The purpose of the present study was to explore the therapeutic effect of paeoniflorin on BiPN and its possible mechanism.Methods: ELISA was used to measure the level of interleukin-6 (IL6) in the plasma of MM patients, and bioinformatics analysis was used to predict the mechanism underlying the effect of paeoniflorin on peripheral neuropathy. Cell and animal models of BiPN were constructed to evaluate mitochondrial function by measuring cell viability and mitochondrial quality and labeling mitochondria with MitoTracker Green. Nerve injury in mice with BiPN was assessed by behavioral tests, evaluation of motor nerve conduction velocity, hematoxylin-eosin (HE) staining, electron microscopy and analysis of the levels of reactive oxygen species (ROS). Western blotting and immunohistochemistry (IHC) were used to assess the expression of autophagy-related proteins.Results: In MM patients, IL6 levels were positively correlated with the degree of PN. The results of bioinformatics analysis suggested that paeoniflorin ameliorated PN by altering inflammation levels and mitochondrial autophagy. Paeoniflorin increased PC12 cell viability and mitochondrial autophagy levels, alleviated mitochondrial damage, and reduced IL6 levels. In addition, paeoniflorin effectively improved the behavior of mice with BiPN, relieved sciatic nerve injury in mice, increased the expression of LC3II/I, beclin-1, and Parkin in sciatic nerve cells, and increased the expression of LC3B and Parkin in the nerve tissue.Conclusion: The present study confirmed that paeoniflorin significantly ameliorated peripheral neuropathy (PN) caused by bortezomib, possibly by reducing IL6 levels to regulate PARKIN-mediated mitochondrial autophagy and mitochondrial damage.Keywords: multiple myeloma, paeoniflorin, peripheral neuropathy, mitochondria, autophagy

Objective To summarize the effect of adding Lactobacillus reuteri in the treatment plan for diarrheal disease in children, and analyze the potential of probiotics in preventing the occurrence of diarrheal disease. Methods Search for randomized controlled trials of Lactobacillus reuteri for the treatment and prevention of diarrhea in the Pubmed, Web of science, Medline, and Cochrane databases. Data such as the number of diarrhea patients, time, length of stay, clinical symptoms and effect of diarrhea prevention were extracted for meta-analysis. Relative risk and confidence interval (RR and 95% CI) were used as outcome indicators. Results 963 participants in the 9 RCTs came from multiple countries/regions. Compared with placebo/no intervention, the number of diarrhea patients in the Lactobacillus reuteri group was significantly reduced on the day 1 (RR = 0.87, 95%CI: 0.78–0.97) and day 2 (RR = 0.61, 95%CI: 0.44–0.83). Cumulative statistics analysis showed that the effect was stable and significant starting on the 4th day after treatment. A few studies have shown that Lactobacillus reuteri can reduce the time of diarrhea, the number of days with watery stools, and days of hospital stay. However, it has no effect on the occurrence of nosocomial diarrhea (RR = 1.11, 95%CI: 0.68–1.83), rotavirus diarrhea (RR = 1.46, 95%CI: 0.78–2.72), antibiotic-related diarrhea (RR = 1.76, 95%CI: 0.77–4.05), and diarrhea (RR = 1.35, 95%CI: 0.95–1.92). Conclusion Addition of Lactobacillus reuteri in the treatment plan has a significant effect on reducing the number of diarrhea and reducing the symptoms of diarrhea, but has no obvious effect on the prevention of diarrhea. Combining probiotics and improving the ability of probiotics to respond is the focus of attention.

Purpose:Gliomas are brain tumors with dismal prognoses. The standard-of-care treatments for gliomas include surgical resection, radiation, and temozolomide administration; however, they have been ineffective in providing significant increases in median survival. Antigen-specific cancer vaccines and immune checkpoint blockade may provide promising immunotherapeutic approaches for gliomas.Experimental Design:We have developed immunotherapy delivery vehicles based on synthetic high-density lipoprotein (sHDL) loaded with CpG, a Toll-like receptor-9 agonist, and tumor-specific neoantigens to target gliomas and elicit immune-mediated tumor regression.Results:We demonstrate that vaccination with neoantigen peptide-sHDL/CpG cocktail in combination with anti–PD-L1 immune checkpoint blocker elicits robust neoantigen-specific T-cell responses against GL261 cells and eliminated established orthotopic GL261 glioma in 33% of mice. Mice remained tumor free upon tumor cell rechallenge in the contralateral hemisphere, indicating the development of immunologic memory. Moreover, in a genetically engineered murine model of orthotopic mutant IDH1 (mIDH1) glioma, sHDL vaccination with mIDH1 neoantigen eliminated glioma in 30% of animals and significantly extended the animal survival, demonstrating the versatility of our approach in multiple glioma models.Conclusions:Overall, our strategy provides a general roadmap for combination immunotherapy against gliomas and other cancer types.

A bifactor model with a general (p) factor reflecting shared variance and specific factors reflecting additional variance in individual symptoms has been introduced to explain common co-occurrence among anxiety, depression and schizotypy. However, longitudinal evidence is lacking and the validity of bifactor modeling is debatable. The current study aimed to examine the presence of the p factor together with specific factors in accounting for relationships between anxiety, depression and schizotypy both cross-sectionally and longitudinally, and to investigate the relationship between these factors and rumination. A validated sample of university students were surveyed on levels of anxiety, depression, schizotypy and rumination at baseline (N = 2291), one year (N = 1833) and two years (N = 1656). Models were estimated using exploratory structural equation modeling (ESEM) and compared at each time point. Longitudinal invariance of the best-fitting model was examined and all potential within- and between-factor stability pathways were tested in an SEM framework. A bifactor model with a p factor and four specific factors (representing residual information of composite anxiety and depression, cognitive-perceptual, interpersonal and disorganized schizotypy respectively) consistently outperformed a correlated-factors model. The bifactor structure appeared longitudinally stable. Within-factor stabilities were moderate, and between-factor pathways reflected a few significant interactions, mostly involving the p factor. Rumination was independently associated with p and four specific factors at each time point. Therefore, there is a p factor accounting for concurrent and sequential co-occurrence of anxiety, depression and schizotypy. Rumination explained partly the p and specific factors. Transdiagnostic interventions should target rumination.

An amphoteric compound of betaine is demonstrated as an electrolyte additive for aqueous zinc batteries. It forms hydrogen bonds with water and preferentially adsorbs on the Zn surface, which inhibits side reactions and ensures uniform Zn deposition.

In order to predict the temperature change of Laoshan scenic area in Qingdao more accurately, a new back propagation neural network (BPNN) prediction model is proposed in this study. Temperature change affects our lives in various ways. The challenge that neural networks tend to fall into local optima needs to be addressed to increase the accuracy of temperature prediction. In this research, we used an improved genetic algorithm (GA) to optimize the weights and thresholds of BPNN to solve this problem. The prediction results of BPNN and GA-BPNN were compared, and the prediction results showed that the prediction performance of GA-BPNN was much better. Furthermore, a screening test experiment was conducted using GA-BPNN for multiple classes of meteorological parameters, and a smaller number of parameter sets were identified to simplify the prediction inputs. The values of running time, root mean square error, and mean absolute error of GA-BPNN are better than those of BPNN through the calculation and analysis of evaluation metrics. This study will contribute to a certain extent to improve the accuracy and efficiency of temperature prediction in the Laoshan landscape.

DNA-wrapped single walled carbon nanotube (SWNT) conjugates have distinct optical properties leading to their use in biosensing and imaging applications. A critical limitation in the development of DNA-SWNT sensors is the current inability to predict unique DNA sequences that confer a strong analyte-specific optical response to these sensors. Here, near-infrared (nIR) fluorescence response data sets for ∼100 DNA-SWNT conjugates, narrowed down by a selective evolution protocol starting from a pool of ∼10sup10/supunique DNA-SWNT candidates, are used to train machine learning (ML) models to predict DNA sequences with strong optical response to neurotransmitter serotonin. First, classifier models based on convolutional neural networks (CNN) are trained on sequence features to classify DNA ligands as either high response or low response to serotonin. Second, support vector machine (SVM) regression models are trained to predict relative optical response values for DNA sequences. Finally, we demonstrate with validation experiments that integrating the predictions of ensembles of the highest quality neural network classifiers (convolutional or artificial) and SVM regression models leads to the best predictions of both high and low response sequences. With our ML approaches, we discovered five DNA-SWNT sensors with higher fluorescence intensity response to serotonin than obtained previously. Overall, the explored ML approaches, shown to predict useful DNA sequences, can be used for discovery of DNA-based sensors and nanobiotechnologies.

The role of the intermolecular H-bond in organic semiconductors is paradoxical, it can inhibit thermal fluctuation, enhance the charge transport stability, but can also affect the crystal growth morphology and thus reduce the device performance.

Multi-colored pigments based on rare earth (RE) with the formula of LiRE(MoO4+δ)2 (RE = Ce, Pr, Nd, Er) were developed via the solid-state reaction method. Related characterization techniques were used for investigating the crystal structure, morphology characteristic, reflecting property, and color performance of the synthesized pigment. The prepared pigment of LiRE(MoO4+δ)2 (RE = Ce, Pr, Nd, Er) shows four colors, namely orange-yellow (Ce), bright green (Pr), lavender (Nd) and pink (Er). Moreover, all of the pigments possess high NIR reflectance in the range of 95–98% at 1100 nm. Remarkably, the average NIR reflectance of LiCe(MoO4+δ)2 pigment reaches up to 95.28%, and the NIR solar reflectance is also up to 95.02%. Besides, the pigment of LiCe(MoO4+δ)2 exhibits good yellow hue (b* = 75.08). The application practical of LiCe(MoO4+δ)2 coated on galvanized sheet exhibits excellent coloring performance. The designed pigments show chemical stabilities in the acid/alkali testing. All of above, the superior performances of color and reflectance from LiRE(MoO4+δ)2 provide visual enjoyment for people and exhibit potential for energy savings.

Ion-absorbed rare earth ores radioactive residues (IREORR) are a class of waste residue from the production of rare earth elements (REEs). Because of its radioactive dose, IREORR are usually stored in waste warehouses. IREORR are difficult to be disposed of. However, it contains relatively high concentrations of REEs, which can be considered as a valuable secondary resource. In this paper, a novel process is developed for the separation of thorium (Th) and recovery of REEs from IREORR hydrochloric acid leachate with primary amine N1923 and Cyanex® 572, respectively. The effects of sulfate concentration, extractant concentration and pH on N1923 extraction in hydrochloric acid solution were investigated in detail. The results show that the extraction capacity of N1923 can be improved by adding sulfate to the solution and increasing the concentration of N1923. Acidity has little effect on the extraction of Th when pH is higher than 1. As for the stripping, REEs are more easily stripped from loaded organic phase than Th, and nitric acid is a better stripping agent than hydrochloric acid. Combined with the extraction of Cyanex® 572 for REEs, a fractional extraction experiment for separating Th and enriching of REEs was performed. The yield of Th is higher than 99.9% and the concentration of REEs is enriched to 183.84 g/L.

Nutritional metal ions play critical roles in many important immune processes. Hence, the effective modulation of metal ions may open up new forms of immunotherapy, termed as metalloimmunotherapy. Here, we demonstrate a prototype of cancer metalloimmunotherapy using cyclic dinucleotide (CDN) stimulator of interferon genes (STING) agonists and Mn2+. We screened various metal ions and discovered specific metal ions augmented STING agonist activity, wherein Mn2+ promoted a 12- to 77-fold potentiation effect across the prevalent human STING haplotypes. Notably, Mn2+ coordinated with CDN STING agonists to self-assemble into a nanoparticle (CDN–Mn2+ particle, CMP) that effectively delivered STING agonists to immune cells. The CMP, administered either by local intratumoural or systemic intravenous injection, initiated robust anti-tumour immunity, achieving remarkable therapeutic efficacy with minute doses of STING agonists in multiple murine tumour models. Overall, the CMP offers a new platform for local and systemic cancer treatments, and this work underscores the great potential of coordination nanomedicine for metalloimmunotherapy. The stimulation of interferon genes (STING) pathway with STING agonists such as cyclic dinucleotides (CDNs) has emerged as a promising immunotherapeutic approach. Here, the authors show that Mn2+ can amplify the STING-promoted anti-tumour immune response in challenging murine tumour models by coordinating with CDNs and self-assembling into nanoparticles that can be delivered locally and systemically.
