Roadway maintenance of the gob-side entry driving with narrow pillars is a key issue that needs to be considered in underground mining. In this paper, a thick-anchored dual-layer locking (TADL) supporting technique is proposed, in which, the flexible bolt and the cable are used as the main bodies of the foundation supporting layer and the deep reinforcement layer, respectively. In the instantaneous deformation stage after excavation, TADL is applied to form a thick-anchored bearing structure in time, aiming to achieve the following effects: (1) prevent the delamination of the roadway at the initial stage of excavation, (2) avoid the reduction of the bearing capacity of the surrounding rock structure, and (3) effectively control the plastic zone development and surrounding rock deformation of roof strata. Finally, the mining conditions of gob-side entry driving with narrow pillars are selected for engineering application and verification. The results of numerical simulation and field industrial tests show that the TADL support technique can effectively control the deformation of surrounding rock and drive away tensile stress. Compared with the original support scheme, the roof displacement of the roadway is significantly reduced by more than 16% after TADL support. This study provides a reference for the stability control of roadway roofs under similar conditions.

The gob-side roadway technology is widely used in coal mining in China, as it can improves the recovery rate of coal resources and alleviate the shortage of mining replacement. However, it is still difficult to control the stability of surrounding rock in gob-side roadway under deeper mining depth and more complex stress environment. With the auxiliary transportation roadway 30207 (ATR 30207) of the Muduchaideng Coal Mine as engineering background, this paper introduces the engineering case of gob-side roadway characterized by deep buried, long-distance, and large section. Through comprehensive methods, we studied and put forward the efficient excavation-support cooperation technology for surrounding rock of gob-side roadway. The research shows that the head-on cutting of gob-side roadway causes the stress to transfer and gradually adjust in the heading face to form a steady-state advanced abutment stress field. The coupling effect of the advanced abutment stress and the lateral abutment stress of goaf are the main reasons for repeated hole collapse of anchor cable drilling in solid coal rib, development and expansion of the roof separation fracture, and the low excavation speed. Supposed that upon the heading face support is completed, the next excavation-support cycle is quickly started during the early stage of stress adjustment, and the time effect can be fully utilized to alleviate the disturbance intensity of the advanced abutment stress in the heading face, as well as reduce the damage of rock mass. This paper puts forward the time-effect collaborative control countermeasures of the roof and rib in gob-side roadway of “hole collapse anchor cable of solid coal rib lag construction and waits for pressure relief and rapid excavation-support cycle in the heading face.” Engineering practice shows that the mine pressure behavior of roadway roof and solid coal rib is effectively controlled, and the roadway forming speed is greatly improved, which can provide reference for the efficient excavation-support design of gob-side roadway under the conditions of deep buried, long-distance, and large section in similar deep mines.

Background The role and extent of the effects of short-term behavioral factors on the risk of hemorrhagic stroke (HS) are unclear. This study aimed to assess and quantify behavioral trigger factors (BTFs) for HS and identify the differences in BTFs between Chinese and other populations. Methods A case-crossover study was performed from March 2021 to February 2022. New-onset HS patients were recruited from two university hospitals in China. The patients were interviewed to evaluate their exposure to 20 potential BTFs during the predefined risk and control periods and to estimate the odds ratios (ORs) and 95% confidence intervals (CIs). A comprehensive literature review was conducted to synthesize the evidence. Results A total of 284 patients with HS were included (150 with intracerebral hemorrhage and 134 with subarachnoid hemorrhage). Multivariate regression analysis showed that straining for defecation (OR: 3.06; 95% CI: 1.01–8.40), weightlifting (OR: 4.82; 95% CI: 1.02–22.83), overeating (OR: 4.33; 95% CI: 1.24–15.21), heavy physical exertion (OR: 3.02; 95% CI: 1.18–7.78), and chess/cards/mahjong games (OR: 2.51; 95% CI: 1.05–6.01) were associated with an increased risk within 2 hours before HS onset, and critical life events (OR: 3.81; 95% CI: 1.06–13.74) were associated with an increased risk 7 days before the onset of HS. Exposure to anger (OR: 3.17; 95% CI: 1.73–5.81) and heavy physical exertion (OR: 2.12; 95% CI: 1.65, 2.74) showed an increased risk of HS events after the pooled analysis. Conclusions Several behavioral activities and mood modifications are associated with the onset of HS. In addition to the common BTFs, Chinese patients have specific BTFs due to their habits and customs distinct from those of different populations in other regions. Background EGFR tyrosine kinase (TKIs) are recommend as the first-line treatment for non-small cell lung cancer (NSCLC) patients with EGFR mutation. However, some patients experience aggressive progression with a progression-free survival (PFS) less than 6 months on the first-line EGFR TKI therapy. Therefore, our study is to analyze the potential influencing factors including clinical features, biomarkers, concomitant mutations et al. Methods A total of 1073 NSCLC patients with EGFR mutation in a multi-center study from January 2019 to December 2021. The datum pathological and molecular characteristics were collected. The area under the receiver operating characteristic (ROC) curve was used to evaluate the predictive effect of Ki-67 on the first-line TKI. The curve of PFS was conducted by Kaplan–Meier method and tested by bilateral log-rank. Cox regression model was used to predict and evaluate PFS of different variables. Chi-square or Fisher analysis was used for correlation between groups. Results 55 patients who show aggressive progression (PFS ≤ 6 months) on the first-line TKI therapy were analyzed in this study, while 71 with slow progression (PFS > 6 months). Concomitant mutations including AXIN2, P2CG and RAD51C mutations occurred only in the aggressively progressive group (P = 0.029). Correlation between Ki-67 index and the aggressive progression of the first-line TKI therapy was significant statistically different (P Conclusion NSCLC harbored EGFR and concomitant mutations (such as AXIN2, PLCG2 and RAD51C), and/or Ki-67 high expression may indicate the aggressive progression to the first-line EGFR-TKI.

Aims Phosphorus (P)-binding colloids (1–450 nm) have been identified as highly mobile and bioavailable P in terrestrial ecosystems. However, the bioavailability of varied colloidal P (Pcoll) sizes remains unclear, especially in situ soils. The purpose of this study was to evaluate the differential contribution of various-sized colloids to plant available P in the rhizosphere and the phosphatase response patterns. Methods In the 45-day rice rhizotron experiment, we applied different size fractioned biogas slurry (BS) colloids with the same total P concentration, including nano-sized colloids (1–20 nm), fine-sized colloids (20–220 nm), and medium-sized colloids (220–450 nm). Colloidal P organic and inorganic fractions, colloidal minerals were investigated in corresponding treatments. Soil zymography was performed during the cultivation period, to examine the processes of enzymic hydrolysis in temporal and spatial dynamics. The biomass and P content of roots and shoots were also recorded. Results The relative bioavailability of colloidal P (RBAcoll) was the highest (64%) for nano-sized BS colloids addition. The phosphatase activities and hotspot areas were found to be significantly 1) correlated with RBAcoll, 2) increased by the colloid-free (truly dissolved P) and nano-sized BS fractions, and 3) affected by the plant growth stage. Conclusion We found that the addition of nano-sized BS colloids increased P uptake by plants and improved the Pcoll bioavailability in the rhizosphere, in which the phosphatase-catalyzed hydrolysis of organic Pcoll played an important role. Nano-sized BS-derived colloids may be an effective substitute for the use of liquid phosphorus fertilizer.

Background: Diabetes nephropathy (DN) is a growing public health concern worldwide. Renal dysfunction impairment in DN is intimately linked to ER stress and its related signaling pathways. Nonetheless, the underlying mechanism and biomarkers for this function of ER stress in the DN remain unknown. Methods: Microarray datasets were retrieved from the Gene Expression Omnibus (GEO) database, and ER stress-related genes (ERSRGs) were downloaded from the MSigDB and GeneCards database. We identified hub ERSRGs for DN progression by intersecting ERSRGs with differentially expressed genes and significant genes in WGCNA, followed by a functional analysis. After analyzing hub ERSRGs with three machine learning techniques and taking the intersection, we did external validation as well as developed a DN diagnostic model based on the characteristic genes. Immune infiltration was performed using CIBERSORT. Moreover, patients with DN were then categorized using a consensus clustering approach. Eventually, the candidate ERSRGs-specific small-molecule compounds were defined by CMap. Results: Several biological pathways driving pathological injury of DN and disordered levels of immune infiltration were revealed in the DN microarray datasets and strongly related to deregulated ERSRGs by bioinformatics multi-chip integration. Moreover, CDKN1B, EGR1, FKBP5, GDF15, and MARCKS were identified as ER stress signature genes associated with DN by machine learning algorithms, demonstrating their potential as DN biomarkers. Conclusions: Our research sheds fresh light on the function of ER stress in DN pathophysiology and the development of early diagnostic and ER stress-related treatment targets in patients with DN.

BackgroundThe intestinal tract serves as a critical regulator for nutrient absorption and overall health. However, its involvement in anti-parasitic infection and immunity has been largely neglected, especially when a parasite is not transmitted orally. The present study investigated the colonic histopathology and functional reprogramming in mice with intraperitoneal infection of the larval Echinococcus granulosus (E. granulosus).ResultsCompared with the control group, the E. granulosus–infected mice exhibited deteriorated secreted mucus, shortened length, decreased expression of tight junction proteins zonula occludens-1 (ZO-1), and occludin in the colon. Moreover, RNA sequencing was employed to characterize colonic gene expression after infection. In total, 3,019 differentially expressed genes (1,346 upregulated and 1,673 downregulated genes) were identified in the colon of infected mice. KEGG pathway and GO enrichment analysis revealed that differentially expressed genes involved in intestinal immune responses, infectious disease-associated pathways, metabolism, or focal adhesion were significantly enriched. Among these, 18 tight junction-relative genes, 44 immune response-associated genes, and 23 metabolic genes were annotated. Furthermore, mebendazole treatment could reverse the colonic histopathology induced by E. granulosus infection.ConclusionsIntraperitoneal infection with E. granulosus induced the pathological changes and functional reprogramming in the colon of mice, and mebendazole administration alleviated above alternations, highlighting the significance of the colon as a protective barrier against parasitic infection. The findings provide a novel perspective on host-parasite interplay and propose intestine as a possible target for treating parasitic diseases that are not transmitted orally.

As a naturally occurring cytolytic peptide, melittin (Mel) has strong cytolytic activity and is a potent therapeutic peptide for cancer therapy. However, the serious hemolytic activity of Mel largely impedes its clinical applications. In this work, based on the strong interactions between proteins/peptides and polyphenols, we develop a tannic acid–Fe 3+ metal–phenolic network (MPN)-based strategy that can convert Mel from foe to friend via shielding its positive charges and reducing its hemolytic activity. Besides, an immune adjuvant resiquimod (R848) is also introduced for immunostimulation, affording the final Mel- and R848-coloaded nanodrug. The Mel-caused membrane disruption can induce immunogenic cell death for immunostimulation, R848 can act as an immune adjuvant to further facilitate the immunostimulatory effect, and the tannic acid–Fe 3+ MPN-mediated Fenton reaction can produce reactive oxygen species for cancer treatment. Further experiments reveal that the nanodrug can effectively cause immunogenic cell death of tumor cells and arouse robust intratumoral and systemic antitumor immunostimulation. In the bilateral tumor-bearing mouse models, the nanodrug considerably destroys the primary tumor and also boosts the abscopal effect to ablate the distant tumor. Collectively, the MPN-facilitated “foe-to-friend” strategy may promote the practical applications of Mel and foster the development of cancer immunotherapeutics.

In this paper, the whole genome of the multidrug-resistant Aeromonas hydrophila MX16A was comprehensively analyzed and compared after sequencing by PacBio RS II. To shed light on the drug resistance mechanism of A. hydrophila MX16A, a Kirby-Bauer disk diffusion method was used to assess the phenotypic drug susceptibility. Importantly, resistance against β-lactam, sulfonamides, rifamycins, macrolides, tetracyclines and chloramphenicols was largely consistent with the prediction analysis results of drug resistance genes in the CARD database. The varied types of resistance genes identified from A. hydrophila MX16A revealed multiple resistance mechanisms, including enzyme inactivation, gene mutation and active effusion. The publicly available complete genomes of 35 Aeromonas hydrophila strains on NCBI, including MX16A, were downloaded for genomic comparison and analysis. The analysis of 33 genomes with ANI greater than 95% showed that the pan-genome consisted of 9556 genes, and the core genes converged to 3485 genes. In summary, the obtained results showed that A. hydrophila exhibited a great genomic diversity as well as diverse metabolic function and it is believed that frequent exchanges between strains lead to the horizontal transfer of drug resistance genes.

This paper studies the influence of trace elements Ag, Bi and Ni on the solid-liquid electromigration (S-L EM) interface diffusion behavior in solder joints. The solder joint Sn0.7Ag0.5Cu (SAC0705), Sn3.0Ag0.5Cu (SAC305) and Sn0.7Ag0.5Cu3.5Bi0.05Ni (SAC0705+BiNi) are taken as the research object. The interfacial intermetallic compound (IMC) growth and evolution behavior were studied after S-L EM. Meanwhile, the influences of the trace elements Ag, Bi and Ni on interface diffusion were analyzed in the solder joint during S-L EM. The results show that the interfacial IMC grains can be refined by adding trace elements Ag, Bi and Ni in solder during reflowing. The addition of Bi and Ni changed the composition and shape of the interfacial IMC. After adding Bi and Ni to the SAC0705 solder alloy, the interfacial IMC changed from polygonal cylindrical Cu6Sn5 to ellipsoidal (Cu,Ni)6Sn5. The addition of Ag, Bi and Ni inhibited the growth of the cathodic interfacial IMC and improved the resistance to the S-L EM performance of the solder joints. Compared with Cu/SAC0705/Cu solder joints, the cathodic interfacial IMC grain size of Cu/SAC0705+BiNi/Cu solder joint decreased by 87%, and the thickness of the cathodic Cu substrate decreased by 29% after S-L EM.

Non-invasive and cost-effective diagnosis of gastric cancer is essential to improve outcomes. Aim of the study was to establish a neural network model based on patient demographic data and serum biomarker panels to aid gastric cancer diagnosis. A total of 295 patients hospitalized in Nanjing Drum Tower hospital diagnosed with gastric cancer based on tissue biopsy, and 423 healthy volunteers were included in the study. Demographical information and tumor biomarkers were obtained from Hospital Information System (HIS) as original data. Pearson's correlation analysis was applied on 574 individuals’ data (training set, 229 patients and 345 healthy volunteers) to analyze the relationship between each variable and the final diagnostic result. And independent sample t test was used to detect the differences of the variables. Finally, a neural network model based on 14 relevant variables was constructed. The model was tested on the validation set (144 individuals including 66 patients and 78 healthy volunteers). The predictive ability of the proposed model was compared with other common machine learning models including logistic regression and random forest. Tumor markers contributing significantly to gastric cancer screening included CA199, CA125, AFP, and CA242 were identified, which might be considered as important inspection items for gastric cancer screening. The accuracy of the model on validation set was 86.8% and the F1-score was 85.0%, which were better than the performance of other models under the same condition. A non-invasive and low-cost artificial neural network model was developed and proved to be a valuable tool to assist gastric cancer diagnosis.

Developing a universal nanoplatform for efficient delivery of various drugs to target sites is urgent for overcoming various biological barriers and realizing combinational cancer treatment. Nanogels, with the advantages of both hydrogels and nanoparticles, may hold potential for addressing the above issue. Here, a dual-responsive nanogel platform (HPC nanogel) is constructed using β-cyclodextrin-conjugated hyaluronic acid (HA-βCD), polyethyleneimine (PEI), and cisplatin. HA-βCD and PEI compose the skeleton of the nanogel, and cisplatin molecules provide the junctions inside the skeleton, thus affording a multiple interactions-based nanogel. Besides, HA endows the nanogel with hyaluronidase (HAase)-responsiveness, and cisplatin guarantees the glutathione (GSH)-responsive ability, which make the nanogel a dual-responsive platform that can degrade and release the loaded drugs when encountering HAase or GSH. Additionally, the HPC nanogel possesses excellent small-molecule drug and protein loading and intracellular delivery capabilities. Especially, for proteins, their intracellular delivery via nanogels is not hindered by serum proteins, and the enzymes delivered into cells still maintain their catalytic activities. Furthermore, the nanogel can codeliver different cargoes to achieve "cocktail" chemotherapeutic efficacy and realize combination cancer therapy. Overall, the HPC nanogel can serve as a multifunctional platform capable of delivering desired drugs to treat cancer or other diseases.

The H9N2 avian influenza virus (AIV) is currently widespread worldwide, posing a severe threat to the poultry industry and public health. Reassortment is an important way for influenza viruses to adapt to a new host. In 2007, the PB2 gene of H9N2 AIV in China was reassorted, and the DK1-like lineage replaced the F/98-like lineage, forming a dominant genotype of G57. This genotype and its reassortants (such as H7N9, H10N8 and H5N6) showed higher mammalian adaptation, and caused increased human infections. However, the adaptive mechanisms of the DK1-like lineage PB2 gene remain unclear. Here, we confirmed that the PB2 lineage of the H9N2 AIV currently prevalent in China still belongs to the DK1-like lineage and, compared with the previously predominant F/98-like lineage, the DK1-like lineage PB2 gene significantly enhances H9N2 AIV to mammalian adaptation. Through transcriptomic analysis and qRT–PCR and western blot experiments, we identified a host factor, sphingosine kinase 1 (SphK1), that is closely related to viral replication. SphK1 inhibits the replication of DK1-like PB2 gene H9N2 AIV, but the ability of SphK1 protein to bind DK1-like PB2 protein is weaker than that of F/98-like PB2 protein, which may contribute to H9N2 AIV containing the DK1-like PB2 gene to escape the inhibitory effect of host factor SphK1 for efficient infection. This study broadens our understanding of the adaptive evolution of H9N2 AIV and highlights the necessity to pay close attention to the AIV that contains the adaptive PB2 protein in animals and humans.

Most of insertion/deletion polymorphisms are diallelic molecular markers characterized as small amplicon sizes, high inter-population diversities, and low mutation rates, which make them the promising genetic markers in biogeographic ancestor inference field. The developmental validations of a 39 ancestry informative marker-insertion/deletion (AIM-InDel) panel and the genetic polymorphic investigations of this panel were performed in the Shaanxi Han population of China. The developmental validation included the optimizations of PCR-related indicators, repeatability, reproducibility, precision, accuracy, sensitivity, species specificity, stability of the panel, and the abilities in analyzing degraded, casework, and mixture samples, and the present results demonstrated that this 39 AIM-InDel panel was robust, sensitive, and accurate. For the population diversity analyses, the combined discrimination power value of 38 AIM-InDel loci except for rs36038238 locus was 0.999999999931257, indicating that this novel panel was highly polymorphic, biogeographic informative, and could be also used in individual identifications in the Shaanxi Han population.

The primary premise of autonomous railway inspection using unmanned aerial vehicles is achieving autonomous flight along the railway. In our previous work, fitted centerline-based unmanned aerial vehicle (UAV) navigation is proven to be an effective method to guide UAV autonomous flying. However, the empirical parameters utilized in the fitting procedure lacked a theoretical basis and the fitted curves were also not coherent nor smooth. To address these problems, this paper proposes a skeleton detection method, called the dynamic-weight parallel instance and skeleton network, to directly extract the centerlines that can be viewed as skeletons. This multi-task branch network for skeleton detection and instance segmentation can be trained end to end. Our method reformulates a fused loss function with dynamic weights to control the dominant branch. During training, the sum of the weights always remains constant and the branch with a higher weight changes from instance to skeleton gradually. Experiments show that our model yields 93.98% mean average precision (mAP) for instance segmentation, a 51.9% F-measure score (F-score) for skeleton detection, and 60.32% weighted mean metrics for the entire network based on our own railway skeleton and instance dataset which comprises 3235 labeled overhead-view images taken in various environments. Our method can achieve more accurate railway skeletons and is useful to guide the autonomous flight of a UAV in railway inspection.

Background: Hemorrhagic stroke (HS) is a sudden-onset disease with high mortality and disability rates, and it is crucial to explore the triggers of HS. In this study, we analyzed individual triggers for HS to provide a basis for HS prevention and intervention. Methods: A systematic search of five databases was conducted until December 2022. Studies on HS-related individual triggers conducted using a case-crossover study or self-controlled case series design were included in the descriptive summary and comprehensive evidence synthesis of each trigger. Results: A total of 39 studies were included after the screening, and 32 trigger factor categories were explored for associations. Potential trigger factors for HS were as follows: Antiplatelet (odd ratio (OR), 1.10; 95% confidence interval (CI), 1.00–1.21) and anticoagulant (OR, 5.43; 95% CI, 2.04–14.46) medications, mood stabilizers/antipsychotics (OR, 1.33; 95% CI, 1.07–1.65), infections (OR, 2.15; 95% CI, 1.73–2.67), vaccinations (relative risk, 1.11; 95% CI, 1.02–1.21), physical exertion (OR, 2.08; 95% CI, 1.58–2.74), cola consumption (OR, 5.45; 95% CI, 2.76–10.76), sexual activity (OR, 7.49; 95% CI, 2.23–25.22), nose blowing (OR range, 2.40–56.40), defecation (OR, 16.94; 95% CI, 3.40–84.37), and anger (OR, 3.59; 95% CI, 1.56–8.26). No associations were observed with illicit drug use (OR, 2.05; 95% CI, 0.52–8.06) or cigarette smoking (OR, 0.81; 95% CI, 0.52–1.24) and HS. Conclusions: Individual triggers, including several medications, infections, vaccinations, and behaviors, may trigger HS onset. Direct control measures for behavioral triggers can play a crucial role in preventing HS. High-risk populations should receive personalized therapies and monitoring measures during the medication treatment to balance the risk of acute HS and the basic diseases.

The management of post-liver resection recurrence is often the life-limiting factor in HCC treatment. While much has been published on intrahepatic recurrence and lung metastasis, there is a relative lack of data on intraabdominal extrahepatic metastasis (EHM). We sought to evaluate the outcomes of patients post-resection of intraabdominal EHM and assess preoperative factors predictive of early recurrence post-metastasectomy. We performed a retrospective review of 25 consecutive patients who underwent metastasectomy for intraabdominal EHM from 2003 to 2016 at our institution. Of the 25 cases of EHM, 16 were in the peritoneum, 3 in the adrenal glands, 3 in the large bowel, 1 in the spleen, 1 in the pancreas and 1 in the omentum. Median overall survival was 27 months (IQR 15–89 months). Twenty-one patients (84%) developed recurrence post-metastasectomy of EHM of which 12 patients experienced early recurrence within 12 months. The median time to recurrence post-metastasectomy was 11(IQR 15.5) months. Multivariate analysis demonstrated both hepatitis B (11 (91.6%) versus 4 (44.4%), p = 0.00) status and high tumour grade (8 (66.6%) versus 3 (25%), p = 0.004) to be significant independent predictors of early recurrence. Patients who experienced early recurrence had a significantly shorter median overall survival (18 months (95% CI 12.9–23.0)) compared to those who did not (89 months (95% CI 24.8–153.1), p = 0.004). Patients with EHM who underwent metastasectomy had a median overall survival of 27 months. Hepatitis B positivity and high primary tumour grade were preoperative predictors of futile surgery. All 7 patients who had both hepatitis B and high tumour grade experienced early recurrence post-metastasectomy.

A neurotoxin is essentially a protein that mainly acts on the nervous system; it has a selective toxic effect on the central nervous system and neuromuscular nodes, can cause muscle paralysis and respiratory paralysis, and has strong lethality. According to their principle of action, neurotoxins are divided into presynaptic neurotoxins and postsynaptic neurotoxins. Correctly identifying presynaptic and postsynaptic nerve toxins provides important clues for future drug development and the discovery of drug targets. Therefore, a predictive model, Neu_LR, was constructed in this paper. The monoMonokGap method was used to extract the frequency characteristics of presynaptic and postsynaptic neurotoxin sequences and carry out feature selection, then, based on the important features obtained after dimensionality reduction, the prediction model Neu_LR was constructed using a logistic regression algorithm, and ten-fold cross-validation and independent test set validation were used. The final accuracy rates were 99.6078 and 94.1176%, respectively, which proved that the Neu_LR model had good predictive performance and robustness, and could meet the prediction requirements of presynaptic and postsynaptic neurotoxins. BackgroundWe previously reported that the larval Echinococcus granulosus (E. granulosus) infection can expand the population of regulatory B cells in mice, thereby inhibiting the anti-infective immunity. However, the underlying mechanism is still largely unknown. This study further investigated the holistic transcriptomic profiles of total splenic B cells following the chronic infection of the parasite.MethodsThe infection model of larval E. granulosus was established by intraperitoneal inoculation with 2000 protoscolexes. Magnetic-Activated Cell Separation (MACS) was used to isolate the total splenic B cells. RNA sequencing was performed to screen the differentially expressed genes (DEGs) after infection. The expression of selected DEGs was verified using qRT-PCR. Gene Ontology (GO) analysis, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, and Co-expression network analysis were applied to predict these DEGs' underlying biological processes, pathways, and interactions respectively.ResultsA total of 413 DEGs were identified in larval E. granulosus infected B cells, including 303 up- and 110 down-regulated genes. Notably, most DEGs related to inflammation and chemotaxis were significantly upregulated after infection. In line with these changes, significant expression upregulation of DEGs associated with fatty acid oxidation, lipid synthesis, lipolysis, lipid transport, and cholesterol biosynthesis, were observed in infected B cells. Co-expression network analysis showed an intimate interaction between these DEGs associated with immune and metabolism.ConclusionsThe present study revealed that the larval E. granulosus infection induces metabolic reprogramming of B cells, which provides a novel clue to clarify the immunoregulatory mechanism of B cells in parasitic infection.

Obtaining portal vein access is a challenging step in TIPS creation, particularly in patients with portal vein thrombosis (PVT). We report the use of an intrahepatic arterial targeting wire to provide a target for portal vein puncture in 20 patients with cirrhosis and complete obliterative PVT who underwent TIPS placement. Technical success rate of TIPS placement was 90% (18/20). The median number of punctures was three. No severe complication occurred.

Purpose: To determine the therapeutic effects of kidney yang-tonifying, blood-activating and phlegmeliminating methods on steroid-induced osteonecrosis of femoral head (SONFH), and their mechanisms of action.Methods: A rabbit model of SONFH was established. Six rabbit groups were used: control, SONFH (given normal saline); tongluo shenggu group, kidney-tonifying group, blood-activating group, and phlegm--eliminating group, which received tongluo shenggu capsule (0.28 g/kg/day), yougui yin (YGY, 2.52 g/kg/day), taohong siwu decoction (TSD, 2.38 g/kg/day) and linggui zhugan decoction (LZD,1.54 g/kg/day), respectively, for 8 weeks. The following serum indices were assayed on the 2nd, 4th and 8th weeks: total cholesterol, triglycerides, thromboxane B2 (TXB2), 6-keto-PGF1α, osteoprotegerin (OPG), and receptor activator of nuclear factor kappa-Β ligand (RANKL). Compound marker consists of two different types of genetic markers, like deletion/insertion polymorphism and single nucleotide polymorphism in the genomic region of 200 bp, and microhaplotype consists of a series of closely linked single nucleotide polymorphisms in a small DNA segment (300 bp), which show great potential for human identifications and mixture analyses. In this study, we initially selected 23 novel genetic markers comprising 10 microhaplotypes and 13 compound markers according to previously reported single nucleotide polymorphism or deletion/insertion polymorphism loci. Genetic distributions of these 23 loci in different continental populations showed that they could be used as valuable loci for forensic human identification purpose. Besides, high informativeness values (0.1) were observed in six loci which could be further employed for forensic ancestry analyses. Finally, 18 loci were successfully developed into a multiplex panel and detected by the next generation sequencing (NGS) technology. Further analyses of these 18 loci in the studied Shaanxi Han population showed that 15 loci exhibited relatively high expected heterozygosities (0.5). Cumulative power of discrimination (0.999 999 999 99 4835) of these 18 loci revealed that the multiplex panel could also be utilized for human identifications in the studied Shaanxi Han population.

The peroxynitrite ion (ONOO- ) has important roles in many biological processes. We have developed a multicolor ONOO- -sensing probe (SiONNOH) that undergoes deprotonation and desilylation processes, which result in several changes in the emission wavelengths. In response to different concentrations of ONOO- , the probe exhibits fluorescence changes from pink (595 nm at 2 eq. ONOO- ) to green (540 nm at 6 eq. ONOO- ) via orange (3 eq. ONOO- ) and yellow (4 eq. ONOO- ) under physiological conditions until no fluorescence signal is observed after ONOO- is completely eliminated by lipoic acid. The probe shows the high selectivity for ONOO- and the limit of detection is calculated to be 1.27 μM. Moreover, the probe shows the capacity to monitor the concentration ranges of ONOO- through multicolor fluorescence in living cells, which will greatly facilitate the rapid detection of ONOO- concentration ranges by the naked eye under a UV light without any precision instrumentation.

In this study, a small set of ancestry informative SNPs was selected to differentiate African, European, East and South Asian samples, which was detected by the next-generation sequencing technology. A total of 127 Chinese Shaanxi Han individuals were collected as test samples. No statistically significant linkage disequilibrium of any pair of loci or departure from Hardy-Weinberg equilibrium of each locus was observed in the test population. To evaluate the performance of ancestry assignment using this panel, admixture analysis, principal component analysis, and likelihood ratio calculations were conducted based on the 1000 genome data and test samples. All populations were clustered into four groups, African, European, South and East Asian populations, which were consistent with their geographical origins. The pairwise fixation index (FST ) between populations from different continental groups ranged from 0.140 to 0.621 with average 0.415, and the pairwise FST between populations from the same continent ranged from 0.000 to 0.056 with average 0.012. The likelihood ratio results of 125 test individuals indicated that their ancestry components were highly possible from East Asia. In conclusion, this small set of ancestry informative SNPs can be used as a reliable tool to identify and quantify ancestry components of unknown samples.

Introduction. Fine-needle aspiration (FNA) biopsies are fundamental procedures in the diagnoses of thyroid tumours. Reports of needle tract tumour seeding, however, question its practice regarding patient safety and potentially jeopardizing its widespread usage. Case Report. We describe a case of a 50-year-old lady with known multinodular goitre, and previous fine-needle aspiration (FNA) biopsies of her thyroid nodules in 2010, who developed palpable right neck nodules 8 years after the initial FNA. Imaging and histological biopsies revealed suspicious right sternocleidomastoid (SCM) nodules that are likely needle tract tumour deposits. She underwent a total thyroidectomy with central compartment clearance and excision of the right SCM nodules and received radioactive iodine therapy thereafter. Discussion. Contrary to other forms of malignancies, needle tract seeding is an uncommon occurrence for thyroid cancers. Nevertheless, there is speculation regarding its potential in cutaneous spread of malignancy with studies investigating its optimal techniques and application. Conclusion. While FNA remains an indisputable tool in the management of thyroid tumours, precautions must be taken to safeguard patient safety and improve patient outcomes.

Predicting the biogeographical ancestries of populations and unknown individuals based on ancestry-informative markers (AIMs) has been widely applied in providing DNA clues to criminal investigations, correcting the factor of population stratification in genome-wide association studies (GWAS), and working as the basis of predicting the externally visible characteristics (EVCs) of individuals. The present study chose Chinese Xinjiang Kazak (XJK) group as research object using a 165 AIM-SNPs panel via next generation sequencing (NGS) technology to reveal its ancestral information and genetic background by referencing the populations’ data from 1000 Genomes Phase 3. After the Bonferroni correction, there were no significant deviations at the 165 AIM-SNP loci except two loci with homozygote in the studied XJK group. Ancestry information inference and populations genetic analyses were conducted basing on multiplex statistical methods such as forensic statistical parameter analyses, estimation of the success ratios with cross-validation, population tree, principal component analysis (PCA), and genetic structure analysis. The present results revealed that XJK group had the admixed ancestral components of East Asian and European populations with the ratio of about 62:37.
