Neuroinflammation caused by microglial activation and consequent neurological impairment are prominent features of diabetes-associated cognitive impairment (DACI). Microglial lipophagy, a significant fraction of autophagy contributing to lipid homeostasis and inflammation, had mostly been ignored in DACI. Microglial lipid droplets (LDs) accumulation is a characteristic of aging, however, little is known about the pathological role of microglial lipophagy and LDs in DACI. Therefore, we hypothesized that microglial lipophagy could be an Achilles’s heel exploitable to develop effective strategies for DACI therapy. Here, starting with characterization of microglial accumulation of LDs in leptin receptor-deficient (db/db) mice and in high-fat diet and STZ (HFD/STZ) induced T2DM mice, as well as in high-glucose (HG)-treated mice BV2, human HMC3 and primary mice microglia, we revealed that HG-dampened lipophagy was responsible for LDs accumulation in microglia. Mechanistically, accumulated LDs colocalized with the microglial specific inflammatory amplifier TREM1 (triggering receptor expressed on myeloid cells 1), resulting in the buildup of microglial TREM1, which in turn aggravates HG-induced lipophagy damage and subsequently promoted HG-induced neuroinflammatory cascades via NLRP3 (NLR family pyrin domain containing 3) inflammasome. Moreover, pharmacological blockade of TREM1 with LP17 in db/db mice and HFD/STZ mice inhibited accumulation of LDs and TREM1, reduced hippocampal neuronal inflammatory damage, and consequently improved cognitive functions. Taken together, these findings uncover a previously unappreciated mechanism of impaired lipophagy-induced TREM1 accumulation in microglia and neuroinflammation in DACI, suggesting its translational potential as an attractive therapeutic target for delaying diabetes-associated cognitive decline. ObjectiveThe purpose of this study was to evaluate the feasibility and outcomes of conduction system pacing (CSP) in patients with heart failure (HF) who had a severely reduced left ventricular ejection fraction (LVEF) of less than 30% (HFsrEF).MethodsBetween January 2018 and December 2020, all consecutive HF patients with LVEF ResultsSeventy patients (66 ± 8.84 years; 55.7% male) with a mean LVEF of 23.2 ± 3.23%, LVEDd of 67.33 ± 7.47 mm and LVESV of 212.08 ± 39.74 ml were included. QRS configuration at baseline was CLBBB in 67.1% (47/70) of patients and non-CLBBB in 32.9%. At implantation, the CSP threshold was 0.6 ± 0.3 V @ 0.4 ms and remained stable during a mean follow-up of 23.43 ± 11.44 months. CSP resulted in significant LVEF improvement from 23.2 ± 3.23% to 34.93 ± 10.34% (P P P = 0.045) was associated with echocardiographic response. The proportions of clinical and echocardiographic responses in the CLBBB group were higher than those in the non-CLBBB group but without significant statistical differences.ConclusionsCSP is feasible and safe in patients with HFsrEF. CSP is associated with a significant improvement in clinical and echocardiographic outcomes, even for patients with non-CLBBB widened QRS.

Grapevine is one of the most economically important crops worldwide. However, the previous versions of the grapevine reference genome consisted of thousands of fragments with missing centromeres and telomeres, which limited the accessibility of the repetitive sequences, the centromeric and telomeric regions, and the inheritance of important agronomic traits in these regions. Here, we assembled a telomere-to-telomere (T2T) gap-free reference genome for the pinot noir cultivar (PN40024) using the PacBio HiFi long reads. The T2T reference genome (PN_T2T) was 69 Mb longer with 9026 more genes identified than the 12X.v2 version (Canaguier et al., 2017). We annotated 67% repetitive sequences, 19 centromeres and 36 telomeres, and incorporated gene annotations of previous versions into the PN_T2T. We detected a total of 377 gene clusters, which showed associations with complex traits, such as aroma and disease resistance. Even though the PN40024 sample had been selfed for nine generations, we still found nine genomic hotspots of heterozygous sites associated with biological processes, such as the oxidation-reduction process and protein phosphorylation. The fully annotated complete reference genome, therefore, provides important resources for grapevine genetics and breeding. Introduction: The Bangong-Nujiang Suture Zone (BNSZ) in central Tibet is a remnant of the Bangong-Nujiang Ocean that records its entire Wilson Cycle. The model of divergent double-sided subduction (DDS) is crucial for elucidating the evolution of tectonomagmatic activity on both sides of the BNSZ and for understanding why no high-pressure metamorphic rocks occur in the BNSZ. However, the DDS geodynamics remain poorly constrained. In particular, there is a lack of reports on magmatic rocks directly associated with slab sinking in the DDS terminal stage.Methods: This study presents new geochronological, geochemical, and isotopic data for the Early Cretaceous bimodal volcanic rocks around the Wuga Co area.Results: The bimodal volcanic rocks are divided into the Wuga Co rhyolites (SiO2 = 77.0–79.0 wt%) and the Wuga Co basaltic andesites (SiO2 = 53.9–55.5 wt%). The isotopic values of the Wuga Co basaltic andesites with low (87Sr/86Sr)i values (+0.7040 to +0.7044) and high εNd(t) values (+3.8 to +4.1) lie among three endmembers (the BNO sediments in accretionary wedge, depleted mantle and the BNO slab).Discussion: These values indicate the partial melting of a mantle peridotite that interacted with the subducted slab and sediment in the accretionary wedge, which was caused by the sinking of the Bangong-Nujiang oceanic slab. The Wuga Co rhyolites (108 Ma) have low (87Sr/86Sr)i values (+0.703 to +0.706), high εNd(t) values (+2.25 to +2.49), and high εHf(t) values (+5.6 to +10.0). These values indicate that the rhyolite formed by partial melting of juvenile basaltic crust. This study also collected Hf isotope data from both sides of the BNSZ to constrain its evolution. Our results show that the εHf(t) values of magma on both sides of the BNSZ were elevated simultaneously at 130 Ma, which may be caused by the Bangong-Nujiang oceanic slab rupture. Based on these new data, we propose that the Bangong-Nujiang oceanic slab ruptured from the two overlying terranes at approximately 130 Ma and subsequently sank into the mantle at approximately 108 Ma.

Hydrogen peroxide (H2O2), an endogenous reactive oxygen species, may cause a series of physical and chemical processes and various diseases in excess in an organism. In this article, simple solvent stirring is reported to synthesize a europium-metal organic framework (Eu-MOF) at room temperature for the detection of H2O2. Eu-MOF emits characteristic red fluorescence, but with the addition of H2O2, significant quenching occurred due to the reduced energy transfer from organic ligand to Eu caused by electrophilic and nucleophilic properties of H2O2. Under the optimized conditions, Eu-MOF showed high sensitivity and selectivity with linearity (R2 > 0.99) from 1 to 150 μM and a limit of detection (LOD) of 0.65 μΜ and was employed for the determination of H2O2 in human serum. In addition, Eu-MOF based portable test strips were developed to monitor H2O2 content with the aid of a smartphone color analysis application, consuming only 5 μL of samples. Thus, the real-time determination of H2O2 was achieved utilizing this portable device.

The balance of protein production and consumption in muscles depends to a large extent on normal mitochondrial function. Mitochondrial dysfunction is inseparable from the occurrence of myopathy. This study explores the relationships between statin-induced myopathy, steroid myopathy, and skeletal muscle illness caused by alcohol addiction, as well as the relationship between these conditions and mitochondrial abnormalities.Web of Science (WOS) central archive was analyzed for alcoholic myopathy research papers from 1999 to 2021, CiteSpace and WOS databases were used for evaluation the number of written articles, distribution of publications by region, research organizations, co-cited lit-erature analysis, and keyword identification.A total of 1,255 publications were collected after screening, with the number of published articles continually increasing. The annual average number of publications is 54.56. Six countries publish the majority of the literature. The United States has published 383 papers in total, which places it first among all countries. It also has the most centrality, meaning that other countries value its scientific achievements more. There are 34 core authors and 238 papers published. Through cluster analysis, there are 9 categories that are significant clusters. Key words of co-occurrence research show that keywords such as nanoparticles, apoptosis, mitochondrial disorders, and inflammation are very common.

Through innovatively introducing the receding horizon into probabilistic model checking, an online strategy synthesis method for multi-robot systems from local automatons is proposed to complete complex tasks that are assigned to each robot. Firstly, each robot is modeled as a Markov decision process which models both probabilistic and nondeterministic behavior. Secondly, the task specification of each robot is expressed as a linear temporal logic formula. For some tasks that robots cannot complete by themselves, the collaboration requirements take the form of atomic proposition into the LTL specifications. And the LTL specifications are transformed to deterministic rabin automatons over which a task progression metric is defined to determine the local goal states in the finite-horizon product systems. Thirdly, two horizons are set to determine the running steps in automatons and MDPs. By dynamically building local finite-horizon product systems, the collaboration strategies are synthesized iteratively for each robot to satisfy the task specifications with maximum probability. Finally, through simulation experiments in an indoor environment, the results show that the method can synthesize correct strategies online for multi-robot systems which has no restriction on the LTL operators and reduce the computational burden brought by the automaton-based approach.

Visual features play a key role in indoor positioning and navigation services as the main semantic information to help people understand the environment. However, insufficient semantic constraint information and mismatching localization without building map have hindered the ubiquitous application services. To address the problem, we propose a smartphone indoor scene features recognition localization method with building map semantic constraints. First, based on Geographic Information System and Building Information Modeling techniques, a geocoded entity library of building Map Location Anchor (MLA) is constructed, which is able to provide users with “immersive” meta-building-map and semantic anchor constraints for mobile phone positioning when map matching. Second, using the MYOLOv5s deep learning model improved on indoor location scenario, the nine types of ubiquitous anchor features in building scenes are recognized in real time by acquiring video frames from the smartphone camera. Lastly, the spatial locations of the ubiquitous indoor facilities obtained using smartphone video recognition are matched with the MLA P3P algorithm to achieve real-time positioning and navigation. The experimental results show that the MLA recognition accuracy of the improved MYOLOv5s is 97.2%, and the maximum localization error is within the range of 0.775 m and confined to the interval of 0.5 m after applying the Building Information Modeling based Positioning and Navigation road network step node constraint, which can effectively achieve high positioning accuracy in the building indoor scenarios with adequate MLA and road network constraint.

DnaJs are the common molecular chaperone proteins with strong structural and functional diversity. In recent years, only several DnaJ family members have been found to be able to regulate leaf color, and it remains to be explored whether there are other potential members that also regulate this character. Here, we identified 88 putative DnaJ proteins from Catalpa bungei, and classified them into four types according to their domain. Gene-structure analysis revealed that each member of CbuDnaJ family had same or similar exon-intron structure. Chromosome mapping and collinearity analysis showed that tandem and fragment duplication occurred in the process of evolution. Promoter analyses suggested that CbuDnaJs might be involved in a variety of biological processes. The expression levels of DnaJ family members in different color leaves of Maiyuanjinqiu were respectively extracted from the differential transcriptome. Among these, CbuDnaJ49 was the largest differentially expressed gene between the green and yellow sectors. Ectopic overexpression of CbuDnaJ49 in tobacco showed that the positive transgenic seedlings exhibited albino leaves, and the contents of chlorophyll and carotenoid were significantly reduced compared with those of wild type. The results suggested that CbuDnaJ49 played an important role in regulating leaf color. This study not only identified a novel gene of DnaJ family members regulating leaf color, but also provided new germplasm for landscaping.

IntroductionDiscretionary salt (added in cooking at home or at the table) is a source of sodium and iodine in New Zealand. The amount of discretionary salt consumed in a population has implications on policies regarding sodium and iodine. Sodium intake from discretionary salt intake has not been quantified in New Zealand. The aim of this study was to estimate the proportion of total sodium that comes from discretionary salt in adults using the lithium-tagged salt method.MethodsA total of 116 healthy adults, who were not pregnant or breastfeeding, regularly consume home-cooked meals and use salt during cooking or at the table, aged 18–40 years from Dunedin, New Zealand were recruited into the study. The study took place over a 9-day period. On Day 1, participants were asked to collect a baseline 24-h urine to establish their normal lithium output. From Day 2 to Day 8, normal discretionary salt was replaced with lithium-tagged salt. Between Day 6 and Day 8, participants collected another two 24-h urine samples. A 24-h dietary recall was conducted to coincide with each of the final two 24-h urine collections. Urinary sodium was analysed by Ion-Selective Electrode and urinary lithium and urinary iodine were analysed using Inductively Coupled Plasma Mass Spectrometry. The 24-h dietary recall data was entered into Xyris FoodWorks 10. All statistical analysis were conducted using Stata 17.0.ResultsA total of 109 participants with complete 24-h urine samples were included in the analysis. From the 24-h urine collections, the median urinary excretion of sodium and iodine was 3,222 mg/24 h (25th, 75th percentile: 2516, 3969) and 112 μg/24 h (82, 134). The median estimated sodium intake from discretionary salt was 13% (25th, 75th percentile: 7, 22) of the total sodium intake or 366 mg/24 h (25th, 75th percentile: 186, 705).ConclusionThe total sodium intake was higher than the suggested dietary target of 2,000 mg/day. In this sample of healthy adults 18 to 40 years old, 13% of total sodium intake derived from discretionary salt. Discretionary salt is an additional source of iodine if iodised salt is used. Policies to reduce sodium intake is recommended to include a range of strategies to target discretionary and non-discretionary sources of salt and will need to take into account the contribution of iodine from discretionary salt intake.

There has a major problem in the current theory of hypothesis testing in which no unified indicator to evaluate the goodness of various test methods since the cost function or utility function usually relies on the specific application scenario, resulting in no optimal hypothesis testing method. In this paper, the problem of optimal hypothesis testing is investigated based on information theory. We propose an information-theoretic framework of hypothesis testing consisting of five parts: test information (TI) is proposed to evaluate the hypothesis testing, which depends on the a posteriori probability distribution function of hypotheses and independent of specific test methods; accuracy with the unit of bit is proposed to evaluate the degree of validity of specific test methods; the sampling a posteriori (SAP) probability test method is presented, which makes stochastic selections on the hypotheses according to the a posteriori probability distribution of the hypotheses; the probability of test failure is defined to reflect the probability of the failed decision is made; test theorem is proved that all accuracy lower than the TI is achievable. Specifically, for every accuracy lower than TI, there exists a test method with the probability of test failure tending to zero. Conversely, there is no test method whose accuracy is more than TI. Numerical simulations are performed to demonstrate that the SAP test is asymptotically optimal. In addition, the results show that the accuracy of the SAP test and the existing test methods, such as the maximum a posteriori probability, expected a posteriori probability, and median a posteriori probability tests, are not more than TI.

Phase change materials, which show different electrical characteristics across the phase transitions, have attracted considerable research attention for their potential electronic device applications. Materials with metal-to-insulator or charge density wave (CDW) transitions such as VO2 and 1T-TaS2 have demonstrated voltage oscillations due to their robust bi-state resistive switching behavior with some basic neuronal characteristics. BaTiS3 is a small bandgap ternary chalcogenide that has recently reported the emergence CDW order below 245 K. Here, we report on the discovery of DC voltage / current-induced reversible threshold switching in BaTiS3 devices between a CDW phase and a room temperature semiconducting phase. The resistive switching behavior is consistent with a Joule heating scheme and sustained voltage oscillations with a frequency of up to 1 kHz has been demonstrated by leveraging the CDW phase transition and the associated negative differential resistance. Strategies of reducing channel sizes and improving thermal management may further improve the device performance. Our findings establish BaTiS3 as a promising CDW material for future energy-efficient electronics, especially for neuromorphic computing.

APT detection is difficult to detect due to the long-term latency, covert and slow multistage attack patterns of Advanced Persistent Threat (APT). To tackle these issues, we propose TBDetector, a transformer-based advanced persistent threat detection method for APT attack detection. Considering that provenance graphs provide rich historical information and have the powerful attacks historic correlation ability to identify anomalous activities, TBDetector employs provenance analysis for APT detection, which summarizes long-running system execution with space efficiency and utilizes transformer with self-attention based encoder-decoder to extract long-term contextual features of system states to detect slow-acting attacks. Furthermore, we further introduce anomaly scores to investigate the anomaly of different system states, where each state is calculated with an anomaly score corresponding to its similarity score and isolation score. To evaluate the effectiveness of the proposed method, we have conducted experiments on five public datasets, i.e., streamspot, cadets, shellshock, clearscope, and wget_baseline. Exploring the origin of the pseudogap is important for the understanding of superconductivity in cuprates. Here we report a systematical experimental study on the phonon vibrational properties of Tl$_2$Ba$_2$Ca$_{n-1}$Cu$_n$O$_{2n+4+δ}$ ($n$=1,2,3) single crystals based on the Raman scattering measurements over the temperature range from 10 to 300 K. The temperature evolution of the frequency and linewidth of the observed phonon modes in each member of this family does not follow the expected self-energy effect when entering the superconducting state. Instead, these phonon modes exhibit a universal softening behavior below the temperature around 150 K, which is higher above the superconducting transition. From the comparison with the existing experimental data for various orders, we find that the observed starting temperature for the phonon softening corresponds to the onset opening temperature of the pseudogap. This finding indicates a large lattice effect in the pseudogap state and the non-negligible spin-phonon coupling for such a phonon softening., 12 pages, 6 figures

Deep learning methods have advanced quickly in brain imaging analysis over the past few years, but they are usually restricted by the limited labeled data. Pre-trained model on unlabeled data has presented promising improvement in feature learning in many domains, including natural language processing and computer vision. However, this technique is under-explored in brain network analysis. In this paper, we focused on pre-training methods with Transformer networks to leverage existing unlabeled data for brain functional network classification. First, we proposed a Transformer-based neural network, named as BrainNPT, for brain functional network classification. The proposed method leveraged token as a classification embedding vector for the Transformer model to effectively capture the representation of brain network. Second, we proposed a pre-training framework for BrainNPT model to leverage unlabeled brain network data to learn the structure information of brain networks. The results of classification experiments demonstrated the BrainNPT model without pre-training achieved the best performance with the state-of-the-art models, and the BrainNPT model with pre-training strongly outperformed the state-of-the-art models. The pre-training BrainNPT model improved 8.75% of accuracy compared with the model without pre-training. We performed an umbrella meta-analysis to explore the factors that influence the efficacy of immune checkpoint inhibitor (ICI) therapy. We systematically searched three databases (PubMed, Web of Science and Embase) up to 20 February 2023. Extracting the effect size and 95% confidence intervals for overall survival (OS), progression-free survival (PFS) and the objective response rate (ORR). A total of 65 articles were included. We identified the following factors that benefit ICI therapy: smoking status (PFS: 0.72 [0.62, 0.84], p < .001), chemotherapy (PFS: 0.68 [0.58, 0.79], p < .001), expression of programmed cell death ligand 1(PD-L1) (≥1%, ≥5%, or ≥10%) (≥1%: 0.76 [0.71,0.82], p < .001; ≥5%: 0.62 [0.52, 0.74], p < .001; ≥10%: 0.42 [0.30, 0.59], p < .001). In this paper, we propose a practically efficient model for securely computing rank-based statistics, e.g., median, percentiles and quartiles, over distributed datasets in the malicious setting without leaking individual data privacy. Based on the binary search technique of Aggarwal et al. (EUROCRYPT \textquotesingle 04), we respectively present an interactive protocol and a non-interactive protocol, involving at most $\log ||R||$ rounds, where $||R||$ is the range size of the dataset elements. Besides, we introduce a series of optimisation techniques to reduce the round complexity. Our computing model is modular and can be instantiated with either homomorphic encryption or secret-sharing schemes. Compared to the state-of-the-art solutions, it provides stronger security and privacy while maintaining high efficiency and accuracy. Unlike differential-privacy-based solutions, it does not suffer a trade-off between accuracy and privacy. On the other hand, it only involves $O(N \log ||R||)$ time complexity, which is far more efficient than those bitwise-comparison-based solutions with $O(N^2\log ||R||)$ time complexity, where $N$ is the dataset size. Finally, we provide a UC-secure instantiation with the threshold Paillier cryptosystem and $\Sigma$-protocol zero-knowledge proofs of knowledge.

WRKY transcription factors (TF) control a variety of biological processes in higher plants, including abiotic and biotic stress responses. WRKY genes have been identified and functionally characterized in many plant species during the past ten years, but despite this progress, few investigations have focused on this TF family in licorice (Glycyrrhiza uralensis Fisch.). This study identified 72 WRKY genes from the licorice genome and classified them into three groups. The gene structure analysis indicated that the GuWRKY genes contain from one to eight introns, with the majority having two introns and three exons. Conserved motif analysis identified the conserved WRKY domain in most GuWRKYs. Further analysis of the promoter region revealed that almost all GuWRKYs had plant hormone- or stress-responsive cis-regulatory elements, with differences in the number of elements among the groups. The role of 10 selected GuWRKYs in regulating abiotic stress responses was further explored via quantitative real-time PCR (qRT-PCR) after low-temperature (4 °C), salt (150 mmol·L−1 NaCl), drought (15% polyethylene glycol 6000) treatments at 3, 6, 12, and 24 h. Thus, our study of the WRKY gene family in licorice provides a basis for further research on the structure and function of WRKY genes.

Objective: The generalizability of clinical large language models is usually ignored during the model development process. This study evaluated the generalizability of BERT-based clinical NLP models across different clinical settings through a breast cancer phenotype extraction task. Materials and Methods: Two clinical corpora of breast cancer patients were collected from the electronic health records from the University of Minnesota and the Mayo Clinic, and annotated following the same guideline. We developed three types of NLP models (i.e., conditional random field, bi-directional long short-term memory and CancerBERT) to extract cancer phenotypes from clinical texts. The models were evaluated for their generalizability on different test sets with different learning strategies (model transfer vs. locally trained). The entity coverage score was assessed with their association with the model performances. Results: We manually annotated 200 and 161 clinical documents at UMN and MC, respectively. The corpora of the two institutes were found to have higher similarity between the target entities than the overall corpora. The CancerBERT models obtained the best performances among the independent test sets from two clinical institutes and the permutation test set. The CancerBERT model developed in one institute and further fine-tuned in another institute achieved reasonable performance compared to the model developed on local data (micro-F1: 0.925 vs 0.932). Conclusions: The results indicate the CancerBERT model has the best learning ability and generalizability among the three types of clinical NLP models. To date, empiric therapy for patients with unexplained recurrent pregnancy loss (URPL) is not precise. Studies have shown that URSAs are associated with Th1/Th2 and Th17/Treg immune imbalances at the maternal-fetal interface. The immunosuppressant cyclosporine A (CsA) is widely used in patients with organ transplantation or autoimmune diseases, and it has a good safety profile in pregnant women. However, high-quality evidence for CsA treatment of URSAs is lacking. Our purpose with this study is to evaluate the efficacy and safety of CsA for improving pregnancy outcomes in patients with URSAs and to explore the role of CsA in regulating the immune balance.Methods/Design: We expect to officially initiate our study at the Taizhou People’s Hospital in March 2022. We defined the live birth rate as the primary outcome, and the secondary outcomes include the rates of successful pregnancy, miscarriage, pregnancy complications, and adverse pregnancy outcomes, and newborn birth weights. Patients who meet URSA eligibility criteria will be randomized in a 1:1 ratio into either a study group receiving CsA 2 weeks after fertilization or a control group receiving placebo at 2 weeks after fertilization (the women in both groups will receive the relevant treatment for 6 months). In addition, we will collect peripheral blood samples of the participants before and after the treatments, and we will isolate mononuclear cells and measure cytokine levels (IFN-γ, TNF-α, IL-2, IL-10, IL-6, IL-4) and Th1/Th2, Th17/Treg ratios.Discussion: This is the first randomized controlled trial to evaluate the clinical and immunomodulatory effects of CsA on the pregnancy outcomes of women with URSA and our results will provide evidence to evaluate the use of CsA as a treatment for women with URSAs.Keywords: cyclosporine A, live birth rate, recurrent abortion of unknown cause, Th1, Th2

Surface modification of cellulose nanocrystals (CNC) is essential for improving their reactivity and adsorption capacity. Oxidation, as a conventional modification method of CNC, has strong reaction conditions, which may partially destroy the structure of CNCs. Herein, we propose a mild cellulose modification method, in which diethylenetriaminepentaacetic acid (DPTA) was grafted onto CNC with an aminosilane coupling agent. To facilitate the enrichment and recovery of adsorbents, we prepared magnetic CNC before grafting DPTA, and finally obtained magnetic carboxylic functionalized CNC (MCNC-DPTA). The effects of the reaction parameters on the adsorption process were systematically studied by transmission electron microscopy, X-ray diffraction, Vibration sample magnetometry, Fourier transform infrared spectroscopy, and X-ray photoelectron spectroscopy. Additionally, the effects of the solution pH, the initial concentration of Pb, and the adsorption time on the adsorption properties of the materials were evaluated. Furthermore, the structure, morphology, and thermal properties of the solution were systematically studied. After 6 cycle tests, the adsorption amount of Pb(II) by MCNC-DPTA was only reduced by 7%. These results show that MCNC-DPTA is an environment-friendly adsorption material with an excellent Pb(II) removal effect.

Aims: This study investigated the feasibility and accuracy of real-time three-dimensional (3D) echocardiographic transilluminated imaging (TrueVue Glass) in left atrial appendage (LAA) anatomical morphology and artificial intelligence (AI)-assisted 3D automated LAA measurement (3D Auto LAA) software in the preoperative evaluation of LAA occlusion (LAAO) in patients with atrial fibrillation (AF).Method and results: Thirty-seven patients with AF were selected. Two-dimensional (2D) and real-time 3D transesophageal echocardiography (RT3D-TEE) were performed preoperatively, using conventional 3D, the new 3D TrueVue Glass mode, and cardiac computed tomography angiography (CCTA) to assess and type the morphology of LAA. Physiological parameters were measured using traditional 2D and 3D manual (3D Manual LAA), 3D Auto LAA, and CCTA. TrueVue Glass for LAA outer contour display was compared with CCTA. Comparisons were based on correlation and consistency in measuring the maximum diameter (LZ max), minimum diameter (LZ min), area (LZ area), and circumference (LZ cir) of LAA landing zone (LZ). Times and variabilities were compared. The concordance rate for external shape of LAA was 97.14% between TrueVue Glass and CCTA. 3D Auto LAA and 3D Manual LAA have a stronger correlation and higher consistency in all parameters. 3D Auto LAA showed higher intra- and interobserver reproducibility and allowed quicker analysis (p < 0.05). LAAO was performed in 35 patients (94.59%), and none of which had serious adverse events.Conclusion: TrueVue Glass is the first non-invasive and radiation-free visualization of the overall external contour of LAA and its adjacent structures. 3D Auto LAA simplifies the measurement, making the preoperative assessment more efficient and convenient while ensuring the accuracy and reproducibility. A combination of the two is feasible for accurate and rapid assessment of LAA anatomy and physiology in AF patients and has practical application in LAAO.

Quantitative analysis of root development is becoming a preferred option in assessing the function of hidden underground roots, especially in studying resistance to abiotic stresses. It can be enhanced by acquiring non-destructive phenotypic information on roots, such as rhizotrons. However, it is challenging to develop high-throughput phenotyping equipment for acquiring and analyzing in situ root images of root development. In this study, the RhizoPot platform, a high-throughput in situ root phenotyping platform integrating plant culture, automatic in situ root image acquisition, and image segmentation, was proposed for quantitative analysis of root development. Plants (1-5) were grown in each RhizoPot, and the growth time depended on the type of plant and the experimental requirements. For example, the growth time of cotton was about 110 days. The imaging control software (RhizoAuto) could automatically and non-destructively image the roots of RhizoPot-cultured plants based on the set time and resolution (50-4800 dpi) and obtain high-resolution (>1200 dpi) images in batches. The improved DeepLabv3+ tool was used for batch processing of root images. The roots were automatically segmented and extracted from the background for analysis of information on radical features using conventional root software (WinRhizo and RhizoVision Explorer). Root morphology, root growth rate, and lifespan analysis were conducted using in situ root images and segmented images. The platform illustrated the dynamic response characteristics of root phenotypes in cotton. In conclusion, the RhizoPot platform has the characteristics of low cost, high-efficiency, and high-throughput, and thus it can effectively monitor the development of plant roots and realize the quantitative analysis of root phenotypes in situ.

CKS (cyclin-dependent kinase subunit) is one of cell cycle genes, and cell cycle in apple (Malus × domestic Borkh.) fruit controlled cell number and fruit size. In order to explore CKS expression during the development of apple fruit size, the cultivar ‘Fuji’ and ‘Ralls’ whose fruit size had obvious difference were used for MdCKS1 gene cloning and expressing. In ‘Fuji’ and ‘Ralls’, the greatest growth rates of fruit size and weight occurred during 7–21 days after full bloom (DAFB), but in ‘Fuji’ during 21–35 DAFB the growth rates of fruit size and weight was still faster, and ‘Fuji’ remained much rapider growth rate of fruit size and weight than ‘Ralls’ within 7–35 DAFB, so the majority of cell production in ‘Ralls’ occurred between 7–21 DAFB and in ‘Fuji’ between 7–35 DAFB, and the biggest difference of fruit size and weight between ‘Fuji’ and ‘Ralls’ occurred at 35 DAFB. MdCKS1 sequence was identical with MdCKS1;1 (CO868566) and apple CKS1 (EB141951). MdCKS1 expression peak in ‘Fuji’ was two-fold higher than in ‘Ralls’; MdCKS1 expression peak in ‘Ralls’ occurred at 21 DAFB and in ‘Fuji’ at 35 DAFB which coincided respectively with rapid growth rate of early fruit. During 140–161 DAFB, ‘Fuji’ fruit had MdCKS1 expression peak again, but ‘Ralls’ not; ‘Fuji’ fruit size and weight had a larger extent of increase than ‘Ralls’, and the second significant difference of fruit size and weight between ‘Fuji’ and ‘Ralls’ occurred at 161 DAFB. In callus, MdCKS1 expression was much higher in ‘Fuji’ than in ‘Ralls’, up-regulated by auxin (NAA) or cytokinin (6-BA) or co-existence of them, and depressed by lower culture temperature. Altogether, ‘Fuji’ fruit had much rapider growth rate than ‘Ralls’ which resulted in significant difference of fruit size and weight between ‘Fuji’ and ‘Ralls’ respectively at 35 DAFB and 161 DAFB, and contributed to their fruit size difference; MdCKS1 expression in ‘Fuji’ fruit was much higher than in ‘Ralls’ both at early stage for fruit cell division and later stage for fruit cell expansion; MdCKS1 expression in callus was up-regulated by auxin or/and cytokinin. Hence, difference of fruit size between ‘Fuji’ and ‘Ralls’ was correlated with growth rate during fruit development, MdCKS1 expression correlated positively with growth rate and cell production of early fruit, and involved in fruit size which maybe was regulated by endogenous auxin or/and cytokine level of genotype.

Red-fleshedapples are preferredbecause of their high content of phenolics and antioxidants in peel and pulp. Herein, we evaluated the mechanisms of apple peel polyphenolic extracts (APP) and apple flesh polyphenolic extracts (AFP) from the new red-fleshed apple in inhibiting cell proliferation and inducing apoptosis on human breast cancer MDA-MB-231 cells. The antiproliferative activities were determined by the CCK8 assay. The expression of proteins was determined using Western blot. We found that the content of polyphenols and flavonoids in APP was significantly higher than that in AFP, and 14 main phenolic compounds in APP and AFP were quantified using UPLC-MS/MS techniques. Besides, the significant inhibition effects of APP and AFP were achieved through Akt pathway by inducing apoptosis (significantly upregulating reactive oxygen species [ROS] levels, and downregulating expression of pAkt, pBad, Bcl-2, promoting Cytochrome c release, activating Cle-Caspase 9, and inducing expressions of Cle-Caspase 3 and Cle-PARP), and inducing G0/G1 cell cycle arrest (increased expressions of p-p53 and p21 and decreased expressions of PCNA and Cyclin D1). And the inhibition effect of APP was stronger than that of AFP. These results suggest that AFP and APP may be excellent sources of natural chemicals for treating triple-negative breast cancer MDA-MB-231 cells. PRACTICAL APPLICATION: The effects of antiproliferation of phenolic extracts from red-fleshed apple peels and flesh on human breast cancer MDA-MB-231 cells were evaluated. The data may clarify the functional parts of red-fleshed apple and provide some basis for scientific researchers and consumers to recognize and exploit red-fleshed apple.

Mining pressure behavior in the process of longwall panel face passing through the parallel abandoned roadways (PARs) is different from the ordinary longwall panel face. It is easy to induce the accident of roof falling, coal wall spalling, and crush accident of shield. In order to reduce the occurrence of mine pressure accidents and ensure safe mining, a new mining method named “swing-inclined” mining method was proposed and was employed in the E13103 of Cuijiazhai coal mine. Based on the process of the longwall panel face passing through the PARs, a long-span and multisupport mass-structure model of the roof was established. The maximum support capacity of shield was calculated combined with stability relation between “roof-shield-PAR-‘similar pillar (SP)’-coal wall.” It provided the basis for determining the reasonable support capacity of shield. Moreover, the sensitivity analysis of influenced factors to the maximum support capacity of shield was carried out by using Matlab software. The sensitivity analysis results indicated that different factors had a different effect on the support capacity of shield. And, the process of passing through the PARs can be divided into 3 stages, depending on the relation between support capacity of shield and width of SP. In different stages, the change degree of support capacity of shield was different. The support capacity of shield is mainly influenced by the hanging distance of the main roof and the horizontal distance between the support point of the coal wall and the breaking position of the main roof. By on-site measurement, the sensitivity analysis results were verified.

Northern corn leaf blight, caused by the fungal pathogen Exserohilum turcicum, is a major disease of maize. The first major locus conferring resistance to E. turcicum race 0, Ht1, was identified over 50 years ago, but the underlying gene has remained unknown. We employed a map-based cloning strategy to identify the Ht1 causal gene, which was found to be a coiled-coil nucleotide-binding, leucine-rich repeat (NLR) gene, which we named PH4GP-Ht1. Transgenic testing confirmed that introducing the native PH4GP-Ht1 sequence to a susceptible maize variety resulted in resistance to E. turcicum race 0. A survey of the maize nested association mapping genomes revealed that susceptible Ht1 alleles had very low to no expression of the gene. Overexpression of the susceptible B73 allele, however, did not result in resistant plants, indicating that sequence variations may underlie the difference between resistant and susceptible phenotypes. Modelling of the PH4GP-Ht1 protein indicated that it has structural homology to the Arabidopsis NLR resistance gene ZAR1, and probably forms a similar homopentamer structure following activation. RNA sequencing data from an infection time course revealed that 1 week after inoculation there was a threefold reduction in fungal biomass in the PH4GP-Ht1 transgenic plants compared to wild-type plants. Furthermore, PH4GP-Ht1 transgenics had significantly more inoculation-responsive differentially expressed genes than wild-type plants, with enrichment seen in genes associated with both defence and photosynthesis. These results demonstrate that the NLR PH4GP-Ht1 is the causal gene underlying Ht1, which represents a different mode of action compared to the previously reported wall-associated kinase northern corn leaf blight resistance gene Htn1/Ht2/Ht3.

Objective: Propofol is the most widely administered intravenous anesthetic to induce deep sedation for gastroscopy procedures. Coadministration of nalbuphine can provide analgesia and sedation to patients experiencing visceral pain, thereby decreasing the amount of propofol needed and reducing the risk of propofol-induced adverse events. We carried out this study to determine the median effective dose (ED50) of propofol in combination with different dosages of nalbuphine and the optimal dosage of nalbuphine during painless gastroscopy.Methods: We recruited sixty-five patients aged 18–60 years who underwent elective painless gastroscopy. A total of sixty-one patients were allocated randomly to the N1 group (nalbuphine 0.1 mg/kg) or N2 group (nalbuphine 0.15 mg/kg). Three minutes after administration of nalbuphine, patients received a preset dose of propofol at 2.0 mg/kg with a dose gradient of 0.2 mg/kg according to Dixon’s “up-and-down” method. The primary outcome was the ED50 of propofol in combination with nalbuphine. Hemodynamic parameters, recovery time, pain score, and adverse events were recorded as secondary outcomes.Results: The ED50 of propofol in the N2 group was significantly lower than that observed in the N1 group (p < 0.01). Using probit analysis, the ED50 and ED95 of propofol were 1.632 mg/kg and 2.759 mg/kg in the N1 group and 1.111 mg/kg and 2.243 mg/kg in the N2 group, respectively. The incidence of hypotension in the N2 group was lower than that in the N1 group (p < 0.05), and the recovery time was shorter than that of the N1 group (p < 0.05).Conclusion: In adult patients, 0.15 mg/kg nalbuphine led to a significant reduction in the ED50 and ED95 of propofol during gastroscopy. This dose of nalbuphine also reduced the incidence of hypotension and shortened the recovery time. Therefore, nalbuphine (0.15 mg/kg) combined with propofol is a safe option for enhancing recovery after painless gastroscopy in adult patients.Clinical Trial Registration: [https://www.chictr.org.cn/edit.aspx?pid=126699&htm=4], identifier [ChiCTR2100053204].

Interpersonal relationship is one of the important factors affecting college students’ mental health. The relationship between interpersonal relationship and college students’ mental health has also become a large number of academic research topics. In order to explore whether there is a correlation between optimism and college students’ mental health, and if so, what kind of situation it presents. Based on literature review, mathematical statistics and questionnaire survey, this study optimized the iterative process of clustering algorithm. Extract valuable parts from a large amount of precipitation of students’ psychological data, establish data models, and provide decision-making guidance for managers. The results show that there are significant differences between optimists and pessimists in optimistic factors and pessimistic factors. Optimists score significantly higher on optimistic factors than pessimists, while pessimists score significantly lower than pessimists. Conclusion optimism can significantly alleviate life stress and intervene psychological crisis.

Most real-world optimization problems have multiple objectives. A system designer needs to find a policy that trades off these objectives to reach a desired operating point. This problem has been studied extensively in the setting of known objective functions. However, we consider a more practical but challenging setting of unknown objective functions. In industry, optimization under this setting is mostly approached with online A/B testing, which is often costly and inefficient. As an alternative, we propose Interactive Multi-Objective Off-policy Optimization (IMO^3). The key idea of IMO^3 is to interact with a system designer using policies evaluated in an off-policy fashion to uncover which policy maximizes her unknown utility function. We theoretically show that IMO^3 identifies a near-optimal policy with high probability, depending on the amount of designer's feedback and training data for off-policy estimation. We demonstrate its effectiveness empirically on several multi-objective optimization problems.

Chronic ethanol abuse is a systemic disorder and a risk factor for acute respiratory distress syndrome (ARDS) and chronic obstructive pulmonary disease (COPD). However, the mechanisms involved are unknown. One explanation is that ethanol produces damaging reactive oxygen species (ROS) and disturbs the balance of mitochondria within the lungs to promote a pro-injury environment. We hypothesized that targeting an antioxidant to the mitochondria would prevent oxidative damage and attenuate EtOH-LPS-induced lung injury. To test this, we investigated the effects of mitochondria-targeted ubiquinone, Mitoquinone (MitoQ) on ethanol-sensitized lung injury induced by LPS. Lung inflammation, ROS, mitochondria function, and mitophagy were assessed. We demonstrated that chronic ethanol feeding sensitized the lung to LPS-induced lung injury with significantly increased reactive oxygen species ROS level and mitochondrial injury as well as lung cellular NLRP3 inflammasome activation. These deleterious effects were attenuated by MitoQ administration in mice. The protective effects of MitoQ are associated with decreased cellular mitophagy and NLRP3 inflammasome activation in vivo and in vitro. Taken together, our results demonstrated that ethanol aggravated LPS-induced lung injury, and antioxidant MitoQ protects from EtOH-LPS-induced lung injury, probably through reducing mitophagy and protecting mitochondria, followed by NLRP3 inflammasome activation. These results will provide the prevention and treatment of ethanol intake effects with new ideas.

Infrared remote-sensing images have irreplaceable value in military and civilian research, such as remote surveillance and military reconnaissance. However, under the conditions of complex scenes, infrared ship detection still faces great challenges. Most importantly, because of the limited hardware resource, the spaceborne satellites usually have weak computational processing ability, which makes the traditional convolution neural network (CNN)-based detection algorithms difficult to show their power. In terms of the above facts, this article proposes a high-performance but low-computation and storage-efficient ship detection algorithm to adapt the severe spaceborne environment. Overall, our method contains the following technical steps: 1) we first present a novel iterative precise segmentation of land and sea algorithm to preprocess the complex and diverse remote-sensing scenes; 2) the multivariate Gaussian distribution is then selected to extract the ship target candidate regions to guarantee the detection recall; 3) we adopt the optical panchromatic data to assist the limited infrared data training; and 4) the cascade decision of multisource features including global and local cues is next utilized to gradually eliminate false alarms. Due to the high efficiency, the proposed method can implement well on the hardware platform of DSP and field-programmable gate array (FPGA) architecture. We conduct a series of experiments and compare with the state-of-the-art object detection algorithms. Experimental results show that our method has fewer parameters but can achieve strong detection robustness against the noise, cloud and reef interfere, which verifies the effectiveness of the proposed method.

Myopathy of the skeletal muscles is caused by excessive misuse of ethanol and affects half to two-thirds of pathological alcohol abusers. It is possible to identify alcohol-induced skeletal muscle disruptions as either 'acute or chronic'. Medium to moderate alcohol has positive or defensive effects (such as cardiovascular) on some organ systems, but long-term drinking and acute toxicity can adversely impact multiple organ systems and potentially increase mortality. Therefore we need a detailed understanding of the study status of alcoholic myopathy. References for researching alcoholic myopathy can be given by the review of this article.Search the Web of Science (WOS) central archive for alcoholic myopathy research papers from 2000 to 2020, and use CiteSpace and WOS databases for their own literature statistics techniques to evaluate the number of written articles, research organizations, citations to literature and identification of keywords.A total of 947 publications were collected after screening and the number of articles published grew year by year. The average number of released publications is 47.355. Most documents have been released by a total of 7 countries. Among them, 397 papers were published by the US, ranked first among all nations. Its intermediate centrality is also the largest, suggesting that other countries have more recognition of its scientific findings. The papers published by Emory Univ and Kobe Univ are well regarded in the field, among the top 7 academic institutions with publication volumes. There are 28 core authors and a total of 263 written articles. Highly cited papers are classified into 40 categories, of which 11 are prominently clustered categories. Co-occurrence study of keywords reveals that keywords such as skeletal muscle, oxidative stress, ethanol, alcohol, etc. are very common. The literature on alcoholic myopathy is primarily focused in Western countries, and autophagy, ROS and antioxidants are the latest study hotspots.

Electrical impedance tomography (EIT) has been used by researchers across several areas because of its low-cost and no-radiation properties. Researchers use complex conductivity in bioimpedance experiments to evaluate changes in various indicators within the image target. The diverse volumes and edges of biological tissues and the large impedance range impose dedicated demands on hardware design. The EIT hardware with a high signal-to-noise ratio (SNR), fast scanning and suitable for the impedance range of the image target is a fundamental foundation that EIT research needs to be equipped with. Understanding the characteristics of this technique and state-of-the-art design will accelerate the development of the robust system and provide a guidance for the superior performance of next-generation EIT. This review explores the hardware strategies for EIT proposed in the literature.

Soil salinization is one of the most important abiotic stresses adversely affecting plant growth and yield. In this study, we cloned and characterized a putative salt tolerance gene encoding a bHLH (basic helix-loop-helix) transcription factor, MdbHLH106L (MdbHLH106-like), in the genome of apple (Malus domestica Borkh.). The expression level and promoter activity of MdbHLH106L are increased under salt stress. Overexpressing MdbHLH106L in ‘Orin’ calli can promote the expression level of MdNHX1 (Na+/H+ exchanger 1). Further analysis showed that MdbHLH106L is able to bind the MdNHX1 promoter. Moreover, we also confirmed the interaction between MdbHLH106L and MdZAT10. In conclusion, MdbHLH106L participates in the salt-stress signaling pathway and may mediate salt tolerance by activating the MdNHX1 promoter and interacting with MdZAT10. Our findings may serve as a theoretical basis for further investigating molecular mechanisms of stress responses and for enriching genetic resources of stress-tolerant apple cultivars.

In this paper, we propose a tightly-coupled SLAM system fused with RGB, Depth, IMU and structured plane information. Traditional sparse points based SLAM systems always maintain a mass of map points to model the environment. Huge number of map points bring us a high computational complexity, making it difficult to be deployed on mobile devices. On the other hand, planes are common structures in man-made environment especially in indoor environments. We usually can use a small number of planes to represent a large scene. So the main purpose of this article is to decrease the high complexity of sparse points based SLAM. We build a lightweight back-end map which consists of a few planes and map points to achieve efficient bundle adjustment (BA) with an equal or better accuracy. We use homography constraints to eliminate the parameters of numerous plane points in the optimization and reduce the complexity of BA. We separate the parameters and measurements in homography and point-to-plane constraints and compress the measurements part to further effectively improve the speed of BA. We also integrate the plane information into the whole system to realize robust planar feature extraction, data association, and global consistent planar reconstruction. Finally, we perform an ablation study and compare our method with similar methods in simulation and real environment data. Our system achieves obvious advantages in accuracy and efficiency. Even if the plane parameters are involved in the optimization, we effectively simplify the back-end map by using planar structures. The global bundle adjustment is nearly 2 times faster than the sparse points based SLAM algorithm.

Exosomes are a new way of the communication between the tumor cell and macrophage in the micro-environment. The macrophage can be induced to different phenotypes according to the different tumors. In the present study, long-chain noncoding RNA HOTAIR (lncRNA HOTAIR) was highly expressed in LSCC and exosomes. The pathway of exosomal lncRNA HOTAIR inducing macrophage to M2 polarization in the LSCC was investigated. The carcinoma tissues and adjacent tissues were collected from 104 LSCC cases, and the positive relationship between CD163-/CD206-M2 macrophage infiltration and clinical phase, lymph node spreading and pathological phase in LSCC was observed. To examine the role of exosomal lncRNA HOTAIR, macrophages were co-cultured with LSCC-exosomes of high lncRNA HOTAIR expression or transferred with HOTAIR mimics. It was suggested that exosomal lncRNA HOTAIR can induce macrophages to M2 polarization by PI3K/p-AKT/AKT signaling pathway. Furthermore, exo-treated M2 macrophages facilitate the migration, proliferation, and EMT of LSCC.
