BackgroundThe worldwide dissemination of K. pneumoniae isolates is a significant public health concern, as these organisms possess a unique capacity to acquire genetic elements encoding both resistance and hypervirulence. This study aims to investigate the epidemiological, resistance, and virulence characteristics of K. pneumoniae isolates that carry both virulence plasmids and blaOXA-48-like genes in a tertiary hospital in China.MethodsA total of 217 clinical isolates of carbapenem-resistant K. pneumoniae (CRKP) were collected between April 2020 and March 2022. The antimicrobial susceptibility test was conducted to evaluate the drug resistance profile. All isolates were screened for the presence of genes encoding carbapenemases (blaKPC, blaNDM, blaIMP, blaVIM, and blaOXA-48-like), ESBLs genes (blaCTX-M, blaSHV, blaTEM), and virulence plasmid pLVPK-borne genes (rmpA, rmpA2, iucA, iroB, and peg344) using polymerase chain reaction (PCR) amplification. Clonal lineages were assigned using multilocus sequence typing (MLST) and pulsed-field gel electrophoresis (PFGE). The plasmid incompatibility groups were identified using PCR-based replicon typing (PBRT). The transferability of carbapenemase-encoding plasmids and pLVPK-like virulence plasmids was assessed via conjugation. The plasmid location of rmpA2 was determined using S1-Pulsed Field Gel Electrophoresis (S1-PFGE) and southern blotting hybridization. The virulence potential of the isolates was assessed using the string test, capsular serotyping, serum killing assay and a Galleria mellonella larval infection model.ResultsOf the 217 CRKP clinical isolates collected, 23% were identified as carrying blaOXA-48-like genes. All blaOXA-48-like isolates exhibited resistance to commonly used clinical antimicrobial agents, except for ceftazidime/avibactam, colistin, tigecycline, trimethoprim-sulfamethOXAzole, polymyxin B, and nitrofurantoin. The main common OXA-48-like carbapenemase enzymes were found to be blaOXA-181 and blaOXA-232. MLST and PFGE fingerprinting analysis revealed clonal transmission and plasmid transmission. OXA-48-like producing CRKP isolates mainly clustered in K64 ST11 and K47 ST15. Results of the string Test, serum killing assay (in vitro) and Galleria mellonella infection model (in vivo) indicated hypervirulence. PBRT showed that the blaOXA-181 and blaOXA-232 producing hypervirulent carbapenem-resistant Klebsiella pneumoniae (Hv-CRKP) were mainly carried on ColE-type, IncF, and IncX3. Eight clinical isolates of hv-CRKP were identified as carrying three carbapenem-resistant genes (blaKPC, blaOXA-181 or OXA-232, and blaNDM-1). Moreover, Southern blotting hybridization revealed that all eight isolates had a pLVPK-like virulent plasmid (138.9–216.9 kb) with an uneven number and size of plasmid.ConclusionIn our investigation, we have observed the emergence of hv-CRKP carrying blaOXA-48-like genes, which identified two genetic relationships: clonal transmission and plasmid transmission. PBRT analysis showed that these genes were mainly carried on ColE-type, IncF, and IncX3 plasmids. These isolates have been shown to be hypervirulent in vitro and in vivo. Additionally, eight clinical isolates of hv-CRKP were identified as carrying three carbapenem-resistant genes (blaKPC, blaOXA-181 or OXA-232, and blaNDM-1) and carrying a pLVPK-like virulent plasmid. Hence, our findings highlight the need for further investigation and active surveillance of hypervirulent OXA-48-like producing Hv-CRKP isolates to control their transmission.

The generation and exploitation of hot spots play a crucial role in the fabrication of highly sensitive surface-enhanced Raman scattering (SERS) substrates. In this work, silver nanosphere (AgNS) array substrates with various morphologies of polystyrene (PS) core/Ag shell were fabricated by reactive ion etching and glancing angle deposition technique on PS sphere templates. The electromagnetic field distribution (hot spots) of AgNS array was calculated by the finite difference time domain method. R6G and MB were used as probe molecules to investigate the effect of structure spacing on electric field enhancement. Under 785 nm excitation, an AgNS substrate with a gap of approximately 20 nm exhibited the best SERS enhancement, and the limit of detection reached 5×10-8 M for both analytes. These results serve as a reference for the development of a new highly sensitive SERS sensors.

Background Extragonadal germ cell tumours (EGGCTs) originated in Shoulder are extremely rare, with 1 case described in the literature. We report a case of a patient with a primary Right Shoulder mixed EGGCT. Case presentation A 36-year-old male patient was hospitalized for 6 months due to progressive right shoulder swelling accompanied by pain. Subsequently, the right shoulder tumor was removed entirely. Gross pathological examination showed that the size of the tumor mass was about 14 × 10 × 6 cm.Mutations were observed in ENPEP (4q25), ZCCHC11, RREB1 (6p24.3), CKAP4 (12q23.3), and other genes were detected by whole exome sequencing. Histology revealed a mixed EGGCT of the Right Shoulder with immature teratoma and yolk sac tumour. The patient went through 6 cycles of chemotherapy. After 7 months of follow-up, the patient is recurrence. Conclusion The primary MEGCT of the shoulder is an extremely rare condition. However, the recurrence and metastasis rates are high. Therefore, further research is necessary to determine this rare disease’s genetic and clinical characteristics to develop an effective treatment plan.

In this study, the effect of bacterial cellulose nanofibers (BCNF) incorporation on the structural and rheological properties of casein gels was investigated, where the mixed BCNF and casein gels were prepared by adding gluconic acid δ-lactone (GDL) to acidify the mixed polymer solutions at 3.0% casein concentration (w/v) and varying BCNF concentrations (0–0.5%, w/v). By changing the addition amount of GDL, the mechanical and structural properties of the mixed gels were studied at above, near and below the electric point (pI) of the casein. At pH above the pI of the casein, the introduction of BCNF initially increased the gel strength, but further addition of BCNF weakened the mixed gels. At near and below the pI of the casein, the incorporation of BCNF continuously increased the gel strength. Besides, all gels showed good structural homogeneity, without macroscopic phase separation occurring, which indicated good compatibility of BCNF with the casein gels.

The structural characteristics of litchi shell-derived carbon are conducive to regulation and modification. Using biomass waste litchi shells as carbon source and nano-silicon particles to prepare silicon-carbon composite materials to relieve the volume effect of silicon in the charge and discharge process, litchi shell-derived activated carbon (LAC) with high specific surface area (1011.115 m2 g−1) and high porosity was obtained from litchi shell as silicon buffer matrix by simple high-temperature calcination and activation of ZnCl2. A silicon-carbon composite material (3D LAC@Si) with an embedded cladding structure was prepared with a high-energy ball milling process. In the electrochemical performance test, 3D LAC@Si as the negative electrode of a lithium-ion battery showed a high lithium storage capacity of 834.4 mAh g−1 and a high coulombic efficiency of 98.34% after cycling 100 cycles at a current density of 0.2A g−1.

Extranodal marginal zone lymphoma of mucosa-associated lymphoid tissue (MALT lymphoma) are a group of diseases with marked heterogeneity, including clinical, immunohistochemical, and molecular heterogeneity. The disease remains unspecified in the genetic landscape with only a few sequencing studies to date; however, systematic studies of alimentary canal MALT lymphoma have not been reported. To better understand the genetics of this tumor, targeted sequencing in a group of 31 cases (including 2 esophageal, 2 colonic, 4 small intestinal, and 23 gastric cases) were performed. We found epigenetic regulation (DNMT3A, KMT2D, KMT2A, EP300, TET2, etc.), signaling pathways (APC, CHD8, TNFAIP3, TNFRSF14, ZAP70, NF1,), and tumor suppressor genes (TP53, BCORL1, FOXO1, ATM, etc.) involved. Moreover, we found MTOR gene mutations in 16% of the cases that made these patients more prone to recurrence and metastasis than those with MTOR wild type genes. More interestingly, ARID2 mutations were detected in 32% of all the cases, and the mutation rate was higher and statistically significant in Helicobacter pylori (Hp)-negative patients in the gastric group. Therefore, this study found that MTOR and ARID2 gene mutations have pathogenic and prognostic implications.

Pulmonary arterial hypertension (PAH) is a devastating disorder characterized by excessive proliferation and vasoconstriction of small pulmonary artery vascular smooth muscle cells (PASMCs). Coptidis rhizoma (CR) because of the complexity of the components, the underlying pharmacological role and mechanism of it on PAH remains unknown. In this article, the network pharmacological analysis was used to screen the main active constituents of CR and the molecular targets that these constituents act on. Then, we evaluated the importance of berberine and quercetin (biologically active components of CR) on the proliferation and migration of PASMCs and vascular remodeling in experimental models of PAH. Our results showed that berberine and quercetin effectively inhibited the proliferation and migration of hypoxia-induced PASMCs in a manner likely to be mediated by the suppression of MAPK1, NADPH oxidase 4 (NOX4), and cytochrome P450 1B1 (CYP1B1) expression. Furthermore, berberine and quercetin treatment attenuates pulmonary hypertension, reduces right ventricular hypertrophy, and improves pulmonary artery remodeling in monocrotaline-induced pulmonary hypertension in rat models. In conclusion, this research demonstrates CR might be a promising treatment option for PAH, and the network pharmacology approach can be an effective tool to reveal the potential mechanisms of Chinese herbal medicine.

Most previous affective studies use facial expression pictures, music or movie clips as emotional stimuli, which are either too simplified without contexts or too dynamic for emotion annotations. In this work, we evaluate the effectiveness of oil paintings as stimuli. We develop an emotion stimuli dataset with 114 oil paintings selected from subject ratings to evoke three emotional states (i.e., negative, neutral and positive), and acquire both EEG and eye tracking data from 20 subjects while watching the oil paintings. Furthermore, we propose a novel affective model for multimodal emotion recognition by 1) extracting informative features of EEG signals from both the time domain and the frequency domain, 2) exploring topological information embedded in EEG channels with graph neural networks (GNNs), and 3) combining EEG and eye tracking data with a deep autoencoder neural network. From the exper-iments, our model obtains an averaged classification accuracy of 94.72 % ± 1.47 %, which demonstrates the feasibility of using oil paintings as emotion elicitation material.

In the mechanical analysis of steel structures, whether it is static analysis or dynamic analysis, it is necessary to establish the structural stiffness matrix first. In the process of building the structural stiffness matrix, the same element usually has different node code connection orders, and it has never been argued whether the different connection orders of the same element will have an effect on the building of the stiffness matrix. In this study, the influence of the difference in the node connection order on the construction of the element stiffness matrix is studied. First, the structural element stiffness matrix in the global coordinate system is established when the node connection order is different. It is found that the element stiffness matrix in the global coordinate system is indeed inconsistent for the same element with different connection orders. In this study, the elements of the established element stiffness matrix are extracted into the global stiffness matrix of the structural system based on the law of energy conservation; it is found that the global stiffness matrix finally established by using two different connection relationships is the same. The research results of the example show that in the stress analysis of steel structures, selecting different node connection sequences to establish the structural stiffness matrix will obtain the element stiffness matrix under different global coordinate systems. However, through the aggregation process of the global stiffness matrix of the structural system, the global stiffness matrix obtained is consistent, so the different connection sequences of nodes will not affect the stress analysis of steel structures. The example further analyzes the static stress and dynamic responses of the steel structure. The conclusions of this study provide a reliable theoretical basis for the situation that the order of node connections need not be consistent in the finite element modeling of steel structures and are of reference value for the finite element modeling of steel structures.

Background Mucormycosis is a rare but serious opportunistic fungal infection that occurs in immunocompromised individuals, especially those with diabetic ketoacidosis. Presently, early diagnosis of the disease remains a challenge for clinicians. Case presentation The patient, a 68-year-old woman with type 2 diabetes mellitus, was admitted with paroxic sharp pain in the left upper abdomen. CT imaging revealed a patchy hypodense shadow of the spleen with wedge-shaped changes. The patient was not considered early for fungal infection. The diagnosis of spleen mucormycosis was not confirmed until pathological biopsy after splenectomy. After surgery, blood glucose level was controlled, acidosis was corrected, and antifungal therapy was effective. Conclusions We report here, for the first time ever, a case of isolated splenic mucormycosis secondary to diabetic ketoacidosis that was diagnosed and treated with antifungal drugs and splenectomy. Following splenectomy, the presence of splenic mucormycosis was confirmed when characteristic mycelia were observed in a tissue biopsy. As the location of any fungal infection is extremely relevant for treatment options and prognoses, early diagnosis and clinical intervention can greatly affect outcomes and prognoses for patients.

PurposeThe purpose of this paper is to recognize and label the faults in wind turbines with a new density-based clustering algorithm, named contour density scanning clustering (CDSC) algorithm.Design/methodology/approachThe algorithm includes four components: (1) computation of neighborhood density, (2) selection of core and noise data, (3) scanning core data and (4) updating clusters. The proposed algorithm considers the relationship between neighborhood data points according to a contour density scanning strategy.FindingsThe first experiment is conducted with artificial data to validate that the proposed CDSC algorithm is suitable for handling data points with arbitrary shapes. The second experiment with industrial gearbox vibration data is carried out to demonstrate that the time complexity and accuracy of the proposed CDSC algorithm in comparison with other conventional clustering algorithms, including k-means, density-based spatial clustering of applications with noise, density peaking clustering, neighborhood grid clustering, support vector clustering, random forest, core fusion-based density peak clustering, AdaBoost and extreme gradient boosting. The third experiment is conducted with an industrial bearing vibration data set to highlight that the CDSC algorithm can automatically track the emerging fault patterns of bearing in wind turbines over time.Originality/valueData points with different densities are clustered using three strategies: direct density reachability, density reachability and density connectivity. A contours density scanning strategy is proposed to determine whether the data points with the same density belong to one cluster. The proposed CDSC algorithm achieves automatically clustering, which means that the trends of the fault pattern could be tracked.

Common prosperity is an essential requirement of socialism and an important feature of China’s modernization. Based on panel data from 159 prefecture-level cities in China between 2010 and 2019, combined with a fixed-effects model, mediation regression model, and spatial Durbin model, this study empirically examines the impact and mechanism of labor mobility on common prosperity. The study found that (1) labor mobility can significantly promote the realization of common prosperity, and this conclusion is robust. (2) Labor mobility has a significant mediating effect on the improvement in the common prosperity level by promoting economic growth spillovers and return effects. (3) The impact of labor mobility on common prosperity is heterogeneous, with regions with lower mobility costs and more developed economies showing stronger promotion effects. Based on the research findings, policy recommendations include breaking down barriers to mobility, protecting the rights of farmers, and establishing regional cooperation mechanisms.

The enhancement of aerodynamic performance of airfoil using Gurney flap was demonstrated and due to its effects of lift increasing and stall suppression on airfoil, Gurney flap has positively functioned in a wide range of engineering. Though there had been numerous scholars who aroused wide concern on geometric parameters of Gurney flap for airfoil, the investigation of geometric design of Gurney flap for straight-bladed vertical axis wind turbine is absent. In current research, a comprehensive numerical study on the effect of geometric parameters of Gurney flap on performance enhancement of straight-bladed vertical axis wind turbine. Before a validation between numerical results and experimental data was carried out, a novel mathematical model of resistance torque of struts was put forward to increase the reliability of numerical model. The results show that the Gurney flap can remarkably promote the aerodynamic performance of vertical axis wind turbine with a decreased rotational velocity. The maximum improvement can reach up to 21.32%. In upstream area, Gurney flap can considerably increase the blade tangential force. Short-Gurney-flap blade can effective weaken the deficiency of aerodynamic loss in downstream area. The 0.75%-chord-length height is most appropriate for the straight-bladed vertical axis wind turbine in this paper. The aerodynamic load with Gurney flap is not sensitive to width. The design with 0.12%-chord-length width is the most suitable value through analyzing the power coefficient curve.

The rising prevalence of cardiovascular disease has become a global health concern. The occurrence of cardiovascular disease is the result of long-term interaction of many risk factors, one of which is diabetes. As a novel anti-diabetic drug, DPP4 inhibitor has been proven to be cardiovascular safe in five recently completed cardiovascular outcome trials. Accumulating studies suggest that DPP4 inhibitor has potential benefits in a variety of cardiovascular diseases, including hypertension, calcified aortic valve disease, coronary atherosclerosis, and heart failure. On the one hand, in addition to improving blood glucose control, DPP4 inhibitor is involved in controlling cardiovascular risk factors. On the other hand, DPP4 inhibitor directly regulates the occurrence and progression of cardiovascular diseases through a variety of mechanisms. In this review, we summarize the recent advances of DPP4 in cardiovascular disease, aiming to discuss DPP4 inhibitor as a potential option for cardiovascular therapy.

As a multidrug-resistant pathogen

Background CD20-negative diffuse large B-cell lymphoma is a very rare and heterogeneous invasive cancer characterized by chemical resistance and poor prognosis. Primary CD20-negative diffuse large B-cell lymphoma of the central nervous system is even rarer, presenting great challenges in pathological diagnosis and clinical treatment. Case presentation We report a case of primary CD20-negative diffuse large B-cell lymphoma of the CNS in a 54-year-old woman admitted to the hospital with a headache lasting more than 10 days. CT and MRI scans showed right temporal lobe lymphoma. Microscopically, large infiltrating lymphoid cells that induced brain tissue damage were observed. Immunohistochemistry showed that the tumor cells were CD79a+, PAX-5+, MUM1+, and CD20-. The patient was diagnosed with lymphoma and transferred to an oncology hospital for chemotherapy. However, because the disease progressed rapidly, the patient died only after two rounds of chemotherapy. Conclusions To the best of our knowledge, this is one of the first reported cases of unclassifiable CD20-negative diffuse large B-cell lymphoma located in the CNS. This case report aims to deepen the understanding of clinicopathological features of this type of lymphoma and expand the scope of this disease.

Efficient drug loading, tumor targeting, intratumoral penetration, and cellular uptake are the main factors affecting the effectiveness of drug delivery systems in oncotherapy. Based on the tumor microenvironment, we proposed to develop Curcumin (Cur)-loaded matrix metalloproteinase (MMP)-responsive nanoparticles (Cur-P-NPs) by static electricity, to enhance tumor targeting, cellular uptake, and drug loading efficiency. These nanoparticles combine the properties of both PEG-peptides (cleaved peptide + penetrating peptide) and star-shaped polyester (DPE-PCL) nanoparticles. Cur-P-NPs displayed good entrapment efficiency, drug loading and biocompatibility. Additionally, they showed an enhanced release rate, cellular uptake, and anti-proliferative activity by activating peptides under the simulated tumor microenvironment. Furthermore, intraperitoneal injection of losartan (LST) successfully enhanced intratumoral drug penetration by collagen I degradation. In vivo studies based on the systematic administration of the synergistic LST + Cur-P-NPs combination to mice confirmed that combined antitumor therapy with LST and Cur-P-NPs could further improve intratumor distribution, enhance anticancer efficacy, and reduce the toxicity and side effects. Therefore, LST + Cur-P-NPs represent a new and efficient system for clinical oncotherapy.

Migration is considered to be a key step in the data processing of the seismic advancement predictions of tunnels, and it directly affects the final interpretations. Therefore, with the goal of addressing the limitations of the current acoustic reverse‐time migration processes, as well as deepening the understanding of the interactions between different wave modes in traditional elastic reverse‐time migration, a vector P‐ and S‐wave reverse‐time migration method is proposed in this study. Consideration is given to the actual tunnel spaces, and forward calculations and reverse‐time extrapolations of the wavefields are carried out based on the first‐order velocity‐stress elastic wave equation of P‐ and S‐wave separation and a high‐order staggered‐grid algorithm. The imaging conditions of a source‐normalized cross‐correlation are used to realize the imaging. Then, a Laplace filtering method is adopted to suppress the low‐frequency artefacts. Numerical simulations of the weak interlayer and karst cave models show that the proposed method had the ability to more effectively suppress noise and achieve high‐precision imaging of tunnel spaces, when compared with using horizontal or vertical components to directly obtain the images. The positions and shapes within the tunnel spaces can be accurately imaged regardless of whether they are lithologic interfaces or local geological bodies. This is found to improve the resolution and accuracy of the tunnel advancement predictions. Finally, the proposed method is successfully applied to the real data processing, and the effects are found to be satisfactory.

Y-chromosome-specific short tandem repeat loci (Y-STRs) are commonly analysed in forensic science for paternity testing, familial searches, and, in sexual assault cases, to determine male DNA identity from mixed sources with high background female DNA content. The Microreader 40Y ID System is a six-dye multiplex amplification kit that contains 17 Y-STR loci from the Yfiler Plus PCR Amplification Kit and the powerplex Y23 system (DYS19, DYF385a/b, DYS389I/II, DYS390, DYS391, DYS392, DYS393, DYS437, DYS438, DYS439, DYS448, DYS456, DYS458, DYS549, DYS635(Y GATA C4), DYS643, Y GATA H4, DYS460, DYS481, DYS533, DYF387S1, DYS449, DYS518, DYS570, DYS576, and DYS627), plus six high polymorphic loci (DYS444, DYS447, DYS557, DYS596, DYS527 a/b) as well as 4 additional candidate Y-STR loci (DYS593, DYF404S1, DYS645) and a Y-Indel loci (Rs2032678), thereby providing greater efficiency, compatibility, and accuracy. The Microreader 40Y ID System can directly amplify markers from blood or saliva on filter paper or FTA cards, without template extraction or purification, and can also be used for extracted DNA templates. To verify the efficiency and accuracy of the kit, the Microreader 40Y ID System was validated by investigating sensitivity, amplification conditions, male-male and male-female mixtures, PCR inhibition, species specificity, reproducibility, and efficacy with degraded samples. The Y-STR loci were also tested using 437 male samples from Tibet, Han, and Yi. The Microreader 40Y ID System was able to compensate for some of the shortcomings of Y-STR markers in practical applications, such as cost and profile interpretation, and fully meets the domestic Y chromosome database construction specifications and requirements.

Microwave-synthesized SiO2-reinforced B-N-co-doped reduced graphene oxide (SiO2-B-N-GO) nanocomposites were characterized by X-ray photon spectroscopy (XPS), X-ray diffraction (XRD), infrared (IR) spectroscopy, and transmission electron microscopy/energy dispersive X-ray (TEM/EDX) analysis. The tribological properties of the SiO2-B-N-GO prepared as anti-wear additives for enhanced lubrication were studied using a four-ball tester. The experiment results indicated that SiO2-B-N-GO exhibits excellent load-carrying, anti-wear, and anti-friction properties in a base oil, especially at the optimal concentration of additives at 0.15 wt%. The wear scar diameter decreased from 0.70 to 0.37 mm and the coefficient of friction was reduced from 0.092 to 0.070, which reductions are attributed to the formation of B-N and graphene layer tribofilms of several tens of nanometers in thickness that prevented direct contact between metals.

Toxicity and environmental issues have elicited research attention regarding the need to prepare a green flame retardant with high flame retardancy. Here, a supermolecular self-assembly technology was used to prepare nickel phytate as shell materials aggregated on aminated silica nanotemplates through electrostatic interactions as a green novel flame retardant (Ni@SiO2-PA). After incorporating the obtained core–shell structured Ni@SiO2-PA into epoxy resin (EP), the supermolecular shell effectively enhanced the adhesive property between the nanoparticles and the EP matrix. The thermal stability was improved, and the peak heat release rate decreased significantly after introducing the well-characterized Ni@SiO2-PA. The absorbance intensity of the toxic aromatic compounds also decreased. Moreover, the char yield of the EP composites was improved because of the synergetic coupled effects between the nickel phytate supermolecules and SiO2 nanotemplates. The possible fire-retardancy mechanism was hypothesized as follows. The crosslinking structure of the silica initially enabled the formation of a polymer network to prevent further decomposition. The N–P synergistic flame-retardancy system then generated a gas barrier and P-rich intumescent char. Besides, char-residue generation was catalyzed by introducing Ni2+, which isolated the heat and the exchange between oxygen and the matrix. Overall, this study proposes a novel green flame retardant that may enable significant improvements in preparing environmentally friendly organic–inorganic materials with applications in the fields of flame-retardant composites.

Sauce-flavored Daqu exhibits different colors after being stacked and fermented at high temperatures. Heiqu (black Daqu, BQ) with outstanding functions is difficult to obtain because its formation mechanism is unclear. In this study, we compared the metabolites in different types of Daqu using ultra-high-performance liquid chromatography triple quadrupole mass spectrometry to explore the formation process of BQ. We found that 251 differential metabolites were upregulated in BQ. Metabolic pathway analysis showed that “tyrosine metabolism” was enriched, and most metabolites in this pathway were differential metabolites upregulated in BQ. The tyrosine metabolic pathway is related to enzymatic browning and melanin production. In addition, the high-temperature and high-humidity fermentation environment of sauce-flavored Daqu promoted an increase in the melanoidin content via a typical Maillard reaction; thus, the melanoidin content in BQ was much higher than that in Huangqu and Baiqu. By strengthening the Maillard reaction precursor substances, amino acids, and reducing sugars, the content of Daqu melanoidin increased significantly after simulated fermentation. Therefore, the enzymatic browning product melanin and Maillard reaction product melanoidin are responsible for BQ formation. This study revealed the difference between BQ and other types of Daqu and provides theoretical guidance for controlling the formation of BQ and improving the quality of liquor.

Atherosclerosis-related cardiovascular diseases are leading causes of mortality worldwide, characterized by the development of endothelial cell dysfunction, increased oxidized low-density lipoprotein uptake by macrophages, and the ensuing formation of atherosclerotic plaque. Local blood flow patterns cause uneven atherosclerotic lesion distribution, and endothelial dysfunction caused by disturbed flow is an early step in the development of atherosclerosis. The present research aims to elucidate the mechanism underlying the regulation of Neuropilin 2 (NRP2) under low shear stress (LSS) in the atheroprone phenotype of endothelial cells. We observed that NRP2 expression was significantly upregulated in LSS-stimulated human umbilical vein endothelial cells (HUVECs) and in mouse aortic endothelial cells. Knockdown of NRP2 in HUVECs significantly ameliorated cell apoptosis induced by LSS. Conversely, overexpression of NRP2 had the opposite effect on HUVEC apoptosis. ObjectiveTo determine if the early response assessments can predict the long-term efficacy of anti-vascular endothelial growth factor (VEGF) treatment for macular edema secondary to retinal vein occlusion (RVO-ME).MethodsA retrospective study of patients with diagnosis of RVO-ME and intravitreal anti-VEGF treatment was conducted. Clinical characteristics including age, gender, disease subtype and disease duration were recorded at baseline. The best corrected visual acuity (BCVA and logMAR), intraocular pressure (IOP), and central macular thickness (CMT) were recorded at baseline, 2 weeks, and every month (months 1–6) after injection. Further, we compared the early response assessments between the cured group (6-month CMT ≤ 250 μm) and the uncured group (6-month CMT > 250 μm).ResultsA total of 164 eyes in 164 patients (77 male and 87 female) were included. At each post-injection time point, both BCVA and CMT are significantly decreased from baseline (all P < 0.001). Spearman’s test showed that 2-week CMT reduction rate after the first injection was negatively correlated with BCVA at 6 months (r = −0.359, P < 0.001). Compared with the uncured group (47 cases), the cured group (117 cases) was younger (59.53 ± 11.68 vs. 65.19 ± 13.10 years old, P < 0.01), had more BRVO patients (76.1% vs. 44.7%, P < 0.01), a shorter disease duration (1.92 ± 2.43 vs. 5.05 ± 4.32 months, P < 0.01), lower baseline CMT (527.09 ± 154.95 vs. 768.96 ± 287.75 μm, P < 0.01), and lower baseline BCVA (0.86 ± 0.44 vs. 1.31 ± 0.51, P < 0.01). At each post-injection time point, the cured group had lower CMT and BCVA values when compared to the uncured group (all P < 0.01), and the 2-week CMT reduction rate was identified as the earliest response time to predict the long-term treatment efficacy. Moreover, ROC curve analysis indicated that a 2-week CMT reduction rate >37% yielded the best cut-off point for predicting the long-term cure of anti-VEGF treatment at 6 months (P < 0.001). Multivariable logistic regression confirmed that the 2-week CMT reduction rate >37% was independently associated with the 6-month cured rate (OR = 9.639, 95% Cl = 1.030–90.227, P = 0.047).ConclusionAge, disease duration, baseline CMT, and baseline BCVA are associated with visual outcomes at 6-month of anti-VEGF treatment for RVO-ME. The “2-week CMT reduction rate >37%” after the first injection is an independent factor to predict better long-term outcomes.
