One of the greatest challenges in integrated circuit design is the repeated executions of computationally expensive SPICE simulations, particularly when highly complex chip testing/verification is involved. Recently, pseudo-transient analysis (PTA) has shown to be one of the most promising continuation SPICE solvers. However, the PTA efficiency is highly influenced by the inserted pseudo-parameters. In this work, we proposed BoA-PTA, a Bayesian optimization accelerated PTA that can substantially accelerate simulations and improve convergence performance without introducing extra errors. Furthermore, our method does not require any pre-computation data or offline training. The acceleration framework can either speed up ongoing, repeated simulations (e.g., Monte-Carlo simulations) immediately or improve new simulations of completely different circuits. BoA-PTA is equipped with cutting-edge machine learning techniques, such as deep learning, Gaussian process, Bayesian optimization, non-stationary monotonic transformation, and variational inference via reparameterization. We assess BoA-PTA in 43 benchmark circuits and real industrial circuits against other SOTA methods and demonstrate an average of 1.5x (maximum 3.5x) for the benchmark circuits and up to 250x speedup for the industrial circuit designs over the original CEPTA without sacrificing any accuracy.

To predict mutations in TP53 and PIK3CA genes in breast cancer using ultrasound (US) signatures and clinicopathology.In this study, we developed and trained a model in 386 breast cancer patients to predict TP53 and PIK3CA mutations. The clinicopathological and US characteristics (including two-dimensional and color Doppler US) were investigated. Statistically significant variables were used to build predictive models, then a combined model was developed using the multivariate logistic regression analysis.Univariate and multivariate analyses revealed that calcifications on US was an independent predictor of TP53 mutation (p0.05), whereas diameter on US and US type were independent predictors of PIK3CA mutation in breast cancer (all p0.05). Meanwhile, Luminal B/Human epidermal growth factor receptor two-positive (HER2+), HER2+/estrogen receptor-negative (ER-), and triple-negative breast cancer (TNBC) subtypes were strong predictors of TP53 mutation (odds ratio [OR] = 3.13, 3.18, 3.44, respectively, all p0.05). HER2+/ER- and TNBC subtypes were negative predictors of PIK3CA mutation (OR = 0.223, 0.241, respectively, all p0.05). The areas under curves (AUCs) for PIK3CA mutation in the training set increased from 0.553-0.610 to 0.741 in the multivariate model that combined US features and molecular subtype, with a sensitivity and specificity of 80.6% and 58.7%, respectively. The application of the multivariate model in the validation set achieved acceptable discrimination (AUC = 0.715). For TP53 mutation, the AUC was 0.653.US is a non-invasive modality to recognize the presence of TP53 and PIK3CA mutation. The models combined with US features and molecular subtype have implications for the practical application of predicting gene mutation for individual decision-making regarding treatment planning.

The changes of intestinal microbiota are closely related to the growth and development of animals. The current study is aimed at exploring the composition of the microbial community of pigs at different growth stages. Fresh fecal samples of three-breed hybrid pigs at three developmental stages (60, 120, and 180 days of age) were collected. The microbial composition was analyzed based on the 16S rDNA gene of bacteria Illumina NovaSeq sequencing platform. The results showed that the intestinal microbiota of pigs was distributed in 22 phyla, 46 classes, 84 orders, 147 families, and 287 genera. Firmicutes, Bacteroides, Spirochaetae, Proteobacteria, and Actinobacteria were the dominant phyla. Lactobacillus, Streptococcus, SMB53, Oscillospira, and Prevotella were the dominant genera. Among them, the abundance of Lactobacillus and SMB53 increased first and then decreased, while the change of Oscillospira was opposite. In addition, the abundance of Streptococcus increased while that of Prevotella decreased gradually. Moreover, with the increase of time and body weight, the microbial diversity showed a decreasing trend. In conclusion, the intestinal microbial composition of the three-breed hybrid pigs was relatively stable during the fattening stage, but there were significant differences in abundance.

Summary In the full life cycle of a well, thermal and mechanical loads may yield serious issues for the cement sheath integrity. However, the information for the integrity assessment, such as temperature and strain, is difficult to acquire underground. In this study, a full-scale experimental facility is used, allowing us to mimic the casing-cement sheath-formation (CCSF) system of a well. The system is monitored by fiber Bragg grating (FBG), enabling a real-time, high-accuracy, nondestructive measurement of temperature and strain inside the cement sheath in the sequence of setting and completion stage. Our observation reveals that the temperature of the cement sample cured in the mold is 22.3°C higher than the curing temperature; however, this temperature difference is not observed in the cement sheath cured in the CCSF system. This implies that the data obtained from the cement sample may overestimate the early-age performance of the cement sheath. Besides, the FBG measures a free strain of the tested cement during the hydration to be −370 με. This shrinkage can yield an internal stress in the CCSF system, which leads the cement sheath to swell circumferentially during the setting stage. During the completion stage, when the cement sheath is subjected to cyclic loading at three casing pressure levels, (i i.e., 10, 20, and 50 MPa), the maximum increment of circumferential strain reaches 160, 270, and 850 με, respectively. A plastic strain is observed for the 50 MPa pressure level, but not for the two other pressure levels (10 and 20 MPa). Unlike the observations in cyclic loading tests on cement samples, the plastic strain in the CCSF system accumulates linearly in the first 10 cycles and then increases slowly afterward. This difference is suggested to be attributed to the redistribution of internal stress along with the accumulation of plastic strain. Finally, the strains measured by the FBG are validated by the simulation, demonstrating the promising applicability of the FBG technology for monitoring the integrity of cement sheath.

BACKGROUND: The disorder of LINC01207 has a significant regulatory effect on cancers, nevertheless its role in non-small cell lung cancer (NSCLC) have not been illustrated. This study investigated the regulatory effect of LINC01207 on NSCLC and clarify its molecular mechanism. METHODS: Bioinformatics analysis was used to find the target lncRNA, miRNA and mRNA. LncBase and TargetScan databases predicted the relationship between LINC01207, miR-525-5p and ARHGAP11A. Dual-luciferase reporter gene assay and RNA binding protein immunoprecipitation assay were used to verify the binding relationship between genes. Fluorescence in situ hybridization assay was used to localize the expression of LINC01207 in NSCLC tissue. qRT-PCR and Western blot assays were used to measure the expression of LINC01207, miR-525-5p and ARHGAP11A. CCK-8 assay, Transwell assay and flow cytometry assay were used to detect NSCLC cell abilities. Mouse xenograft models further determined the effect of LINC01207 on the growth of NSCLC in vivo. RESULTS: LINC01207 was up-regulated in NSCLC tissue and cells, which was mainly localized in the cytoplasm. LINC01207 knockdown could inhibit the proliferation, migration and invasion of cancer cells and induce cell apoptosis. In addition, silencing LINC01207 could suppress tumor growth in vivo. LINC01207 could sponge and inhibit the expression of miR-525-5p in NSCLC cells, and inhibiting LINC01207 and miR-525-5p simultaneously could reverse the effect of miR-525-5p on the progression of NSCLC cells. Further study on downstream target genes showed that miR-525-5p could restrain the expression of ARHGAP11A, and then affect the progression of NSCLC. LINC01207 acting as a competing endogenous RNA (ceRNA) could regulate the expression of ARHGAP11A by competitively binding with miR-525-5p. CONCLUSION: LINC01207 regulates the progression of NSCLC by regulating the miR-525-5p/ARHGAP11A axis as a ceRNA and plays a carcinogenic role. In conclusion, our study elucidates the mechanism of LINC01207 regulating the progression of NSCLC, and provides a new idea for the diagnosis and treatment of NSCLC guided by lncRNA.

Pancreatic ductal adenocarcinoma (PDAC) usually presents infrequent infiltration of T lymphocytes. The known immune-checkpoint inhibitors to date focus on activating T cells and manifest limited effectiveness in PDAC. SIGLEC15 was identified as a novel tumor-associated macrophage (TAM)-related immune-checkpoint in other cancer types, while its immunosuppressive role and clinical significance remained unclear in PDAC. Background Even though a fair amount of studies focus on depression among college students, the effect of perceived parenting styles on the incidence of major depressive disorder (MDD) among representative freshmen in Chinese context is scarcely studied. The aim of this study is to investigate the effect of parenting styles on MDD in Chinese freshmen. Methods A total of 9,928 Chinese freshmen were recruited in 2018. 6985 valid questionnaires were collected at one-year follow-up. Composite International Diagnostic Interview 3.0 (CIDI-3.0) was used for the diagnosis of MDD. Egna Minnen Beträffande Uppfostran (EMBU) questionnaire and Beck Depression Inventory-II (BDI-II) were used to assess parenting styles and baseline depressive symptoms, respectively. The associations between parenting styles and MDD incidence was analyzed with logistic regression. Results The incidence of MDD in freshmen was 2.23% (95%CI: 1.91-2.60%). Maternal overprotection (OR = 1.03, 95%CI: 1.01–1.05) and disharmony relationship between parents (OR = 2.35, 95% CI: 1.42–3.89) increased the risk of new-onset MDD in freshmen, respectively. Mild depressive symptoms (OR = 2.06, 95%CI: 1.06–4.02), moderate (OR = 4.64, 95%CI: 2.55–8.44) and severe depressive symptoms (OR = 7.46, 95%CI: 2.71–20.52) at baseline increased the risk of new-onset MDD. Conclusions Maternal overprotection, disharmony relationship between parents and baseline depressive symptoms are risk factors for new-onset MDD in Chinese freshmen.

BackgroundLOX family members are reported to play pivotal roles in cancer. Unlike their enzymatic activities in collagen cross-linking, their precise cancer functions are unclear. We revealed that LOXL4 is highly upregulated in breast cancer cells, and we thus sought to define an unidentified role of LOXL4 in breast cancer.MethodsWe established the MDA-MB-231 sublines MDA-MB-231-LOXL4 mutCA and -LOXL4 KO, which stably overexpress mutant LOXL4 that loses its catalytic activity and genetically ablates the intrinsic LOXL4 gene, respectively. In vitro and in vivo evaluations of these cells’ activities of cancer outgrowth were conducted by cell-based assays in cultures and an orthotopic xenograft model, respectively. The new target (s) of LOXL4 were explored by the MS/MS analytic approach.ResultsOur in vitro results revealed that both the overexpression of mutCA and the KO of LOXL4 in cells resulted in a marked reduction of cell growth and invasion. Interestingly, the lowered cellular activities observed in the engineered cells were also reflected in the mouse model. We identified a novel binding partner of LOXL4, i.e., annexin A2. LOXL4 catalyzes cell surface annexin A2 to achieve a cross-linked multimerization of annexin A2, which in turn prevents the internalization of integrin β-1, resulting in the locking of integrin β-1 on the cell surface. These events enhance the promotion of cancer cell outgrowth.ConclusionsLOXL4 has a new role in breast cancer progression that occurs via an interaction with annexin A2 and integrin β-1 on the cell surface.

We adopted Sequence MASSarray to identify genotyping, and used immunohistochemistry (IHC) to test the expression of estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor-2 (HER-2) in tumor tissue.Results: We found that rs16999593 (TC/CC vs TT: adjusted OR=1.38, 95% CI: 1.03– 1.84, p=0.030) was associated with an increased risk of BC, while rs2228611 was related to a decreased BC risk (GA/AA vs GG: adjusted OR=0.74, 95% CI: 0.56– 0.97, p=0.030). Intestinal microbiota not only participates in the digestion and absorption of nutrients, but also plays an important role in regulating host metabolism and health. The current study aimed to explore the intestinal microbiota characteristics in pigs infected with African swine fever. Below the same term, fresh fecal samples of sick and healthy pigs were collected. Inflammation and immunity play a major role in the development of hypertension, and a potential correlation between host mucosal immunity and inflammatory response regulation. We explored the changes of intestinal mucosal microbiota in hypertensive rats induced by high-salt diet and the potential link between the intestinal mucosal microbiota and inflammation in rats. Therefore, we used PacBio (Pacific Bioscience) SMRT sequencing technology to determine the structure of intestinal mucosal microbiota, used enzyme-linked immunosorbent assay (ELISA) to determined the proinflammatory cytokines and hormones associated with hypertension in serum, and used histopathology methods to observe the kidney and vascular structure. We performed a potential association analysis between intestinal mucosal characteristic bacteria and significantly different blood cytokines in hypertensive rats induced by high-salt. The results showed that the kidney and vascular structures of hypertensive rats induced by high salt were damaged, the serum concentration of necrosis factor-α (TNF-α), angiotensin II (AngII), interleukin-6 (IL-6), and interleukin-8 (IL-8) were significantly increased (p p p > 0.05). From the perspective of gut microbiota, high-salt diet leads to significant changes in intestinal mucosal microbiota. Bifidobacterium animalis subsp. and Brachybacterium paraconglomeratum were the dominant differential bacteria in intestinal mucosal, with the AUC (area under curve) value of Bifidobacterium animalis subsp. and Brachybacterium paraconglomeratum were 1 and 0.875 according to ROC (receiver operating characteristic) analysis. Correlation analysis showed that Bifidobacterium animalis subsp. was correlated with IL-6, IL-8, TNF-α, and Ang II. Based on our results, we can speculated that high salt diet mediated chronic low-grade inflammation through inhibited the growth of Bifidobacterium animalis subsp. in intestinal mucosa and caused end-organ damage, which leads to hypertension.

BACKGROUND There has been a growing need for mental health services in adolescents. However, access for this population to professional service providers is limited thus leaving a gap between the demand and availability of mental healthcare resources. OBJECTIVE The goal of this study is to evaluate the functionality and effectiveness of a school digital mental health clinic (DMHC) provided by a psychiatric hospital in China. METHODS Use of an app driven DHMC at three secondary schools in Wenzhou, China between January and July 2021 was evaluated. Qualitative analysis was conducted to determine the app’s functional effectiveness while quantitative on the volume of counseling calls delivered, number of calls per hundred students, length of call, and reasons for the calls. Mini-course videos offered by the DMHC was analyzed by topics and viewing time. RESULTS The DMHC’s designed functions are well aligned with required factors defined in the literature. Majority of the counseling calls ((58%) and mini-course video viewing (62%) occurred at non-business hours. More senior high students used the DMHC than junior. Reasons for counseling calls include difficulties in learning, interpersonal relationships, and emotional distress. Students were mostly interested in three topics of the mini-course videos, emotional assistance, personal growth and family relationships. The DMHC provided crisis intervention for three students with high risks for suicide and self-injury. CONCLUSIONS The DMHC serves as a complementary solution particularly during non-business hours to address the mental healthcare needs of adolescents. Extension of the DMHC into more schools, more urgently senior high schools is recommended.

Background Migration can be linked to the transmission of COVID-19. COVID-19 vaccine uptake and hesitancy among rural-to-urban migrant workers in China, the largest group of internal migrants in the world, has not been characterized. Objective To investigate COVID-19 vaccine uptake and identify vaccine hesitancy-associated factors among rural-to-urban migrant workers in the first round of COVID-19 vaccination in China. Methods A cross-sectional questionnaire-based survey was conducted, including 14,917 participants. Socio-demographics, COVID-19 vaccine uptake, vaccine hesitancy and its associated factors based on Vaccine Hesitancy Determinants Matrix (VHDM) were applied for the survey. Data were principally analyzed by logistic regression analysis. Results The COVID-19 vaccine uptake and vaccine hesitancy rates were 7.1% and 57.7%, respectively. Vaccine hesitancy was strongly associated with VHDM, including individual factors (female, higher annual income and fewer medical knowledge), group factors (less family support, friend support and public opinion support), COVID-19 epidemic factors (lower fatality, infection and emotional distress) and vaccine factors (less vaccine necessity, vaccine safety, vaccine efficacy, vaccine importance and vaccine reliability). Conclusion The VHDM model has the potential utility in efforts to reduce COVID-19 vaccine hesitancy. Greater efforts should be put into addressing positive predictors associated with vaccine hesitancy.

Pseudomonas aeruginosa is one of the most important foodborne pathogens that can persist in leafy green vegetables and subsequently produce biofilms. In this study, the synergistic effect of thymoquinone and nisin in reducing biofilm formation of P. aeruginosa on lettuce was evaluated, and their anti-virulence and anti-biofilm mechanisms were also investigated. At concentrations ranging from 0.5 to 2 mg/ml, thymoquinone inhibited the production of autoinducers and virulence factors, and enhanced the susceptibility of P. aeruginosa biofilms to nisin as evidenced by the scanning electron microscopy and confocal laser scanning microscopy. Integrated transcriptomics, metabolomics, and docking analyses indicated that thymoquinone treatment disrupted the quorum sensing (QS) system, altered cell membrane component, and down-regulated the expressions of genes related to virulence, efflux pump, and antioxidation. The changed membrane component and repressed efflux pump system enhanced membrane permeability and facilitated the entrance of nisin into cells, thus improving the susceptibility of biofilms to nisin. The dysfunctional QS and repressed antioxidant enzymes lead to the enhancement of oxidative stress. The enhanced oxidative stress disrupted energy metabolism and protein metabolism and ultimately attenuated the virulence and pathogenicity of P. aeruginosa PAO1. Our study indicated that thymoquinone has the potential to function as a QS-based agent to defend against foodborne pathogens in combination with nisin.

A mixed dihedral group is a group $H$ with two disjoint subgroups $X$ and $Y$, each elementary abelian of order $2^n$, such that $H$ is generated by $X\cup Y$, and $H/H'\cong X\times Y$. In this paper, for each $n\geq 2$, we construct a mixed dihedral $2$-group $H$ of nilpotency class $3$ and order $2^a$ where $a=(n^3+n^2+4n)/2$, and a corresponding graph $\Sigma$, which is the clique graph of a Cayley graph of $H$. We prove that $\Sigma$ is semisymmetric, that is, ${\rm Aut}(\Sigma)$ acts transitively on the edges, but intransitively on the vertices, of $\Sigma$. These graphs are the first known semisymmetric graphs constructed from groups that are not $2$-generated (indeed $H$ requires $2n$ generators). Additionally, we prove that $\Sigma$ is locally $2$-arc-transitive, and is a normal cover of the `basic' locally $2$-arc-transitive graph ${\rm K}_{2^n,2^n}$. Autonomous drone racing is becoming an excellent platform to challenge quadrotors' autonomy techniques including planning, navigation and control technologies. However, most research on this topic mainly focuses on single drone scenarios. In this paper, we describe a novel time-optimal trajectory generation method for generating time-optimal trajectories for a swarm of quadrotors to fly through pre-defined waypoints with their maximum maneuverability without collision. We verify the method in the Gazebo simulations where a swarm of 5 quadrotors can fly through a complex 6-waypoint racing track in a 35m * 35m space with a top speed of 14m/s. Flight tests are performed on two quadrotors passing through 3 waypoints in a 4m * 2m flight arena to demonstrate the feasibility of the proposed method in the real world. Both simulations and real-world flight tests show that the proposed method can generate the optimal aggressive trajectories for a swarm of autonomous racing drones. The method can also be easily transferred to other types of robot swarms.

Background: Cirrhosis is a chronic progressive liver disease. Hepatocyte injury leads to liver dysfunction. At present, antiviral drugs are mainly used to inhibit the replication of hepatitis virus to block liver fibrosis. Tenofovir disoproxil fumarate is a novel nucleotide reverse transcriptase inhibitor, which can eliminate hepatitis B virus and control the deterioration of chronic hepatitis B. Objective To investigate the efficacy and mechanism of Xuefu Zhuyu Decoction combined with tenofovir disoproxil fumarate tablets in the treatment of hepatitis B cirrhosis of liver depression and blood stasis. Methods A total of 150 patients with hepatitis B cirrhosis who were treated in our hospital from January 2019 to June 2021 with the dialectical type of "liver stagnation and blood stasis" were selected and divided into groups according to their treatment plan. 75 cases in the control group were given Fu Tenofovir disoproxil marate tablets were treated, and 75 patients in the observation group were treated with Xuefu Zhuyu Decoction combined with tenofovir disoproxil fumarate tablets. Count the total effective rates and adverse reactions of the two groups, record the scores of TCM syndromes, liver hemodynamics, fibrosis, liver function, hepatitis B virus deoxyribonucleic acid (HBV-DNA), MMP1, TIMP1, TIMP1/MMP1 changes. Results The curative effect of the observation group was higher than that of the control group, which was statistically significant (P0.05). After treatment, PW and HV-HA in the observation group were higher than those in the control group (P0.05). Compared with before treatment, the scores of the two groups of chest and hypothermia distended, abdominal distension, mental fatigue, depression, irritability, and amitrati decreased (P0.05). Conclusion Xuefu Zhuyu Decoction combined with tenofovir disoproxil fumarate tablets is effective in treating hepatitis B cirrhosis of liver depression and blood stasis, and its mechanism may be related to improving liver hemodynamics, regulating fibrosis, liver function and other indicators related.

Background Protein interactions is the foundation of all metabolic activities of cells, such as apoptosis, immune response and metabolic pathway. In order to optimize the performance of protein interaction prediction, a coding method based on normalized difference sequence characteristics (NDSF) of amino acid sequences is proposed. Methods By using the positional relationships between amino acids in the sequences and the correlation characteristics between sequence pairs, NDSF is jointly encoded. Using principal component analysis (PCA) and local linear embedding (LLE) dimensionality reduction methods, the coded 174-dimensional human protein sequence vector is extracted by using sequence features. This study compares the classification performance of four ensemble learning methods (AdaBoost, Extra trees, LightGBM, XGBoost) applied to PCA and LLE features, and uses cross-validation and grid search methods to find the best combination of parameters. Results The accuracy of NDSF is generally higher than that of MOS coding method, and the loss and coding time can be greatly reduced. The bar chart of feature extraction shows that the classification accuracy is significantly higher when using the linear dimensionality reduction method PCA than the nonlinear dimensionality reduction method LLE. After classification with XGBoost, the model accuracy reaches 99.2%, which is the best performance among all models. Conclusions NDSF combined with PCA and XGBoost may be an effective strategy for classifying different human protein interactions.
