This article proposes a competitive-evolving-committee proactive secret sharing. Every participant in the system has the opportunity to become a member of the holding committee and have sufficient anonymity. During the life cycle of serving as the holding committee members, they only send one message in the protocol without excessive interaction, and achieve receiver strong anonymity with a capability-based encryption scheme different from most public-key encryption schemes, at present named RiddleEncryption, which is also proposed in this paper. In RiddleEncryption the sender does not need to pay attention to the specific identity of the receiver but focuses on what kind of capability the receiver should have. Nobody can determine this kind of capability at the beginning of the system establishment. This article aims at depositing a secret in a distributed manner (e.g., blockchain) without excessive trust and to emphasize more anonymity and capability. The scheme can be used in the dynamic groups, authentication management, rights abuse prevention, and so on.

Blueberry leaf spots and stem cankers caused by Pestalotiopsis spp. have become a serious threat for the production of blueberry in Sichuan Province. To characterize the etiology of the diseases connected with these fungi, samples showing leaf spot and stem canker symptoms were collected from the 12 main blueberry-growing areas of Sichuan Province from 2015 to 2020 and used for pathogen isolation. In total, 91 fungal isolates were obtained with preliminary morphological identification and 48 representative strains were selected for further pathogenicity test and molecular identification. Four species, including Pestalotiopsis clavispora (Neopestalotiopsis clavispora) (57.14%), P. trachicarpicola (28.57%), P. chamaeropis (13.19%), and P. adusta (1.10%), were identified based on conidial morphology, cultural characteristics, and phylogenetic analysis of the internal transcribed spacer region, partial sequence of the β-tubulin gene, and the translation elongation factor 1-α. Pathogenicity tests showed that four species were pathogenic to leaves and stems of blueberry. Among them, P. clavispora (N. clavispora) was the most aggressive as the predominant species to cause both leaf spot and stem canker. P. trachicarpicola and P. chamaeropis were mainly isolated from leaves but also pathogenic to stems. P. adusta was only isolated from stems but also pathogenic to leaves. To the best of our knowledge, this is the first report of P. chamaeropis and P. adusta as pathogens causing leaf spots and stem canker on blueberry. The results provide helpful information in disease diagnosis and management of blueberry.

Due to the increasing bacterial resistance to conventional antibiotics, developing safe and effective approaches to combat infections caused by bacteria and biofilms has become an urgent clinical problem. Recently, carbon dots (CDs) have received great attention as a promising alternative to conventional antimicrobial agents due to their excellent antimicrobial efficacy and biocompatibility. Although CDs have been widely used in the field of antibacterial applications, their antibacterial and antibiofilm mechanisms have not been systematically discussed. This review provides a systematic overview on the complicated mechanisms of antibacterial and antibiofilm CDs based on recent development.

Background Gout is usually found in patients with atrial fibrillation (AF). K+ efflux is a common trigger of NLRP3 inflammasome activation which is involved in the pathogenesis of AF. We investigated the role of the K+ channel Kv1.5 in monosodium urate crystal (MSU)-induced activation of the NLRP3 inflammasome and electrical remodeling in mouse and human macrophages J774.1 and THP-1, and mouse atrial myocytes HL-1. Methods and Results Macrophages, primed with lipopolysaccharide (LPS), were stimulated by MSU. HL-1 cells were incubated with the conditioned medium (CM) from MSU-stimulated macrophages. Western blot, ELISA and patch clamp were used. MSU induced caspase-1 expression in LPS-primed J774.1 cells and IL-1β secretion, suggesting NLRP3 inflammasome activation. A selective Kv1.5 inhibitor, diphenyl phosphine oxide-1 (DPO-1), and siRNAs against Kv1.5 suppressed the levels of caspase-1 and IL-1β. MSU reduced intracellular K+ concentration which was prevented by DPO-1 and siRNAs against Kv1.5. MSU increased expression of Hsp70, and Kv1.5 on the plasma membrane. siRNAs against Hsp70 were suppressed but heat shock increased the expression of Hsp70, caspase-1, IL-1β, and Kv1.5 in MSU-stimulated J774.1 cells. The CM from MSU-stimulated macrophages enhanced the expression of caspase-1, IL-1β and Kv1.5 with increased Kv1.5-mediated currents that shortened action potential duration in HL-1 cells. These responses were abolished by DPO-1 and a siRNA against Kv1.5. Conclusions Kv1.5 regulates MSU-induced activation of NLRP3 inflammasome in macrophages. MSUrelated activation of NLRP3 inflammasome and electrical remodeling in HL-1 cells are via macrophages. Kv1.5 may have therapeutic value for diseases related to gout-induced activation of the NLRP3 inflammsome, including AF.

Soybean (Glycine max L.) is one of the important oilseed and vegetable crop worldwide and provides the main source of vegetable oil and proteins for human and livestock (Hartman et al. 2011). In October 2021, approximately 35% of soybean pods suffered from anthracnose in the farmer’s field in Chongzhou, Sichuan Province, China (103°40'12"E, 30°37'48"N), and the occurrence area accounted for about 3.3 hm2. Symptoms of soybean were characterized by yellow spots at the initial stage, gradually expanded into dark brown spots, and eventually amounts of small black particles were densely arranged in the wheel shape on dead spots. Diseased spots of soybean pods were cut into pieces and sequentially sterilized in 75% alcohol for 30 s, 4% sodium hypochlorite for 30 s, sterile water for 3 times. After that, these pieces were placed on potato dextrose agar (PDA), and incubated at 25±2°C in the dark for 5-7 days. Single spore was separately picked and transferred to a fresh PDA plate to obtain pure culture isolates. Total six pure isolates were collected, and among them the hyphae of representative isolate 8-B were initially white, turned grey gradually on PDA medium, and the colonial reverse were radiating, whorled or a mixture of both. Conidia of 8-B were septate, hyaline, unicellular, cylindrical, obtusely rounded at both ends with 1 or 2 oil balls inside, and 10.5-17.6 µm in length and 7.0 µm-3.6 µm in width (n=100). The conidial appressoria were brown subspherical, 6.9 µm-13.3 µm in length and 5.6 µm-10.1 µm (n=50) in width. Based on morphological and cultural characteristics, the isolate 8-B was tentatively identified as Colletotrichum gloeosporioides species complex（Weir et al. 2012). To test pathogenicity, the mycelial plugs were inoculated on 20 detached soybean pods at full seed (R6) stage, and three areas of each pod were lightly scratched using a needle prior to inoculation. As controls, the PDA plugs were attached to the pinned-treated pods. Three independent replicates were conducted for control and inoculated pods, respectively. All pods were incubated in a greenhouse at 25 ± 2°C with a relative humidity of approximately 90%. After 4-5 days post-inoculation, typical anthracnose lesions were observed on the inoculated pods while the control pods remained healthy only with small wound spots. The pathogen re-isolated from all the inoculated pods were morphologically identical to the inoculation isolate (8-B). For further molecular verification, the six gene fragments including the internal transcribed spacer (ITS), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), chitin synthase 1 (CHS-1), actin (ACT), β-tubulin 2 (TUB2) and calmodulin (CAL) were amplified and sequenced (Weir et al. 2012, Damm et al. 2012), and the obtained sequences were deposited in GenBank (Accession numbers ON960278, ON685214, ON964475, ON974476, ON685215 and ON964477, respectively). All six gene sequences of 8-B had a high identity to C. fructicola (the stand isolate ICMP 18581) with the accession numbers ON960278 (100%), ON974476 (96%), ON685214 (99%), ON964475 (99%), ON685215 (100%), and ON964477 100%), respectively. Anthracnose disease caused by C. fructicola has previously been reported to affect a range of plant hosts worldwide (Guarnaccia et al. 2017). However, it is still unknown on C. fructicola causing anthracnose in soybean in China. This study firstly reports C. fructicola as the causal agent of anthracnose on soybean in the country, and provides a theoretical basis for the diagnosis and control of this disease.

Bacterial infection and excessive reactive oxygen species (ROS) remain challenging factors contributing to the delayed healing of chronic wounds. Although various antibacterial and antioxidant hydrogel dressings have been developed to accelerate wound healing, multifunctional hydrogels fabricated by rationally designing and introducing carbonized polymer dots (CPDs) have rarely been reported. Herein, inspired by the mussel biomimetic approach, we synthesized 3,4-dihydroxybenzaldehyde functionalized chitosan (DFC), and then the polymeric precursor was pyrolyzed into CPDs with abundant amino and catechol groups on the surface, which endowed it with a highly positively charged surface that could activate the photothermal effect under near-infrared (NIR) light irradiation. Finally, the nanocomposite hydrogel (PVA@CPDs) was simply constructed by directly mixing polyvinyl alcohol (PVA) with CPDs, utilizing the freeze-thaw cycle method to form a gel, in which, CPDs as a center of polyfunctional nanoparticles drove the formation of PVA microcrystalline crosslinking and endowed the PVA substrate with versatile functionalities. Remarkable and comprehensive improvements in the swelling behavior, mechanical properties and adhesive strength of the hydrogel could be conveniently achieved with the suitable loading of CPDs. The

H. Mutabilis (Cotton rose or confederate rose) is a deciduous shrub in the Malvaceae family, with ornamental, medicinal and edible values (Fan et al. 2015). In May to August 2020, 40.4% of potted plants of H. mutabilis were found to have root and stalk rot in Chengdu Botanical Garden (E104°7′28″, N30°45′57″). At first the leaves of affected H. mutabilis turned yellow and wilted, followed by the stem and root cortex became dark brown and rotten. Finally, the whole plant died within two months. Root and stem produced white mycelium when the humidity exceeded 90%. Samples taken from the lesions were surface disinfested for 3 min in 4% sodium hypochlorite, rinsed in sterile water and plated on potato dextrose agar (PDA), 35 single-spore cultures with similar morphology isolated from symptomatic tissues were obtained and subcultured. After seven days at 25°C in the dark, the mycelium of a representative culture MFR1 covered the entire plate surface (9 cm diameter). The aerial mycelium of cultures were white and fluffy at first and produced lavender pigment on the back of the cultures in the later stage. After seven days, the cultures produced abundant sickle-shaped macroconidia which have 3 to 5 septations and some oval or oblong microconidia which have 0 to 1 septation. Macroconidia 22.35~46.67 μm (mean 32.11 μm) in length and 4.32~7.72 μm (mean 5.21 μm) in width (n = 100). Microconidia 7.10~21.85 μm (mean 11.62 μm) in length and 2.76~6.84 μm (mean 4.20 μm) in width (n = 100). Based on these characteristics, isolates were tentatively identified as Fusarium sp. (Crous et al. 2021). Pathogenicity was tested on 1-year-old potted seedlings of H. mutabilis by root-zone irrigation inoculation in Sichuan Agricultural University (Jia et al.2019). Conidia suspension (1×107conidia/mL,collected from MFR1 )was poured into the soil along the plant roots. The same amount of distilled water was poured around the roots of the control plants. All inoculated and control plants were incubated in the greenhouse (about 25 ± 2°C). The experiment was performed three times. Within 25 days after inoculation, all plants inoculated with pathogens showed symptoms similar to those in the field, whereas the controls remained symptomless. The pathogen was reisolated from all inoculated plants, and the cultural and morphological characteristics were the same as those of the original isolate. After DNA extraction and PCR amplification, the translation elongation factor 1-alpha (TEF) and RNA polymerase II second largest subunit (RPB2) genes of a representative culture MFR1, were sequenced (O’Donnell et al. 2010) and deposited in GenBank (accession numbers OK334295 and ON316728, respectively). The TEF and RPB2 sequences were 99.7% and 99.39% identical to those of F. oxysporum (MN892354 and MZ198892). The result was confirmed by multilocus phylogenetic analysis. Through morphological identification and molecular analyses, the pathogen was identified as F. oxysporum. F. oxysporum is known to infect cotton (Dowd et al.2004), soybean (Ellis et al.2016) and banana (Fourie et al.2011) among other hosts, but it is the first report of F. oxysporum infecting H. mutabilis in China or worldwide. This disease seriously reduced the survival rate of H. mutabilis and may become an important reason to hinder the increase of H. mutabilis in potted seedlings stage. Moreover, the findings will provide theoretical basis to solve the bottleneck problem affecting the popularization and propagation of H. mutabilis.

Broadband optical frequency comb (OFC) generation based on a single electro-absorption modulator (EAM) is proposed. The EAM is driven by a radio frequency (RF) multi-frequency signal generated by a multiplication coupler composed of an electrical power splitter and an arithmetic circuit. Thus the number of comb-lines of the generated OFC can be increased. A complete theoretical model of OFC generation by an EAM driven by nth power of the RF source is established, and the performance of the OFC is analyzed by using OptiSystem software. The results show that, the number of comb-lines of the OFC is positively correlated with the number of multiplication of the RF source signal. The frequency spacing of the comb-lines is twice the frequency of the RF source signal and is tunable by adjusting the frequency of the RF source signal. Increasing chirp factor and modulation index of EAM could increase the number of comb-lines of the generated OFC. The amplitude of the RF source signal had little impact on the flatness of the OFC and the average OFC power. The scheme developed is not only simple and low-cost, but also can produce a large number of comb-lines. Graphical Abstract

The enhancement of anonymity for blockchain users received much attention. However, in some cases tracing user’s identity is also very important, especially to expose illegal transactions. In this paper, we propose an identity tracing scheme and implement it using a simple and efficient proof method. In particular, we design a new signature scheme that is used to generate a user’s certificate, and present a binding method for the user’s certificate and trading public key. Users only need to register once with the supervision center, and then the center may trace sender’s identity of the transactions on the blockchain. Our scheme is suitable to blockchain-based systems that explicitly use public/private key pairs, and does not affect the anonymity property of the original system.

It is very challenging to design nanomaterials with both excellent antibacterial activity and cytocompatibility when facing bacterial infection. Serum uric acid (UA) is taken up by endothelial cells and reduces the level of nitric oxide (NO) by inhibiting its production and accelerating its degradation. Cytosolic and plasma xanthine oxidase (XO) generates superoxide and also decreases the NO level. Thus, hyperuricemia is associated with impaired endothelial function. Hyperuricemia is often associated with vascular diseases such as chronic kidney disease (CKD) and cardiovascular disease (CVD). It has long been debated whether hyperuricemia is causally related to the development of these diseases. The 2020 American College of Rheumatology Guideline for the Management of Gout (ACR2020) does not recommend pharmacological treatment of hyperuricemia in patients with CKD/CVD. In contrast, the Japanese Guideline on Management of Hyperuricemia and Gout (JGMHG), 3rdedition, recommends pharmacological treatment of hyperuricemia in patients with CKD. In a FREED study on Japanese hyperuricemic patients with CVD, an XO inhibitor, febuxostat, improved the primary composite endpoint of cerebro-cardio-renovascular events, providing a rationale for the use of urate-lowering agents (ULAs). Since a CARES study on American gout patients with CVD treated with febuxostat revealed increased mortality, ACR2020 recommends switching to different ULAs. However, there was no difference in the mortality of Japanese patients between the febuxostat-treated group and the placebo or allopurinol-treated groups in either the FEATHER or FREED studies.

Two experiments were conducted to evaluate the effects of adding multi-enzyme on the available energy concentration and standardized ileal digestibility (SID) of amino acids of double-low rapeseed expellers (RSE) and rapeseed meal (RSM) fed to growing pigs. In Experiment 1, a total of 36 growing barrows were fed six diets in a replicated 3 × 2 factorial design. Diets included a corn–soybean meal basal diet, RSE diet, and RSM diet with or without multi-enzyme (1000 U g−1 of cellulase, 10 000 U g−1 of xylanase, 2000 U g−1 of glucanase, and 10 000 U g−1 of protease). Bacterial biofilm infection is perplexing people’s life. Systemic administration of antibiotics usually causes adverse effects and bacterial resistance. How to make antibacterial agents quickly penetrate and eradicate the biofilm without adverse effects is an urgent problem. Here, polyamine-functionalized carbon quantum dots (CQDs), abbreviated as CQD600, CQD1w and CQD2.5w, are synthesized by a simple hydrothermal treatment of citric acid and branched polyethyleneimine (bPEI) with different molecular weight (MW). It is found that the antibacterial activity is promoted with high MW of bPEI, which is due to the protonated amines on the surface of the CQDs. Moreover, in terms of eradicating mature Staphylococcus aureus biofilm, CQD2.5w possesses significantly higher activity than CQD600 and CQD1w because of stronger electrostatic interactions and longer surface coronas. Additionally, the cytotoxicity test exhibits the L929 cells can keep at high survival rates even treated with 500 µg mL−1 CQDs for 3 days. In this study, the surface chemistry related to antibacterial activity and low cytotoxicity give the chance in designing and developing better CQDs for biofilm infection.

We propose a novel, to the best of our knowledge, 1 × 5 broadband power splitter based on the photonic crystal. The Powell algorithm is used to reverse-design the proposed broadband power splitter. The results show that the transmittance of each output port of the broadband photonic crystal power splitter can be adjusted by changing the radii and offsets of the dielectric rods at the junction area of each waveguide. According to the target splitting ratio, the reverse design of the structural parameters using the Powell algorithm significantly improves the optimization efficiency and splitting performance of the broadband power splitter. The designed power splitters have a wide working bandwidth of 1525–1565 nm, a flexible and designable power splitting ratio, excellent splitting performance, and a compact size, which have great application prospects in all-optical communication networks, high-density photon integration, and other fields.

This study investigated the effects of adipose-derived stem cell sheets on a rat model of detrusor underactivity.Adipose-derived stem cell sheets were prepared from the subcutaneous adipose tissue of male Lewis rats. Female Lewis rats were assigned into four groups: control, sham operation, cryo-injury, and cryo-injury+sheet (n = 8 per group). Rats in the cryo-injury+sheet group were implanted with ASC sheets 3 days after cryo-injury induction and underwent cystometry 7 days later. Subsequently, reverse transcription-polymerase chain reaction (RT-PCR) and histopathological examinations were performed. Cell sheets expressing the green fluorescent protein were prepared and transplanted to confirm the viability and differentiation of the sheets. Fluorescence was confirmed using a fluorescence stereomicroscope on days 3, 7, 14, 21, and 28 after sheet implantation, and tissue immunostaining was performed.Cystometry showed that sheet implantation improved the maximum intravesical pressure (P = 0.009) and the residual urine volume (P = 0.011). Furthermore, RT-PCR indicated that the mRNA levels of the angiogenic factors vascular endothelial growth factor and hepatocyte growth factor were significantly higher in the cryo-injury+sheet group than in the cryo-injury group (P = 0.045, P = 0.037, respectively). Histologically, sheet implantation resulted in an improvement in inflammation and increased the number of blood vessels. Green fluorescent protein-positive cells fused with von Willebrand factor-positive cells and differentiated into blood vessels 7 days after sheet implantation.Adipose-derived stem cell sheets transplanted into the bladder of cryo-injured rats differentiated into blood vessels and restored bladder contractile function 7 days after transplantation.

Among the several independent risk factors for atrial fibrillation (AF), hyperuricemia has been widely accepted as associated with the incidence of paroxysmal or persistent AF, as well as with the risk of AF in patients undergoing cardiovascular surgery. The electrophysiological mechanism of AF involves electrical remodeling of the arrhythmogenic substrate and abnormal automaticity as trigger. Both electrical and structural remodeling mediated by oxidative stress derived from either xanthine oxidoreductase (XOR), soluble uric acid (UA) or monosodium urate (MSU) crystals might be plausible explanations for the association of AF with hyperuricemia. XOR generates reactive oxygen species (ROS) that lead to atrial structural remodeling via inflammation. Soluble UA accumulates intracellularly through UA transporters (UAT), shortening the atrial action potential via enhanced expression and activity of Kv1.5 channel proteins. Intracellular accumulation of soluble UA generates ROS in atrial myocytes via nicotinamide adenine dinucleotide phosphate oxidase, which phosphorylates ERK/Akt and heat shock factor 1 (HSF1), thereby increasing transcription and translation of Hsp70, which stabilizes Kv1.5. In macrophages, MSU activates the NLRP3 inflammasome and proteolytic processing mediated by caspase-1 with enhanced interleukin (IL)-1β and IL-18 secretion. Use of an XOR inhibitor, antioxidants, a UAT inhibitor such as a uricosuric agent, and an NLRP3 inflammasome inhibitor, might become a potential strategy to reduce the risk of hyperuricemia-induced AF, and control serum UA level.

In this paper, we proposed a novel ultra-flat broadband microwave frequency comb (MFC) generation based on optical frequency comb (OFC) with a multiple-quantum-well electro-absorption modulator (MQW-EAM) in critical state. The scheme is simple and easy to adjust. The performance of the MFC generation scheme is investigated using software Optisystem. The results show that the comb spacing of MFC can be adjusted from 5 to 20 GHz by changing RF signal’s frequency and the MFC is almost independent on the linewidth of the tunable laser diode. The performance of the MFC can be improved by reasonably increasing the voltage of the RF, the small-signal gain of the Erbium-doped fiber amplifier (EDFA) and the responsivity of the photodetector. The MFC generated by this scheme has 300 GHz effective bandwidth with 15 comb lines, whose power variation is 0.02 dB, when the components’ parameters in the Optisystem are set as follows: the power of tunable laser diode (TLD) is 0 dBm, the wavelength is 1552.52 nm, and linewidth is 1 MHz; RF signal’s frequency is 20 GHz and the voltage is 10 V; the reverse bias voltage of MQW-EAM is 6.92 V; the small-signal gain of the EDFA is 40 dB; the responsivity of the photodetector (PD) is 1 A/W.

Background Intracellular uric acid is known to increase the protein level and channel current of atrial Kv1.5; however, mechanisms of the uric acid-induced enhancement of Kv1.5 expression remain unclear. Methods and Results: The effects of uric acid on mRNA and protein levels of Kv1.5, as well as those of Akt, HSF1 and Hsp70, in HL-1 cardiomyocytes were studied by using qRT-PCR and Western blotting. The uptake of uric acid was measured using radio-labeled uric acid. The Kv1.5-mediated channel current was also measured by using patch clamp techniques. Uric acid up-taken by HL-1 cells significantly increased the level of Kv1.5 proteins in a concentration-dependent manner, with this increase abolished by an uric acid transporter inhibitor. Uric acid slowed degradation of Kv1.5 proteins without altering its mRNA level. Uric acid enhanced phosphorylation of Akt and HSF1, and thereby increased both transcription and translation of Hsp70; these effects were abolished by a PI3K inhibitor. Hsp70 knockdown abolished the uric acid-induced increases of Kv1.5 proteins and channel currents. Conclusions Intracellular uric acid could stabilize Kv1.5 proteins through phosphorylation of Akt and HSF1 leading to enhanced expression of Hsp70.

A four-channel routing switch based on a square lattice 2-D silicon photonic crystal with four point defects and five line defects is proposed. In this scheme, the properties that magnetic permeability of the ferrite material changing with external magnetic field and the coupling characteristics between line defect and point defect are used. The proposed device is theoretically analyzed and numerically investigated by using plane wave method (PWM) and finite-difference time-domain (FDTD). The results show that the four-channel routing switch can be realized by applying different intensity of magnetic fields. The extinction ratios of the four channels are all more than 13dB, the transmittance rates are higher than 93%, the insertion losses are lower than 0.288dB, and steady times are less than 50 ps. The proposed THz routing switch has potential applications in terahertz wave integrated circuit fields in the future.

We propose a scheme of optical frequency comb (OFC) generation based on cascaded Mach–Zehnder modulator (MZM) and electroabsorption modulator (EAM) with Gaussian-shaped pulse signal. In the proposed scheme, the MZM and the nonzero-chirp EAM are driven by a periodic Gaussian-shaped pulse signal that is controlled with binary sequences, and then an OFC is output from EAM. The theoretical model of the OFC generation is developed by improving the theory of electroabsorption modulation. The performance of OFC is investigated. The results show that the frequency spacing of the OFC can be tuned in the range of 1 to 10 GHz, the number of effective comb-lines is 35, and the power variation of comb-lines is 2.14 dB.

Cell-based therapy using adipose-derived stem cells (ADSCs) has emerged as a novel therapeutic approach to treat heart failure after myocardial infarction (MI). The purpose of this study was to determine whether inhibition of α1-adrenergic receptors (α1-ARs) in ADSCs attenuates ADSC sheet-induced improvements in cardiac functions and inhibition of remodeling after MI. ADSCs were isolated from fat tissues of Lewis rats. In in vitro studies using cultured ADSCs, we determined the mRNA levels of vascular endothelial growth factor (VEGF)-A and α1-AR under normoxia or hypoxia and the effects of norepinephrine and an α1-blocker, doxazosin, on the mRNA levels of angiogenic factors. Hypoxia increased α1-AR and VEGF mRNA levels in ADSCs. Norepinephrine further increased VEGF mRNA expression under hypoxia; this effect was abolished by doxazosin. Tube formation of human umbilical vein endothelial cells was promoted by conditioned media of ADSCs treated with the α1 stimulant phenylephrine under hypoxia but not by those of ADSCs pretreated with phenylephrine plus doxazosin. In in vivo studies using rats with MI, transplanted ADSC sheets improved cardiac functions, facilitated neovascularization, and suppressed fibrosis after MI. These effects were abolished by doxazosin treatment. Pathway analysis from RNA sequencing data predicted significant upregulation of α1-AR mRNA expression in transplanted ADSC sheets and the involvement of α1-ARs in angiogenesis through VEGF. In conclusion, doxazosin abolished the beneficial effects of ADSC sheets on rat MI hearts as well as the enhancing effect of norepinephrine on VEGF expression in ADSCs, indicating that ADSC sheets promote angiogenesis and prevent cardiac dysfunction and remodeling after MI via their α1-ARs.

Optical frequency comb (OFC) has important applications in measurement, communication, military and other fields. Usually, OFC needs to be designed according to different applications. However, the existing methods to design the operating parameters of the OFC generators are time-consuming, inefficient, and difficult to achieve optimal results. In this paper, a novel method of inversely designing OFC using deep learning, which is real-time and can improve the performance of the generated OFC, is proposed and applied to an OFC generator based on a single dual-drive Mach–Zehnder modulator. In this method, according to the required target OFC, the trained neural network can be used to inversely design the corresponding parameters. Using this inverse design method, the generated OFC not only is highly consistent with the target OFC, but also has the programmability of comb-line number, comb-line power, side mode suppression ratio, and comb spacing. Moreover, the proposed method can be utilized for more complicated OFC generator, and is an inspiration for efficient design of OFC.

We know that compressive sensing can establish stable sparse recovery results from highly undersampled data under a restricted isometry property condition. In reality, however, numerous problems are coherent, and vast majority conventional methods might work not so well. Recently, it was shown that using the difference between $\ell_1$- and $\ell_2$-norm as a regularization always has superior performance. In this paper, we propose an adaptive $\ell_p$-$\ell_{1-2}$ model where the $\ell_p$-norm with $p\geq 1$ measures the data fidelity and the $\ell_{1-2}$-term measures the sparsity. This proposed model has the ability to deal with different types of noises and extract the sparse property even under high coherent condition. We use a proximal majorization-minimization technique to handle the nonconvex regularization term and then employ a semismooth Newton method to solve the corresponding convex relaxation subproblem. We prove that the sequence generated by the semismooth Newton method admits fast local convergence rate to the subproblem under some technical assumptions. Finally, we do some numerical experiments to demonstrate the superiority of the proposed model and the progressiveness of the proposed algorithm.

The increasing popularity of blockchain-based cryptocurrencies has revitalized the search for efficient Byzantine fault-tolerant (BFT) protocols. Many existing BFT protocols can achieve good performance in fault-free cases but suffer severe performance degradation when faults occur. This is also a problem with DiemBFT. To mitigate performance attacks in DiemBFT, we present an improved BFT protocol with optimal liveness called the Golden Snitch. The core idea is to introduce unbiased randomness in leader selection and improve the voting mechanism to protect honest leaders from being dragged down by the previous leader. The performance of the Golden Snitch is evaluated through experiments, turning out it outperforms DiemBFT in the presence of faults.

Aiming at the difficulty of integrated repair of osteochondral tissue, we designed a hybrid hydrogel scaffold that mimicked the microenvironment of osteochondral niches. Besides, the nano-hydroxyapatite (nHAP) was specially introduced into the hydrogel for its natural ability to promote bone regeneration. The hydrogel also exhibited good toughness (7500 KJ/m3), strength (1000 kPa), viscoelasticity, and in vitro cell experiments showed that hydrogels had quite good cytocompatibility (near 100 % viability). The results of the three-dimensional (3D) cell culture also proved that the survival rate of the cells in the hybrid hydrogels doped with nHAP and dispersion were the highest. In vitro RT-qPCR experiments proved that after being cultured in hydrogel scaffolds doped with nHAP, bone mesenchymal stem cells (BMSCs) could express genes related to osteoblasts and chondrocytes. As a result, this hydrogel provides a general for developing alternative materials applicable for stem cells differentiation and even osteochondral tissue engineering.

Three experiments were conducted to investigate the effects of dietary crude protein (CP) level and N-carbamylglutamate (NCG) supplementation on apparent total tract digestibility (ATTD) and ileal digestibility of nutrients and digestive enzyme activity of jejunum in growing pigs. In experiment 1, 10 Duroc × Landrace × Yorkshire barrows (initial BW: 48.7 kg) were allotted to a three-period switchback design with five experimental diets and two replicate pigs per diet in each period. Diets were categorized as high CP (HP, 18% CP), moderate low CP (MLP, 15% CP), very low CP (VLP, 12% CP), and MLP and VLP with 0.1% NCG supplementation. Feces and urine were collected from day 6 to day 11 after a 5-d adaptation period. The DE, ME, and ATTD of GE, OM, CP, NDF, ADF, and P decreased (P < 0.01) with a reduction of dietary CP, but no effect of dietary treatments on pig daily N retention was detected. The NCG supplementation increased (P < 0.01) DE and ATTD of ADF of the VLP diet. In experiment 2, 10 jejunal-cannulated Duroc × Landrace × Yorkshire barrows (initial BW: 44.5 kg) were fed five diets for three periods as experiment 1. Jejunal fluid was collected on days 6 and 8 after a 5-d adaptation period. The digestive enzymes activity was not affected by dietary CP level, except for α-amylase, for which there was a decrease (P < 0.01) in pigs fed VLP diets compared to HP and MLP diets. In experiment 3, 12 ileal-cannulated Duroc × Landrace × Yorkshire barrows (initial BW: 46.7 kg) were allotted to a three-period switchback design with six diets and two replicate pigs per diet in each period. The six experimental diets consisted of five experimental diets as experiment 1 and one N-free diet. Ileal digesta was collected from day 6 to day 8 after a 5-d adaptation period. Results indicated that apparent ileal digestibility (AID) of CP and P and ileal digestibility of Arg, His, Ile, Leu, Phe, and all dispensable AA, except Pro, decreased (P < 0.01) in pigs fed VLP diet compared to HP and MLP diets, but AID of GE, OM, EE, NDF, and ADF were not affected. The supplementation of NCG in the VLP diet increased (P < 0.01) the AID of CP and ileal digestibility of Arg, His, Leu, Phe, Val, Ser, and Tyr. In conclusion, reducing dietary CP level decreased nutrient digestibility, but improved the efficiency of dietary N utilization and reduced N emission. Moderate reduction of dietary CP level had a minimal effect on nutrient digestibility and digestive enzyme activity. Additionally, NCG supplementation plays a beneficial effect on nutrient digestion only if the dietary CP level is extremely lowered.

A novel microwave photonic single-passband filter with an improved out-of-band rejection and an extended frequency tuning range is proposed and demonstrated, which is achieved by using birefringence of a phase-shifted fiber Bragg grating (PSFBG). If two polarization states of two phase-modulated signals with opposite modulation indices from a polarization modulator (PolM) are controlled to be aligned with two principal axes of the polarization-maintaining PSFBG (PM-PSFBG), respectively, two single-passband or two single-notch filters with slight frequency interval are achieved, depending on the position of the optical carrier relative to the notches of the PM-PSFBG. Through subtraction of the two filters, the out-of-band rejection is significantly improved, and the frequency tuning range is greatly extended. Analytical simulations are performed using an established theoretical model, which are verified by an experiment.

The objective of this experiment was to determine the effects of increased fiber content in diets on heat production (HP) and NE:ME ratio and to determine the NE content and NE:ME ratio of full-fat rice bran (FFRB), defatted rice bran (DFRB), corn gluten feed (CGF), and corn germ meal (CGM) fed to growing barrows using indirect calorimetry (IC). Thirty growing barrows (28.5 ± 2.4 kg BW) were allotted in a completely randomized design to 5 dietary treatments that included a corn–soybean meal basal diet and 4 experimental diets with a constant ratio of corn and soybean meal (difference method) containing 30% FFRB, DFRB, CGF, and CGF. Pigs were housed in individual metabolism crates for 20 d including 14-d adaptation to the diet and 6 d to determine the HP and total collection of feces and urine in respiration chambers. Pigs were fed their respective diets at 550 kcal ME·kg BW(0.60−1)·d(−1) on the basis of BW measured on days 0, 7, and 14. The apparent total tract digestibility (ATTD) of DM, GE, and OM were greater (P < 0.01) in pigs fed the basal diet. The ATTD of DM, GE, and OM in pigs fed the DFRB diet were lesser (P < 0.01) when compared with those fed the basal and FFRB diets. The ATTD of ether extract (EE) in pigs fed the FFRB diet was greater (P < 0.01) compared with those fed basal, DFRB, CGF, and CGM diets. The HP adjusted for the same ME intake was greater (P < 0.01) in pigs fed the DFRB, CGF, and CGM diets compared with those fed basal and FFRB diets. The NE:ME ratio in pigs fed the FFRB diet was greater (P < 0.01) when compared with those fed the DFRB, CGF, and CGM diets. The NE content of FFRB, DFRB, CGF, and CGM determined using the IC method were 2,952, 1,100, 1,747, and 2,079 kcal/kg DM, respectively. The NE content of FFRB, CGF, and CGM determined using the IC method were 3.5%, 3.8%, and 1.8% greater, respectively, than the predicted values, whereas NE content of DFRB determined using the IC method was 2.1% lower than the predicted values. In conclusion, pigs fed the fiber-rich ingredients had greater HP and lower nutrient digestibility. However, pigs fed FFRB diets containing greater fat content had a lower heat increment and, therefore, higher utilization efficiency. The NE:ME ratio ranged from 71.6% to 82.4%. The NE of FFRB, DFRB, CGF, and CGM determined using the IC method were 2,952, 1,100, 1,747, and 2,079 kcal/kg DM, respectively.

The human ether-a-go-go-related gene (hERG) encodes the α subunit of a rapidly activating delayed-rectifier potassium (IKr) channel. Mutations of the hERG cause long QT syndrome type 2 (LQT2). Acetylation of lysine residues occurs in a subset of non-histone proteins and this modification is controlled by both histone acetyltransferases and deacetylases (HDACs). The aim of this study was to clarify effects of HDAC(s) on wild-type (WT) and mutant hERG proteins. WThERG and two trafficking-defective mutants (G601S and R752W) were transiently expressed in HEK293 cells, which were treated with a pan-HDAC inhibitor Trichostatin A (TSA) or an isoform-selective HDAC6 inhibitor Tubastatin A (TBA). Both TSA and TBA increased protein levels of WThERG and induced expression of mature forms of the two mutants. Immunoprecipitation showed an interaction between HDAC6 and immature forms of hERG. Coexpression of HDAC6 decreased acetylation and, reciprocally, increased ubiquitination of hERG, resulting in its decreased expression. siRNA against HDAC6, as well as TBA, exerted opposite effects. Immunochemistry revealed that HDAC6 knockdown increased expression of the WThERG and two mutants both in the endoplasmic reticulum and on the cell surface. Electrophysiology showed that HDAC6 knockdown or TBA treatment increased the hERG channel current corresponding to the rapidly activating delayed-rectifier potassium current (IKr) in HEK293 cells stably expressing the WT or mutants. Three lysine residues (K116, K495 and K757) of hERG were predicted to be acetylated. Substitution of these lysine residues with arginine eliminated HDAC6 effects. In HL-1 mouse cardiomyocytes, TBA enhanced endogenous ERG expression, increased IKr, and shortened action potential duration. These results indicate that hERG is a substrate of HDAC6. HDAC6 inhibition induced acetylation of hERG which counteracted ubiquitination leading its stabilization. HDAC6 inhibition may be a novel therapeutic option for LQT2.

Sparse regression plays a very important role in statistics, machine learning, image and signal processing. In this paper, we consider a high-dimensional linear inverse problem with \begin{document}$ \ell^0 $\end{document}-\begin{document}$ \ell^2 $\end{document} penalty to stably reconstruct the sparse signals. Based on the sufficient and necessary condition of the coordinate-wise minimizers, we design a smoothing Newton method with continuation strategy on the regularization parameter. We prove the global convergence of the proposed algorithm. Several numerical examples are provided, and the comparisons with the state-of-the-art algorithms verify the effectiveness and superiority of the proposed method.

Polyoxometalate H5PMo10V2O40/SiO2 (HPMoV/SiO2) nanofibers with a mesoporous structure were fabricated through combined electrospinning and surfactant-directing pore formation technique. These heterogeneous nanofibers were characterized by a wide variety of techniques to show mesoporous structure, high surface area, excellent stability, nanofiber morphology, highly efficient activity and selectivity towards the oxidation of starch and 5-hydroxymethylfurfural (5-HMF) either by H2O2 or by oxygen. POM/SiO2 nanofibers were suitable for oxidation of starch by H2O2 and oxidation of 5-HMF by O2 with high efficiency of 0.58 mol per 100 g carboxyl content and 89.2% yield of DFF; these results are comparable with those obtained using homogeneous H5PMo10V2O40 and HPMoV/SiO2 without nanofiber morphology. Furthermore, HPMoV/SiO2 nanofibers could be easily recycled and reused at least ten times with no significant loss of catalytic activity due to their nanofiber morphology.

A novel scheme of blind frequency resolved optical gating (Blind-FROG) measurement for optical arbitrary waveforms, based on sum frequency effect in periodically poled lithium niobate (PPLN) optical waveguide, is proposed. The proposed scheme is very simple, low cost and good noise performance. Furthermore, it can simultaneously measure two arbitrary, potentially very complex pulses. Through theoretical analysis and numerical simulation, the performance of the scheme is studied. The results show that, the accuracy of measurement degrades with the increase of the two optical arbitrary waveforms complexity; Nevertheless, the smaller the polarization periodic mismatching is, the higher the measurement accuracy becomes; And the accuracy of the complete measurement system also can be improved by increasing the number of iterations.

Background Pilsicainide, classified as a relatively selective Na + channel blocker, also has an inhibitory action on the rapidly-activating delayed-rectifier K + current ( I Kr ) through human ether-a-go-go-related gene (hERG) channels. We studied the effects of chronic exposure to pilsicainide on the expression of wild-type (WT) hERG proteins and WT-hERG channel currents, as well as on the expression of mutant hERG proteins, in a heterologous expression system. Methods HEK293 cells stably expressing WT or mutant hERG proteins were subjected to Western blotting, immunofluorescence microscopy and patch-clamp experiments. Results Acute exposure to pilsicainide at 0.03–10μM influenced neither the expression of WT-hERG proteins nor WT-hERG channel currents. Chronic treatment with 0.03–10μM pilsicainide for 48h, however, increased the expression of WT-hERG proteins and channel currents in a concentration-dependent manner. Chronic treatment with 3μM pilsicainide for 48h delayed degradation of WT-hERG proteins and increased the channels expressed on the plasma membrane. A cell membrane-impermeant pilsicainide derivative did not influence the expression of WT-hERG, indicating that pilsicainide stabilized the protein inside the cell. Pilsicainide did not influence phosphorylation of Akt (protein kinase B) or expression of heat shock protein families such as HSF-1, hsp70 and hsp90. E4031, a chemical chaperone for hERG, abolished the pilsicainide effect on hERG. Chronic treatment with pilsicainide could also increase the protein expression of trafficking-defective mutant hERG, G601S and R752W. Conclusions Pilsicainide penetrates the plasma membrane, stabilizes WT-hERG proteins by acting as a chemical chaperone, and enhances WT-hERG channel currents. This mechanism could also be applicable to modulations of certain mutant-hERG proteins.

This study proposes a novel scheme of a crosscorrelation frequency-resolved optical gating (X-FROG) measurement for an optical arbitrary waveform (OAW) based on the sum frequency generation (SFG) effect of a periodically poled lithium niobate (PPLN) waveguide. Based on the SFG effect and combined with the principal component generalized projects algorithm on a matrix, the theory model of the scheme is established. Using Matlab, the proposed OAW measurement X-FROG scheme using the PPLN waveguide is simulated and studied. Simulation results show that a rectangular pulse is a suitable gate pulse because of its low errors. Moreover, the increased complexity of OAW and phase mismatch decrease measurement accuracy.

Carbon quantum dots derived from gentamicin sulfate are directly developed by facile calcination at different temperatures. A promising nanomaterial, CQD180, even shows a much superior antibacterial activity compared with the antibiotic counterpart and low drug resistance, by preserving the active structure of the starting materials and providing an additional antibacterial mode arising from the positive charge surface and induced reactive oxygen species simultaneously. Moreover, CQD180 can effectively disrupt mature Staphylococcus aureus biofilm and selectively eliminate bacteria over mammalian cells.

Many blockchain researches focus on the privacy protection. However, criminals can leverage strong privacy protection of the blockchain to do illegal crimes (such as ransomware) without being punished. These crimes have caused huge losses to society and users. Implementing identity tracing is an important step in dealing with issues arising from privacy protection. In this paper, we propose a blockchain traceable scheme with oversight function (BTSOF). The design of BTSOF builds on SkyEye (Tianjun Ma et al., Cryptology ePrint Archive 2020). In BTSOF, the regulator must obtain the consent of the committee to enable tracing. Moreover, we construct a non-interactive verifiable multi-secret sharing scheme (VMSS scheme) and leverage the VMSS scheme to design a distributed multi-key generation (DMKG) protocol for the Cramer-Shoup public key encryption scheme. The DMKG protocol is used in the design of BTSOF. We provide the security definition and security proof of the VMSS scheme and DMKG protocol.

This study investigated the effects of modulation of the amino acid profile on growth performance and gut health in weaned pigs fed an antibiotic-free, low-protein diet. In experiment 1, 5 treatments were included: a control diet with antibiotics; a low-protein diet with antibiotics; a low-protein diet without antibiotics (LP); a LP diet with 10% more dietary essential amino acids (LP110); and an LP110 diet with 12% more dietary Met + Cys, Thr and Trp. The intestinal digestive enzyme activity and morphology were improved with the increase in dietary essential amino acid levels, while the growth performance was decreased, indicating that the dietary amino acid level was too high. In experiment 2, all 5 treatments of experiment 1 were included, plus a LP diet with 5% more dietary essential amino acids (LP105) and an LP105 diet with 6% more dietary Met + Cys, Thr and Trp. The LP105 treatment showed optimal feed efficiency, a reduced plasma endotoxin concentration, and an increased fecal lactate concentration and increased abundances of Prevotellaceae and Roseburia bacteria. Our results demonstrate that the optimal amino acid profile in an antibiotic-free, low-protein diet can efficiently improve growth performance and gut health and modulate the fecal microbial structure in weaned pigs.

A tunable dual-frequency optoelectronic oscillator (OEO) based on a tunable dual-passband microwave photonic filter (DPMPF) is proposed and demonstrated. The DPMPF is based on phase-to-intensity modulation (PM-IM) conversion and stimulated Brillouin scattering (SBS). Two pump lightwaves are generated through carrier-suppression doublesideband modulation (CS-DSB) in a Mach-Zehnder modulator (MZM) to generate two SBS gain regions in a single mode fiber. Two SBS gain regions act on the phase modulation signal to achieve a dual-passband filter. Through simply varying the frequency of the radio frequency (RF) signal used for CS-DSB, the two central frequencies of the OEO can be tuned with the frequency interval kept constant. In addition, adjusting the frequency of tunable optical source launched to the MZM, the frequency interval of the OEO can be tuned. In the experiment, the dual-frequency OEOs with frequency intervals of 0.2 GHz and 1.2 GHz are achieved, respectively. The frequency tuning range from 3 to 8 GHz is demonstrated. The dual-frequency OEO with the frequency interval tuned is also achieved.
