High throughput genomics has greatly facilitated identification of genetic variants. However, determining which variants contribute to disease causation is challenging with more than half of all missense variants now classified asvariants of uncertain significance(VUS). A VUS leaves patients and their clinicians unable to utilize the variant information in clinical decision-making. In long QT syndrome type 2,KCNH2channel function is directly associated with disease presentation. Therefore, functional phenotypingof KCNH2variants can provide direct evidence to aid variant classification. Here, we investigated the expression of all codon variants in exon 2 ofKCNH2using a massively parallel trafficking assay and for a subset of 458 single nucleotide variants compared the results with peak tail current density and gating using automated patch clamp electrophysiology. Trafficking could correctly classify loss of peak tail current density variants with an AUC reaching 0.94 compared to AUCs of 0.75 to 0.8 forin silicovariant classifiers. We suggest massively parallel trafficking assays can provide prospective and accurate functional assessment for all missense variants inKCNH2and most likely many other ion channels and membrane proteins.

Background: Ethnomedicinal knowledge is highly significant for persistence of human health care. Different studies have shown that medicinal plants are considered as curing agent for digestive disorders in Pakistan. The aim of present review is to explore this ethnomedicinal information all over the Pakistan and to investigate the ecological status of these species in the country.Methodology: Online literatures on ethnomedicinal plants used to treat digestive complaints in Pakistan were searched and gathered using online bibliographic databases including PubMed, Scopus, Google Scholar, Web of Science and Flora of Pakistan.Results: The review documented 178 medicinal plant species, belonging to 59 families, used for different digestive ailments. The majority of species were herbs (60%), used in drug preparation. The most frequent part used in preparation of these remedies is leaves (23%), followed by roots (19%).Conclusion: In conclusion, the study provides enormous ethno-medicinal knowledge and due to the unsustainable activities of the people of Pakistan, some medicinal plants need to be conserved for the future.Key words: Ethno-medicine, medicinal plants, gastrointestinal diseases, ecological status.

Application of solid sorbents for CO2 capture and separation has been under consideration since last decades owing to the promising feature of repeatable use, low cost of regeneration and thermo physical stability. However, the capturing capacity, selectivity and the overall performance of adsorbents at different temperatures and pressures still require further investigation to make these materials capable of replacing the mono-ethanol system. Here in we report synthesis, characterization and application of covalent organic polymers. Capturing capacity of COPs at low temperature and pressure has been experimented for two COPs materials (namely COP-9, COP-10) for CO2 and N2 adsorption.
