It has been previously shown that zinc-finger transcription factor Gata3 has dynamic expression within the inner ear throughout embryonic development and is essential for cochlear neurosensory development. However, the temporal window to which Gata3 is required for the formation of the cochlear neurosensory epithelia remains unclear. To investigate the role of Gata3 on cochlear neurosensory development in the late prosensory stages, we used the Sox2-creERT2 mouse line to target and conditionally delete Gata3 at E11.5 before the cells have fully committed to a neurosensory fate. While the inner ears of Sox2-creERT2: Gata3 f/f mice appear morphologically normal, the sensory cells in the organ of Corti are partially lost and disorganized in a basal to apical gradient with the apex demonstrating the more severe phenotype. Additionally, spiral ganglion neurons display aberrant peripheral projections, such as increased distances between radial bundles and disorganization upon reaching the organ of Corti. Furthermore, heterozygous Sox2-creERT2: Gata3 f/+ mice show a reduced phenotype in comparison to the homozygous mutant, supporting the concept that Gata3 is not only required for proper formation at the later proneurosensory stage, but also that a specific level of Gata3 is required. Therefore, our studies confirm that Gata3 plays a time-sensitive and dose-dependent role in the development of sensory cells in the late proneurosensory stages.

Proper development of the O2-sensing system is essential for survival. Here, we characterized the development of the O2-sensing system in the mangrove rivulus (Kryptolebias marmoratus), an amphibious fish that transitions between hypoxic aquatic environments and O2-rich terrestrial environments. We found that NECs formed in the gills and skin of K. marmoratus during embryonic development and that both NEC populations are retained from the embryonic stage to adulthood. We also found that the hyperventilatory response to acute hypoxia was present in embryonic K. marmoratus, indicating that functional O2-sensing pathways are formed during embryonic development. We then exposed embryos to aquatic normoxia, aquatic hyperoxia, aquatic hypoxia, or terrestrial conditions for the first 30 days of embryonic development and tested the hypothesis that environmental O2 availability during embryonic development modulates the development of the O2-sensing system in amphibious fishes. Surprisingly, we found that O2 availability during embryonic development had little impact on the density and morphology of NECs in the gills and skin of K. marmoratus. Collectively, our results demonstrate that, unlike the only other species of fish in which NEC development has been studied to date (i.e., zebrafish), NEC development in K. marmoratus is largely unaffected by environmental O2 levels during the embryonic stage, indicating that there is interspecies variation in O2-induced plasticity in the O2-sensing system of fishes.

Heat-stressed dairy cows on pasture will compete for resources that aid cooling, but it is not known how heat stress affects the competition for water by indoor-housed cows. The aim of this observational study was to evaluate how heat stress affects the behavior of indoor-housed cows at the drinker at both group and cow levels. For 3 wk after calving, cows were housed in a dynamic group of 20 animals in a pen with 12 electronic feed bins, 2 electronic water bins, and 24 freestalls. A total of 69 lactating Holstein dairy cows were enrolled over the 59-d study. The electronic water bins recorded time spent at the drinker, frequency of visits, water intake, and competitive events for 24 h/d. Competitive events were quantified using the number of replacements (recorded when there was a ≤29-s interval between 2 cows sequentially visiting the same drinker). The number of replacements a cow was involved in was used to determine her level of competitive success at the drinker (low, medium, high). The temperature–humidity index (THI) was recorded by the local weather station, and moving averages for daily maximum THI over a 3-d period were calculated. For the analysis of time spent at the drinker, frequency of visits, and water intake, the measures from all cows were averaged to create 1 observation per day, and the number of replacements at the drinker was summed. A linear regression was performed to determine the relationship between THI and group-level drinking behavior. At the cow level, a repeated measures mixed model, with fixed effects of level of competitive success, milk yield, and 3-d maximum THI and a first-order autoregressive covariance structure, was used to determine how increasing THI affects the drinking behavior of individual cows based on their level of competitive success. Feed intake was included as a fixed effect in the water intake model. We found that, with increasing THI, cows drank more water, spent more time at the drinker, made more visits to the drinker, and engaged in more competitive events at the drinker. In exploratory analysis, we found that cows with low competitive success at the drinker shifted their drinking behavior to avoid the drinker at the hottest and most competitive time of day. These results indicate that behavior can be used to indicate when cows feel hot. These measures may be of practical value in deciding when to provide cooling, especially for farms where attendance at the drinker can be monitored electronically.

Conspecific negative density dependence (CNDD) promotes tree species diversity by reducing recruitment near conspecific adults due to biotic feedbacks from herbivores, pathogens, or competitors. While this process is well-described in tropical forests, tests of temperate tree species range from strong positive to strong negative density dependence. To explain this, several studies have suggested that tree species traits may help predict the strength and direction of density dependence: for example, ectomycorrhizal-associated tree species typically exhibit either positive or weaker negative conspecific density dependence. More generally, the strength of density dependence may be predictably related to other species-specific ecological attributes such as shade tolerance, or the relative local abundance of a species. To test the strength of density dependence and whether it affects seedling community diversity in a temperate forest, we tracked the survival of seedlings of three ectomycorrhizal-associated species experimentally planted beneath conspecific and heterospecific adults on the Prospect Hill tract of the Harvard Forest, in Massachusetts, USA. Experimental seedling survival was always lower under conspecific adults, which increased seedling community diversity in one of six treatments. We compared these results to evidence of CNDD from observed sapling survival patterns of 28 species over approximately 8 years in an adjacent 35-ha forest plot. We tested whether species-specific estimates of CNDD were associated with mycorrhizal association, shade tolerance, and local abundance. We found evidence of significant, negative conspecific density dependence (CNDD) in 23 of 28 species, and positive conspecific density dependence in two species. Contrary to our expectations, ectomycorrhizal-associated species generally exhibited stronger (e.g., more negative) CNDD than arbuscular mycorrhizal-associated species. CNDD was also stronger in more shade-tolerant species but was not associated with local abundance. Conspecific adult trees often have a negative influence on seedling survival in temperate forests, particularly for tree species with certain traits. Here we found strong experimental and observational evidence that ectomycorrhizal-associating species consistently exhibit CNDD. Moreover, similarities in the relative strength of density dependence from experiments and observations of sapling mortality suggest a mechanistic link between negative effects of conspecific adults on seedling and sapling survival and local tree species distributions.

We present a 3-D finite element (FE) model of the chinchilla's inner ear consisting of the entire cochlea structure and the vestibular system. The reaction of the basilar membrane to the head rotation and the reaction of ampulla to the stapes movement were investigated. These results demonstrate the existence of hearing-vestibular system interaction. They provide an explanation to the clinical finding on the coexistence between hearing loss and equilibration dysfunction. It is a preliminary, yet critical step toward the development of a comprehensive FE model of an entire ear for mechano-acoustic analysis.

The ability to tolerate environmental change may decline as fishes age. We tested the hypothesis that ageing influences the scope for phenotypic flexibility in the mangrove rivulus (Kryptolebias marmoratus), an amphibious fish that transitions between two vastly different environments, water and land. We found that older fish (4–6 years old) exhibited marked signs of ageing; older fish were reproductively senescent, had reduced fin regenerative capacity and body condition, and exhibited atrophy of both oxidative and glycolytic muscle fibers relative to younger adult fish (1–2 years old). However, age did not affect routine O2 consumption. We then acclimated adult fish (1–6 years) to water (control) or air for 10 days to assess the scope for phenotypic flexibility in response to terrestrial exposure. In support of our hypothesis, we found that older air-acclimated fish had a diminished scope for gill remodeling relative to younger fish. We also found that older fish exhibited poorer terrestrial locomotor performance relative to younger adult fish, particularly when acclimated to air. Our results indicate that ageing diminishes skeletal muscle integrity and locomotor performance of amphibious fishes, and may, therefore, impair terrestrial foraging ability, predator avoidance, or dispersal across the terrestrial environment. Remarkably, older fish voluntarily left water to a similar degree as younger fish despite the age-related deterioration of traits important for terrestrial life.

The objective of this study was to determine whether data from an electronic drinking system that tracks individual animal attendance with ear tag transponders could be used to detect social competition between dairy cows at the drinker. Specifically, we sought to identify the interval between one cow leaving the drinker and another cow taking her place that most accurately identified competitive replacements (when physical contact initiated by one cow causes the other to entirely remove her head from the drinker and the initiator subsequently places her head in the same drinker). The optimal interval to accurately identify replacements at the drinker was determined using a receiver operating characteristic curve and by evaluating the sensitivity (Se), specificity (Sp), and confusion matrix values (true positives, true negatives, false positives, false negatives) of different thresholds identified by 3 metrics: the Youden index, the point closest to (0,1) on the curve, and the point where Se roughly equals Sp. Lactating Holstein dairy cows (n = 20) were monitored for 4 consecutive 24-h periods by video recording and with an electronic drinking system. Two periods were used to identify the optimal interval (the baseline set), and the other 2 periods were used to validate the interval (the validation set). The occurrence of a replacement identified by video was paired with the interval between drinking events of 2 cows at the same electronic drinker to identify the interval that best predicted replacement events. Based on the low prevalence of replacement events compared with nonreplacement events, the interval with the lowest number of false positives was considered optimal. Of the 3 potential metrics, the point on the receiver operating characteristic curve where Se and Sp were roughly equal yielded the fewest false positives. The optimal interval to identify replacements at the drinker was ≤29 s, with 82% Se and 83% Sp in the baseline set. This interval was found to have 85% Se and 89% Sp when applied to the validation set. These results indicate that social competition between dairy cows at the drinker can be accurately measured with an electronic drinking system.

Proper development of the O

The interaction between developmental plasticity and the capacity for reversible acclimation (phenotypic flexibility) is poorly understood, particularly in organisms exposed to fluctuating environments. We used an amphibious killifish (Kryptolebias marmoratus) to test the hypotheses that organisms reared in fluctuating environments (i) will make no developmental changes to suit any one environment because fixing traits to suit one environment could be maladaptive for another, and (ii) will be highly phenotypically flexible as adults because their early life experiences predict high environmental variability in the future. We reared fish under constant (water) or fluctuating (water-air) environments until adulthood and assessed a suite of traits along the oxygen (O2) cascade (e.g., neuroepithelial cell density and size, cutaneous capillarity, gill morphology, ventricle size, red muscle morphometrics, terrestrial locomotor performance). To evaluate the capacity for phenotypic flexibility, a subset of adult fish from each rearing condition were then air-exposed for 14 days before the same traits were measured. In support of the developmental plasticity hypothesis, traits involved with O2 sensing and uptake were largely unaffected by water-air fluctuations during early life, but we found marked developmental changes in traits related to O2 transport, utilization, and locomotor performance. In contrast, we found no evidence supporting the phenotypic flexibility hypothesis. Adult fish from both rearing conditions exhibited the same degree of phenotypic flexibility in various O2 sensing- and uptake-related traits. In other cases, water-air fluctuations attenuated adult phenotypic flexibility despite the fact that phenotypic flexibility is hypothesized to be favoured when environments fluctuate. Overall, we conclude that exposure to environmental fluctuations during development in K. marmoratus can dramatically alter the constitutive adult phenotype, as well as diminish the scope for phenotypic flexibility in later life.

Belgium has seen more foreign fighters leave for the ISIS ‘caliphate’ than any other western European country in per capita figures. But among Belgian foreign fighters, as well as among Belgian terrorists who did not travel abroad, a stunning majority have been Belgian-Moroccan. What factors can explain this phenomenon? Both groups live in relatively similar socio-economic settings, experience stigmatization and discrimination, and both are, relative to broader Belgian society, poorly integrated. However, in investigating this question, an interesting caveat emerges vis-a-vis the ‘integration deficit’ theory: Belgian-Turks, who are found to be comparatively less integrated than the Belgian-Moroccans, are underrepresented in extremist milieus. Recent investments in earthquake engineering research have produced an array of experimental equipment and testing capabilities worldwide. Laboratories are often equipped with shake tables, ranging from uni-axial tables to six-degree-of-freedom tables to multiple table arrays. These tables are capable of providing interface boundary conditions for substructure real-time hybrid simulation (RTHS). In the simplest case, the lower stories of a shear building are simulated numerically while the upper stories tested experimentally. Even this simple case reveals the challenges of RTHS using shake tables. Shake tables are highly nonlinear devices, making modeling and control a challenging task. Furthermore, the mass of the test specimen is typically large relative to the capacity of the table, leading to substantial coupling of the table and specimen dynamics. These challenges are exacerbated by the loop of action and reaction between numerical and experimental components in RTHS. Any delay or lag in the realization of the desired table trajectory and measurement of the base shear can introduce inaccuracies and instabilities into the loop. This research investigates the challenges of RTHS using shake tables through a simple uni-axial shake table and shear building specimen. A model-based shake table control approach is successfully implemented for online acceleration tracking. A Kalman filter is used to reduce measurement noise in the RTHS loop without introducing phase lag. Numerical and experimental substructures with low damping are selected to demonstrate the robustness of the proposed framework for a challenging RTHS scenario. Even for shake tables with large control-structure interaction and structures with low damping, the proposed framework is robust, reliable, and uses readily available equipment, providing a new experimental tool for laboratories with modest experimental testing capabilities.

Mechanical loading is an essential stimulus for skeletal tissues. Osteocytes are primarily responsible for sensing mechanical stimuli in bone and for orchestrating subsequent responses. This is critical for maintaining homeostasis, and responding to injury/disease. The osteocyte mechanotransduction pathway, and the downstream effects it mediates, is highly complex. In vivo models have proved invaluable in understanding this process. This review summarizes the commonly used models, as well as more recently developed ones, and describes how they are used to address emerging questions in the field. Minimally invasive animal models can be used to determine mechanisms of osteocyte mechanotransduction, at the cell and molecular level, while simultaneously reducing potentially confounding responses such as inflammation/wound-healing. The details of osteocyte mechanotransduction in bone are gradually becoming clearer. In vivo model systems are a key tool in pursing this question. Advances in this field are explored and discussed in this review.

Long chain acyl CoA synthetase (ACSL)-dependent upper small intestinal lipid metabolism activates preabsorptive pathways to regulate metabolic homeostasis, but whether changes in the upper small intestinal microbiota alter specific fatty acid-dependent pathways to impact glucose homeostasis remains unknown. We here first find that upper small intestinal infusion of Intralipid, oleic acid, or linoleic acid preabsorptively increases glucose tolerance and lowers glucose production in rodents. High-fat feeding impairs preabsorptive fatty acid sensing and reduces upper small intestinal Lactobacillus gasseri levels and ACSL3 expression. Transplantation of healthy upper small intestinal microbiota to high-fat fed rodents restores Lactobacillus gasseri levels and fatty acid sensing via increased ACSL3 expression, while Lactobacillus gasseri probiotic administration to non-transplanted high-fat fed rodents is sufficient to restore upper small intestinal ACSL3 expression and fatty acid sensing. In summary, we unveil a glucoregulatory role of upper small intestinal Lactobacillus gasseri that impacts an ACSL3-dependent glucoregulatory fatty acid sensing pathway. Disclosure P.V. Bauer: None. Vascular smooth muscle cells’ primary function is to maintain vascular homeostasis through active contraction and relaxation. In diseases such as hypertension and atherosclerosis, this function is inhibited concurrent to changes in the mechanical environment surrounding vascular smooth muscle cells. It is well established that cell function and extracellular mechanics are interconnected; variations in substrate modulus affect cell migration, proliferation, and differentiation. To date, it is unknown how the evolving extracellular mechanical environment of vascular smooth muscle cells affects their contractile function. Here, we have built upon previous vascular muscular thin film technology to develop a variable-modulus vascular muscular thin film that measures vascular tissue functional contractility on substrates with a range of pathological and physiological moduli. Using this modified vascular muscular thin film, we found that vascular smooth muscle cells generated greater stress on substrates with higher moduli compared to substrates with lower moduli. We then measured protein markers typically thought to indicate a contractile phenotype in vascular smooth muscle cells and found that phenotype is unaffected by substrate modulus. These data suggest that mechanical properties of vascular smooth muscle cells’ extracellular environment directly influence their functional behavior and do so without inducing phenotype switching.

The pubovisceral muscles (PVM) help form the distal maternal birth canal. It is not known why 13% of vaginal deliveries end in PVM tears, so insights are needed to better prevent them because their sequelae can lead to pelvic organ prolapse later in life. In this paper we provide the first quantification of the variation in in vivo viscoelastic properties of the intact distal birth canal in healthy nulliparous women using Fung’s Quasilinear Viscoelastic Theory and a secondary analysis of data from a clinical trial of constant force birth canal dilation to 8 cm diameter in the first stage of labor in 26 nullipara. We hypothesized that no significant inter-individual variation would be found in the long time constant, τ(2), which characterizes how long it takes the birth canal to be dilated by the fetal head. That hypothesis was rejected because τ(2) values ranged 20-fold above and below the median value. These data were input to a biomechanical model to calculate how such variations affect the predicted length of the active second stage of labor as well as PVM tear risk. The results show there was a 100-fold change in the predicted length of active second stage for the shortest and longest τ(2) values, with a noticeable increase for τ(2) values over 1,000 seconds. The correlation coefficent between predicted and observed second stage durations was 0.51. We conclude that τ(2) is a strong theoretical contributor to the time a mother has to push in order to deliver a fetal head larger than her birth canal, but a weak predictor of PVM tear risk.

The responsiveness of glucose sensing per se to regulate whole-body glucose homeostasis is dependent on the ability of a rise in glucose to lower hepatic glucose production and increase peripheral glucose uptake in vivo. In both rodents and humans, glucose sensing is lost in diabetes and obesity, but the site(s) of impairment remains elusive. Here, we first report that short-term high-fat feeding disrupts hypothalamic glucose sensing to lower glucose production in rats. Second, leptin administration into the hypothalamus of high-fat–fed rats restored hypothalamic glucose sensing to lower glucose production during a pancreatic (basal insulin)-euglycemic clamp and increased whole-body glucose tolerance during an intravenous glucose tolerance test. Finally, both chemical inhibition of hypothalamic lactate dehydrogenase (LDH) (achieved via hypothalamic LDH inhibitor oxamate infusion) and molecular knockdown of LDHA (achieved via hypothalamic lentiviral LDHA shRNA injection) negated the ability of hypothalamic leptin infusion to enhance glucose sensing to lower glucose production in high fat–fed rats. In summary, our findings illustrate that leptin enhances LDHA-dependent glucose sensing in the hypothalamus to lower glucose production in high-fat–fed rodents in vivo.

Remarkable changes must occur in the pelvic floor muscles and tissues comprising the birth canal to allow vaginal delivery. Despite these preparatory adaptations, approximately 13% of women who deliver vaginally for the first time (nulliparas) sustain tears near the origin of the pubovisceral muscle (PVM) which can result in pelvic organ prolapse later in life. To investigate why these tears occur, it is necessary to quantify the viscoelastic behavior of the term pregnant human birth canal. The goal of this study was to quantify the in vivo material properties of the human birth canal, in situ, during the first stage of labor and compare them to published animal data. The results show that pregnant human, ovine and squirrel monkey birth canal tissue can be characterized by the same set of constitutive relations; the interspecies differences were primarily explained by the long time constant, τ(2), with its values of 555 seconds, 1110 seconds, and 2777 seconds, respectively. Quantification of these viscoelastic properties should allow for improved accuracy of computer models aimed at understanding birth-related injuries.

The gut microbiota alters energy homeostasis. In parallel, metformin regulates upper small intestinal sodium glucose cotransporter-1 (SGLT1), but whether changes of the microbiota or SGLT1-dependent pathways in the upper small intestine mediate metformin action is unknown. Here we report that upper small intestinal glucose sensing triggers an SGLT1-dependent pathway to lower glucose production in rodents. High-fat diet (HFD) feeding reduces glucose sensing and SGLT1 expression in the upper small intestine. Upper small intestinal metformin treatment restores SGLT1 expression and glucose sensing while shifting the upper small intestinal microbiota partly by increasing the abundance of Lactobacillus. Transplantation of upper small intestinal microbiota from metformin-treated HFD rats to the upper small intestine of untreated HFD rats also increases the upper small intestinal abundance of Lactobacillus and glucose sensing via an upregulation of SGLT1 expression. Thus, we demonstrate that metformin alters upper small intestinal microbiota and impacts a glucose-SGLT1-sensing glucoregulatory pathway.

As America entered the twentieth century, a number of women contributed to the popular elocution movement through their publication of compilations of recitation, dialogues, tableaux, and other elocutionary genres. An examination of woman-authored elocutionary compilations reveals a nascent feminism: Through their selection of pieces that examine women's changing roles and celebrate women's accomplishments—both within and beyond the domestic sphere—women compilers encouraged novice women speakers to rethink their gendered societal roles.

Acquired clitoral enlargement is a rare condition resulting from a variety of etiologies, including tumors and excess androgens. Few cases of nonmalignant schwannoma, a benign tumor of the peripheral nerve sheath, have been reported in the literature as causes of clitoral enlargement in patients without known neurofibromatosis. These painless, slow-growing tumors rarely recur once excised. We present the initial investigation of a patient with a large clitoral schwannoma and subsequent treatment with partial vulvectomy. The workup, including advanced pelvic imaging for diagnosis and surgical planning, as well as removal of the clitoral tumor with preservation of functional tissue and restoration of normal vulvar anatomy despite a large excision, is demonstrated.

Although butterflies are a common topic of study in many early childhood classrooms, integrating art production broadens the scope of the study and allows children to deepen their knowledge and understanding through creative self-expression. This article presents a set of integrated activities that focus on helping children fully grasp the process of metamorphosis by making observations, asking questions, and using visual arts to communicate ideas. In all the activities, children actively engage in discussing the life cycle of the butterfly and using concept-specific vocabulary to explore relationships among the butterflies' different stages, as well as relationships between butterflies and their environment.

Because of the emphasis on high-stakes testing, art has often become a neglected subject. Research indicates that the teaching and integration of art increases academic performance and promotes engagement in other disciplines. Science provides stimulating potential for learning content, practicing observational skills, and expanding students' creativity. Through Gyotaku—an ancient Japanese practice of fish printing—students can learn about biology, fish anatomy and habitats, and elements of art. This article provides teachers with ideas for the integration of art with science in a meaningful and fun way. The authors have facilitated this activity in classrooms from kindergarten to college. The levels of supervision, assistance, and depth of content vary with the age of the learner.

C(4) plants have been reported to have Rubiscos with higher maximum carboxylation rates (kcat(CO(2))) and Michaelis-Menten constants (K(m)) for CO(2) (K(c)) than the enzyme from C(3) species, but variation in other kinetic parameters between the two photosynthetic pathways has not been extensively examined. The CO(2)/O(2) specificity (S(C/O)), kcat(CO(2)), K(c), and the K(m) for O(2) (K(o)) and RuBP (K(m-RuBP)), were measured at 25 degrees C, in Rubisco purified from 16 species of Flaveria (Asteraceae). Our analysis included two C(3) species of Flaveria, four C(4) species, and ten C(3)-C(4) or C(4)-like species, in addition to other C(4) (Zea mays and Amaranthus edulis) and C(3) (Spinacea oleracea and Chenopodium album) plants. The S(C/O) of the C(4) Flaveria species was about 77 mol mol(-1), which was approximately 5% lower than the corresponding value in the C(3) species. For Rubisco from the C(4) Flaverias kcat(CO(2)) and K(c) were 23% and 45% higher, respectively, than for Rubisco from the C(3) plants. Interestingly, it was found that the K(o) for Rubisco from the C(4) species F. bidentis and F. trinervia were similar to the C(3) Flaveria Rubiscos (approximately 650 microM) while the K(o) for Rubisco in the C(4) species F. kochiana, F. australasica, Z. mays, and A. edulis was reduced more than 2-fold. There were no pathway-related differences in K(m-RuBP). In the C(3)-C(4) species kcat(CO(2)) and K(c) were generally similar to the C(3) Rubiscos, but the K(o) values were more variable. The typical negative relationships were observed between S(C/O) and both kcat(CO(2)) and K(c), and a strongly positive relationship was observed between kcat(CO(2)) and Kc. However, the statistical significance of these relationships was influenced by the phylogenetic relatedness of the species.

Evidence continues to emerge detailing the myriad of ways the gut microbiota influences host energy homeostasis. Among the potential mechanisms, short chain fatty acids (SCFAs), the byproducts of microbial fermentation of dietary fibers, exhibit correlative beneficial metabolic effects in humans and rodents, including improvements in glucose homeostasis. The underlying mechanisms, however, remain elusive. We here report that one of the main bacterially produced SCFAs, propionate, activates ileal mucosal free fatty acid receptor 2 to trigger a negative feedback pathway to lower hepatic glucose production in healthy rats in vivo. We further demonstrate that an ileal glucagon-like peptide-1 receptor-dependent neuronal network is necessary for ileal propionate and long chain fatty acid sensing to regulate glucose homeostasis. These findings highlight the potential to manipulate fatty acid sensing machinery in the ileum to regulate glucose homeostasis.

Because levator ani (LA) muscle injuries occur in approximately 13% of all vaginal births, insights are needed to better prevent them. In Part I of this paper, we conducted an analysis of the bony and soft tissue factors contributing to the geometric “capacity” of the maternal pelvis and pelvic floor to deliver a fetal head without incurring stretch injury of the maternal soft tissue. In Part II, we quantified the range in demand, represented by the variation in fetal head size and shape, placed on the maternal pelvic floor. In Part III, we analyzed the capacity-to-demand geometric ratio, g, in order to determine whether a mother can deliver a head of given size without stretch injury. The results of a Part I sensitivity analysis showed that initial soft tissue loop length (SL) had the greatest effect on maternal capacity, followed by the length of the soft tissue loop above the inferior pubic rami at ultimate crowning, then subpubic arch angle (SPAA) and head size, and finally the levator origin separation distance. We found the more caudal origin of the puborectal portion of the levator muscle helps to protect it from the stretch injuries commonly observed in the pubovisceral portion. Part II fetal head molding index (MI) and fetal head size revealed fetal head circumference values ranging from 253 to 351 mm, which would increase up to 11 mm upon face presentation. The Part III capacity-demand analysis of g revealed that, based on geometry alone, the 10th percentile maternal capacity predicted injury for all head sizes, the 25th percentile maternal capacity could deliver half of all head sizes, while the 50th percentile maternal capacity could deliver a head of any size without injury. If ultrasound imaging could be operationalized to make measurements of ratio g, it might be used to usefully inform women on their level of risk for levator injury during vaginal birth.

Despite significant progress in understanding the homeostatic regulation of energy balance, successful therapeutic options for curbing obesity remain elusive. One potential target for the treatment of obesity is via manipulation of the gut-brain axis, a complex bidirectional communication system that is crucial in maintaining energy homeostasis. Indeed, ingested nutrients induce secretion of gut peptides that act either via paracrine signaling through vagal and non-vagal neuronal relays, or in an endocrine fashion via entry into circulation, to ultimately signal to the central nervous system where appropriate responses are generated. We review here the current hypotheses of nutrient sensing mechanisms of enteroendocrine cells, including the release of gut peptides, mainly cholecystokinin, glucagon-like peptide-1, and peptide YY, and subsequent gut-to-brain signaling pathways promoting a reduction of food intake and an increase in energy expenditure. Furthermore, this review highlights recent research suggesting this energy regulating gut-brain axis can be influenced by gut microbiota, potentially contributing to the development of obesity.

The inspiration for this article was not conceived over lunch, but more specifically over the box in which the lunches were packaged. Five colleagues brainstormed ideas for teaching a unit on trees for the third grade that would fit in our boxes. The disciplines involved were art, children's literature, science, social studies, and technology. Each person decorated the lunch box for their appropriate discipline and placed instructional items inside. Technology was infused throughout the unit.

Computer technology can be integrated into science inquiry activities to increase student motivation and enhance and expand scientific thinking. Fifth-grade students used the visual thinking tools in the Kidspira-tion® software program to generate and represent a web of hypotheses around the question, "What affects the distance a marble rolls?" The software encouraged flexible and creative thinking as students generated hypotheses, performed preliminary tests of their ideas, easily edited and revised their web, and created pictorial representations to anchor their thinking. The Kidspiration® software can be used in many different science inquiries and can help teachers meet the unique needs of visual learners as well as ESL students.

In 1876, Julia and Annie Gregory Thomas founded a Conservatory of Elocution, Acting, and Physical Culture in Manhattan, New York, where students learned to train their voices and bodies to become more expressive in acting, reading, and speaking.2 As Thomas and Thomas’s comment reveals, physical training was an integral part of their elocutionary program. They initially developed physical exercises for “pupils who came to the Conservatory for the study of elocution” who had “ailments” or “deformities” that a ected their vocal tone and production. Eventually, however, Thomas and Thomas came to believe that such exercises were not only valuable for correcting “ailments” or “deformities,” but for aiding students’ overall expressive development. Their exercises promoted “harmony of action between the moving part and the part to be moved, or in other words, harmony of action between soul and body” (original emphasis). Such physical exercises would make one “fi t for every duty both mental and bodily.”3
