This work presents a suite of measurement techniques for characterizing the dielectric loss tangent across a wide frequency range from $\sim$1 GHz to 150 GHz using the same test chip. In the first method, we fit data from a microwave resonator at different temperatures to a model that captures the two-level system (TLS) response to extract and characterize both the real and imaginary components of the dielectric loss. The inverse of the internal quality factor is a second measure of the overall loss of the resonator, where TLS loss through the dielectric material is typically the dominant source. The third technique is a differential optical measurement at 150 GHz. The same antenna feeds two microstrip lines with different lengths that terminate in two microwave kinetic inductance detectors (MKIDs). The difference in the detector response is used to estimate the loss per unit length of the microstrip line. Our results suggest a larger loss for SiN$_x$ at 150 GHz of ${\mathrm{\tan \delta\sim 4\times10^{-3}}}$ compared to ${\mathrm{2.0\times10^{-3}}}$ and ${\mathrm{\gtrsim 1\times10^{-3}}}$ measured at $\sim$1 GHz using the other two methods. {These measurement techniques can be applied to other dielectrics by adjusting the microstrip lengths to provide enough optical efficiency contrast and other mm/sub-mm frequency ranges by tuning the antenna and feedhorn accordingly.

Line Intensity Mapping (LIM) is a new observational technique that uses low-resolution observations of line emission to efficiently trace the large-scale structure of the Universe out to high redshift. Common mm/sub-mm emission lines are accessible from ground-based observatories, and the requirements on the detectors for LIM at mm-wavelengths are well matched to the capabilities of large-format arrays of superconducting sensors. We describe the development of an R = 300 on-chip superconducting filter-bank spectrometer covering the 120--180 GHz band optimized for future mm-LIM experiments, focusing on SPT-SLIM, a pathfinder LIM instrument for the South Pole Telescope. Radiation is coupled from the telescope optical system to the spectrometer chip via an array of feedhorn-coupled orthomode transducers. Superconducting microstrip transmission lines then carry the signal to an array of channelizing half-wavelength resonators, and the output of each spectral channel is sensed by a lumped element kinetic inductance detector (leKID). Key areas of development include incorporating new low-loss dielectrics to improve both the achievable spectral resolution and optical efficiency and development of a robust fabrication process to create a galvanic connection between ultra-pure superconducting thin-films to realize multi-material (hybrid) leKIDs. Experiments deploying large arrays of transition-edge sensors (TESs) often require a robust method to monitor gain variations with minimal loss of observing time. We propose a sensitive and non-intrusive method for monitoring variations in TES responsivity using small square waves applied to the TES bias. We construct an estimator for a TES's small-signal power response from its electrical response that is exact in the limit of strong electrothermal feedback. We discuss the application and validation of this method using flight data from SPIDER, a balloon-borne telescope that observes the polarization of the cosmic microwave background with more than 2000 TESs. Planar ortho-mode transducers (OMTs) are a commonly used method of coupling optical signals between waveguides and on-chip circuitry and detectors. While the ideal OMT-waveguide coupling requires minimal disturbance to the waveguide, when used for mm-wave applications the waveguide is typically constructed from two sections to allow the OMT probes to be inserted into the waveguide. This break in the waveguide is a source of signal leakage and can lead to loss of performance and increased experimental systematic errors. Here we report on the development of new OMT-to-waveguide coupling structures with the goal of reducing leakage at the detector wafer interface. The pixel to pixel optical leakage due to the gap between the coupling waveguide and the backshort is reduced by means of a protrusion that passes through the OMT membrane and electrically connects the two waveguide sections on either side of the wafer. High frequency electromagnetic simulations indicate that these protrusions are an effective method to reduce optical leakage in the gap by ~80% percent, with a ~60% filling factor, relative to an standard OMT coupling architecture. Prototype devices have been designed to characterize the performance of the new design using a relative measurement with varying filling factors. We have measured the quasiparticle generation-recombination (GR) noise in aluminium lumped element kinetic inductors with a wide range of detector volumes at various temperatures. The basic detector consists of meandering inductor and interdigitated capacitor fingers. The inductor volume is varied from 2 to 153 {\mu}m^{3} by changing the inductor width and length to maintain a constant inductance. We started with measuring the power spectrum density (PSD) of the detectors frequency noise which is a function of GR noise and we clearly observed the spectrum roll off at 10 kHz which corresponds to the quasiparticle lifetime. Using data from a temperature sweep of the resonator frequency we convert the frequency fluctuation to quasiparticle fluctuation and observe its strong dependence on detector volume: detectors with smaller volume display less quasiparticle noise amplitude. Meanwhile we observe a saturated quasiparticle density at low temperature from all detectors as the quasiparticle life time {\tau}qp approaches a constant value at low temperature.

The separation and optimization of noise components is critical to microwave-kinetic inductance detector (MKID) development. We analyze the effect of several changes to the lumped-element inductor and interdigitated capacitor geometry on the noise performance of a series of MKIDs intended for millimeter-wavelength experiments. We extract the contributions from two-level system noise in the dielectric layer, the generation-recombination noise intrinsic to the superconducting thin-film, and system white noise from each detector noise power spectrum and characterize how these noise components depend on detector geometry, material, and measurement conditions such as driving power and temperature. We observe a reduction in the amplitude of two-level system noise with both an elevated sample temperature and an increased gap between the fingers within the interdigitated capacitors for both aluminum and niobium detectors. We also verify the expected reduction of the generation-recombination noise and associated quasiparticle lifetime with reduced inductor volume. This study also iterates over different materials, including aluminum, niobium, and aluminum manganese, and compares the results with an underlying physical model.

Study question Is it possible to select spermatozoa using progesterone gradients? Does chemotaxis improve sperm selection compared to a classical density gradient or swim up? Summary answer Spermatozoa selected using progesterone gradients yield higher percentages of high quality spermatozoa with lower DNA fragmentation and better morphology compared to classic sperm selection. What is known already There are few studies which aim at improving sperm selection, even though it is known to be a vital parameter for IVF success. DNA integrity is fundamental for embryo development although it is currently underestimated. Progesterone has been suggested to be a major chemoattractant for spermatozoa in the fallopian tubes and therefore using gradients in a clinical setting may lead to a more physiologically improved selection method. Study design, size, duration 19 normo spermic patients (according to W.H.O. parameters) were selected. After standard semen analysis the sample was split in two halves. One half was treated using classical swim up and the remaining half was treated on a progesterone gradient. The treated samples were then tested for fragmentation, motility and morphology. Sperm motility was analyzed using SCA computer system. DNA fragmentation using the TUNEL assay. Participants/materials, setting, methods The patients enrolled in the study were all normospermic. The results were compared using ANOVA one way. Progesterone gradients was obtaining using an horizontal density gradients with added progesterone. Main results and the role of chance The percentages of rapid progressive spermatozoa were 12,89 ± 4,50%; 56,37 ± 13,61% and 29,84±12,52% in the untreated sample (US), treated sample (TS) and control group (C)respectively; p < 0,005. Normal sperm morphology rates were 3,474 ± 1,46; 5,500 ±1,57 and 3,684 ± 1,15 in US, TS and C respectively. Sperm fragmentation rates were 9,77 ± 9,012,50 ± 2,66 and 7,94 ± 6,68 in US, TS and C respectively. Limitations, reasons for caution The number of normospermic patients enrolled in the study was limited. Moreover, the sperm concentration obtained in this kind of selection is lower than in the conventional techniques and therefore may be harder to use in patients with a very low sperm count. Wider implications of the findings Our data shows a significant improvement in sperm selection using chemotaxis gradient compared to the control group by all the parameters considered in this study. Chemotaxis gradients may be useful in a clinical setting to improve spermatozoa selection, lowering the rates of sperm with damaged DNA potentially increasing embryo quality. Study question Does enhancing oxygen availability during dynamic in vitro culture of bovine ovarian cortical tissue (BOCT) improve follicle growth and health? Summary answer Enhancing oxygen availability during dynamic in vitro culture of BOCT in perifusion bioreactors (PB) does improve follicle health and yield to secondary follicles What is known already Oxygen availability has been demonstrated to represent a key factor in follicle health and growth during in vitro culture of bovine and human ovarian cortical tissue (HOCT) under static culture conditions. Disruption of solutes gradients and application of physiological fluid mechanical stress, through in vitro dynamic culture of HOCT in a newly designed perifusion bioreactor have been shown to further enhance follicle growth and health. As it shows striking similarities with human, bovine folliculogenesis is considered a valuable model to study follicle growth in vitro Study design, size, duration Bovine ovaries from animals aged 8-24 months were collected at slaughterhouse. In each experiment (n = 3), BOCT strips from the same ovary were cultured for 6 days in perifusion bioreactors (PB, dynamic culture) and conventional dishes (CD, static culture). Culture outcome in static culture was analysed and compared to two bioreactor configurations in which medium oxygenation was kept low by using a standard tube reservoir (StPB) or was enhanced by using a gas-permeable dish reservoir (PB+O2). Participants/materials, setting, methods Slices of BOCT 0.5mm thick were cut with a tissue slicer and chopped into 1x1mm strips. In each experiment, fresh (D0) and cultured tissue (groups of ten strips) were analyzed. Follicle stages and health were assessed by histology (hematoxylin-eosin staining). Follicle viability was estimated by labelling with live-dead far-red and propidium iodide followed by clearing before analysis at the confocal laser scanning microscope. Main results and the role of chance Overall, 2417 follicles were analyzed (histology, 1476; viability, 941). At day 0 most follicles were primordial (primordial, 88.7%; primary, 10.6%; secondary, 0.7%), and had good quality (grade 1-2, 92.2%; grade 3, 7.8%), and high viability (91.8%). The role of FSH on the progression of primordial to secondary follicles in ovarian organ culture is still debated. Study design, size, duration Bovine ovaries from animals aged 8-24 months were collected at a slaughterhouse. In each experiment (n = 3), BOCS of varying thickness collected from the same ovary were cultured with or without FSH in gas-permeable dishes for 10 and 15 days. Participants/materials, setting, methods Slices either 0.5mm or 1mm thick were cut with a tissue-slicer and were chopped into 1x1mm2 strips. BOCS were cultured for 10 or 15 days at the same tissue/medium volume ratio (groups of either five 1mm strips or ten 0.5mm strips in 5ml medium) with 0, 1, or 5ng/ml FSH. Follicle stages were assessed by histology. Follicle viability was estimated by labeling with live-dead far red and propidium iodide at the confocal laser scanning microscope. Main results and the role of chance Overall, 2314 follicles were analyzed (histology, 998; viability, 1316). At day 0 most follicles were primordial (primordial, 89.4%; primary, 8.7%; secondary, 1.9%), and had a high viability (94.69%). The best follicle growth and viability was observed in 0.5mm thin BOCS cultured with 5ng/ml FSH. In particular, when compared to 1mm thick BOCS cultured with 5ng/ml FSH, 0.5mm thin BOCS cultured with 5ng/ml FSH showed a higher and significant proportion of secondary follicles at day 10 (0.5 vs 1mm, % secondary follicles: 26.5 vs 10, P These results demonstrate that smaller BOCS thickness and 5ng/ml FSH supplementation significantly improve the growth and health of secondary follicles. Limitations, reasons for caution Although the bovine is considered a reliable model for human folliculogenesis, the study should be validated on human ovarian tissue. Wider implications of the findings A limiting step in the production of mature oocytes starting from primordial follicles is the low yield to secondary follicles at the end of organ culture. Given the similarities between bovine and human folliculogenesis, the best culture conditions herein identified could contribute to the refinement of human in vitro folliculogenesis. We present the first measurements of asteroids in millimeter wavelength (mm) data from the South Pole Telescope (SPT), which is used primarily to study the cosmic microwave background (CMB). We analyze maps of two $\sim270$ deg$^2$ sky regions near the ecliptic plane, each observed with the SPTpol camera $\sim100$ times over one month. We subtract the mean of all maps of a given field, removing static sky signal, and then average the mean-subtracted maps at known asteroid locations. We detect three asteroids$\text{ -- }$(324) Bamberga, (13) Egeria, and (22) Kalliope$\text{ -- }$with signal-to-noise ratios (S/N) of 11.2, 10.4, and 6.1, respectively, at 2.0 mm (150 GHz); we also detect (324) Bamberga with S/N of 4.1 at 3.2 mm (95 GHz). We place constraints on these asteroids' effective emissivities, brightness temperatures, and light curve modulation amplitude. Our flux density measurements of (324) Bamberga and (13) Egeria roughly agree with predictions, while our measurements of (22) Kalliope suggest lower flux, corresponding to effective emissivities of $0.66 \pm 0.11$ at 2.0 mm and $, 21 pages, 9 figures

We present measurements of the $E$-mode ($EE$) polarization power spectrum and temperature-$E$-mode ($TE$) cross-power spectrum of the cosmic microwave background using data collected by SPT-3G, the latest instrument installed on the South Pole Telescope. This analysis uses observations of a 1500 deg$^2$ region at 95, 150, and 220 GHz taken over a four month period in 2018. We report binned values of the $EE$ and $TE$ power spectra over the angular multipole range $300 \le \ell < 3000$, using the multifrequency data to construct six semi-independent estimates of each power spectrum and their minimum-variance combination. These measurements improve upon the previous results of SPTpol across the multipole ranges $300 \le \ell \le 1400$ for $EE$ and $300 \le \ell \le 1700$ for $TE$, resulting in constraints on cosmological parameters comparable to those from other current leading ground-based experiments. We find that the SPT-3G dataset is well-fit by a $\Lambda$CDM cosmological model with parameter constraints consistent with those from Planck and SPTpol data. From SPT-3G data alone, we find $H_0 = 68.8 \pm 1.5 \mathrm{km\,s^{-1}\,Mpc^{-1}}$ and $\sigma_8 = 0.789 \pm 0.016$, with a gravitational lensing amplitude consistent with the $\Lambda$CDM prediction ($A_L = 0.98 \pm 0.12$). We combine the SPT-3G and the Planck datasets and obtain joint constraints on the $\Lambda$CDM model. The volume of the 68% confidence region in six-dimensional $\Lambda$CDM parameter space is reduced by a factor of 1.5 compared to Planck-only constraints, with only slight shifts in central values. We note that the results presented here are obtained from data collected during just half of a typical observing season with only part of the focal plane operable, and that the active detector count has since nearly doubled for observations made with SPT-3G after 2018.

Modern CMB analysis pipelines regularly employ complex time-domain filters, beam models, masking, and other techniques during the production of sky maps and their corresponding angular power spectra. However, these processes can generate couplings between multipoles from the same spectrum and from different spectra, in addition to the typical power attenuation. Within the context of pseudo-$C_\ell$ based, MASTER-style analyses, the net effect of the time-domain filtering is commonly approximated by a multiplicative transfer function, $F_{\ell}$, that can fail to capture mode mixing and is dependent on the spectrum of the signal. To address these shortcomings, we have developed a simulation-based spectral correction approach that constructs a two-dimensional transfer matrix, $J_{\ell\ell'}$, which contains information about mode mixing in addition to mode attenuation. In this work we describe upgrades to the Spider balloon-borne telescope in preparation for its second flight, currently planned for December 2021. The Spider instrument is optimized to search for a primordial B-mode polarization signature in the cosmic microwave background at degree angular scales. During its first flight in 2015, Spider mapped ~10% of the sky at 95 and 150 GHz. The payload for the second Antarctic flight will incorporate three new 280 GHz receivers alongside three refurbished 95- and 150 GHz receivers from Spider's first flight. In this work we discuss the design and characterization of these new receivers, which employ over 1500 feedhorn-coupled transition-edge sensors. We describe pre-flight laboratory measurements of detector properties, and the optical performance of completed receivers. O alto nível de periculosidade das atividades e os índices de acidentes são alarmantes no cenário nacional. A fim de reduzir os riscos envolvidos, o corpo técnico atua na revisão e padronização de instruções de trabalho, homologação de novos equipamentos de proteção, constante treinamento das equipes e um rígido controle de atividades executadas. Porém, por serem processos essencialmente manuais, estes estão sujeitos à falha humana. Este projeto de pesquisa, portanto, propõe a aplicação de tecnologia de sensoriamento vestível na prevenção de ações e condições que precedem incidentes e que é definida em um conceito de pirâmide de DuPont como desvio de procedimento. Parâmetros físicos, fisiológicos e ambientais são monitorados e alimentam uma plataforma corporativa. Os resultados obtidos promovem inovação no sistema de gestão de riscos e, assim, criam um cenário totalmente novo, tecnológico e conectado para a prevenção de acidentes e de doenças.

We present component-separated maps of the primary cosmic microwave background/kinematic Sunyaev–Zel’dovich (SZ) amplitude and the thermal SZ Compton-y parameter, created using data from the South Pole Telescope (SPT) and the Planck satellite. These maps, which cover the ∼2500 deg2 of the southern sky imaged by the SPT-SZ survey, represent a significant improvement over previous such products available in this region by virtue of their higher angular resolution ( 1 .′ 25 for our highest-resolution Compton-y maps) and lower noise at small angular scales. In this work we detail the construction of these maps using linear combination techniques, including our method for limiting the correlation of our lowest-noise Compton-y map products with the cosmic infrared background. We perform a range of validation tests on these data products to test our sky modeling and combination algorithms, and we find good performance in all of these tests. Recognizing the potential utility of these data products for a wide range of astrophysical and cosmological analyses, including studies of the gas properties of galaxies, groups, and clusters, we make these products publicly available at http://pole.uchicago.edu/public/data/sptsz_ymap and on the NASA/LAMBDA website.

A new data logger based on Raspberry-Pi to monitor the locomotion of Arctic invertebrates has been made, tested and deployed in field. The device uses infrared sensors to check in vivo the invertebrates picking up the locomotor activity, data are collected for the analysis. Thanks to the Raspberry Pi capabilities and features, the instrument proved to be suitable for extreme scenarios such as the polar environment, offering good performances obtained at a very low price. Preliminary test made in field have demonstrated its reliability. Some experimental considerations and the trend of the biological rhythms of the tadpole shrimp’s locomotor activity are also discussed.

We provide an overview of the science case, instrument configuration and project plan for the next-generation ground-based cosmic microwave background experiment CMB-S4, for consideration by the 2020 Decadal Survey.

The electron signals from the field ionization of two closely-spaced Rydberg states of \mbox{rubidium-85} are separated using quantum control. In selective field ionization, the state distribution of a collection of Rydberg atoms is measured by ionizing the atoms with a ramped electric field. Generally, atoms in higher energy states ionize at lower fields, so ionized electrons which are detected earlier in time can be correlated with higher energy Rydberg states. However, the resolution of this technique is limited by the Stark effect. As the electric field is increased, the electron encounters numerous avoided Stark level crossings which split the amplitude among many states, thus broadening the time-resolved ionization signal. Previously, a genetic algorithm has been used to control the signal shape of a single Rydberg state. The present work extends this technique to separate the signals from the $34s$ and $33p$ states of rubidium-85, which are overlapped when using a simple field ramp as in selective field ionization.

In this paper we explore the possibility of using transition edge sensor (TES) detectors in multi-mode configuration in the focal plane of the Short Wavelength Instrument for the Polarization Explorer (SWIPE) of the balloon-borne polarimeter Large Scale Polarization Explorer (LSPE) for the Cosmic Microwave Background (CMB) polarization. This study is motivated by the fact that maximizing the sensitivity of TES bolometers, under the augmented background due to the multi-mode design, requires a non trivial choice of detector parameters. We describe 280 GHz bolometric detector arrays that instrument the balloon-borne polarimeter SPIDER. A primary science goal of SPIDER is to measure the large-scale B-mode polarization of the cosmic microwave background in search of the cosmic-inflation, gravitational-wave signature. 280 GHz channels aid this science goal by constraining the level of B-mode contamination from galactic dust emission. We present the focal plane unit design, which consists of a 16$\times$16 array of conical, corrugated feedhorns coupled to a monolithic detector array fabricated on a 150 mm diameter silicon wafer. Detector arrays are capable of polarimetric sensing via waveguide probe-coupling to a multiplexed array of transition-edge-sensor (TES) bolometers. The SPIDER receiver has three focal plane units at 280 GHz, which in total contains 765 spatial pixels and 1,530 polarization sensitive bolometers. A new inexpensive data logger specific designed to monitor the behavior of small animals in extreme scenario will be presented. The new data logger is based on common, commercial affordable and reliable devices that assure inexpensiveness and, at the same time, could be the base for further reasonings and developments with the aim to improve the device to face stressing scenarios such as that aerospace. Preliminary test realized in laboratory and in field have demonstrated its reliability.

Part of the problem in interpreting Smith within the framework of the subject-object polarity is that the term objective is used in various senses which are not always carefully discriminated. Thus objective knowledge may be, in the first place, knowledge of objects construed as external facts or things. Respecting this first sense of objectivity as knowledge of external objects arrived at by an epistemologica! approach that looks upon them as neutral, impersonal things, I believe Smith's critique is in the main incontestable. As early as 1968 in "What is Comparative Religion Comparing?"2 I was a convert to Smith's view that the proper study of humankind's religiousness is the meanings ascribed to religious facts, meanings that are at the same time the meanings ascribed to gods, human, history, and nature. The use of DNA restriction fragment length polymorphisms (RFLP) to analyze variable number of tandem repeat (VNTR) sequences dispersed in the human genome, has become a powerful tool for the study of population genetics due to the very substantial polymorphism involved. Because the markers usually employed for twin zygosity determination (such as sex combination, placentation, HLA typing, blood group antigens, etc) may not be uniformly informative, we propose the use of synthetic olygonucleotides, representing VNTR “core” sequences, for the determination of zygosity in twins.

The mechanisms of sperm adhesion and release within the mammalian oviduct are still poorly understood. In this in vitro study, a previously developed adhesion assay was used to analyze the effects of heparin, N-desulfated heparin, fucoidan, dextran sulfate, and dextran on bovine sperm-oviductal cell adhesion and release. Results showed that 1) all sulfated glycoconjugates were powerful inhibitors of sperm binding to oviductal monolayers in a dose-dependent manner, whereas N-desulfated heparin and dextran had no effect; 2) sperm pretreatment with heparin and fucoidan markedly inhibited adhesion; 3) treatment of oviductal monolayers with heparinase I, II, or sodium chlorate (an inhibitor of sulfation) had no effect on sperm adhesion; 4) sulfated glycoconjugates were also powerful and quick inducers of sperm release from oviductal monolayers; and 5) addition of sulfated glycoconjugates to the cocultures caused a sudden increase of bound-sperm flagellar beat frequencies, followed by a release of highly motile sperm. In conclusion, these data support the hypothesis that sulfated glycoconjugates may act as signals that induce sperm release and migration from the oviductal reservoir.

The mammalian oviduct plays a key role in sperm storage, capacitation, and selection. Specific oviduct secretions and/or binding to oviductal cells are thought to be responsible for the extension of the fertile life span of sperm. In this in vitro study, a quantitative assay for sperm binding was developed to analyze the mechanisms of sperm-oviductal cell adhesion and release in the bovine species. Distribution and acrosomal status of sperm bound to in vitro-cultured ampullary and isthmic cell monolayers were followed until the time of sperm release by means of fluorescence labeling techniques. In order to understand whether release is due to surface changes of sperm or oviductal cells, double incubation experiments with unlabeled and Hoechst-labeled sperm have been performed. Main findings demonstrate that (1) only acrosome-intact sperm bind specific bovine oviductal epithelial cells; (2) acrosomes of bound sperm are preserved intact over time; and (3) release of unreacted sperm is likely to be due to changes of the sperm surface, probably triggered by capacitation. These findings support the hypothesis that binding to oviductal cells is essential for preserving the sperm fertilization competence during the interval from the onset of estrus to ovulation.

In the present ultrastructural study intercellular bridges, connecting somatic granulosa cells to oocyte, have been detected for the first time and their modifications have been followed during Raja oogenesis. Intercellular bridges make their first appearance in small previtellogenic follicles as connecting devices between small cells and the oocyte. Later on, when the follicular epithelium becomes polymorphic and multilayered, for the presence of small, large, and pyriform-like cells, intercellular bridges link the oocyte and the different granulosa cells. Intercellular bridges contain ribosomes, whorl of membranes, mitochondria and vacuoles. Such cytoplasmic components are present also in the cell apex of large and pyriform-like cells thus suggesting, in agreement with other species (Motta et al. J. Exp. Zool., 1996;276:223-241) they may flow toward the oocyte. In this regard the presence of intercellular bridges during the oogenesis of cartilagineous fish may represent a crucial event of the active cooperation between granulosa cells and the oocyte.

A simple method for light microscope screening of pre-embedded, gold-labeled samples, by silver enhancing deplasticized semithin epoxy sections is presented. This method is of great help in visualizing the topography of gold labeled sites at low magnification and for selecting labeled sample regions for subsequent electron microscope analysis on serial ultrathin sections. The method was developed during a study of lectin binding sites on the egg surface of the anuran amphibian Discoglossus pictus.

A longstanding but inchoate preoccupation of mine is the geography of religions. How are landscape and religious faith related? In the context of this question, the definition of religion with which I work entails two considerations: first, what landscape features are incorporated into traditional religious symbol systems? I shall make a few observations about this geographic symbology below. Second, what is the existential connection between landscape and personal faith, i.e. between physical land features and human perceptions, values and commitments. It is this latter enquiry that is the main concern of this paper.

What concept of time is required to make sense of statements like ‘Jesus is the Eternal Contemporary’ or ‘There is a real presence of Christ in the sacramental action’?

A rough circumscription of our area of study will be achieved when I say that by images I mean the kind of thing about which Ian Crombie and Austin Farrer have written in Faith and Logic and Farrer also in The Glass of Vision. In the latter work Farrer cites as examples of religious images the Kingdom of God, the Son of Man, the Israel of God and the ‘infinitely complex and fertile image of sacrifice and communion, of expiation and covenant’. Farrer's images are essentially verbal pictures whose function is to interpret and elucidate revelatory events. He writes:The great images interpreted the events of Christ's ministry, death and resurrection, and the events interpreted the images; the interplay of the two is revelation. Certainly the events without the images would be no revelation at all, and the images without the events would remain shadows on the clouds. The events by themselves are not revelation, for they do not by themselves reveal the divine work which is accomplished in them: the martyrdom of a virtuous Rabbi and his miraculous return are not of themselves the redemption of the world.

A case of double bilateral renal vessels in both twins of a MZ pair is reported for the first time. Even in non-twins, the anomalies reported so far involved the inferior polar arteries in agreement with the embryological development. The two male twins, examined at the age of 14 years, had simultaneously developed a marked hypertension at the age of 7 years. Zygosity was determined by blood group and HLA analysis and various clinical tests were carried out to diagnose the condition. It is suggested that the anomaly is the result of a genetically induced early block in embryonic development.

Normal hemopoietic cell differentiation and proliferation is critically dependent upon marrow stromal elements. Long-term liquid culture of marrow provides a model for the study of stromal function. We evaluated the effects of radiation and 5-fluorouracil (5-FU) on various aspects of long-term murine hemopoietic cell growth and stromal function. Exposure of C57BL/6J murine adherent cells from long-term marrow cultures to varying doses of irradiation (0-1000 rad) in vitro resulted in the elaboration of growth factors stimulating granulocyte, macrophage, megakaryocyte, mixed megakaryocyte-granulocyte macrophage, and blast colonies. This increased production of growth factors appears to be related to the ablation of normal granulocyte production in the culture system since addition of normal stroma to irradiated stroma blocks growth factor production. Two cell types appear to be mediating stromal factor production and support of liquid culture hemopoiesis: a macrophagelike cell and an alkaline-phosphatase-positive epithelioid cell. Exposure of these two cell types to pokeweed mitogen results in marked enhancement of growth factor production. Furthermore, a cell line isolated from normal murine stroma produced an activity capable of acting at an early hemopoietic stem cell level and of inducing secondary marrow cell lines. The establishment of cultures from 5-FU-treated animals revealed that chemotherapy-depleted marrow was capable of establishing adequate stromal function and that the residual surviving stem cells had a higher than normal proliferative rate. In addition, the function of granulocytes derived from this post-5-FU marrow was normal. Thus, it appears that both chemotherapy and radiation exposure of marrow results in an enhanced capacity of stromal elements to produce growth factors and support hemopoiesis and that post-5-FU marrow represents an enriched source of high proliferative potential bone marrow stem cells.

These data suggest that two types of radioresistant adherent stromal cells are adequate for maintenance of long term hemopoiesis in the Dexter culture system. One cell type appears to be a typical adherent macrophage while the other is an alkaline phosphatase positive epitheloid cell possibly representative of the adventitial reticular cell of the bone marrow. These two cell types seem to be clearly defined by the in-vivo irradiation studies and less clearly defined in the in-vitro irradiation experiments. These data do not exclude other cell types as playing important roles in modulating hemopoiesis but do suggest that these major types are probably playing important roles in maintaining stem cells in long term liquid cultures. In addition, data suggests that the epitheloid cell directly nurtures hemopoietic cells on its surface while the macrophages may serve a separate function. A number of growth factors are produced by these two cell types which appear to include CSF-1, a granulocyte CSA separate from CSF-1 and a megakaryocyte CSA separate from both the GM-CSA and CSF-1 (Table 5). Thus the present data suggest that there are at least three (formula; see text) separate hemopoietic growth factors produced. The FDC-P1 activity produced by irradiated stroma would appear most likely to be GM-CSA-II. The fact that lectins enhanced production of these activities is intriguing but the cell type on which the lectins are acting has not as yet been defined. It appears that lithium also acts upon adherent marrow stromal cells to induce production of myeloid regulatory growth factors. Lithium appears to stimulate both normal and irradiated stroma to produce hemopoietic maintenance and growth factors and the present data suggests that factors active on pre IL-3 cells, HPP-CFC, CFU-D, CFU-meg, and CFU-S are all induced from these stromal cells by lithium. Whether the lithium induced factors and the factors derived from irradiated stroma represent a number of different growth factors or one or two critical regulatory molecules is at present unclear. The synergistic activity derived from the TC-1 cell line is of particular interest in this regard. This CSF-1 dependent activity is capable of acting on a very primitive stem cell to induce impressive proliferation and differentiation. In addition an activity in TC-1 conditioned media which may be synergistic activity appears capable of inducing adherent marrow cell lines which then can subsequently produce more of the same factor, a classic autocrine system.(ABSTRACT TRUNCATED AT 400 WORDS)
