Most genotype XII newcastle disease viruses (NDVs) were isolated from poultry, chickens, or geese, with the exception of one subtype, XIIa NDV, which was isolated from a peacock. Here, two subtype XIIb NDVs, francolin/China/GX01/2017 and francolin/China/GX02/2017 (GX01 and GX02 hereafter), were isolated from francolins, which are resident birds in southern China. GX01 and GX02 were characterized as velogenic NDVs. Based on the weaker pathogenicity of these viruses in chickens, the amino acid sequences of seven proteins from genotype XII NDVs were compared, which revealed 17, 40, 15, 7, 32, 25, and 31 variations in the NP, P, M, F, HN, L, and V proteins, respectively, some of which could be responsible for this decreased pathogenicity. Epidemiological and phylogenetic analyses suggest that subtype XIIb NDVs have multiple transmission chains, and that resident birds may be involved in this process as intermediate hosts in which viruses keep evolving. Because of the increased pathogenicity of subtype XIIb NDVs, the protective efficacy of GX01 as an inactivated vaccine was evaluated and compared with that of two commercial inactivated vaccines in chickens. The results showed that the subtype XIIb NDVs could be candidate genotype-matched vaccine strains against genotype XII NDVs.

The density-based applied spatial clustering algorithm is an algorithm based on high-density interconnected regions, which discovers class clusters of arbitrary shapes in noisy data sets and is widely used. However, it suffers from slow computation speed due to large-scale disk I/O and clustering bias due to uneven density class clusters and poor parameter search ability. To address these problems, a parallel density clustering algorithm based on an improved fruit fly optimization algorithm and Spark memory iteration is proposed. The proposed algorithm first divides the data grid using an irregular dynamic density region partitioning strategy. Then, a hybrid fruit fly particle swarm algorithm based on a genetic optimization mechanism is proposed to achieve dynamic optimization seeking for parameters in local clustering to improve the clustering effect of local clustering. Finally, the local merging of samples in irregularly bounded grid cells under each partition is achieved by designing a custom clustering merging strategy. The experiments show that the improved algorithm is generally applicable to the clustering of different shape class clusters and larger scale data and has obvious improvement in accuracy and parallel efficiency.

Chicken parvovirus (ChPV), chicken infectious anaemia virus (CIAV) and fowl adenovirus serotype 4 (FAdV-4) are avian viruses that have emerged in recent years and have endangered the global poultry industry, causing great economic loss. In this study, a multiplex fluorescence-based loop-mediated isothermal amplification (mLAMP) assay for detecting ChPV, CIAV and FAdV-4 was developed to simultaneously diagnose single and mixed infections in chickens. Three primer sets and composite probes were designed according to the conserved regions of the NS gene of ChPV, VP1 gene of CIAV and hexon gene of FAdV-4. Each composite probe was labelled with a different fluorophore, which was detached to release the fluorescence signal after amplification. The target viruses were distinguished based on the colour of the mLAMP products. The mLAMP assay was shown to be sensitive, with detection limits of 307 copies of recombinant plasmids containing the ChPV target genes, 749 copies of CIAV and 648 copies of FAdV-4. The assay exhibited good specificity and no cross-reactivity with other symptomatically related avian viruses. When used on field materials, the results of the mLAMP assay were in 100% agreement with those of the previously published PCR assay. The mLAMP assay is rapid, economical, sensitive and specific, and the results of amplification are directly observable by eye. Therefore, the mLAMP assay is a useful tool for the clinical detection of ChPV, CIAV and FAdV-4 and can be applied in rural areas.

A hydrophobic surface effectively enhances the photoelectrochemical CO2 reduction activity at the three-phase interface of Cu2O nanowires in an aqueous electrolyte.

Electroacupuncture (EA) can protect against acute urinary retention (AUR); however, the underlying mechanism remains unclear. Non-vesicular ATP release mediated by transient receptor potential (TRP) channels were identified as a key contributor to signaling in urothelial cells. In this study, the AUR model was established by urethral outlet obstruction in female Sprague-Dawley rats. EA was performed at SP6 and BL32 for 0.5 h prior to induction of AUR. EA reduced TRPV1 expression and urinary ATP concentrations in rat bladder, decreased the peak intravesical pressure during AUR, and attenuated abnormal voiding patterns and bladder pathological injury induced by AUR. Besides, 179 patients who experienced postoperative urinary retention were recruited and found that EA reduced urinary ATP concentrations and accelerated the recovery of spontaneous voiding. These observations indicate that EA exerts protection against AUR-induced bladder dysfunction by reducing urinary ATP concentrations through the regulation of TRPV1.

To compare the safety and efficacy of step-by-step debulking Microwave Ablation (MWA) with Transarterial Chemoembolization (TACE) monotherapy for huge (≥10 cm in diameter) unresectable hepatocellular carcinoma (HCC) after TACE refractoriness.This is a multi-center retrospective study carried out on 599 patients with huge unresectable HCC who received TACE as first-line therapy at five hospitals from January 2009 to December 2018. A total of 103 patients with TACE refractoriness were divided into two cohorts: monthly step-by-step debulking MWA (Median follow-up period was 24.3 months and median OS and PFS were significantly longer in the MWA group than in the TACE group (OS 21.0 vs. 11.7 months, PFS 6.1 vs. 3.0 months, bothStep-by-step debulking MWA can significantly improve long-term OS and PFS in patients with huge unresectable HCCs compared with repeated TACE after TACE refractoriness. Key PointThe debulking MWA therapy provides significantly longer OS and PFS than continued TACE for patients with huge unresectable HCCs after TACE-refractory, especially with complete tumor ablation.The most common complications were fever (48.1%) and pain (46.2%) in the MWA group. Two major complications (abdominal infection) were recorded in the MWA group, which recovered after symptomatic treatment.During the course of repeated MWAs, liver hyperplasia appeared mainly after the second MWA procedure and the average maximum increased RLV/TLV rate was 16.3%±12.7%.

The GX2020-019 strain of fowl adenovirus serotype 4 (FAdV-4) was isolated from the liver of chickens with hydropericardium hepatitis syndrome in Guangxi Province, China, and was purified by plaque assay three times. Pathogenicity studies showed that GX2020-019 can cause typical FAdV-4 pathology, such as hydropericardium syndrome and liver yellowing and swelling. Four-week-old specific pathogen-free (SPF) chickens inoculated with the virus at doses of 103 median tissue culture infectious dose (TCID50), 104 TCID50, 105 TCID50, 106 TCID50, and 107 TCID50 had mortality rates of 0, 20, 60, 100, and 100%, respectively, which were lower than those of chickens inoculated with other highly pathogenic Chinese isolates, indicating that GX2020-019 is a moderately virulent strain. Persistent shedding occurred through the oral and cloacal routes for up to 35 days postinfection. The viral infection caused severe pathological damage to the liver, kidney, lung, bursa of Fabricius, thymus, and spleen. The damage to the liver and immune organs could not be fully restored 21 days after infection, which continued to affect the immune function of chickens. Whole genome analysis indicated that the strain belonged to the FAdV-C group, serotype 4, and had 99.7–100% homology with recent FAdV-4 strains isolated from China. However, the amino acid sequences encoded by ORF30 and ORF49 are identical to the sequences found in nonpathogenic strains, and none of the 32 amino acid mutation sites that appeared in other Chinese isolates were found. Our research expands understanding of the pathogenicity of FAdV-4 and provides a reference for further studies.

In this paper, the Seismic risk in the middle of the Bayan Har block based on the profile likelihood estimation of the generalized Pareto distribution is studied. Earthquake samples in the middle of the Bayan Har block in the late 60 years are analyzed. The result shows that the area experiences an earthquake above 7.0 every 5 years. The theoretical upper limit of magnitude is 8.7, indicating frequent geological activities in this area. The profile likelihood estimation is consistent with the estimation of the maximum likelihood of the parameters. The confidence interval of the short-term return level is the same for the two methods. But with the increasing return period, the confidence interval obtained by the profile likelihood estimation is shifted to the right as a whole compared to that of the Delt method. The ratio of the confidence interval length on both sides of the return level will increase. This asymmetry is effective for the uncertainty of magnitude prediction.

Purpose：AFP appears to be negative about 30% of overall hepatocellular carcinoma (HCC). Our study aimed to develop a nomogram model to diagnose AFP negative HCC (AFPN-HCC). Patients and methods: The training set and the external validation set consisted of 516 and 456 objects. LASSO, univariate and multivariable logistic regression were performed to construct the model and then transformed into a visualized nomogram. We further used the receiver operating characteristic (ROC) curves, the calibration curve, decision curve analysis (DCA) and clinical impact curve (CIC) for validation. Results:Four variables included age, PIVKA-II, platelet (PLT) counts and prothrombin time(PT) were selected to establish the nomogram. The area under the curve (AUC) of the ROC to distinguish AFPN-HCC patients was 0.937(95%CI, 0.892-0.938) in training set and 0.942(95%CI, 0.921-0.963) using the validation set and indicated satisfactory discriminative ability of the model. The calibration plots showed favorable consistency between the prediction of the nomogram and actual observations. DCA and CIC showed that the nomogram was clinically useful. Conclusions:Our model was effective for discrimination of AFPN-HCC from control subsets, and might be helpful for the diagnosis for AFPN-HCC.

BackgroundWhether irAEs can predict the efficacy of PD-1 inhibitors in cholangiocarcinoma (CCA) has not been assessed. Therefore, this study aims to investigate the correlation between irAEs and the therapeutic effect of PD-1 inhibitors combination therapy in patients with advanced CCA.MethodsAll patients with CCA who were consecutively admitted to the inpatient unit of our hospital and received PD-1 inhibitors combination therapy between September 2020 and April 2022 were screened. In total, 106 patients with CCA were screened out. We then followed up these patients until October 2022. Due to perioperative use (n=28), less than 2 cycles of PD-1 inhibitor therapy (n=9), incomplete data (n=8) and no pathological report (n=2), 59 patients were included in the final analysis. The patients were divided into the irAEs cohort and the non-irAEs cohort according to whether they experienced irAEs or not. The Log-Rank test was performed to compare the difference in survival time between these two cohorts. We then applied multivariate COX regression analysis to investigate whether irAEs were independent prognostic factors for survival in patients with advanced CCA.ResultsFinally, 32 patients were included in the irAEs cohort and 27 patients in the non-irAEs cohort. A total of 32 patients (54.2%) had any-grade irAEs, of which 4 patients (6.8%) had grade 3-4 irAEs. The most common irAEs were thyroid toxicity (30.5%) and dermatologic toxicity (30.5%). There were no notable differences in demographics and clinical characteristics between the irAEs and non-irAEs cohorts, except for total bilirubin level (P=0.026) and relapse (P=0.016). The disease control rate (DCR) in the irAEs cohort was higher than in the non-irAEs cohort (90.6% vs 70.4%, P=0.047). Median overall survival (OS) and median progression-free survival (PFS) were better in the irAEs cohort than in the non-irAEs cohort (OS: 21.2 vs 10.0 months, PConclusionIrAEs correlated with improved DCR, OS, and PFS in advanced CCA patients receiving PD-1 inhibitors combination therapy.

BackgroundThe emergence of highly drug-resistant K. pneumoniae, has become a major public health challenge. In this work, we aim to investigate the diversity of species and sequence types (STs) of clinical Klebsiella isolates and to characterize the prevalence and structure of class 1 integrons.MethodsBased on the whole genome sequencing, species identification was performed by 16S rRNA gene homology and average nucleotide identity (ANI) analysis. STs were determined in accordance with the international MLST schemes for K. pneumoniae and K. variicola. Integron characterization and comparative genomic analysis were performed using various bioinformatic tools.ResultsSpecies identification showed that the 167 isolates belonged to four species: K. pneumoniae, K. variicola subsp. variicola, K. quasipneumoniae and K. aerogenes. Thirty-six known and 5 novel STs were identified in K. pneumoniae, and 10 novel STs were identified in K. variicola subsp. variicola. Class 1 integrons were found in 57.49% (96/167) of the isolates, and a total of 169 resistance gene cassettes encoding 19 types of resistance genes, including carbapenem resistance gene (blaIPM-4) and class D β-lactamases gene (blaOXA-1 and blaOXA-10), were identified. Among the 17 complete genomes, 29 class 1 integrons from 12 groups were found, only 1 group was encoded on chromosomes. Interestingly, one plasmid (pKP167-261) carrying two copies of approximately 19-kb IS26-Int1 complex resistance region that contains an integron and a multidrug resistance gene fragment.ConclusionThe results of this work demonstrated that the species and STs of the clinical Klebsiella isolates were more complex by the whole genome sequence analysis than by the traditional laboratory methods. Finding of the new structure of MGEs related to the resistance genes indicates the great importance of deeply exploring the molecular mechanisms of bacterial multidrug resistance.

The present study aimed to investigate whether leucine (Leu) alleviates oxidative injury in bovine intestinal epithelial cells (BIECs) induced by hydrogen peroxide (HBIECs were treated with HThese results indicate that Leu promotes the relative expression of antioxidant enzymes (SOD2, CAT and GPx1) and phase II detoxification enzymes (HO-1) by upregulating nuclear Nrf2 and activating the Nrf2 signaling pathway, thus enhancing the antioxidant capacity of cells. © 2022 Society of Chemical Industry.

A superior allele of wheat gene TaGL3.3-5B was identified and could be used in marker-assisted breeding in wheat. Identifying the main genes which mainly regulate the yield-associated traits can significantly increase the wheat production. In this study, gene TaGL3.3 was cloned from common wheat according to the sequence of OsPPKL3. A SNP in the 8th exon of TaGL3.3-5B, T/C in coding sequence (CDS), which resulted in an amino acid change (Val/Ala), was identified between the low 1000-kernel weight (TKW) wheat Chinese Spring and the high TKW wheat Xinong 817 (817). Subsequently, association analysis in the mini-core collection (MCC) and the recombinant inbred lines (RIL) revealed that the allele TaGL3.3-5B-C (from 817) was significantly correlated with higher TKW. The high frequency of TaGL3.3-5B-C in the Chinese modern wheat cultivars indicated that it was selected positively in wheat breeding programs. The overexpression of TaGL3.3-5B-C in Arabidopsis resulted in shorter pods and longer grains than those of wild-type counterparts. Additionally, TaGL3.3 expressed a tissue-specific pattern in wheat as revealed by qRT-PCR. We also found that 817 showed higher expression of TaGL3.3 than that in Chinese Spring (CS) during the seed development. These results demonstrate that TaGL3.3 plays an important role in the formation of seed size and weight. Allele TaGL3.3-5B-C is associated with larger and heavier grains that are beneficial to wheat yield improvement.

This study aimed to identify potential drug targets for lung adenocarcinoma (LUAD) patients with COVID-19.Methods: LUAD samples were obtained from public databases. Differentially expressed genes (DEGs) related to COVID-19 were screened. Protein–protein interactions among COVID-19-related genes, the traditional Chinese medicine (TCM) and TCM target genes were analyzed by CytoScape. The correlation between tumor microenvironment and COVID-19 target genes were assessed by Pearson correlation analysis. Unsupervised consensus clustering was conducted to categorize molecular subtypes.Results: We filtered 26 COVID-19 target genes related to TCM for LUAD. Interleukin (IL)-17 signaling pathway and tumor necrosis factor (TNF) signaling pathway were significantly enriched in these 26 genes. A strong correlation was found between COVID-19 target genes and tumor microenvironment (TME), cell death. Importantly, interleukin-1beta (IL1B) was identified as a core gene in the protein–protein interactions (PPI) network. Based on the 26 target genes, two molecular subtypes showing distinct overall survival, TME and response to target therapy were developed.Conclusions: This study explored 26 COVID-19 target genes, which could serve as potential therapeutic drug targets for LUAD. IL1B was verified as a critical target for developing new molecular drugs. Objective. To evaluate the influence of sinusoidal vibration (50-Hertz) stimulation on the uterus of osteoporotic rats. Methods. We constructed an osteoporosis rat model by ovariectomy (OVX). 36 3-month-old Sprague Dawley rats were randomly divided into the control group, vibrating group, sham operation group, sham operation vibrating group, OVX group, and OVX vibrating group ( n = 6 per group). Rats started to vibrate one week after the operation: one 10 minutes 50-Hertz sinusoidal vibration per day, except for Saturday and Sunday. In the second, 8, and 12 week after vibration stimulation, rats were sacrificed in batches. And then, the uteruses were taken out to measure the wet weight and calculate uterus relative wet weight. Results. Compared with the control group, OVA induced a significant increase in wet weight and relative wet weight in rat uterus. The vibration was to the uterus wet weight and the uterus relative wet weight in ovariectomized rats and at the same time had no significant effect, but the 12-week prolonged vibration can significantly reduce the uterus wet weight and the uterus relative wet weight in ovariectomized rats than 2 weeks. Conclusions. The uterus wet weight and the uterus relative wet weight were increased in the OVA-induced osteoporosis rats. The 50-Hertz sinusoidal vibration had no significant effect on the uterus wet weight and the uterus relative wet weight in the ovariectomized rats at the same time, but 12 weeks of vibration can significantly reduce the uterine wet weight and uterine relative wet weight of ovariectomized rats. And the uterus relative wet weight can be used as a new indicator of stimulating the uterus.

Port-A-Cath systems (PCS) are safe and convenient devices for long-term infusion in patients with malignant tumors. This study retrospectively analyzed the complications from PCS and their management.Data of 1695 adults (641 males and 1054 females) with malignant tumors who underwent PCS implantation in our center from January 1, 2009 to December 31, 2019 who had complete follow-up records were collected in this study. The early and late complications and corresponding treatments were studied.A total of 1716 PCSs were implanted; 21 patients underwent 2 implantations each. The success rate was 100% and no severe complications occurred during implantation. The overall occurrence rate of post-implantation complications was 18.5% (318/1716); 5.5% (94/1716) were early complications and 13.0% (224/1716) were late complications. A total of 451 PCSs were removed, of which 398 were removed due to the end of chemotherapy, while 53 were removed because of complications. A total of 4 deaths occurred from these complications.The incidence of intra- and post-operative complications is low. In most cases, complications can be effectively controlled without the removal of the PCS and regular follow-up and maintenance are critical.

The starch composition of tubers affects the quality and yield of yam, and it is of great significance to deeply investigate the mechanism of starch accumulation in yam. In this study, two yam varieties with significant differences in starch content, Bikeqi (Dioscorea opposita Bikeqi) and Dahechangyv (Dioscorea opposita Dahechangyu), were used as materials. The transcriptome of tubers from seven developmental stages were sequenced using RNA-seq and Iso-seq. Differential genes closely contacted with the metabolism of starch and sucrose were screened and the functions were analysed. The results showed that, the starch content of the two yam varieties reached the maximum at 150 and 165 days respectively, and their difference was the largest at 150 days. Total 21623 unigenes were annotated, and 8378 differential genes were identified within varieties by comparing each developmental stage with the beginning of tuber expansion (90 days), respectively. A total of 5177 differential genes were identified in two varieties in the same period. GO and KEGG were used to analyze these differential genes, a variety of key genes closely related to starch and sucrose metabolism were screened. These results help us to better understand the mechanism of starch accumulation in yam.

Plasmonic Ag-decorated Cu2O nanowires for efficient photoelectrochemical CO2 reduction to multi-carbon product acetic acid in an aqueous electrolyte.

As a green and environmental protection method, the self-repairing technology has great expectations to be met. In this study, the influence of ettringite on the crack self-repairing of cement-based materials in a hydraulic environment was systematically analyzed. First, the composition and pore characteristics of the self-repairing carrier was studied by XRD, XRF, and the mercury injection method. The composition was basically consistent with cement-based materials, and the structure of the self-repairing carrier was relatively dense. Second, the products were analyzed by SEM and EDS, and it was found that the distribution range of crystal size was from 1 to 20 µm, the cube and flocculent accumulation was calcite, while the rod and flake crystals were ettringite. Finally, the repair effect on the surface and inside of the crack was demonstrated by the crack area repair rate and water permeability resistance. Compared with single microbial mineralization, the synergistic repair effect of microbial mineralization and ettringite formation was better in water permeability resistance. With the further increase of water pressure, its advantages would be more obvious.

Background Genome-wide sequencing may extensively identify potential pathogenic variants, which helps to understand mechanisms of tumorigenesis, but such study has not been reported in benzene-induced leukemia (BIL). Methods We recruited 10 BIL patients and conducted the whole-exome sequencing on their peripheral blood samples. The obtained sequencing data were screened for potential pathogenic and novel variants, then the variants-located genes were clustered to identify cancer-related pathways. Shared or recurrent variants among the BIL cases were also identified and evaluated for their potential functional impact. Results We identified 48,802 variants in exons in total, 97.3% of which were single nucleotide variants. After filtering out variants with minor allele frequency ≥ 1%, we obtained 8667 potentially pathogenic variants, of which 174 were shared by all the BIL cases. The identified variants located in genes that could be significantly enriched into certain cancer-related pathways such as PI3K-AKT signaling pathway and Ras signaling pathway. We also identified 1010 novel variants with no record in the Genome Aggregation Database and in dbSNP, and one of them was shared by 90% cases. The recurrent and novel variant caused a missense mutation in SESN3. Conclusions We examined variations of the whole exome in BIL patients for the first time. The commonly shared variants implied a relation with BIL, and the recurrent and novel variant might be specifically related to BIL. The related variants may help unravel the carcinogenic mechanisms of BIL.

Paper and plastic bags have been mainly used in fruit bagging of pineapple production. However, they are often discarded as agricultural wastes after harvest of pineapple fruit and threaten environment. In this study, effects of white non-woven fabric bag, expandable polyethylene bag, black plastic bag and kraft paper bag on fruit quality of pineapple cv. ‘MD-2’ were investigated. Results showed that fruits with fruit bagging showed no sunburn. When expandable polyethylene bag was used, fruits of pineapple cv. ‘MD-2’ showed better external qualities, nutrition qualities and texture properties. Expandable polyethylene bag made of degradable and recyclable materials could be recommended as a promising practice to reduce agricultural waste in pineapple production. RESUMO: Sacos de papel e plástico têm sido usados principalmente no ensacamento de frutas na produção de abacaxi. No entanto, eles são frequentemente descartados como resíduos agrícolas, após a colheita da fruta do abacaxi, e ameaçam o meio ambiente. Neste estudo, os efeitos da sacola de tecido, não tecido branco, sacola de polietileno expansível, sacola plástica preta e sacola de papel artesanal na qualidade do fruto do abacaxi cv. ‘MD-2’ foram investigados. Os resultados mostraram que as frutas ensacadas não apresentaram queimaduras solares. Quando foi utilizada a bolsa de polietileno expansível, as frutas de abacaxi cv. ‘MD-2’ mostraram melhores qualidades externas, qualidades nutricionais e propriedades de textura. O saco de polietileno expansível feito de materiais degradáveis e recicláveis pode ser recomendado como uma prática promissora para reduzir o desperdício agrícola na produção de abacaxi.

Background & aims Although the prevalence of nonalcoholic fatty liver disease (NAFLD) has risen dramatically to 25% of the adult population worldwide, there are as yet no approved pharmacological interventions for the disease due to uncertainty about the underlying molecular mechanisms. It is known that mitochondrial dysfunction is an important factor in the development of NAFLD. Mitochondrial antiviral signaling protein (MAVS) is a critical signaling adaptor for host defenses against viral infection. However, the role of MAVS in mitochondrial metabolism during NAFLD progression remains largely unknown. Approach & results Based on expression analysis, we identified a marked downregulation of MAVS in hepatocytes during NAFLD progression. By employing MAVS global knockout and hepatocyte-specific MAVS knockout mice, we found that MAVS is protective against diet-induced NAFLD. MAVS deficiency induces extensive mitochondrial dysfunction during NAFLD pathogenesis which was confirmed as impaired mitochondrial respiratory capacity and membrane potential. Metabolomics data also showed the extensive metabolic disorders after MAVS deletion. Mechanistically, MAVS interacts with the N-terminal stretch of voltage-dependent anion channel 2 (VDAC2), which is required for the ability of MAVS to influence mitochondrial function and hepatic steatosis. Conclusions In hepatocytes, MAVS plays an important role in protecting against NAFLD by helping to regulate healthy mitochondrial function. These findings provide new insights regarding the metabolic importance of conventional immune regulators and support the possibility that targeting MAVS may represent a new avenue for treating NAFLD.
