T-type calcium channel blockers have been previously shown to protect glomeruli from hypertension by regulating renal arteriolar tone. To examine whether blockade of these channels has a role in protection against tubulointerstitial damage, we used a stereo-selective T-type calcium channel blocker R(-)-efonidipine and studied its effect on the progression of this type of renal injury in spontaneously hypertensive rats that had undergone subtotal nephrectomy. Treatment with racemic efonidipine for 7 weeks significantly reduced systolic blood pressure and proteinuria. The R(-)-enantiomer, however, had no effect on blood pressure but significantly reduced proteinuria compared to vehicle-treated rats. Both agents blunted the increase in tubulointerstitial fibrosis, renal expression of alpha-smooth muscle actin and vimentin along with transforming growth factor-beta (TGF-beta)-induced renal Rho-kinase activity seen in the control group. Subtotal nephrectomy enhanced renal T-type calcium channel alpha1G subunit expression mimicked in angiotensin II-stimulated mesangial cells or TGF-beta-stimulated proximal tubular cells. Our study shows that T-type calcium channel blockade has renal protective actions that depend not only on hemodynamic effects but also pertain to Rho-kinase activity, tubulointerstitial fibrosis, and epithelial-mesenchymal transitions.

症例は41歳男性.慢性骨髄性白血病でインターフェロンーα2b (IFN一α2b) 500万単位/日の投与を受け, 39カ月後に蛋白尿が, 55カ月後には腎機能障害が出現し,漸次増悪した. 66カ月後, Ccrは44m1/minと低下し,一日lg前後の蛋白尿,血小板減少およびハプトグロビンの低値を認めた.腎生検所見から本例の腎障害に溶血性尿毒症症候群類似の細小血管レベルでの内皮細胞障害の関与が示唆され,原因としてIFN-αの関与が強く疑われた.

The objective of this study was to estimate non-additive genetic effects of heterosis and epistatic loss on percent of live spermatozoa in admixed Swiss Fleckvieh bulls, a composite of Simmental and Holstein Friesian cattle. Heterosis is the additional gain in productivity or fitness of crossbred progeny over the mid purebred parents mean which arises from intra-locus gene interaction. Epistatic effects generally reduce productivity or fitness due to lack of gene interactions of genes from different breeds, which is called epistatic loss. Bovine SNP chip data of were used to predict locus specific breed origin of alleles along the autosomes of 815 admixed bulls as well as 147 Holstein Friesian and 207 Simmental bulls representing the parental breeds. The breed proportions for admixed bulls based on 32,899 SNP were used to calculate breed heterozygosity and epistatic loss, considering additive by additive effects for 1,000,000 random pairs of loci. The average Holstein Friesian ancestry in admixed bulls was estimated to be 0.82. Results of fitting different linear mixed models showed that including breed heterozygosity and epistatic loss improved the model fitness (ΔAIC > 3). The heterosis effect and epistatic loss were estimated 2.5(±1.39) % and -0.65(±1.68) % of live spermatozoa, respectively. High correlation (0.97) between breed heterozygosity and epistatic loss values indicate strong confounding of these effects in the model, indicating that it is not possible to properly separate these effects.

Three rat mAb, RR3-15, RR3-16, and RR3-18, were established by fusing spleen cells from a rat immunized with the male Ag-specific cytolytic T cell clone, OH6, to mouse myeloma cells. The mAb was identified by their capacity to focus the cytolytic activity of the OH6 CTL clone on nonspecific target cells via FcR-FcR interaction. That all three mAb recognized the OH6 TCR was confirmed by immunoprecipitation studies in which each antibody precipitated a 90 kDa disulfide-linked heterodimer characteristic of the TCR. Surface immunofluorescence staining of a panel of T cell lines and splenic T cell populations showed that RR3-16 reacted not only to the OH6 T cell clone but also to a minor fraction of normal T cells. This reactivity was found to be due to the expression of a gene in the V alpha 3 family. However, RR3-16 did not react with all T cell lines and clones known to express genes from the V alpha 3 family. cDNA sequences of three independent RR3-16+ T cell hybridomas analyzed by polymerase chain reaction were identical to the previously published V alpha 3 sequence of the CTL clone C9. Thus, the mAb RR3-16 is specific for a single member of the TCR V alpha 3 gene family. Analysis of the expression of RR3-16+ TCR in CD4+ and CD8+ subsets of peripheral T cells demonstrated preferential expression on CD8+ T cells, suggesting regulated expression of this particular TCR V alpha gene.

We conducted a GWAS for ALS using the single-step methodology to take advantage of 1632 pedigree records, 892 phenotypes and 372 genotypes (173 662 markers) for which only 12% of the dogs had both phenotypes and genotypes available. Our analysis revealed associations towards the growth differentiation factor 9 gene (GDF9), which is known to regulate oocyte maturation. The trait heritability was estimated at 43.1%, from which approximately 15% was accountable by the GDF9 locus alone. Therefore, markers flanking GDF9 explained approximately 6.5% of the variance in ALS. Analysis of WGSs revealed two missense substitutions in GDF9, one of which (g.11:21147009G>A) affected a highly conserved nucleotide in vertebrates. The derived allele A was validated in 111 dogs and shown to be associated with decreased ALS (−0.75 ± 0.22 puppies per litter). The variant was further predicted to cause a proline to serine substitution. The affected residue was immediately followed by a six-residue deletion that is fixed in the canine species but absent in non-canids. We further confirmed that the deletion is prevalent in the Canidae family by sequencing three species of wild canids. Since canids uniquely ovulate oocytes at the prophase stage of the first meiotic division, requiring maturation in the oviduct, we conjecture that the amino acid substitution and the six-residue deletion of GDF9 may serve as a model for insights into the dynamics of oocyte maturation in canids.
