüsü Histoloji Embriyoloji Yüksek Lisans Tezi, 2019. Organ gelişiminde ve pek çok moleküler biyolojik olaylarda Wnt β-katenin sinyal yolağının etkisi bilinmektedir. Wnt sinyal yolağı, hedef genlerin transkripsiyonunda ve bir çok genin ekspresyonunda görev almaktadır. Bu sebep ile Wnt sinyal yolağının aktivasyonun veya inhibisyonunun beyin kök hücre belirteçlerinin üzerinde etkisinin olduğu düşünülmektedir. Bu çalışma da, Wnt β-katenin sinyal yolağının fetal sinir ve beyin kök hücre belirteçleri açısından bir fark oluşturup oluşturmadığının tespit edilmesi amaçlandı. Bu amaç ile, üç gruptan oluşan bir deney dizaynı oluşturuldu. Bu gruplar; gebe kontrol grubu, gebe DMSO grubu ve porcupine inhibitörü uygulanan gebe grubudur. Fetal gelişimin 13.5, 15.5, 17.5 gününde anne karnından alınan fetusların ağırlıkları ve boyları değerlendirildi. 13.5, 15.5, 17.5 günlük fetusların beyin dokularında Wnt/β-katenin sinyal yolağının inhibitörü olan LGK 974 (Porcupine inhibitörü) uygulanması sonrasında, β-katenin ifadesinin değişimine, S100B beyin belirtecine, Lgr5 kök hücre belirteci proteinlerine, immünohistokimya ile ifade düzeylerine bakıldı. Böylece Wnt/ β-katenin sinyal yolağının embriyonik sinir ve beyin gelişiminde etkisi değerlendirildi. Ayrıca beyin dokularına Toluidin blue ve Hematoksilen-Eozin boyaması yapılarak morfolojik olarak değerlendirildi. Yapılan deney sonuçlarına göre, Porcupine inhibitörü uygulanan fetüslerin ağırlıkları ve boylarında diğer gruplara oranla istatistiksel olarak anlamlı bir düşüş saptanmıştır. İmmünohistokimya sonuçlarına göre; Porcupine inhibitör grubunda, Kontrol ve DMSO gruplarına oranla β-katenin, S100B, Lgr5 ifadesinde istatistiksel olarak anlamlı değişiklere rastlanmadı. Hematoksilen-Eozin ve Toluidin Blue boyama ile morfolojileri tüm gruplar arasında olağan görülmüştür. Bu sonuçlar doğrultusunda Wnt/ β-katenin sinyal yolağının embriyo gelişimi üzerine anlamlı etkileri olduğu bulunurken, bu yolak ile ilgili beyin gelişiminde rol alan gen ifade düzeylerinde istatistiksel olarak anlamlı bir değişiklik gözlenmedi. The Wnt signaling pathway is involved in the transcription of the target genes and expression of many genes. Therefore, activation or inhibition of the Wnt signaling pathway is thought to have an effect on brain stem cell markers. In this study, it was aimed to determine whether Wnt signaling pathway made a difference in terms of fetal nerve and brain stem cell markers. For this purpose, an experimental design consisting of three groups was formed. These groups are; pregnant control group, pregnant DMSO group and porcupine inhibitör applied pregnant group. The weights and lengths of fetuses taken from the mother's abdomen at 13.5, 15.5, 17.5-days of fetal development were evaluated. Immunohistochemical expression levels of β-catenin, S100B, Lgr5 genes were evaluated in the brain tissues of 13.5, 15.5, 17.5-day fetuses after the administration of LGK 974 (Porcupine inhibitor) which is an inhibitor of Wnt / β-catenin signaling pathway. Thus, the effects of the Wnt / β-catenin signaling pathway on the embryonic nerve and brain development was evaluated. In addition, Toluidin blue and Hematoxylin-Eosin staining were performed to brain tissues and evaluated morphologically. According to the results of the experiment, the weight and lengths of the fetuses, treated with Porcupine inhibitör, decreased statistically significant compared to the other groups. According to the results of immunohistochemistry; in the porcupine inhibitor group, there were no statistically significant changes in β-catenin, S100B, Lgr5 expression compared to the control and DMSO groups. Morphology of all groups was common for hematoxylin-eosin and Toluidine Blue staining. According to these results, we found that Wnt / β-catenin signaling pathway had significant effects on embryo development, but we could not observe any statistically significant change in gene expression levels involved in this pathway. Amaç: Telomerler, ökaryotik kromozomların uç kısımlarında bulunan tekrarlı DNA dizileridir. Biyolojik yaşlanmaya bağlı olarak memeli ovaryumlarında yaşlanmakta ve bu süreçte telomerlerin kısaldığı bilinmektedir. Telomerlerdeki bu kısalmanın moleküler alt yapısı tam olarak bilinmemektedir. Bu çalışmanın amacı telomerlerin uzatılmasında ve telomerlerin bütünlüğünün korunmasında görevli genlerin postnatal hayatın farklı yaşlarındaki fare ovaryumlarında belirlenmesidir. Yöntem: Farelerde normal ovaryal gelişimin erken (1. ve 2. hafta), genç (3. ve 4. hafta), ergin (5. ve 6. hafta), geç ergin (16. ve 18. hafta) ve yaşlı (52., 60. ve 72. hafta) dönem olmak üzere 5 grup (n=3) oluşturuldu. Fare ovaryumlarında Tert, Terc, Trf1, Trf2 ve Pot1a genlerinin mRNA ekspresyonu qRT-PCR ile, protein ekspresyonu ise immünohistokimya ile belirlenmiştir. Elde edilen veriler, one way ANOVA testi ile istatistiksel açıdan değerlendirildi.Bulgular: Analizler sonucunda erken dönem ovaryum dokularındaki Tert mRNA ekspresyonu genç, ergin, geç ergin ve yaşlı dönemlere göre daha yüksek bulunmuştur. Terc mRNA ekspresyon ise erken dönemden yaşlı döneme doğru azaldığı tespit edildi. Trf1 ekspresyonu gruplar arasında farklılık göstermezken; Trf2 ekspresyonu genç ve ergin dönemlerden farklı olarak geç ergin dönemde en düşük düzeyde bulundu. Yaşlı dönemdeki Pot1a ekspresyonu, geç ergin döneme göre daha yüksek bulunmuştur. Gruplar arasında göreli TERT ekspresyonu, erken dönemden yaşlı döneme doğru azalmış, fakat bu azalma anlamlı olarak bulunmamıştır. TRF1 ekspresyonunda istatistiksel olarak bir fark belirlenmemiştir. TRF2 ekspresyonu, erken dönemde yaşlı döneme göre daha düşük bulunmuştur. Erken dönemden geç ergin döneme doğru benzer düzeylerde seyreden POT1 ekspresyonu, geç ergin dönemde azalmakta ve yaşlı dönemde ise yükselmektedir. Sonuç: Ovaryal yaşlanmaya bağlı olarak telomerde gözlenen kısalmada, Tert, Terc, Trf2 ve Pot1a genlerindeki ekspresyonel değişimlerin etkili olabileceği düşünülmektedir. Objective: Telomeres are repetitive DNA sequences found at the ends of eukaryotic chromosomes. Due to biological aging mammalian ovaries are getting aged and telomeres shorten during this process. The molecular background of telomeres shortening is not fully understood. The aim of this study was to determine the genes involved in elongating telomeres and maintaining the integrity of telomeres in the mouse ovaries at different ages of postnatal life.Method: Five groups were formed based on the ovarian development terms in mice as follows: early (1- and 2-week old), young (3- and 4-week old), adult (5- and 6-week old), late adult (16- and 18-week old) and aged (52-, 60- and 72-week old). Tert, Terc, Trf1, Trf2 and Pot1a mRNA expression has been determined by qRT-PCR and their protein expression was characterized by using immunohistochemistry in the mouse ovaries. The statistical significance was evaluated with one way ANOVA test.Results: Tert expression in early group was found to be significantly higher than that of young, adult, late adult and aged groups. The Terc expression decreased from early to aged groups. Although Trf1 expression did not differ among groups, Trf2 expression was at the lowest levels in the late adult groups when compared to young and adult groups. Pot1a expression in the aged group was significantly higher than that of in the late adult groups. The relative TERT expression among the groups decreased from early to aged groups, but this change was not found significant. No statistically significant difference was found in TRF1 expression among groups. TRF2 expression was significantly lower in the early group in comparision to the aged group. The POT1 expression decreased in the late adult groups and increased in the aged group.Conclusion: The expressional changes in Tert, Terc, Trf2 and Pot1a genes are thought to have an effect on telomere shortening during ovarian aging. Amaç: İmplantasyon embriyonun maternal uterusa yerleşme sürecidir. Zigotun blastosiste gelişmesi ve uterusun reseptif hale gelmesinin senkronize olması kritik öneme sahiptir. Sirtuinler, yüksek derecede korunmuş histon/protein deasetilazlardır. Sirtuin 1 oksidatif strese direnç, apoptoz, yaşlanma, inflamasyon gibi süreçlerde rol alır. Sirtuin 1 bu fonksiyonları FoxO gibi transkripsiyon faktörlerini deasetile ederek gerçekleştirir. Parakuat, oksidatif stres oluşturan bir ajandır. Çalışmamızda parakuat ve Sirtuin 1 aktivatörü SRT1720 uygulanan gebe farelerde Sirtuin 1, FoxO1 ve FoxO1'in ekspresyonunu düzenlediği antioksidan olan MnSOD miktarını belirlemeyi amaçladık.Yöntem: Gebeliğin 5. günündeki Balb/C ırkı farelerle kontrol, vehicle, parakuat, SRT1720 ve SRT1720 + parakuat grupları şöyle oluşturuldu: hiçbir enjeksiyonun yapılmadığı kontrol grubu, gebeliğin 1-4. günlerinde SRT1720 ve parakuatın çözücülerinin enjekte edildiği vehicle grubu, gebeliğin 4. günü 30 mg/kg parakuatın enjekte edildiği parakuat grubu, gebeliğin 1-4. günlerinde 50 mg/kg SRT1720'nin enjekte edildiği SRT1720 grubu, gebeliğin 1-4. günlerinde 50 mg/kg SRT1720 ile gebeliğin 4. gününde 30 mg/kg parakuatın enjekte edildiği SRT1720 + parakuat grubu. Tüm gruplardaki fareler gebeliğin 5. günü sabahı Chicago mavisi enjeksiyonundan sonra sakrifiye edilerek implantasyon ve inter-implantasyon bölgeleri ile uterus dokuları alındı. Bu dokularda immünohistokimya ve Western blot ile FoxO1, Sirtuin 1; Western blot ile asetile lizin; ELISA ile ROT/RNT ve MNSOD miktarları belirlendi.Bulgular: Sirtuin 1 nükleer, FoxO1 hem nükleer hem sitoplazmik ekspresyon göstermiştir. Parakuat uygulanan farelerde implantasyon gerçekleşmemiştir. SRT1720 uygulanan gruplarda Sirtuin 1 ve FoxO1 ekspresyonları artmıştır. SRT1720 uygulanan gruplarda Sirtuin 1 aktivasyonuna bağlı FoxO1 deasetilasyonu sonucunda MnSOD miktarı artarak ROT/RNT miktarını azaltmıştır. Sonuç: Parakuat ve SRT1720'nin birlikte uygulandığı farelerin bazılarında MnSOD artışının ROT/RNT'yi ortadan kaldırmasıyla embriyo implantasyonu gerçekleşmiştir. Synchronization of development of zygote to blastocyst and uterus becomes receptive are critical. Sirtuins are highly conserved histone/protein deacetylases. Sirtuin1 is involved in processes such as resistance to oxidative stress, apoptosis, aging and inflammation. Sirtuin1 performs these functions by deacetylating transcription factors such as FoxO. Paraquat is an oxidative stress generating agent. We aimed to determine amount of Sirtuin1, FoxO1 and antioxidant MnSOD regulated by FoxO1 in pregnant mice treated with paraquat and Sirtuin1 activator SRT1720.Method: On 5th day of pregnancy, control, vehicle, paraquat, SRT1720 and SRT1720+paraquat groups formed by Balb/C mice were as follows: control group with no injection; vehicle group SRT1720 and paraquat solvents injected on days 1-4 of pregnancy; paraquat group injected with 30 mg/kg of paraquat 4th day of pregnancy, SRT1720 group injected with 50 mg/kg SRT1720 on days 1-4 of pregnancy; SRT1720+paraquat group injected with 50 mg/kg SRT1720 on days 1-4 of pregnancy and 30 mg/kg paraquat on 4th day of pregnancy. Mice in all groups were sacrificed after Chicago blue injection on day 5 morning of pregnancy, implantation and inter-implantation sites, uteri were taken. FoxO1 and Sirtuin1 were detected with immunohistochemistry and Western blot; acetylated lysine were detected with Western blot; ROS/RNS and MnSOD were detected with ELISA.Results: Sirtuin1 showed nuclear, FoxO1 showed both nuclear and cytoplasmic expression. There was no implantation in paraquat group. Sirtuin1 and FoxO1 expressions were increased in SRT1720 treated groups. As a result of FoxO1 deacetylation due to Sirtuin1 activation in SRT1720 treated groups, increased MnSOD caused ROT/RNT decrease.Conclusion: In some of mice treated with paraquat and SRT1720, embryos implanted with increase of MnSOD by eliminating ROT/RNT. Amaç:Hücreler arası köprüler somatik hücrelerde hücre bölünmesinin sitokinez evresinde oluşan geçici yapılardır. Farede farklılaşan germ hücreleri arasında TEX14 proteininin varlığı ile kalıcı yapılara dönüşen hücreler arası köprülerin, diploid dişi germ hücrelerindeki rolü belirsizliğini korumaktadır. Çalışmamızda, germ hücrelerinde hücreler arası köprü içermeyen Tex14 mutant (Tex14-/-) fareler kullanılarak, hücreler arası köprülerin mayozun senkronizasyonu ve germ hücre havuzunun kurulmasındaki rolü üç boyutlu (3D) analiz yöntemi kullanılarak araştırılmıştır.Yöntem:Wholemount immünofloresan yöntemi ile Tex14+/+ ve Tex14-/- ovaryumlarda germ hücreleri mayoz ve apoptoz belirteçleri ile işaretlendi. Imaris programı ve MATLAB komut dizisi ile ovaryumların 3D haritası oluşturuldu. Ayrıca apoptozun grup/küme analizi gerçekleştirildi. Tex14+/+ ve Tex14-/- ovaryumlarda kimyasal stres oluşturulduktan sonra hücreler arası köprülerin varlığında ve yokluğunda hücre eliminasyonu değerlendirildi.Bulgular:Ovaryumda mayozun ilk olarak radyal bir dalga halinde başladığı, anterior-posterior (AP) mayotik dalganın ise E13.5'tan itibaren etkili olduğu izlendi. Tex14-/- ovaryumlarda, radyal mayotik dalganın olmadığı, sadece AP dalganın korunduğu ve Tex14-/- germ hücrelerinin Tex14+/+'e kıyasla mayoza daha erken başladığı dikkat çekti. Germ hücre eliminasyonunun gerçekleştiği dönemde Tex14-/- ovaryumlarda germ hücre sayısının azaldığı, apoptozun arttığı ve Tex14+/+'den farklı olarak, Tex14-/- germ hücrelerinin ovaryumda rastgele elimine edildiği izlendi. Stres indüklendiğinde ise germ hücre kaybının oransal olarak Tex14+/+'de Tex14-/-'e göre artmış olduğu tespit edildi.Sonuç:Çalışmamızda, ovaryumda hücreler arası köprülerin radyal mayotik dalganın düzenlenmesinde ve apoptoz düzeyinin belirlenmesindeki rolleri literatürde ilk defa ortaya konulmuştur. Fetal germ hücre gelişiminin araştırılması, ovaryan rezervin kurulmasında etkili olan mekanizmaları anlamamızı sağlayarak, infertilite, subfertilite ve oositlerde sıklıkla izlenen anöploidi ile ilgili bilinmeyenlere ışık tutacaktır. Objective: Intercellular bridges (ICBs) are transient structures which form during cytokinesis step of somatic cell division. They transform into permanent structures with the presence of TEX14 protein in differentiating mice germ cells. The role of ICBs in mammalian female germ cell development is still unclear. In our study, we analyzed the role of ICBs in the synchronization of meiosis and establishment of germ cell pool by using 3D analyzing method and Tex14 mutant (Tex14-/-) mice, which lack ICBs in germ cells.Method: Wholemount immunofluorescent staining was performed, Tex14+/+ and Tex14-/- germ cells were labelled with meiosis and apoptosis markers. 3D maps of ovaries were created by using Imaris software and MATLAB. Clustering analysis of germ cells was also applied. Germ cell elimination was analyzed in the absence or presence of ICBs in Tex14+/+ and Tex14-/- ovaries which were subjected to chemical stress.Results: Meiosis first started as a radial meiotic wave and anterior-posterior (AP) meiotic wave was effective in the ovaries earliest at E13.5. The radial wave was absent and pre-mature meiotic entry was observed in Tex14-/-. Germ cell numbers were decreased and germ cell death were increased in Tex14-/- compared to Tex14+/+ when germ cell elimination occurred through apoptosis. Unlike Tex14+/+, Tex14-/- germ cells were eliminated randomly. When ovaries were subjected to chemical stress, further increase of apoptosis following chemical stress was modest in Tex14-/- compared to Tex14+/+ ovaries.Conclusion:In our study, the roles of ICBs in the regulation of radial meiotic wave and in calibrating the threshold of apoptosis in germ cells in the developing ovary were revealed for the first time. Studying germ cell development in great detail will allow us to gain deeper understanding in mechanisms which play a role during the establishment of ovarian reserve and will enlighten infertility, subfertility and aneuploidy defects in females. 113

Amaç: CTCFL, literatürde BORİS (Brother of the Regulator of Imprinted Sites), CCCTC-bağlanma faktörü-benzeri protein ya da CTCF-T olarak ifade edilen bir proteindir. Ctcfl'in aşırı ekspresyonunun embriyogenez sırasında, vasküler bozukluklar, çoklu organ patolojileri ve neonatal ölüm ile bağlantılı olduğu ortaya konulmuştur. Bu noktadan hareketle çalışmamızda, doksisiklin ile Ctcfl gen ekspresyonunun indüklendiği fare embriyonik kök hücrelerinde (EKH), Ctcfl'in aşırı ekspresyonunun in vitro endotel yönünde farklanmada etkisinin olup olmadığının araştırılması hedeflenmiştir.Yöntem: Çalışmamızda Ctcfl transgenik ve transgenik olmayan fare EKH'leri kullanıldı. Kültür medyumuna 2 μg/ml doksisiklin ilavesi ile Ctcfl gen indüksiyonu gerçekleştirildi. Kültür sonrasında ektopik gen ekspresyonunun sağlanıp sağlanmadığı qRT-PCR ile mRNA, Western Blot ile protein düzeyinde değerlendirildi. Ektopik Ctcfl ekspresyonunun yapıldığı transgenik deney grubu ile transgen içermeyen ve gen indüksiyonunun yapılmadığı kontrol grupları çalışmaya dahil edildi. Her grup kendi içerisinde endotel yönünde farklandırılan ve farklandırılmayan şeklinde iki gruba ayrıldı. Faz-kontrast mikroskopik değerlendirme ile endotelyal hücre farklanması gruplar arasında karşılaştırıldı. Flow sitometri yöntemi ile endotelyal farklanmanın erken dönem belirteçlerinden Flk1 ve VE-kaderin proteinleri analiz edildi. Ayrıca Flk1, VE-kaderin, CD31, CD34, VEGF, Flt1 protein ekspresyonları immünofluoresan boyanma ile değerlendirildi.Bulgular: Doksisiklin ile indüklenen grupta Ctcfl gen ve protein ekspresyonları, kontrol gruplarına kıyasla anlamlı bir şekilde yüksek bulundu. Deney grupları arasında Flk1, VE-kaderin, CD31, CD34, VEGF ve Flt1 proteinlerinin ekspresyon paternleri açısından farklılık izlenmedi. Flow sitometri sonuçlarında, Flk1 ve VE-kaderin proteinleri gruplar arasında benzer ortalama fluoresan yoğunluk değerleri gösterdi.Sonuç: Bulgularımızda ektopik Ctcfl ekspresyonunun, in vitro endotel farklanmada Flk1 ve VE-kaderin ekspresyonları açısından belirgin bir etkisinin olmadığı gözlendi. Overexpression of Ctcfl has been shown to be associated with vascular disorders, multiple organ pathologies and neonatal death during embryogenesis. Thus, in the present work, we aimed to investigate whether or not overexpression of Ctcfl induced by doxycycline would affect in vitro endothelial differentiation in Ctcfl transgenic mouse embryonic stem cells (ESCs).Method: Ctcfl transgenic and non-transgenic mouse embryonic stem cells were used. Ctcfl gene induction was performed by adding 2 μg/ml doxycycline to the culture media. Ectopic gene expression was confirmed by qRT-PCR at mRNA and by Western Blot method at protein level after culture. The transgenic group with ectopic Ctcfl gene expression and control groups with no transgene and no gene induction were included in the study. Each group was also divided into two subgroups that cells differentiated into endothelial cells or not. Phase-contrast microscopic evaluation was performed between groups for endothelial cell differentiation. Expression of early markers for endothelial differentiation, Flk1 and VE-cadherin, were analyzed by flow cytometry. Besides, expression of Flk1, VE-kaderin, CD31, CD34, VEGF and Flt1 proteins were assessed by immunofluorescence staining.Results: The Ctcfl gene and protein expressions in the doxycycline-induced group were found to be significantly higher than control groups. There were no differences in the expression patterns for Flk1, VE-cadherin, CD31, CD34, VEGF and Flt1 proteins between experimental groups. Flow cytometry results demonstrated that Flk1 and VE-cadherin proteins showed similar mean fluorescence intensity values in each group. Conclusion: According to our results, ectopic Ctcfl expression has no obvious effects on in vitro endothelial cells differentiation by means of Flk1 and VE-cadherin expressions. Amaç: Bu çalışma dağ nanesinin (Mentha caucasica) Japon bıldırcınlarının ince bağırsakları üzerindeki etkisinin ışık mikroskobik seviyede belirlenmesi amacıyla yapıldı. Gereç ve Yöntem: Çalışmada 1günlük 30 adet Japon bıldırcını kullanıldı. Bıldırcınlar, her biri 10 adet olmak üzere üç gruba ayrıldı. Hayvanlar 6 hafta boyunca kontrol, N1 (%0,75 oranında nane) ve N2 (%1,5 oranında nane) rasyonları ile beslendiler. Çalışma sonunda servikal dislokasyonla öldürülen hayvanlardan duodenum, jejunum ve ileum doku örnekleri alındı. Rutin histolojik işlemlerden sonra alınan seri kesitler Crossmon'un üçlü boyaması ve Periyodik Asit Schiff (PAS) reaksiyonu ile boyandılar. Tüm seri kesitler ışık mikroskobu ile değerlendirildikten sonra dijital kamera ile fotoğraflar çekildi ve kaydedildi. Materials and Methods: One day-old Japanese quail chicks (n=30) were used in the study. The quails were randomly divided into 3 groups which consist of 10 animals for each.as control, M1 (0,75% mint content) and M2 (1,5% mint content). Animals were fed diets containing different mint content for six weeks. At the end of the study, the birds were killed by cervical dislocation. The duodenum, jejunum and ileum samples were collected. Bu çalışmada zebra çiklit (Amatitlania nigrofasciata) ventrikülünün morfolojik ve histolojik özellikleri ışık mikroskobu ile incelenmiştir. Ventrikül, piramit biçimlidir ve ventrikül duvarı epikardiyum, miyokardiyum ve endokardiyum tabakalarından meydana gelmiştir. Epikardiyum ve endokardiyum tek katlı yassı epitel ile ince bir bağ dokudan oluşmuştur. Daha kalın bir tabaka olan miyokardiyum, kolayca ayırt edilebilen iki tabaka halinde izlenmiştir; dış tabaka, sıkı şekilde organize olmuşken iç tabaka, dallanmış kalp kası demetleri arasında yer alan iri trabekülleri ile süngerimsi görünüme sahiptir. Ventrikülün piramidal biçimi ve kas düzenlenişi metabolik aktivitenin önemli bir parametresi olarak değerlendirilerek tartışılmıştır., The morphological and histological structures of the ventricle of convict cichlid (Amatitlania nigrofasciata) were described light microscopically. The ventricle was pyramidal in shape, and its wall was comprised of epicardium, myocardium, and endocardium. Epicardium and endocardium were thin and had similar histological structure that consisted of simple squamous epithelium and connective tissue. The thicker layer, myocardium, was composed of two layers differentiated easily: the outer part was compactly organized while the inner one was observed as spongy in appearance with large trabeculae oriented between branched bundles of cardiac muscle. The pyramidal shape and myoarchitectural features of the ventricle were evaluated and discussed as being the most important histological parameter of metabolic activity which is depended on myocardial functions.

Amaç: CCM1, CCM2 ve CCM3 genlerinde tanımlanan mutasyonlar sporadik ya da kalıtsal olup santral sinir sisteminde görülen serebral kavernoz malformasyonların (SKM) sebebidir. Bu genlerin fonksiyonları hala tam olarak anlaşılamamasına rağmen; hücre-hücre bağlantılarında, migrasyonda, hücrenin oryantasyonunda ve apoptozunda önemli rollerinin olduğu gösterilmiştir. SKM proteinleri hücre içerisinde üçlü özel bir yapı oluşturmakta ve adezyon molekülleri ile hücre iskeleti arasında bağlantı kurmaktadırlar. Bu bilgilere göre; çalışmamızda, serebral kavernoz malformasyonda tanımlanmış ve daha önce primer tümör ve metastazlardaki etkisi araştırılmamış CCM1, CCM2 ve CCM3 proteinlerinin hem primer tümörlerde hem de akciğer ve karaciğer gibi metastatik dokulardaki ekspresyonlarını değerlendirmeyi amaçladık.Materyal ve Yöntem: Çalışmamızda; 4T1 (metastatik meme kanseri hücre hattı) hücrelerinin karaciğer ve kalp dokularına metastazları sonrasında elde edilen 4TLM ve 4THM hücreleri kullanıldı. Hücreler 500.000 hücre/fare olacak şekilde 8-10 haftalık Balb/C dişi farelerin meme dokularına ortotopik olarak enjekte edildi. Enjeksiyondan 26 gün sonra fareler sakrifiye edilerek primer tümörleri, akciğer ve karaciğer dokuları çıkartıldı, parafin bloklar hazırlandı. CCM1, CCM2 ve CCM3 proteinlerinin ekspresyonları immünohistokimya ve western-blot tekniği ile değerlendirildi. Tüm bulgular tümör enjeksiyonu yapılmamış kontrol hayvanları ile kıyaslanarak yorumlandı.Bulgular: Primer tümör dokularında üç proteinin ekspresyonu da gözlendi. Tümör enjekte edilen farelerin akciğer ve karaciğerlerinde CCM1 ve CCM2 benzer şiddette ekspresyon patternine sahipken; CCM3 daha yoğun bir immunoreaksiyon sergiledi. Metatatik dokuların özellikle metastaz alanlarında CCM proteinlerinin reaksiyonuna rastlanmadı. Bu alanlarda gözlenen nadir immunreaksiyonların kupfer hücrelere ait olduğu görüldü.Sonuç: CCM proteinlerinin primer tümörlere kıyasla metastatik dokulardaki ekspresyon farkları ve metastatik alanlarda ekspresyonlarının görülmeyişi; bu proteinlerin metastaz da rol oynayabileceğini ve invaze olan tümör hücrelerinin adezif bağlantı yapılarını değiştirerek kendi mikroçevrelerini yaratabileceğini düşündürmektedir. Objective: Sporadic or familial mutations in at least three genes, CCM1, CCM2, CCM3, cause Cerebral cavernous malformations (CCMs) which mostly occurs in the central nervous system. The precise functions of these genes are not fully understood. Studies show that the proteins are thought to have important roles on cell-cell junctions, migration, and apoptosis. All three proteins can be found in a same complex within the cell and they maintain stabilization of cell junctions. However; the role of CCM proteins on tumor development and metastasis has not been explained yet. Therefore, the aim of the study was to evaluate the expressions of CCM1, CCM2, CCM3 proteins in primary breast cancer tumor and highly metastatic organs lung and liver.Method: In our study; 4TLM, 4THM cell lines obtained from the metastasis of 4T1 (metastatic breast cancer cells) cells to liver, heart tissues were used. 4TLM, 4THM (500.000 cell/mouse) cells were injected to breast tissues of Balb/C female mice (8-10 weeks old) orthotopically. After 26 days, the primary tumors, liver and lung tissues were obtained and paraffin blocks were prepared. CCM1, CCM2 and CCM3 immunoreactions were evaluated by immunohistochemistry. All the findings were interpreted by comparing control animals that were not injected with tumor cells.Results: It was observed three CCM protein expressions in primary tumor tissues of all tumor injected animals. The expressions of CCM1 and CCM2 in lung and liver tissues were similar to the other. CCM2 expressions were vulnerable but, CCM3 had higher immunoreactivity in both groups. Interestingly, CCM proteins were not seen in metastatic tumor mass in liver tissues. But, CCM proteins were mainly observed in Kupffer cells and macrophages.Conclusion: The loss of CCM expressions in metastatic tissues and metastatic tumor mass which may change adherent junctions' stability, create it's own microenvironment and facilitate metastasis to distant organs. Therefore; CCM proteins might have a role new perspective of carcinogenesis. 74

Doğurganlığı koruma teknikleri, kanser gibi yıkıcı kemoterapi ve radyoterapi uygulamaları içeren hastalıkların tedavisi sonrasında, kısırlık ve hormonal yetmezliklerle başetmede önemli bir seçenektir. Üremeye yardımcı tekniklerin kullanıma sunduğu embriyo, oosit ve ovaryum kriyoprezervasyonu, kadın hastaların ovaryum rezervlerinin ve doğurganlıklarının, bu tedaviler öncesinde korunması için bir kolaylık sağlamaktadır. Ancak, bekâr ve pre-pubertal hastalarda kendisini en fazla kanıtlamış yöntem olan embriyo kriyoprezervasyonu seçeneği uygun değildir. Kanser tanısı sebebiyle bir an önce tedavi planına başlanması gereken hastalar için oosit toplama süreci zaman kaybına neden olmaktadır. Hormon uyarısıyla tümör dokusunu indükleme riskinin olduğu durumlarda (örneğin östrojen reseptörü pozitif olan meme kanseri olguları) ovaryum dokusu kriyoprezervasyonu tek seçenek olarak karşımıza çıkmaktadır ve günümüzde bildirilmiş 60 kadar gebelik olgusu mevcuttur. Primordiyal folikülden primer foliküle geçişte, kontrollü aktive edici ve baskılayıcı rol oynayarak folikül havuzunun korunmasında rol alan sinyal yolakları birbirinden bağımsız ancak dengeli bir şekilde çalışmalıdır. Baskılayıcı ve aktivatör genlerin dengeli çalışmadığı durumlarda, klinikte prematür ovaryan yetmezlik olarak bilinen, foliküllerin kitlesel aktivasyonu ile folikül havuzunun yarısından fazlasının hatta daha büyük kısmının erkenden tükenmesine sebep olmaktadır. Bununla bağlantılı olarak, ovaryum kriyoprezervasyonu ve transplantasyonu sonrasında geciken anjiyogenez ve oluşan hipoksi ovaryumda primordiyal folikülden primer foliküle geçişte görev alan intrensek mekanizmaları bozuyor olabilir. Ovaryum doku kriyoprezervasyonu ve transplantasyonu sonrasında meydana gelen folikül kaybının nedenleri arasında foliküllerin büyümelerinin baskılanma mekanizmasının bozulmuş olabileceğini araştırdığımız bu çalışmada pTEN, Tsc1, p27 ve AMH moleküllerinin ekspresyonlarını değerlendirdik. Bulgularımız, literatüre benzer şekilde dondurulup çözme ve transplantasyon sonrasında ovaryumda primordiyal folikül havuzunun azaldığını gösterdi. pTEN, Tsc1, p27 ve AMH moleküllerinin ekspresyonlarını değerlendirdiğimizde K grubu ile DÇ grubunda bir fark gözlenmezken, T ve DÇT gruplarında bu moleküllerin pTEN, Tsc1 ve p27 ekspresyonlarının primordiyal foliküllerde, AMH ekspresyonunun ise büyüyen foliküllerde neredeyse tamamen ortadan kalktığı gözlendi. Ovaryum kriyoprezervasyonu ve transplantasyonu sonrasında, primordiyal folikülden primer foliküle geçişte rol oynayan baskılayıcı moleküllerin ekspresyonlarını araştırdığımız bu çalışma ile literatürde ilk kez bu konuyu değerlendirmiş oluyoruz. Sonuçlarımız, ovaryum doku kriyoprezervasyonu ve transplantasyonu sonrasında primordiyal folikül kaybı ile ilişkili sürece yeni bir yaklaşım sunmaktadır ve primordiyal folikülden primer foliküle geçişte fren mekanizmasının adeta bozulduğunu işaret etmektedir. Primordiyal folikülden primer foliküle geçişte rol oynayan aktivatörler ve baskılayıcıların bu süreçte fonksiyonel değerlendirmelerini yaptığımız yeni çalışmalar ovaryum kriyoprezervasyonu ve transplantasyonu ile ilişkili süreçleri daha da aydınlatabilecektir. Fertility preservation techniques are crucial options for compete with infertility and hormonal insufficiency or cancer theraphies including destructive implementation such as chemotheraphy and radiotheraphy. Embryo, oocyte and ovarian tissue cryopreservation that offered by the use of assisted reproductive techniques are simplicity for preserving ovarian reserve of female patients for improving their fertility. However, even if embryo cryopreservation method has evidence-based methodology but not feasible for unmarried for prepubertal female patients. Oocyte pick-up process means waste of time for female cancer patients whose immediately need to start scheduling their theraphy. Ovarian tissue cryopreservation is unique option in some cases of tumor tissue induction risk via hormonal stimulation (e.g. estrogen receptor positive breast cancer cases). It is already known that currently methods for cancer theraphies are increased survival rate of human race. But, it's not easy to handle complications on reproductive systems. Ovarian cryopreservation is unprecedented fertility method for the group of chosen patients. Although increasing cancer incidence, a decreasing the proportion of patients who died because of cancer. Most irreversable complication from these cell destructive theraphies are damages through reproductive cells and organs. Fertility preservation theraphies would be offered to patients meanwhile and carry out with most feasible option and nearly 60 cases of gestation are announced to present day. It is restricted evidence based information about injury mechanisms due to retransplanting frozen tissue into body. Transitioning from primordial to primer follicle form, there are controlled activators and suppessors for balancing signalisation of reserving primordial follicle pool like a seesaw. In some cases of imbalancing these activators and supressors, result in activation of follicles globally then burn-out high percentage of primordial follicle pool, known as `premature ovarian insufficiency` (POI) at clinically. In respect of this, late angiogenesis and period of hipoxia may damage intrinsic mechanisms of ovary. In our study, we evaluated expressions of pTEN, Tsc1, p27 and AMH molecules for asking reasons of follicle lost after ovarian tissue cryopreservation and transplantation. Our findings show that, primordial follicles are decreasing after frozing-thawing and transplantation litterateurly correlate. We criticize expressions of pTEN, Tsc1, p27 molecules for primordial follices but growing follicles for AMH molecules. There is not much difference between K and DÇ group. Between T and DÇT group, we almost detected hardly any expressions of these molecules. With our study, we firstly point out expressions of supressor mechanisms at ovarian cryopreservation and transplantation period. Our data represents new aspects underlying process of primordial follicle activation after ovarian cryopreservation and transplantation. New functional studies about activator and inhibitor mechanisms about primordial to primer follicle transition will provide new point of view about preserving follicle pool. 68

Amaç: Kronik glomerüler ve tübülointerstisyel fibrozis, son dönem böbrek yetmezliğinin en yaygın sebebidir. Böbrek, sınırlı koşullarda organın kendisini yenilemesine izin veren intrinsik rejeneratif kapasiteye sahiptir. Mezenkimal kök hücre (MKH) tedavisi, böbrek hasarının rejenerasyonu için umut verici sonuçlar sunmaktadır. MKH'lerin terapötik etkilerinin gösterilebilmesi açısından ölçülebilir biyobelirteçler büyük önem taşımaktadır. Adiponektin, kronik böbrek yetmezliği (KBY) gelişimi için önerilen biyobelirteçlerden biridir. Çalışmamızda, sıçanlara subtotal nefrektomi yaparak kronik böbrek yetmezliği oluşturuldu ve insan plasentasından elde edilen mezenkimal kök hücreler enjekte edilerek böbrek hastalığında önemli bir biyobelirteç olabileceği önerilen adiponektinin rolü ve bu rol üzerine mezenkimal kök hücrelerin etkisi incelendi.Yöntem: Çalışmamızda ilk olarak term plasentalarındaki amniyon membranından mezenkimal kök hücrelerin izolasyonu gerçekleştirildi. Elde edilen hücrelerin flow sitometre ile karakterizasyonu yapıldı ve ardından mezenkimal hücrelerin adiposit, kondrosit ve osteositlere yönlendirilmesi sağlandı. Deney gruplarını oluşturmak için Wistar türü erkek ratların sol böbreklerine ligasyon yapıldıktan iki hafta sonra sağ böbrekleri tamamen alınarak 5/6 nefrektomi yapılmış oldu. Daha sonra bazı gruplara kök hücre enjeksiyonu uygulandı. Tüm gruplarda Adiponektin, Adiponektin Reseptör 1 (AdipoR1) ve Fibronektin'in protein düzeyleri western blot yöntemi ile, mRNA ekspresyonları ise Real-Time PCR yöntemi ile tespit edildi. Ayrıca, Adiponektin, sinyalizasyonu açısından AMPK fosforilasyonu Western Blot yöntemi ile değerlendirildi. Son olarak da adiponektinin idrar ve serum düzeyleri sıçana özgü ELISA kiti kullanılarak ölçüldü.Bulgular: Adiponektin, AdipoR1 ve Fibronektin protein ve mRNA ekspresyonlarında ve AMPK fosforilasyonunda nefrektomi yapılan gruplarda kontrol gruplarına kıyasla bir artma gözlenirken; 5/6 nefrektomi yapıldıktan sonra kök hücre verilmiş olan sıçanlarda, kök hücre verilmemiş KBY'li gruplar ile karşılaştırıldığında, istatistiksel olarak anlamlı bir şekilde azalma belirlendi. Ayrıca, kök hücre verilen gruplarda serum adiponektin düzeylerinde istatistiksel olarak anlamlı bir artma gözlenirken, idrar adiponektin düzeylerinde ise anlamlı bir azalma tespit edildi.Sonuç: Sonuç olarak, kan ve idrarda ölçülebilir bir biyobelirteç olan adiponektin kronik böbrek hastalığının ilerlemesinde önemli bir role sahiptir. Objective: Chronic glomerular and tubulointerstitial fibrosis is the most common cause of end-stage renal failure. The kidney has an intrinsic regenerative capacity that allows the organ to regenerate itself under limited conditions. Mesenchymal stem cell therapy offers promising results for regeneration of kidney damage. measurable biomarkers are of great importance for demonstrating the therapeutic effects of mesenchymal stem cells. Adiponectin is one of the recommended biomarkers for the development of chronic renal failure. In our study, we performed subtotal nephrectomy in rats to establish chronic renal failure and injected mesenchymal stem cells from human placenta to investigate the role of proposed adiponectin and the effect of mesenchymal stem cells on this role as an important biomarker in renal disease.Methods: In our study, mesenchymal stem cells were isolated from the amnion membrane of term placentas. The obtained cells were characterized by flow cytometry and then mesenchymal cells were directed to adipocytes, chondrocytes and osteocytes. In order to construct the experimental groups, 5/6 nephrectomy was performed after two weeks after the ligation of the left kidneys of male Wistar rats. Some groups were then injected with stem cells. Protein levels of Adiponectin, Adiponectin Receptor 1 (AdipoR1) and Fibronectin were determined by western blot method and mRNA expressions by Real-Time PCR method in all groups. In addition, adiponectin was evaluated for signaling by AMPK phosphorylation by Western Blot method. Finally, urine and serum levels of adiponectin were measured using a rat specific ELISA kit.Findings: Adiponectin, AdipoR1 and Fibronectin protein and mRNA expressions and AMPK phosphorylation were increased in the nephrectomy groups compared to the control groups. After 5/6 nephrectomy, there was a statistically significant decrease in stem cell-injected rats compared with groups with untreated stem cells. In addition, there was a statistically significant increase in serum adiponectin levels and a significant decrease in urinary adiponectin levels in the stem cell group.Conclusion: As a result, adiponectin, a measurable biomarker in blood and urine, plays an important role in the progression of chronic kidney disease. mesenchymal stem cells may play a therapeutic role in terms of clinical trials and design of experimental systems in chronic renal failure.Key words: Mesenchymal stem cell, chronic renal failure, adiponectin, fibrosis. 111

Embriyo fallop tüplerinden uterusa doğru yol alırken implantasyona hazırlanır ve endometriyuma implante olana kadar geçirdiği süreç preimplantasyon dönem embriyo gelişimi olarak tanımlanır. Preimplantif dönemdeki embriyo, yarıklanma bölünmelerine başladığında; 1 hücreli, 2 hücreli, 4 hücreli, 8 hücreli, morula ve blastosist aşamasındaki embriyoyu oluşur. Preimplantif embriyo gelişim süreci, maternal çevrede bulunan büyüme faktörleri tarafından üretilen sinyallere bağlı olarak pek çok farklı hücresel olay tarafından etkilenmektedir. PI3K/Akt sinyal yolağı, büyümeyi ve gelişmeyi destekleyen pek çok sinyal yolağından birisidir. FoxO transkripsiyon faktörleri, nükleusta görev yaparlar ve aktiviteleri PI3K/Akt sinyal yolağı tarafından düzenlenir. Memelilerde, FoxO1, FoxO3a, FoxO4 ve FoxO6 olmak üzere dört adet FoxO ailesi üyesi tanımlanmıştır. FoxO'lar; glikoz metabolizması, enerji homeostazı, hücresel farklanma, oksidatif strese karşı dayanıklılık, hücre siklusunun duraklaması, DNA tamiri ve apoptozun düzenlenmesi, gibi süreçlerde önemli roller oynarlar. Tüm bu bilgiler, bir yandan çoğalırken diğer yandan farklanan ve mikroçevresi sürekli değişirken ortama adapte olmaya çalışan preimplantif gelişim sürecindeki embriyoda, FoxO transkripsiyon faktörlerinin de ekspre olabileceği sorusunu akla getirmektedir. Bu çalışmanın amacı, preimplantasyon embriyo gelişim sürecinde FoxO transkripsiyon faktörlerinin ekspre olup olmadığının, eğer oluyor ise hücredeki lokalizasyonlarının belirlenmesidir. Çalışmamızda 5-6 haftalık Balb/C ırkı dişi fareler kullanılarak hormonal indüksiyon (PMSG ve hCG) ile; profaz I, metafaz I ve metafaz II evresindeki oositler ve erişkin Balb/C ırkı erkek fareler ile çiftleşme sonucunda elde edilen 1 hücreli, 2 hücreli, 4 hücreli, 8 hücreli, morula ve blastosist aşamasındaki embriyolar fallop tüpleri ve uteruslarından toplanmıştır. İmmünfloresan yöntemi kullanılarak; FoxO1, FoxO3 ve FoxO4 proteinlerinin ekspresyonları ve lokalizasyonları ortaya konmuştur. Çalışmamız; profaz I, metafaz I ve metafaz II oositlerinde ve 1 hücreli, 2 hücreli, 4 hücreli, 8 hücreli, morula ve blastosist aşamasındaki embriyolarda, FoxO1, FoxO3 ve FoxO4 proteinlerinin farklı düzeylerde ekspre edildiğini gösteren literatürdeki ilk çalışmadır. Sonuçlarımız, in vivo embriyo gelişimi sürecinde sitoplazmada ekspre olduğu belirlenen FoxO transkripsiyon faktörlerinin özellikle in vitro embriyo gelişimi sırasında veya stres koşullarında (pH, oksijen düzeyi, ısı değişimleri gibi) ekspresyonlarının ne şekilde olabileceği ile ilgili yapılabilecek çalışmalar için temel oluşturmuştur. Özellikle yardımcı üreme tekniklerinde embriyo gelişim süreçlerinde FoxO transkripsiyon faktörlerinin araştırılması ile ilgili ileri çalışmalar laboratuvarımızda devam etmektedir. After fertilization, while passing through the fallopian tubes to enter the uterus, embryo gets prepared for implantation. The period between fertilization and implantation is defined as preimplantation embryo development. Fertilized zygote starts to cleave and develops into 2 cell, 4 cell, 8 cell embryo, morula, and blastocyst. Preimplantation embryo developmental period is affected by various cellular events that depend on signals generated by growth factors present in the maternal environment. The PI3K/Akt pathway is one of these pathways that support growth and development. FoxO transcription factors are members of a subgroup of Forkhead family which are characterized by a conserved DNA-binding domain and function in the nucleus. Their activities are regulated by PI3K/Akt signalling pathway. In mammals, there are four FoxO genes, FoxO1, 3, 4, and 6. FoxO transcription factors are at the interface of crucial cellular processes, orchestrating programs of gene expression that regulate apoptosis, cell-cycle arrest, and oxidative stress resistance, DNA repair, glucose metabolism, energy homeostasis and differentiation. In the presence of growth factors, FoxO transcription factors are localized in the cytoplasm. Under stress conditions such as oxidative stress and genotoxic stress, FoxO transcription factors move to the nucleus and trigger transcriptional activities of their target genes. Studies have shown that FoxO molecules play important roles and protect fertility in male and female reproduction. Additionally, it has been shown that the PI3K/Akt signaling pathway, the main regulator of FoxOs', is present both in the oocyte and in the preimplantation embryo. Thus, the aim of the present study is to investigate whether FoxO transcription factors are present during oocyte maturation and preimplantation embryo development. Female Balb/C mice were superovulated with PMSG and hCG and then prophase I, metaphase I, metaphase II oocytes, 1 cell embryo, 2 cell embryo, 4 cell embryo, 8 cell embryo, morula and blastocyst were collected from their fallopian tubes and uterus. Presence and localizations of FoxO1, FoxO3 and FoxO4 proteins have been determined with immunofluorescence staining. Our results have confirmed that FoxO1, FoxO3 and FoxO4 proteins are differentially expressed in prophase I, metaphase I, metaphase II oocytes and in 1 cell embryo, 2 cell embryo, 4 cell embryo, 8 cell embryo, morula and blastocyst. Our findings are the first results in the literature showing that FoxO transcription factors are present during both oocyte and embryo in vivo maturation. These findings may provide basic information for planning future studies which investigate whether these factors are affected or not when the embryo faces stress conditions (such as pH, oxygen level or temperature changes) during in vitro maturation, especially in the IVF laboratories. Based on the findings of the present study, new studies are in progress in our laboratory. 68

Metotreksat, kanser tedavisinde sıklıkla kullanılan kemoterapotik bir maddedir. Ayrıca testis üzerinde de toksik bir etki göstermektedir. Hücre ve dokular, meydana gelen bu toksik etkilerden kurkumin gibi antioksidan sistemlerle korunabilirler. Bu çalışmada, metotreksat ile oluşturulan sıçan testis hasarına karşı kurkuminin etkilerinin ortaya konulması amacıyla 28 adet Spraque-Dawley türü sıçan kullanıldı. Sıçanlar her grupta 7 erişkin erkek sıçan olacak şekilde kontrol, 20 mg/kg metotreksat, 200 mg/kg kurkumin, 20 mg/kg metotreksat+200 mg/kg kurkumin verilen grup olmak üzere 4 gruba ayrıldı. Deney sonunda sıçanların vücut ve testis ağırlıkları ölçüldü ve karşılaştırmalar yapıldı. Sol testisler doku takip işlemi için Bouin çözeltisi içerisine, sağ testisler ise %10' luk formaldehit içerisine alındı ve rutin histolojik işlemlerden sonra bloklandı. Elde edilen parafin bloklardan 3 µm kalınlığında seri kesitler alındı ve testislerden alınan kesitler Hematoksilin + Eozin ve Periyodik Asit-Schiff+Hematoksilin ile boyanarak mikroskobik incelemeleri yapıldı. Çalışmanın sonucunda elde ettiğimiz bulgulara göre, metotreksatın vücut ağırlığını azalttığı, testiste seminifer tübüllerde ve hücrelerde hasara yol açarak spermatogenezi etkilediği gözlendi. Metotreksat ile birlikte verilen kurkuminin ise testiste oluşan bu hasarı azalttığı gözlendi.Anahtar Sözcükler : Metotreksat, Kurkumin, Sıçan, Testis Methotrexate is a frequently used chemotherapeutic agent, it also has toxic effect on testes. Cells and tissues may be protected from these toxic effects by antioxidant substances like curcumin. In this study, 28 Sprague - Dawley rats were used to determine the effects of curcumin on metotrexate induced testicular damage in rats. Rats were divided into 4 groups as control, 20 mg/kg methotrexate, 200 mg/kg curcumin, 20 mg/kg methotrexate + 200 mg/kg curcumin, including 7 adult male rats in each group. At the end of administration period, body and testes weights were measured and compared to each other. Left testes were put into Bouin solution, right testes were put into 10% formaldehyde for tissue processing and blocked after the routine histological procedures. Serial sections with 3μm thickness were obtained from those paraffin blocks and microscopical examinations were performed on sections of testes after stained by Hematoxyline + Eosine and Periodic Acid- Schiff + Hematoxyline. Results showed that methotrexate causes the loss of body weight in rats. Methotrexate also affects the spermatogenesis by damaging the seminiferous tubule and cells. On the other hand, it was observed that administration of curcumin together with methotrexate might reduce this damage in rat testicle.Key words : Methotrexate, Curcumin, Rat, Testes 120

Meme kanseri kadınlarda sıkça görülen ve tedavi edilmezse ölüme sebebiyet verebilen bir kanser türüdür. Meme kanseri metastaz yapma yeteneği sebebiyle tehlikeli bir hastalıktır. Metastaz; tümörün bulunduğu yerden ayrılıp başka bir organda yeni bir tümör oluşturmasıdır. Metastazın gerçekleşebilmesi için tümör önce bulunduğu dokuda büyüyüp gelişmeli daha sonra da damarlar yardımıyla uzak organlara göç etmelidir. Tümör metastazı sırasında yeni gideceği çevredeki normal yapıyı bozarak kendine bir ortam yaratmalıdır. Bu yeniden düzenlenmede de hücre dışı matriks proteinleri de görevalmaktadır.Nefronektin; epiteliyal mezenkimal geçişte rol oynadığı bilinen bir hücre dışı matriks proteinidir. Tümör gelişiminde rol alabileceği düşünülen nefronektinin metastatik tümörler üzerindeki etkisini gösteren bir çalışma henüz yapılmamıştır. Bu nedenle çalışmamızda metastatik ve metastatik olmayan hücre hatlarını kullanarak oluşturulan primer tümörlerde ve metastaz alanlarında nefronektin ekspresyonunu değerlendirmeyi amaçladık. Çalışmamızda metastatik olduğu bilinen 4TLM ve 4THM; ve metastatik karakterde olmayan 67NR hücre hatları enjekte edilen farelerden 12. ve 25. günlerde alınan pimer tümör, akciğer ve karaciğer dokuları, ışık mikroskobu ve western blot araştırmaları için hazırlandı. Elde edilen kesitlerde nefronektin ve Ki67 proteinleri için immünohistokimya metodu uygulanarak bu proteinlerin dokulardaki dağılımları belirlendi. Western blot tekniği kullanılarak da nefronektin ve Ki67 ekspresyon miktarları protein düzeyinde araştırıldı. Bulgularımıza göre; nefronektin ekspresyonunun primer tümör, akciğer ve karaciğerlerde, metastazın yoğunluğuna bağlı olarak azaldığı gözlendi. Ayrıca metastazın ileri evresi olan 25. günde, metastazın henüz gerçekleşmediği 12. güne göre nefronektin ekspresyonunda bir azalma olduğu görüldü. Proliferasyon belirteci olan Ki67'nin; metastatik hücre hatlarından alınan dokularda yoğun olarak lokalize olduğu; ayrıca metastatik olmayan hücre hattından alınan dokularda da yoğun olmasa da varlığı belirlendi. Çalışmamız; metastatik yetenekteki hücre hatlarından elde edilen primer tümör, akciğer ve karaciğer dokularında nefronektin ekspresyon değişimini gösteren literatürdeki ilk çalışmadır. Elde edilen bulgular; hücrelerin dokuya invaze olabilmeleri ve hücre dışı matriks düzenini değiştirerek metastaz yapabilmelerinde nefronektin proteininin bağlantı noktalarında ayrılmaları sebebiyle ekspresyonunun düştüğü ve hücre dışı matriksdeki yerleşiminin bozulduğu ihtimalini düşündürmektedir. Özellikle metastazın yoğun olduğu alanlarda ekspresyonunun düşmesi bu proteinin metastatik süreçte görevli olma ihtimalini akla getirmektedir. Breast cancer, which can cause of death if its not treated, is commonly occurs in women . One of the reason of breast cancer as a dangerous illness high quality of the its metastatic ability. Metastasis means that; a tumor split from an organ and create a new tumor one another organ. For existing metastasis firstly tumor grow up in settled tissue and then migrate to a new organ via blood vessels. During the tumor metastasis, tumor create a new environment with disturb to normal tissue structure a new tissue. So during the reprogramming to extracellular matrix proteins also play a role.Nephronectin, an extracellular matrix proteins and as a play a role epitelial mesenchymal transition. There is no working about the effect of nephronectins which can take a role in tumor grows on metastatic tumors. So, in this study we aimed to determine the nephronectin expressions by using metastatic and non-metastatic cell lines in primary tumors and metastasis.In our study, 4TLM and 4THM cell lines, known as metastatic, and 67NR cell lines, known as non-metastatic, is ortotopicaly injected to mice. Primary tumors, lung and liver tissues, taken in 12th and 25th day, is prepared for light microscopy and western blot investigations. Immunohistochemistry methods are implied for nephronectin and Ki67 proteins in tissue sections; and localization of this proteins are determined. The protein expression levels of nephronectin and Ki67 were also investigated by western blot.According to our results, decreased of nephronectin expressions were observed in primary tumors, lung and liver in 4TLM and 4THM injected mice. Moreover there is a notablu decrease of nephronectin expressions 25thday ,high level of metastasis, than 12th day, not yet metastasis. Ki67 as a proliferation marker is highly localizated in metastatic cell lines tissues. It is also seemed not highly, in non-metastatic cell line tissues. Ki67 expression levels is clearly observed in tissue taken from 25th day than the tissue taken of 12th day.In our study we showed for the first time in literature that the expression levels of nephronectin demonstrated in primary tumors, lung and liver tissues which is obtained from metastatic cell lines. Result of our study shows that the possibility of that cancer cells can disturb the localization of nephronectin proteins in extracellular matrix for invasion to tissue and reprogramming extracellular matrix to metastasis. Bir immünofilin olan FK506'ya bağlanan protein 52 (FKBP52), steroid hormon reseptör aktivitesini değiştiren bir Hsp90 koşaperonudur. FKBP52'nin reseptör-şaperon kompleksleri ile işbirliği sonucu reseptörün hormona bağlanması artar. Daha önce yapılan çalışmalarda CD1 ve C57BL6/129 genetik zeminli Fkbp52 geni çıkarılmış (Fkbp52-/-) dişi farelerde ovulasyon ve fertilizasyonun normal olduğu ancak implantasyonun gerçekleşmediği gözlenmiştir. Fkbp52-/- dişi farelere gebeliklerinin 2-4. günleri boyunca Progesteron (P4) enjeksiyonu yapıldığında CD1 genetik zeminli olanlarda implantasyon gerçekleşmekte ancak C57BL6/129 genetik zeminli olanlarda gerçekleşmemektedir.P4 enjeksiyonu yapılan CD1 genetik zeminli Fkbp52-/- farelerde implantasyon gerçekleşmesine karşın C57BL6/129 genetik zeminli olanlarda gerçekleşmemesi, CD1 Fkbp52-/- farelerde gebeliğin terme ulaşabilmesi için daha fazla P4 düzeylerine gereksinim duyulması, plasentada Fkbp52 eksprese edilmesine karşın Pgr'nin eksprese edilmemesi bilgilerinden yola çıkarak FKBP52'nin steroid hormon P4'ün reseptörü olan Progesteron reseptörü (PR) aktivitesini optimize etmekten başka bir fonksiyonu olabileceği hipotezini kurduk.Yaptığımız 2 boyutlu ayrım jel elektroforezi ve kütle spektrometresi sonucunda Fkbp52-/- uterusta P4-PR sinyalinden bağımsız olarak sadece antioksidan protein Peroksiredoksin6 (PRDX6)'nın azaldığını bulduk. Bu proteinin spesifik olarak uterusta azaldığını Northern Blot yöntemi ile gösterdik. Ayrıca 8-izoprostan assay oksidatif stresin uterusa spesifik olduğunu gösterdik.Normalde implantasyonun %100 oranında gerçekleştiği P4 uygulanan Fkbp52-/- farelere oksidatif stres indükleyici ajan parakuat enjeksiyonu bu fareleri oksidatif strese daha hassas hale getirerek implantasyonu büyük ölçüde inhibe etti. Bu farelere parakuatın yanı sıra N-Asetil sistein ve ?-Tokoferol antioksidanlarının uygulanması bu farelerdeki implantasyon başarısını artırdı.Desiduada FKBP52 ile PRDX6 arasında fiziksel etkileşim olduğunu immünopresipitasyon yöntemiyle gösterdik.Fkbp52-/- fareler ile in vivoda elde ettiğimiz bulguları in vitroda fare embriyonik fibroblast çalışmaları ile doğruladık.Elektron mikroskopi incelemeleri ile Fkbp52-/- farelerin uterusunda vahşi tip uterusa oranla ultrastrüktürel düzeyde değişiklikler olduğunu gösterdik.Çalışmamız, FKBP52'nin PR kompleksindeki koşaperon fonksiyonunun yanı sıra uterusta PRDX6 ile etkileşip oksidan/antioksidan düzeylerini etkileyerek gebeliği oksidatif stresten koruduğunu göstermiştir. FK506 binding protein (FKBP52) is an immunophilin which serves as a Hsp90 cochaperone to influence steroid hormone receptor function. Association of FKBP52 with receptor-chaperone complex increases the binding of receptor-hormone binding. Previous studies showed that Fkbp52-/- mice on CD1 and C57BL6/129 genetic backgrounds have normal ovulation and fertilization but have complete implantation failure. P4 supplementation rescued implantation in CD1 Fkbp52?/? females but not in C57BL6/129 Fkbp52?/? females. Hence P4 supplementation can rescue implantation in CD1 Fkbp52?/? females but not in C57BL6/129 Fkbp52?/? females, CD1 Fkbp52?/? females need higher amount of P4 to maintain pregnancy to full term and expression of Fkbp52 in the placenta without Pgr we hypothesized that FKBP52 should have another function independent of its PR cochaperone activity.As a result of two dimensional difference gel electrophoresis and mass spectrometry, antioxidant protein Peroxiredoxin6 (PRDX6) was found to be the only protein that downregulated in Fkbp52-/- uterus independent of P4-PR signaling. We confirmed specific downregulation of Prdx6 in the uterus by Northern Blotting. In addition we demonstrated that oxidative stress was specific to the uterus via 8-isoprostane assay.Injection of paraquat which is an oxidative stress inducing agent to P4 treated Fkbp52-/- mice that normally have %100 implantation inhibited implantation substantially because these mice became more vulnerable to oxidative stress by paraquat injection. Treatment of these mice with N-acetylcysteine and ?-tocopherol antioxidants in addition to paraquat increased implantation success.We showed physical interaction between FKBP52 and PRDX6 in the decidua via immunoprecipitation.We confirmed our in vivo findings about Fkbp52-/- mice also by in vitro studies performed on mouse embryonic fibroblasts.We showed changes in the uteri of Fkbp52-/- mice compared to wild type mice at ultrastructural level via electron microscopy.Our study showed that in addition to its cochaperone function in PR complex FKBP52 also interacts with PRDX6 to balance oxidant/antioxidant status in the uterus and protects pregnancy. 181
