Genetics ,Plant Science ,Biotechnology - Abstract
Background Traditional CRISPR/Cas9 systems that rely on U6 or U3 snRNA promoters (RNA polymerase III-dependent promoters) can only achieve constitutive gene editing in plants, hampering the functional analysis of specifically expressed genes. Ribozyme-mediated CRISPR/Cas9 systems increase the types of promoters which can be used to transcribe sgRNA. Therefore, such systems allow specific gene editing; for example, transcription of the artificial gene Ribozyme-sgRNA-Ribozyme (RGR) is initiated by an RNA polymerase II-dependent promoter. Genetic transformation is indispensable for editing plant genes. In certain plant species, including pyrethrum, genetic transformation remains challenging to do, limiting the functional verification of novel CRISPR/Cas9 systems. Thus, this study’s aim was to develop a simple Agrobacterium rhizogenes-mediated hairy root transformation system to analyze the function of a ribozyme-mediated CRISPR/Cas9 system in pyrethrum. Results A hairy root transformation system for pyrethrum is described, with a mean transformation frequency of 7%. Transgenic hairy roots transformed with the pBI121 vector exhibited significantly increased beta-glucuronidase staining as a visual marker of transgene expression. Further, a ribozyme-based CRISPR/Cas9 vector was constructed to edit the TcEbFS gene, which catalyzes synthesis of the defense-related compound (E)-β-farnesene in pyrethrum. The vector was transferred into the hairy roots of pyrethrum and two stably transformed hairy root transgenic lines obtained. Editing of the TcEbFS gene in the hairy roots was evaluated by gene sequencing, demonstrating that both hairy root transgenic lines had DNA base loss at the editing target site. Gas chromatography–mass spectrometry showed that the (E)-β-farnesene content was significantly decreased in both hairy root transgenic lines compared with the empty vector control group. Altogether, these results show that RGR can be driven by the CaMV35S promoter to realize TcEbFS gene editing in pyrethrum hairy roots. Conclusion An A. rhizogenes-mediated hairy root transformation and ribozyme-mediated CRISPR/Cas9 gene editing system in pyrethrum was established, thereby facilitating gene editing in specific organs or at a particular developmental stage in future pyrethrum research.

To study the difference of long non coding RNA (lncRNA) expression profile in bone marrow specimens of children with acute leukemia (AL) and other hematological disease children with normal bone marrows as controls, to screen the lncRNA related with childhood hematological diseases, and to explore the expression of lncRNA AC002454.1 and its clinical significance in AL children. The microarray gene chip technology was used to statistically analyze the lncRNA in bone marrow cells of newly diagnose AL children and control children. Ninty-seren differentially expressed lncRNAs were selected. The bone marrow specimens of ALL children (21 cases), AML children (22 cases) and control children (21 cases) were verified and compared by using qRT-PCR; then the lncRNA with maximum differential expression-lncRNA AC002454.1 was selected and used to analyze the relation of relative expression level with clinical indicators. The microarray gene chip detection showed that 1 884 differentially expressed lncRNA were found in ALL children, and 4 289 differentically expressed lncRNA were found in AML children. The results confirming these differentically expressed lncRNA by qRT-PCR showed that 9 lncRNA expression were significantly up-regulated in ALL children, and 12 lncRNA expression were significantly up-regulated in AML children. Among these up-regulated lncRNA, the difference of AC002454.1 expression was most significant in ALL and AML children (P<0.05, P<0.01). The detection showed that there was a significant difference, in AC002454.1 relative expression level of newly diagnosed T-ALL and B-ALL children (P<0.01), moreover, this difference also was found in ALL and AML children (P<0.05). The detection analysis showed that there was no statistical difference in AC002454.1 relative expression level among the different sex, age, WBC count at initial diagnosis, chromosome, fusion gene, and risk stratification (P>0.05 for all). The lncRNA expression profile of AL children has been gained by using the lncRNA microarray gene chip technicology. AC002454.1 the significantly high expression exist in AL children, which relates with immunotyping and prognosis of AL children in a certain degree.

In this paper, we describe our submissions to the WMT20 shared task on parallel corpus filtering and alignment for low-resource conditions. The task requires the participants to align potential parallel sentence pairs out of the given document pairs, and score them so that low-quality pairs can be filtered. Our system, Volctrans, is made of two modules, i.e., a mining module and a scoring module. Based on the word alignment model, the mining module adopts an iterative mining strategy to extract latent parallel sentences. In the scoring module, an XLM-based scorer provides scores, followed by reranking mechanisms and ensemble. Our submissions outperform the baseline by 3.x/2.x and 2.x/2.x for km-en and ps-en on From Scratch/Fine-Tune conditions, which is the highest among all submissions., Comment: WMT 2020

In this paper, we study automatic question generation, the task of creating questions from corresponding text passages where some certain spans of the text can serve as the answers. We propose an Extended Answer-aware Network (EAN) which is trained with Word-based Coverage Mechanism (WCM) and decodes with Uncertainty-aware Beam Search (UBS). The EAN represents the target answer by its surrounding sentence with an encoder, and incorporates the information of the extended answer into paragraph representation with gated paragraph-to-answer attention to tackle the problem of the inadequate representation of the target answer. To reduce undesirable repetition, the WCM penalizes repeatedly attending to the same words at different time-steps in the training stage. The UBS aims to seek a better balance between the model confidence in copying words from an input text paragraph and the confidence in generating words from a vocabulary. We conduct experiments on the SQuAD dataset, and the results show our approach achieves significant performance improvement.

Compositional structures between parts and objects are inherent in natural scenes. Modeling such compositional hierarchies via unsupervised learning can bring various benefits such as interpretability and transferability, which are important in many downstream tasks. In this paper, we propose the first deep latent variable model, called RICH, for learning Representation of Interpretable Compositional Hierarchies. At the core of RICH is a latent scene graph representation that organizes the entities of a scene into a tree structure according to their compositional relationships. During inference, taking top-down approach, RICH is able to use higher-level representation to guide lower-level decomposition. This avoids the difficult problem of routing between parts and objects that is faced by bottom-up approaches. In experiments on images containing multiple objects with different part compositions, we demonstrate that RICH is able to learn the latent compositional hierarchy and generate imaginary scenes.

A high-quality work–family balance (WFB) can enhance job satisfaction and superior task performance, as well as reduce absenteeism and turnover rate, thereby enabling organisations to improve the effectiveness and commitment of their employees. This research investigates the features of quality WFB for female employees in a casino hotel in Macau. Many female employees continue to face the double burden of their careers and families. Long working hours, shift duties, demanding customers and stressful environment have become neglected phenomena in the casino business and could lead to an unbalanced WFB. Female employees experience disappointments and frustrations when work prevents them from providing care to their families. Previous studies have determined that male employees have better WFB than female employees. In particular, male employees know how to deal with their emotions, face responsibilities, take action and solve problems. By contrast, females are challenged to achieve WFB. Over the past decades, practitioners in the main hospitality industry have become increasingly interested in the female workforce. The high levels of family responsibility and shift work that females handle may cause them to experience high levels of work-family roles. Compared with male employees, female employees display lower levels of shift work tolerance but report more fatigue and sleepiness whilst working in a risk-exposed environment. This qualitative study involves face-to-face interviews with 30 female casino hotel employees (e.g., dealers, service staff, supervisors and managers) from 8 large major casino hotels. These respondents include single and married female frontline employees. Content analysis is performed to elaborate the female employees’ perception of WFB and identify their challenges in achieving such balance. Recommendations are provided for HR practitioners in terms of the appropriate policies and strategies to improve WFB of female casino hotel employees. Therefore, the proper approach will enable casino hotels to retain the best talents and increase the productivity and competitiveness of their organisation.

Having been widely used in a VoIP system, the ITU-T G.729 standard is of high computational complexity, a heavy load for a real-time operation on a low-end smart phone or other embedded devices even with the G.729A standard. In this paper, there are three methods proposed for an efficient complexity reduction of G.729A, i.e., a combined local optimal pulse method, employed to search an algebraic codebook, a discontinuous pitch estimation, used to analyze the open-loop pitch, and the hybrid two-stage vector quantization (HTVQ), assigned to quantize the line spectral frequency (LSF) coefficients. It is validated by experimental results that the improved version of G.729A can be easily implemented on a smart phone (312 MHz CPU, 64 MB RAM) in real-time operation. In total 80.94% of computational load can be thus eliminated in comparison with the G.729A and a satisfactory speech quality is maintained as intended, an excellent efficiency demonstration by the methods proposed.

To investigate the relationship between genetic polymorphism in exon 12 C1236T, exon 21 G2677T/A and exon 26 C3435T of the multidrug resistance 1 (MDR1) gene and the risk of childhood acute lymphocytic leukemia (ALL).A total of 176 patients with ALL and a cohort of 170 matched healthy subjects were included. SNaPshot SNP typing was used to determine the genotypes of MDR1 C1236T, G2677T/A, C3435T. Based on the clinical data, the relationship between genetic polymorphism of MDR1 and the risk of childhood ALL was analyzed. There was significant difference in the distribution of genotype of MDR1 C3435T between the group of controls and cases. The mutant homozygous TT genotype was found to be associated with occurrence of ALL (P = 0.000; OR = 4.504). The data show evidence of pairwise linkage disequilibrium between the three common SNPs (C1236T-G2677T/A-C3435T). The haplotypes of TTT, TGC, CGC and CAC were predominant. The haplotype CGT distributed significantly differently between the groups of controls and cases (P = 0.034). The frequency of the haplotype TTT/TTT in the high risk group was higher than the other groups (P = 0.037).The present findings suggest that 3435C→T polymorphism in MDR1 gene may be a genetic susceptibility factor for ALL. The haplotype of MDR1 (C1236T-G2677T/A-C3435T) could be the clinical parameter at diagnosis.

Pruning operators are used to reduce the search tree in logic programming computation. Pruning operators typically are not declarative, result in a number of semantic problems. We focus on the pruning operator in Godel, called commit, which can be used to prune away parts of a search tree and not destroy the declarative semantics of program. We propose the method to realize the control facility in logic programming language which can support the fully implementation of Godel.

To investigate the expression of matrix metalloproteinases (MMPs) and tissue inhibitors of metalloproteinases (TIMPs) in the sclera of lamina cribrosa in rat chronic elevated intraocular pressure (IOP) and its the relationship with the pathogenesis of glaucoma. It was an experimental study. Chronic ocular hypertension (OHT) model was induced in the left eyes of 43 Wistar rats by cauterizing episcleral venous while the right eyes were used as control. The rats were divided into three groups: 13 for immunohistology, 15 for Western blotting, and 15 for RT-PCR by randomly computerized assignment and sacrificed at week 12 after OHT induction. The expression of MMP-2, MMP-9, TIMP-1, and TIMP-2 in the sclera lamina cribrosa was detected using immunohistology, Western blotting, and RT-PCR technique, respectively. Repeated measures ANOVA of two-factor Studies in SAS statistic software were used to analyze the data. After 12 weeks, compared with control eyes, the IOP of rat OHT eyes was significantly (F = 1519.67, P0.01) elevated. The expression of MMP-2 and MMP-9 was increased in cell nucleus and cytoplasm in sclera lamina cribrosa in rat OHT when compared to control eyes with negative expression. No expressions of TIMP-1, TIMP-2 in sclera lamina cribrosa were found in both OHT and control eyes. The mean gray-scale values of MMP-2, MMP-9, TIMP-1, and TIMP-2 in OHT were significantly (F = 405.55, F = 436.11, F = 1167.77, and F = 4629.64, P0.01) higher than control eye by Western blotting (193.88 +/- 8.84 vs 117.38 +/- 10.76, 202.65 +/- 8.37 vs 134.13 +/- 5.06, 283.63 +/- 5.65 vs 186.88 +/- 7.14, and 284.75 +/- 5.50 vs 183.0 +/- 5.58), while by RT-PCR (200.50 +/- 3.25 vs 181.88 +/- 9.36, 200.13 +/- 2.95 vs 181.75 +/- 5.85, 201.88 +/- 3.14 vs 179.25 +/- 9.21, and 195.50 +/- 3.55 vs 179.75 +/- 7.12), respectively. The increased expression of MMP-2, MMP-9, TIMP-1, and TIMP-2 in lamina cribrosa in rat OHT eye indicates that MMPs and TIMPs may be involved in the pathogenesis of glaucoma.

This paper is concerned with the design of type systems for logic programming so as to satisfy the requirements of modern logic programming. The design of type systems is based on the language Godel which has a strongly type system based on many-sorted logic with parametric polymorphism. The definitions of the basic logic programming concepts of terms, atoms, programs are given in the setting of polymorphic many-sorted logic. In particular, an unification algorithm for typed predicates is proposed for the compiler construction of Godel.

The rapid growth of weblog provides a rich resource for social community mining. In this paper, we put forward a novel research problem of recommendation under the circumstance of a blog system. Most of the previous works usually focus on recommendation for registered blogger, few present a comprehensive investigations on recommendation for potential bloggers who have not provided personalized information in blog system. With the rapid popularity of social networks in recent years, non-personalized recommendation has become one of big challenges in different scenarios. We present a algorithm on the inhomogeneous graph to calculate the important value of an object, and then combined with similarity value to do recommendation. The algorithms which we proposed learn multi parameters in the random walk model, which is also very important in searching problems. The experimental results show that our proposed approaches can achieve good performance.

Mixed-model straight/U-shaped assembly line has been recognized as a relevant component of Just-In-Time (JIT) production line system. For this system, “Heijunka” design is also challenged as both the task assignment and the production sequence affect the workload imbalance among workstations. In this context and recognizing uncertain task time environment that is often observed in actual manufacturing scene, this research addresses the Line Balancing Problem (LBP) and the Product Sequencing Problem (PSP) jointly and proposes a mathematical model with stochastic task time which is subjected to normal distribution. The objectives of this model are to maximize line efficiency and to minimize the variation of work overload time. A Multi-objective Genetic Algorithm (MOGA) and an Ameliorative Structure of Multi-objective Genetic Algorithm (ASMOGA) with Priority-based Chromosome (PBC) are applied to solve this problem. At last, this research conducts an experimental simulation on a set of benchmark problems to verify the outperformance of the proposed algorithm.
