Checkpoint kinase 1 (CHK1; encoded by CHEK1) is an essential gene that monitors DNA replication fidelity and prevents mitotic entry in the presence of under-replicated DNA or exogenous DNA damage. Cancer cells deficient in p53 tumor suppressor function reportedly develop a strong dependency on CHK1 for proper cell cycle progression and maintenance of genome integrity, sparking interest in developing kinase inhibitors. Pharmacological inhibition of CHK1 triggers B-Cell CLL/Lymphoma 2 (BCL2)-regulated cell death in malignant cells largely independently of p53, and has been suggested to kill p53-deficient cancer cells even more effectively. Next to p53 status, our knowledge about factors predicting cancer cell responsiveness to CHK1 inhibitors is limited. Here, we conducted a genome-wide CRISPR/Cas9-based loss-of-function screen to identify genes defining sensitivity to chemical CHK1 inhibitors. Next to the proapoptotic BCL2 family member, BCL2 Binding Component 3 (BBC3; also known as PUMA), the F-box protein S-phase Kinase-Associated Protein 2 (SKP2) was validated to tune the cellular response to CHK1 inhibition. SKP2 is best known for degradation of the Cyclin-dependent Kinase Inhibitor 1B (CDKN1B; also known as p27), thereby promoting G1-S transition and cell cycle progression in response to mitogens. Loss of SKP2 resulted in the predicted increase in p27 protein levels, coinciding with reduced DNA damage upon CHK1-inhibitor treatment and reduced cell death in S-phase. Conversely, overexpression of SKP2, which consequently results in reduced p27 protein levels, enhanced cell death susceptibility to CHK1 inhibition. We propose that assessing SKP2 and p27 expression levels in human malignancies will help to predict the responsiveness to CHK1-inhibitor treatment.

Background After the 2002/2003 SARS outbreak, 30% of survivors exhibited persisting structural pulmonary abnormalities. The long-term pulmonary sequelae of coronavirus disease 2019 (COVID-19) are yet unknown, and comprehensive clinical follow-up data are lacking. Methods In this prospective, multicentre, observational study, we systematically evaluated the cardiopulmonary damage in subjects recovering from COVID-19 at 60 and 100 days after confirmed diagnosis. We conducted a detailed questionnaire, clinical examination, laboratory testing, lung function analysis, echocardiography, and thoracic low-dose computed tomography (CT). Results Data from 145 COVID-19 patients were evaluated, and 41% of all subjects exhibited persistent symptoms 100 days after COVID-19 onset, with dyspnea being most frequent (36%). Accordingly, patients still displayed an impaired lung function, with a reduced diffusing capacity in 21% of the cohort being the most prominent finding. Cardiac impairment, including a reduced left ventricular function or signs of pulmonary hypertension, was only present in a minority of subjects. CT scans unveiled persisting lung pathologies in 63% of patients, mainly consisting of bilateral ground-glass opacities and/or reticulation in the lower lung lobes, without radiological signs of pulmonary fibrosis. Sequential follow-up evaluations at 60 and 100 days after COVID-19 onset demonstrated a vast improvement of both, symptoms and CT abnormalities over time. Conclusion A relevant percentage of post-COVID-19 patients presented with persisting symptoms and lung function impairment along with pulmonary abnormalities more than 100 days after the diagnosis of COVID-19. However, our results indicate a significant improvement in symptoms and cardiopulmonary status over time., 100 days after COVID-19 onset, a high portion of patients exhibit persisting symptoms and cardiopulmonary impairment. Still, a marked improvement of symptoms, pulmonary function, and CT pathologies was found at follow-up.

Carboligations catalyzed by aldolases or thiamine diphosphate (ThDP)-dependent enzymes are well-known in biocatalysis to deliver enantioselective chain elongation reactions. A pyruvate-dependent aldolase (2-oxo-3-deoxy-6-phosphogluconate aldolase [EDA]) introduces a chiral center when reacting with the electrophile, glyoxylic acid, delivering the (S)-enantiomer of (4S)-4-hydroxy-2-oxoglutarate [(S)-HOG]. The ThDP-dependent enzyme MenD (2-succinyl-5-enol-pyruvyl-6-hydroxy-3-cyclohexene-1-carboxylate synthase (SEPHCHC synthase)) enables access to highly functionalized substances by forming intermolecular C?C bonds with Michael acceptor compounds by a Stetter-like 1,4- or a benzoin-condensation 1,2-addition of activated succinyl semialdehyde (ThDP adduct formed by decarboxylation of 2-oxoglutarate). MenD-catalyzed reactions are characterized by high chemo- and regioselectivity. Here, we report (S)-HOG, in situ formed by EDA, to serve as new donor substrate for MenD in 1,4-addition reactions with 2,3-trans-CHD (2,3-trans-dihydroxy-cyclohexadiene carboxylate) and acrylic acid. Likewise, (S)-HOG serves as donor in 1,2-additions with aromatic (benzaldehyde) and aliphatic (hexanal) aldehydes. The production of hydrolases from cereals has been examined in order to investigate food-derived enzymes as an alternative source to microbial enzymes for the use in food processes. For that, the influence of temperature on the pretreatment, imbibition and germination of barley and wheat grains was determined by measuring the β-glucosidase, β-galactosidase and lipase activities using a design of experiments. The evaluation of the statistical model showed an increase of the β-glucosidase activity with low imbibition and low germination temperature for barley grains and low imbibition and high germination temperature for wheat grains. The maximum β-glucosidase activity in wheat extracts was (585±151) nkat per g of dry mass (dm), while in barley extracts it was (109±15) nkat per g of dm. The maximum β-galactosidase activities in barley and wheat extracts were (34±12) and (63±23) nkat per g of dm, respectively. The maximum lipase activities of (6.7±0.1) and (4.6±4.4) nkat per g of dm in barley and wheat extracts, respectively, were rather low compared to the glycosidase activities. The extracts were also tested for other hydrolase activities (e.g. peptidase and α-amylase activities). The insights obtained enable the basis for the potential use of cereal hydrolases in food processing, which might be attractive to consumers.

In der hier vorliegenden Studie finden sich die Schnittflachen zwischen Wissen, Konnen und Handlungspraxis von padagogischen MitarbeiterInnen in den Handlungsfeldern der auserschulischen Kinder- und Jugendarbeit rekonstruiert und hinsichtlich der habituellen beruflichen Profile fallkontrastierend verdichtet und theoretisch kodiert. Trotz einiger Unscharferelationen und noch auszubuchstabierender theoretischer Bezugspunkte ist gezeigt, das die in der auserschulischen Kinder- und Jugendarbeit tatigen AkademikerInnen ihre berufliche Praxis nicht ausnahmslos und wesentlich uber wissenschaftlich kodifiziertes Wissen abstutzen und begrunden, das absolvierte Studium fur die Performanz der beruflichen Praxis nicht durchgangig als notwendig ansehen und auch nur potentiell zur Abstutzung von Sprachspielen heranziehen.

Diplom-PadagogInnen, SozialpadagogInnen und die MitarbeiterInnen in den Handlungsfeldern der Sozialen Arbeit mit anderen Ausbildungen stehen in ihrer beruflichen Praxis vor der Bewaltigung eines gemeinsamen Problems. Im Gegensatz zu anderen, klassischen Professionen konnen sie das »Praktisch-Werden« ihrer berufsorientierten Kompetenzen uber nur schwach normierte oder ritualisierte Schablonen abstutzen, weil keine berufsspezifische Professionalisierungstypik vorliegt, die erstens die beruflichen Integrationsprozesse steuert, zweitens in der beruflichen Praxis eine strukturell verankerte, habituelle Sicherheit bereitstellt und daruber drittens wissenschaftliches Wissen berufsorientiert kodifiziert. Mit anderen Worten: Den in den Handlungsfeldern der modernen sozialpadagogischen Arbeit der auserschulischen Kinder- und Jugendarbeit professionell Engagierten stehen weder anerkannte, kollektive, generalisierte disziplinare Wissensressourcen noch ethisch-normative Zeichen- und Bewertungssysteme und keine Netzwerke von vorstrukturierten Handlungsfolien und institutionell gebundenen Regeln abrufbar bereit, die die beruflichen Handlungen und subjektiven Habitualisierungen steuern und berufstypisch kanalisieren.

Der 32jahrige Diplom-Sozialpadagoge Dieter Brattke leitet seit vier Jahren die Jugendfreizeiteinrichtung einer niedersachsischen Kleinstadt, die in einem grosstadtischen Einzugsgebiet liegt. In Hamburg geboren — sein Vater war zum Zeitpunkt seiner Geburt als Diakon in einem Seemannsheim beschaftigt — werden die Orte des Aufwachsens durch die berufliche Mobilitat des Vaters bestimmt. In Frankfurt leitet der Vater anschliesend ein Aussiedlerheim, darauf folgend obliegt ihm die Leitung des erzieherischen Dienstes in einem familienersetzenden Jungenheim der Evangelischen Kirche, dem »Sanderhof« in Niedersachsen. Hier verbringt der inzwischen funfjahrige Dieter Brattke auch den Grosteil seiner Kindheit und Jugend. Seine Mutter, eine gelernte Kinderkrankenschwester, ubernimmt neben ihrer Tatigkeit als Hausfrau ehrenamtliche Aufgaben im hauswirtschaftlichen Bereich des Jugendheimes. Erst als Dieter Brattkes sechs Jahre jungere Schwester, die heute im Ausland lebt, zur Schule geht, nimmt sie eine Halbtagstatigkeit in einem Kindergarten auf.

Gewissermasen als einer der ersten Vorlaufer der gegenwartigen professionsorientierten erziehungswissenschaftlichen und sozialwissenschaftlichen Vergewisserungen kann die in den zwanziger Jahren dieses Jahrhunderts publizierte Analyse »Erziehung als Beruf« von Aloys Fischer angesehen werden. Wird in den gegenwartigen Diskussionen dem Verberuflichungsprozes und der damit einhergehenden Zunahme von erziehungswissenschaftlichen Begrundungen padagogischen Handelns durchaus eine, wenn auch unklare Bedeutung fur die Ausbuchstabierung von padagogischen Handlungszusammenhangen zugestanden, betrachtete Aloys Fischer die Verberuflichung padagogischer Raume zuruckhaltender. Die enormen Institutionalisierungs-, Ausdifferenzierungs- und Modernisierungsschube der schulischen, aber auch der auserschulischen Handlungsfelder vor Augen (vgl. Dewe/Ferchhoff/Radtke 1992), sah er das eigentlich »Padagogische«, die naturhafte, familiale Erziehung, schon Mitte der zwanziger Jahre durch die »Professionalisierung der Erziehung« (Fischer 1961, S. 163) bedroht, weil sie eine »Menschenleistung« darstellt, die nicht einfach an Professionelle abzutreten ist (vgl. Hornstein/Luders 1989, S. 749). »Durch die Entstehung und Entwicklung der Erziehung als Beruf scheint die Erziehung selbst geradezu von innen heraus bedroht.
