Alpha-lactalbumin (α-LA) and binding of zinc cations to protein were studied. Molecular characteristics of protein was determined by MALDI-TOF/MS and electrophoresis SDS-PAGE, and also, for complexes, it was determined by spectroscopic techniques (ATR-FT-IR and Raman) and microscopic techniques (SEM along with an EDX detector and also TEM). The pH dependence of zeta potential of α-LA was determined in saline solution. The zinc binding to the protein mechanism was investigated; zinc binding to protein kinetics, the molecular modeling by the DFT method, and electron microscopy (SEM and TEM) for microstructure observation were performed. The experiments performed indicate a quick binding process (equilibrium takes place after 2 min of incubation) which occurs onto the surface of α-LA. Zinc cations change the conformation of the protein and create spherical particles from the morphological point of view. DFT studies indicate the participation of acidic functional groups of the protein (aspartic acid and glutamic acid residues), and these have a decisive influence on the interaction with zinc cations. Application studies of general toxicity and cytotoxicity and bioavailability were conducted.

A method was developed to determine 23 biologically active compounds, including 13 flavonoids and 10 phenolic acids, in various morphological parts (flowers, leaves, stems, seeds, and roots I and III cut) of the lucerne plant using a simple, fast high performance liquid chromatography with tandem mass spectrometry detection. The separation was achieved using a pentafluorophenyl stationary phase and a gradient elution with 0.1% formic acid in water and acetonitrile. The calibration curves exhibited good linearity (r2 value from 1.0 to 0.9925). The limit of detection (LOD) and limit of quantification (LOQ) for 23 compounds range from 0.01-50 ng/mL and 0.033-165 ng/mL, respectively. The main flavonoids found in the extracts were apigenin (765.96 ± 29.08 µg/g), luteolin (545.21 ± 14.64 µg/g), quercetin (169.90 ± 16.69 µg/g), and esculetin (130.99 ± 6.72 µg/g). Salicylic acid (2960.89 ± 11.00 µg/g) and 4-hydroxybenzoic acid (1561.42 ± 38.62 µg/g) were the most abundant in the analyzed extracts. The roots of the third cut and the stems were found to be the richest in polyphenolic compounds. These findings suggest that different morphological parts of lucerne could be used as food additives or supplement components due to their nutritional properties.

The purpose of this study was to review the possibility of using supercritical CO2 as a green and sustainable technology for microbial inactivation of raw material for further application in the food industry. The history of the development of supercritical CO2 microbial inactivation has been widely described in this article. The fundamental scientific part of the process like mechanism of bactericidal action of CO2 or inactivation of key enzymes were characterized in detail. In summary, this study provides an overview of the latest literature on the use of supercritical carbon dioxide in microbial inactivation of food raw materials and products.

The presence of certain microorganisms in dairy products or silage is highly desirable. Among them are probiotic strains of lactic acid bacteria (LAB), which show many beneficial features, including antimicrobial properties that support the development of beneficial microflora; in addition, owing to their biochemical activity, they influence the nutritional, dietary, and organoleptic properties of food products. Before being placed on the market, each strain requires separate testing to determine its probiotic properties and effectiveness. The aim of this study was to isolate LAB strains from a pickled beetroot sample that could be used in the dairy industry and with the potential to be considered as a probiotic in the future. A novel synthetic method for enantioselective C2-functionalization of 3-hydroxychromenones promoted by N-heterocyclic carbenes via the formation of α,β-unsaturated acyl azolium intermediates, which occurs with Coates–Claisen rearrangement is established. This synthetic strategy enabled the rapid assembly of enantiomerically enriched δ-hydroxychromenone-derived esters/amides under mild conditions with good to excellent yields and broad substrate scope.

The aim of the study was to investigate the process of Fe3+ binding to bLTF. Moreover, the physicochemical characterization of the respective supersaturated complex was studied. The knowledge should be important for the description of processes that may take place in dairy products fortified with iron. Additionally, the synthesized complex can be utilized as a dietary supplement for the treatment of iron deficiency anemia (IDA). Finally, it was shown that formation of supersaturated iron-protein structures which include LTF often accompanies development of neurodegenerative diseases such as Alzheimer or Parkinson. Thus, the study can reveal some aspects of its pathogenesis process. The methodology of the investigation comprised the utilization of batch sorption study and applying Freundlich and Langmuir models. The complex also was characterized by numerous techniques: spectrometric (ICP-MS), spectroscopic (UV–Vis, ATR-FTIR), electron microscopy (TEM–EDX), SDS-PAGE. Based on obtained results the potential mechanisms of iron interaction with protein were described. Moreover, the molecular docking was applied to visualize possible metal binding sites. The respective complex contains ≈ 33.0 mg/g of iron which is nearly 50 Fe3+ per one protein molecule. The cytotoxicity of the obtained complex was evaluated by MTT reduction and LDH release assays on Caco-2 and nL929 cell lines.

In the face of the growing demand for functional food, the search for new sources of lactic acid bacteria (LAB) becomes a priority. In our research, we used multiplied culture conditions followed by identification via the matrix-assisted laser desorption ionization-time of flight mass spectrometry for seeking LAB strains in plant- and animal-derived sources. Furthermore, the selected LAB isolates were examined for their proteolytic activity as well as antimicrobial action against different bacterial pathogens. The applied method appeared to be useful tool for searching LAB strains within different types of the biological matrices. The best source of the LABs was from calf. Comparing properties of the two selected LABs, those isolated from calf demonstrated the greatest proteolytic and antibacterial properties suggesting that gastrointestinal microbiota are the most valuable LAB source. Nevertheless, second selected strain derived from pickled cucumber juice may be also treated as a promising source of potential probiotic strains.

Matrix-assisted laser desorption/ionization - imaging mass spectrometry is an alternative tool, which can be implemented in order to obtain and visualize the "omic" signature of tissue samples. Its application to clinical study enables simultaneous imaging-based morphological observations and mass spectrometry analysis. Application of fully informative material like tissue allows obtaining the complex and unique profile of analyzed samples. This knowledge leads to diagnosing disease, studying the mechanism of cancer development, selecting the potential biomarkers as well as correlating obtained images with prognosis. Nevertheless, it is worth noticing that this method is found to be objective but the result of the analysis is mainly influenced by the sample preparation protocol, including the collection of biological material, its preservation, and processing. However, the application of this approach requires a special sample preparation procedure. The main goal of the study is to present the current knowledge on the clinical application of matrix-assisted laser desorption/ionization with imaging mass spectrometry in cancer research, with particular emphasis on the sample preparation step. For this purpose, several protocols based on cryosections and formalin-fixed paraffin-embedded tissue were compiled and compared, taking into account the measured metabolites of potential diagnostic importance for a given type of cancer.

A novel synthetic method for enantioselective C2-functionalization of 3-hydroxychromenones promoted by N-heterocyclic carbenes

A study of the mechanism responsible for the zearalenone (ZEA) neutralization by lactic acid bacteria Lactococcus lactis 56 and L929 cell line was carried out by determination of the kinetics of the binding process. In the case of prokaryotic cells the biosorption process was non-linear and three steps were identified. The maximum efficiency of zearalenone binding to L. lactis was almost 30% and no metabolites were observed. In turn, for eukaryotic cells only two steps of the binding process were differentiated, and the efficiency of zearalenone binding was 53.99%. Furthermore, L929 cell line metabolizes zearalenone to α-ZOL and β-ZOL. Additionally, Fourier transform infrared spectroscopy (FTIR) was used for description of the structural changes at the protein and lipid level, while Matrix Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry (MALDI-TOF-MS) was applied to detect changes at the molecular level.

Background A retrospective analysis was performed to investigate the survival outcomes in pediatric acute lymphoblastic leukemia (ALL) based on time period. We hypothesized that improvement has been obtained with the time-dependent therapeutic era and rise in the gross domestic product (GDP) and Human Development Index (HDI). Materials and methods Data from 710 children who were treated for ALL between 1958 and 2018 at a single pediatric center were analyzed for probability of 5-year overall survival (pOS), event-free survival (pEFS) and relapse risk (pRR). Time periods were defined by the treatment protocols used in seven consecutive therapeutic eras. Results Over the 60-year period analyzed, pOS increased from 1.2% to 90.7%, pEFS from 1.2% to 86.6%, and pRR decreased from 98.8% to 9.9% for patients treated in the past decade. Risk of mortality for patients who received chemotherapy and hematopoietic cell transplant was reduced to 9.9% in the recent era, however, no statistically significant survival difference was found between patients treated with stem cell transplant and those not. Conclusion The therapeutic era, related to improved GDP and HDI, was a statistically significant predictor of increased OS from ALL.

The aim of this study was to evaluate chemical and biological potential of garden cress (Lepidium sativum L.) to receive valuable plant extracts with potential application in pharmacy or food industry. Four techniques of extraction and three environmentally friendly solvents such as water, supercritical CO2 and ethanol have been tested. Biological activity and chemical profile were evaluated in obtained extracts. GC/MS analysis showed that SFE extract from dried sprouts of L. sativum was especially rich in such glucosinolate derivatives as benzyl cyanide and benzyl thiocyanate. However, the extract obtained from freeze-dried sprouts by SFE with addition of 96% ethanol as co-solvent was especially rich in flavonoids and simultaneously exhibited the best antimicrobial activity. Comparison of MALDI-TOF-MS spectra of all obtained extracts clearly indicates that both SFE and maceration with water are the most selective techniques of extraction due to the lowest level of interfering substances with high molecular masses.

A simple method for preparing AgNPs/clay nanocomposites using an adsorption process without any reducing agent was developed in which saponite iron-rich clay was both the solid inorganic support and reducing agent. Silver adsorption by ion exchange of silver ions and saponite ferrous ions resulted in simultaneous silver reduction and silver nanoparticle formation. The maximum loading of silver was determined as 48 mg/g (4.8 mass %). Microscopy showed a homogeneous distribution of sphere-like silver nanoparticles which are composed from smaller crystallites in the form of twinned triangular prisms. The silver particle sizes ranged from 1 nm to 50 nm but predominantly between 8 and 10 nm. The optimum pH range for silver immobilization on saponite support was between 4 and 8. Characterization of the clay samples and synthesized AgNPs/saponite nanocomposites was performed by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), energy-dispersive X-ray spectroscopy (EDX), Fourier transform infrared spectroscopy (FTIR), porosimetry (low temperature nitrogen adsorption-desorption) and zeta potential measurements. The antibacterial activities of raw saponite and AgNPs/saponite nanocomposite samples were tested against clinical relevant Gram-positive Staphylococcus aureus, Staphylococcus epidermidis, and Gram-negative Escherichia coli, Pseudomonas aeruginosa and Proteus mirabilis bacteria by the well diffusion method.

Solidago gigantea Ait. (goldenrod) is widely spread in Poland and has a traditional usage as medicinal plant due to the content of bioactive compounds. The main aims of this research were to obtain the optimal conditions of supercritical fluid extraction (SFE) of S. gigantea with carbon dioxide as a solvent in the quarter-technical plant and to scale-up the process to half-technical, where both works in a high range of pressure - up to 100 MPa. Response surface methodology (RSM) was used to analyze and evaluate the influence of process parameters: pressure (P, MPa), temperature (T, K) and solvent flow rate (F, kg h−1) on output variables. The selected output variables were yield, total phenolics content, total chlorophylls content and lipids content. The optimal parameters obtained at quarter-technical plant, for a chosen response – yield, were: T = 353.15 K (80 °C), P = 72.31 MPa and F = 6.97 kg h−1. Scale-up extractions were conducted according to various criteria at half-technical plant under optimal conditions. Extraction with simultaneously constant ratio of the solvent flow rate to the mass of the feedstock (Q CO2/M, kg h−1 kg−1) and a constant ratio of solvent mass to mass of the batch (S CO2/M, kg kg−1) ended with the best results: mass of obtained extract was 0.311 kg (7.17% DM – dry matter). Analysis of biological properties of the obtained product showed that it exhibits antibacterial activity and very low level of cytotoxicity and therefore, it can be safely used in pharmaceutical and cosmetics formulations.

The article describes a possible application of Sc-CO2 extraction for isolation of polyphenols from germinated lupin seeds. Efficiency of germination duration was valorized, and the obtained results suggest that six-day-long germination process provides samples with the highest polyphenolic yield. Optimization of extraction parameters was done by using response surface methodology (RSM). Extracts obtained under optimal extraction conditions (73 °C, 147 bar, 16.1% of co-solvent) were characterized in terms of their chemical composition and biological activity. The yield of extraction was 3.25% while total phenolic content was 14.56 mg GAE/g. HPLC-ESI-MS/MS analysis confirmed the presence of different polyphenolic compounds in which apigenin and fisetin were proven to be dominant compounds. The GC–MS analysis revealed that the obtained extract is also rich in fatty acids, phytosterols and lupeol. Further tests determined the antioxidant, antiradical, antimicrobial and cytotoxic potential of samples. Fluorescence investigation was used to visualize the effect of Lupinus luteus extract on viability of bacterial cells. In addition, it has been shown that the extract tested in anti-bacterial concentrations does not exhibit cytotoxicity.

Mycotoxins are toxic metabolites produced by mold fungi, which commonly contaminate cereal crops. These compounds include zearalenone (ZEA), which may disturb the proper functioning of the endocrine system in mammals. The metabolism of ZEA plays a key role in its toxic properties. The type and amount of produced metabolites may contribute to both the reduction and increase in its pathogenic effect. Therefore, it is extremely important to investigate the possible pathways of zearalenone metabolism. The electrochemical approach may prove alternatives for the in vitro and in vivo methods. For this reason, in this study the electrochemical, theoretical and in vitro assays were applied to determine the possible ZEA metabolism mechanism. Electrochemical processes were conducted in the EC/ESI-MS system. The application of HPLC-MS/MS allowed to characterize the obtained electrochemical products specific for phase I and II. Appropriate conditions for redox identification processes were optimized with regard to such parameters as the potential value, mobile phase and working electrode. In addition, the data obtained by instrumental techniques for ZEA metabolism were compared with those obtained by in vitro methods for HepG2 cell lines. Furthermore, results from quantum mechanical calculations allowed to explain the mechanism and the conformational stability of the reduction products.

The aim of this study was to isolate LAB strains from a pickled beetroot sample that could be used in the dairy industry and with the potential to be considered as a probiotic in the future. Two strains identified using the MALDI technique were selected—Lactococcus lactis and Weissella cibaria. The optimal growth conditions of the strains were determined, and their proteolytic properties were assessed with the use of the o-PA reagent and spectrophotometry. The lipid profile was analyzed using the SALDI (surface-assisted laser desorption/ionization) technique and silver nanoparticles. High-performance liquid chromatography was used to assess the ability of the strains to synthesize beneficial metabolites, such as B vitamins (B2, B3, and B9) or lactic acid, and gas chromatography was used to analyze the substances responsible for organoleptic properties. Moreover, the ability to inhibit the growth of pathogenic strains was also tested in the selected strains. Both tested strains demonstrated the desired properties of starter cultures for future use in functional food production, showing that fermented plant products can serve as valuable potential probiotic sources.

A facile approach for the synthesis of a silver nanostructured layer for application in surface-assisted laser desorption/ionization mass spectrometry of low-molecular-weight biomolecules was developed using electrochemical deposition. The deposition was carried out using the following silver salts: trifluoroacetate, acetate and nitrate, varying the voltage and time. The plate based on trifluoroacetate at 10 V for 15 min showed intense SALDI-MS responses for standards of various classes of compounds: fatty acids, cyclitols, saccharides and lipids at a concentration of 1 nmol/spot, with values of the signal-to-noise ratio ≥50. The values of the limit of detection were 0.71 µM for adonitol, 2.08 µM for glucose and 0.39 µM for palmitic acid per spot. SEM analysis of the plate showed anisotropic flower-like microstructures with nanostructures on their surface. The reduced chemical background in the low-mass region can probably be explained by the absence of stabilizers and reducing agents during the synthesis. The plate synthesized with the developed approach showed potential for future use in the analysis of low-molecular-weight compounds of biological relevance. The absence of the need for the utilization of sophisticated equipment and the synthesis time (10 min) may benefit large-scale applications of the layer for the detection of various types of small biomolecules.

Butter is an important source of essential fatty acids, lipid-soluble vitamins, and antioxidants in the diet. However, this study showed that the presence of the Lacticaseibacillus paracasei strain has a great influence on the fatty acid profile as well as provitamin D3 and vitamin D3 content in the cream—the raw material from which the butter is obtained. The addition of this lactic acid bacteria enriches the cream in 9-hexadecenoic acid, oleic acid, octadeca-9,12-dienoic acid, and conjugated linoleic acid, which exhibit antimutagenic and anticarcinogenic properties. Moreover, a higher level of monounsaturated fatty acids can extend the shelf life of butter in the future. In the present work, we observed that the presence of lactic acid bacteria contributed to an increase in the level of provitamin D after 6 h of incubation and an increase in the levels of vitamin D3 after 24 and 48 h. Fatty acid profiles and the content of vitamins were largely dependent on the presence of light and mixing, which are probably associated with the status of lipid peroxidation.

The aim of this study was to develop an effective Medicago sativa extraction method using enzyme-assisted supercritical fluid extraction (EA-SFE). The response surface methodology (RSM) based on Box-Behnken design was chosen to optimize the parameters of extraction process, i.e., extraction temperature, pressure and percentage of co-solvent for total phenolics content and antioxidants. Commercial enzyme preparation kemzyme was applied to M. sativa leaves. The extraction of antioxidant phenolics in optimal conditions from the enzymatically hydrolyzed M. sativa leaves was carried out by supercritical fluid extraction (SFE) with 96% ethanol as a co-solvent. Optimal extraction parameters for TPC and the antioxidant activity were: the temperature of 68 °C, the pressure of 205 bar and 15.5% of the co-solvent addition. For the sake of comparison, conventional extraction was performed. The obtained results showed that the use of the commercial enzyme formulation allowed the content of phenolic compounds in the leaves extract increase as compared to the control. HPLC-MS/MS characterization revealed that extraction of enzyme digested material was much more efficient (142.69 ± 5.11 μg/g) than extraction of non-digested material (94.17 ± 4.80 μg/g) or the use of conventional method (57.42 ± 2.25 μg/g). Our work offers an effective enzyme-assisted supercritical fluid extraction protocol for the enhanced release of biological active compounds from plant cells.

Background/aim Immune recovery is a key factor in the management of patients after allogeneic hematopoietic stem cell transplantation (allo-HSCT). This study analyzed the factors contributing to immune reconstitution after allo-HSCT. Patients and methods Overall, 65 children with malignant or non-malignant diseases were included in multivariate analyses. Results The following factors contributed to a faster immune recovery: peripheral blood as a stem cell source and reactivation of CMV infection for CD3+ and CD4+ lymphocyte subpopulations; reactivation of CMV infection for CD8+ subset; donor EBV-IgG+ and no EBV reactivation for CD19 lymphocytes; recipient age below 10 years and peripheral blood as a stem cell source for NK cells. For CD2 and CD4/CD8 ratio no factor was significant in multivariate analysis. Conclusion Patients receiving a graft from an EBV-IgG-positive donor and not having early EBV post-transplant viremia show faster recovery of the B-cells, while patients with early CMV-DNA-emia have a better re-establishment of T-cell subsets.

In this review, recent advances in the methods of pre-treatment of plant material for the extraction of secondary metabolites with high biological activity are presented. The correct preparation of the material for extraction is as important as the selection of the extraction method. This step should prevent the degradation of bioactive compounds as well as the development of fungi and bacteria. Currently, the methods of preparation are expected to modify the particles of the plant material in such a way that will contribute to the release of bioactive compounds loosely bonded to cell wall polymers. This review presents a wide range of methods of preparing plant material, including drying, freeze-drying, convection drying, microwave vacuum drying, enzymatic processes, and fermentation. The influence of the particular methods on the structure of plant material particles, the level of preserved bioactive compounds, and the possibility of their release during the extraction were highlighted. The plant material pre-treatment techniques used were discussed with respect to the amount of compounds released during extraction as well their application in various industries interested in products with a high content of biologically active compounds, such as the pharmaceutical, cosmetics, and food industries.

In this paper, a study of the cytotoxicity of bare and functionalized zinc oxide nanoparticles (ZnO NPs) is presented. The functionalized ZnO NPs were obtained by various types of biological methods including microbiological (intra- and extracellular with Lactobacillus paracasei strain), phytochemical (Medicago sativa plant extract) and biochemical (ovalbumin from egg white protein) synthesis. As a control, the bare ZnO NPs gained by chemical synthesis (commercially available) were tested. The cytotoxicity was measured through the use of (3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) dye as well as lactate dehydrogenase (LDH) assays against murine fibroblast L929 and Caco-2 cell lines. As a complementary method, scanning electron microscopy (SEM) was performed to assess the morphology of the tested cells after treatment with ZnO NPs. The microscopic data confirmed the occurrence of apoptotic blebbing and loss of membrane permeability after the administration of all ZnO NPs. The reactive oxygen species (ROS) concentration during the cell lines’ exposure to ZnO NPs was measured fluorometrically. Additionally, the photocatalytic degradation of methylene blue (MB) dye in the different light conditions, as well as the antioxidant activity of bare and functionalized ZnO NPs, is also reported. The addition of all types of tested ZnO NPs to methylene blue resulted in enhanced rates of photo-degradation in the presence of both types of irradiation, but the application of UV light resulted in higher photocatalytic activity of ZnO NPs. Furthermore, bare (chemically synthetized) NPs have been recognized as the strongest photocatalysts. In the context of the obtained results, a mechanism underlying the toxicity of bio-ZnO NPs, including (a) the generation of reactive oxygen species and (b) the induction of apoptosis, is proposed.

Medicago sativa L. (M. sativa) is a source of many valuable secondary metabolites. Extraction yield and the concentration of phenolics, flavonoids, and saponins, as well as antioxidant potential were determined in extracts from different parts of M. sativa obtained using extraction methods such as maceration, ultrasound-assisted extraction (UAE), accelerated solvent extraction (ASE), and supercritical fluid extraction (SFE). The concentrations of the listed groups of compounds were spectrophotometrically determined and confirmed by HPLC-MS. The results showed that ASE of flowers with 70% ethanol (EtOH) provided the highest yield of extraction (47.5 ± 4.0%), whereas the lowest yield was obtained in stems (4.0 ± 0.2%). The 70% EtOH extract from flowers showed the highest phenolic content [48.4 ± 4.6 mg gallic acid equivalents/g dry matter (DM)], as well as the highest antioxidant activity. The highest total flavonoid content (139.0 ± 7.1 mg rutin equivalents/g DM) was observed in the extract from leaves obtained through SFE. This extract was also especially rich in saponins [622.2 ± 30.3 mg oleanolic acid equivalents (OAE)/g DM]. However, the lowest compound content was observed in maceration extracts from stems (54.6 ± 27.0 mg OAE/g DM). The results suggest that EtOH extracts from alfalfa flowers and SFE extracts from M. satvia leaves, especially, may serve as potential sources of natural antioxidants for nutraceuticals, food additives, and cosmetic ingredients.

Zinc oxide (ZnO), as a material with attractive properties, has attracted great interest worldwide, particularly owing to the implementation of the synthesis of nano-sized particles. High luminescent efficiency, a wide band gap (3.36eV), and a large exciton binding energy (60meV) has triggered intense research on the production of nanoparticles using different synthesis methods and on their future applications. ZnO nanomaterials can be used in industry as nano-optical and nano-electrical devices, in food packaging and in medicine as antimicrobial and antitumor agents. The increasing focus on nano zinc oxide resulted in the invention and development of methods of nanoparticles synthesis. Recently, various approaches including physical, chemical and biological ("green chemistry") have been used to prepare ZnO nanocomposites with different morphologies. The obtained nanoparticles can be characterized with a broad range of analytical methods including dynamic light scattering (DLS), electron microscopy (TEM, SEM), UV-VIS spectroscopy, X-ray diffraction (XRD) or inductively coupled plasma with mass spectrometry (ICP-MS). With these it is possible to obtain information concerning the size, shape and optical properties of nanoparticles. ZnO NPs exhibit attractive antimicrobial properties against bacteria (Gram-positive and Gram-negative) and fungi. Zinc oxide nanocomposites show also selective toxicity toward normal and cancerous cells, which is explained by reactive oxygen formation (ROS). Yet despite the potentially interesting antitumor activity of ZnO nanoparticles, it has been proven that they can be also cytotoxic and genotoxic for multiple types of human cells (i.e. neuronal or epithelial cells). This paper reviews the methods of synthesizing zinc oxide nanocomposites as well as their characteristics, antimicrobial activity and cytotoxicity against normal and tumor cells.

The aim of the study was to neutralize zearalenone by lactic acid bacteria (LAB) such as Lactococcus lactis and Bifidobacterium sp. and investigate the mechanism of zearalenone (ZEA) binding. Neutralization of ZEA by LAB was confirmed by identification of binding kinetics and spectroscopic studies such as Fourier transform infrared spectroscopy (FT-IR) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). The obtained results showed that the kinetic process of zearalenone binding to L. lactis is not homogeneous but is expressed with an initial rapid stage with about 90% of ZEA biosorption and with a much slower second step. In case of Bifidobacterium sp., the neutralization process is homogeneous; the main stage can be described with about 88% of ZEA biosorption. MALDI-TOF-MS measurements and FTIR analysis confirmed the uptake of zearalenone molecules by bacterial species. Moreover, the assessment of dead and live lactic acid bacteria cells after zearalenone treatment was performed using fluorescence microscopy. The resistance of pathogenic bacteria to antibiotics has become a serious problem. The emphasis is placed on the development of new, effective antimicrobial strategies. One of them is the use of AgNPs in association with antibiotic drugs. The aim of this study was to obtain silver nanoparticles functionalized with ampicillin and to investigate the mechanism of binding antibiotics to nanoparticle using high-performance liquid chromatography approach. To confirm the occurrence of silver nanoparticles functionalization, FTIR, MALDI-TOF MS, and DLS analysis and zeta potential measurements were performed. Moreover we assessed the antibacterial activity of biologically synthesized nanoparticles functionalized with ampicillin against a range of gram (+) and gram (-) bacteria strains such as Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, Staphylococcus epidermidis, and Escherichia coli.

Medicago sativa L. (lucerne or alfalfa) is a species which is generally regarded as a cheap source of valuable protein not only for animal fodder but also for nourishment provided in the poorest and developing countries. Moreover, this plant has a long tradition of use in folk medicine. Numerous studies indicate that besides protein, M. sativa synthesizes a variety of secondary metabolites. Among secondary metabolite classes produced by alfalfa, the saponins and flavonoids are of most interest and well characterized. In the current review, we have summarized their chemical structure and method of identification. The potential of alfalfa extracts in medicine and pharmacology has been highlighted. Furthermore, new ways of utilizing flavonoids as well as saponins in agriculture and horticulture are discussed. Collected data can be the starting point and inspiration for scientists of various specialties for future research on this plant. This review draws attention to the necessity of developing modern analytical tools for identification and quantification of individual M. sativa phytochemicals.

The aim of this research was to select parameters for supercritical extraction with CO2 of Medicago sativa L., considered as functional food, in quarter-technical plant, providing the highest concentration of bioactive polar constituents and simultaneously maintaining the highest efficiency of the process. For the purpose of optimization, mathematical statistics was used. Qualitative analysis of products was performed with supercritical fluid chromatography (SFC). The SFC analysis revealed a proper separation of flavonoids and phenolics acids for dedicated TFC and TPC optimal parameters. The obtained results have proved that it is a possibility to extract polar compounds with non-polar solvent under higher values of pressure and temperature and to enrich product with desired group of bioactive compounds with proper optimization. The proposed extraction technique allows to obtain on an industrial scale, using an environmentally friendly solvent, a preparation rich in biologically active nutrients that can be implemented in the cosmetics, pharmaceutical and food industries.

Background/aim Neurofibromatosis type 1 (NF1) is characterized by the occurrence of multisystem tumors, among which the most characteristic are optic pathway gliomas (OPGs) and plexiform neurofibromas (PNFs). With the development of new anticancer drugs targeting the immune system, it is important to examine the immunological status of patients with NF1. Furthermore, the immune system has been suggested as a probable modulator of NF1-associated phenotypes. The objective of this study was the analysis of lymphocyte subset populations with respect to the presence of PNFs and OPGs. Patients and methods Fifty-three patients with NF1 diagnosed with OPG/PNF were analyzed for lymphocyte subpopulations. Results Significantly lower levels of B-cells, T-cells and natural killer (NK) cells were observed in the group of patients with PNFs compared to those with OPG. Conclusion Our observation may indicate a correlation between weakened functioning of the immune system and the formation of PNFs.

Saponins in plant extracts were indirectly determined by estimation of the content of sapogenins. The first step of determination is extraction with high efficiency. One conventional extraction technique (maceration) and two modern ones (accelerated solvent extraction and supercritical fluid extraction) were compared. Methanol and ethanol were used as solvents or co-solvents. The results were supported by statistical analysis. Saponins were extracted from leaves, roots, and sprouts of Medicago sativa. Acid hydrolysis, purification, and determination by high-performance liquid chromatography with evaporative light scattering detector were used. The content of sapogenins was the highest in the roots. Smaller amounts of sapogenins were found in sprouts and the smallest ones in leaves. The main ingredient was medicagenic acid with mean concentration of 621.8 µg/g in roots, 456.7 µg/g in sprouts, and 471.3 µg/g in leaf extract. The highest content of sapogenins in extract was obtained after maceration with methanol; however, this method is nonselective in relation to biologically active compounds. Due to the possibility of using the obtained extracts with sapogenins in the cosmetic or pharmaceutical industry, the selection of extraction techniques and solvents is a very important aspect. Additionally, the chosen technique should be considered eco-friendly and consistent with the assumptions of "green chemistry."

This work reports the effect of silver bionanoparticles (Bio(AgNPs) synthesized by Actinobacteria CGG 11n on selected Gram (+) and Gram (-) bacteria. Flow cytometry, classical antibiogram method and fluorescent microscopy approach was used for evaluation of antimicrobial activity of Bio(AgNPs) and their combination with antibiotics. Furthermore, the performed research specified the capacity of flow cytometry method as an alternative to the standard ones and as a complementary method to electromigration techniques. The study showed antibacterial activity of both BioAgNPs and the combination of antibiotics/BioAgNPs against all the tested bacteria strains in comparison with a diffusion, dilution and bioautographic methods. The synergistic effect of antibiotics/BioAgNPs combination (e.g. kanamycin, ampicillin, neomycin and streptomycin) was found to be more notable against Pseudomonas aeruginosa representing a prototype of multi-drug resistant "superbugs" for which effective therapeutic options are very limited.

Although silver nanoparticles are the most widespread product of nanotechnology, the mechanisms underlying AgNP microbial toxicity remain the subject of intense debate. In this study, Bacillus subtilis has been used as model organism to elucidate the molecular interactions between this class of bacteria and different forms of silver such as nanoparticles, nanoparticles functionalized with tetracycline and silver ions. For this purpose, we carried out transmission electron microscopy and MALDI-TOF MS analysis of cells treated with silver nanoparticles (AgNPs, AgNPs functionalized with tetracycline, combination of AgNPs with tetracycline) and silver ions as well as we measured the level of reactive oxygen species. The data demonstrate that B. subtilis exhibits high resistance to silver nanoparticles and this phenomenon is associated with following processes: (I) initiation of endospore formation, (II) reduction of free Ag+ released from nanoparticles and (III) modification of the AgNPs surface. However, high silver ions concentration appeared to be very toxic to studied strain of bacterium. MALDI-TOF MS analysis revealed that the spectra of B. subtilis cells treated with silver ions are significantly different from spectra of control cells and cell treated with AgNPs and antibiotic which can suggest that silver ions in the highest degree modify bacterial components.

Saponins are widespread secondary metabolites with various beneficial properties: fungicidal, antibacterial, antiviral, and anticancer. Alfalfa saponin molecules contain mainly: medicagenic acid, hederagenin, bayogenin, and soyasapogenol B. Structural diversity of saponins makes their determination in Medicago sativa extracts very difficult. The most popular determination technique is high-performance liquid chromatography applied with evaporative light scattering detection. Qualitative and quantitative analysis of sapogenins from Medicago sativa by high-performance liquid chromatography with evaporative light scattering detection required hydrolysis and purification of extracts obtained by supercritical fluid extraction. Hydrolysis of saponins with concentrated hydrochloric acid provided high concentration of medicagenic acid. In the purification process, satisfactory results were obtained for solid-phase extraction using octadecyl. Recoveries were from 71 to 99% with a standard deviation from 2 to 8. Hydrolysis with concentrated hydrochloric acid was the only method that allowed identification of all four analyzed sapogenins. Moreover, it is characterized by a short time of preparation, simplicity of execution, a small amount of the sample and solvents. The hydrolysis and purification methods coupled with high-performance liquid chromatography and evaporative light scattering detection can be successfully used for qualitative and quantitative analysis of the main saponins present in Medicago sativa plant extracts obtained by supercritical fluid extraction.

The effect of a different pH on Saccharomyces cerevisiae cells modified with calcium ions was investigated by the capillary zone electrophoresis technique. For the identification of the wild strain of S. cerevisiae, the ribosomal nucleic acid sequencing and internal transcribed spacer sequencing as well as spectrometric approach were applied. The potentiometric titration and Fourier transform infrared spectroscopy have shown the occurrence of active functional groups such as carboxyl, amine/hydroxyl, phosphate/hydrogen phosphate groups on the surface of native yeast cells. Moreover, the spectroscopy study in a medium infrared range was carried out to identify the functional groups of yeast cells that participate in calcium ions binding interaction. Furthermore, the microscopic and spectrometric analysis shows that the pH value of the calcium ions solution has a significant effect on the intensity yeast cells clumping. Additionally, the impact of yeast cell clumping on the electrophoretic behaviours was examined. The modification of surface functional groups by calcium ions significantly affected the efficiency of electrophoretic separation. However, these changes did not affect the accuracy of S. cerevisiae identification by MALDI equipment with BioTyper platform. These results form the analytical solution for coupling of electrophoresis and MALDI-TOF MS technique.

Medicago sativa L. (M. sativa) is a source of many valuable secondary metabolites. Extraction yield and the concentration of phenolics, flavonoids, and saponins, as well as antioxidant potential were determined in extracts from different parts of M. sativa obtained using extraction methods such as maceration, ultrasound-assisted extraction (UAE), accelerated solvent extraction (ASE), and supercritical fluid extraction (SFE). The concentrations of the listed groups of compounds were spectrophotometrically determined and confirmed by HPLC-MS. The results showed that ASE of flowers with 70% ethanol (EtOH) provided the highest yield of extraction (47.5 ± 4.0%), whereas the lowest yield was obtained in stems (4.0 ± 0.2%). The 70% EtOH extract from flowers showed the highest phenolic content [48.4 ± 4.6 mg gallic acid equivalents/g dry matter (DM)], as well as the highest antioxidant activity. The highest total flavonoid content (139.0 ± 7.1 mg rutin equivalents/g DM) was observed in the extract from leaves obtained through SFE. This extract was also especially rich in saponins [622.2 ± 30.3 mg oleanolic acid equivalents (OAE)/g DM]. However, the lowest compound content was observed in maceration extracts from stems (54.6 ± 27.0 mg OAE/g DM). The results suggest that EtOH extracts from alfalfa flowers and SFE extracts from M. satvia leaves, especially, may serve as potential sources of natural antioxidants for nutraceuticals, food additives, and cosmetic ingredients.

There has been growing interest in the application of supercritical solvents over the last several years, many of the applications industrial in nature. The purpose of plant material extraction is to obtain large amounts of extract rich in the desired active compounds in a time-sensitive and cost-effective manner. The productivity and profitability of a supercritical fluid extraction (SFE) process largely depends on the selection of process parameters, which are elaborated upon in this paper. Carbon dioxide (CO2) is the most desirable solvent for the supercritical extraction of natural products. Its near-ambient critical temperature makes it suitable for the extraction of thermolabile components without degradation. A new approach has been adopted for SFE in which the solubility of nonpolar supercritical CO2 can be enhanced by the addition of small amounts of cosolvent.

New (1R)-camphor-derived triazolium salts, incorporating a spirocyclic system, are described. Benzoin condensation of aromatic aldehydes, mediated by these salts affords α-hydroxyketones in good yields and moderate enantiomeric excesses.

Novel chiral N-heterocyclic carbenes derived from (−)-β-pinene have been synthesized and used as efficient catalysts for the intramolecular Stetter reaction. Mono-, di-, and trisubstituted 4-chromanone derivatives have been obtained almost quantitatively and with high enantioselectivity up to 99:1 er.

The main aim of this study was to investigate the possibility of using Solidago gigantea Ait., as a source of fatty acids. S. gigantea, by contrast to rapeseed or sunflower seeds, is not commonly used plant material to obtain these compounds. For that purpose, the proper optimization of supercritical carbon dioxide extraction (SFE-scCO2) of S. gigantea at quarter-technical plant was investigated to provide the highest concentration of FAME. SFE-scCO2 is a green technology and it is very efficient technique for the extraction of non-polar compounds. Response surface methodology (RSM) was used to analyze and evaluate the influence of chosen input variables: pressure (P, MPa), temperature (T, K) and solvent flow rate (F, kg h−1) on FAME content in products. The output variable in the obtained extract was determined by GC–MS. The optimal parameters obtained at quarter-technical plant were T =313.79 K (40.64 °C), P = 22.22 MPa and F =6.46 kg h−1. Finally, the scale-up procedure of optimal conditions from the quarter-technical to half-technical plant was conducted to substantiate the correctness of the optimization process and to facilitate the transfer of the process to industry scale. Scale-up procedure of optimal conditions was conducted according to various criteria and it ended successfully. Extraction with constant ratio of solvent mass to mass of the batch (S CO2/M, kg kg−1) with the final result with confidence interval: 273.22 mg g−1 DM proved the correctness of the optimization proces. The obtained amount of FAME concentrations allowed to conclude that S. gigantea can be considered as a source of fatty acids. Finally, obtained optimal parameters can be implemented in an industry plant.
