2. Data from Chemosensitization of B-Cell Lymphomas by Methylseleninic Acid Involves Nuclear Factor-κB Inhibition and the Rapid Generation of Other Selenium Species

- Abstract
Although recent reports suggest that selenium can modulate the activity of cytotoxic drugs, the mechanism underlying this activity remains unclear. This has been investigated using a panel of human B-cell lymphoma cell lines. The cytotoxic effects of chemotherapeutic agents (e.g., doxorubicin, etoposide, 4-hydroperoxycyclophosphamide, melphalan, and 1-β-d-arabinofuranosylcytosine) were increased by up to 2.5-fold when combined with minimally toxic concentrations (EC5-10) of the organic selenium compound, methylseleninic acid (MSA). DNA strand breaks were identified using comet assays, but the measured genotoxic activity of the combinations did not explain the observed synergistic effects in cell death. However, minimally toxic (EC10) concentrations of MSA induced a 50% decrease in nuclear factor-κB (NF-κB) activity after an exposure of 5 h, similar to that obtained with the specific NF-κB inhibitor, BAY 11-7082. Combinations of BAY 11-7082 with these cytotoxic drugs also resulted in synergism, suggesting that the chemosensitizing activity of MSA is mediated, at least in part, by its effects on NF-κB. Basal intracellular selenium concentration was higher in a MSA-sensitive cell line. After exposure to MSA, methylselenocysteine and selenomethionine were identified as the main intracellular species generated. Volatile selenium species, trapped using solid-phase microextraction fibers, were identified as dimethylselenide and dimethyldiselenide. These volatile species are thought to be the most biologically active forms of selenium. Taken together, these results show that the NF-κB pathway is one target for MSA underlying the interaction between MSA and chemotherapy. These data encourage the further clinical development of selenium as a potential modulator of cytotoxic drug activity in B-cell lymphomas. [Cancer Res 2007;67(22):10984–92]

3. Data from AML1/ETO Proteins Control POU4F1/BRN3A Expression and Function in t(8;21) Acute Myeloid Leukemia

- Abstract
A variety of genetic lesions, including chromosomal translocations, internal tandem duplications, and mutations, have been described in acute myeloid leukemia (AML). Expression profiling has shown that chromosomal translocations, in particular, are associated with distinctive patterns of gene expression. AML exhibiting the translocation t(8;21), which fuses the AML1 and ETO genes, has such a characteristic expression profile. One gene whose expression is highly correlated with the presence of the AML1/ETO fusion is POU4F1, which encodes the POU homeodomain transcription factor BRN3A. Here we show using specific siRNA in t(8;21) cells and overexpression studies in progenitor cells that AML1/ETO promotes expression of POU4F1/BRN3A. This effect requires DNA-binding function of AML1/ETO, and accordingly, AML1/ETO is bound to the POU4F1 locus in t(8;21) cells. Functionally, whereas overexpression of Brn3a in murine hematopoietic progenitor cells induces terminal myeloid differentiation, coexpression of AML1/ETO or AML1/ETO9a blocks this effect. Furthermore, Brn3a reduction by shRNA impairs AML1/ETO-induced immortalization of murine progenitors. In summary, we identify POU4F1/BRN3A as a novel potential upregulated AML1/ETO target gene whose dramatically high expression may cooperate with AML1/ETO in t(8;21) cells. Aims. We produce a clean and well-characterised catalogue of objects within 100 pc of the Sun from the Gaia Early Data Release 3. We characterise the catalogue through comparisons to the full data release, external catalogues, and simulations. We carry out a first analysis of the science that is possible with this sample to demonstrate its potential and best practices for its use., Methods. Theselection of objects within 100 pc from the full catalogue used selected training sets, machine-learning procedures, astrometric quantities, and solution quality indicators to determine a probability that the astrometric solution is reliable. The training set construction exploited the astrometric data, quality flags, and external photometry. For all candidates we calculated distance posterior probability densities using Bayesian procedures and mock catalogues to define priors. Any object with reliable astrometry and a non-zero probability of being within 100 pc is included in the catalogue., Results. We have produced a catalogue of 331 312 objects that we estimate contains at least 92% of stars of stellar type M9 within 100 pc of the Sun. We estimate that 9% of the stars in this catalogue probably lie outside 100 pc, but when the distance probability function is used, a correct treatment of this contamination is possible. We produced luminosity functions with a high signal-to-noise ratio for the main-sequence stars, giants, and white dwarfs. We examined in detail the Hyades cluster, the white dwarf population, and wide-binary systems and produced candidate lists for all three samples. We detected local manifestations of several streams, superclusters, and halo objects, in which we identified 12 members of Gaia Enceladus. We present the first direct parallaxes of five objects in multiple systems within 10 pc of the Sun., Conclusions. We provide the community with a large, well-characterised catalogue of objects in the solar neighbourhood. This is a primary benchmark for measuring and understanding fundamental parameters and descriptive functions in astronomy.

Aims. We aim to demonstrate the scientific potential of the Gaia Early Data Release 3 (EDR3) for the study of different aspects of the Milky Way structure and evolution and we provide, at the same time, a description of several practical aspects of the data and examples of their usage., Methods. We used astrometric positions, proper motions, parallaxes, and photometry from EDR3 to select different populations and components and to calculate the distances and velocities in the direction of the anticentre. In this direction, the Gaia astrometric data alone enable the calculation of the vertical and azimuthal velocities; also, the extinction is relatively low compared to other directions in the Galactic plane. We then explore the disturbances of the current disc, the spatial and kinematical distributions of early accreted versus in situ stars, the structures in the outer parts of the disc, and the orbits of open clusters Berkeley 29 and Saurer 1., Results. With the improved astrometry and photometry of EDR3, we find that: (i) the dynamics of the Galactic disc are very complex with oscillations in the median rotation and vertical velocities as a function of radius, vertical asymmetries, and new correlations, including a bimodality with disc stars with large angular momentum moving vertically upwards from below the plane, and disc stars with slightly lower angular momentum moving preferentially downwards; (ii) we resolve the kinematic substructure (diagonal ridges) in the outer parts of the disc for the first time; (iii) the red sequence that has been associated with the proto-Galactic disc that was present at the time of the merger with Gaia-Enceladus-Sausage is currently radially concentrated up to around 14 kpc, while the blue sequence that has been associated with debris of the satellite extends beyond that; (iv) there are density structures in the outer disc, both above and below the plane, most probably related to Monoceros, the Anticentre Stream, and TriAnd, for which the Gaia data allow an exhaustive selection of candidate member stars and dynamical study; and (v) the open clusters Berkeley 29 and Saurer 1, despite being located at large distances from the Galactic centre, are on nearly circular disc-like orbits., Conclusions. Even with our simple preliminary exploration of the Gaia EDR3, we demonstrate how, once again, these data from the European Space Agency are crucial for our understanding of the different pieces of our Galaxy and their connection to its global structure and history.

In Las Cumbres Observatory’s global network, the 1-meter telescopes are in greatest demand. Beginning in 2019, we undertook an investigation into the focus stability of these telescopes. We also refined the procedures for setting and maintaining optimal focus. The investigation showed the sensitivity of the telescopes’ optical components to temperature variations. The temperatures within the domes, as well as along the telescopes, are rarely stable for the first 80-120 minutes of science observations. A consequence is that start-of-night focus corrections must later be countered by additional corrections after thermalization. We report the various improvements to telescope focus that our investigation has spawned. The start-of-night focus corrections are now made immediately before science observations begin. Focus checks are now monitored during the night and trigger actionable alerts if they fail. Finally, we report on our effort to use the guide cameras as real-time focus sensors.

Context. Gaia Early Data Release 3 (Gaia EDR3) provides accurate astrometry for about 1.6 million compact (QSO-like) extragalactic sources, 1.2 million of which have the best-quality five-parameter astrometric solutions., Aims. The proper motions of QSO-like sources are used to reveal a systematic pattern due to the acceleration of the solar systembarycentre with respect to the rest frame of the Universe. Apart from being an important scientific result by itself, the acceleration measured in this way is a good quality indicator of the Gaia astrometric solution., Methods. Theeffect of the acceleration was obtained as a part of the general expansion of the vector field of proper motions in vector spherical harmonics (VSH). Various versions of the VSH fit and various subsets of the sources were tried and compared to get the most consistent result and a realistic estimate of its uncertainty. Additional tests with the Gaia astrometric solution were used to get a better idea of the possible systematic errors in the estimate., Results. Our best estimate of the acceleration based on Gaia EDR3 is (2.32 +/- 0.16) x 10(-10) m s(-2) (or 7.33 +/- 0.51 km s(-1) Myr-1) towards alpha = 269.1 degrees +/- 5.4 degrees, delta = -31.6 degrees +/- 4.1 degrees, corresponding to a proper motion amplitude of 5.05 +/- 0.35 mu as yr(-1). This is in good agreement with the acceleration expected from current models of the Galactic gravitational potential. We expect that future Gaia data releases will provide estimates of the acceleration with uncertainties substantially below 0.1 mu as yr(-1).

We present observations of comet 67P/Churyumov-Gerasimenko acquired in support of the $Rosetta$ mission. We obtained usable data on 68 nights from 2014 September until 2016 May, with data acquired regularly whenever the comet was observable. We collected an extensive set of near-IR $J$, $H$, and $Ks$ data throughout the apparition plus visible-light images in $g'$, $r'$, $i'$, and $z'$ when the comet was fainter. We also obtained broadband $R$ and narrowband $CN$ filter observations when the comet was brightest using telescopes at Lowell Observatory. The appearance was dominated by a central condensation and the tail until 2015 June. From 2015 August onwards there were clear asymmetries in the coma, which enhancements revealed to be due to the presence of up to three features (i.e., jets). The features were similar in all broadband filters; $CN$ images did not show these features but were instead broadly enhanced in the southeastern hemisphere. Modeling using the parameters from Vincent et al. (2013) replicated the dust morphology reasonably well, indicating that the pole orientation and locations of active areas have been relatively unchanged over at least the last three apparitions. The dust production, as measured by $A(0^{\circ})f{\rho}$ peaked $\sim$30 days after perihelion and was consistent with predictions from previous apparitions. $A(0^{\circ})f{\rho}$ as a function of heliocentric distance was well fit by a power-law with slope $-$4.2 from 35-120 days post-perihelion. We detected photometric evidence of apparent outbursts on 2015 August 22 and 2015 September 19, although neither was discernible morphologically in this dataset., Comment: Accepted by MNRAS; 15 pages, 6 figures, 2 tables

On 15 February 2013, the asteroid 367943 Duende (2012 DA14) experienced a near-Earth encounter at an altitude of 27,700 km or 4.2 Earth radii. We present here the results of an extensive, multi-observatory campaign designed to probe for spectral and/or rotational changes to Duende due to gravitational interactions with the Earth during the flyby. Our spectral data reveal no changes within the systematic uncertainties of the data. Post-flyby lightcurve photometry places strong constraints on the rotation state of Duende, showing that it is in non-principal axis rotation with fundamental periods of P_1 = 8.71 +/- 0.03 and P_2 = 23.7 +/- 0.2 hours. Multiple lightcurve analysis techniques, coupled with theoretical considerations and delay-doppler radar imaging, allows us to assign these periods to specific rotational axes of the body. In particular we suggest that Duende is now in a non-principal, short axis mode rotation state with a precessional period equal to P_1 and oscillation about the symmetry axis at a rate equal to P_2. Temporal and signal-to-noise limitations inherent to the pre-flyby photometric dataset make it difficult to definitively diagnose whether these periods represent a change imparted due to gravitational torques during the flyby. However, based on multiple analysis techniques and a number of plausibility arguments, we suggest that Duende experienced a rotational change during the planetary encounter with an increase in its precessional rotation period. Our preferred interpretation of the available data is that the precession rate increased from 8.4 hours prior to the flyby to 8.7 hours afterwards. A companion paper by Benson et al. (2019) provides a more detailed dynamical analysis of this event and compares the data to synthetic lightcurves computed from a simple shape model of Duende. Work in time-domain astronomy necessitates robust, automated data processing pipelines that operate in real time. We present the BANZAI pipeline which processes the thousands of science images produced across the Las Cumbres Observatory Global Telescope (LCOGT) network of robotic telescopes each night. BANZAI is designed to perform near real-time preview and end-of-night final processing for four types of optical CCD imagers on the three LCOGT telescope classes. It performs instrumental signature removal (bad pixel masking, bias and dark removal, flat-field correction), astrometric fitting and source catalog extraction. We discuss the design considerations for BANZAI, including testing, performance, and extensibility. BANZAI is integrated into the observatory infrastructure and fulfills two critical functions: (1) real-time data processing that delivers data to users quickly and (2) derive metrics from those data products to monitor the health of the telescope network. In the era of time-domain astronomy, to get from these observations to scientific results, we must be able to automatically reduce data with minimal human interaction, but still have insight into the data stream for quality control., 9 Pages, 6 figures

A stellar occultation by the large trans-Neptunian object (90482) Orcus was predicted to occur on 2017 March 07. Observations were made at five sites in North and South America. High-speed, visible-wavelength images were taken at all sites, in addition to simultaneous K-band images at one location. Solid-body occultations were observed from two sites. Post-event reconstruction suggested an occultation of two different stars observed from two different sites. Follow-up, speckle imaging at Gemini Observatory revealed a second star, which verified that the occulting body in both cases was Orcus` satellite, Vanth. The two single-chord detections, with an anomalously large timing delay in one chord, have lengths of 291+/-125 km and 434.4+/-2.4 km. The observations, combined with a non-detection at a nearby site, allow a tight constraint of 443+/-10 km to be placed on Vanth`s size (assuming it is spherical). A 3-{\sigma} upper limit of 1-4 {\mu}bar (depending on constituent) is found for a global Vanth atmosphere. The immersion and emersion profiles are slightly different, with atmospheric constraints 40 percent higher on immersion than on emersion. No rings or other material were detected within ten thousand kms of Vanth, and beyond 8010 km from Orcus, to the tightest optical depth limit of approximately 0.1 at 5 km scale. The occultation probed as close as 5040 km from Orcus, placing an optical depth limit of approximately 0.3 at 5 km scale on any encircling material at that distance., Comment: 37 Pages; 12 Figures; 3 Tables

The purpose of this work was to modernize recommendations for evaluation, staging, and response assessment of patients with Hodgkin lymphoma (HL) and non-Hodgkin lymphoma (NHL). A workshop was held at the 11th International Conference on Malignant Lymphoma in Lugano, Switzerland, in June 2011, that included leading hematologists, oncologists, radiation oncologists, pathologists, radiologists, and nuclear medicine physicians, representing major international lymphoma clinical trials groups and cancer centers. Clinical and imaging subcommittees presented their conclusions at a subsequent workshop at the 12th International Conference on Malignant Lymphoma, leading to revised criteria for staging and of the International Working Group Guidelines of 2007 for response. As a result, fluorodeoxyglucose (FDG) positron emission tomography (PET)–computed tomography (CT) was formally incorporated into standard staging for FDG-avid lymphomas. A modification of the Ann Arbor descriptive terminology will be used for anatomic distribution of disease extent, but the suffixes A or B for symptoms will only be included for HL. A bone marrow biopsy is no longer indicated for the routine staging of HL and most diffuse large B-cell lymphomas. However, regardless of stage, general practice is to treat patients based on limited (stages I and II, nonbulky) or advanced (stage III or IV) disease, with stage II bulky disease considered as limited or advanced disease based on histology and a number of prognostic factors. PET-CT will be used to assess response in FDG-avid histologies using the 5-point scale. The product of the perpendicular diameters of a single node can be used to identify progressive disease. Routine surveillance scans are discouraged. These recommendations should improve evaluation of patients with lymphoma and enhance the ability to compare outcomes of clinical trials.

Las Cumbres Observatory Global Telescope Network (LCOGT) has deployed a homogeneous telescope network of nine 1-meter telescopes to four locations in the northern and southern hemispheres, with a planned network size of twelve 1-meter telescopes at 6 locations. This network is very versatile and is designed to respond rapidly to target of opportunity events and also to perform long term monitoring of slowly changing astronomical phenomena. The global coverage of the network and the apertures of telescope available make LCOGT ideal for follow-up and characterization of Solar System objects (e.g. asteroids, Kuiper Belt Objects, comets, Near-Earth Objects (NEOs)) and additionally for the discovery of new objects.We are using the LCOGT network to confirm newly detected NEO candidates produced by the major sky surveys such as Catalina Sky Survey (CSS) and PanSTARRS (PS1&2) and several hundred targets are now being followed per year. An increasing amount of time is being spent to obtain follow-up astrometry and photometry for radar-targeted objects and those on the Near-Earth Object Human Space Flight Accessible Targets Study (NHATS) or Asteroid Retrieval Mission (ARM) lists in order to improve the orbits, determine the light curves and rotation periods and improve the characterization. This will be extended to obtain more light curves of other NEOs which could be targets. Recent results have included the first period determinations for several of the Goldstone-targeted NEOs. We are in the process of building a NEO follow-up portal which will allow professionals, amateurs and Citizen Scientists to plan, schedule and analyze NEO imaging and spectroscopy observations and data using the LCOGT Network and to act as a co-ordination hub for the NEO follow-up efforts.

Gain of function mutations in the H3K27 methyltransferase EZH2 represent a promising therapeutic target in germinal center lymphomas. In this study, we assessed the frequency and distribution of EZH2 mutations in a large cohort of patients with follicular lymphoma (FL) (n = 366) and performed a longitudinal analysis of mutation during the disease progression from FL to transformed FL (tFL) (n = 33). Mutations were detected at 3 recurrent mutation hot spots (Y646, A682, and A692) in 27% of FL cases with variant allele frequencies (VAF) ranging from 2% to 61%. By comparing VAF of EZH2 with other mutation targets (CREBBP, MLL2, TNFRSF14, and MEF2B), we were able to distinguish patients harboring clonal EZH2 mutation from rarer cases with subclonal mutations. Overall, the high incidence of EZH2 mutations in FL and their stability during disease progression makes FL an appropriate disease to evaluate EZH2 targeted therapy.

Background: Women treated with supradiaphragmatic radiotherapy (sRT) for Hodgkin lymphoma (HL) at young ages have a substantially increased breast cancer risk. Little is known about how menarcheal and reproductive factors modify this risk. We report the discovery of WASP-10b, a new transiting extrasolar planet (ESP) discovered by the WASP Consortium and confirmed using NOT FIES and SOPHIE radial velocity data. A 3.09 day period, 29 mmag transit depth, and 2.36 hour duration are derived for WASP-10b using WASP and high precision photometric observations. Simultaneous fitting to the photometric and radial velocity data using a Markov-chain Monte Carlo procedure leads to a planet radius of 1.28R_J, a mass of 2.96M_J and eccentricity of ~0.06. WASP-10b is one of the more massive transiting ESPs, and we compare its characteristics to the current sample of transiting ESP, where there is currently little information for masses greater than ~2M_J and non-zero eccentricities. WASP-10's host star, GSC 2752-00114 (USNO-B1.0 1214-0586164) is among the fainter stars in the WASP sample, with V=12.7 and a spectral type of K5. Between 1969 and 1999, 420 patients (age > 60 years) with newly diagnosed AML were managed at St Bartholomew's Hospital (SBH), London, UK. Sixty-nine percent of patients received therapy with curative intent Eighty-eight patients (31%) of the latter achieved complete remission (CR), representing an overall CR rate of 21%. Treatment failure due to early death (ED) and resistant disease (RD) occurred in 50 and 19%, respectively. With median follow up of 11 years, actuarial survivals at 1,3 and 5 years were 20, 7 and 4%, respectively, the median survival of the entire cohort was 2 months. For patients who achieved CR, median survival was significantly better than that of patients in whom treatment failed (14 vs. 6 months). Over the 30 years, CR rate and the relative incidence of RD both increased from 13 to 45%, and 3 to 27%, respectively, whilst ED rate reduced from 84 to 27%. Multivariate analysis showed that treatment era, hepatosplenomegaly and increasing age predicted for reduced CR rate and OS. Although elderly patients with AML are characterised by a poor response to intensive chemotherapy, significant improvements in supportive care and the delivery of intensive treatment have led to improved CR rates and OS. New therapeutic strategies and a greater awareness of prognostic factors may further improve clinical outcome in this important group of patients.

Around the time of its perihelion passage the observability of 67P/Churyumov-Gerasimenko from Earth was limited to very short windows each morning from any given site, due to the low solar elongation of the comet. The peak in the comet's activity was therefore difficult to observe with conventionally scheduled telescopes, but was possible where service/queue scheduled mode was possible, and with robotic telescopes. We describe the robotic observations that allowed us to measure the total activity of the comet around perihelion, via photometry (dust) and spectroscopy (gas), and compare these results with the measurements at this time by Rosetta's instruments. The peak of activity occurred approximately two weeks after perihelion. The total brightness (dust) largely followed the predictions from Snodgrass et al. 2013, with no significant change in total activity levels from previous apparitions. Purpose To investigate breast cancer risk after supradiaphragmatic radiotherapy administered to young women with Hodgkin's lymphoma (HL) in a much larger cohort than previously to provide data for patient follow-up and screening individualized according to treatment type, age, and time point during follow-up. Patients and Methods Breast cancer risk was assessed in 5,002 women in England and Wales treated for HL with supradiaphragmatic radiotherapy at age < 36 years from 1956 to 2003, who underwent follow-up with 97% completeness until December 31, 2008. Results Breast cancer or ductal carcinoma in situ developed in 373 patients, with a standardized incidence ratio (SIR) of 5.0 (95% CI, 4.5 to 5.5). SIRs were greatest for those treated at age 14 years (47.2; 95% CI, 28.0 to 79.8) and continued to remain high for at least 40 years. The maximum absolute excess risk was at attained ages 50 to 59 years. Alkylating chemotherapy or pelvic radiotherapy diminished the risk, but only for women treated at age ≥ 20 years, not for those treated when younger. Cumulative risks were tabulated in detail; for 40-year follow-up, the risk for patients receiving ≥ 40 Gy mantle radiotherapy at young ages was 48%. Conclusion This article provides individualized risk estimates based on large numbers for patients with HL undergoing follow-up after radiotherapy at young ages. Follow-up of such women needs to continue for 40 years or longer and may require more-intensive screening regimens than those in national general population programs. Special consideration is needed of potential measures to reduce breast cancer risk for girls treated with supradiaphragmatic radiotherapy at pubertal ages.

The relative merits of reduced-intensity allogeneic stem cell transplantation (RISCT) for high-risk indolent lymphoid malignancies are emerging, although the preferred conditioning regimen to manage the risks of graft-versus-host disease (GVHD) is not clearly defined. Here we report the outcome of 73 patients with lymphoid malignancies who received RISCT with a fludarabine/cyclophosphosphamide conditioning regimen and a median follow-up of 3 years. Median age was 54 years. Forty-eight per cent of patients had previously undergone autologous stem cell transplantation with a median of three prior therapies. Non-relapse mortality at 3 years was 19% but only 5% for patients with multiple myeloma (MM). Three-year overall survival and current progression-free survival was 67% and 63% respectively. Grade 2-4 acute GVHD occurred in 14% of patients while 49% had chronic GVHD requiring systemic immunosuppression. The preparatory regimen in this study has the advantage of reduced acute GVHD and low mortality, notably in patients with MM. In addition, this strategy provides long-term disease control in a significant proportion of patients with particular benefit in those with high-risk follicular lymphoma.

Inherited risk determinants for follicular lymphoma (FL) have recently been described in the immune gene-rich human leukocyte antigen region on chromosome 6p. The known importance of host immune response to FL survival led us to evaluate these germline factors in FL outcome. We confirm the association of single nucleotide polymorphisms rs10484561 (P = 3.5 × 10−9) and rs6457327 (P = .008) with risk of FL and demonstrate that rs6457327 predicts both time to (P = .02) and risk of (P < .01) FL transformation independently of clinical variables, including the Follicular Lymphoma International Prognostic Index.

Follicular lymphoma has considerable clinical heterogeneity, and there is a need for easily quantifiable prognostic biomarkers. Microvessel density has been shown to be a useful prognostic factor based on numerical assessment of vessel numbers within histologic sections in some studies, but assessment of tumor neovascularization through angiogenic sprouting may be more relevant. We therefore examined the smallest vessels, single-staining structures measuring less than 30 μm2 in area, seen within histologic sections, and confirmed that they were neovascular angiogenic sprouts using extended focal imaging. Tissue microarrays composing diagnostic biopsies from patients at the extremes of survival of follicular lymphoma were analyzed with respect to numbers of these sprouts. This analysis revealed higher angiogenic activity in the poor prognostic group and demonstrated an association between increased sprouting and elevated numbers of infiltrating CD163+ macrophages within the immediate microenvironment surrounding the neovascular sprout.

Leukemia-initiating cells (LICs) in acute myeloid leukemia (AML) are believed to be restricted to the CD34+ fraction. However, one of the most frequently mutated genes in AML is nucleophosmin (NPM), and this is associated with low CD34 expression. We, therefore, investigated whether NPM-mutated AMLs have LICs restricted to the CD34+ fraction. We transplanted sorted fractions of primary NPM-mutated AML into immunodeficient mice to establish which fractions initiate leukemia. Approximately one-half of cases had LICs exclusively within the CD34− fraction, whereas the CD34+ fraction contained normal multilineage hematopoietic repopulating cells. Most of the remaining cases had LICs in both CD34+ and CD34− fractions. When samples were sorted based on CD34 and CD38 expression, multiple fractions initiated leukemia in primary and secondary recipients. The data indicate that the phenotype of LICs is more heterogeneous than previously realized and can vary even within a single sample. This feature of LICs may make them particularly difficult to eradicate using therapies targeted against surface antigens.

We report the discovery of a new transiting close-in giant planet, WASP-24 b, in a 2.341 day orbit, 0.037 AU from its F8-9 type host star. By matching the star's spectrum with theoretical models, we infer an effective temperature T-eff = 6075 +/- 100 K and a surface gravity of log g = 4.15 +/- 0.10. A comparison of these parameters with theoretical isochrones and evolutionary mass tracks places only weak constraints on the age of the host star, which we estimate to be 3.8(-1.2)(+1.3) Gyr. The planetary nature of the companion was confirmed by radial velocity measurements and additional photometric observations. These data were fit simultaneously in order to determine the most probable parameter set for the system, from which we infer a planetary mass of 1.071(-0.038)(+0.036) M-Jup and radius 1.3(-0.037)(+0.039) R-Jup.

To date, studies on T cells in acute myeloid leukemia (AML) have been limited to flow cytometric analysis of whole peripheral blood mononuclear cell (PBMC) specimens or functional work looking at the impact of AML myeloblasts on normal or remission T cells. This lack of information on T cells at the time of presentation with disease is due in part to the difficulty in isolating sufficiently pure T cells from these specimens for further study. Negative immunomagnetic selection has been the method of choice for isolating immune cells for functional studies due to concerns that binding antibodies to the cell surface may induce cellular activation, block ligand-receptor interactions or result in immune clearance. In order specifically to study T cells in presentation AML specimens, we set out to develop a method of isolating highly pure CD4 and CD8 T cells by negative selection from the peripheral blood (PB) of newly diagnosed AML patients. This technique, unlike T cell selection from PB from normal individuals or from patients with chronic lymphocytic leukaemia, was extremely problematic due to properties of the leukaemic myeloblasts. A successful method was eventually optimized requiring the use of a custom antibody cocktail consisting of CD33, CD34, CD123, CD11c and CD36, to deplete myeloblasts.

Quantitative methylation profiling was performed using the Illumina GoldenGate Assay in untreated Follicular Lymphoma (FL) (164), paired pre- and post-transformation FL (20), benign haematopoietic (24) samples and purified B & T cells from two FL cases. Methylation values allowed separation of untreated FL samples from controls with one exception based primarily on tumour-specific gains of methylation typically occurring within CpG islands. Genes which are targets for epigenetic repression in stem cells by Polycomb Repressor Complex 2 were significantly overrepresented among hypermethylated genes. 191 combined collections of granulocytes (PMN) and platelets (PLT) obtained by discontinuous cytapheresis were given to 23 patients with acute myeloid leukaemia (AML). The circulation in recipients of the concomitantly transfused cells was compared by measuring the % recovery of PMN at 1 h, and of PLT at 1 and 20 h. The recovery of both cells was greatly reduced in transfusions accompanied by a transfusion reaction. Fever in recipients had no effect on the recovery of either cell, but PMN recovery was significantly higher in patients receiving oral prednisolone. Alloimmunization to PLT, defined as recovery levels less than 15% at 1 h and/or less than 10% at 20 h, was recorded in 42 transfusions, and PMN recovery was significantly reduced in this group. In transfusions where PMN recovery was 'low' (less than 5%); the recovery of the coincidentally transfused PLT was significantly reduced only in transfusions accompanied by a reaction. These results emphasize the deleterious effects of transfusion reactions and suggest that donor PMN frequently circulate less well in recipients alloimmunized to PLT from the same donor.

The t(14;18)(q32;q21) chromosomal translocation induces BCL2 protein overexpression in most follicular lymphomas. However the expression of BCL2 is not always homogeneous and may demonstrate a variable degree of heterogeneity. This study analysed BCL2 protein expression pattern in 33 cases of t(14;18)-positive follicular lymphomas using antibodies against two different epitopes (i.e. the widely used antibody BCL2/124 and an alternative antibody E17). 16/33 (49%) cases demonstrated strong BCL2 expression. In 10/33 (30%) cases, BCL2 expression was heterogeneous and in some of these, its loss appeared to be correlated with cell proliferation, as indicated by Ki67 expression. Double immunofluorescence labelling confirmed an inverse BCL2/Ki67 relationship, where in 24/28 (86%) cases cellular expression of BCL2 and Ki67 was mutually exclusive. In addition, seven BCL2 'pseudo-negative' cases were identified in which immunostaining was negative with antibody BCL2/124, but positive with antibody E17. Genomic DNA sequencing of these 'pseudo-negative' cases demonstrated eleven mutations in four cases and nine of these were missense mutations. It can be concluded that in follicular lymphomas, despite carrying the t(14;18) translocations, BCL2 protein expression may be heterogeneous and loss of BCL2 could be related to cell proliferation. Secondly, mutations in translocated BCL2 genes appear to be common and may cause BCL2 pseudo-negative immunostaining.

Purpose To determine the clinical relevance of Wilms’ tumor 1 (WT1) gene mutations in acute myeloid leukemia (AML) with normal karyotype (NK). Patients and Methods Exons 7 and 9 of WT1 were screened in samples from 470 young adult NK AMLs using a combination of direct sequencing and high-resolution capillary electrophoresis. Results Overall, 51 mutations were detected in 47 cases (10%): 46 frameshift mutations with insertion/deletion of one to 28 base pairs in exon 7 (n = 45) or exon 9 (n = 1), with a median mutant level of 45% (range, 8% to 86%), and five substitutions in exon 9: D396N (n = 3), H397Y (n = 1) and H397Q (n = 1). Patients with WT1 mutations had an inferior response to induction chemotherapy compared with wild-type cases (complete remission rate, 79% v 90%, odds ratio [OR] = 3.02; 95% CI, 1.17 to 7.82; P = .02), a higher rate of resistant disease (15% v 4%; OR = 9.33; 95% CI, 2.38 to 36.6; P = .001), an increased cumulative incidence of relapse (67% v 43%, hazard ratio [HR] = 3.02; 95% CI, 1.69 to 5.38; P = .0008), with a reduction in both relapse-free survival (22% v 44%; HR = 2.16; 95% CI, 1.32 to 3.55; P = .005) and overall survival (26% v 47%; HR = 1.91; 95% CI, 1.23 to 2.95; P = .007) at 5 years. In multivariate analysis, which included FLT3 internal tandem duplication and NPM1 mutation status, the presence of a WT1 mutation remained an independent adverse prognostic factor. Conclusion WT1 mutations are a negative prognostic indicator in NK AML and may be suitable for the development of targeted therapy.

A platelet cross-matching procedure has been assessed for selecting compatible donors for alloimmunized patients. This confirms the clinical value of combining an indirect platelet immunofluorescence test (PIFT) with a lymphocytotoxicity test (LCT) in predicting the survival of single-donor platelets. There was good agreement between the PIFT cross-match and post-transfusion platelet recovery. Compatibility in the LCT alone was insufficient for platelet donor selection, as this test did not detect all antibodies affecting platelet survival. Positive LCT and PIFT cross-matches indicated the presence of HLA antibodies. Inclusion of an indirect lymphocyte immunofluorescence test (LIFT) helped to classify the platelet antibody when the LCT cross-match was negative. In such cases, parallel positive findings with the LIFT and PIFT suggested a cytotoxic-negative antibody of probable HLA specificity active against platelets. Disparity between the LIFT and PIFT was also observed; a strongly positive PIFT along with a weak reaction in the LIFT suggested that a platelet-specific antibody was responsible for the poor platelet survival in these cases. This study has also shown the presence in multitransfused patients of LIFT-positive antibodies not reacting in the LCT and PIFT, which do not affect the survival of transfused platelets. A positive granulocyte cross-match was demonstrated in patients with febrile rigors associated with compatible platelet transfusions. Splenectomy and steroids may improve the survival of incompatible platelets depending on the nature of the platelet antibody.

37. Co-ordinated Follow-Up of Transiting Planet Candidates with Robotic Telescope Facilities

There are now several large photometric surveys scanning millions of stellar light-curves for signs of planetary transits. All produce large candidate lists with a high false alarm rate, so that further observations are required to confirm new detections. One such survey, SuperWASP, produced ~150 candidates during the 2007–2008 season. Here we describe our campaign to follow-up 86 of these candidates using the robotic facilities of Las Cumbres Observatory Global Telescope Network and the Tenagra-II robotic telescope in Arizona. The aim of these observations was to eliminate false positives as far as possible ahead of spectroscopic follow-up and to provide additional photometry to help characterise the surviving targets.

Although recent reports suggest that selenium can modulate the activity of cytotoxic drugs, the mechanism underlying this activity remains unclear. This has been investigated using a panel of human B-cell lymphoma cell lines. The cytotoxic effects of chemotherapeutic agents (e.g., doxorubicin, etoposide, 4-hydroperoxycyclophosphamide, melphalan, and 1-β-d-arabinofuranosylcytosine) were increased by up to 2.5-fold when combined with minimally toxic concentrations (EC5-10) of the organic selenium compound, methylseleninic acid (MSA). DNA strand breaks were identified using comet assays, but the measured genotoxic activity of the combinations did not explain the observed synergistic effects in cell death. However, minimally toxic (EC10) concentrations of MSA induced a 50% decrease in nuclear factor-κB (NF-κB) activity after an exposure of 5 h, similar to that obtained with the specific NF-κB inhibitor, BAY 11-7082. Combinations of BAY 11-7082 with these cytotoxic drugs also resulted in synergism, suggesting that the chemosensitizing activity of MSA is mediated, at least in part, by its effects on NF-κB. Basal intracellular selenium concentration was higher in a MSA-sensitive cell line. After exposure to MSA, methylselenocysteine and selenomethionine were identified as the main intracellular species generated. Volatile selenium species, trapped using solid-phase microextraction fibers, were identified as dimethylselenide and dimethyldiselenide. These volatile species are thought to be the most biologically active forms of selenium. Taken together, these results show that the NF-κB pathway is one target for MSA underlying the interaction between MSA and chemotherapy. These data encourage the further clinical development of selenium as a potential modulator of cytotoxic drug activity in B-cell lymphomas. [Cancer Res 2007;67(22):10984–92]

Transiting extrasolar planets constitute only a small fraction of the range of stellar systems found to display periodic, shallow dimmings in wide-field surveys employing small-aperture camera arrays. Here we present an efficient selection strategy for follow-up observations, derived from analysis of the light curves of a sample of 67 SuperWASP targets that passed the selection tests we used in earlier papers, but which have subsequently been identified either as planet hosts or as astrophysical false positives. We determine the system parameters using Markov-chain Monte Carlo analysis of the SuperWASP light curves. We use a constrained optimisation of chi-squared combined with a Bayesian prior based on the main-sequence mass and radius expected from the 2MASS J-H colour. The Bayesian nature of the analysis allows us to quantify both the departure of the host star from the main-sequence mass-radius relation and the probability that the companion radius is less than 1.5 Jupiter radii. When augmented by direct light curve analyses that detect binaries with unequal primary and secondary eclipses, and objects with aperture blends that are resolved by SuperWASP, we find that only 13 of the original 67 stars, including the three known planets in the sample, would qualify for follow-up. This suggests that planet discovery "hit rates" better than one-in-five should be achievable. In addition, the stellar binaries that qualify are likely to have astrophysically interesting stellar or sub-stellar secondaries.

We have performed photometric observations of nearly 7 million stars with 8 < V < 15 with the SuperWASP-North instrument from La Palma between 2004 May-September. Fields in the RA range 17-18hr, yielding over 185,000 stars with sufficient quality data, have been searched for transits using a modified box least-squares (BLS) algorithm. We find a total of 58 initial transiting candidates which have high S/N in the BLS, show multiple transit-like dips and have passed visual inspection. Analysis of the blending and inferred planetary radii for these candidates leaves a total of 7 transiting planet candidates which pass all the tests plus 4 which pass the majority. Purpose The aim of this retrospective analysis was to determine the outcome of patients with follicular lymphoma who received myeloablative therapy supported by autologous bone marrow transplantation as consolidation of second or subsequent remission, with a minimum follow-up of 12 years. Patients and Methods One hundred twenty-one adults received cyclophosphamide (CY) and total-body irradiation (TBI) supported by autologous bone marrow transplantation, with the marrow mononuclear cell fraction having been treated with monoclonal antibodies and complement. Data from St Bartholomew's Hospital and Dana-Farber Cancer Institute were combined for the purpose of this analysis because the patients were treated in an identical manner. Results Fifty-seven patients are alive, 41 without progression between 9 and 19 years; 64 patients have died, 20 without progression. With a median follow-up of 13.5 years, 60 patients have developed recurrent lymphoma. There is an apparent plateau on the remission duration curve at 48% at 12 years. Survival of patients treated in second remission was significantly longer than the survival of patients treated later in the course of the illness. Both remission duration and overall survival were also significantly longer for patients treated in second remission compared with an age-matched, remission-matched group of patients treated at St Bartholomew's Hospital before the introduction of this treatment. However, use of CY+TBI was associated with a significant risk of secondary myelodysplasia and secondary acute myeloblastic leukemia, resulting in 15 patient deaths. Conclusion These mature data confirm that prolonged freedom from recurrence may be achieved with myeloablative therapy and that a plateau on the curve seems to emerge with long follow-up.

Predictive mapping of indoor radon potential often requires the use of additional datasets. A range of geological, geochemical and geophysical data may be considered, either individually or in combination. The present work is an evaluation of how much of the indoor radon variation in south west England can be explained by four different datasets: a) the geology (G), b) the airborne gamma-ray spectroscopy (AGR), c) the geochemistry of topsoil (TSG) and d) the geochemistry of stream sediments (SSG). The study area was chosen since it provides a large (197,464) indoor radon dataset in association with the above information. Geology provides information on the distribution of the materials that may contribute to radon release while the latter three items provide more direct observations on the distributions of the radionuclide elements uranium (U), thorium (Th) and potassium (K). In addition, (c) and (d) provide multi-element assessments of geochemistry which are also included in this study. The effectiveness of datasets for predicting the existing indoor radon data is assessed through the level (the higher the better) of explained variation (% of variance or ANOVA) obtained from the tested models. A multiple linear regression using a compositional data (CODA) approach is carried out to obtain the required measure of determination for each analysis. Results show that, amongst the four tested datasets, the soil geochemistry (TSG, i.e. including all the available 41 elements, 10 major – Al, Ca, Fe, K, Mg, Mn, Na, P, Si, Ti - plus 31 trace) provides the highest explained variation of indoor radon (about 40%); more than double the value provided by U alone (ca. 15%), or the sub composition U, Th, K (ca. 16%) from the same TSG data. The remaining three datasets provide values ranging from about 27% to 32.5%. The enhanced prediction of the AGR model relative to the U, Th, K in soils suggests that the AGR signal captures more than just the U, Th and K content in the soil. The best result is obtained by including the soil geochemistry with geology and AGR (TSG + G + AGR, ca. 47%). However, adding G and AGR to the TSG model only slightly improves the prediction (ca. +7%), suggesting that the geochemistry of soils already contain most of the information given by geology and airborne datasets together, at least with regard to the explanation of indoor radon. From the present analysis performed in the SW of England, it may be concluded that each one of the four datasets is likely to be useful for radon mapping purposes, whether alone or in combination with others. The present work also suggest that the complete soil geochemistry dataset (TSG) is more effective for indoor radon modelling than using just the U (+Th, K) concentration in soil.

One hundred and seven untreated patients with acute myelogenous leukemia (AML) were admitted to St. Bartholomew's Hospital between the 10th October 1970 and the 31st January 1973. Before receiving drugs to induce remission they were allocated alternatively into 2 groups to decide their remission treatment, a group to receive chemotherapy alone and a group to receive the same chemotherapy with immunotherapy. The patients were then given induction chemotherapy and 45 of them attained complete remission. All patients in remission then received chemotherapy consisting of 5 days treatment every 28 days. Patients receiving immunotherapy were also given multiple weekly intradermal injections of irradiated stored AML cells and Glaxo BCG using a Heaf gun. There were 19 patients in the group which received only chemotherapy during remission; 7 of these patients remain alive (median survival after attaining remission--303 days) and only 5 are still in their first remission (median remission length 188 days). Twenty three patients were allocated to receive immunotherapy during remission in addition to chemotherapy and 16 remain alive (median 545 days) and 8 are in their first remission (median 312 days). The difference in survival of the 2 groups is significant with a sigma value of 0.003.

We present a model of the stellar populations in the fields observed by one of the SuperWASP-N cameras in the 2004 observing season. We use the Besancon Galactic model to define the range of stellar types and metallicities present, and populate these objects with transiting extra-solar planets using the metallicity relation of Fischer & Valenti (2005). We investigate the ability of SuperWASP to detect these planets in the presence of realistic levels of correlated systematic noise (`red noise'). We find that the number of planets that transit with a signal-to-noise ratio of 10 or more increases linearly with the number of nights of observations. Based on a simulation of detection rates across 20 fields observed by one camera, we predict that a total of 18.6 \pm 8.0 planets should be detectable from the SuperWASP-N 2004 data alone. The best way to limit the impact of co-variant noise and increase the number of detectable planets is to boost the signal-to-noise ratio, by increasing the number of observed transits for each candidate transiting planet. Purpose To examine the immune microenvironment in diagnostic follicular lymphoma (FL) biopsies and evaluate its prognostic significance. Patients and Methods Immunohistochemistry was used to study numbers and location of cells staining positive for immune cell markers CD4, CD7, CD8, CD25, CD68, forkhead box protein P3 (FOXP3), T-cell intracellular antigen-1, and Granzyme B in tissue microarrays of paraffin-embedded, diagnostic lymph node biopsies taken from 59 FL patients who lived less than 5 years (short-survival group; n = 34) and more than 15 years (long-survival group; n = 25). Results CD4 and FOXP3 expression were significantly different between the two groups. Samples from the long-survival group were more likely than those from the short-survival group to have CD4+ staining cells and to have FOXP3-positive cells in a perifollicular location. Conclusion This study has identified differences in immune cell composition of the diagnostic FL lymph node immune microenvironment and these have the potential for use as prognostic biomarkers in a routine histopathology setting.

Fourteen patients with Hodgkin's disease (two previously untreated, 12 following relapse or with refractory disease) were treated with a combination chemotherapy regimen comprising chlorambucil, vinblastine, procarbazine, prednisolone, etoposide, vincristine and adriamycin administered on days 1-8. Recombinant human granulocyte-macrophage colony stimulating factor (rhGM-CSF) (mammalian glycosylated, Sandoz/Schering-Plough) was administered after alternate cycles of chemotherapy from day 10 for 7 days by continuous intravenous (i.v.) infusion in 12 patients in a dose finding study (dose: 2 micrograms/kg/day in four patients, 4 micrograms/kg/day in four patients and 8 micrograms/kg/day in four patients) and by daily subcutaneous (s/c) injections in two patients (8 micrograms/kg/day). There was a rapid peripheral leucocytosis following the rhGM-CSF, reaching a peak at 1-2 days in 12/14 patients. The initial leucocytosis was composed of neutrophils followed by a rise in immature myeloid cells. There was no difference observed in the duration or depth of the nadir following chemotherapy or in the rate of recovery of peripheral white cell counts between cycles with and without rhGM-CSF in patients treated with 2 and 4 micrograms/kg/day. At the dose of 8 micrograms/kg/day, 3/6 patients had a shorter nadir duration in the cycle with rhGM-CSF, compared with cycle without rhGM-CSF. There was no difference in frequency of infection in cycles with and without rhGM-CSF. Following chemotherapy, six patients achieved clinical remission, six partial remission and two had progressive disease.
