Accurate and early diagnosis of bean common mosaic virus (BCMV) in Phaseolus vulgaris tissues is critical since the pathogen can spread easily and have long-term detrimental effects on bean production. The use of resistant varieties is a key factor in the management activities of BCMV. The study reported here describes the development and application of a novel SYBR Green-based quantitative real-time PCR (qRT-PCR) assay targeting the coat protein gene to determine the host sensitivity to the specific NL-4 strain of BCMV. The technique showed high specificity, validated by melting curve analysis, without cross-reaction. Further, the symptoms development of twenty advanced common bean genotypes after mechanical BCMV-NL-4 infection was evaluated and compared. The results showed that common bean genotypes exhibit varying levels of host susceptibility to this BCMV strain. The YLV-14 and BRS-22 genotypes were determined as the most resistant and susceptible genotypes, respectively, in terms of aggressiveness of symptoms. The accumulation of BCMV was analyzed in the resistant and susceptible genotypes 3, 6, and 9 days following the inoculation by the newly developed qRT-PCR. The mean cycle threshold (Ct) values showed that the viral titer was significantly lower in YLV-14, which was evident in both root and leaf 3 days after the inoculation. The qRT-PCR thus facilitated an accurate, specific, and feasible assessment of BCMV accumulation in bean tissues even in low virus titers, allowing novel clues in selecting resistant genotypes in the early stages of infection, which is critical for disease management. To the best of our knowledge, this is the first study of a successfully performed qRT-PCR to estimate BCMV quantification.

The adzuki bean

Silicon (Si), despite being abundant in nature, is still not considered a necessary element for plants. Si supplementation in plants has been extensively studied over the last two decades, and the role of Si in alleviating biotic and abiotic stress has been well documented. Owing to the noncorrosive nature and sustainability of elemental Si, Si fertilization in agricultural practices has gained more attention. In this review, we provide an overview of different smart fertilizer types, application of Si fertilizers in agriculture, availability of Si fertilizers, and experiments conducted in greenhouses, growth chambers, and open fields. We also discuss the prospects of promoting Si as a smart fertilizer among farmers and the research community for sustainable agriculture and yield improvement. Literature review and empirical studies have suggested that the application of Si-based fertilizers is expected to increase in the future. With the potential of nanotechnology, new nanoSi (NSi) fertilizer applications may further increase the use and efficiency of Si fertilizers. However, the general awareness and scientific investigation of NSi need to be thoughtfully considered. Thus, we believe this review can provide insight for further research into Si fertilizers as well as promote Si as a smart fertilizer for sustainability and crop improvement.

The conventional plant breeding evaluation of large sets of plant phenotypes with precision and speed is very challenging. Thus, consistent, automated, multifaceted, and high-throughput phenotyping (HTP) technologies are becoming increasingly significant as tools to aid conventional breeding programs to develop genetically improved crops. With rapid technological advancement, various vegetation indices (VIs) have been developed. These VI-based imaging approaches, linked with artificial intelligence and a variety of remote sensing applications, provide high-throughput evaluations, particularly in the field of precision agriculture. VIs can be used to analyze and predict different quantitative and qualitative aspects of vegetation. Here, we provide an overview of the various VIs used in agricultural research, focusing on those that are often employed for crop or vegetation evaluation, because that has a linear relationship to crop output, which is frequently utilized in crop chlorophyll, health, moisture, and production predictions. In addition, the following aspects are here described: the importance of VIs in crop research and precision agriculture, their utilization in HTP, recent photogrammetry technology, mapping, and geographic information system software integrated with unmanned aerial vehicles and its key features. Finally, we discuss the challenges and future perspectives of HTP technologies and propose approaches for the development of new tools to assess plants’ agronomic traits and data-driven HTP resolutions for precision breeding.

The N-terminal domain of the Pseudomonas sp. FB15 phytase increases low-temperature activity and catalytic efficiency. In this study, the 3D structure of the N-terminal domain was predicted and substitutions for the amino acid residues of the region assumed to be the active site were made. The activity of mutants, in which alanine (A) was substituted for the original residue, was investigated at various temperatures and pH values. Significant differences in enzymatic activity were observed only in mutant E263A, suggesting that the amino acid residue at position 263 of the N-terminal domain is important in enzyme activity.

Silicon (Si) is absorbed and accumulated by some plant species; it has been shown to improve plant growth and performance. The beneficial role of Si in plants is based on the fundamental assumptions, and the biological function of Si is still being researched due to its complex nature, distinctiveness, and interaction. The present study included two distinct experiment sets: a screening test and an advanced test. In the initial examination, we used 21 soybean (Glycine max L.) cultivars. Following the evaluation, we chose four cultivars to investigate further. In particular, the positive response cultivars, Taeseon and Geomjeongsaeol, showed a 14% increase in net photosynthesis (PN), and a 19–26% increase in transpiration in Si-treated plants when compared to the control plants. Si-treated Taeseon, Geomjeongsaeol, and Somyongkong, Mallikong cultivars showed significant differences in root morphological traits (RMTs) and root system architecture (RSA) when compared to the control plants. Taeseon and Geomjeongsaeol showed a 26 and 46% increase in total root length (TRL) after Si application, respectively, compared to the control, whereas Mallikong and Somyongkong showed 26 and 20% decrease in TRL after Si treatment, respectively, compared to the control. The Si application enhanced the overall RMTs and RSA traits in Taeseon and Geomjeongsaeol; however, the other two cultivars, Somyongkong and Mallikong, showed a decrease in such RMTs and RATs. Furthermore, to understand the underlying molecular mechanism and the response of various cultivars, we measured the Si content and analyzed the gene expression of genes involved in auxin transport and root formation and development. We showed that the Si content significantly increased in the Si-treated Somyongkong (28%) and Taeseon (30%) compared to the control cultivars. Overall, our results suggested that Si affects root development as well as the genes involved in the auxin synthesis, transport pathway, and modulates root growth leading to cultivar-dependent variation in soybeans.

Corn oil was heated using an infrared cooker, an air fryer, and a cooking oven at similar temperatures, and oxidative stability and physicochemical properties including moisture content, temperature change, the profile of headspace volatiles, formaldehyde and acetaldehyde of the heated oils were compared. Corn oil heated using the air fryer showed the lowest degree of oxidation, followed by that heated using the infrared cooker and the cooking oven. However, the content of headspace volatiles in 120 min heated oil using the infrared cooker was higher by 2.57 and 5.37 times than that in oil heated using the cooking oven and the air fryer, respectively. The profiles of formaldehyde and acetaldehyde in oils showed patterns inconsistent with those of headspace volatile and oxidation parameters. Generally, the air fryer-treated oil underwent slow lipid oxidation, whereas oil from the infrared cooker had more volatiles and imparted odor to foods.

The adzuki bean Vigna angularis (Wild.) is an important leguminous crop cultivated mainly for food purposes in Asian countries; it represents a source of carbohydrates, digestible proteins, minerals, and vitamins. Aquaporins (AQPs) are crucial membrane proteins involved in the transmembrane diffusion of water and small solutes in all living organisms, including plants. In this study, we used the whole genome sequence of the adzuki bean for in silico analysis to comprehensively identify 40 Vigna angularis aquaporin (VaAQP) genes and reveal how these plants react to drought stress. VaAQPs were compared with AQPs from other closely-related leguminous plants, and the results showed that mustard (Brassica rapa) (59), barrel medic (Medicago truncatula) (46), soybean (Glycine max) (66), and common bean (Phaseolus vulgaris L.) (41) had more AQP genes. Phylogenetic analysis revealed that forty VaAQPs belong to five subfamilies, with the VaPIPs (fifteen) subfamily the largest, followed by the VaNIPs (ten), VaTIPs (ten), VaSIPs (three), and VaXIPs (two) subfamilies. Furthermore, all AQP subcellular locations were found at the plasma membrane, and intron–exon analysis revealed a relationship between the intron number and gene expression, duplication, evolution, and diversity. Among the six motifs identified, motifs one, two, five, and six were prevalent in VaTIP, VaNIP, VaPIP, and VaXIP, while motifs one, three, and four were not observed in VaPIP1-3 and VaPIP1-4. Under drought stress, two of the VaAQPs (VaPIP2-1 and VaPIP2-5) showed significantly higher expression in the root tissue while the other two genes (VaPIP1-1 and VaPIP1-7) displayed variable expression in leaf tissue. This finding revealed that the selected VaAQPs might have unique molecular functions linked with the uptake of water under drought stress or in the exertion of osmoregulation to transport particular substrates rather than water to protect plants from drought. This study presents the first thorough investigation of VaAQPs in adzuki beans, and it reveals the transport mechanisms and related physiological processes that may be utilized for the development of drought-tolerant adzuki bean cultivars.

Bentazone is a broadleaf post-emergence herbicide widely used for crop production that inhibits photosynthetic activity, resulting in phytotoxicity and injury in plants. Evaluating and identifying herbicide-tolerant genotypes is a critical step in plant breeding programs. In this study, we determined the reaction of 138 Korean soybean cultivars to bentazone using visual evaluation, and selected cultivars were further evaluated to determine the effects of bentazone on physiological parameters. For physiological parameters, we measured the normalized difference vegetation index (NDVI) from hyperspectral reflectance images. From 2 to 4 DAT, the NDVI for two sensitive cultivars was between 0.60 and 0.69, while the NDVI for tolerant cultivars was between 0.70 and 0.86. Photosynthesis rate (A), transpiration (E), stomatal conductance (gsw), and total conductance of CO2 (gtc) were measured using chlorophyll fluorescence. Visual score evaluation showed that moderate bentazone-tolerant cultivars were predominant among the Korean cultivars. For physiological measurements, differences in NDVI were detected between bentazone-tolerant and -sensitive cultivars 2 days after treatment (DAT). However, the A, E, gsw, and gtc levels dramatically decreased 1 DAT in the sensitive cultivars. This study provides insights into the tolerance and sensitivity of soybeans to bentazone.

Drought is a major threat worldwide for crop production, especially due to the rapid climate changes. Current drought solutions involve improving irrigation system, rainwater harvesting, damming, cloud seeding, and changes of cultivation methods. Despite effective, each solution has economic, environmental, and temporal drawbacks. Among all solutions, the most effective, inexpensive and manageable method is the use of drought-tolerant cultivars via plant breeding. However, conventional plant breeding is a time-consuming and laborious task, especially for phenotypic data acquisition of target traits of numerous progenies. High-throughput phenotyping (HTP) is a recently developed method and has potential to overcome the mentioned issues. HTP offers massive, accurate, rapid, and automatic data acquisition in the breeding procedure and can be a breakthrough for developing drought resistant/tolerant cultivars. This study introduces various methods of HTP to detect drought stress, which can accelerate the breeding processes of drought-tolerant cultivars to provide helpful guidelines for breeders and researchers to choose appropriate methods.

To assess the utility of three-dimensional double-echo steady-state with water excitation (3D-DESS-WE) imaging for localizing deep-seated parotid tumors in relation to the facial nerve. A prospective study comparing the surgical outcomes of parotidectomy with or without 3D-DESS-WE sequence is currently enrolling the patients. Magnetic resonance imaging data from the first 25 patients with 3D-DESS-WE sequence were reviewed. Visibility of the intraparotid facial nerve was independently assessed by two neuroradiologists. The diagnostic performance of the 3D-DESS-WE sequence for prediction of deep lobe involvement was compared with that of two conventional methods based on the retromandibular vein line (RMVL) and facial nerve line (FNL). The relationship between the tumor and the main trunk of the facial nerve was also evaluated on the 3D-DESS-WE sequence. On 3D-DESS-WE images, the main trunk, temporofacial division, and cervicofacial division of the intraparotid facial nerve were visualized in 100% (25/25), 48% (12/25), and 36% (9/25) of patients, respectively. The diagnostic accuracy of the 3D-DESS-WE sequence for prediction of deep lobe involvement was 92% (23/25), which was significantly superior to that of the RMVL (68% [17/25]; p = 0.008) and FNL (64% [16/25]; p = 0.004) methods. The relationship between the tumor and the main trunk of the facial nerve was correctly predicted in 92% (23/25) of 3D-DESS-WE images. By direct visualization of the facial nerve, the 3D-DESS-WE sequence improved the preoperative localization of the intraparotid facial nerve in deep-seated parotid tumors. This information may help better surgical planning for deep-seated parotid tumors.

The current unpredictable climate changes are causing frequent and severe droughts. Such circumstances emphasize the need to understand the response of plants to drought stress, especially in rice, one of the most important grain crops. Knowledge of the drought stress response components is especially important in plant roots, the major organ for the absorption of water and nutrients from the soil. Thus, this article reviews the root response to drought stress in rice. It is presented to provide readers with information of use for their own research and breeding program for tolerance to drought stress in rice.

I examine the IPO underpricing phenomenon through the lens of incomplete contracts, where holdup is measured with the degree of IPO underpricing. Competing theories set forth a reason for why one type of IPO contracting (Firm Commitment Offering) is a more efficient contract than another (Best Efforts Offering). In particular, I highlight how i) allocation of bargaining power and ii) default option in the renegotiation process can be mapped to key features of the contracts. This paper demonstrates the general applicability of contract theories to study a well-known phenomenon of IPO underpricing.

In this paper, the control problem of excess-loop length of strip-caster is proposed. To realize an accurate loop length control, a linear CCD camera is adopted and an heuristic algorithm for loop length inference is developed. In addition to these, a simple but effective adaptive fuzzy control algorithm which is asymptotically stable is proposed as the control algorithm. By implementing these in a specially designed computer control systems and applying them to a pilot strip-casting machine, the proposed methods are shown to be reliable and give good performances. The proposed systems have been implemented at the pilot strip-casting machine in POSCO.
