Within the fields of regenerative medicine and tissue engineering, the development of tough hydrogel biomaterials is a challenging topic that has received much attention over the past few years. Recently, a method was developed to synthesize tough combinatorial poly(urethane-isocyanurate) (PUI)-type hydrogels by the trimerization of mixtures of NCO-functionalized prepolymers. As this synthesis approach allows a large degree of freedom in terms of polymer network design with a high level of control over the polymer network structure, the resulting systems are ideally suited for studying structure–property relations in tough hydrogel systems. In this work, we aim to systematically investigate the influence of introducing a hydrophobic component into a PUI polymer network on the mechanical properties of the resulting PUI hydrogels. Additionally, the effect of (the degree of) crystallinity of the hydrophobic network component is investigated. For this, two series of combinatorial PUI hydrogels are synthesized, based on a hydrophilic poly(ethylene glycol) prepolymer and increasing amounts of either a crystallizable hydrophobic prepolymer (poly(ε-caprolactone)) or an amorphous hydrophobic prepolymer (poly(propylene glycol)). It is shown that the toughness of amorphous PUI hydrogels is hardly influenced by the hydrophobic content, whereas the toughness of semi-crystalline PUI hydrogels strongly increases with increasing hydrophobic content. Also, the toughness of the latter hydrogels increases further with increasing degree of crystallinity of the hydrogel. Finally, it is shown that the semi-crystalline PUI hydrogels are promising materials for biomedical adhesive and coating applications, as well as for load-bearing biomedical applications within the fields of tissue engineering and regenerative medicine.

Hydrogels present a great number of advantages, such as their swelling capacity or their capability to mimic tissues, which make them very interesting biomaterials. However, one of their main disadvantages is their lack of good mechanical properties, which could limit some of their applications. Several strategies have been carried out to develop hydrogels with enhanced mechanical properties, but many of the suggested synthetic pathways to improve this property are expensive and time consuming. In this work, we studied an easy synthetic path to produce tough hydrogels based on different maleic anhydride copolymers crosslinked with polyethylenglycol. The effect of the comonomers in the mechanical properties has been studied, their excellent mechanical properties, good swelling behavior and thermal stability being remarkable. In addition, in order to evaluate their possible applications as scaffolds or in wound healing applications, microsized fibers have been fabricated by electrospinning.

Artificial shape-morphing hydrogels are emerging toward various applications, spanning from electronic skins to healthcare. However, the low freezing and drying tolerance of hydrogels hinder their practical applications in challenging environments, such as subzero temperatures and arid conditions. Herein, we report on a shape-morphing system of tough organohydrogels enabled by the spatially encoded rigid structures and its applications in conformal packaging of “island–bridge” stretchable electronics. To validate this method, programmable shape morphing of Fe (III) ion-stiffened Ca-alginate/polyacrylamide (PAAm) tough organohydrogels down to −50 °C, with long-term preservation of their 3D shapes at arid or even vacuum conditions, was successfully demonstrated, respectively. To further illustrate the potency of this approach, the as-made organohydrogels were employed as a material for the conformal packaging of non-stretchable rigid electronic components and highly stretchable liquid metal (galinstan) conductors, forming a so-called “island–bridge” stretchable circuit. The conformal packaging well addresses the mechanical mismatch between components with different elastic moduli. As such, the as-made stretchable shape-morphing device exhibits a remarkably high mechanical durability that can withstand strains as high as 1000% and possesses long-term stability required for applications under challenging conditions.

In the field of actuator materials, hydrogels that undergo large volume changes in response to external stimuli have been developed for a variety of promising applications. However, most conventional hydrogels are brittle and therefore rupture when they are stretched to moderate strains (~50%). Thus, gels to be used for actuators still require improved mechanical properties and actuation performance. In this study, we synthesized a tough and thermo-sensitive hydrogel with a large actuation force by forming interpenetrating networks between covalently crosslinked poly(N-isopropylacrylamide) and ionically crosslinked alginate. Poly(N-isopropylacrylamide) was used as a thermo-sensitive actuation material, and alginate was found to enhance the mechanical properties of the hydrogels. Due to the enhanced elastic modulus and energy dissipation in the hybrid gel, the toughness was increased by a factor of 60 over that of pure PNIPAAm gel. Further, based on the results showing that the hybrid gel exhibits an actuation force that is seven times higher than that of pure PNIPAAm gel, the hybrid gel is more applicable to real actuators.

Biological tissues hinge on blood perfusion and mechanical toughness to function. Injectable hydrogels that possess both high permeability and toughness have profound impacts on regenerative medicine but remain a long‐standing challenge. To address this issue, injectable, pore‐forming double‐network hydrogels are fabricated by orchestrating stepwise gelation and phase separation processes. The interconnected pores of the resulting hydrogels enable direct medium perfusion through organ‐sized matrices. The hydrogels are amenable to cell encapsulation and delivery while promoting cell proliferation and spreading. They are also pore insensitive, tough, and fatigue resistant. When tested in biomimetic perfusion bioreactors, the hydrogels maintain physical integrity under prolonged, high‐frequency biomechanical stimulations (>6000 000 cycles at 120 Hz). The excellent biomechanical performance suggests the great potential of the new injectable hydrogel technology for repairing mechanically dynamic tissues, such as vocal folds, and other applications, such as tissue engineering, biofabrication, organs‐on‐chips, drug delivery, and disease modeling., A design and methodology to form injectable, in situ pore‐forming, tough, and cytocompatible hydrogels are reported. Such hydrogels combine superior permeability and toughness, enabling direct medium perfusion through organ‐sized matrices. They are immune against extreme biomechanical loads and auspicious to cells. This work advances the injectable hydrogel technology and opens new opportunities in tissue engineering, drug/cell delivery, biofabrication, microfluidics, and organs‐on‐chips.

Development of mechanically durable and biologically inductive hydrogels is a major challenge for load-bearing applications such as engineered cartilage. Dissipative capacity of articular cartilage is central to its functional behavior when submitted to loading. While fluid frictional drag is playing a significant role in this phenomenon, the flow-dependent source of dissipation is mostly overlooked in the design of hydrogel scaffolds. Herein, we propose an original strategy based on the combination of fluidic and polymeric dissipation sources to simultaneously enhance hydrogel mechanical and mechanobiological performances. The nondestructive dissipation processes were carefully designed by hybrid cross-linking of the hydrogel network and low permeability of the porous structure. It was found that intrachain and pore water distribution in the porous hydrogels improves the mechanical properties in high water fractions. In contrast to widely reported tough hydrogels presenting limited load support capability at low strain values, we obtained stiff and dissipative hydrogels with unique fatigue behavior. We showed that the fatigue resistance capability is not a function of morphology, dissipation level, and stiffness of the viscoelastic hydrogels but rather depends on the origin of the dissipation. Moreover, the preserved dissipation source under mechanical stimulation maintained a mechanoinductive niche for enhancing chondrogenesis owing to fluid frictional drag contribution. The proposed strategy can be widely used to design functional scaffolds in high loading demands for enduring physiological stimuli and generating regulatory cues to cells.

Inspired by the toughening mechanism of double-network (DN) gels, tough hydrogel composites with a sandwich structure were fabricated from photoresponsive polymers. By copolymerization of hydrophilic monomers, 2-ureidoethyl methacrylate (UM), and photoresponsive hydrophobic monomers, (2-nitrobenzyloxycarbonylaminoethyl methacrylate (NBOC)) at high concentrations, physical hydrogels that are soft and highly stretchable are formed due to the hydrophobic associations of NBOC, serving as dynamic crosslinkers. By UV irradiation, the physical crosslinking switches into chemical crosslinking, and the soft physical hydrogels transform into rigid and less stretchable chemical hydrogels. By UV curing the surface layers of the physical hydrogels, we prepared hydrogel composites having a sandwiched structure with two rigid outer layers and a soft inner layer. The molecular-level continuous interfaces and matched swelling ratios between the layers ensure the macroscale hydrogel composites’ high strength and toughness with a DN gel effect. The outer layers fracture preferentially at deformation, playing a role like the first network of a DN gel, while the inner layer maintains the integrity, playing a role resembling the second network. The evolution of the fracture morphology of the rigid layers gives useful insight into the internal fracture process of DN gels.

Electrically conductive hydrogels (ECHs) are attracting much interest in the field of biomaterials science because of their unique properties. However, effective incorporation and dispersion of conductive materials in the matrices of polymeric hydrogels for improved conductivity remains a great challenge. Here, we demonstrate highly transparent, electrically conductive, stretchable tough hydrogels modified by single-walled carbon nanotubes (SWCNTs). Two different approaches for the fabrication of SWCNT/hydrogel structures are examined: a simple SWCNT film transfer onto the as-prepared hydrogel and the film deposition onto the pre-stretched hydrogel. Functionality of our method is confirmed by scanning electron microscopy along with optical and electrical measurements of our structures while subjecting them to different strains. Since the hydrogel-based structures are intrinsically soft, stretchable, wet, and sticky, they conform well to a human skin. We demonstrate applications of our material as skin-like passive electrodes and active finger-mounted joint motion sensors. Our technique shows promise to accelerate the development of biointegrated wearable electronics.

Hydrogels are often employed for tissue engineering and moistening applications. However, they are rarely used for load-bearing purposes because of their limited stiffness and the stiffness-toughness compromise inherent to them. By contrast, nature uses hydrogel-based materials as scaffolds for load-bearing and protecting materials by mineralizing them. Inspired by nature, the stiffness or toughness of synthetic hydrogels has been increased by forming minerals, such as CaCO3, within them. However, the degree of hydrogel reinforcement achieved with CaCO3 remains limited. To address this limitation, we form CaCO3 biominerals in situ within a model hydrogel, poly(acrylamide) (PAM), and systematically investigate the influence of the size, structure, and morphology of the reinforcing CaCO3 on the mechanical properties of the resulting hydrogels. We demonstrate that especially the structure of CaCO3 and its affinity to the hydrogel matrix strongly influence the mechanical properties of mineralized hydrogels. For example, while the fracture energy of PAM hydrogels is increased 3-fold if reinforced with individual micro-sized CaCO3 crystals, it increases by a factor of 13 if reinforced with a percolating amorphous calcium carbonate (ACC) nano-structure that forms in the presence of a sufficient quantity of Mg2+. If PAM is further functionalized with acrylic acid (AA) that possesses a high affinity towards ACC, the stiffness of the hydrogel increases by a factor 50. These fundamental insights on the structure-mechanical property relationship of hydrogels that have been functionalized with in situ formed minerals has the potential to enable tuning the mechanical properties of mineralized hydrogels over a much wider range than what is currently possible.
