Due to the huge potential energy associated with water storage in reservoirs, dam-break floods are often catastrophically destructive for people and structures downstream. This study aims to simulate and compare floods generated under various dam-break scenarios and their downstream impacts, taking Pingshuijiang Reservoir in southeastern China as an example. A two-dimensional hydrodynamic model is used to simulate the downstream evolution of floods under three dam-break scenarios, and the breach flood and downstream inundation process are analyzed. Gradual failure of the main dam leads to near-total inundation of the nearby town over c. 1 h, allowing time for warning and evacuation. Instantaneous failure of the main dam results in larger peak flow, greater submergence depth and faster inundation (20 min), leaving little time for warning/evacuation. Instantaneous failure of the auxiliary dam generates a much lower peak flow magnitude and, although the town is still largely submerged within 45 min, the shallow water depth and low velocity are conducive to rescue/evacuation. The results show significant variation in flood process and submergence due to dam size and failure mode that provide guidance for dam-break flood risk assessment and disaster avoidance planning.

4. Clinical Efficacy of Subhypothermia in the Treatment of Neonatal Hypoxic-Ischemic Encephalopathy Combined with Myocardial Damage

Objective. To investigate the efficacy of subhypothermia in the treatment of neonatal hypoxic-ischemic encephalopathy (HIE) combined with myocardial damage. Methods. 136 children with HIE and myocardial damage admitted to our hospital from October 2019 to October 2021 were included in the study and were divided into study group and control group of 68 cases each according to the random number table method. The control group was given conventional treatment, and the study group was treated with subhypothermia therapy on top of the control group. Comparing the effects of treatment between the two groups. The serum levels of S-100β protein, Tau protein, neuron-specific enolase (NSE), creatine kinase (CK), creatine kinase isoenzyme MB (CK-MB), lactate dehydrogenase (LDH), alpha-hydroxybutyrate dehydrogenase (ɑ-HBDH), myoglobin (Myo), cardiac trophic factor-1 (CT-1), cardiac troponin I (cTnI), superoxide dismutase (SOD), reactive oxygen species (ROS), glutathione peroxidase (GSH-Px), interleukin-1β (IL-1β), interleukin-8 (IL-8), and tumor necrosis factor-ɑ (TNF-ɑ) were measured in both groups before and after treatment, respectively. Results. The total effective rate was higher in the study group (88.24%) than in the control group (72.06%) ( P < 0.05 ). After treatment, the serum levels of S-100β protein, Tau protein, NSE, CK, CK-MB, LDH, ɑ-HBDH, Myo, CT-1, cTnI, ROS, IL-1β, IL-8, and TNF-ɑ were reduced in both groups, and the study group was lower than the control group ( P < 0.05 ). The serum levels of SOD and GSH-Px were higher in both groups after treatment than before treatment and were higher in the study group than in the control group ( P < 0.05 ). Conclusion. Subhypothermia treatment of children with HIE combined with myocardial injury can further improve the hypoxic-ischemic state; reduce myocardial damage, oxidative stress, and inflammatory response; and has a good overall efficacy.

5. Deep‐learning‐based removal of autofluorescence and fluorescence quantification in plant‐colonizing bacteria in vivo

Fluorescence microscopy is common in bacteria-plant interaction studies. However, strong autofluorescence from plant tissues impedes in vivo studies on endophytes tagged with fluorescent proteins. To solve this problem, we developed a deep-learning-based approach to eliminate plant autofluorescence from fluorescence microscopy images, tested for the model endophyte Azoarcus olearius BH72 colonizing Oryza sativa roots. Micrographs from three channels (tdTomato for gene expression, green fluorescent protein (GFP) and AutoFluorescence (AF)) were processed by a neural network based approach, generating images that simulate the background autofluorescence in the tdTomato channel. After subtracting the model-generated signals from each pixel in the genuine channel, the autofluorescence in the tdTomato channel was greatly reduced or even removed. The deep-learning-based approach can be applied for fluorescence detection and quantification, exemplified by a weakly expressed, a cell-density modulated and a nitrogen-fixation gene in A. olearius. A transcriptional nifH::tdTomato fusion demonstrated stronger induction of nif genes inside roots than outside, suggesting extension of the rhizosphere effect for diazotrophs into the endorhizosphere. The pre-trained convolutional neural network model is easily applied to process other images of the same plant tissues with the same settings. This study showed the high potential of deep-learning-based approaches in image processing. With proper training data and strategies, autofluorescence in other tissues or materials can be removed for broad applications.

7. Satellite-based dry-wet seasonal changes of OCS surface budgets over the Amazon rainforests

- Abstract
Carbonyl sulfide (OCS) is the most dominant sulfur-containing species in the atmosphere and is an important tracer of the terrestrial gross primary productivity as it is involved only in photosynthesis. Biomass burning and terrestrial uptakes by plants and soil is the primary terrestrial source and sink of OCS, respectively. The Amazon basin alone accounts for 10% of the global biomass burning emission and 33% of the global plant/soil uptake. However, both terms are sensitive to water stress, heat stress, and the associated wildfires in the dry seasons. Here, we estimate the dry-wet seasonal difference of the terrestrial OCS budget over the Amazon region by constraining the NCAR MOZART4 chemistry-transport model with the mid-tropospheric OCS abundances retrieved from NASA’s Thermal Emission Spectrometer (TES) measurements during 2004 and 2012. Our perturbative calculations show that biomass-burning emissions that are predominant in the south rim of the Amazon have more influence on the mid-tropospheric OCS over the southeast subtropical Amazon. In comparison, the plant/soil uptakes that are predominant in the tropical Amazon have more influence over the northwest tropical Amazon. This dipole spatial pattern helps distinguish the mid-tropospheric OCS seasonal variability due to biomass-burning emissions and plant/soil uptakes.

8. Improvement of straw decomposition and rice growth through co-application of straw-decomposing inoculants and ammonium nitrogen fertilizer

Background The growth of rice is reduced by the slow decomposition of accumulated straw, which competes with rice for soil nitrogen nutrient. In recent year, straw-decomposing inoculants (SDIs) that can accelerate straw decomposition and ammonium nitrogen (N) fertilizer that can quickly generate available N is increasingly adopted in China. However, it is still unknown whether the N demand of straw decomposition and crop growth can be simultaneously met through the co-application of SDIs and ammonium N fertilizer. Results In this study, we investigated the effect of the co-application of SDIs and ammonium bicarbonate on decomposition rate of wheat straw, rice growth and rice yield over two consecutive years in rice-wheat rotation system. Compound fertilizer (A0) was used as control. The ratios of ammonium bicarbonate addition were 20% (A2), 30% (A3) and 40% (A4), respectively, without SDIs or with SDIs (IA2, IA3, IA4). Our results revealed that without SDIs, compared with A0, straw decomposition rate, rice growth and yield were improved under A2; However, under A3, rice yield was decreased due to the slow decomposition rate of straw and limited growth of rice during late growth stage. Combining SDIs and N fertilizer increased straw decomposition rate, rice growth rate and yield more than that of N fertilizer alone, especially under IA3. Compared with A0, straw decomposition rate, tiller number, aboveground biomass, leaf area index, root length, and nitrogen use efficiency were significantly increased by 16%, 8%, 27%, 12%, 17%, and 15% under IA3. Consequently, the average rice yield of IA3 was increased to 10,856 kg/ha, which was 13% and 9% higher, respectively, than of A0 and A2. Conclusion Our results indicated that ammonium bicarbonate application alone carried a risk of nutrient deficiency during late growth stage and yield decline. Therefore, the co-application of SDIs and 30% ammonium N fertilizer substitution can be a favorable practice to simultaneously accelerate straw decomposition and increase rice crop growth.

9. The causal effects of leisure screen time on irritable bowel syndrome risk: result from a Mendelian randomization study

- Abstract
Associations between leisure sedentary behavior (especially leisure screen time, LST) and irritable bowel syndrome (IBS) have been reported, but causality is unclear. Here, the two-sample Mendelian randomization was performed to investigate the causal association between LST and IBS. Two recently published genome-wide association studies (GWASs) including a total of 1,190,502 people from Europe were used as our data source. Inverse variance weighting (OR = 1.120, 95% CI 1.029–1.219) and weighted median (OR = 1.112, 95% CI 1.000–1.236) analyses revealed an association between the genetically predicted risk of IBS and LST. There was no evidence of pleiotropy in the sensitive analysis (MR-Egger, p = 0.139). After removing potentially confounding single nucleotide polymorphisms, similar results were found using inverse variance weighting (OR = 1.131, 95% CI 1.025–1.248) and weighted median (OR = 1.151, 95% CI 1.020–1.299), as well as in the validation analyses using inverse variance weighting (OR = 1.287, 95% CI 0.996–1.662). This study provided support for a possible causal relationship between leisure screen time and IBS. This information can be used to gain insight into the prevention and treatment of disease.

Hot dip galvanizing can effectively prevent the corrosion of steel. With the increasing demand of industry for steel strip, higher requirements are put forward for stable and continuous operation of submerged roller bearings in hot dip galvanizing line. The existing sliding bearings for sink roll of galvanizing line mainly depend on imports, and their service life is about two weeks. Two weeks later, the sliding bearing failed, so it is necessary to stop the line and replace the new bearing, wasting a lot of material cost and time cost. In this paper, zinc liquid corrosion test, high temperature pin-and-disc dry friction test and corrosion wear test were carried out on the materials of sliding bearing pair of submerged roller, mainly alumina ceramic bushing and three kinds of metal sleeves (alloy A, alloy B and 316L stainless steel) under simulated actual working conditions. The status of corrosion and wear in bearing failure was compared and analyzed, and the hydrodynamic lubrication effect formed by zinc liquid under test conditions was considered, and the main failure mechanism was comprehensively analyzed.

11. SDN: Semantic Decoupling Network for Temporal Language Grounding

Temporal language grounding (TLG) is one of the most challenging cross-modal video understanding tasks, which aims at retrieving the most relevant video segment from an untrimmed video according to a natural language sentence. The existing methods can be separated into two dominant types: 1) proposal-based and 2) proposal-free methods, where the former conduct contextual interactions and the latter localizes timestamps flexibly. However, the constant-scale candidates in proposal-based methods limit the localization precision and bring extra computational costs. In contrast, the proposal-free methods perform well on high-precision metrics-based on the fine-grained features but suffer from a lack of coarse-grained interactions, which cause degeneration when the video becomes complex. In this article, we propose a novel framework termed semantic decoupling network (SDN) that combines the advantages of proposal-based and proposal-free methods and overcomes their defects. It contains three key components: 1) semantic decoupling module (SDM); 2) context modeling block (CMB); and 3) semantic cross-level aggregation module (SCAM). By capturing the video-text contexts in multilevel semantics, the SDM and CMB effectively utilize the benefits of proposal-based methods. Meanwhile, the SCAM maintains the merit of proposal-free methods in that it localizes timestamps precisely. The experiments on three challenge datasets, i.e., Charades-STA, TACoS, and ActivityNet-Caption, show that our proposed SDN method significantly outperforms recent state-of-the-art methods, especially the proposal-free methods. Clinical cancer trials are designed to collect radiographic measurements of each patient’s baseline and residual tumor burden at regular intervals over the course of study. For solid tumors, the extent of reduction in tumor size following treatment is used as a measure of a drug’s antitumor activity. Statistical estimation of treatment efficacy routinely reduce the longitudinal assessment of tumor burden to a binary outcome describing the presence versus absence of an objective tumor response as defined by RECIST criteria. The objective response rate (ORR) is the predominate method for evaluating an experimental therapy in a single-arm trial. Additionally, ORR is routinely compared against a control therapy in phase III randomized controlled trials. The longitudinal assessments of tumor burden are seldom integrated into a formal statistical model, nor integrated into mediation analysis to characterize the relationships among treatment, residual tumor burden, and survival. This article presents a frameworkfor landmark mediation survival analyses devised to incorporate longitudinal assessment of tumor burden. R2 effect-size measures are developed to quantify the survival treatment mediation effects using longitudinal predictors. Analyses are demonstrated with applications to two colorectal cancer trials. Survival prediction is compared in the presence versus absence of longitudinal analysis. Simulation studies elucidate settings wherein patterns of tumor burden dynamics require longitudinal analysis.

The Benalcazar-Bernevig-Hughes (BBH) model [Science 357, 61 (2017)], featuring bulk quadrupole moment, edge dipole moments, and corner states, is a paradigm of both higher-order topological insulators and topological multipole insulators. In this work, we generalize the BBH model to arbitrary dimensions and demonstrate the bulk multipole moment. In our demonstration, the analytical solution of corner states can be directly constructed, which possesses a unified and elegant form. Based on the corner states solution and chiral symmetries analysis, we develop a general boundary projection method to extract the boundary Hamiltonians, which thoroughly reveals the boundary-localized multipole moments of lower dimension, the hallmark of topological multipole insulators. Our work facilitates the unified understanding of topological multipole insulators and unveils their cascade hierarchy versatilely.

The bolometric Bond albedo is a fundamental parameter of planets and moons. Here, combined observations from the Cassini spacecraft and the Hubble Space Telescope are used to determine the bolometric Bond albedo of Enceladus. We provide the full-disk reflectance of Enceladus across all phase angles (0° -180°) from 150 nm to 5131 nm, a spectral range that accounts for nearly all incoming solar power. Considering the distribution of the monochromatic Bond albedo over wavelength, we find a value of 0.76 ± 0.03 for Enceladus' bolometric Bond albedo. The corresponding optical characteristics (e.g., geometric albedo and phase function), which are closely related to Enceladus' surface properties, are also investigated. The wavelength-dependent nature of Enceladus' Bond albedo suggests that the bolometric Bond albedos of other icy moons, if they are mainly determined by the visible observations only, should be carefully considered. Our new measurements of bolometric Bond albedo can be used to better determine the radiant energy budget of Enceladus and further constrain the internal heat flow, a critical driving force for the water plumes on Enceladus.

18. Predicting outcomes of phase III oncology trials with Bayesian mediation modeling of tumor response

Pivotal cancer trials often fail to yield evidence in support of new therapies thought to offer promising alternatives to standards-of-care. Conducting randomized controlled trials in oncology tends to be considerably more expensive than studies of other diseases with comparable sample size. Moreover, phase III trial design often takes place with a paucity of survival data for experimental therapies. Experts have explained the failures on the basis of design flaws which produce studies with unrealistic expectations. This article presents a framework for predicting outcomes of phase III oncology trials using Bayesian mediation models. Predictions, which arise from interim analyses, derive from multivariate modeling of the relationships among treatment, tumor response, and their conjoint effects on survival. Acting as a safeguard against inaccurate pre-trial design assumptions, the methodology may better facilitate rapid closure of negative studies. Additionally the models can be used to inform re-estimations of sample size for under-powered trials that demonstrate survival benefit via tumor response mediation. The methods are applied to predict the outcomes of two colorectal cancer studies. Simulation is used to evaluate and compare models in the absence versus presence of reliable surrogate markers of survival.

To assess the problem of sewage treatment under the condition of low carbon sources, we carried out a study of activated sludge and a biofilm symbiosis system (IFAS). The occurrence characteristics and interaction law of microorganisms in two phases of sludge membrane under low carbon source conditions were discussed, and their niche and influence on treatment efficiency were clarified. Through a pilot-scale experiment in actual water plants, the biofilm characteristics, sludge membrane activity, and succession law of flora were analyzed, and the microbial structure and interaction in sludge membrane in two phases under the control of different activated sludge ages were compared. The results showed that the sludge concentration in the reactor increased with the increase in SRT under variable SRT. Because the microbial concentration in SRT-H was much higher than that in SRT-L, the competition between mud films in SRT-H was more intense than that in SRT-L, and the pollutant removal efficiency in SRT-H was lower than that in SRT-L. Under the condition of low-carbon feed water, the sludge activity in the IFAS process decreased with the increase in SRT. Under the condition of low SRT(5 d), the nitrification, denitrification, phosphorus accumulation, and phosphorus absorption rate of activated sludge increased by 122%, 88%, 34%, and 44%, respectively, compared with that of high SRT (25 d). However, SRT had little effect on biofilm activity, and there was little difference in nitrification activity and denitrification activity between the two SRTs. Microbial sequencing analysis showed that the functional bacteria of the IFAS process were enriched and transferred with the change in SRT between the two phases of mud membrane. A novel cloud point extraction was developed for the pre-concentration and determination of Cu(II), Zn(II) and Fe(III) in bottled mineral water samples by flame atomic absorption spectrometry. The procedure was based on the formation of a charge transfer ion-pairing agent between the 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol (5-Br-PADAP) and tetraphenylborate (TPB) in the presence of 0.1 mol L−1 HCl, and the selective cloud point extraction was achieved after its complexation with target metals. Under the optimum conditions, good linearity was observed in the concentration ranges of 10–160 μg L−1 for Cu(II), 2–30 μg L−1 for Zn(II) and 20–300 μg L−1 for Fe(III). In a 50 mL of sample, concentration factors of 38.4, 39.0 and 29.9 and limits of detection of 0.44, 0.14 and 0.78 μg L−1 for Cu(II), Zn(II) and Fe(III) were obtained, respectively. Compared with single complexing reagent, the ion pair (5-Br-PADAP-TPB) significantly enhanced the enrichment factor of each analyte and tolerance levels of interference ions and obtained the lower detection limit. The proposed method was validated via the analysis of certified reference materials and successfully used for the determination of Cu(II), Zn(II) and Fe(III) in ten brands of bottled mineral water samples. All samples contained lower levels of Cu(II), Fe(III) and Zn(II) according to the maximum allowable concentration recommended by Chinese legislation. And recoveries for Cu(II), Zn(II) and Fe(III) were found to be between 95.0 and 105%; the relative standard deviation of this method was 0.620−1.67%

ObjectiveAs monotherapy is insufficient for some patients, the existing fixed-dose combination (FDC) requires two or more daily administrations with declining adherence. The present study compared the efficacy and safety of netarsudil/latanoprost FDC with monotherapy of its individual components in patients with glaucoma.MethodsA systematic literature search was performed for studies comparing netarsudil/latanoprost fixed-dose combination (FDC) vs. monotherapy in patients with glaucoma. The primary endpoints included intraocular pressure (IOP), intraocular pressure reduction percentage (IOPR%) and adverse events (AEs).ResultsThree randomized controlled trial studies (RCTs) involving 1,692 patients (FDC: 556, netarsudil: 577, latanoprost: 559) were included in this meta-analysis. FDC was more effective than netarsudil, with significantly lower diurnal IOP over three time points (8:00 a.m., 10:00 a.m., 4:00 p.m.), mean diurnal IOP (MD = −2.36 [−3.08, −1.63], P < 0.00001) and higher IOPR% (MD = 9.60 [7.86, 11.33], P < 0.00001). When comparing FDC with latanoprost, both mean diurnal IOP (MD = −1.64 [−2.05, −1.23], P < 0.00001) and diurnal IOP across 3 time points were significantly lower with FDC than with latanoprost, while FDC induced significantly higher IOPR% (MD = 6.09 [4.40, 7.77], P < 0.00001). Incidence of total AEs was similar between netarsudil and FDC, but higher with FDC than with latanoprost.ConclusionNetarsudil/latanoprost FDC appears to be superior to netarsudil or latanoprost alone, with better ocular hypotensive effects. T-cell lymphomas are aggressive lymphomas that often resist current therapy options or present with relapsed disease, making the development of more effective treatment regimens clinically important. Previously, we have shown that CD4 CAR can effectively target T-cell malignancies in preclinical studies. As IL-15 has been shown to strengthen the anti-tumor response, we have modified CD4 CAR to secrete an IL-15/IL-15sushi complex. These CD4-IL15/IL15sushi CAR T cells and NK92 cells efficiently eliminated CD4+ leukemic cell lines in co-culture assays. Additionally, CD4-IL15/IL15sushi CAR out-performed CD4 CAR in in vivo models, demonstrating a benefit to IL-15/IL-15sushi inclusion. In a Phase I clinical trial, CD4-IL15/IL15sushi CAR T cells were tested for safety in three patients with different T-cell lymphomas. Infusion of CD4-IL15/IL15sushi CAR T cells was well-tolerated by the patients without significant adverse effects and led to the remission of their lymphomas. Additionally, infusion led to the depletion of CD4+ Treg cells and expansion of CD3+CD8+ T cells and NK cells. These results suggest that CD4-IL15/IL15sushi CAR T cells may be a safe and effective treatment for patients with relapsed or refractory T-cell lymphomas, where new treatment options are needed.

A method for simultaneous preconcentration and determination of mercury species in water and soil samples was established using high-performance liquid chromatography with hydride generation atomic fluorescence spectrometry after ultrasound-assisted dual-cloud point extraction. The extraction process was divided into two steps. In the first cloud point extraction, inorganic mercury and methylmercury formed chelates with sodium diethyldithiocarbamate and were extracted into Triton X-114 micelles. In the second stage, a displacement reaction between sodium diethyldithiocarbamate-inorganic mercury/methylmercury and l-cysteine occurred, and the analytes entered the l-cysteine aqueous solution under ultrasonication. This aqueous solution was directly introduced to the high-performance liquid chromatography with hydride generation atomic fluorescence spectrometry and the detection was completed within 6 min. Under the optimum experimental conditions, the linear range was 0.10-5.0 μg/L (r ≥0.9993) for inorganic mercury and methylmercury, and the enhancement factors were 15.7 for inorganic mercury and 6.35 for methylmercury. The limits of detection for inorganic mercury and methylmercury were 0.004 and 0.016 μg/L, respectively. The approach was successfully applied to the determination of trace inorganic mercury and methylmercury in water and soil samples with good recoveries (85.3-110%). This method solved the problem of peak fusion of the two analytes and was successfully applied to the speciation analysis of mercury.

Background Childhood overweight and obesity have become significant public health challenges worldwide. The present study aimed to investigate whether caregivers’ feeding behaviour and children's eating behaviour were associated with the weight status of preschool children in China. Methods A cross‐sectional questionnaire was administered to 912 caregivers of preschool children from April to July 2016. Caregivers’ feeding behaviours were assessed by the Chinese Preschooler's Caregiver Feeding Behaviour Scale. Children's eating behaviours were evaluated using the Chinese Preschooler's Eating Behaviour Questionnaire. After controlling for demographic characteristics, multiple linear regression and logistic regression analyses were performed to evaluate the relationship between caregivers’ feeding behaviour, children's eating behaviour and children's body mass index (BMI). Results The results showed that weight concerns on the part of caregivers (β = 0.53) and food responsiveness on the part of children (β = 0.93) were positively correlated with children's BMI, whereas caregivers’ responsibility for feeding (β = −0.68) and children's external eating (β = −0.53) were negatively correlated with BMI. A 2.5-D miniaturized substrate integrated electromagnetic bandgap (SI-EBG) is proposed for parallel plate cavity mode suppression for the low frequency microwave circuit packaging. The proposed 2.5-D SI-EBG unit cell consists of a circular patch and four via arrays. In this 2.5-D SI-EBG unit cell, via arrays are introduced to increase the capacitance and the inductance of the unit cell, which contributes to lower cavity mode suppression cutoff frequency. Dispersion diagram and mode analysis of the unit cell are given to explain its working mechanism. The electric size of the unit cell is reduced to $0.107{l} _{0}\times 0.107{l}_{0}\times 0.038{l}_{0}$ , and its stopband fractional bandwidth (FBW) is 42%. Finally, an experimental prototype of such miniaturized SI-EBG packaging structure is fabricated by PCB technique for a microstrip meander line to demonstrate its cavity mode suppression performance.

A half-air-filled substrate-integrated groove-gap waveguide is proposed for millimeter-wave and terahertz applications. The waveguide is constructed by a ball grid array using a flip-chip technique without metallic vias with lightweight and low profile. The possible leakage perpendicular to the groove is prevented by solder balls as modeled by an equivalent circuit. Besides, an inline transition between the proposed waveguide and a microstrip line is proposed, fabricated, and measured. The design concept and working mechanism are explained. Good agreement between simulation and measurement is observed. The simulation shows that the proposed waveguide can cover the entire ${W}$ -band (70–118 GHz), and the measurement demonstrates that the proposed transition can operate at 90–98.8 GHz.

Significance The radiant energy budget is a fundamental metric for planets. Based on the observations from multiple missions, we provide a global picture of Mars’ emitted power. Furthermore, we estimate the radiant energy budget of Mars, which suggests that there are energy imbalances at the time scale of Mars’ seasons. Such energy imbalances provide a new perspective to understanding the generating mechanism of dust storms. Mars’ radiant energy budget is assumed to be balanced at all time scales in current models and theories, but our analyses show that the energy budget is not balanced, at least at the time scale of Mars’ seasons. Therefore, current theories and models should be revisited with the newly revealed energy characteristics.

OBJECTIVE: To study the clinical efficacy of ultrasound-guided endoscopic retrograde appendicitis therapy in children with appendix-related chronic abdominal pain. METHODS: A retrospective analysis was performed on the medical data of 30 children with the chief complaint of chronic abdominal pain who were admitted from August 2019 to May 2021. All the children were found to have inflammation of the appendix or intracavitary stool and fecalith by ultrasound and underwent ultrasound-guided endoscopic retrograde appendicitis therapy. The medical data for analysis included clinical manifestations, endoscopic findings, white blood cell count, neutrophil percentage, length of hospital stay, and cure rate. RESULTS: Among the 30 children with chronic abdominal pain, there were 13 boys (43%) and 17 girls (57%), with a mean age of (9±3) years (range 3-15 years) at diagnosis. The median duration of the disease was 12 months, and the median length of hospital stay was 3 days. The children had a median white blood cell count of 6.7×10(9)/L and a neutrophil percentage of 50%±13%. Fecalith and a large amount of feces were flushed out of the appendix cavity for 21 children (70%) during surgery. The follow-up rate was 97% (29/30), and the median follow-up time was 11 months (range 5-26 months). Of the 29 children, abdominal pain completely disappeared in 27 children (93%). CONCLUSIONS: Ultrasound-guided endoscopic retrograde appendicitis therapy is effective in children with chronic abdominal pain caused by feces or fecalith in the appendix cavity.

Purpose Network analysis has been widely used in psychometrics over the past decade, yet it is unknown that whether this methodology could be applied in the field of child health assessment such as caregivers’ feeding behavior and child eating behavior. Our study leveraged network psychometrics method to estimating and examining the network structure of Chinese Preschoolers’ Caregivers’ Feeding Behavior Scale (CPCFBS), and compared the applicability of network methods in the feeding behavior scale. Methods The CPCFBS was previously applied in a sample of 768 preschoolers’ caregivers, used to estimate the structure of feeding behavior networks. Network structure was estimated with Gaussian Graphical Model. Dimensionality was detected using Exploratory Graph Analysis (EGA). The network structural consistency was tested using EGA bootstrap. The network structure was compared with the original structure using model fit indices and reliability. Results A seven-dimensional EGA network was explored after rearranging four items and deleting one item with unstable structural consistency. The absolute fit and relative fit of EGA structure were better than the original structure. The EGA structure had nearly same values of the reliability with the original structure. Conclusion Our study presented a novel perspective for feeding behavior analytical strategies, and demonstrated that network analysis was applicable and superior in exploring the structure of feeding behavior scales. Endoscopic retrograde appendicitis therapy (ERAT) is an emerging endoscopic treatment modality for acute uncomplicated appendicitis (AUA) supported by several case series. However, to date, systematic studies have not been conducted in children and the prospective comparative data are lacking. Moreover, due to a concern for future malignancy risk in children from ionizing radiation, we used contrast-enhanced ultrasound (CEUS) instead of endoscopic retrograde appendiceal radiography (ERAR). Therefore, we conducted a prospective, randomized control clinical trial to compare the modified ERAT (mERAT) to antibiotic therapy in children with AUA. The aim of this study was to evaluate the safety and feasibility and of mERAT in the treatment of hospitalized children with AUA. Children with AUA, confirmed by ultrasonography and or abdominal computed tomography, were consecutively enrolled from October 2018 to February, 2020. They were randomly assigned to receive mERAT or routine antibiotic treatment. Patients were followed until May, 2020. Th primary outcome variable was the duration of relief of the abdominal pain after treatment. We collected patient’s demographics, ultrasonic imaging findings, colonoscopy findings, and treatment outcomes of the mERAT and adverse even associated with mERAT. A total of 83 children were enrolled. 36 were randomized to mERAT and 47 to antibiotics treatment. All children in the mERAT group had endoscopic confirmed acute uncomplicated appendicitis, and there were no significant complications. However, 9 of patients in antibiotic group were poor responsive to treatment and switched to mERAT. The overall success rate of treatment with mERAT (100%) was significantly higher than that of antibiotics (80.9%) (P = 0.004). The median time to discharge was significantly shorter in mERAT group than in antibiotics treatment group [6.0 ± 1.76 days] (P = 0.004). mERAT provide a new alternative therapeutic option for childhood with AUA, especially for families who are reluctant to undergo an appendectomy.

Summary We develop a Bayesian design method for a clinical program where an investigational product is to be studied concurrently in a set of clinical trials involving related diseases with the goal of demonstrating superiority to a control in each. Mixture priors are constructed by eliciting conjugate priors based on pessimistic and enthusiastic predictions for the data to be observed for each disease and then by eliciting mixture weights for all possible configurations of the pessimistic and enthusiastic priors across the diseases to be studied. The proposed approach provides a robust framework for information borrowing in settings where the diseases may have endpoints based on different data types. We show via simulation that operating characteristics based on the proposed design framework are favorable compared to those based on information borrowing designs using the Bayesian hierarchical model which is poorly suited for information borrowing when there are different data types underpinning the endpoints across which information is to be borrowed.

Superconducting qubits provide a promising path toward building large-scale quantum computers. The simple and robust transmon qubit has been the leading platform, achieving multiple milestones. However, fault-tolerant quantum computing calls for qubit operations at error rates significantly lower than those exhibited in the state of the art. Consequently, alternative superconducting qubits with better error protection have attracted increasing interest. Among them, fluxonium is a particularly promising candidate, featuring large anharmonicity and long coherence times. Here, we engineer a fluxonium-based quantum processor that integrates high qubit-coherence, fast frequency-tunability, and individual-qubit addressability for reset, readout, and gates. With simple and fast gate schemes, we achieve an average single-qubit gate fidelity of 99.97% and a two-qubit gate fidelity of up to 99.72%. This performance is comparable to the highest values reported in the literature of superconducting circuits. Thus our work, for the first time within the realm of superconducting qubits, reveals an approach toward fault-tolerant quantum computing that is alternative and competitive to the transmon system.

Dielectric loss is one of the major decoherence sources of superconducting qubits. Contemporary high-coherence superconducting qubits are formed by material systems mostly consisting of superconducting films on substrate with low dielectric loss, where the loss mainly originates from the surfaces and interfaces. Among the multiple candidates for material systems, a combination of titanium nitride (TiN) film and sapphire substrate has good potential because of its chemical stability against oxidization, and high quality at interfaces. In this work, we report a TiN film deposited onto sapphire substrate achieving low dielectric loss at the material interface. Through the systematic characterizations of a series of transmon qubits fabricated with identical batches of TiN base layers, but different geometries of qubit shunting capacitors with various participation ratios of the material interface, we quantitatively extract the loss tangent value at the substrate-metal interface smaller than $8.9 \times 10^{-4}$ in 1-nm disordered layer. By optimizing the interface participation ratio of the transmon qubit, we reproducibly achieve qubit lifetimes of up to 300 $\mu$s and quality factors approaching 8 million. We demonstrate that TiN film on sapphire substrate is an ideal material system for high-coherence superconducting qubits. Our analyses further suggest that the interface dielectric loss around the Josephson junction part of the circuit could be the dominant limitation of lifetimes for state-of-the-art transmon qubits.
