The nature of glaciation (bipolar vs. unipolar) during the Eocene–Oligocene transition (EOT) remains unresolved. Here, we report the occurrence of frost marks, ice-rafted debris (IRD), and glendonites from the Upper Eocene to Lower Oligocene Niubao Formation (Fm.) deposited in a proglacial lake above glaciolacustrine conglomerates and diamictite facies in the Lunpola Basin, central Tibetan Plateau (CTP). Magnetostratigraphy dates these cryospheric deposits to ca. 36.2–31.8 Ma, synchronous with a stratigraphic interval containing IRD offshore of SE Greenland and in the Barents, Chukchi, and Laptev Seas, suggesting a strong continental-oceanic coupling. Our results provide robust continental evidence for intermittent cryospheric processes in the midlatitude Northern Hemisphere during the late Eocene and EOT. The global cold snap EOT-1 influenced already glacierized high-altitude mountains, lowering equilibrium line altitudes (ELAs) of glaciers and leading to local development of ice fields, ice caps, and valley glaciers with proglacial lake systems, such as the one recorded in the Niubao Fm. The record of IRD, glendonites, and frost marks before the onset of EOT-1 points to an active cryosphere on a plateau already elevated by ca. 36.2 Ma.

The main objective of this study is to investigate the language features and conventions of the political speech. Therefore, a study of it in the light of Systemic-functional Grammar will help to gain an in-depth understanding of the interpersonal meaning and the thoughts of national leaders in political speeches.

Adenoid basal carcinoma (ABC) of the cervix is a rare and low-incidence low-grade cervical cancer. In our practice, we encountered a case of cervical ABC with squamous differentiation and high-grade squamous intraepithelial lesion (HSIL) with gland involvement in the peripheral cervix. Reviewing relevant literature and analyzing its clinical manifestations, pathological morphology, and immunohistochemical characteristics would help deepen the understanding of this malignant tumor, so as to make a comprehensive diagnosis with differential diagnosis and prevent misdiagnosis.

Objective: To explore the mechanism by which macrophages secrete CCL22 to promote the metastasis of triple-negative breast cancer under hypoxic conditions. Methods: 20 ng/mL mass concentration of phorbol 12-myristate 13-acetate (PMA) cell culture medium, 4?,6-diamidino-2-phenylindole (DAPI), dimethyl sulfoxide (DMSO), trypsin digestion solution, CCL18 Kit , Interleukin (IL)-10 Kit, CCL17 Kit, CCL22 Kit, TRIzolTM Reagent Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) Kit, triple-negative breast cancer cells MDA-MB-231 and BT-549, as well as other reagents were used to culture triple-negative breast cancer (TNBC) cells MDA-MB-231 and BT-549 as well as human mononuclear cells THP-1, analyze and observe the metastasis of triple-negative breast cancer cells to the lungs, the secretion of CCL22, the migration of triple-negative breast cancer, and the situation of CCR4. Results: Compared with normal tumor-associated macrophage (TAM), hypoxic TAM further promoted the migration of tumor cells. The number of tumor metastases in the lungs, induced by hypoxic TAM, was significantly higher than that of normoxic TAM. Hypoxia can significantly stimulate the expression of CCL22. CCL22 can significantly promote the migration of MDA-MB-231 and BT-549 cells. The expressions of CCR3, CCR4, and CCR5 in tumor tissues were significantly increased compared with normal tissues, in which CCR4 showed the most significant increase. Conclusion: TAM cultured under hypoxia significantly enhanced the migration ability of TNBC cells and promoted the metastasis of cancer cells to the lungs in vivo. The hypoxic condition induced TAM to secrete CCL22; the expression of CCL22 receptor, CCR4, in breast cancer tissues was significantly higher than that in normal tissues.

General academic English has been attached greater importance in higher education in China. This paper focuses on the teaching effect of general academic English in North China Electric Power University（Baoding）from the perspective of PCT（Projects+Cases+Texts）. Through the analysis of students’ questionnaire and interview, it is proved that PCT is an effective way in general academic English teaching. This paper aims to provide practical experience for the further development of general academic English curriculum.

Background. Hematopoietic stem cell transplantation (HSCT) has become the main treatment for acute myeloid leukemia (AML) and has been studied in many systematic reviews (SRs), but strong conclusions have not been drawn yet. Objective. This study aimed to summarize and critically evaluate the methodological and evidence quality of SRs and meta-analysis on this topic. Methods. PubMed, Embase, the Cochrane Library, and Web of Science were searched for SRs/meta-analyses regarding HSCT for AML. Two reviewers assessed the quality of SRs/meta-analyses in line with AMSTAR-2 and evaluated the strength of evidence quality with the grading of the evaluation system (GRADE) for concerned outcomes independently. Results. 12 SR/Meta articles were included, and the AMSTAR-2 scale showed that the quality grade of all articles was low or very low. GRADE results showed 29 outcomes, 2 of which were high, 12 were moderate, and 15 were low. Limitations and inconsistency were the most important factors leading to degradation, followed by imprecision and publication bias. Allo-SCT had better OS and DFS benefits than auto-SCT and significantly reduced the relapse in intermediate-risk AML/CR1 patients. Auto-SCT was associated with lower TRM than allo-SCT but generally had higher relapse. The results should be confirmed further for the low or moderate evidence quality. Conclusion. Current SRs show that allo-SCT in the treatment of AML might improve the OS, RFS, and DFS. Auto-SCT has significantly lower TRM but higher RR. Whether bone marrow transplantation is superior to nonmyeloablative chemotherapy remains to be evaluated. Meanwhile, the quality of methodology needs to be further improved. The intensity of evidence was uneven, and the high-quality evidence of outcomes was lacking. Considering the limitations of our overview, more rigorous and scientific studies are needed to fully explore the efficacy of different interventions of HSCT in AML, and clinicians should be more cautious in the treatment.

Electric vehicle deployment shows promising potentials in promoting cleaner energy utilization and reducing carbon emission. Due to increasing carbon neutral pressure and market competition from transportation sector, government agencies and public bodies (GAPBs) have emphasized the significance of electric vehicle adoption through supplier selection. Consequently, GAPBs must consider a reasonable criteria system and a comprehensive supplier selection framework and rationally select the electric vehicle supplier that matches their practical needs in terms of economic, social, environmental, and technical factors. This paper provides insights into electric vehicle supplier selection (EVSS) from the perspective of GAPBs using an integrated multi-criteria decision-making (MCDM) framework based on best-worst method (BWM) and fuzzy ViseKriterijumska Optimizacija I Kompromisno Resenje (VIKOR). Initially, 14 critical factors from economic, social, environmental, and technical dimensions are identified as the criteria by literature review and experts' opinions. Then, a comprehensive decision framework using the integrated MCDM approach is proposed. To validate the applicability and feasibility of the proposed framework, a case study is launched and analyzed. It emerges that bad environmental record, cost, quality, service, and environmental initiatives are the most important criteria in EVSS for GAPBs with the weight values of 0.1995, 0.1172, 0.1219, 0.0708, and 0.2553. The comparative analysis and the sensitivity analysis are performed for verifying the reliability of the proposed framework. The work helps to understand the electric vehicle supplier selection criteria and makes methodological decision-making support for GAPBs.

Hepatitis B virus (HBV) is a major risk factor for the development and progression of hepatocellular carcinoma (HCC). It has been reported that viral infection can interfere with the expression of cellular microRNA (miRNA) to affect oncogenesis. In this study, we showed that miR-520c-3p was upregulated in liver tumor specimens, and we revealed that HBV infection enhanced the expression of miR-520c-3p through the interaction of viral protein HBV X protein (HBx) with transcription factor CREB1. We further showed that miR-520c-3p induced by HBV transfection/infection caused epithelial-mesenchymal transition (EMT). Using the miRNA target prediction database miRBase and luciferase reporter assays, we identified PTEN as a novel target gene of miR-520c-3p and miR-520c-3p directly targeted PTEN's 3'-untranslated region. Moreover, we discovered that HBV promoted EMT via the miR-520c-3p-PTEN to activate AKT-NFκB signaling pathway, leading to increased HCC migration and invasion. Importantly, miR-520c-3p antagomir significantly represses invasiveness in HBx-induced hepatocellular xenograft models. Our findings indicate that miR-520c-3p is a novel regulator of HBV and plays an important role in HCC progression. It may serve as a new biomarker and molecular therapeutic target for HBV patients.

In recent years, there are increasingly more standardized trainings for doctors based on the standardized training outline that is suitable for different grades. Hierarchical training has gradually formed a relatively sound training mode. In reflecting students’ learning effect, there is a need for an ideal quality feedback system in the standardized practical skills training for clinical pathology residents. This feedback system can help students achieve the training objectives through understanding and grasping the learning content.

This research, on the basis of outcome-based educational theory, using Unit 10 in Book 1, New Experiencing English Viewing, Listening and Speaking by Higher Education Press as the teaching content, aims to design a syllabus for college English class. Focusing on results’ orientation of student, this paper is intended to promote the development of college English class, further improve the efficiency of college English class and also hope to provide substantial support for college English teaching studies.

The Report on the Work of the Chinese Government in 2021 emphasised that stable employment is the foundation of national development. Therefore, adjustment of the employment structure is one of the main routes for sustainable development of resource-based cities. However, the impact of employment structure on sustained economic growth, particularly in heterogeneous regions, has not yet been determined. This study analyses China’s employment structure’s spatial evolution, using panel data from 2004 to 2018 of 115 prefecture-level resource-based cities. It explores the driving factors and spatial effects of employment structure changes on economic growth through an extended two-sector economic growth model and spatial econometric model, and proposes solutions for heterogeneous regions. The results show that the labour productivity of the employed population in the secondary industry is the most important factor affecting economic growth, but the spatial effects of employment structure adjustment on economic growth are different in heterogeneous regions. They further reveal that improving the productivity of the employed population in the secondary industry and building an industrial system according to regional advantages are the top priorities for developing the sustainable economy of resource-based cities.

This paper investigates the water quality system using the Piper diagram method to qualitatively analyze the distribution, characteristics, and laws of typical ion content. It also compares the differences between ion components of each aquifer with the closeness of the water body to be discriminated by combining statistical characteristic values. The R factor is utilized to simplify the index attributes, reduce input information dimension, and construct the coupled R-SVM discriminative model of Zhaogezhuang Mine. The input index is established as typical ion content, and the output is water source type. The discriminative model automatically establishes the mapping relationship between water quality indexes and discriminative criteria by learning the inherent property law between water quality samples. The independence components after dimensionality reduction are used as new discriminative indexes. The accuracy of the coupled model classification was 90.90% in the verification of the water source discrimination example of Zhaogezhuang mine. The coupled quantitative discriminant model based on R-factor and support vector machine provides an auxiliary verification and scientific decision for qualitative water chemistry analysis and provides a new idea for water source identification. Compared with the traditional qualitative way of water chemistry characteristics, Fisher function discrimination method, and single support vector machine model, this method improved the accuracy by more fully exploiting the internal laws of the data.

The decentralized manycore architecture is broadly adopted by neuromorphic chips for its high computing parallelism and memory locality. However, the fragmented memories and decentralized execution make it hard to deploy neural network models onto neuromorphic hardware with high resource utilization and processing efficiency. There are usually two stages during the model deployment: one is the logical mapping that partitions parameters and computations into small slices and allocate each slice into a single core with limited resources; the other is the physical mapping that places each logical core to a physical location in the chip. In this work, we propose the mapping limit concept for the first time that points out the resource saving upper limit in logical and physical mapping. Furthermore, we propose a closed-loop mapping strategy with an asynchronous 4D model partition for logical mapping and a Hamilton loop algorithm (HLA) for physical mapping. We implement the mapping methods on our state-of-the-art neuromorphic chip, TianjicX. Extensive experiments demonstrate the superior performance of our mapping methods, which can not only outperform existing methods but also approach the mapping limit. We believe the mapping limit concept and the closed-loop mapping strategy can help build a general and efficient mapping framework for neuromorphic hardware.

Messenger RNA (mRNA) stability and translational efficiency are two crucial aspects of the posttranscriptional process that profoundly impact protein production in a cell. While it is widely known that ribosomes produce proteins, studies during the past decade have surprisingly revealed that ribosomes also control mRNA stability in a codon-dependent manner, a process referred to as codon optimality. Therefore, codons, the three-nucleotide words read by the ribosome, have a potent effect on mRNA stability and provide cis -regulatory information that extends beyond the amino acids they encode. While the codon optimality molecular mechanism is still unclear, the translation elongation rate appears to trigger mRNA decay. Thus, transfer RNAs emerge as potential master gene regulators affecting mRNA stability. Furthermore, while several factors related to codon optimality have been identified in yeast, the orthologous genes in vertebrates do not necessary share the same functions. Here, we discuss codon optimality findings and gene regulation layers related to codon composition in different eukaryotic species. ObjectivesWe developed and tested a deep learning (DL) framework applicable to color Doppler echocardiography for automatic detection and quantification of atrial septal defects (ASDs).BackgroundColor Doppler echocardiography is the most commonly used non-invasive imaging tool for detection of ASDs. While prior studies have used DL to detect the presence of ASDs from standard 2D echocardiographic views, no study has yet reported automatic interpretation of color Doppler videos for detection and quantification of ASD.MethodsA total of 821 examinations from two tertiary care hospitals were collected as the training and external testing dataset. We developed DL models to automatically process color Doppler echocardiograms, including view selection, ASD detection and identification of the endpoints of the atrial septum and of the defect to quantify the size of defect and the residual rim.ResultsThe view selection model achieved an average accuracy of 99% in identifying four standard views required for evaluating ASD. In the external testing dataset, the ASD detection model achieved an area under the curve (AUC) of 0.92 with 88% sensitivity and 89% specificity. The final model automatically measured the size of defect and residual rim, with the mean biases of 1.9 mm and 2.2 mm, respectively.ConclusionWe demonstrated the feasibility of using a deep learning model for automated detection and quantification of ASD from color Doppler echocardiography. This model has the potential to improve the accuracy and efficiency of using color Doppler in clinical practice for screening and quantification of ASDs, that are required for clinical decision making.

Background: As distinct marker of proliferating cells, chromatin assembly factor-1 (CAF-1) was critical in DNA replication. However, there is paucity information about the clinical significance, functions and co-expressed gene network of CHAF1A, the major subunit in CAF-1, in cancer.Methods: Bioinformatic analysis of CHAF1A and its co-expression gene network were performed using various public databases. Functional validation of CHAF1A was applied in breast cancer.Results: Overexpression of CHAF1A was found in 20 types of cancer tissues. Elevated expression of CHAF1A was positively correlated with breast cancer progression and poor patients’ outcome. The analysis of co-expression gene network demonstrated CHAF1A was associated with not only cell proliferation, DNA repair, apoptosis, but cancer metabolism, immune system, and drug resistance. More importantly, higher expression of CHAF1A was positively correlated with immunosuppressive microenvironment and resistance to endocrine therapy and chemotherapy. Elevated expression of CHAF1A was confirmed in breast cancer tissues. Silencing of CHAF1A can significantly inhibit cell proliferation in MDA-MB-231 cells.Conclusion: The current work suggested that overexpression of CHAF1A can be used as diagnostic and poor prognostic biomarker of breast cancer. Higher expression of CHAF1A induced fast resistance to endocrine therapy and chemotherapy, it may be a promising therapeutic target and a biomarker to predict the sensitivity of immunotherapy in breast cancer.

ObjectivePrecise monitoring of cotton leaves’ nitrogen content is important for increasing yield and reducing fertilizer application. Spectra and images are used to monitor crop nitrogen information. However, the information expressed using nitrogen monitoring based on a single data source is limited and cannot consider the expression of various phenotypic and physiological parameters simultaneously, which can affect the accuracy of inversion. Introducing a multi-source data-fusion mechanism can improve the accuracy and stability of cotton nitrogen content monitoring from the perspective of information complementarity.MethodsFive nitrogen treatments were applied to the test crop, Xinluzao No. 53 cotton, grown indoors. Cotton leaf hyperspectral, chlorophyll fluorescence, and digital image data were collected and screened. A multilevel data-fusion model combining multiple machine learning and stacking integration learning was built from three dimensions: feature-level fusion, decision-level fusion, and hybrid fusion.ResultsThe determination coefficients (R2) of the feature-level fusion, decision-level fusion, and hybrid-fusion models were 0.752, 0.771, and 0.848, and the root-mean-square errors (RMSE) were 3.806, 3.558, and 2.898, respectively. Compared with the nitrogen estimation models of the three single data sources, R2 increased by 5.0%, 6.8%, and 14.6%, and the RMSE decreased by 3.2%, 9.5%, and 26.3%, respectively.ConclusionThe multilevel fusion model can improve accuracy to varying degrees, and the accuracy and stability were highest with the hybrid-fusion model; these results provide theoretical and technical support for optimizing an accurate method of monitoring cotton leaf nitrogen content.

Based on the development concept and requirements of the new liberal arts construction, this paper proposes specific measures to improve the basic ability of public management talents cultivation and promote the process of the new liberal arts construction in universities.

ObjectivesThe association between coffee consumption and the risk of metabolic syndrome (MetS) remains inconsistent. The aim of this study was to evaluate the association between coffee intake and components of MetS.MethodA cross-sectional survey including 1,719 adults was conducted in Guangdong, China. Data on age, gender, education level, marriage status, body mass index (BMI), current smoking and drinking status and breakfast habit, coffee consumption type, and daily servings were derived based on 2-day, 24-h recall. MetS were assessed according to the International Diabetes Federation definition. Multivariable logistic regression was conducted to examine the association between the coffee consumption type, daily servings, and the components of MetS.ResultsRegardless of the coffee type, compared with non-coffee consumers, coffee consumers had higher odds ratios (ORs) of the elevated fasting blood glucose (FBG) in both men [OR: 3.590; 95% confidence intervals (CI): 2.891–4.457] and women (OR: 3.590; 95% CI: 2.891–4.457). In women, the risk of elevated blood pressure (BP) was 0.553 times (OR: 0.553; 95% CI: 0.372–0.821, P = 0.004) for people who drank total coffee > 1 serving/day than for non-coffee drinkers.ConclusionIn conclusion, regardless of type, coffee intake is associated with an increased prevalence of FBG in both men and women, but has a protective effect on hypertension only in women.

Incorporating mixed ion is a frequently used strategy to stabilize black-phase formamidinum lead iodide perovskite for high-efficiency solar cells. However, these devices commonly suffer from photoinduced phase segregation and humidity instability. Herein, we find that the underlying reason is that the mixed halide perovskites generally fail to grow into homogenous and high-crystalline film, due to the multiple pathways of crystal nucleation originating from various intermediate phases in the film-forming process. Therefore, we design a multifunctional fluorinated additive, which restrains the complicated intermediate phases and promotes orientated crystallization of α-phase of perovskite. Furthermore, the additives in-situ polymerize during the perovskite film formation and form a hydrogen-bonded network to stabilize α-phase. Remarkably, the polymerized additives endow a strongly hydrophobic effect to the bare perovskite film against liquid water for 5 min. The unencapsulated devices achieve 24.10% efficiency and maintain >95% of the initial efficiency for 1000 h under continuous sunlight soaking and for 2000 h at air ambient of ~50% humid, respectively.

Utilization of specific codons varies between organisms. Cancer represents a model for understanding DNA sequence evolution and could reveal causal factors underlying codon evolution. We found that across human cancer, arginine codons are frequently mutated to other codons. Moreover, arginine limitation—a feature of tumor microenvironments—is sufficient to induce arginine codon–switching mutations in human colon cancer cells. Such DNA codon switching events encode mutant proteins with arginine residue substitutions. Mechanistically, arginine limitation caused rapid reduction of arginine transfer RNAs and the stalling of ribosomes over arginine codons. Such selective pressure against arginine codon translation induced an adaptive proteomic shift toward low-arginine codon–containing genes, including specific amino acid transporters, and caused mutational evolution away from arginine codons—reducing translational bottlenecks that occurred during arginine starvation. Thus, environmental availability of a specific amino acid can influence DNA sequence evolution away from its cognate codons and generate altered proteins.

G-quadruplex (G4) is a kind of peculiar nucleic acid secondary structure formed by DNA or RNA, which is considered as a fundamental feature of the genome. Many proteins can specifically bind to G4 structures. There is increasing evidence that G4-protein interactions involve in the regulation of important cellular processes such as DNA replication, transcription, RNA splicing and translation. Besides, G4-protein interactions have been proved to be potential targets for disease treatment. In order to unravel the detailed bioregulatory mechanisms of G4-binding proteins (G4BPs), biochemical methods for detecting G4-protein interactions with high specificity and sensitivity are highly demanded. Here, we review recent advances in the screening and validation of new G4BPs and bioregulatory tools of G4BPs.
