Background: The three chick wing digits represent a classical example of a pattern specified by a morphogen gradient. Here we have investigated whether a mathematical model of a Shh gradient can describe the specification of the identities of the three chick wing digits and if it can be applied to limbs with more digits. Results: We have produced a mathematical model for specification of chick wing digit identities by a Shh gradient that can be extended to the four digits of the chick leg with Shh-producing cells forming a digit. This model cannot be extended to specify the five digits of the mouse limb. Conclusions: Our data suggest that the parameters of a classical-type morphogen gradient are sufficient to specify the identities of three different digits. However, to specify more digit identities, this core mechanism has to be coupled to alternative processes, one being that in the chick leg and mouse limb, Shh-producing cells give rise to digits; another that in the mouse limb, the cellular response to the Shh gradient adapts over time so that digit specification does not depend simply on Shh concentration.

Chicken talpid(3) mutant embryos have a wide range of Hedgehog-signalling related defects and it is now known that the talpid(3) gene product encodes a novel protein essential for Hedgehog signalling which is required for both activator and repressor functions of Gli transcription factors (Davey, M.G., Paton, I.R., Yin, Y., Schmidt, M., Bangs, F.K., Morrice, D.R., Gordon-Smith, T., Buxton, P., Stamataki, D., Tanaka, M., Münsterberg, A.E., Briscoe, J., Tickle, C., Burt, D.W. (2006). The chicken talpid(3) gene encodes a novel protein essential for Hedgehog signalling. Genes Dev 20 1365-77). Haemorrhaging, oedema and other severe vascular defects are a central aspect of the talpid(3) phenotype (Ede, D.A. and Kelly, W.A (1964a). Developmental abnormalities in the head region of the talpid(3) mutant fowl. J. Embryol. exp. Morp. 12:161-182) and, as Hedgehog (Hh) signalling has been implicated in every stage of development of the vascular system, the vascular defects seen in talpid(3) are also likely to be attributable to abnormal Hedgehog signalling. Gene expression of members of the VEGF and Angiopoietin families of angiogenic growth factors has been linked to haemorrhaging and oedema and we find widespread expression of VEGF-D, rigf and Ang2a in the talpid(3) limb. Furthermore, ectopic expression of these genes in talpid(3) limbs points to regulation via Gli repression rather than activation. We monitored specification of vessel identity in talpid(3) limb vasculature by examining expression of artery-specific genes, Np1 and EphrinB2, and the vein-specific genes, Np2a and Tie2. We show that there are supernumerary subclavian arteries in talpid(3) limb buds and abnormal expression of an artery-specific gene in the venous submarginal sinus, despite the direction of blood flow being normal. Furthermore, we show that Shh can induce Np1 expression but has no effect on Np2a. Finally, we demonstrate that induction of VEGF and Ang2a expression by Shh in normal limb buds is accompanied by vascular remodelling. Thus Hedgehog signalling has a pivotal role in the cascade of angiogenic events in a growing embryonic organ which is similar to that proposed in tumours.

Urodele amphibians are the only vertebrates that can regenerate amputated limbs, even as adults. However, we have previously shown that amputated chick wing bud stumps can be induced to ((regenerate)) and to form a complete set of correctly-patterned skeletal elements, following implantation of beads soaked in fibroblast growth factor-4 (FGF-4). We have now performed Dil injection experiments to determine which cells contribute to FGF-4-induced chick wing bud ((regenerates)). We show that the FGF-4-induced outgrowth of the regenerating wing bud stump is comprised of mesenchyme cells that originate from a region within 200 microm of the FGF-4 bead, and that cells proximal to the bead move distally.

Pattern formation is the developmental process that leads to the spatial ordering of cell differentiation. We have explored the problem of pattern formation in the development of the face of chick embryos. At early stages, the developing face consists of a series of small buds of tissue, the facial primordia that encircle the primitive mouth. The concepts of positional information provide a framework for considering how the patterns of differentiated cells are generated in the face. We suggest that the cranial neural crest cells must first be informed to which facial primordium they belong and then of their position within that primordium. The cells of the early primordia appear indistinguishable. However, when the mesenchyme cells are placed in high-density culture, cartilage differentiates. The extent and pattern of cartilage differentiation is characteristic for the cell population of each facial primordium. Myogenic cells also differentiate in the cultures, but the proportion of myogenic cells is independent of the extent of chondrogenesis. Within the facial primordia, a set of epithelial-mesenchymal interactions appears to be required for outgrowth and pattern formation along the proximodistal axis of the chick beaks. In culture, face epithelium locally inhibits cartilage differentiation and suggests that another set of epithelial-mesenchymal interactions may be involved in cell patterning. The mechanisms involved in specifying the mediolateral axis of the face, for example, the midpoint of the upper beak, are not known. Vitamin A derivatives, collectively known as retinoids, affect the development of the face of chick embryos and lead to a specific facial defect. Upper beak development is inhibited but the lower beak develops normally. The response to retinoids could be related to the specification of cells to belong to the facial primordium that will form the upper beak. Alternatively, retinoids may interfere with positional cues that operate to inform cells of their position within that primordium.

Developing chicken wings homozygous for the talpid (ta3/ta3) mutation are polydactylous and have defects in the establishment of their anteroposterior polarity. We analysed the expression domains of the posteriorly restricted homeobox Hox-4 genes in such mutant wings. The Hox-4 genes are now expressed right across the anteroposterior axis instead of being expressed just posteriorly. This correlates well with the absence of clear morphological differences between the talpid3 digits and reinforces the idea that vertebrate Hox-4 genes are involved in setting up the limb anteroposterior asymmetry.

The products of Hox-4 genes appear to encode position in developing vertebrate limbs. In chick embryos, a number of different signalling regions when grafted to wing buds lead to duplicated digit patterns. We grafted tissue from the equivalent regions in mouse embryos to chick wing buds and assayed expression of Hox-4 genes in both the mouse cells in the grafts and in the chick cells in the responding limb bud using species specific probes. Tissue from the mouse limb polarizing region and anterior primitive streak respecify anterior chick limb bud cells to give posterior structures and lead to activation of all the genes in the complex. Mouse neural tube and genital tubercle grafts, which give much less extensive changes in pattern, do not activate 5’-located Hox-4 genes. Analysis of expression of Hox-4 genes in mouse cells in the grafted signalling regions reveals no relationship between expression of these genes and strength of their signalling activity. Endogenous signals in the chick limb bud activate Hox-4 genes in grafts of mouse anterior limb cells when placed posteriorly and in grafts of mouse anterior primitive streak tissue. The activation of the same gene network by different signalling regions points to a similarity in patterning mechanisms along the axes of the vertebrate body.
