Following the trend of finding better thermoelectric materials among synthetic analogs of copper–chalcogenide minerals, we have synthesized iron-bearing colusites of a general formula Cu26−xFexV2Sn6S32. They crystallize in the cubic space group P-43n with the unit cell parameter increasing linearly with the iron content. At a low iron concentration, the crystal structure features disorder manifested by an anti-site effect and a shift of a part of the tin atoms from their ideal positions, which is absent for higher iron contents. The magnetization and 57Fe/119Sn Mössbauer studies showed that, for x = 1, iron is present as Fe3+, whereas for x > 1, Fe2+ and Fe3+ coexist. Additionally, weak antiferromagnetic interactions between iron atoms and fast on the 57Fe Mössbauer time scale (107–109 s−1) electron transfer between adjacent Fe2+ and Fe3+ centers were revealed. Thermoelectric studies showed that iron-bearing colusites are p-type semiconductors with low thermal conductivity stemming from their complex crystal structure and structural disorder. The highest ZT of 0.78 at 700 K was found for the x = 1 iron content, where iron is present as Fe3+ only.

Necrotizing enterocolitis (NEC) remains a devastating disease that affects preterm infants. Hydrogen sulfide (H2S) donors have been shown to reduce the severity of NEC, but the optimal compound has yet to be identified. We hypothesized that oral H2S-Mesalamine (ATB-429) would improve outcomes in experimental NEC, and its benefits would be dependent on endothelial nitric oxide synthase (eNOS) pathways. NEC was induced in 5-day-old wild-type (WT) and eNOS knockout (eNOSKO) pups by formula feeding and stress. Four groups were studied in both WT and eNOSKO mice: 1) breastfed controls, 2) NEC, 3) NEC + 50 mg/kg mesalamine, and 4) NEC + 130 mg/kg ATB-429. Mesalamine and ATB-429 doses were equimolar. Pups were monitored for sickness scores and perfusion to the gut was measured by Laser Doppler Imaging (LDI). After euthanasia of the pups, intestine and lung were hematoxylin and eosin-stained and scored for injury in a blind fashion. TLR4 expression was quantified by Western blot and IL-6 expression by ELISA. P < 0.05 was significant. Both WT and eNOSKO breastfed controls underwent normal development and demonstrated milder intestinal and pulmonary injury compared with NEC groups. For the WT groups, ATB-429 significantly improved weight gain, reduced clinical sickness score, and improved perfusion compared with the NEC group. In addition, WT ATB-429 pups had a significantly milder intestinal and pulmonary histologic injury when compared with NEC. ATB-429 attenuated the increase in TLR4 and IL-6 expression in the intestine. When the experiment was repeated in eNOSKO pups, ATB-429 offered no benefit in weight gain, sickness scores, perfusion, intestinal injury, pulmonary injury, or decreasing intestinal inflammatory markers. An H2S derivative of mesalamine improves outcomes in experimental NEC. Protective effects appear to be mediated through eNOS. Further research is warranted to explore whether ATB-429 may be an effective oral therapy to combat NEC.

Полученные данные являются важным инструментом не только для уточнения прогнозно-поисковых критериев золоторудного месторождения углеродистых черносланцевых толщ, но также будут способствовать увеличению извлечения золота из руды за счет корректировки технологической схемы обогащения. Основная цель: изучение минеральной формы золота в рудах. Объектом исследования являются руды Северной зоны Вернинского месторождения. Методы: стандартные петрографические и минераграфические анализы, а также сканирующая электронная микроскопия. Результаты. Проведенными исследованиями установлено, что оруденение представлено двумя морфологическими типами: вкрапленный и прожилково-вкрапленный. Значительная часть рудной минерализации приурочена ко второму типу. Выявлено, что золото представлено в виде самородного с высокой пробой, большая его часть (75…80 %) приурочена к прожилково-вкрапленному типу. Самородное золото диагностировано преимущественно в ассоциации с арсенопиритом и пиритом. Многообразие форм выделений является одной из специфических особенностей самородного золота на данном месторождении, при этом отмечается господство в рудах золота неправильной формы. Для вариации диагностированных форм отмечается общая характерная черта - подчиненность полостям, существовавшим ранее, к началу или во время кристаллизации самого золота. При этом генезис данных полостей и непосредственно их очертания, а также расположение в рудах достаточно различны. Одной из наиболее распространенных морфологических разновидностей золота в рудах является проволочковидная форма, отмечающаяся в трещинах катаклаза в арсенопирите, в меньшей степени диагностируются амебообразные и каплевидные формы, образующие многочисленные мельчайшие включения самородного металла в рудах. The relevance of the research is caused by poor knowledge of the material composition of the ores of the Verninskoe deposit. The data obtained are an important tool not only for clarifying the predictive and prospecting criteria for gold ore deposits of carbonaceous black shale strata, but also will help increase gold extraction from ore by adjusting the enrichment process scheme. The main aim of the research is to study the mineral form of gold in ores. The object of the research is the ores of the Northern zone of the Verninskoe deposit. Research methods included standard petrographic and mineragraphic analyzes, as well as scanning electron microscopy. Results. Conducted researches have established that mineralization is represented by two morphological types: disseminated and veindisseminated. A significant part of ore mineralization is confined to the second type. It was revealed that gold is presented in the form of native gold and has a high fineness, most of it (75...80 %) is confined to the vein-disseminated type. Native gold is diagnosed mainly in association with arsenopyrite and pyrite. The variety of forms of segregations is one of the specific features of native gold at this deposit, while gold of irregular shape predominates in the ores. For the variation of the diagnosed forms, a common characteristic feature is noted - subordination to cavities that existed earlier, at the beginning or during the crystallization of gold itself. At the same time, the genesis of these cavities and their outlines and location in the ores are quite different. One of the most common morphological varieties in ores is a wire-like form, observed in cataclasis cracks in arsenopyrite; to a lesser extent, amoeba-like and tear-shaped forms are diagnosed, forming numerous tiny inclusions of gold in ores.

In vitro or in vivo fluorescence imaging based on quantum dots (QDs) has shown promise for the noninvasive diagnosis of atherosclerosis. However, simultaneous in vitro and in vivo imaging remains challenging due to the limitation of the current synthesis method of dual-emission QDs (dual-emitting hybrid QDs, and broad-spectrum emitting QDs). Herein, we fabricate a dual-emission (visible region and near-infrared region emission) QDs (ZAISe/ZnS) via the "bottom to up" method of a quaternary inorganic compound for the foam cells and atherosclerosis plaque imaging simultaneously without the intricate size modulation and the strict optical filter requirements. The oil-soluble ZAISe/ZnS is further encapsulated with bovine serum albumin (BSA) to realize phase transfer and ultimately possess the inflammation-targeting properties via biomimetic treatment with MMV (macrophage-derived micro-vesicle). The results first indicate that the as-constructed ZAISe/ZnS@BSA@MMV could accurately locate the foam cells and conduct long-term imaging of the atherosclerotic plaque, which provides a new strategy for the early and noninvasive diagnosis of atherosclerosis.

Multifunctional core-shell hybrids formed by integration of metal-organic framework (MOF) and functional materials have attracted extensive attention as promising theranostic nanoplatforms due to their combined novel properties and enhanced therapeutic efficacy. Recently, the second near-infrared (NIR-II, 1000-1700 nm) laser-induced photothermal therapy (PTT) as compared to the NIR-I(700-950 nm) laser-induced PTT has displayed improved therapeutic effects owing to its merits that include deeper tissue penetration and increased maximum permissible exposure. Herein, a novel core-shell hollow copper sulfide@metal-organic framework (HCuS@MIL-100) has been successfully fabricated by a layer-by-layer technique for the first time and their collective theranostic effects are investigated in vitro and in vivo. In this platform, the inner HCuS was applied as the NIR-II photothermal agent with excellent NIR-II absorption feature, leading to impressive photothermal effects under irradiation by 1064 nm light. With MIL-100 as the shell, HCuS@MIL-100 not only displayed optimal biocompatibility but also presented superior T2 magnetic resonance imaging (MRI) ability. In the current study multifunctional hollow core-shell HCuS@MIL-100 are fabricated for the MRI-guided PTT. This study also offers a facile and effective strategy for the development of novel theranostic platforms with high efficiency through the integration of MOFs and functional materials.

Ti-incorporated mesoporous materials have widespread applications in photocatalysis. Their adjustable pores could accommodate dyes like alizarin red S (ARS) to circumvent the lack of visible light response. Herein, Ti-MCM-41 was obtained to anchor visible light-capturing ARS, forming ARS-Ti-MCM-41. The ARS-Ti-MCM-41 was screened for the selective photocatalytic oxidation of organic sulfides. To improve the stability of the anchored ARS, electron transfer was orchestrated by a mediator trimethylamine (TMA, 2 mol%) illuminated by blue LEDs. Phenyl methyl sulfide could be almost entirely converted into phenyl methyl sulfoxide with 99% of selectivity within 18 min. In addition, Ti-MCM-41 was beneficial for the anchored ARS, which in turn guaranteed good recycling performance of ARS-Ti-MCM-41. The solvent trifluoroethanol enabled the stability of TMA and facilitated the highly selective formation of the target sulfoxides. This work sheds light on the vast possibility for visible light photocatalysis of dye-mesoporous materials.

BackgroundBurns are severe injuries often associated with impaired wound healing. Impaired healing is caused by multiple factors, including dysregulated inflammatory responses at the wound site. Interestingly, montelukast, an antagonist for cysteinyl leukotrienes and U.S. Food and Drug Administration approved for treatment of asthma and allergy, was previously shown to enhance healing in excision wounds and to modulate local inflammation.MethodsIn this study, the authors examined the effect of montelukast on wound healing in a mouse model of scald burn injury. Burn wound tissues isolated from montelukast- and vehicle-treated mice at various times after burn injury were analyzed for wound areas ( n = 34 to 36), reepithelialization ( n = 14), inflammation ( n = 8 to 9), and immune cell infiltration ( n = 3 to 6) and proliferation ( n = 7 to 8).ResultsIn contrast to previously described beneficial effects in excision wounds, this study shows that montelukast delays burn wound healing by impairing the proliferation of keratinocytes and endothelial cells. This occurs largely independently of inflammatory responses at the wound site, suggesting that montelukast impairs specifically the proliferative phase of wound healing in burns. Wound healing rates in mice in which leukotrienes are not produced were not affected by montelukast.ConclusionMontelukast delays wound healing mainly by reducing the proliferation of local cells after burn injury.Clinical relevance statementAlthough additional and clinical studies are necessary, our study suggests that burn patients who are on montelukast may exhibit delayed healing, necessitating extra observation.

Although sulfide perovskites usually require high-temperature syntheses, we demonstrate that organosulfides can be used in the milder syntheses of halide perovskites. The zwitterionic organosulfide, cysteamine (CYS;+NH3(CH2)2S–), serves as both the X–site and A+site in the ABX3halide perovskites, yielding the first examples of 3D organosulfide-halide perovskites: (CYS)PbX2(X–= Cl–or Br–). Notably, the band structures of (CYS)PbX2capture the direct bandgaps and dispersive bands of APbX3perovskites. The sulfur orbitals compose the top of the valence band in (CYS)PbX2, affording unusually small direct bandgaps of 2.31 and 2.16 eV for X–= Cl–and Br–, respectively, falling in the ideal range for the top absorber in a perovskite-based tandem solar cell. Measurements of the carrier dynamics in (CYS)PbCl2suggest carrier trapping due to defects or lattice distortions. The highly desirable bandgaps, band dispersion, and improved stability of the organosulfide perovskites demonstrated here motivate the continued expansion and exploration of this new family of materials, particularly with respect to extracting photocurrent. Our strategy of combining the A+and X–sites with zwitterions may offer more members in this family of mixed-anion 3D hybrid perovskites.

Aims: A simple, transition-metal-free C-S coupling protocol for the synthesis of aryl thioethers is reported. Background: Sulfur-containing moieties are ubiquitous in pharmaceutical drugs and materials and therefore methods for their construction are of great importance. One approach entails the catalytic coupling of an aryl halohydrocarbon with a thiol, but the transition metal catalysts usu-ally used are prone to poisoning by participating sulfur species and efficient catalysis is usually only achieved after complex ligand optimization. Objective: New transition-metal-free approaches to the synthesis of C-S bonds are urgently need. Method: We screened the reaction conditions such as alkali, crown ether, solvent, temperature, etc., tested the compatibility of the reaction substrate, and analyzed the mechanism process. Result: The optimized reaction conditions were determined to be 1.0 equiv of aryl halides and 1.2 equiv of thiols at 110 ℃ in toluene with K2CO3 (1.5 equiv) as a base, promoted by 10 mol% dicyclohexano-18-crown-6. Up to 33 examples of thioethers were synthesized under transition-metal-free conditions in good to excellent yields. Conclusion: We have developed a simple and efficient method for the C-S cross-coupling of a wide variety of (hetero)aryl halides and thiols mediated by dicyclohexano-18-crown-6 and with-out the need for transition-metal catalyst. In addition, the preparation and gram-scale experi-ments of a variety of drug molecules further verify the practicability of our developed method.

Methanotrophs are bacteria capable on growing on methane as their sole carbon source. They may provide a promising route for upgrading natural gas into more valuable fuels and chemicals. However, natural gas may contain significant quantities of hydrogen sulfide. Little is known about how hydrogen sulfide affects the growth and physiology of methanotrophs aside from a few studies showing that it is inhibitory. This study investigated how hydrogen sulfide affects the growth and physiology of the model methanotroph, Methylococcus capsulatus Bath. Growth studies demonstrated that hydrogen sulfide inhibits the growth of M. capsulatus Bath when the concentration exceeds 0.5% (v/v). To better understand how hydrogen sulfide is inhibiting the growth of M. capsulatus Bath, transcription and metabolite concentrations were profiled using RNA sequencing and gas chromatography-mass spectrometry, respectively. Our analysis of the differentially expressed genes and changes in metabolite concentrations suggests that hydrogen sulfide inhibits cellular respiration. The cells respond to sulfide stress in part by increasing the rate of sulfide oxidation and by increasing the expression of sulfide quinone reductase and a putative persulfide dioxygenase. In addition, they reduce the expression of the native calcium-dependent methanol dehydrogenase and increase the expression of XoxF, a lanthanide-dependent methanol dehydrogenase. While the reason of this switch in unknown, XoxF has previously been shown to be induced by lanthanides or nitric oxide in methanotrophs. Collectively, these results further our understanding of how methanotrophs respond to sulfide stress and may aid in the engineering of strains resistant to hydrogen sulfide. Channels and sewers are commonly used to collect sewage during extensively rural areas. The sewage and bacterial characteristics of rural sewage collection systems can influence their operation and maintenance performance which further affect appropriate system decision. In this study, eight rural sewage collection systems (four each of channels and sewers) were applied to evaluate the sewage quality, bacterial characteristics, and their differences of two kinds of collection systems. The results indicate that significantly distinction existed between the rural sewage collection systems of channels and sewers. Sewage in channels had higher suspended solid (SS) concentration but lower sulfide concentration than that in sewers. The SS, sulfate, and chemical oxygen demand (COD) removal capacity in channels was nearly 3.5, 4.0, and 0.6 times than those in sewers. At least 14 genera and 18 species of bacteria showed significantly distinction between channels and sewers even their main phylum, genus, and species of bacteria communities was Proteobacteria (∼70.3%), Acinetobacter (∼22.3%), and Pseudomonas fragi (∼13.8%), respectively. The structural characteristics and bacterial function caused the difference between channels and sewers. Overall, this study revealed the intrinsic and essential differences of channels and sewers, providing basic and meaningful data for rural sewage collection systems decision.

Preparation conditions have a vital effect on the structure of alumina-supported hydrodesulfurization (HDS) catalysts. To explore this effect, we prepared two NiMoS/Al2O3 catalyst samples with the same target composition using different chemical sources and characterizing the oxidic NiMo precursors and sulfided and spent catalysts to understand the influence of catalyst structure on performance. The sample prepared from ammonium heptamolybdate and nickel nitrate (sample A) contains Mo in the oxidic precursor predominantly in tetrahedral coordination in the form of crystalline domains, which show low reducibility and strong metal-support interactions. This property influences the sulfidation process such that the sulfidation processes of Ni and Mo occur tendentially separately with a decreased efficiency to form active Ni-Mo-S particles. Moreover, inactive unsupported MoS 2 particles or isolated NiS., species are formed, which are either washed off during catalytic reaction or aggregated to larger particles as seen in scanning transmission electron microscopy/energy-dispersive X-ray spectroscopy (STEM/EDX). The oxidic precursor of the sample synthesized using nickel carbonate and molybdenum trioxide as metal sources (sample B), however, contains Mo in octahedral coordination and shows higher reducibility of the metal species as well as weaker metal-support interactions than that of sample A; these properties allow an efficient sulfidation of Mo and Ni such that formation of active Ni-Mo-S particles is the main product. Ptychographic X-ray computed tomography (PXCT) and STEM and EDX measurements show that the structure formed during sulfidation is stable under operation conditions. Fatty acid synthase (FASN), a sole cytosolic enzyme responsible for de-novo lipid synthesis, is overexpressed in cancer but not in normal non-lipogenic tissues. FASN has been targeted, albeit no such inhibitor has been approved. Proton pump inhibitors (PPIs), approved for digestive disorders, were found to inhibit FASN with anticancer activities in attempting to repurpose Food and Drug Administration-approved drugs. Indeed, PPI usage benefited breast cancer patients and increased their response rate. Due to structural similarity, we thought that their metabolites might extend anticancer effects of PPIs by inhibiting FASN. Here, we tested this hypothesis and found that 5-hydroxy lansoprazole sulfide (5HLS), the end lansoprazole metabolite, was more active than lansoprazole in inhibiting FASN function and regulation of NHEJ repair of oxidative DNA damage via PARP1. Surprisingly, 5HLS inhibits the enoyl reductase, whereas lansoprazole inhibits the thioesterase of FASN. Thus, PPI metabolites may contribute to the lasting anticancer effects of PPIs by inhibiting FASN.

The first, novel solid-state membrane sensor for Zn(II) determination is developed based on ZnS nanoparticles. ZnS nanoparticles are synthesized by chemical co-precipitation and investigated via X-ray diffraction, transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR) and impedance study. X-ray diffraction shows that the prepared ZnS nanoparticles have an average domain size of 5.72 nm, which is very close to the particle size obtained from TEM observations (6.30 nm). The ZnS nanoparticles are pressed into disks and examined as electroactive solid-state membrane. Solid-state membrane and coated wire sensors are fabricated. They display linear responses over concentration ranges of 1.0 × 10−5 to 1.0 × 10−1 mol L−1 Zn2+ ions with cationic slopes of 28.9±0.2 and 25.9±0.2 mV decade−1 for the solid-state membrane and coated wire sensors, respectively. The lower limits of detection are 2.86 × 10−6 and 4.60 × 10−6 mol L−1 Zn2+ ions for the solid-state membrane and coated wire sensors, respectively. The response time for the two sensors is instantaneous (1 s), and the useful lifetimes for the solid-state membrane and coated wire sensors are long (10 and 6 months, respectively). The solid-state membrane sensor is utilized for the quantification of Zn(II) ions in brass alloys and pharmaceutical preparations.

Abstract Monotopic membrane-bound flavoproteins, sulfide:quinone oxidoreductases (SQRs), have a variety of physiological functions, including sulfide detoxification. SQR enzymes are classified into six groups. SQRs use the flavin adenine dinucleotide (FAD) cofactor to transfer electrons from sulfide to quinone. A type VI SQR of the photosynthetic purple sulfur bacterium, Thiocapsa roseopersicina (TrSqrF), has been previously characterized, and the mechanism of sulfide oxidation has been proposed. This paper reports the characterization of quinone binding site (QBS) of TrSqrF composed of conserved aromatic and apolar amino acids. Val331, Ile333, and Phe366 were identified near the benzoquinone ring of enzyme-bound decylubiquinone (dUQ) using the TrSqrF homology model. In silico analysis revealed that Val331 and Ile333 alternately connected with the quinone head group via hydrogen bonds, and Phe366 and Trp369 bound the quinones via hydrophobic interactions. TrSqrF variants containing alanine (V331A, I333A, F366A) and aromatic amino acid (V331F, I333F, F366Y), as well as a C-terminal α-helix deletion (CTD) mutant were generated. These amino acids are critical for quinone binding and, thus, catalysis. Spectroscopic analyses proved that all mutants contained FAD. I333F replacement resulted in the lack of the charge transfer complex. In summary, the interactions described above maintain the quinone molecule’s head in an optimal position for direct electron transfer from FAD. Surprisingly, the CTD mutant retained a relatively high level of specific activity while remaining membrane-anchored. This is a unique study because it focuses on the QBS and the oxidative stage of a type VI sulfide-dependent quinone reduction. Aquifer groundwater quality is largely controlled by sediment composition and physical heterogeneity, which commonly sustains a unique redox gradient pattern. Attenuation of heavy metals within these heterogeneous aquifers is reliant on multiple factors, including redox conditions and redox-active species that can further influence biogeochemical cycling. Here, we simulated an alluvial aquifer system using columns filled with natural coarse-grained sediments and two domains of fine-grained sediment lenses. Our goal was to examine heavy metal (Ni and Zn) attenuation within a complex aquifer network and further explore nitrate-rich groundwater conditions. The fine-grained sediment lenses sustained reducing conditions and served as a sink for Ni sequestration─in the form of Ni-silicates, Ni-organic matter, and a dominant Ni-sulfide phase. The silicate clay and sulfide pools were also important retention mechanisms for Zn; however, Ni was associated more extensively with organic matter compared to Zn, which formed layered double hydroxides. Nitrate-rich conditions promoted denitrification within the lenses that was coupled to the oxidation of Fe(II) and the concomitant precipitation of an Fe(III) phase with higher structural distortion. A decreased metal sulfide pool also resulted, where nitrate-rich conditions generated an average 20% decrease in solid-phase Ni, Zn, and Fe. Ultimately, nitrate plays a significant role in the aquifer's biogeochemical cycling and the capacity to retain heavy metals.

Methane mitigation from the oil and gas (OG) sector represents a key near-term global climate action opportunity. Recent legislation in the United States requires updating current methane reporting programs for oil and gas facilities with empirical data. While technological advances have led to improvements in methane emissions measurements and monitoring, the overall effectiveness of mitigation strategies rests on quantifying spatially and temporally varying methane emissions more accurately than the current approaches. In this work, we demonstrate a quantification, monitoring, reporting, and verification framework that pairs snapshot measurements with continuous emissions monitoring systems (CEMS) to reconcile measurements with inventory estimates and account for intermittent emission events. We find that site-level emissions exhibit significant intraday and daily emission variations. Snapshot measurements of methane can span over 3 orders of magnitude and may have limited application in developing annualized inventory estimates at the site level. Consequently, while official inventories underestimate methane emissions on average, emissions at individual facilities can be higher or lower than inventory estimates. Using CEMS, we characterize distributions of frequency and duration of intermittent emission events. Technologies that allow high sampling frequency such as CEMS, paired with a mechanistic understanding of facility-level events, are key to an accurate accounting of short-duration, episodic, and high-volume events that are often missed in snapshot surveys and to scale snapshot measurements to annualized emissions estimates.

Bioleaching of metal sulfides is performed by diverse microorganisms. The dissolution of metal sulfides occurs via two chemical pathways, either the thiosulfate or the polysulfide pathway. These are determined by the metal sulfides’ mineralogy and their acid solubility. The microbial cell enables metal sulfide dissolution via oxidation of iron(II) ions and inorganic sulfur compounds. Thereby, the metal sulfide attacking agents iron(III) ions and protons are generated. Cells are active either in a planktonic state or attached to the mineral surface, forming biofilms. This review, as an update of the previous one (Vera et al., 2013a), summarizes some recent discoveries relevant to bioleaching microorganisms, contributing to a better understanding of their lifestyle. These comprise phylogeny, chemical pathways, surface science, biochemistry of iron and sulfur metabolism, anaerobic metabolism, cell–cell communication, molecular biology, and biofilm lifestyle. Despite the development of sterilization, the study of reactive oxygen species (ROS) based disinfection mechanism is still under restrictions due to the lack of direct evidence. The developmental and epileptic encephalopathies encompass a group of rare syndromes characterised by severe drug-resistant epilepsy with onset in childhood and significant neurodevelopmental comorbidities. The latter include intellectual disability, developmental delay, behavioural problems including attention-deficit hyperactivity disorder and autism spectrum disorder, psychiatric problems including anxiety and depression, speech impairment and sleep problems. Classical examples of developmental and epileptic encephalopathies include Dravet syndrome, Lennox-Gastaut syndrome and tuberous sclerosis complex. The mainstay of treatment is with multiple anti-seizure medications (ASMs); however, the ASMs themselves can be associated with psychobehavioural adverse events, and effects (negative or positive) on cognition and sleep. We have performed a targeted literature review of ASMs commonly used in the treatment of developmental and epileptic encephalopathies to discuss the latest evidence on their effects on behaviour, mood, cognition, sedation and sleep. The ASMs include valproate (VPA), clobazam, topiramate (TPM), cannabidiol (CBD), fenfluramine (FFA), levetiracetam (LEV), brivaracetam (BRV), zonisamide (ZNS), perampanel (PER), ethosuximide, stiripentol, lamotrigine (LTG), rufinamide, vigabatrin, lacosamide (LCM) and everolimus. Bromide, felbamate and other sodium channel ASMs are discussed briefly. Overall, the current evidence suggest that LEV, PER and to a lesser extent BRV are associated with psychobehavioural adverse events including aggressiveness and irritability; TPM and to a lesser extent ZNS are associated with language impairment and cognitive dulling/memory problems. Patients with a history of behavioural and psychiatric comorbidities may be more at risk of developing psychobehavioural adverse events. Topiramate and ZNS may be associated with negative effects in some aspects of cognition; CBD, FFA, LEV, BRV and LTG may have some positive effects, while the remaining ASMs do not appear to have a detrimental effect. All the ASMs are associated with sedation to a certain extent, which is pronounced during uptitration. Cannabidiol, PER and pregabalin may be associated with improvements in sleep, LTG is associated with insomnia, while VPA, TPM, LEV, ZNS and LCM do not appear to have detrimental effects. There was variability in the extent of evidence for each ASM: for many first-generation and some second-generation ASMs, there is scant documented evidence; however, their extensive use suggests favourable tolerability and safety (e.g. VPA); second-generation and some third-generation ASMs tend to have the most robust evidence documented over several years of use (TPM, LEV, PER, ZNS, BRV), while evidence is still being generated for newer ASMs such as CBD and FFA. Finally, we discuss how a variety of factors can affect mood, behaviour and cognition, and untangling the associations between the effects of the underlying syndrome and those of the ASMs can be challenging. In particular, there is enormous heterogeneity in cognitive, behavioural and developmental impairments that is complex and can change naturally over time; there is a lack of standardised instruments for evaluating these outcomes in developmental and epileptic encephalopathies, with a reliance on subjective evaluations by proxy (caregivers); and treatment regimes are complex involving multiple ASMs as well as other drugs.

The widespread resistance of clinically relevant bacteria against established antibiotics emphasizes the urgent need for novel therapeutics. In this context, wound infections constitute a specific challenge, as most systemically applied antibiotics are insufficiently available at the site of infection. Therefore, the local treatment of infected wounds poses a particular challenge regarding the appropriate release kinetics of actives and their residence time in the wound bed. Consequently, design and development of novel, drug-loaded wound dressings constitute a major research focus for the effective treatment of wound infections. In this study, we employed electrospinning to design drug-loaded wound dressings, incorporating the therapeutically promising antimicrobial peptide tyrothricin. By parallel electrospinning, we combined different ratios of water-soluble polyvinylpyrrolidone and water-insoluble methacrylate copolymer (EudragitE), in order to take advantage of their specific mechanical stability and dissolution properties. We fabricated fiber mats constituting mechanically stable wound dressings with a controlled drug release profile, combining an initial burst release above the minimal inhibitory concentration of known wound pathogens and a subsequent prolonged antimicrobial effect of the active ingredient. Antimicrobial activity against Staphylococcusaureus and Staphylococcusepidermidis was successfully proven, thereby introducing our tyrothricin-loaded fiber mats as a promising prospective therapy against typical wound-associated pathogens.

The ligand-directed (LD) chemistry provides powerful tools for site-specific modification of proteins. We utilized a peptide with an appended methionine (Met) as a ligand; then, the Met thioether was modified into sulfonium which enabled a proximity induced group transfer onto protein cysteine in the vicinity upon peptide-target binding. The sulfonium warhead could be easily constructed with unprotected peptides, and the transferable group scope was conducted on model protein PDZ and its ligand peptides. In addition, a living cell labeling was successfully achieved.

Clinical trials of antidepressants often fail to demonstrate their efficacy versus placebo, suggesting that patient selection based on physician ratings of depression may contribute to a high placebo response.In the CCT-004 trial of vortioxetine, central monitoring was employed to compare physician and patient ratings of depression and anxiety at baseline and over time to identify factors contributing to a large placebo response, as well as to explore the potential of a unique patient-rated clinical measure combining QIDS-J and Himorogi Self-rating Anxiety Scale (HSAS), to contribute to optimal patient selection at baseline and patient monitoring over time.The CCT-004 trial showed similar trends between the QIDS-J and MADRS (Montgomery-Åsberg Depression Rating Scale) ratings. It was suggested that central monitoring of the QIDS-J and MADRS ratings of depression and anxiety symptoms helped reduce the baseline score inflation by calling the study sites' attention to discrepancies between these ratings at baseline; it also allowed these ratings to be assessed for their concordance over time. Of note, MDD patients with baseline QIDS-J scores ≥11/HSAS ≤19 were associated with the smallest placebo response, with the effect size being larger than that for those with QIDS-J scores ≤10/HSAS ≥20.The use of both physician and patient ratings of depression and anxiety symptoms at baseline and over time, as well as their central monitoring, helped minimize the baseline score inflation and optimize patient monitoring over time, and allowed the antidepressant to be evaluated for its full therapeutic potential.

Owing to their ultrathin two-dimensional structure and efficient catalytic ability for persulfate activation, graphene-based nanocarbons exhibit considerable application potential in fabricating carbonaceous composite membranes for in situ catalytic oxidation to remove organic pollutants. This approach offers significant advantages over conventional batch systems. However, the relationships between the physicochemical properties of carbon mats and performance of graphene-based catalytic membranes in water purification remain ambiguous. Herein, we summarize the main mechanisms of in situ catalytic oxidation and the facile fabrication strategies of carbonaceous composite membranes. Different factors influencing the performance of graphene-based catalytic membranes are comprehensively discussed. The defective level, heteroatom doping, and stacking morphology of carbon mats and operational conditions during filtration play critical roles in the oxidative degradation of target pollutants. Long-term operation leads to the deterioration of catalytic activity and transmembrane pressure, especially in the complex water matrix. Finally, the present challenges and future perspectives are presented to improve the anti-fouling performance and catalytic stability of membranes and develop scalable fabrication methods to promote the engineering applications of in situ catalytic oxidation in real water purification.

Class II lanthipeptide synthetases (LanM enzymes) catalyze the installation of multiple thioether bridges into genetically encoded peptides to produce macrocyclic lanthipeptides, a class of biologically active natural products. Collectively, LanM enzymes install thioether rings of different sizes, topologies, and stereochemistry into a vast array of different LanA precursor peptide sequences. The factors that govern the outcome of the LanM-catalyzed reaction cascade are not fully characterized but are thought to involve both intermolecular interactions and intramolecular conformational changes in the [LanM:LanA] Michaelis complex. To test this hypothesis, we have combined AlphaFold modeling with hydrogen-deuterium exchange mass spectrometry (HDX-MS) analysis of a small collection of divergent LanM/LanA systems to investigate the similarities and differences in their conformational dynamic properties. Our data indicate that LanA precursor peptide binding triggers relatively conserved changes in the structural dynamics of the LanM dehydratase domain, supporting the existence of a similar leader peptide binding mode across the LanM family. In contrast, changes induced in the dynamics of the LanM cyclase domain were more highly variable between enzymes, perhaps reflecting different peptide-cyclase interactions and/or different modes of allosteric activation in class II lanthipeptide biosynthesis. Our analysis highlights the ability of the emerging AlphaFold platform to predict protein-peptide interactions that are supported by other lines of experimental evidence. The combination of AlphaFold modeling with HDX-MS analysis should emerge as a useful approach for investigating other conformationally dynamic enzymes involved in peptide natural product biosynthesis.

Aims. To observe the clinical efficacy of self-made Lifei Dingchuan decoction combined with western medicine in the treatment of cough variant asthma (phlegm-heat accumulation in the lung syndrome). Materials and Methods. The clinical data of 90 patients with cough variant asthma who were hospitalized in the Department of Respiratory Medicine of our hospital from January 2020 to April 2022 were selected as the research objects, and they were equally divided into the observation group and the reference group according to different treatment methods, 45 cases in each group. The group was treated with traditional montelukast sodium chewable tablet and salmeterol fluticasone mixed powder inhalation, and the observation group was treated with self-made Lifei Dingchuan decoction on the basis of the control group, saturation, pH, partial pressure of oxygen in arterial blood, partial pressure of carbon dioxide, length of stay, and hospitalization costs. Results. After the patients underwent self-made Lifei Dingchuan decoction, there were significant differences between the observation group and the reference group in terms of heart rate, respiratory rate, blood oxygen saturation, pH value, arterial blood oxygen partial pressure, carbon dioxide partial pressure, and within the group. There was a statistical difference ( P < 0.05 ). The adverse reactions in patients with cough variant asthma after treatment showed that the red throat, shortness of breath, chest tightness, and dry mouth in the observation group were significantly different from those in the control group ( P < 0.05 ). After investigation, follow-up, and statistics, the hospitalization time, hospitalization cost, asthma exacerbation control time, effective rate, and recurrence rate were compared between the two groups, and the differences between the two groups were statistically significant ( P < 0.05 ). Conclusion. The study on the clinical efficacy and low hospitalization cost of the self-prepared lung and asthma-restorative soup in patients with cough variant asthma significantly improved the patients’ arterial oxygen saturation, acid-base value, arterial partial pressure of oxygen, and partial pressure of carbon dioxide and effectively controlled the heart rate and respiratory rate with high safety, which is worth further promotion.

1,1′-Bi-2-naphthol (BINOL)-derived chiral bifunctional sulfide and selenide catalysts that possess a hydroxy group are known to be effective catalysts for enantioselective bromolactonizations. When applied to asymmetric iodolactonizations of 4-pentenoic acids, these catalysts yield chiral γ-butyrolactone products that are important compounds in medicinal chemistry. Although chiral bifunctional selenides have shown good catalytic performances in enantioselective iodolactonizations, reactions with BINOL-derived chiral sulfide catalysts unexpectedly gave iodolactonization products in nearly racemic forms. The roles of chalcogenide moieties and hydroxy groups on bifunctional catalysts were investigated, and the importance of both a selenide moiety and a hydroxy group on chiral bifunctional selenide catalysts to achieve enantioselective iodolactonizations was clarified. An optimized chiral bifunctional selenide catalyst was applied to the asymmetric synthesis of chiral γ-butyrolactones and phthalides. The capabilities to manipulate light-matter interaction at the nanoscale lie at the core of many promising photonic applications. Optical nanoantennas, made of metallic or dielectric materials, have seen a rapid development for their remarkable optical properties facilitating the coupling of electromagnetic waves with subwavelength entities. However, high-throughput and cost-effective fabrication of these nanoantennas is still a daunting challenge. In this work, we provide a versatile nanofabrication method capable of producing large scale optical nanoantennas with different shapes. It is developed from colloidal lithography with no dry etching required. Furthermore, both metallic and all-dielectric nanoantennas can be readily fabrication in a high-throughput fashion. Au and Si nanodisks were fabricated and employed to assemble heterostructures with monolayer tungsten disulfide. Strong coupling is observed in both systems between plasmon modes (Au nanodisks) or anapole modes (Si nanodisks) with excitons. We believe that this nanofabrication method could find a wide range of applications with the diverse optical nanoantennas it can engineer.

Active hydrothermal travertine systems are ideal environments to investigate how abiotic and biotic processes affect mineralization mechanisms and mineral fabric formation. In this study, a biogeochemical characterization of waters, dissolved gases, and microbial mats was performed together with a mineralogical investigation on travertine encrustations occurring at the outflow channel of a thermal spring. The comprehensive model, compiled by means of TOUGHREACT computational tool from measured parameters, revealed that mineral phases were differently influenced by either abiotic conditions or microbially driven processes. Microbial mats are shaped by light availability and temperature gradient of waters flowing along the channel. Mineralogical features were homogeneous throughout the system, with euhedral calcite crystals, related to inorganic precipitation induced by CO
