1. Genesis of the 1.45 Ga Kratz Spring Iron Oxide-Apatite Deposit Complex in Southeast Missouri, USA: Constraints from Oxide Mineral Chemistry

Abstract

The U.S. state of Missouri contains seven major and numerous lesser Fe oxide deposits within the 1.47 Ga St. Francois Mountains terrane. These deposits have been previously described as iron oxide-apatite (IOA) and iron oxide-copper-gold (IOCG) deposits and are speculated to contain significant resources of critical minerals, most notably rare earth elements and cobalt. One of the less-studied deposits in the region is the 1.455 Ga Kratz Spring deposit. The deposit consists of two steeply dipping magnetite bodies beneath 450 m of sedimentary cover. The genesis of the Kratz Spring deposit and its relationship to nearby IOA-IOCG deposits remains poorly constrained. To better understand the formation of the Kratz Spring deposit, we integrated stratigraphic, petrographic, and bulk rock studies with in situ trace element and Fe isotope chemistry of magnetite and hematite. Our data show that the Kratz Spring deposit is hydrothermal in origin but is divided into two subdeposits according to different fluid sources and formation conditions: (1) a deep but cooler hydrothermal Kratz Spring South deposit with a juvenile fluid source and (2) a shallow but hotter magmatic-hydrothermal Kratz Spring North deposit with variable fluid sources. Our genetic model suggests the two Kratz Spring deposits are local expressions of the same mineralization system; i.e., the Kratz Spring South deposit is a distal, lower-temperature offshoot of the feeder system that formed the Kratz Spring North deposit. Understanding the magmatic-hydrothermal plumbing system that formed Missouri’s IOA-IOCG deposits is important to guide critical mineral exploration efforts in the region.

Abstract

ObjectiveThe Seraph®100 Microbind Affinity Blood Filter® (Seraph 100) is an extracorporeal medical countermeasure that can remove many pathogens from blood, including the SARS-CoV-2 virus. The aim of this study was to evaluate safety and efficacy of Seraph 100 treatment for severe coronavirus disease 2019 (COVID-19).DesignMulticenter retrospective observational cohort study.SettingIntensive care units across four of thirteen participating sites who have completed data extraction.PatientsCritically ill COVID-19 patients treated with Seraph 100 under an Emergency Use Authorization (n=53) and historical control patients who met criteria for treatment (n=46).InterventionExtracorporeal treatment with the Seraph 100 filter.Measurements and Main ResultsAt baseline, the median age was 61 years, 72.7% were male, and 59.6% required mechanical ventilation. The groups were matched in terms of sex, race/ethnicity, body mass index, APACHE II score, need for mechanical ventilation, and other COVID-19 treatments. However, patients in the Seraph 100 group were younger with a median age of 61 years (IQR 42-65) compared to controls who had a median age of 64 (IQR 56-68, p=0.036). The Seraph 100 group also had a lower median Charlson comorbidity index (2, IQR 0-3) compared to control patients (3, IQR 2-4, p=0.006). Mortality was lower in the Seraph 100 treated group compared to the historical controls (37.7% vs 67.4%, respectively, p=0.003). Multivariable logistic regression analysis yielded an odds ratio of 0.27 (95% confidence interval 0.09-0.79, p=0.016). Of the 53 patients treated with Seraph 100, only 1 patient experienced a serious adverse event (transient hypotension at the start of the treatment which required a brief period of vasopressor support).ConclusionsThese data suggest that broad spectrum, pathogen agnostic, extracorporeal blood purification technologies can be safely and effectively deployed to meet new pathogen threats as an adjunct to standard treatments while awaiting the development of directed pharmacologic therapies and/or vaccines.

We quantified the genetic sharing of 25 brain disorders from genome-wide association studies of 265,218 patients and 784,643 control participants and assessed their relationship to 17 phenotypes from 1,191,588 individuals. Psychiatric disorders share common variant risk, whereas neurological disorders appear more distinct from one another and from the psychiatric disorders. We also identified significant sharing between disorders and a number of brain phenotypes, including cognitive measures. Further, we conducted simulations to explore how statistical power, diagnostic misclassification, and phenotypic heterogeneity affect genetic correlations. These results highlight the importance of common genetic variation as a risk factor for brain disorders and the value of heritability-based methods in understanding their etiology. 13 p.

Abstract

Item does not contain fulltext BACKGROUND: Bococizumab is a humanized monoclonal antibody that inhibits proprotein convertase subtilisin-kexin type 9 (PCSK9) and reduces levels of low-density lipoprotein (LDL) cholesterol. We sought to evaluate the efficacy of bococizumab in patients at high cardiovascular risk. METHODS: In two parallel, multinational trials with different entry criteria for LDL cholesterol levels, we randomly assigned the 27,438 patients in the combined trials to receive bococizumab (at a dose of 150 mg) subcutaneously every 2 weeks or placebo. The primary end point was nonfatal myocardial infarction, nonfatal stroke, hospitalization for unstable angina requiring urgent revascularization, or cardiovascular death; 93% of the patients were receiving statin therapy at baseline. The trials were stopped early after the sponsor elected to discontinue the development of bococizumab owing in part to the development of high rates of antidrug antibodies, as seen in data from other studies in the program. The median follow-up was 10 months. RESULTS: At 14 weeks, patients in the combined trials had a mean change from baseline in LDL cholesterol levels of -56.0% in the bococizumab group and +2.9% in the placebo group, for a between-group difference of -59.0 percentage points (P/=70 mg per deciliter [1.8 mmol per liter] and the median follow-up was 7 months), major cardiovascular events occurred in 173 patients each in the bococizumab group and the placebo group (hazard ratio, 0.99; 95% confidence interval [CI], 0.80 to 1.22; P=0.94). In the higher-risk, longer-duration trial (in which the patients had a baseline LDL cholesterol level of >/=100 mg per deciliter [2.6 mmol per liter] and the median follow-up was 12 months), major cardiovascular events occurred in 179 and 224 patients, respectively (hazard ratio, 0.79; 95% CI, 0.65 to 0.97; P=0.02). The hazard ratio for the primary end point in the combined trials was 0.88 (95% CI, 0.76 to 1.02; P=0.08). Abstract

Spinal epidural abscess (SEA) is rare, especially in children, in which fewer than 90 cases have been reported. We present three case examples illustrating the complex course before diagnosis and treatment. All patients had back pain, but two had pain of the abdomen or extremity dominating the clinical picture. Magnetic resonance imaging (MRI) was found to be the best diagnostic test. All had surgery with full recovery. A literature review of 58 cases since 1945 was done and showed that 76% of children had neurological compromise before surgery, and the overall mortality rate in children was 12%.

Abstract

Sarcomas--like leukemias, which are also mesodermal malignancies--carry biological significance disproportionate to their clinical frequency. Identification of mutations and translocations associated with these tumors has illuminated aberrant signaling pathways that cause these diseases, determine their behavior, and are therapeutic targets. Activated receptor-associated tyrosine kinase c-kit, mutated in most gastrointestinal stromal tumors, has proven a clinically effective target for enzyme inhibition. A translocation involving a single gene family, consisting of EWS and related genes, has been identified in five different sarcomas, and its chimeric protein products could prove similarly amenable to inhibitors. Resolution of the histopathological complexity is being aided by data from molecular and chromosomal characterization. Improvements in imaging, definition of prognostic factors, and surgical and radiotherapeutic treatment have resulted in improved local control. Continued progress will depend on further adapting the rapidly evolving technologies of genomics and proteomics. It will also depend upon accurate histopathological diagnosis based on validated reagents and consistent methodologies applied to adequate tissue samples derived from patients with complete clinical data. Finally, multicenter, coordinated trials, such as those that occurred with assessment of imatinib mesylate in metastatic gastrointestinal stromal tumors, will assure the most rapid reductions in morbidity and mortality.

Oxygen equilibria, ligand-binding kinetics and some other physicochemical properties are reported for erythrocruorins of two intertidal polychaetes:Marphysa sanguinea, which inhabits simple, relatively stagnant burrows, andDiopatra cuprea, which inhabits impermeable, parchment-like tubes that are vigorously ventilated.Marphysa erythrocruorin has a higher O2 affinity, which is less pH dependent (at pH 7.3 and 20øC, half-saturation O2 tension,P 50, and Bohr factor, =logP 50/pH, are 0.8 mm Hg and -0.25, respectively) than the corresponding parameters (P 50=5.5; =-0.86) inDiopatra (Figs. 1 and 2). In contrast to vertebrate haemoglobins, inorganic salts increase erythrocruorin O2 affinity (Fig. 3). The kinetic rates of ligand binding and dissociation ofMarphysa andDiopatra erythrocruorins (Tables 1 and 2) correlate well with the measured O2 affinities and appear to illustrate basic molecular adaptations of the two species to their respective micro-environmental conditions.

Abstract

We report on an emerging application focused on the design of resilient long reach passive optical networks using combinatorial optimisation techniques. The objective of the application is to determine the optimal position and capacity of a set of metro nodes. We specifically consider dual parented networks whereby each customer must be associated with two metro nodes. An important property of such a placement is resilience to single node failure. Therefore excess capacity should be provided at each metro node in order to ensure that customers can be redistributed amongst the metro sites. Our application, as well as finding optimal node placements, can compute the minimum level of excess capacity on all metro nodes. In this paper we present three alternative approaches to optimal metro node placement. We present a detailed analysis of the impact of different placement approaches on the distribution of excess capacity throughout the network. We show that preferential distributions occur in practice, based on a case-study in Ireland. Finally we show that load and excess capacity provision are independent of each other.

Abstract

Preference Inference involves inferring additional user preferences from elicited or observed preferences, based on assumptions regarding the form of the user's preference relation. In this paper we consider a situation in which alternatives have an associated vector of costs, each component corresponding to a different criterion, and are compared using a kind of lexicographic order, similar to the way alternatives are compared in a Hierarchical Constraint Logic Programming model. It is assumed that the user has some (unknown) importance ordering on criteria, and that to compare two alternatives, firstly, the combined cost of each alternative with respect to the most important criteria are compared; only if these combined costs are equal, are the next most important criteria considered. The preference inference problem then consists of determining whether a preference statement can be inferred from a set of input preferences. We show that this problem is coNP-complete, even if one restricts the cardinality of the equal-importance sets to have at most two elements, and one only considers non-strict preferences. However, it is polynomial if it is assumed that the user's ordering of criteria is a total ordering; it is also polynomial if the sets of equally important criteria are all equivalence classes of a given fixed equivalence relation. We give an efficient polynomial algorithm for these cases, which also throws light on the structure of the inference.
