Dislocations play a critical role in metal forming processes, and accurate values of dislocation density are important in modelling these processes. However, direct determination of the dislocation density is challenging. In this study, electron backscatter diffraction is used to estimate the evolution of geometrically necessary dislocation density as a function of plastic strain, strain rate and temperature in hot compression of AA7050 alloy. The geometrically necessary dislocation density was found to increase at a higher strain rate and lower temperature; the higher dislocation density in these samples promoted continuous dynamic recrystallisation leading to grain refinement. At lower strain rates and higher temperatures, the dislocation densities were lower and dislocations formed into walls, channels and cells. These observations agree with accepted theories of dislocation evolution and demonstrate the capability of electron backscatter diffraction to provide representative dislocation density values as well as comprehensive information linking plastic flow with microstructural evolution.Keywords: electron backscatter diffraction (EBSD), geometrically necessary dislocations (GNDs), hot deformation, AA7050

Objective: As one of the most prevalent psychiatric disorders, the exact pathogenesis of depression remains elusive. Therefore, there is an urgent need to identify novel antidepressants for effective treatment. MicroRNA-124 (miR-124), the most abundant miRNA in brain tissue, plays a key effect on adult neurogenesis and neuronal differentiation. However, the mechanism of miR-124 in depression has not been clarified so far. The aim of this study is to provide broad insight into the mechanisms underlying depression. Methods: In the study, we used the forced swim test (FST), the tail suspension test (TST), and a Chronic Social Defeat Stress (CSDS) mice model of depression. Quantitative real-time reverse transcription PCR (qRT-PCR), western blotting, immunofluorescence and virus-mediated gene transfer were used together. The level of plasma corticosterone in mice was analyzed by Enzyme Linked Immunosorbent Assay (ELISA). Results: It was found that CSDS robustly increased the level of miR-124 in the hippocampus. Genetic knockdown of hippocampal miR-124 produced significant antidepressant-like effects in the CSDS model of depression. Furthermore, AAV-siR-124-EGFP treatment increased the level of plasma corticosterone in CSDS-induced mice. Moreover, it was found that the antidepressant-like effects induced by miR-124 inhibition required the hippocampal BDNF-TrkB system. Conclusion: Hippocampal miR-124 participated in the pathogenesis of depression by regulating BDNF biosynthesis and was a feasible antidepressant target.

Bone scaffolds require both good bioactivity and mechanical properties to keep shape and promote bone repair. In this work, T-ZnOw enhanced biphasic calcium phosphate (BCP) scaffolds with triply periodic minimal surface (TPMS)-based double-layer porous structure were fabricated by digital light processing (DLP) with high precision. Property of suspension was first discussed to obtain better printing quality. After sintering, T-ZnOw reacts with β-tricalcium phosphate (β-TCP) to form Ca19Zn2(PO4)14, and inhibits the phase transition to α-TCP. With the content of T-ZnOw increasing from 0 to 2 wt%, the flexural strength increases from 40.9 to 68.5 MPa because the four-needle whiskers can disperse stress, and have the effect of pulling out as well as fracture toughening. However, excessive whiskers will reduce the cure depth, and cause more printing defects, thus reducing the mechanical strength. Besides, T-ZnOw accelerates the deposition of apatite, and the sample with 2 wt% T-ZnOw shows the fastest mineralization rate. The good biocompatibility has been proved by cell proliferation test. Results confirmed that doping T-ZnOw can improve the mechanical strength of BCP scaffolds, and keep good biological property, which provides a new strategy for better bone repair.

As the weakest segment within frozen soil, the interface between ice and frozen soil undertakes the link of load transmission. The mechanical parameters between ice and frozen soil influence the deformation and strength of frozen soil. To investigate the mechanical behaviors of ice-frozen soil interface, the cryogenic direct shear tests were carried out and the deformation, strength properties of interface were analyzed. The shear stress of interface was described by generalized hyperbolic model and the physical meaning of the model parameters were defined. Based on the generalized hyperbolic model, the shear stiffness of interface was derived. The effect of temperature on the strength, shear stiffness of interface under different initial moisture contents, initial void ratios and normal stresses were discussed. The three-dimensional finite element model was established using COMSOL Multiphysics, and the mechanical behaviors of ice frozen soil interface were researched.

ObjectiveCentral precocious puberty (CPP) is a rare condition that causes early sexual development in children. Although the cure is effective, the etiology of central precocious puberty is unclear.MethodsIn total, 10 girls with central precocious puberty and same number of age-matched female controls were enrolled. Plasma samples were collected from each participant and subjected to untargeted metabolomics and lipidomics. Student's t-tests were employed to compare the mean of each metabolite and lipid. Furthermore, orthogonal partial least-squares discriminant analysis was conducted and the variable importance in the projection was calculated to identify differentially expressed metabolites or lipids. Subsequent bioinformatics was conducted to investigate the potential function of differentially expressed metabolites and lipids.ResultsFifty-nine differentially expressed metabolites were identified based on the criteria used (variable importance in the projection >1 and a P value ConclusionThe present study showed that antibiotic overuse, increased meat consumption, and obesity may have potential roles in the development of central precocious puberty in girls. Several metabolites have diagnostic value but further research is required.

BackgroundMultiple biomarkers have been identified by previous studies to diagnose acute kidney injury (AKI). Moreover, combination of biomarkers with conventional criteria to define AKI substages so that we can identify high-risk patients and improve diagnostic accuracy were recommended. Our study aimed to explore the incidence of AKI substages defined by serum cystatin C (CysC), determine whether AKI substages were associated with worse outcomes.MethodsWe prospectively included 2519 pediatric patients (Results507 (20.8%) patients developed SCr-AKI, with 337 (13.8%) in stage 1, 77(3.2%) in stage 2 and 93 (3.8%) in stage 3 respectively. Of the 1925 patients without SCr-AKI, 256 (14.3%) met the definition of sub-AKI. Of the 507 patients with SCr-AKI, 281 (55.4%) patients were defined as AKI substage A, while others (226, 44.6%) were defined as AKI substage B. After adjusting for BSA, neonates, STAT mortality score≥4, previous sternotomy and CPB time>120min, the postoperative LOIS, LOHS and DMV were prolonged with increasing hospitalization expense (PConclusionsOur analysis indicates defining AKI with both CysC and SCr might more significantly affecting clinical outcome associations in pediatric patients undergoing cardiac surgery. Moreover, the serum CysC cutoff of 1.29mg/dL postoperatively is a valuable threshold for AKI risk assessment to define AKI subtypes.
