Childhood adversity is associated with higher adult weight, but few investigations prospectively test mechanisms accounting for this association. Using two socioeconomically high-risk prospective longitudinal investigations, the Minnesota Longitudinal Study of Risk and Adaptation (MLSRA; N = 267; 45.3% female) and the Fragile Families and Child Wellbeing Study (FFCWS; n = 2,587; 48.5% female), pathways between childhood adversity and later body mass index (BMI) were tested using impulsivity, emotion dysregulation, and overeating as mediators. Childhood adversity from 0 to 5 years included four types of adversities: greater unpredictability, threat/abuse, deprivation/neglect, and low socioeconomic status. Parents reported on child impulsivity, emotion dysregulation, and overeating. Height and weight were self-reported and measured at 32 and 37 years in MLSRA and at 15 years in FFCWS. FFCWS results indicated that threat, deprivation, and low socioeconomic status predicted greater impulsivity and emotion dysregulation at 5 years, which in turn predicted greater overeating at 9 years and higher BMI z-score at 15 years. Early unpredictability in FFCWS predicted higher BMI through greater impulsivity but not emotion dysregulation at age 5. MLSRA regression results replicated the threat/abuse → emotion dysregulation → overeating → higher BMI pathway. These findings suggest that different dimensions of early adversity may follow both similar and unique pathways to predict BMI.

To ensure safe flight under snowy conditions, aircraft manufacturers must demonstrate that each engine and its air inlet system can operate throughout the flight power range of the engine (including idling) in both falling and blowing snow conditions. This study is part of an effort to develop models for snow accretion. The focus here is on the modeling of snowflake transport by the carrier airflow and more specifically on the modeling of the drag coefficient of snowflakes. Based on comparisons between numerical and experimental results, we show that, provided that the complex shape of the snowflakes is accounted for, the models initially developed for ice crystal icing can also be used to estimate the drag coefficient of a snowflake. We also propose a methodology to estimate the geometric parameters of the drag models from two-dimensional images of the particles, which in general are the only available experimental data.

Individual differences in the quality of early experiences with primary caregivers have been reliably implicated in the development of socioemotional adjustment and, more recently, physical health. However, few studies have examined the development of such associations with physical health into the adult years. Prior research has demonstrated that teacher-student relationships characterized by high levels of closeness and low levels of conflict are associated with higher levels of academic achievement among children. At the same time: (a) some research suggests that the quality of teacher-student relationships in part reflects the quality of early caregiving, and (b) the observed quality of early care by primary caregivers robustly predicts subsequent academic achievement. Given the potential for associations between teacher-student relationship quality and academic achievement to thus be confounded by the quality of early parenting experiences, the present study examined to what extent children’s experiences in early life with primary caregivers (i.e., ages 3 to 42 months) and relationships with teachers during grade school (i.e., Kindergarten to Grade 6) were uniquely associated with an objective assessment of academic achievement at age 16 years in a sample born into poverty ( N = 267; 45% female; 65% White/non-Hispanic; 41% of mothers did not complete high school). Early maternal sensitivity, though a strong predictor of later academic achievement, was not reliably associated with either teacher-reports or interview-based assessments of teacher-student relationship quality in grade school. Nonetheless, early maternal sensitivity and teacher-student relationship quality were each uniquely associated with later academic achievement, above and beyond key demographic variables. Taken together, the present results highlight that the quality of children’s relationships with adults at home and at school independently, but not interactively, predicted later academic achievement in a high-risk sample.

An unsettled question in attachment theory and research is the extent to which children's attachment patterns with mothers and fathers jointly predict developmental outcomes. In this study, we used individual participant data (IPD) meta-analysis to assess whether early attachment networks with mothers and fathers are associated with children's internalizing and externalizing behavioral problems. Following a pre-registered protocol, data from 9 studies and 1,097 children (mean age: 28.67 months) with attachment classifications to both mothers and fathers were included in analyses. We used a linear mixed effects analysis to assess differences in children's internalizing and externalizing behavioral problems as assessed via the average of both maternal and paternal reports based on whether children had two, one, or no insecure (or disorganized) attachments. Results indicated that children with an insecure attachment relationship with one or both parents were at higher risk for elevated internalizing behavioral problems compared with children who were securely attached to both parents. Children whose attachment relationships with both parents were classified as disorganized had more externalizing behavioral problems compared to children with either one or no disorganized attachment relationship with their parents. Across attachment classification networks and behavioral problems, findings suggest (a) an increased vulnerability to behavioral problems when children have insecure or disorganized attachment to both parents, and (b) that mother-child and father-child attachment relationships may not differ in the roles they play in children's development of internalizing and externalizing behavioral problems.

Ice accretion is often caused by the nucleation and freezing of a supercooled water drop after its impact on working surfaces, such as aircraft wings, transmission lines or fan blades, it threatens the safe operation of the equipment and even leads to catastrophic failures. Therefore, it is crucial to better understand the physics of supercooled drop impact. The main subject of this study is the observation of a supercooled drop impact and measurements of a freezing time. The experiments are performed in a vertical wind tunnel, installed in a cold room. The phenomenon is observed with a high-speed video system. In the experiments, the air velocity has been varied from 0 to 22.5 m/s. Additionally, three substrates of different roughness have been investigated. It has been found that higher roughness of the substrate leads to a higher probability of nucleation after impact. Additionally, the higher air velocity also prompts earlier nucleation inception. Furthermore, intriguing cavities and voids formed from the drop breakup upon impact on rough substrates have been observed, which have an effect on the nucleation as well.

Unresolved states of mind regarding experiences of loss/abuse (U/d) are identified through lapses in the monitoring of reasoning, discourse, and behavior surrounding loss/abuse in response to the Adult Attachment Interview. Although the coding system for U/d has been widely used for decades, the individual indicators of unresolved loss/abuse have not been validated independently of the development sample. This study examined the psychometric validity of U/d, using individual participant data from 1,009 parent-child dyads across 13 studies. A latent class analysis showed that subsets of commonly occurring U/d indicators could differentiate interviewees with or without unresolved loss/abuse. Predictive models suggested a psychometric model of U/d consisting of a combination of these common indicators, with disbelief and psychologically confused statements regarding loss being especially important indicators of U/d. This model weakly predicted infant disorganized attachment. Multilevel regression analysis showed no significant association between ratings of unresolved other trauma and infant disorganized attachment, over and above ratings of unresolved loss/abuse. Altogether, these findings suggest that the coding system of U/d may have been overfitted to the initial development sample. Directions for further articulation and optimization of U/d are provided.

One parameter frequently considered to be relevant for superspreading of trisiloxane surfactants is surface tension kinetics. In the scientific literature, some experimental results reported for trisiloxane surfactants are in contradiction with fundamental concepts of surfactant monomer diffusion. Therefore, maximum bubble pressure tensiometry has been used to determine dynamic surface tension (DST) of two types of trisiloxane surfactants: superspreader and non-superspreader. Results show that both surfactants behave similarly at concentrations below critical micelle concentration (CMC), as expected. The CMC curves, as determined by drop shape analysis, confirmed that the more hydrophilic non-superspreader has a higher CMC as compared to the more hydrophobic superspreader. Accordingly, the lower surfactant monomer concentration of the superspreader results in a higher DST than the non-superspreader at the same surface age. So, in contrary to claims in the literature, there is nothing mysterious or unexpected concerning the surface tension behavior of trisiloxane surfactants. Graphical Abstract

Spray cooling of solid substrates is one of the methods used in various industrial processes such as forging, quenching or other metallurgical applications, electronics, pharmaceutical industry, medicine, or for cooling of powerful electrical devices. Spray cooling is governed by various hydrodynamic and thermodynamic processes, like drop impact, heat conduction in the substrate and convection in the spreading drops, and different regimes of boiling. The problem of modeling spray cooling becomes even more challenging if the liquid is multicomponent. The presence of components with various physicochemical properties (surfactants, binders, dispersed particles, etc.) can significantly affect the entire process of spray impact, as well as the outcome of the known cooling regimes and could lead to a formation of a thin deposited layer on the substrate. In this experimental study, spray impact onto a substrate, initially heated to temperatures significantly exceeding the liquid saturation point, is visualized using a high-speed video system. The heat transfer associated with spray impact is characterized using an array of thermocouples installed in a thick metal target. As a working fluid, a mixture of a distilled water and industrial white lubricant was used. It is observed that the presence of very small concentrations of lubricant augments the heat flux dramatically, particularly at high wall temperatures, at which usually film boiling is observed for spray cooling by using distilled water. Three main mechanisms lead to the increase of heat flux and shift of the Leidenfrost point. They are caused by the significant viscosity increase of the evaporating lubricant solutions, by an increase of the substrate wettability and by the emergence of stable liquid sheets between bubbles, preventing their coalescence and percolation of the vapor channels.

Aging is associated with functional decline of hematopoietic stem cells (HSC) as well as an increased risk of myeloid malignancies. We performed an integrative characterization of epigenomic and transcriptomic changes, including single-cell RNA sequencing, during normal human aging. Lineage−CD34+CD38− cells [HSC-enriched (HSCe)] undergo age-associated epigenetic reprogramming consisting of redistribution of DNA methylation and reductions in H3K27ac, H3K4me1, and H3K4me3. This reprogramming of aged HSCe globally targets developmental and cancer pathways that are comparably altered in acute myeloid leukemia (AML) of all ages, encompassing loss of 4,646 active enhancers, 3,091 bivalent promoters, and deregulation of several epigenetic modifiers and key hematopoietic transcription factors, such as KLF6, BCL6, and RUNX3. Notably, in vitro downregulation of KLF6 results in impaired differentiation, increased colony-forming potential, and changes in expression that recapitulate aging and leukemia signatures. Thus, age-associated epigenetic reprogramming may form a predisposing condition for the development of age-related AML.Significance:AML, which is more frequent in the elderly, is characterized by epigenetic deregulation. We demonstrate that epigenetic reprogramming of human HSCs occurs with age, affecting cancer and developmental pathways. Downregulation of genes epigenetically altered with age leads to impairment in differentiation and partially recapitulates aging phenotypes.This article is highlighted in the In This Issue feature, p. 983

Despite the widespread utilization of Patient-Derived Xenografts (PDXs) as preclinical platforms in lung cancer research, there are concerns regarding their capability to accurately represent the tumor's clinical and molecular features across sequential passages. In this study, we established a Non-Small Cell Lung Cancer (NSCLC) PDX model in NSG-SGM3 mice and assessed clinical and preclinical factors throughout subsequent passages. Our cohort consisted of 40 NSCLC patients, which were used to successfully create 20 patient-specific PDX models in NSG-SGM3 mice. We found that the main factors that contributed to the growth of the engrafted PDX in mice were a higher grade or stage of disease, in contrast to a long duration of chemotherapy treatment which was negatively correlated with PDX propagation. Successful PDX growth was also linked to poorer prognosis and overall survival, while growth pattern variability was affected by the tumor's aggressiveness, primarily affecting the first passage. Pathology analysis showed preservation of histological type and grade; however, Whole Exome Sequence (WES) analysis revealed genomic instability in advanced passages, leading to the inconsistency of clinically relevant alterations between PDXs and biopsies. Multiple clinical and preclinical factors affect the engraftment success, growth kinetics, and tumor stability of patient-specific NSCLC PDXs; thus, their use for prolonged treatment evaluation studies remains questionable.

Since the technology of direct injected engines has become more and more prevalent, combustion anomalies such as pre-ignition events have been occurring with increasing frequency. The interaction between the fuel-spray and the oil wetted cylinder liner is considered to be a partial cause for these phenomena. The incoming fuel jet can lead to a detachment of secondary droplets, which may affect pre-ignition events due to their changed flammability properties. To characterize these secondary droplets, spatially and temporally highly resolved optical investigations on the fuel-oil-film-interaction are carried out in this work. Using laser induced fluorescence and two high-speed cameras both fluids are optically separated. Thereby the secondary droplets can be detected and characterized in size, velocity vector, and composition.

Este es un estudio de caso con enfoque cuali-cuantitativo y nivel descriptivo, sobre cómo los estudiantes perciben el uso del gestor bibliográfico colaborativo digital. El estudio ayuda a reconocer la relevancia de utilizar dicho gestor para cualificar la investigación y la producción académica, desde proyectos que desarrollan estudiantes de la maestría en Tecnologías Digitales Aplicadas a la Educación. Participaron 103 estudiantes de los módulos: Caracterización de la investigación aplicada y Propuesta de investigación aplicada. Los resultados dejan ver que los tesistas valoran positivamente el gestor como dispositivo tecnológico clave para la gestión de la información y del conocimiento; a su vez, permiten inferir que esta herramienta informática en mención facilita la búsqueda, clasificación y análisis de registros bibliográficos, traduciéndose en adecuadas toma de decisiones y mejora en el desempeño tanto académico como de investigación.

Lung cancer is the most common cancer-related cause of death worldwide, most of which are non-small cell lung cancers (NSCLC). Epidermal growth factor receptor (EGFR) mutations are common drivers of NSCLC. Treatment plans for NSCLC, specifically adenocarcinomas, rely heavily on the presence or absence of specific actionable driver mutations. Liquid biopsy can guide the treatment protocol to detect the presence of various mechanisms of resistance to treatment. We report three NSCLC EGFR mutated cases, each treated with Osimertinib in a combination therapy regimen to combat resistance mechanisms. The first patient presented with EGFR L858R/L833V compound mutation with MET amplification alongside CEP85L-ROS1 fusion gene, the second with EGFR exon 19del and MKRN1-BRAF fusion, and the last EGFR L858R/V834L compound mutation with MET amplification. Each regimen utilized a tyrosine kinase inhibitor or monoclonal antibody in addition to osimertinib and allowed for a prompt and relatively durable treatment response.

Impacts of the COVID-19 pandemic in the United States have been exacerbated by pre-existing inequities in resources and opportunities, leaving the most vulnerable to face a multitude of hardships. The goal of the current study was to characterise COVID-19-related stressful life events in specific life domains and to identify the sociodemographic characteristics of individuals who are more likely to experience such events. Participants (n = 372, 57% female) in a follow-up study of the NICHD Study of Early Child Care and Youth Development completed the Epidemic-Pandemic Impacts Inventory (June-August 2020) to assess COVID-19-related stressors. Sociodemographic factors (gender, race/ethnicity, socioeconomic status and wealth) were examined simultaneously as predictors of the number of stressful life events in separate categories of work/finances, home life, social activity, health and healthcare, adjusted for covariates (household size, community COVID-19 transmission risk). In negative binomial regression analyses, being female (vs. male) predicted a 31%, 64%, 13% and 94% increase in the number of stressful life events in domains of work/finances, home life, social activity and healthcare, respectively, whereas each one standard deviation increase in wealth predicted a 17%, 16% and 21% reduction in the number of stressful life events in domains of work/finances, COVID-19 infection and healthcare, respectively. Findings highlight the pronounced and far-reaching impacts of the COVID-19 pandemic on women as well as the unique role wealth may play in lessening such impacts. This new knowledge may be leveraged to develop intervention and policy-related strategies to remediate impacts of COVID-19-related stressors on those most vulnerable.

La teoría y la investigación sobre el apego se utilizan en muchos contextos aplicados, incluidos los juzgados de familia, pero hay malentendidos muy extendidos y que a veces dan lugar a aplicaciones erróneas. El objetivo de este documento de consenso es, por lo tanto, mejorar la comprensión, contrarrestar la desinformación y dirigir el uso de la teoría del apego por parte de los juzgados de familia en la dirección de un apoyo basado en la evidencia, especialmente en lo que respecta a la protección de niñas y niños y las decisiones sobre su custodia. El artículo se divide en dos partes. En la primera abordamos los problemas relacionados con el uso de la teoría y la investigación sobre apego en los juzgados de familia, analizando las causas de tales problemas. Para ello examinamos las aplicaciones de la teoría del apego en los juzgados de familia en el marco del concepto de interés superior de niños y niñas, analizamos los malentendidos relacionados con la teoría del apego e identificamos los factores que han obstaculizado su adecuada aplicación. En la segunda parte ofrecemos recomendaciones para la aplicación de la teoría y la investigación sobre apego. Para ello establecemos tres principios del apego: la necesidad de niñas y niños de cuidadoras y cuidadores habituales que no maltraten, el valor de la continuidad de cuidados de calidad suficientemente buena y los beneficios de las redes de relaciones de apego. También analizamos la idoneidad de las evaluaciones de los estilos de apego y del comportamiento de los cuidadores para fundamentar la toma de decisiones de los juzgados de familia. Llegamos a la conclusión de que las evaluaciones de quienes cuidan deberían ocupar un lugar central. Aunque no hay consenso entre nosotros sobre el uso de las evaluaciones del estilo de apego para fundamentar las decisiones sobre custodia y protección infantil, dichas evaluaciones son actualmente las más adecuadas para orientar y dirigir las intervenciones de apoyo. Por último, proporcionamos orientaciones para guiar la futura colaboración en la investigación interdisciplinaria.

The dynamics of solid particles of complex shapes such as airborne snowflakes are governed by aerodynamic drag forces that are a function of the relative velocity of the particle in the flow and the particle drag coefficient, which depends on the particle geometry and its orientation. In this study, artificial snowflakes are produced by additive manufacturing and their drag coefficients are obtained by measuring the terminal velocity in a liquid container, matching the Reynolds number typically encountered in natural occurrences. The experimental results show that the convex hull of the particle is suitable to accurately predict the drag force with existing correlations. Since it is unfeasible to accurately measure the three-dimensional geometries of natural snowflakes, the approximation with the convex hull provides a useful simplification. Furthermore, the known shapes of the artificial snowflakes are used to develop correlations to estimate the most relevant three-dimensional descriptors to predict the drag of snowflakes from a two-dimensional projection onto an arbitrary plane. Aims: To describe a Fabry disease, that it’s diagnosis was only possible through the molecular test Presentation of Case: L.A.P. female, 42 years old, lawyer, seen by the ophthalmology department for routine consultation only with refractive complaints. Fundus of the eye: Mild narrowing with increased vascular brightness and presence of pathological arteriovenous crossings. The rest of the exam was within normal limits. Therefore, a genetic test with the dosage of the α-Gal enzyme was requested, which evidenced the alteration in it, confirming the diagnosis of Fabry disease. Discussion: A Fabry Disease (FD) is an inborn error of glycosphingolipid (GL) metabolism, resulting from deficient activity of the enzyme alpha-galactosidase A (α -Gal). It has X-chromosome-linked inheritance, affecting mainly males, with an estimated prevalence of 1:40,000 males. The expression of the disease in heterozygous female patients can vary from an asymptomatic condition to a severe systemic disease, like that which occurs in men. Conclusions: The ophthalmological examination played an important role in the diagnosis, as this change is highly suggestive of the disease, in order to avoid erroneous and late diagnoses that can cause consequences for patients with this condition.

BackgroundNon-small cell lung cancer (NSCLC) harboring EGFR exon 20 insertions (EGFRex20ins) is relatively resistant to the existing EGFR tyrosine kinase inhibitors (TKIs). Mobocertinib is a novel TKI that selectively targets EGFRex20ins and has demonstrated therapeutic efficacy in pretreated patients with tumors harboring these mutations.MethodsThis is a retrospective, non-interventional, multicenter real-world study aimed at assessing the efficacy and safety of mobocertinib in patients with EGFRexon20ins who received 160 mg QD monotherapy as part of expanded access. Data collection was based on patients’ records. PET-CT or CT scans were used to measure systemic response, while brain MRIs were used to examine intracranial response as part of the follow-up.Results16 patients were included in this report. Mobocertinib was administered to 31.3% (5) of patients as first-line, 50% (8) as second-line, and 18.7% (3) as a later-line therapy. The median age was 65 years (range, 38-83), 75% (12/16) were female, and 50% (8/16) had brain metastases at baseline before mobocertinib treatment. The objective response rate (ORR) to mobocertinib was 25% (4/16) (1/5 for first line and 3/11 for other lines), disease control rate (DCR) was 75% (12/16) with a follow-up period of 11 months. The median duration of treatment (mDoT) was 5.6 months across all patients, and 8.6 months in responders. Based on the presence or absence of brain metastasis, the mDoT was 14.8 and 5.4 months (p=0.01), respectively. Mobocertinib Grade ≥3 treatment-related adverse events (TRAEs) included diarrhea (19%), nausea (6%) and renal failure (6%). Dose reduction was reported in 25% of cases to 80 mg.ConclusionMobocertinib in compassionate use exhibited an ORR of 25%, which is very similar to that of the phase 2 EXCLAIM study and clearly better than historical data of monochemotherapy or conventional EGFR inhibitors. The greatest benefit was noted in patients without brain metastases, who showed durable effects with mDoT 14.8 months, while intracranial activity was limited. These findings may assist therapeutic considerations, inasmuch as results from the EXCLAIM cohort-3 dedicated to brain lesions are not available yet.
