In this study, we compared the microbiota and volatile organic compounds (VOC) present in the milk obtained from 3 different sheep breeds, namely Merino, Lacaune, and Assaf. Udder milk was collected from 21 animals, 7 from each breed. Bacterial microflora was determined metagenomically by extracting the DNA from the milk and analyzing the V3-V4 region of the 16S rRNA gene. Headspace solid-phase microextraction gas chromatography-mass spectrometry method was used to analyze VOC. The metagenomic analysis revealed (for Merino, Lacaune, and Assaf milk, respectively) Firmicutes (66.32, 69.36, and 57.08%), Actinobacteria (19.09, 7.67, and 19.40%), Proteobacteria (13.76, 21.06, and 22.19%), and Bacteroidetes (0.84, 1.91, and 1.33%) phyla in the milk samples. Lactobacillus was highly abundant in the milk of 3 breeds (29.64, 43.50, and 18.70%). The genera constituting more than 2% of all bacteria in all groups were Jeotgalicoccus (7.19, 5.34, and 10.77%), Enterococcus (5.18, 9.78, and 3.64%), and Corynebacterium (4.08, 3.00, and 13.44%). A total of 32 different VOC were identified by headspace solid-phase microextration analysis with 9, 30, and 24 different compounds from Merino, Lacaune, and Assaf breeds, respectively. Although ketone was the most abundant compound in Merino milk (71.84%), hydrocarbons were the most detected in Lacaune and Assaf milk (37.18% and 55.42%, respectively). A positive correlation was found between acetone, which was detected at the highest level in all groups, with Salinicoccus, Alloiococcus, Psychrobacter, and Dietzia. In addition, a negative correlation was found between the Lactobacillus genus, detected at the highest level in all groups, with methyl cyclopentane, 3-methylheptane, octane, decane, 3,3-dimethyloctane, and dodecane. Thus, differences were observed in the bacterial microflora and VOC in the sheep milk from different breeds under different feeding and breeding conditions.

Amaç: Bu araştırmada, keklik etlerinde Listeria spp. ve patojen tür olan Listeria monocytogenes’in varlığı araştırılarak insidensinin ve dolayısıyla halk sağlığı bakımından öneminin ortaya konması amaçlandı.Gereç ve Yöntem: Araştırmada, 10 erkek ve 10 dişi olmak üzere 20 adet keklik eti kullanıldı. Numuneler, keklik yetiştiriciliği yapılan özel bir çiftlikten temin edildi. Kanatlı kesiminin yapıldığı özel bir kesimhanede kesilip aseptik şartlarda soğuk koşullar altında laboratuara getirilip analize alındı. Listeria spp.’nin izolasyon ve identifikasyonunda Food and Drug Administration (FDA) tarafından bildirilen yöntem kullanıldı.Bulgular: İncelenen 20 keklik numunesinden bir erkek ve bir dişi olmak üzere 2’sinin (%10) Listeria spp. ile kontamine olduğu tespit edildi. Kontamine iki numuneden izole edilen 10 izolatın 5’i (%50) L. innocua, 4’ü (%40) L. grayi ve 1’i (%10) L. welshimeri olarak identifiye edildi. Öneri: Elde edilen bulgular, keklik etinin halk sağlığı açısından risk oluşturabileceğini ve bazı önlemlerin alınması gerektiğini ortaya koymaktadır. Bu amaçla, özellikle yetiştirme ve kesimhane işletmelerinin hijyen kalitesinin yükseltilmesi, çapraz bulaşmaların önlenmesi ve tüketim aşamasına kadar soğuk zincirin sağlanmasının önem arz ettiği kanaatine varılmıştır., Aim: In this study, it was aimed that to determine the presence of Listeria spp. and especially Listeria monocytogenes in partridge meats for putting forth the importance on public health. Materials and Methods: The survey included 10 male and 10 female partridge carcasses. Samples were obtained from a private farm which performs partridge breeding. After the cutting of partridges under aseptic conditions in a private slaughterhouse, samples were brought in to the laboratory and analyzed. Food and Drug Administration (FDA) method was used for the isolation and identification. Results: Two of the (10%) samples analyzed were found to be contaminated with Listeria spp. 10 isolates collected from the contaminated samples. These samples identified as L. innocua (5, 50%), L. grayi (4, 40%) and L. welshimeri (1, 10%). Conclusions: As a result, findings showed us partridge meat may be a public health risk and preventive measures should be taken. For this purpose hygienic conditions have to be proven on breeding and slaughtering enterprises, prevent cross- contamination and kept the cold chain until consumption.

ÖZET: Araştırma, farklı dumanlama teknikleri uygulanmasının Selçuklu Tulum peynirinin kimyasal, mikrobiyolojik ve duyusal niteliklerine etkisini belirlemek amacıyla yapılmıştır. Deneysel olarak dört grup peynir numunesi üretildi. 1. gruptakilere (kontrol grubu) dumanlama işlemi uygulanmadı, 2. gruptaki numuneler % 0.5 oranında sıvı duman içeren solüsyonda beş dakika bekletildi, 3. gruptaki numuneler ise 20±1°C üç saat süreyle doğal dumanlamaya tabii tutuldu. 4. gruptaki numunelerin üretiminde kullanılan süte % 0.01 oranında sıvı duman ilave edildi. Numuneler üretimin 0. ve olgunlaşmanın 15., 30., 60. ve 90. günlerinde kimyasal, mikrobiyolojik ve olgunlaşmanın 30., 60.ve 90. günlerinde duyusal analizlere tabi tutuldu. 4. grup numunelerin % yağ ve asitlik değerleri, diğer gruplardaki numunelere göre daha az, pH değerleri ise daha yüksek bulundu. Kontrol grubu ile farklı dumanlama işlemleri uygulanan numuneler arasında % rutubet, protein, tuz, kül ve su aktivitesi değerleri ile içerdikleri mikroorganizma sayıları bakımından önemli (p>0.05) düzeyde farklılık görülmedi. Duyusal yönden 1. ve 2. gruptaki numunelerin daha çok beğenildiği tespit edildi. Sonuç olarak Selçuklu Tulum peyniri üretiminde, % 0.5 oranında sıvı duman içeren solüsyonda peynirin beş dakika bekletilmesiyle, Türkiye’de üretilen peynirlere çeşitlilik kazandırılabileceği ve peynir tüketiminin artmasına katkıda bulunabileceği kanaatine varıldı.Anahtar Kelimeler : Dumanlama, peynir, kimyasal kalite, mikrobiyolojik kalite, organoleptik kaliteEffect of the Different Smoking Techniques on the Chemical Microbiological and Sensory Properties of Selçuklu Tulum CheeseABSTRACT : The investigation was done to determine the effect of different smoking techniques on chemical, microbiological and sensory properties of Selçuklu Tulum cheese. Four cheese samples were produced experimentally. Any smoking procees was not applied to first group (group 1). The samples in group 2 were kept five minutes in the solution containing 0.5 % liquid smoke. The samples in group 3 were smoked naturally at 20±1°C for three hours. In the production of Group 4, liquid smoke was added to the milk at the rate of 0.01 %. Samples were analysed chemically and microbiologically on the 1st , 15th , 30th , 60th , and 90th days of the ripening and were evaluated organolepticaly at 30th , 60th , and 90th days of the ripening. Fat contents and acidity values of fourth group samples (0.01 % liquid smoke added to milk) were lower than those in the other groups while pH values were higher. There was not any significant (p>0.05) difference in chemical and microbiological results between control (Group 1) and the others (Groups 2, 3, 4). According to the sensory analyses, the samples in the 1st and the 2rd Groups were more preferred. It is concluded that the application of 0.5 % liquid smoke in the production of Selçuklu Tulum cheese could be used successfully in order to obtain a new type of smoked cheese, and may contribute to the increase of cheese consumption in Turkey.Key words: Smoking, cheese, chemical quality, microbiological quality, organoleptical quality

Araştırmada Konya yöresindeki süt işletmelerinde peynir yapımında kullanılan 20 adet hayvansal sıvı peynir mayası,mikrobiyolojik ve kimyasal kalite nitelikleri yönünden incelendi. Numunelerde aerobik mezofilik mikroorganizmave maya/küf sayılarının sırasıyla 0-1.0x103 kob/g ve 0-9.6x10 kob/g arasında değiştiği, diğer taraftan numunelerin hiçbirinde koliform bakterilere, E.co/Iye, S. eureuse ve anaerobik bakterilere rasllanmadığı tespit edildi. Numunelerin tuzve pH değerleri ise sırasıyla %12.91-18.79 ve 5.02-6.10 arasında bulundu. Sonuç olarak, incelenen numunelerin mikrobiyolojik özellikler açısından TS 3844 peynir mayası standardına uygun olduğu, fakat %1O'nun tuz miktarı yönündenTS 3844'e uygun olmadığı tespit edildi., In this research, microbiological and chemical qualities of 20 animal liquid rennet samples, used for cheesemakingin dairy plants in Konya region were determined. As a result of the microbiological analysis, aerobic mesophile,yeast and mould counts of the samples were determined between 0-1.0x103 du/ml and 0-9.6x10 du/ml respectively.None of the samples contained coliform bacteria, E.eoli, S. aureus and anaerobic bacteria. In theconsequence of the chemical analysis salt contents and pH values were determined between 12.91-18.79% and 5.02-6.10, respectively. In conclusion, all of the rennet samples were convenient with microbiological standard in TS 3844(Turkish Rennet Standard), but of the 20 rennet samples 10% for salt contents were under the minimum lirnlt in TS3844.

Bu araştırmada Konya piyasasından temin edilen 30 adet sucuk numunesi kimyasal ve mikrobiyQ.lojik yönünden incelendi. Sucuk numunelerinin mikrobiyolojik muayeneleri sonucunda genel canlı, koliform grubu, Staph­ylococcus-Micrococcus mikroorganizmaları ve maya-küf sayıları, sırasıyla, ortalama 5.7xI06, 7.4xI03, 3.2xl05 ve 6.4x104/g olarak tespit edildi. Kimyasal analizler sonuclında numunelerin ortalama rutubet, yağ, protein, kOI ve tuz miktarları, sırasıyla, %29.49, %30.9, %22.73, %4.33 ve %2.93; pH değeri 5.24 ve su aktivitesi (Aw) değeri 0.806 ola­rak bulundu. Elde edilen bulguların değerlendirilmesinde, sucuk numunelerinin mikrobiyolojik ve kimyasal kalitesInin düşük olduğu ve numunelerin o/..33.33'ünOn mikrobiyoloijk, % 86.67'sinin de kimyasal yönden Tüzük ve Standard'a uymadığı saptandı, In this research, chemical and microbiological properties of 30 Turkish sausage samptes collecled 'rom Konya marKets were determined. At the result of microbiological analyzes, the average count ollotal microorganisms, Coliform groups, Staphylococcus-Micrococcus and yeast and mould were determined as 5.7xl06, 7.4xl03, 3.2xl05 and 6.4xl04/g respectively. In the chemical analyses of the samples, the average values of moisture, fat, protein, ash and salt contents were found to be 29.49 %, 30.9 %, 22.73%, 4.33 % and 2.93 % respectively; pH and water activity (Aw) values were found to be 5.24 and 0.806. During the assesment ol results, microbiological and chemical pro­pertles ol the sausage samples were found to be low quality. According to the results, the sausages (33.33 % in mic­robiologically, 86.67 %in chemically) weren't in accordance with Food Aegulation and Standards

Amaç: Değişik oranlarda rennet kazein eklenerek üretilen kaşar peynirlerinin olgunlaşma süresince kimyasal, mikrobiyolojik ve duyusal özelliklerinde meydana gelen değişimin araştırılması amaçlanmıştır. Gereç ve Yöntem: Sütün yağı %3,0’e ayarlandı, 37˚C’ye ısıtılıp enzim eklendi. Pıhtı işleme, baskılama ve fermantasyondan sonra parçalama işleminde eritme tuzları (%0,1), kaya tuzu (%1,5) ve rennet kazein (%0-kontrol, 0,5, 1,0, 1,5, 2,0) eklendi. Eritme işlemi (65˚C, 5 dak) yapılarak peynir üretildi, vakumla paketlendi, olgunlaştırıldı (8˚C, 90 gün). Örneklerde kuru madde gravimetrik yöntemle, yağ Gerber metoduyla, tuz Mohr yöntemiyle ve titrasyon asitliği % laktik asit olarak belirlendi. Örneklerin Toplam mezofilik aerobik bakteri, laktik asit bakterisi, Stapylococcus aureus, koliform bakteri, maya–küf içeriği incelendi. Duyusal değerlendirme 7 panelist tarafından yapıldı. Kimyasal ve mikrobiyolojik sonuçlar istatistiksel olarak one-way Anova testine, duyusal sonuçlar ise Kruskal-Wallis H testine tabi tutuldu. Bulgular: Peynirler 8˚C’de 90 gün olgunlaştırıldı ve olgunlaştırmanın 1., 30., 60. ve 90. günlerinde kimyasal, mikrobiyolojik ve duyusal analizler yapıldı. Peynirlerin ortalama pH değerleri 5,62-5,76, asitlik %0,55-0,83 L.a., kurumadde %54,93-58,04, tuz %1,22-1,37, kurumaddede tuz %2,21-2,45, yağ %23,67- 25,33 ve kurumaddede yağ %41,20-45,46, TAMB 5,17-5,75 logkob/g, LAB 5,49-6,98 logkob/g, maya-küf sayısı 3,66-4,88 logkob/g olarak belirlendi, S.aureus ve koliform grubu bakteriye rastlanılmadı. Peynirlerin titrasyon asitliğinde önemli farklılık bulundu (p0,05). Duyusal analizlerde dış görünüş bakımından 3 nolu peynirde, iç görünüş bakımından 2 nolu peynirde istatistiki olarak önemli farklılık bulundu (p, Aim: It was aimed to investigate the changes in the chemical, microbiological and sensory properties of kashar cheese produced by adding different rates of rennet casein during ripening. Materials and Methods: Bovine milk (3,0% fat) was heated (37˚C), enzyme was added. After clotting, pressure and fermantation, emulsifying salts (0,1%), salt (1,5%) and rennet casein (%0-control, 0,5, 1,0, 1,5, 2,0) was added. Produced cheese at melting tank (65˚C, 5 min) was packaged and was ripened (8˚C, 90 days). In samples, dry matter was conducted with gravimetric, fat with Gerber, salt with Mohr and titration acidity with % lactic acidity. Total mesophilic aerobic bacteria, lactic acid bacteria, S.aureus, coliform bacteria, yeast-mould contents of the samples were examined. Sensory evaluation was performed by 7 panelists. One-way Anova test and Kruskal-Wallis H test was used for istatistically analysis. Results: Cheeses were ripened at 8˚C and were analyzed during the ripening period (90 days). pH value was found between 5,62-5,76, acidity 0,55-0,83L.a., drymatter 54,93-58,04%, salt 1,22-1,37%, salt in drymatter 2,21-2,45%, fat 23,67-25,33% and fat in drymatter 41,20-45,46%. Mean values were determined to TAMB 5,17-5,75 logcfu/g, LAB 5,49-6,98 logcfu/g, mould-yeast 3,66-4,88 logcfu/g. There was a significant difference (p 0,05) was found on other criteria. Total scores were between 20,62-22,19 point at sensory evaluation.
