Computational study on the catalytic control of endo/exoDiels-Alder reactions by cavity quantum vacuum fluctuations.

Ultracold gases of highly magnetic lanthanide atoms have enabled the realisation of dipolar quantum droplets and supersolids. However, future studies could be limited by the achievable atom numbers and hindered by high three-body loss rates. Here we study density-dependent atom loss in an ultracold gas of ${}^{166}$Er for magnetic fields below $4\ \textrm{G}$, identifying six previously unknown features which display both a strong temperature dependence and also sensitivity to the polarisation and intensity of the light used to optically trap the atoms. This detailed knowledge of the loss landscape allows us to optimise the production of dipolar BECs with more than $2 \times 10^5$ atoms and points towards optimal strategies for the study of large-atom-number dipolar gases in the droplet and supersolid regimes., Comment: 6 pages, 4 figures

Marine heatwaves (MHWs) have increased worldwide in intensity, frequency, and duration, often with severe impacts on ecologically and socio-economically important coastal habitats. Most MHW studies have focused on summer events in specific regions or global open-water analyses with little consideration to seasonality or spatial biogeography of the nearshore zone. We address this research gap by analysing changes to MHWs across seasons and ecoregions, within 12 nautical miles (NM) of New Zealand’s coastline, an area with high biodiversity and endemism. We used season-specific linear trend analysis with temporal breakpoints between 1982 and 2021 to identify the number of MHW events and MHW days, and mean, maximum, and cumulative intensity of MHWs for New Zealand coastal ecoregions. Across ecoregions, 69.5% of 200 trend analyses increased significantly whereas the remaining 30.5% were unaffected. For the significant increases, temporal breakpoints occurred between 1994 and 2012. We conclude that MHWs have become stronger, longer and more frequent across coastal New Zealand, but with some variability within and between regions. Our trend-analyses could enable retrospective analyses of biological tipping-points and inform management practices that may enhance the resilience of ecoregions yet to face MHW breakpoints or those in the initial stages of major climatic changes.

Click chemistry, which refers to chemical reactions that are fast, selective, and with high product yields, has become a powerful approach in organic synthesis and chemical biology. Due to the cytotoxicity of the transition metals employed in click chemistry reactions, a search for novel metal-free alternatives continues. Herein we demonstrate that an optical cavity can be utilized as a metal-free alternative in click chemistry cycloaddition reaction between cyanoacetylene and formylazide using the quantum electrodynamics coupled cluster (QED-CC) method. We show that by changing the molecular orientation with respect to the polarization of the cavity mode(s), the reaction can be selectively catalyzed to form a major 1,4-disubstituted or 1,5-disubstituted product. This work highlights that a cavity has the same effect on the investigated cycloaddition as the transition metal catalysts traditionally employed in click chemistry reactions. We expect our findings to further stimulate research in cavity-assisted click chemistry reactions.

LITMUS was a single-centre, Phase 2a study designed to investigate whether the gene biomarker FGL2/IFNG previously reported for the identification of tolerance in murine models could identify operationally tolerant liver transplant recipients. Multiplex RT-PCR was used to amplify eight immunoregulatory genes in peripheral blood mononuclear cells (PBMC) from 69 adult liver transplant recipients. Patients with PBMC FGL2/IFNG ≥ 1 and a normal liver biopsy underwent immunosuppression (IS) withdrawal. The primary end point was the development of operational tolerance. Secondary end points included correlation of tolerance with allograft gene expression and immune cell markers. Twenty-eight of 69 patients (38%) were positive for the PBMC tolerance biomarker and 23 proceeded to IS withdrawal. Nine of the 23 patients had abnormal baseline liver biopsies and were excluded. Of the 14 patients with normal biopsies, eight (57%) have achieved operational tolerance and are off IS (range 12–57 months). Additional studies revealed that all of the tolerant patients and only one non-tolerant patient had a liver gene ratio of FOXP3/IFNG ≥ 1 prior to IS withdrawal. Increased CD4+ T regulatory T cells were detected both in PBMC and livers of tolerant patients following IS withdrawal. Higher expression of SELE (gene for E-selectin) and lower expression of genes associated with inflammatory responses (GZMB, CIITA, UBD, LSP1, and CXCL9) were observed in the pre-withdrawal liver biopsies of tolerant patients by RNA sequencing. These results suggest that measurement of PBMC FGL2/IFNG may enrich for the identification of operationally tolerant liver transplant patients, especially when combined with intragraft measurement of FOXP3/IFNG. Antipsychotic medications are often used in older adults, but the knowledge base in this population is limited. By use of a large TDM-database we will study exposure, polypharmacy and the relationship between age and serum levels of these medications.

Emerging biotechnologies from fields such as synthetic biology and industrial biotechnology raise challenges for governance. In response, public funders have developed new approaches to govern these technologies before decisions are locked in and products emerge onto the market. Over a decade of experience with these nascent forms of governance, such as Responsible Re- search and Innovation (RRI), shows their value but also the limitations, particularly when im- plemented without consideration of day-to-day working conditions, sector specific distinctions and institutional structures shaping research in the biological sciences. Drawing on three workshops with members of the ERA CoBioTech funding programme, we show how a new approach, grounded in the idea of human capabilities, can help to integrate the skills, knowledge and institutional conditions needed to enact upstream governance in the design of future funding programmes. We identify the goals researchers associated with RRI in the life sciences, outline five sets of capabilities that enable researchers, managers and adminis- trators to practise responsible research and innovation, and unearth a corresponding set of re- sources that these capabilities depend upon. Funders that learn to design programmes to max- imise and expand the five capability sets are likely to enable more substantive forms of upstream governance than before.

We will attempt to replicate 12 effects in a single experimental package across numerous labs. Variations between lab conditions and sample characteristics will be analyzed to investigate how these factors might influence replication success.

We have already started this review in 2012. We did not have a published protocol available at that time. However, already then, we followed very closely the methods of the related Cochrane review “amitriptyline for depression” (Leucht et al. 2012) with a few a priori defined decisions, and we had decided from the start that we would not restrict the review to major depressive disorder defined by operationalized diagnostic criteria and that we would analyze fewer outcomes. This protocol also serves to document our methodology more in detail than in a journal publication.

This article traces the history of India’s first tertiary cancer hospital, Tata Memorial Hospital (TMH). TMH was originally conceived in 1932 as a philanthropic project by the Tatas, an elite Parsi business family in Bombay. The founding of TMH represented a form of philanthro-capitalism which both enabled the Tatas to foster a communal acceptance for big businesses in Bombay and provide the Tatas with the opportunity to place stakes in the emerging nuclear research economy seen as essential to the scientific nationalist sentiment of the post-colonial state. In doing this, the everyday activities of TMH placed a heavy emphasis on nuclear research. In a time when radium for the treatment of cancer was still seen as ‘quackery’ in much of the world, the philanthro-capitalist investment and the interest in nuclear research by the post-colonial state provided an environment where radium medicine was able to be validated. The validation of radiotherapy at TMH influenced how other cancer hospitals in India developed and also provided significant resources for cancer research in early-mid twentieth century India. Ultimately, this article identifies ways in which cancer comes to be seen as relevant in the global south and raises questions on the relationship between local and global actors in setting health priorities.

Area-based conservation is a widely used approach for tackling biodiversity loss and there are ongoing discussions over what is an appropriate global conservation area coverage target. To inform such debates, we need to know the extent and ecological representativeness of the current conservation area network, but this is hampered by gaps in existing global datasets. In particular, while data on privately and communally managed protected areas (PAs) and OECMs (other effective area-based conservation measures) are often available at the national level, it can take many years to incorporate these into official datasets. This suggests a complementary approach is needed, based on selecting a sample of countries and using their national-scale datasets to produce more accurate metrics. However, every country added to the sample increases the costs of data collection, collation and analysis. To address this, here we present a data collection framework underpinned by a spatial prioritisation algorithm, which identifies a minimum set of countries that are also representative of 10 factors that influence conservation area establishment and biodiversity patterns. We then illustrate this approach to identify a representative set of sampling units that cover 10% of the terrestrial realm, selecting 25 countries (choosing the same 10% area at random selected a mean of 162 countries). These sampling units could be the focus of future data collation on different types of conservation area. Analysing these data could produce quicker, more accurate estimates of global conservation area coverage and ecological representativeness, complementing existing international reporting systems.

Boyle's 1662 observation that the volume of a gas is, at constant temperature, inversely proportional to pressure, offered a prototypical example of how an equation of state (EoS) can succinctly capture key properties of a many-particle system. Such relations are now cornerstones of equilibrium thermodynamics. Extending thermodynamic concepts to far-from-equilibrium systems is of great interest in various contexts including glasses, active matter, and turbulence, but is in general an open problem. Here, using a homogeneous ultracold atomic Bose gas, we experimentally construct an EoS for a turbulent cascade of matter waves. Under continuous forcing at a large length scale and dissipation at a small one, the gas exhibits a non-thermal, but stationary state, which is characterised by a power-law momentum distribution sustained by a scale-invariant momentum-space energy flux. We establish the amplitude of the momentum distribution and the underlying energy flux as equilibrium-like state variables, related by an EoS that does not depend on the details of the energy injection or dissipation, or the history of the system. Moreover, we show that the equations of state for a wide range of interaction strengths and gas densities can be empirically scaled onto each other. This results in a universal dimensionless EoS that sets benchmarks for the theory and should also be relevant for other turbulent systems., Comment: 6 pages, 5 figures

We provide an overview of the effects of interactions in Bose-condensed gases. We focus on phenomena that have been explored in ultracold atom experiments, covering both tuneable contact interactions and dipolar interactions. Horizon scanning is intended to identify the opportunities and threats associated with technological, regulatory and social change. In 2017 some of the present authors conducted a horizon scan for bioengineering (Wintle et al., 2017). Here we report the results of a new horizon scan that is based on inputs from a larger and more international group of 38 participants. The final list of 20 issues includes topics spanning from the political (the regulation of genomic data, increased philanthropic funding and malicious uses of neurochemicals) to the environmental (crops for changing climates and agricultural gene drives). The early identification of such issues is relevant to researchers, policy-makers and the wider public.

Conservation area networks in most countries are fragmented and inadequate. To tackle this in England, government policies are encouraging stakeholders to create local-level Nature Recovery Networks. Here we describe work led by a wildlife organisation that used the systematic conservation planning approach to identify a Nature Recovery Network for three English counties and select focal areas within it where they will focus their work. The network was based on identifying core zones to maintain current biodiversity and recovery zones for habitat restoration, meeting area-based targets for 50 priority habitat, landscape, landcover and ecosystem service types. It included the existing designated sites for conservation, which cover 6.05% of the study site, and identified an additional 11.6% of land as core zones and 18% as recovery zones, reflecting the organisation’s broad objective of conserving and connecting 30% of England by 2030. We found that systematic conservation planning worked well in this context, identifying a connected, adequate, representative and efficient network and producing transparent and repeatable results. The analysis also highlighted the pressing need for government agencies to provide national-level guidance and datasets for setting targets and including species data in spatial planning, creating a national framework to inform local action.

Summary box In March 2021, Khan et al published a commentary titled ‘Decolonising global health in 2021: a roadmap to move from rhetoric to reform’ in BMJ Global Health .1 To our knowledge, under the decolonising global health umbrella, this is the first publication to make explicit how to move from theory to practice. However, we express concern over the prescriptive nature of this commentary, namely in its call for ‘metrics,’ ‘checklists,’ and a ‘map’—unironically all tactics that have been and are used by colonisers to assert violence. In this commentary, we directly respond to Khan et al and join the larger discussion on what it may mean for decolonising global health to move from theory to practice. First, we address the definition of decolonisation in decolonial thought, followed by the definition used by Khan et al . Second, we critically deconstruct the suggested “roadmap,” and conclude that these “reforms” will not realise the definition of decolonisation. Finally, we suggest conceptual frameworks that may be better suited to think through decolonisation in the global health industry. Paul Schneider and Robert Smith are PhD students in the Wellcome Trust DTC for Public Health, Economics & Decision Science. They found their first-year research attachment with ‘parkrun UK’ so interesting, they continued to work on it even after they moved on to their PhD research. They tell us more about their work with the organisation that has encouraged thousands of people across the UK to take up the weekly challenge of a 5km run or walk.This case study focuses on their approach to open research using open access publications, open data and open source code.

PurposeWomen with a first-degree family history of breast cancer (FHBC) are sometimes advised to initiate screening mammography when they are 10 years younger than the age at which their youngest relative was diagnosed, despite a lack of unambiguous evidence that this is an effective strategy. It is unknown how often this results in women initiating screening earlier (< 40 years) than screening guidelines recommend for average-risk women.MethodsWe examined screening initiation age by FHBC and age at diagnosis of the youngest relative using data collected by the Breast Cancer Surveillance Consortium on 74,838 first screening mammograms performed between 1996 and 2016.ResultsOf the 74,838 women included in the study, nearly 9% reported a FHBC. Approximately 16.8% of women who initiated mammography before 40 years reported a FHBC. More women with a FHBC than without initiated screening < 40 years (48% vs. 23%, respectively). Among women with a FHBC who initiated screening < 40 years, 65% were 10 years younger than the age at which their relative was diagnosed.ConclusionWomen with a first-degree relative diagnosed with breast cancer were more likely to start screening before 40 years than women reporting no FHBC, especially if their relative was diagnosed before 50 years.

We provide a suite of publicly available spectral data reduction software to facilitate rapid scientific products from time-domain observations. The Automated SpectroPhotometric REDuction (ASPIRED) toolkit is aimed to be as general as possible with high flexibility such that it can work with different instruments. The default settings support typical low-resolution long-slit spectrometer configurations, whilst it also offers a flexible set of functions for users to refine and tailor-make automated pipelines to an instrument's individual characteristics. Automation can provide immediate data reduction to allow adaptive observing strategies, which is particularly important in Time Domain Astronomy. The Mid-Atlantic region of the USA has experienced increasing annual precipitation amounts in recent decades, along with more frequent extreme events of greater magnitude. Unlike many US regions that have suffered increasing drought conditions from higher evapotranspiration demand, positive trends in the Mid-Atlantic accumulated precipitation are greater than the recent increases in reference evapotranspiration. The temporal correlation between precipitation events and soil moisture capacity is essential for determining how the nature of drought has changed in the region. This analysis has shown that soil moisture scarcity has declined in nine of ten subregions of the Mid-Atlantic that were analyzed from 1985 to 2019. Two algorithms were deployed to draw this conclusion: Climatol enabled the use of the FAO-56 Penman-Monteith equation on daily observation station data for which complete records were unavailable, and the second algorithm calculated soil moisture levels on a daily basis, more accurately capturing drought conditions than common methods using weekly or monthly summaries. Although the declining drought trends were not statistically significant, a result of more extreme events and higher evapotranspiration rates, the inclusion of direct data from an expanded set of locations provides greater clarity from the trends, allowing policymakers and landowners to anticipate changes in future Mid-Atlantic irrigation water demand.

Contains fulltext : 244827.pdf (Publisher’s version ) (Open Access) PURPOSE: To review the scientific literature seeking lessons for the COVID-19 era that could be learned from previous health services interruptions that affected the delivery of cancer screening services. METHODS: A systematic search was conducted up to April 17, 2020, with no restrictions on language or dates and resulted in 385 articles. Two researchers independently assessed the list and discussed any disagreements. Once a consensus was achieved for each paper, those selected were included in the review. RESULTS: Eleven articles were included. Three studies were based in Japan, two in the United States, one in South Korea, one in Denmark, and the remaining four offered a global perspective on interruptions in health services due to natural or human-caused disasters. No articles covered an interruption due to a pandemic. The main themes identified in the reviewed studies were coordination, communication, resource availability and patient follow-up. CONCLUSION: Lessons learned applied to the context of COVID-19 are that coordination involving partners across the health sector is essential to optimize resources and resume services, making them more resilient while preparing for future interruptions. Communication with the general population about how COVID-19 has affected cancer screening, measures taken to mitigate it and safely re-establish screening services is recommended. Use of mobile health systems to reach patients who are not accessing services and the application of resource-stratified guidelines are important considerations. More research is needed to explore best strategies for suspending, resuming and sustaining cancer screening programs, and preparedness for future disruptions, adapted to diverse health care systems.

In recognition of a global health governance ecosystem plagued with inequities, this short commentary critically analyses the recent focus on youth ‘inclusion’ in global digital health governance and discusses potential strategies to amplify and include youth voices in accessible and meaningful ways, without museumising them. As global youth representatives and advocates with varied backgrounds and lived experiences in health and technology across the world, we are well-positioned to provide critique on how youth have been engaged thus far in digital health governance as well as provide recommendations.

Background There are approximately 300 cases of enteric fever reported annually from England and Wales; most are imported infections. Clinical management of enteric fever remains a challenge with the emergence of ESBL-producing strains, especially XDR Salmonella Typhi from Sindh, Pakistan. Methods All strains of S. Typhi and Salmonella Paratyphi A isolated from cases presenting with symptoms of enteric fever in England and Wales, between 1 April 2014 and 31 March 2020, were characterized using WGS. Antibiotic susceptibility testing was performed using an agar dilution method. Results ESBL strains contributed to 69 cases of enteric fever (S. Typhi n = 68, S. Paratyphi A n = 1); 68 were imported (Pakistan n = 64, Iraq n = 2, Bangladesh n = 1 and India n = 1). Ages ranged from 1 to 56 years, 36/69 (52%) were children, 52% were female and the duration of hospital stay ranged from 1 to 23 days. The ESBL phenotype was conferred by the presence of blaCTX-M-15 (S. Typhi n = 67 and S. Paratyphi A n = 1) or blaCTX-M-55 (S. Typhi n = 1). An IncY plasmid harbouring blaCTX-M-15 and qnr was detected in 56 strains from Pakistan. The IncY plasmid was absent in the remaining strains and there was evidence of a 4 kb ISEcpl-blaCTX-M-15-tnp gene cassette insertion into the chromosome at one of three integration points. Conclusions Chromosomal integration of blaCTX-M-15 within the XDR Sindh strains may lead to the maintenance of resistance in the absence of antibiotic selection pressure. Empirical treatment of cases of complicated enteric fever returning from Pakistan will henceforth have to include a carbapenem.

Bose-Einstein condensation is a unique phase transition in that it is not driven by inter-particle interactions, but can theoretically occur in an ideal gas, purely as a consequence of quantum statistics. This chapter addresses the question \emph{`How is this ideal Bose gas condensation modified in the presence of interactions between the particles?' } This seemingly simple question turns out to be surprisingly difficult to answer. Leon Mestel was best known for his wide-ranging work on cosmic magnetism, but he also worked on an equally wide range of non-magnetic problems in astrophysics, from star and galaxy formation to white dwarf cooling. Despite his work being primarily theoretical and highly mathematical, he was always aware of all the relevant observational data that both needed to be explained and also provided constraints for his theoretical models. He was internationally recognized as an authority on the influence of magnetic fields in astronomy, receiving a number of significant honours. He also had a deserved reputation for scrupulous honesty and integrity in his work. His life's work culminated in the publication of two editions of a magisterial monograph on stellar magnetism. He collaborated widely, influenced many other researchers and was in great demand as a conference speaker. He was also a conscientious academic, taking his full part in departmental teaching and administration, and a delightful companion and friend to all who knew him. He was well known for his many stories and jokes, which were widely enjoyed. His outside interests included being a long-standing member of the Editorial Board of this publication, Biographical Memoirs of Fellows of the Royal Society.
