Daphnia magna belongs to the Cladocera order and plays an important role in the aquatic ecosystem. With the intensification of water pollution, the wild population of D. magna has declined rapidly in recent years, and insufficient molecular markers have limited effective research and conservation of this species.26 novel microsatellite (SSR) markers were developed in an artificially domesticated D. magna and 12 wild D. magna populations using restriction site-associated DNA sequencing (RAD-seq). The results showed that the observed heterozygosity (Ho) and expected heterozygosity (He) ranged from 0.083 to 0.999 and 0.085 to 0.862, respectively. The PIC ranged from 0.368 to 0.805. These results indicate that the developed SSR marker is highly polymorphic. Nei's genetic identity (H) ranged from 0.0926 to 0.3462. Shannon's Information index (I) ranged from 0.1333 to 0.4799. Genetic distance and Nei's genetic identity analysis, NJ tree diagram analysis, and PCoA analysis were conducted on populations of D. magna from different regions. The results show that the D. magna genetic relationship between Liaoning and Shanxi, Hunan and Anhui, and Beijing and Hainan are relatively close, while the genetic structure of D. magna in Guangdong, Jiangsu, and Sichuan is quite different from other sampling sites. An analysis of population genetic structure divided the D. magna samples into two major groups.These results indicate that the genetic structure of D. magna differs considerably in different regions. Our research results and the newly developed polymorphic SSR markers for D. magna are of great significance in terms of the genetic breeding of D. magna, identification of wild and artificially domesticated populations and conservation genetics research.

This paper studies the user selection problem for a cooperative nonorthogonal multiple access (NOMA) system consisting of a base station, a far user, and N near users. The selected near user receives its own message and assists the far user by relaying the far user’s message. Firstly, we propose a user selection strategy to maximize the selected near user’s data rate while satisfying the quality-of-service (QoS) requirement of the far user. Considering that the channel state information (CSI) of users in actual communication is usually imperfect, we then analyze the outage probability of the NOMA system based on the user selection strategy under imperfect CSI and obtain a closed-form expression. The theoretical analysis shows that the diversity order of the NOMA system under imperfect CSI is 0, which means the multiuser diversity order disappears. In order to improve the impact of imperfect CSI on system performance, we use the deep learning method to identify and classify channels of imperfect CSI and improve the accuracy of CSI. The simulation results show that the theoretical analysis of outage performance is consistent with the numerical results. Compared with the strategy without the deep learning method, the proposed deep learning-based user selection scheme significantly improves the system performance. Furthermore, we verify that our scheme recovers the diversity gain.

Neutrophils are the first defenders of the innate system for injury and infection. They have gradually been recognized as important participants in tumor initiation and development due to their heterogeneity and plasticity. In the tumor microenvironment (TME), neutrophils can exert antitumor and protumor functions, depending on the surroundings. Tumor cells systemically alter intracellular amino acid (AA) metabolism and extracellular AA distribution to meet their proliferation need, leading to metabolic reprogramming and TME reshaping. However, the underlying mechanisms that determine how altered AAs affect neutrophils in TME are less-explored. Here, we identified that abundant glutamate releasing from tumor cells blunted neutrophils' cell-killing effects toward tumor cells in vitro and in vivo. Mass spectrometric detection, flow cytometry, and western blot experiments proved that increased levels of pSTAT3/RAB10/ARF4, mediated by glutamate, were accompanied with immunosuppressive phenotypes of neutrophils in TME. We also discovered that riluzole, an FDA-approved glutamate release inhibitor, significantly inhibited tumor growth by restoring neutrophils' cell-killing effects and decreasing glutamate secretion from tumor cells. These findings highlight the importance of tumor-released glutamate on neutrophil transformation in TME, providing new possible cancer treatments targeting altered glutamate metabolism.

Bi-based electrocatalysts have been widely investigated in the CO

Electrochemically converting nitrate to ammonia is an essential and sustainable approach to restoring the globally perturbed nitrogen cycle. The rational design of catalysts for the nitrate reduction reaction (NO3RR) based on a detailed understanding of the reaction mechanism is of high significance. We report a Cu2O+Co3O4 tandem catalyst which enhances the NH3 production rate by ~2.7-fold com-pared to Co3O4 and ~7.5-fold compared with Cu2O, respectively. Most importantly, we precisely place single Cu2O and Co3O4 cube-shaped nanoparticles individually and together on carbon nano-electrodes provide insight into the mechanism of the tandem catalysis. The structural and phase evolution of the individual Cu2O+Co3O4 nanocubes during NO3RR is unveiled using identical location trans-mission electron microscopy. Combining single-entity electrochemis-try with precise nano-placement sheds light on the dynamic transfor-mation of single catalyst particles during tandem catalysis in a direct way.

SUMMARYV2-glycan/apex broadly neutralizing antibodies (bnAbs) recognize a closed quaternary epitope of the HIV-1 envelope glycoprotein (Env). This closed structure is necessary to elicit apex antibodies and useful to guide maturation of other bnAb classes. To compare antigens designed to maintain this conformation, apex-specific responses were monitored in mice engrafted with a diverse repertoire of B cells expressing the HCDR3 of the apex bnAb VRC26.25. Engineered B cells affinity matured, guiding improvement of VRC26.25 itself. We found that soluble Env (SOSIP) variants differed significantly in their ability to raise anti-apex responses. A transmembrane SOSIP (SOSIP-TM) delivered as an mRNA-lipid nanoparticle elicited more potent neutralizing responses than multimerized SOSIP proteins. Importantly, SOSIP-TM elicited neutralizing sera from B cells engineered with the predicted VRC26.25-HCDR3 progenitor, which also affinity matured. Our data show that HCDR3-edited B cells facilitate efficientin vivocomparisons of Env antigens and highlight the potential of an HCDR3-focused vaccine approach.

Proton exchange membrane fuel cells (PEMFCs) are regarded as one of the promising new carbon mitigation strategies to realize carbon neutrality. However, efficient and robust electrocatalysts are vital for the commercialization of PEMFCs. Herein, three commercial Pt/C electrocatalysts were investigated including a carbon support and Pt nanoparticles (NPs) to identify their merits and disadvantages, which will help end users quickly select catalysts with excellent performances among the many brands of domestic and foreign catalysts to further better study and better utilize them. Subsequently, they were optimized for real automotive application for about 1800 h, and then the variations in the electrocatalysts on the MEA were analysed by transmission electron microscopy (TEM) and X-ray photoelectron spectroscopy (XPS). The mean Pt particle size of the catalysts after operating for 1800 h (cathode, 9.9 ± 3.2 nm) was nearly 4-fold that before use (2.5 ± 0.6 nm), greatly reducing the exposure of metal sites, which was due to the violent three-phase interfacial reaction (ORR) occurring at the cathode side. How to control the frequent occurrence of cyanobacteria, especially the outbreak of toxin-producing Microcystis aeruginosa, has been a subject of constant research. This investigation focused on the effect of Moina mongolica on restricting M. aeruginosa blooms under different variables (temperature, light intensity, and salinity) and its growth at the molecular level. The results of batch experiments showed that the range of M. mongolica feeding rates was from 4.02 ± 0.81 × 103~182.23 ± 5.37 × 103 cells/ind·h in the whole experiment, where the highest feeding rates of larva M. mongolica and adult M. mongolica were 133.21 ± 5.24 × 103 vs. 182.23 ± 5.37 × 103 cells/ind·h at 30 °C, 85.88 ± 0.44 × 103 vs. 143.15 ± 14.07 × 103 cells/ind·h at 3000 lx and 88.18 ± 0.32 × 103 vs. 84.49 ± 4.95 × 103 cells/ind·h at 0‰ salinity, respectively. The results of transcriptomics further demonstrated that the response of M. mongolica to M. aeruginosa toxicity was caused by the downregulation of relevant functional genes (cell components, cell processes, metabolic processes, and protein complexes) and related signaling pathways (apoptosis, phagosome, lysosome, ribosome, oxidative phosphorylation, amino and nucleoside sugar metabolism, and PPAR signaling pathways). The findings show that M. mongolica can be released to low-salinity lakes and coastal areas (the subtropic and temperate zones) to prevent and inhibit M. aeruginosa blooms in the early summer phase. Additionally, the results achieved by the investigation will provide the relevant technology for inhibiting cyanobacteria blooms because M. mongolica even resists the produced toxin by M. aeruginosa.

The efficient nondestructive assessment of quality and homogeneity for two-dimensional (2D) MoS2 is critically important to advance their practical applications. Here, we presented a rapid and large-area assessment method for visually evaluating the quality and uniformity of chemical vapor deposition (CVD)-grown MoS2 monolayers simply with conventional optical microscopes. This was achieved through one-pot adsorbing abundant sulfur particles selectively onto as-grown poorer-quality MoS2 monolayers in a CVD system without any additional treatment. We further revealed that this favorable adsorption of sulfur particles on MoS2 originated from their intrinsic higher-density sulfur vacancies. Based on unadsorbed MoS2 monolayers, superior performance field effect transistors with a mobility of ∼49 cm2 V-1 s-1 were constructed. Importantly, the assessment approach was noninvasive due to the all-vapor-phase and moderate adsorption-desorption process. Our work offers a new route for the performance and yield optimization of devices by quality assessment of 2D semiconductors prior to device fabrication.

Brain pericyte is unique and indispensable part of blood-brain barrier (BBB), and contributes to several pathological processes in traumatic brain injury (TBI). However, the cellular and molecular mechanisms about how pericyte is regulated in damaged brain are largely unknown. Here, we show that neutrophil extracellular traps (NETs) formation induces the appearance of CD11b+ pericytes post TBI. These CD11b+ pericytes subsets are characterized with increased permeability and pro-inflammatory profiles compared to CD11b- pericytes. Moreover, histones from NETs by Dectin-1 facilitate CD11b induction on brain pericytes in PKC-c-Jun dependent manner, conferring neuroinflammation and BBB dysfunction post TBI. These data indicate that "neutrophil-NETs-pericyte" and "histones-Dectin-1-CD11b" are possible mechanisms for pericyte’s activation and dysfunction. Targeting at NETs formation and Dectin-1 are promising ways for TBI treatments.

These research results and the newly developed polymorphic SSR markers for D. magna are of great significance in terms of the genetic breeding of D. magna, identification of wild and artificially domesticated species and conservation genetics research.

Metal oxides or sulfides are considered to be one of the most promising CO2 reduction reaction (CO2 RR) precatalysts, owing to their electrochemical conversion in situ into highly active electrocatalytic species. However, further improvement of the performance requires new tools to gain fine control over the composition of the active species and its structural features [e.g., grain boundaries (GBs) and undercoordinated sites (USs)], directly from a predesigned template material. Herein, we describe a novel electrochemically driven cation exchange (ED-CE) method that enables the conversion of a predesigned CoS2 template into a CO2 RR catalyst, Cu2 S. By means of ED-CE, the final Cu2 S catalyst inherits the original 3 D morphology of CoS2 , and preserves its high density of GBs. Additionally, the catalyst's phase structure, composition, and density of USs were precisely tuned, thus enabling rational design of active CO2 RR sites. The obtained Cu2 S catalyst achieved a CO2 -to-formate Faradaic efficiency of over 87 % and a record high activity (among reported Cu-based catalysts). Hence, this study opens the way for utilization of ED-CE reactions to design advanced electrocatalysts.

There is an on-going search for new earth-abundant electrocatalytic materials, suitable for replacing noble-metals as efficient accelerators of energy–conversion reactions. In this regard, over the last few years, metal–organic framework (MOF)-converted materials have demonstrated promising electrocatalytic properties. Nevertheless, the discovery of new catalytic materials requires development of methods combining high-throughput synthesis and electrochemical-activity screening. To do so, here we couple the synthetical and the analytical virtues of scanning electrochemical microscopy (SECM). Namely, we first utilized an SECM tip electrode to induce spatially confined (μm-scale) electrochemical conversion of cobalt-based ZIF-67 MOFs into patterns of cobalt sulfide with a tuned chemical composition. In turn, the same SECM setup was used to map the H2 evolution activity of the as-formed cobalt sulfide. Hence, the presented method should have great implications for future screening of new electrocatalytic materials for a variety of energy-related applications.

The Ubp family of deubiquitinating enzymes has been found to play important roles in plant-pathogenic fungi, but their regulatory mechanisms are still largely unknown. In this study, we revealed the regulatory mechanism of the deubiquitinating enzyme Ubp3 during the infection process of Magnaporthe oryzae. AUBP3 deletion mutant was severely defective in appressorium turgor accumulation, leading to the impairment of appressorial penetration. During appressorium formation, the mutant was also defective in glycogen and lipid metabolism. Interestingly, we found that nitrogen starvation and rapamycin treatment induced the ribophagy process in M. oryzae, which is closely dependent on Ubp3. In the ∆ubp3 mutant, the ribosome proteins and rRNAs were not well degraded on nitrogen starvation and rapamycin treatment. We also found that Ubp3 interacted with the GTPase-activating protein Smo1 and regulated its de-ubiquitination. Ubp3-dependent de-ubiquitination of Smo1 may be required for Smo1 to coordinate Ras signalling. Taken together, our results showed at least two roles of Ubp3 in M. oryzae: it regulates the ribophagy process and it regulates de-ubiquitination of GTPase-activating protein Smo1 for appressorium-mediated infection.

Electrocatalytic recycling of waste nitrate (NO3−) to valuable ammonia (NH3) at ambient conditions is a green and appealing alternative to the Haber−Bosch process. However, the reaction requires multi-step electron and proton transfer, making it a grand challenge to drive high-rate NH3 synthesis in an energy-efficient way. Herein, we present a design concept of tandem catalysts, which involves coupling intermediate phases of different transition metals, existing at low applied overpotentials, as cooperative active sites that enable cascade NO3−-to-NH3 conversion, in turn avoiding the generally encountered scaling relations. We implement the concept by electrochemical transformation of Cu−Co binary sulfides into potential-dependent core−shell Cu/CuOx and Co/CoO phases. Electrochemical evaluation, kinetic studies, and in−situ Raman spectra reveal that the inner Cu/CuOx phases preferentially catalyze NO3− reduction to NO2−, which is rapidly reduced to NH3 at the nearby Co/CoO shell. This unique tandem catalyst system leads to a NO3−-to-NH3 Faradaic efficiency of 93.3 ± 2.1% in a wide range of NO3− concentrations at pH 13, a high NH3 yield rate of 1.17 mmol cm−2 h−1 in 0.1 M NO3− at −0.175 V vs. RHE, and a half-cell energy efficiency of ~36%, surpassing most previous reports.

The number of active sites and their intrinsic activity are key factors in designing high-performance catalysts for the oxygen evolution reaction (OER). The synthesis, properties, and in-depth characterization of a homogeneous CoNiFeCu catalyst are reported, demonstrating that multimetal synergistic effects improve the OER kinetics and the intrinsic activity. In situ carbon corrosion and Cu leaching during the OER lead to an enhanced electrochemically active surface area, providing favorable conditions for improved electronic interaction between the constituent metals. After activation, the catalyst exhibits excellent activity with a low overpotential of 291.5± 0.5mV at 10 mA cm−2 and a Tafel slope of 43.9 mV dec−1. It shows superior stability compared to RuO2 in 1 m KOH, which is even preserved for 120 h at 500 mA cm−2 in 7 m KOH at 50°C. Single particles of this CoNiFeCu after their placement on nanoelectrodes combined with identical location transmission electron microscopy before and after applying cyclic voltammetry are investigated. The improved catalytic performance is due to surface carbon corrosion and Cu leaching. The proposed catalyst design strategy combined with the unique single-nanoparticle technique contributes to the development and characterization of high-performance catalysts for electrochemical energy conversion.

The growing world population has produced an increasing demand for seafood, and the aquaculture industry is under corresponding pressure to fill this demand. The offshore ecology and environment are under significant threat with the continuous expansion of the scale and intensity of aquaculture. Integrated multi-tropic aquaculture (IMTA) is a healthy and sustainable mariculture model based on ecosystem-level management, and has become popular in recent years. It is an effective way to cope with the significant changes in offshore ecosystems under multiple stressors. Phytoplankton bacteria are essential to maintaining the marine ecosystem’s balance and stability. Investigating the changes in the community structure of marine planktonic bacteria can elucidate the impact of mariculture on the marine ecological environment. This study took the fish-shell IMTA system with natural macroalgae nearby as the object, and monitored the plankton community’s structure in the system’s surface seawater for four quarters from July 2020 to April 2021. The space–time distribution characteristics and influencing factors of the plankton community in the surface water were examined. The results showed no significant difference between the planktonic bacterial communities at different sampling sites. There was also no significant difference in the α-diversity index. However, the dominant species and abundance of planktonic bacteria at the sampling sites differed significantly. Proteobacteria and Bacteroides were the dominant groups of planktonic bacteria. The results of the distance-based redundancy analysis demonstrated that chemical oxygen demand, chlorophyll a, and dissolved oxygen constituted the primary environmental factors affecting the planktonic bacterial community structures. The heatmap also showed that NH4+- N, temperature, and salinity levels were also related to certain planktonic bacteria. This study preliminarily identified the distribution of the surface bacterial plankton community and its response to changes in environmental factors in the sea area near Xiasanhengshan Island. The results provide a preliminary basis for assessing the health and stability of the IMTA system in open sea areas.

Aims Greenhouses are widely used in agriculture systems to shield crops from unfavourable weather to achieve a year-round food supply. In recent years, aquaculture ponds have been placed in greenhouses in many regions. The impacts of the greenhouses on planktonic bacterial communities should be uncovered. Methods and Results In this study, two polyolefin film greenhouses accommodating aquaculture ponds were established and planktonic bacterial communities were compared from samples taken in aquaculture ponds inside and outside the greenhouses, using Illumina 16S rRNA sequencing. Conclusions The results showed there were significant variations in bacterial community structure between indoor and outdoor samples. Obvious differences were also found between two greenhouses, whereas the differences in indoor samples were weaker than outdoor samples. Significantly higher temperature (in summer), pH and permanganate index were found in the outdoor pond samples. Results of redundancy analysis showed that Proteobacteria and Bacteroidota were positively related to the dissolved oxygen, total nitrogen and total phosphorus, and Actinobacteriota were positively related to pH, temperature and permanganate index, whereas Cyanobacteria were positively related to the salinity, conductivity, total dissolved solids and ammonia nitrogen. Significance and Impact of the Study The results of this study revealed that greenhouses significantly influenced planktonic bacterial communities in aquaculture ponds. This study is expected to provide a scientific basis for aquaculture in greenhouses.

Endocytosis plays key roles during infection of plant‐pathogenic fungi, but its regulatory mechanisms are still largely unknown. Here, we identified a putative endocytosis‐related gene, PAL1, which was highly expressed in appressorium of Magnaporthe oryzae, and was found to be important for appressorium formation and maturation. Deletion of PAL1 significantly reduced the virulence of M. oryzae due to defects in appressorial penetration and invasive growth in host cells. The Pal1 protein interacted and colocalized with the endocytosis protein Sla1, suggesting it is involved in endocytosis. The Δpal1 mutant was significantly reduced in appressorium formation, which was recovered by adding exogenous cAMP and 3‐isobutyl‐1‐methylxanthine (IBMX). Moreover, the phosphorylation level of Pmk1 in Δpal1 was also reduced, suggesting Pal1 functions upstream of both the cAMP and Pmk1 signalling pathways. As a consequence, the utilization of glycogen and lipid, appressorial autophagy, actin ring formation, localization of septin proteins, as well as turgor accumulation were all affected in the Δpal1 mutant. Taken together, Pal1 regulates cAMP and the Pmk1 signalling pathway for appressorium formation and maturation to facilitate infection of M. oryzae., Endocytic protein Pal1 of Magnaporthe oryzae contributes to full pathogenicity by regulating cAMP and the Pmk1 signalling pathway for appressorium formation and maturation.

BackgroundBlood-brain barrier (BBB) disruption and leukocyte infiltration are two pathological features post traumatic brain injury (TBI). However, the role of circulating leukocytes in BBB disruption and the crosstalk between them are not fully elucidated. Neutrophil is the most abundant circulating cell type that migrates into brain tissue when TBI occurs instantly, while brain pericyte occupies a strategic position between circulating cell and interstitial space in BBB. Understanding their interactions is essential to provide insight into the intrinsic relationship and identify biological targets for TBI treatments. MethodsBy analyzing brain tissues from TBI patients and mouse TBI model through immunohistochemical method and flow cytometry, we build the relationship between neutrophils, neutrophil extracellular traps (NETs) and brain pericyte. The components of NETs-related medium were investigated by proteomics and metabolomics to decipher the factors directly regulating pericytes. The molecular mechanisms were deeply explored by WB/CHIP/RT-PCR in primary brain pericyte/pericyte cell line MBVP treated with NETs-formed medium or specific NETs components. In mice TBI model, we also explored the possible therapeutic approaches for TBI treatment that targeting at the axis of neutrophil-NETs-pericyte. ResultsNETs formation is highly enhanced post TBI, inducing the appearance of CD11b expressing brain pericyte simultaneously. This novel CD11b+ pericyte subset is characterized with increased permeability and pro-inflammatory profiles. Mechanistically, recognition of histones from NETs by Dectin-1 on pericyte contributes to CD11b induction in protein kinase C (PKC)-c-Jun-dependent manner. Transcription factor c-Jun directly binds to the promoter sequence of CD11b to enhance its expression in pericyte, conferring pericyte activation, BBB disruption and aggravated neutrophil infiltration post TBI. Either inhibiting NETs formation by Cl-Amidine or blocking Dectin-1 by Laminarin are both beneficial for decreasing neutrophil infiltration and brain pericyte activation post TBI. ConclusionsThese results unfold that “neutrophil-NETs-pericyte” and “histones-Dectin-1-CD11b” are possible cellular and molecular mechanisms for building connection between BBB damage and neutrophil infiltration. Targeting at NETs formation and Dectin-1 are promising treatments for improving neurological outcomes of TBI patients.
